The master thesis deals with the issue of the careers of the preschool principals after twenty years in the office. The theoretical section describes the needs of principals, their responsibilities, adult education and motivation for their job. It works on the assumption that the determining factor for the very good principal is his/her personal potential which leads to the fulfillment of the competencies which are necessary for his/her office. The research section search answers to the question of how preschool principals after twenty years in the office look at their own career. The qualitative research was used in which the unstructured interviews with preschool principals were conducted. The findings such as needs of principals after twenty years in the office, their abilities to continue to manage school in the changing society, their plans for further professional growth and their motivation for further work after twenty years in the office are described in the conclusion of the master thesis. Vliv fixačních činidel na kvalitu a kvantitu nukleových kyselin v archivovaných vzorcích tkání / The effect of fixatives on DNA quality and quantity in archival tissue specimen

Formaldehyde is widely used fixative. Its advantages are low cost, simplicity of use and good fixation traits, which are fast tissue penetration, good preservation of morphological structures and compatibility with downstream histological applications. Formaldehyde disadvantages are negative effects on nucleic acids. Formaldehyde solutions modify primary structure of deoxyribonucleic acid (DNA), fragment DNA and create protein-DNA covalent bonds that hinder DNA isolation procedures. Level of negative effects of formaldehyde is dependent on many factors. Effect of formaldehyde chemical composition (formaldehyde dilution, presence of buffer or formic acid) and effect of fixation length were studied in this work. On DNA extracted from fixed tissues, DNA quantity and level of DNA fragmentation were studied by quantitative polymerase chain reaction, fluorescence assay for DNA quantification and by on- chip electrophoresis on bioanalyzer Agilent 2100. Quality and quantity of acquired DNA were tested by DNA profile determination for identification purposes using STR (short tandem repeats) analysis. Results show that of all tested fixatives, buffered 4% formaldehyde is the most suited solution in regards of sufficient amount of DNA and sufficient DNA quality. in English Use of trademarks as keywords for searching the Internet The purpose of this thesis is a comprehensive analysis of the use of trademarks as Internet search engine keywords to trigger advertisements that are displayed on search result pages. The main part of the thesis is devoted to legislation and case law in the US. However, for the comparative purposes, I also address developments concerning keyword advertising in the EU. I chose this topic because it combines traditional elements of law with modern technologies. Moreover, despite its significant economic and legal impacts, I consider this topic as insufficiently researched in the Czech Republic. This thesis consists of six chapters. The first chapter is devoted to a general introduction to trademarks, including the history of trademarks, current legal definitions of trademarks as well as legal frameworks of trademarks law. This chapter also focuses on an element of trademark distinctiveness and analyses functions of trademarks. Use of trade marks as internet search engine keywords Main purpose of my thesis is a comprehensive analysis of the issue of trade mark use as internet search engine keywords in relation to displaying advertisements on the internet search engine's websites, mostly from the point of view of the European Union law. However, I briefly address also legislation and jurisdiction in the USA. I chose the topic of my thesis mostly because it is one of the most discussed issues in the field of trade marks in the last few years, not only thanks to its significant economic importance, but also because of the challenge which this issue brought to the law practice. This completely new form of trade mark use was not anticipated by applicable law and it caused many debates regarding basic principles of trade mark protection. The thesis is composed of eight chapters. The first chapter is an introduction. Chapter two provides basic insight into the issue of keywords. - EN The thesis analyzes the idiolect of the then-presidential candidate Barack Obama in the specific speech situation of the televized debates on the background of the utterances of other candidates in the years 2000, 2004 and 2008. The analysis uses corpus-driven methods to compare the Obama corpus with the reference corpus. The comparison is largely based on the analysis of keywords and their use in context, supplemented by the discussion of their collocations and associated clusters. The results of the analysis, i.e. the principal features Obama's idiolect, are presented in a structured summary, divided into specific areas. Descriptive assessment and functional analysis of problem behaviour among individuals with dementia

Functional analysis is a method of behavioural assessment used to determine the environmental variables maintaining a specific behaviour. While functional analysis is often used with individuals with developmental and intellectual disabilities, problem behaviours in people with dementia are rarely assessed using functional analysis methodology. The purpose of the present study was to conduct descriptive and functional analyses of problem behaviours among individuals with dementia to determine: (a) if a descriptive assessment would identify a function of the challenging behaviour assessed; (b) the extent to which stimuli identified in the descriptive assessment were needed to conduct a functional analysis; and (c) if a standard functional analysis is an effective assessment method for this population. The participants were two individuals with dementia who exhibited inappropriate vocalizations, consisting of swearing and delusional speech. For both participants, the results of the descriptive assessments suggested that the function of the behaviour was positive reinforcement in the form of attention from staff members. The findings of the descriptive analyses were confirmed in the functional analyses. The results suggest that a functional analysis informed by a descriptive analysis can identify the function of problem behaviour in individuals with dementia in a residential setting. / May 2016

Abstract The aim of this study is to examine segregation in the three Swedish municipalities of Uppsala, Gävle and Värmdö. The study attempts to describe the current situation in regards to segregation in the chosen municipalities, the reasons for this and what the municipalities can do to prevent further segregation. In an attempt to find answer to these questions, a literature study has been done and three semi structured interviews with planners working for the municipalities in question. The study shows that different socio economic groups are indeed spatially clustered and a big reason for this is historic decisions in regard to large scale planning and Sweden’s historic housing policies. The interviews show that there are many possible tools for a planner to use when trying to decrease segregation. A large focus is put on creating mixed types of housing in each part of the city and municipality. Creating places for social interaction and good communications are also an important part according to all interviewees. The challenges faced by the planners are lack of economic incentive, the limitations of the planning system and preferences of the population.

Les conditions de vie étudiante, avec les nombreux changements occasionnés, peuvent constituer des sources de stress pour les étudiants avec des répercussions non négligeables sur leur santé et leur qualité de vie. Les objectifs de notre recherche sont d’étudier les tracas quotidiens des étudiants de Grandes Écoles en liens avec la santé perçue et la qualité de vie et d’identifier les ressources pouvant jouer un rôle sur leur vécu psychologique. Nos résultats montrent que les étudiants sont principalement préoccupés par l’éloignement géographique, le manque d’indépendance financière, les examens, les difficultés d’organisation ou encore les problèmes de fatigue et de sommeil avec des effets plus ou moins importants sur leur santé mentale. En effet, plus de 40% des étudiants sont en situation de détresse psychologique avec des problèmes de tensions internes, de concentration ou encore de la tristesse. En revanche, la qualité de vie est plutôt préservée même si certains domaines sont affectés par les préoccupations des étudiants. De plus, nos résultats soulignent l’influence des traits de personnalité et de l’estime de soi dans la relation stress / détresse et mettent en avant l’effet pathogène de certains facteurs comme le Névrosisme et le faible niveau d’estime de soi. Ainsi, notre recherche pose la question des capacités d’adaptation des étudiants et soulève l’importance du rôle du psychologue et du corps professoral dans les besoins de soutien et d’accompagnement des étudiants en souffrance psychique. Gonzalo Torrente Ballester (1910-1999), une des figures de proue de la littérature espagnole du XXème siècle, est surtout connu pour ses narrations. Le théâtre constitue pourtant sa grande passion –la première aussi– car il y consacre les vingt premières années de sa carrière littéraire. L’activité théâtrale de l’auteur galicien au cours des décennies 1930 et 1940, intense et soutenue, nous livre un témoignage de choix sur la situation de la scène espagnole pendant la Seconde République : la coexistence d’un théâtre bourgeois et commercial qui jouit de la faveur du public depuis la fin du XXème siècle et d’un théâtre d’avant-garde, minoritaire, qui ambitionne de rénover la scène théâtrale à grand renfort d’innovations formelles. Les deux premières pièces de Torrente Ballester s’inscrivent dans cette dernière démarche mais un événement historique, la Guerre d’Espagne, entraîne un changement esthétique et thématique majeur. Torrente rejoint un groupe d’intellectuels Phalangistes, le Grupo de Escorial, où se mêlent littérature et politique, un amalgame qui conditionne les pièces –mais aussi les essais et les articles– de notre auteur au tout début des années 1940. Le rêve d’une société idéale éduquée par le théâtre tourne court en 1943 et don Gonzalo se détourne vite du dogmatisme qui caractérise la période 1937-1942. L’auteur galicien retrouve alors une liberté créatrice qui lui permet de composer ses meilleures pièces juste au moment où, faute de mise en scène, il décide d’abandonner l’écriture dramatique pour se consacrer exclusivement à la narration. L’aventure théâtrale de Torrente s’arrête à la fin des années 1940 mais l’expérience –littéraire, politique et humaine– accumulée au cours de cette étape se révèle essentielle pour la brillante carrière de romancier qu’il connaît ensuite. / Gonzalo Torrente Ballester (1910-1999), one of the main figures of the Spanish literature of the 20th century, is best known for his narrative. However, theatre is his greatest passion – and also the very first one – since he devoted the first twenty years of his literary career to theatre. The theatrical activity of the Galician author during the 1930s and the 1940s, hectic and steady, gives us an outstanding account of the Spanish stage during the Second Republic: the coexistence of a commercial and bourgeois theatre appreciated by the public since the end of the 19th century, and a minor avant-garde theatre aspiring to reform the theatrical stage with extensive innovations in the form. The two first plays of Torrente Ballester are in the line of this latest approach but a historical event, the war of Spain, leads to a major thematic and aesthetic change. The playwright meets up with a group of intellectuals of the Spanish Phalanx, the Grupo de Escorial, where literature mingles with politics, a mixture which determines the plays – but also the essays and articles – of our author at the very beginning of the 1940s. The dream of an ideal society educated by theatre ends up in 1943 and Torrente turns away from the dogmatism characteristic of the 1937-1942 period. The Galician author recovers then a creative freedom which allows him to compose his best plays just when, in the absence of staging, he decides to give up dramatic writing to dedicate himself exclusively to narrative. The theatrical adventure of Torrente stops at the end of the 1940s but the literary, political and human experience collected during this stage turns out to be essential to his later brilliant career as a novelist.
