En mötesplats med utsikt och ett öppet rum för insikt : En studie av folkbiblioteksarkitektur och synen på folkbiblioteksrummet - Exemplen Lomma bibliotek och Halmstads stadsbibliotek

The focus of this two year master’s thesis in Library- and Information science is the study of two Swedish Public Library buildings with the examples Lomma and Halmstad. The main goal and purpose of the thesis is to try to uncover which views and values of the public library are expressed in the examined libraries; and to what extent these views manifests are shown in its architecture. The theoretical framework is based on Raymond Williams’s cultural concepts of effective, dominant culture and the two extremities of this culture, called residual and emergent cultures. A second aspect is Yi-Fu Tuans concepts of Space and Place. The study also uses a historical background of Swedish Public Library Buildings. By close reading and textual analyses, the empirical study focuses on the municipalities’ documents surrounding the building of the library buildings in Lomma and Halmstad. The architectural perspective is studied through the architects own documents and architectural journals discussing the built Library Buildings. Results show that the technological advances of the time these libraries were built has put them in a position of change, which also is visible in the architecture. The libraries in Lomma and Halmstad function as civic icons of the community and the architecture is very dependent of the places where they are situated. The expressions of the two libraries differ in details but the overall result show that the values exhibited behind the building of the examined libraries is a part of a historical residual culture, functioning as effective and dominant. The conclusion of the thesis is that the expression of the architecture is however a part of an emergent culture. The buildings aim to comprise the values and display the diverse causes in its architecture. The buildings seek to manifest the ideal activity and working of the modern Public Library.

Der Bibliotheksneubau der Hochschule für Technik, Wirtschaft und Kultur Leipzig

Der Neubau der Hochschulbibliothek der Hochschule für Technik, Wirtschaft und Kultur Leipzig (HTWK) ist ein Beispiel für ein durchgeführtes Bauprojekt des Vorhabens „Infrastruktur an Hochschulen“, finanziert mit Mitteln des Europäischen Fonds für regionale Entwicklung. Ziel der Förderung ist unter anderem, die baulichen Voraussetzungen für eine bessere Vernetzung der Forschung innerhalb der Hochschule, aber auch zu anderen Einrichtungen und Unternehmen zu schaffen. Die HTWK Leipzig konzentriert sich an ihrem Hauptstandort im Leipziger Süden auf beiden Seiten der Karl-Liebknecht-Straße. Ausgehend von dieser Zielstellung wurde für die Hochschul - bibliothek ein Neubau geschaffen, der sowohl in seiner Größe als auch in seiner Ausstattung den gewachsenen Ansprüchen an die Literatur- und Informationsversorgung der sich im Campus konzentrierenden Lehre und Forschung gerecht wird.

Umbau und Erweiterung der Görlitzer Stadtbücherei: Aufbruch in eine neue Epoche

Mit einem dicken schwarzen Aktenordner und seiner etwas nüchtern anmutenden Beschriftung: „Umbau/Modernisierung Altbau und Erweiterungsanbau Jochmannstraße“ begann für die Stadtbibliothek nach vielen Jahren der Bauplanung eine neue Epoche: Am 19.12.2006 wurde „Der erste Spatenstich“ auf dem Nachbargrundstück der historischen Volksbücherei und Lesehalle ausgeführt. Diesem Tag gingen viele entscheidende und spannende Momente voraus, unzählige folgten: Lediglich der Stadtratsbeschluss vom Oktober 2005 oder der Tag der Grundsteinlegung im Februar 2007 sollen hier genannt sein. Ein Informations- und Kommunikationszentrum mit besten Nutzungsbedingungen war schon lange ein Wunsch vieler Görlitzer Bürger – und der konnte jetzt in Erfüllung gehen.

Für die Universitätsbibliothek und das Universitätsarchiv Chemnitz wird ein alter Industriebau modernisiert – die Alte Aktienspinnerei. Das Gebäude behält den Charakter eines Fabrikgebäudes aus dem 19. Jahrhundert und wird eine moderne Bibliothek beherbergen, die rund um die Uhr geöffnet haben wird. Es entsteht eine sehr besondere Bibliothek, die nicht nur ein Speicher für gedruckte und elektronische Medien ist, sondern vielmehr eine Atmosphäre bietet, in der sich wissenschaftlich interessierte Menschen treffen, gedruckte und digitale Informationen finden, miteinander diskutieren und neue Ideen entwickeln können. / An old industrial building is being modernised for the University Library and the University Archive of Chemnitz - the Alte Aktienspinnerei. The building retains the character of a 19th century factory building and will house a modern library that will be open around the clock. A very special library will be created, which will not only be a home for printed and electronic media, but will also provide an atmosphere in which people interested in science can meet, find printed and digital information, discuss with each other and develop new ideas.

A very particular library building will be resulting from this process combining historic elements with state-of-the-art technology. The library is supposed to become an inspiring location for everyone who is interested in gaining scientific information, gathering new insights, meeting other people as well as exchange experiences and ideas - and doing this day and night in a 24/7-library.

DNA metabarcoding for the identification of species within vegetarian food samples

>Magister Scientiae - MSc / Aims DNA metabarcoding has recently emerged as a valuable supplementary tool to ensure food authenticity within the global food market. However, it is widely known that highly processed food samples are one of DNA metabarcoding’s greatest shortfalls due to high DNA degradation, presence of PCR inhibitors and the incomplete removal of several undesirable compounds (such as polysaccharides) that makes the amplification of desired DNA challenging. This project has two main aims, the first of which was to determine and develop a cost and time effective DNA metabarcoding system that could successfully describe to species level the ingredient composition of highly processed vegetarian food products. The DNA metabarcoding system was thoroughly evaluated and tested by combining well-researched primers with varying concentrations into a multiplex reaction. The combination of plant and animal primers selected that yielded the best results were used to determine the species composition in the samples. The second aim is to determine the possible presence of meat contaminants within the highly processed vegetarian food samples. Numerous studies have shown that food adulteration is a wide-spread phenomenon throughout the world due to the economic gains it can provide. Animal primers were introduced into the multiplex reaction to aid in the identification of any meat products that could have been inserted into the vegetarian products to lower the overall cost to company. Methodology Thirty-two highly processed vegetarian food samples were collected in the Cape Town area from local and franchised supermarkets. DNA was extracted using the Chloroform/Isoamyl alcohol method best suited for plant-based samples followed by amplification of the following mini-barcoding regions: the mitochondrial 16S ribosomal rRNA, cytochrome B, tRNALeu – trnL – UAA intron and the ribosomal internal transcribed spacer region – ITS2 for plant and fungi identification. The PCR products were purified using the Qiaquick kit and library preparation and building was conducted using the TruSeq DNA PCR-free Library kit. Final purification was completed using AMPure XP kit and the pooled libraries were sequenced on an Illumina Miseq using 300bp paired-end run. Statistical and bioinformatic analysis on the NGS raw sequence reads was performed in R version 3.6.3. Results The results of the data analysis showed that the cytochrome B primer couldn’t detect any animal DNA in the vegetarian samples, however animal-derived sequences were detected in the positives present, validating the efficacy of the multiplex reaction. Mitochondrial 16S ribosomal rRNA was only able to detect plant-based DNA due to the structural homology between chloroplast and mitochondrial DNA. The fungal ribosomal internal transcribed spacer region – ITS2 detected sequences deriving from “Viridiplantae”. This result could have been due to the fungal and plant ribosomal internal transcribed spacer region – ITS2 sharing a reverse primer during amplification. The trnL region was able to detect the presence of undeclared coriander, mustard and wheat in 8 (29%), 6 (21%) and 5 (18%) samples respectively. Additionally, trnL was able to detect the presence of tobacco in 11 (35%) samples. This could have been due to cross-contamination between samples being co-extracted and amplified at the same time for separate studies. The PITS2 region was able to detect the presence of undeclared barley, mustard and wheat in 8 (25%), 4 (14%) and 4 (14%) samples respectively. Our results show the possibility of DNA metabarcoding for the authentication of a wide range of species present in highly processed vegetarian samples using a single assay. However, further optimization of the technique for the identification of both plant and animal species within vegetarian samples needs to be performed before the wide-spread implementation of this technology would be both feasible and viable. Eliminating primer biases, decreasing the risk of homology between different primers in the same assay as well as preventing the amplification of sequencing of undesirable DNA need to be further explored and ultimately mitigated before DNA metabarcoding can be widely seen as an effective and cost-effective method for authentication and food control.
