No campo, foram capturadas ¿mosca-dos-chifres¿ e coletadas as fezes de bovinos tratados (ivermectina) e não tratados (controle). Para avaliar o desenvolvimento da mosca nos grupos, em condição de temperatura laboratorial e ambiental, os ovos das moscas foram obtidos em laboratório e contadas as larvas inoculadas em fezes bovinas. Considerou-se como resultado dos testes, o número de ¿moscas-dos-chifres¿ emergidas, do número de larvas inicialmente inoculadas. O grupo não tratado obteve maior número de moscas, comparativamente ao grupo tratado, tanto em condição de temperatura laboratorial, como em condição ambiental. O tratamento mensal dos bovinos impediu o desenvolvimento da ¿mosca-dos-chifres¿ por até 30 dias em todos os testes, e no tratamento com única dose, as moscas emergiram a partir do 28º dia após sua aplicação. Para avaliação da ação dos artrópodes predadores, competidores e parasitos da ¿mosca-dos-chifres¿ em fezes bovinas, os ovos das moscas foram inoculados apenas em fezes do grupo não tratado. O número de moscas emergidas das fezes expostas no campo, sob a ação dos insetos da coprofauna bovina, foi significativamente menor (p<0,0021), comparativamente às moscas emergidas das fezes protegidas. Os Dípteros simbovinos no município de Araçatuba, SP, foram similares para os períodos, e entre o grupo tratado e não tratado: Cecidomyidae, Chloropidae, Muscidae, Psychodidae, Sepsidae, Sarcophagidae, Sphaeroceridae e Syrphidae, porém o número, a freqüência, a dominância, a diversidade e a similaridade das famílias, foram influenciadas pelo tratamento / Abstract: The influence of coprophagic insects and the treatment with a anti-helmintic drug (ivermectin) on the development of horn fly (Haematobia irritans) (L.1758), inside bovine feces were evaluated, in Araçatuba county (São Paulo State, Brazil), as well as the influence of these factors on the coprophilic entomofauna. Horn flies and feces were collected in the field from the bovines treated with ivermectin and from untreated bovines (control group). To evaluate the development of these flies in the two groups in conditions of controlled and environmental temperatures the fly eggs were collected, counted and inoculated into bovine feces. The numbers of horn fly that emerged from the feces and the number of larvae primarily inoculated were compared. The untreated group showed a higher number of fly counts, when compared to the treated group (were not observation fly), both in controlled temperature, as well in environmental conditions. The treatment at bovines with ivermectin, performed monthly, prevented the development of horn fly for up to 30 days in all the experiments and, after one treatment with the drug, the flies emerged only on the 28th day. To evaluate the effects of the arthropod predators, competitors and parasites of the horn fly in the bovine feces, flies eggs were inoculated into the feces from the untreated group. The number of flies that emerged from the feces exposed to the field, under the action of coprophilic entomofauna, was significantly smaller (p<0,0001), comparatively to those that emerged from the protected feces. The Diptera associated with bovine dung in Araçatuba municipality, were similar in the treated and untreated groups for studied period: Cecidomyidae, Chloropidae, Muscidae, Psychodidae, Sepsidae, Sarcophagidae, Sphaeroceridae e Syrphidae. A peroxidação foi ineficaz independentemente da concentração de H2O2 utilizada (0,5 - 10 mmol L-1). A fotólise e a peroxidação assistida por luz ultravioleta apresentaram eficiências superiores a 98% em 300 s de reação (dose de 1.410 mJ cm-2). No processo de degradação Fenton, quando utilizado 1,0 mmol L-1de Fe(II) e 5,0 mmol L-1 de H2O2, a eficiência máxima foi de 80% em 60 s. No foto-Fenton as degradações atingiram 99 % em 600 s de reação. Na fotocatálise com TiO2 em suspensão houve uma degradação de 90% para uma dose de radiação de 1.410 mJ cm-2, e 97% para 2.820 mJ cm-2. No processo eletroquímico em 60 s de reação a eficiência foi entre 50 e 60%, e em 600 s entre 60 e 70% em todas as condições de densidade de corrente elétrica (13,6 a 40,9 mA cm-2) e concentrações de eletrólito suporte (0,01 a 0,25 mmol L-1 de K2SO4) avaliadas. O processo fotoeletroquímico apresentou eficiência de degradação próxima a 95% em 300 s de reação, independentemente das concentrações de eletrólito suporte e densidades de corrente elétrica empregadas. De forma geral, as reduções da toxicidade aguda, utilizando-se o microcrustáceo Daphnia similis como organismo-teste, foram diretamente proporcionais às eficiências de degradação. Ao final de 600 s não foi observada toxicidade nas soluções submetidas aos processos de fotólise, peroxidação assistida por luz ultravioleta, reagente de Fenton (1,0 e 5,0 mmol L-1 de Fe(II) e H2O2, respectivamente), foto-Fenton (todas as condições avaliadas) e fotocatálise com TiO2/H2O2 (0,12 e 0,01 mmol L-1 de TiO2 e H2O2, respectivamente) / Abstract: Ivermectin degradation (500 ?g L-1) by peroxidation, photolysis, peroxidation assisted by UV radiation, Fenton's reagent, photo-Fenton, photocatalysis with TiO2 in suspension, electrochemical, and photoelectrochemical processes were evaluated. Estudo preliminar da redução da microfilaremia in vivo de Mansonella ozzardi (Manson, 1897) com uso de ivermectina, utilizando a técnica de filtração em membrana de policarbonato, Lábrea, Amazonas, Brasil. Estudou-se a eficácia do uso de dose única de ivermectina 0,15 mg/kg de peso no tratamento de 74 pacientes com infecção por Mansonella ozzardi. Inicialmente foi realizado a coleta de sangue para o diagnóstico parasitológico utilizando a filtração de sangue em membrana de policarbonato e para análises bioquímicas e hematológicas. Foi realizado a quantificação de microfilárias antes e depois do tratamento seguindo-os até 1 ano. Foi observado uma redução estatisticamente significativa em relação à densidade de microfilárias (&#967;2 de Friedman = 159,00; valor-p < 0, 0001) após 1 ano do uso da medicação, e que não houve alterações laboratoriais e efeitos adversos que comprometessem o uso da ivermectina. Concluiu-se que o fármaco nesta dose é eficaz e seguro, e mantém o \"clearance\" de microfilaremia pelo menos 1 ano após o uso. / Preliminary study of the reduction of microfilaremia in vivo of Mansonella ozzardi (Manson, 1897) with use of ivermectin, using the blood filtration polycarbonate membrane technique, in Labrea, Brazilian Amazon, Brazil. The study focused in the efficacy and tolerability of a single-dose use of ivermectin 0.15 mg / kg body weight in the treatment of 74 patients infected by the filarial worm M. ozzardi. Before and after the parasitological diagnosis by a blood filtration polycarbonate membrane and treatment, anamnesis, clinical examination and blood collection for quantification of microfilariae, biochemical and hematological analysis were done, comparing the outcomes of patients the in first day and after 72 hours and at least following up the patients for 1 year. The study concluded that there was a statistically significant reduction in microfilariaemia density (&#967;2 Friedman = 159, 00; p-value <0, 0001) after 1 year of use of the medication, and that there were no laboratory abnormalities and clinical symptoms that compromised the use of ivermectin. In conclusion, the use of ivermectin is an effective microfilaricide and maintains a suppressive effect for at least 1 year and the adverse reactions are not an obstacle for the treatment.

P2X7-Rezeptoren stellen Schlüsselmoleküle bei der Entstehung und Aufrechterhaltung proinflammatorischer Zustände, chronischer Schmerzen sowie der neuroglialen Kommunikation dar. Ihre Aktivität wird durch eine Vielzahl zellbiologischer Mechanismen beeinflusst. Dazu gehört die allosterische Modulation durch extrazelluläre niedermolekulare Stoffe. Die Entwicklung selektiver und potenter P2X7-Modulatoren ist darum Gegenstand intensiver Forschung. Bisher sind jedoch keine Pharmaka für die klinische Anwendung verfügbar. Die Untersuchung zugelassener pharmakologischer Substanzen in einem akademischen Screening erbrachte eine hohe Trefferrate für P2X7-Rezeptoren. In dieser Arbeit wird die P2X7-Wirkung einiger der potentesten allosterischen Modulatoren genauer charakterisiert. Das Antihistaminikum Clemastin stellt dabei einen positiven allosterischen Modulator dar, der den Rezeptor gegenüber niedrigeren ATP-Konzentrationen sensibilisiert. Ivermectin, ein häufig angewendetes Anthelminthikum, konnte als potenzierender Modulator des humanen P2X7-Rezeptors charakterisiert werden. Mit den Phenothiazinen Prochlorperazin und Trifluoperazin zeigen sich schließlich ZNS-gängige Inhibitoren der ATP-induzierten P2X7-Aktivität, die für weiterführende in vivo-Untersuchungen hilfreiche pharmakologische Werkzeuge darstellen können.:1. Usutu virus (USUV) is an emerging mosquito-borne arbovirus within the genus Flavivirus, family Flaviviridae. Similar to the closely related West Nile virus (WNV), USUV infections are capable of causing mass mortality in wild and captive birds, especially blackbirds. In the last few years, a massive spread of USUV was present in the avian population of Germany and other European countries. To date, no specific antiviral therapies are available. Nine different approved drugs were tested for their antiviral effects on the replication of USUV in vitro in a screening assay. Ivermectin was identified as a potent inhibitor of USUV replication in three cell types from different species, such as simian Vero CCL-81, human A549 and avian TME R. A 2- to 7-log10 reduction of the viral titer in the supernatant was detected at a non-cytotoxic concentration of 5 µM ivermectin dependent on the applied cell line. IC50 values of ivermectin against USUV lineage Africa 3 was found to be 0.55 µM in Vero CCL-81, 1.94 µM in A549 and 1.38 µM in TME-R cells. The antiviral efficacy was comparable between the USUV lineages Africa 2, Africa 3 and Europe 3. These findings show that ivermectin may be a candidate for further experimental and clinical studies addressing the treatment of USUV disease, especially in captive birds.
