Nowadays, when environmental concerns are becoming increasingly important are there great interest in natural materials and recyclability. The possibility of reusing materials with maintained mechanical properties are essential for sustainability. Today produced approximately 90,000 tons of natural fiber reinforced composites in Europe of those are 40,000 tons compression molded of which the automotive industry uses 95%. Natural fiber reinforced composites is recyclable and therefore interesting in many applications. Also, natural fiber reinforced composites is inexpensive, light in weight and shows decent mechanical properties which makes them attractive to manufactures. However, the problem with natural fiber reinforced composites is the poor adhesion between fiber and matrix, the sensitivity of humidity and their low thermal stability. Those problems could be overcome by addition of compatibilizer and reactive filler. This study will examine the technical requirement in order to develop a sustainable and recyclable biocomposite. It investigates the composition of matrix (polypropylene), fiber (flax), compatibilizer (maleic anhydride grafted polypropylene) and reactive filler (CaO) in order to obtain various combinations of stiffness, strength and processability. The two main methods used for preparing samples were compounding and injection molding. Results shows that 20 wt% flax was the optimal fiber content and that maleic anhydride grafted polypropylene is a very good compatibilizer by enhancing the strength significant. Surprisingly was the strength impaired due to the addition of CaO. The composition of 20 wt% flax, 1 wt% maleic anhydride grafted polypropylene and 79 wt% polypropylene is the technically most favorable composition.

Prediktor vlivu aminokyselinových substitucí na funkci proteinů / Predictor of the Effect of Amino Acid Substitutions on Protein Function

This thesis discusses the issue of predicting of the effect of amino acid substitutions on protein funkcion, based on phylogenetic analysis method, inspired by tool MAPP. Significant number of genetic diseases is caused by nonsynonymous SNPs manifested as single point mutations on the protein level. The ability to identify deleterious substitutions could be useful for protein engineering to test whether the proposed mutations do not damage protein function same as for targeting disease causing harmful mutations. However the experimental validation is costly and the need of predictive computation methods has risen. This thesis describes desing and implementation of a new in silico predictor based on the principles of evolutionary analysis and dissimilarity between original and substituting amino acid physico-chemical properties. Developed algorithm was tested on four datasets with 74,192 mutations from 16,256 sequences in total. The predictor yields up to 72 % accuracy and in the comparison with the most existing tools, it is substantially less time consuming. In order to achieve the highest possible efficiency, the optimization process was focused on selection of the most suitable (a) third-party software for calculation of a multiple sequence alignment, (b) overall decision threshold and (c) a set of physico-chemical properties.

Predikce škodlivosti aminokyselinových mutací s využitím metody MAPP / Predicting the Effect of Amino Acid Substitutions on Protein Function Using MAPP Method

This thesis discusses the issue of predicting the effect of amino acid substitutions on protein function using MAPP method. This method requires the multiple sequence alignment and phylogenetic tree constructed by third-party tools. Main goal of this thesis is to find the combination of suitable tools and their parameters to generate the inputs of MAPP method on the basis of analysis on one massively mutated protein. Then, the MAPP method is tested with chosen combination of parameters and tools on two large independent datasets and consequently is compared with the other tools focused on prediction of the effect of mutations. Apart from this the web interface for the MAPP method was created. This interface simplifies the use of the method since the user need not to install any tools or set any parameters.

Recycling of Glass Fiber Composites

Composites are the materials which can be used for a wide variety of applications andproducts such as sports equipment, aerospace and marine because of light and stiffnessproperties. Composites are often made from thermoset resin with glass fibers.In this study, two ways of recycling composites were evaluated, which are microwavepyrolysed composites (MGC) and mechanical composites (GC). These glass fibers weregoing to be compounded with Polypropylene (PP) or Maleic Anhydride ModifiedPolypropylene (MAPP) and then injection moulded the sample by Micro-compounder.In order to get better adhesion to the polymer, a coating was added. The Neoxil 5682-polypropylene water emulsion was evaluated.The samples were characterized by Tensile Testing, Thermogravimetric Analysis (TGA),Different Scanning Calorimetry (DSC), and Dynamic Mechanical Analysis (DMA) to find aoptimum combination of recycled glass fiber reinforced polymer.Microwave pyrolysis is a new research area. The glass fiber, polymer oil and gas can beobtained by heating the composite with microwaves to in an inert atmosphere. Målet med detta projekt var att hjälpa IT-Mästaren att kunna fakturera sina kunder på ett smidigare sätt. Deras dåvarande lösning av kundernas dataanvändning var tidskrävande och ineffektivt, därför ville de automatisera systemet. Det nya systemet skulle användas för att administrera kundernas servrar med hjälp av ett grafiskt gränssnitt och sedan kontakta dem för att få fram olika värden för respektive kund som ska presenteras på IT-Mästarens hemsida. På så sätt skulle de kunna fakturera sina kunder på ett mer effektivt sätt och även få bättre överblick av kundernas användning.  I och med utvecklingen utav detta system så fördjupande jag mig dels i egna kända områden men jag lärde mig också nya kunskaper. Det gällde inte enbart teknisk utveckling utan också undersökning av exempelvis hämtningen av alla data som skulle göras. Jag använde utvalda metoder för att kunna utföra detta projekt på ett smidigt och bra sätt. Metoderna beskrivs i rapporten. / The goal of this project was that the IT-Mästaren wanted to be able to bill their customers in a more comfortable way. Their solution of the customer’s server usage of that time was time consuming and inefficient, therefore they wanted to automate the system. The new system should manage their customer’s servers by using a graphical interface and then contact them to get different values for each customer to be presented at the IT-Mästaren's website. In this way they could bill their customers more efficiently and get a better understanding of their customers’ use.  With the development of this system I immersed in my own known areas and I also learned new knowledge. It was not only about technical development but also investigationamong other thingslike retrieval of all the data that should be collected. I used selected methods in order to perform this project in a smooth and effective manner. The methods are described in the report.
