A possibilidade de obtenção de informações relevantes através domonitoramento hormonal não-invasivo foram fatores determinantes para o delineamento deste projeto, que possibilitou um estudo detalhado dos aspectos comportamentais, reprodutivos e endócrinos do sagüi-detufo-preto (Callithrix penicillata), mantido em cativeiro. Utilizamos6 casais desagüi-de-tufopreto (Callithrix penicillata) mantidos cativos e em pares no Centro de Reabilitação de Animais Silvestres - CRAS, São Paulo. Todos os animais tiveram seus comportamentos sócio-sexuais registrados pelo método de amostragem focal por intervalo de tempo com registros a cada 30 segundos em sessões de 30 minutos, totalizando 60 horas para cada animal. Foram coletadas amostras fecais matinais, seis vezes por semana durante 6 meses, a fim de serem extraídos e dosados os metabólitos fecais de estradiol, progesterona e testosterona pelo método de enzimaimunoensaio (EIE). O objetivo deste trabalho foi descrever e analisar os comportamentos sócio-sexuais e suas respectivas associações com as concentrações de metabólitos fecais de esteróides sexuais em machos e fêmeas de sagui-detufo- preto (Callithrix penicillata), nas diferentes fases do ciclo ovariano.Os resultados mostraram a duração do ciclo ovariano de 24,3 ± 4,1 dias e as maiores concentrações de metabólitos fecais de estradiol e testosterona das fêmeas na fase lútea. A ocorrência dos comportamentos sócio-sexuais se mostrou mais frequente na fase peri-ovulatória do que nas fases folicular e lútea. Não houve diferença significativa destes comportamentos entre as fases folicular e lútea. Os metabólitos fecais de testosterona dos machos não apresentou diferença significativa entre as fases do ciclo ovariano das fêmeas. / The possibility of attaining relevant information through a non-invasive hormonal monitoring allow the detailed study of behavioral, reproductive and endocrine profile of captive black tufted marmoset (Callithrix penicillata). Six couples of captive black tufted marmoset (Callithrix penicillata) were housed at the Centro de Reabilitação de Animais Silvestres - CRAS, São Paulo. Socio-sexual behaviors of all animals were registered using the method of focal interval sampling with observations performed each 30 seconds in 30 minutes sessions, in a total of 60 hours per animal. Fecal samples were collected during the morning, six times a week, for 6 months, in order to extract and measure fecal metabolites of estradiol, progesterone and testosterone by enzyme immunoassay (EIE). The objective of the present study was to describe and analyze the socio-sexual behaviors and correlate them with the fecal concentrations of sexual steroids metabolites in paired male and female black tufted marmoset (Callithrix penicillata) in different moments of the ovarian cycle. Results demonstrated that helengthofovariancycleis24. ± 4.1 days. Also, higher concentrations of fecal metabolites of estradiol and testosterone in females are observed during the luteal phase.The occurrence of socio-sexual behaviors were more frequent during the periovulatory phase when compared to the follicular and luteal phases. A síndrome de emagrecimento progressivo (SEP) é responsável por elevada morbidade e mortalidade de calitriquídeos mantidos em cativeiro em diferentes instituições. Essa síndrome representa um desafio aos médicos veterinários por suas características ainda pouco esclarecidas e são poucos os estudos multidisciplinares que visam à avaliação dos diferentes sistemas, como os órgãos linfo-hematopoiéticos. O objetivo foi caracterizar a evolução e duração da SEP, associando os dados clínicos, laboratoriais e anatomopatológicos dos órgãos linfo-hematopoiéticos de saguis naturalmente acometidos por SEP no cativeiro. Foram analisadas as fichas clínicas, necroscópicas e os resultados das amostras de sangue e urina de 47 saguis doentes, Callithrix spp., machos e fêmeas, que foram a óbito devido a SEP e eram provenientes de dois criadouros diferentes do estado de São Paulo. Os fragmentos dos órgãos linfo-hematopoiéticos, intestino delgado e do fígado foram processados e avaliados. Os resultados caracterizaram que a SEP acomete calitriquídeos de espécies diferentes, adultos, sem predisposição sexual, mantidos sob condições estáveis de manejo por em média 42 meses e a duração clínica varia de 41 dias a 1 ano e 7 meses. As características clínicas na fase inicial foram predominantemente sinais gastrintestinais e na fase terminal, sinais gastrintestinais e extra-intestinais. A anemia macrocítica normo ou hipocrômica com policromasia, esferocitose, presença de corpúsculos de Heinz e hemoglobinúria foi a alteração hematológica mais frequente. As lesões dos órgãos linfo-hematopoiéticos foram características de anemia hemolítica ou foram inespecíficas e reacionais caracterizadas por hiperplasia ou depleção das células da medula óssea, baço e linfonodo e lesões degenerativas no fígado. Na SEP, a associação clínica, laboratorial e anatomopatológica possibilitou a caracterização da evolução e duração clínica, da anemia e das alterações dos órgãos linfo-hematopoiéticos, cujas lesões foram consideradas secundárias à desnutrição crônica e progressiva decorrente da severa enterite atrófica. / Wasting marmoset syndrome (WMS) causes high morbidity and mortality of marmosets and tamarins kept in captivity in different colonies. WMS challenges the veterinarian due to its unclear and not established features and there are few multidisciplinary studies that carried out an evaluation of different systems, such the lymphohematopoietic system. The aim was described the duration and evolution of illness based on an association of clinical, laboratory and pathological aspects of WMS. Medical record, laboratory data and pathological findings were analyzed of 47 Callithrix spp., males and females, sick marmosets that died due to WMS in two different colonies in São Paulo state. Tissue samples of small intestine, lymphohematopoietc system and liver were histological processed and evaluated. The results showed that WMS affects adult marmosets of different species; there are no sex-related differences, and the marmosets are at least 42 months under similar general management at colony. The clinical duration of WMS is from 41 days to 1 year and 7 months. The clinical features were gastrointestinal symptoms in the beginning and extra-gastrointestinal and gastrointestinal signs in the end. Normochromic or hypochromic macrocytic anemia with polychromasia, spherocytes, Heinz bodies, and hemoglobinuria is the common hematological result. The lymphohematopoietic system lesions were the common findings of hemolytic anemia or unspecific and reacting features such as hyperplasia or depletion of cell numbers of bone marrow, spleen and lymph node, and degenerative lesions of liver. The clinical, laboratory and pathological association allowed the characterization of evolution and duration of the WMS, the anemia and the lesions of lymphohematopoietic organs which lesions were considered secondary to chronic and progressive malnutrition as a result of severe atrophic.

Improving the welfare of laboratory-housed primates through the use of positive reinforcement training : practicalities of implementation

Whilst there has been a recent increase in interest in using positive reinforcement training for laboratory-housed primates, there remains a reluctance to put into practice training programmes. Much of this reticence seems to stem from lack of expertise in the running of training programmes, and a perception that training requires a large time investment, with concurrent staff costs. The aim of this thesis was to provide practical recommendations for the use of training programmes in laboratories, providing primate users and carestaff with background information needed to successfully implement training programmes whilst improving the welfare of the animals in their care. Training was carried out with two species, cynomolgus macaques (Macaca fascicularis) and common marmosets (Callithrix jacchus) in three different research laboratories to ensure practicability was as wide ranging as possible. Training success and the time investment required were closely related to the primate's temperament, most notably an individual's willingness to interact with humans, in both common marmosets and cynomolgus macaques. Age and sex however had no effect on an individual's trainability. The training of common marmosets was more successful than that with cynomolgus macaques, possibly due to differences in early experience and socialisation. Positive reinforcement training helped both species to cope with the stress of cage change or cleaning, with the monkeys showing less anxiety-related behaviour following the training programme than before. Involving two trainers in the training process did not affect the speed at which common marmosets learned to cooperate with transport box training, but behavioural observations showed that initial training sessions with a new trainer led to animals experiencing some anxiety. This however was relatively transient. Whilst the training of common marmosets to cooperate with hand capture was possible, there seemed little benefit in doing so as the monkeys did not show a reduced behavioural or physiological stress response to trained capture as compared to hand capture prior to training. However strong evidence was found that following both training and positive human interactions the marmosets coped better with capture and stress was reduced. It is recommended that an increased use of early socialisation would benefit laboratory-housed primates, and would also help improve the success of training. Further, the time investment required shows that training is practicable in the laboratory for both species, and that positive reinforcement training is an important way of improving their welfare likely through reducing boredom and fear.

Studies of natural vocalizations, specially in songbirds, have largely benefited from this tool. Here we used IEGs expression to investigate brain areas activated by the hearing of conspecific common marmoset (Callithrix jacchus) vocalizations and/or utterance of antiphonal vocalizations. Nine adult male common marmosets were housed in sound-attenuating cages. Six animals were stimulated with playbacks of freely recorded natural long distance vocalizations (phee calls and twitters; 45 min. total duration). Three of them vocalized in response (O/V group) and three did not (O/n group). The control group (C) was composed by the remaining animals, which neither heard the playbacks nor spontaneously vocalized. After one hour of the stimulation onset (or no stimulation, in the case of the C group), animals were perfused with 0,9% phosphate-saline buffer and 4% paraformaldehyde. The tissue was coronally sectioned at 20 micro meter in a cryostat and submitted to immunohistochemistry for the IEGs egr-1 and c-fos. Marked immunoreactivity was observed in the auditory cortex of O/V and O/n subjects and in the anterior cingulate cortex, the dorsomedial prefrontal cortex and the ventrolateral prefrontal cortex of O/V subjects. In this study, brain areas activated by vocalizations of common marmosets were investigated using IEGs expression for the first time. Our results with the egr-1 gene indicate that potential plastic phenomena occur in areas related to hearing and uttering conspecific vocalizations. / A express?o de genes imediatos tem sido largamente utilizada na investiga??o de ?reas cerebrais ativadas por est?mulos espec?ficos. Thus, in concurrent visual discrimination of objects the animal must learn that only one of the objects will be rewarded. The concurrent visual discrimination is relatively simple, and already been observed Callithrix jacchus is able to accomplish this task. As yet wasn't seen the influence of the qualitative aspects of the rewards, in the performance of concurrent visual discrimination of objects in nonhuman primates, and as in most tests are used isolated animals, the present study had two stages: at first we had as objective to analyze the influence of the caloric value of the reward on the performance in concurrent visual discrimination of objects in isolated animals; in the second, we had the intention analyze performance of C. jacchus in realization of discrimination task in different social contexts, as well as, analyze the influence of previous experience in task performance. In the first stage (Study 1), the animals were not able to discriminate foods that presented small caloric differences . This incapacity in discriminates the rewards was responsible by generating randomness in task of concurrent visual discrimination of objects. In the second stage (Study 2), observed that, independent of social context in which the task was presented, the performance both of the experienced animals as the inexperienced animals tended to randomness. In the first case, is likely that the pattern of responses of the experienced animals is a reflection of their own performance when they were observed in isolation. In the second case, in turn, the randomness was probably due to the small number of sessions. Although present a pattern of performance similar to inexperienced individuals, we verify that the experienced animals monopolize the food consumption when they were in the presence of inexperienced individuals. This was a consequence of the experienced animals have presented lower latency the approximation of apparatus and, consequently, obtain more food. In turn, the inexperienced animals, when were in the presence of experienced, had to adopt alternative strategies to obtain food. Thus, C. jacchus is able to use the previous information he had about the task of solving their own benefit. / A aprendizagem discriminat?ria ? analisada atrav?s de tarefas instrumentais nas quais o indiv?duo ? recompensado por escolher um item ao inv?s de outro. No primeiro caso, ? prov?vel que o padr?o de respostas dos animais experientes seja um reflexo do seu pr?prio desempenho quando foram observados isoladamente. Photoperiod signaling by melatonin in mammals allows behavioral changes along the year, as in the activity-rest cycle, in mood states and in cognitive performance. The aim of this study was to investigate if common marmoset (Callithrix jacchus) exhibits behavioral changes under short and long photoperiods in a 24h cycle, assessing their individual behaviors, vocal repertoire, exploratory activity (EA), recognition memory (RM) and the circadian rhythm of locomotor activity (CRA). Eight adult marmosets were exposed to a light-dark cycle of 12:12; LD 08:16; LD 12:12 and LD 16:08, sequentially, for four weeks in each condition. Locomotor activity was recorded 24h/day by passive infrared motion detectors above the individual cages. A video camera system was programmed to record each animal, twice a week, on the first two light hours. From the videos, frequency of behaviors was registered as anxiety-like, grooming, alert, hanging position, staying in nest box and feeding using continuous focal animal sampling method. Simultaneously, the calls emitted in the experimental room were recorded by a single microphone centrally located and categorized as affiliative (whirr, chirp), contact (phee), long distance (loud shrill), agonistic (twitter) and alarm (tsik, seep, see). EA was assessed on the third hour after lights onset on the last week of each condition. In a first session, marmosets were exposed to one unfamiliar object during 15 min and 24h later, on the second session, a novel object was added to evaluate RM. Results showed that long days caused a decreased of amplitude and period variance of the CRA, but not short days. Short days decreased the total daily activity and active phase duration. On long days, active phase duration increased due to an advance of activity onset in relation to symmetric days. However, not all subjects started the activity earlier on long days. The activity offset was similar to symmetric days for the majority of marmosets. Results of EA showed that RM was not affected by short or long days, and that the marmosets exhibited a decreased in duration of EA on long days. Frequency and type of calls and frequency of anxiety-like behaviors, staying in nest box and grooming were lower on the first two light hours on long days. Considering the whole active phase of marmosets as we elucidate the results of vocalizations and behaviors, it is possible that these changes in the first two light hours are due to the shifting of temporal distribution of marmoset activities, since some animals did not advance the activity onset on long days. Consequently, the marmosets mean decreased because the sampling was not possible. In conclusion, marmosets synchronized the CRA to the tested photoperiods and as the phase angle varied a lot among marmosets it is suggested that they can use different strategies. A atividade locomotora foi registrada continuamente por sensores de movimento acima das gaiolas. Um sistema de c?meras registrou os comportamentos individuais por 2 horas ap?s o in?cio do claro, duas vezes por semana. A partir das filmagens, foi amostrada a frequ?ncia dos comportamentos ansiosos, cata??o, alerta, pendurado, ficar na caixa ninho e comer pelo m?todo Animal Focal Cont?nuo. Os resultados mostraram que os DL diminu?ram a amplitude e a vari?ncia do per?odo do RCA, mas n?o os DC. Os DC causaram uma diminui??o do total di?rio de atividade e da dura??o da fase ativa. Nos DL, a dura??o da fase ativa aumentou devido a um avan?o no in?cio da atividade em rela??o aos dias sim?tricos. Contudo, nem todos os sujeitos iniciaram a atividade mais cedo nos dias longos. O hor?rio de t?rmino da atividade nos DL foi similar aos DS para a maioria dos sujeitos. Os resultados da AE mostrou que os DL e DC n?o afetaram a MR dos saguis e que os DL reduziram a dura??o da AE. Der Weißbüschelaffe (Callithrix jacchus) und das Metabolische Syndrom: Einfluss von Geschlecht und pränataler Programmierung

Das Metabolische Syndrom (MetSyn) ist gekennzeichnet durch eine Kombination verschiedener kardiovaskulärer Risikofaktoren: Glukoseintoleranz, Adipositas, Dyslipidämie sowie arterielle Hypertonie. Es gilt beim Menschen als eine der Hauptursachen für Herzkreislauferkrankungen und befindet sich weltweit auf enormem Vormarsch. Die Weichen für die Erkrankung werden zum Teil schon vor der Geburt durch eine veränderte Umwelt in utero gestellt. So können Stress oder eine Glukokortikoidbehandlung während der Schwangerschaft zu einem veränderten Phänotyp des Embryos/Fetus führen - mit Konsequenzen für das gesamte spätere Leben. Dieses Phänomen wird als pränatale Programmierung bezeichnet. Neben diesen epigenetischen Effekten spielen u. a. auch geschlechtsabhängige Faktoren eine Rolle für das Risiko, am MetSyn zu erkranken. Die vorliegende Arbeit befasst sich mit den Auswirkungen einer Glukokortikoidbehandlung in der frühen Trächtigkeit sowie dem Einfluss des Geschlechts auf kardiovaskuläre Risikofaktoren im Erwachsenenalter. Als Modelltier für die Studie wurde der Weißbüschelaffe eingesetzt. In einem 2002 stattgefundenen Vorversuch im Deutschen Primatenzentrum in Göttingen wurde tragenden Tieren (F0) eine Woche lang täglich oral Dexamethason verabreicht. Dieses synthetische Glukokortikoid kann die Plazentaschranke passieren. Die drei folgenden in Leipzig gehaltenen Generationen DexF1/2/3W (weibliche Tiere, n = 4/6/2) und DexF2/3M (männliche Tiere, n = 2/4) gingen in die Untersuchung ein. Tiere, die keine Nachkommen der F0-Generation darstellten, bildeten jeweils eine weibliche (ControlW, n = 11) und eine männliche (ControlM, n = 15) Kontrollgruppe und wurden ebenfalls herangezogen, um die Auswirkungen des Geschlechts auf die untersuchten Parameter zu ermitteln. Es wurde ein oraler Glukosetoleranztest (OGTT) durchgeführt (inklusive der Erfassung der Insulinwerte), der Quantitative Insulin Sensitivity Check Index (QUICKI – Maß für die Insulinsensitivität) berechnet sowie Lipidstoffwechselparameter bestimmt. Außerdem fanden wöchentlich Erfassungen des Körpergewichts statt. In mehreren Sitzungen pro Tier wurde der Blutdruck gemessen. Die statistische Auswertung erfolgte mittels Mann-Whitney-U-Test für unabhängige Stichproben. Unterschiede mit einer Irrtumswahrscheinlichkeit p ≤ 0,05 wurden als signifikant angesehen. Im OGTT wies DexF1W im Vergleich zu ControlW 120 Minuten nach oraler Glukoseapplikation eine signifikant niedrigere Insulinkonzentration auf. Da nach 30 und 120 Minuten auch die Glukosekonzentration signifikant erniedrigt war, ist jedoch nicht von einer klinischen Relevanz auszugehen. Weitere Auswirkungen der Dexamethasonapplikation auf die F1- bis F3-Generation konnten nicht beobachtet werden. Beim Vergleich der weiblichen und männlichen Nachkommen unbehandelter Weißbüschelaffen fiel auf, dass weibliche Tiere signifikant höhere Insulinkonzentrationen und damit eine signifikant größere Insulin-AUC (Fläche unter der Kurve) im OGTT zeigten. Ihr QUICKI war signifikant niedriger. Hyperinsulinämie und niedriger QUICKI stellen Symptome einer gestörten Glukoseregulation dar. Die weiblichen Tiere zeigten außerdem eine signifikante Erhöhung hinsichtlich Körpergewicht, VLDL-Triglycerid- und folglich Plasmatriglyceridkonzentrationen. Ihre HDL-Cholesterolwerte waren signifikant niedriger. Diese Kombination einer Hypertriglyceridämie mit niedrigem HDL-Cholesterol wird als atherogene Dyslipidämie bezeichnet. Eine gestörte Glukosehomöostase, eine Adipositas sowie eine atherogene Dyslipidämie stellen kardiovaskuläre Risikofaktoren und wichtige Komponenten des MetSyn dar. Zusammenfassend lässt sich sagen, dass beim Weißbüschelaffen eine Glukokortikoidbehandlung während der frühen Trächtigkeit nicht zum MetSyn der F1- bis F3-Generationen im Erwachsenenalter führte. Hingegen ergab die Untersuchung auf ein geschlechtsabhängiges Erkrankungsrisiko eine eindeutige Prädisposition bei den weiblichen Tieren. Die zu Grunde liegenden Mechanismen dieses Phänomens bleiben Gegenstand weiterer Untersuchungen. / The metabolic syndrome (MetSyn) consists of a cluster of metabolic disorders, characterized by glucose intolerance, obesity, dyslipidemia and hypertension. In humans, it is a major cause for cardiovascular disease. Its worldwide prevalence is increasing. The way for the disease can be paved even before birth. An adverse intrauterine environment due to prenatal stress or an iatrogenic overexposure of the fetus to glucocorticoids can lead to an altered phenotype with consequences for later life. This phenomenon is called prenatal programming. In addition gender specific factors play a leading role for the risk of developing MetSyn. The aim of the present study was to investigate the influence of a glucocorticoid application in early pregnancy and gender on cardiovascular risk factors in adulthood. The common marmoset was used as model species. In a preliminary experiment (2002) at the german primate centre (Göttingen) animals (F0) were orally treated with dexamethasone for one week during early pregnancy. Dexamethasone is a synthetic glucocorticoid that can pass the placental barrier. The following three generation offspring, reared in Leipzig, DexF1/2/3W (female animal, n = 4/6/2) and DexF2/3M (male animal, n = 2/4) were regarded. Animals that were no descendants of the F0 generation built a female (ControlW, n = 11) and a male (ControlM, n = 15) control group and were also regarded for gender-specific risk for MetSyn. An oral glucose tolerance test (OGTT) was carried out (including measurements of insulin concentration), the Quantitative Insulin Sensitivity Check Index (QUICKI – measure of insulin sensitivity) was calculated and parameters of lipid metabolism were investigated. Furthermore, all animals were weighed weekly and blood pressure was monitored at a series of meetings. Statistical analysis was performed by Mann-Whitney-U-Test for independent samples. The level of significance was defined at p ≤ 0.05. DexF1W in comparison to ControlW had a significantly lower insulin concentration 120 minutes after glucose application in the OGTT and a significantly lower glucose concentration 30 and 120 minutes after reaching the sugar solution. These findings did not seem to be clinically relevant. Apart from that, no consequences could be determined in the F1-3 generation offspring after dexamethasone treatment in pregnancy. Regarding gender comparison of untreated common marmosets, female animals had significantly higher insulin concentrations in OGTT and therefore a significantly greater insulin AUC (area under the curve). QUICKI was significantly lower. Hyperinsulinemia and a low QUICKI are symptoms of an impaired glucose regulation. Furthermore, the female animals showed an increase in body weight, VLDL triglycerides and therefore total triglycerides. HDL cholesterol was significantly lower. Hypertriglyceridemia in combination with low HDL cholesterol is called atherogenic dyslipidemia. A disturbed glucose homeostasis, obesity and an atherogenic dyslipidemia are cardiovascular risk factors and important components of MetSyn. In summary, dexamethasone applied in early pregnancy did not lead to metabolic syndrome in the F1-F3 generation offspring of common marmoset in adulthood. However, the female gender was associated with a higher risk of developing the disease. The underlying mechanisms require further investigation.

Einfluss von Stress in der Schwangerschaft auf den Fettstoffwechsel weiblicher Folgegenerationen am Primatenmodell Weißbüschelaffe (Callithrix jacchus)

Wie für viele andere Zivilisationskrankheiten werden auch für Atherosklerose und dadurch verursachte Erkrankungen wie Herzinfarkt oder Schlaganfall die Weichen mitunter schon vor der Geburt gestellt. Pränatale oder fetale Programmierung heißt der Mechanismus, durch den negative Umweltbedingungen während der Schwangerschaft, allen voran der Einfluss von Stresshormonen, auf die Entwicklung des Fetus wirken und die Prädisposition für spätere Erkrankungen schaffen können (SCHWAB 2009, SECKL 2001). Ziel der vorliegenden Arbeit war es, die Auswirkungen von Stress während der Schwangerschaft auf den Fettstoffwechsel der Nachkommen unter besonderer Berücksichtigung bekannter Herz-Kreislauf-Risikofaktoren zu untersuchen. Zu diesem Zweck wurde 28 Weißbüschelaffen (F0) während der Trächtigkeit eine Woche lang täglich Dexamethason (DEX) – ein synthetisches Glucocorticoid (GC), welches die Plazentaschranke passieren kann (TEGETHOFF et al. 2009) – oral verabreicht (BEINDORFF et al. 2006, EINSPANIER et al. 2006c). Die drei weiblichen Folgegenerationen DEX F1 (n = 5), DEX F2 (n = 6) und DEX F3 (n = 3) dieser Tiere wurden untersucht, wobei sich die Medikamentengabe auf die F0-Generation beschränkte und alle weiteren Trächtigkeiten ungestört verliefen. Im Alter von 3,3 bis 5,6 Jahren (DEX F1) bzw. von Geburt an bis 1,5 Jahre (DEX F2, DEX F3) wurden die Tiere wöchentlich gewogen. In Blutproben wurden einerseits Fettsäuren (FS), andererseits Cholesterol (CHOL), Triglyceride (TG) und Lipoproteine gemessen, wobei zwei Methoden – enzymatische Analyse nach Ultrazentrifugation und direkter Assay – zum Einsatz kamen. Alle Resultate wurden denen gesunder Kontrolltiere ähnlichen Alters (n = 12) gegenübergestellt. Die Körpermasse unterschied sich zu keinem Zeitpunkt signifikant zwischen den Nachkommen der mit DEX behandelten Tiere und den Kontrollgruppen. Entweder gab es keinen programmierten Effekt auf das Gewicht oder er wurde durch individuelle Schwankungen, möglicherweise verstärkt durch erhöhte Stressempfindlichkeit oder Hyperaktivität der DEX-Nachkommen (FRENCH et al. 2004, SCHWAB 2009) und damit einhergehende Tendenz zur Gewichtsabnahme (KAPLAN und SHELMIDINE 2010) maskiert. Beide Methoden zur Untersuchung des Lipoproteinprofils erschienen für Weißbüschelaffen geeignet und können für zukünftige Untersuchungen empfohlen werden. Bei den Kontrollgruppen fiel auf, dass ältere Tiere u. a. signifikant mehr LDL- und VLDL-CHOL, aber signifikant weniger HDL-TG und n3-FS hatten als jüngere, was auf ein wie beim Menschen mit dem Alter steigendes Herz-Kreislauf-Risiko (CARLSSON et al. 2010) schließen lässt. Sowohl DEX F2 als auch DEX F3 wiesen signifikant höhere Konzentrationen von LDL-CHOL, signifikant niedrigere Werte von HDL-TG, mehr Gesamt-CHOL sowie einen höheren Quotienten CHOL : HDL-CHOL im Blutplasma auf als die Kontrolltiere. Diese Parameter gehören zu den in der humanmedizinischen Diagnostik genutzten Herz-Kreislauf-Risikofaktoren und die Veränderungen weisen auf eine erhöhte Auftrittswahrscheinlichkeit kardiovaskulärer Erkrankungen hin (KANNEL et al. 1994, LUSIS et al. 2004, NCEP 2002). Zusätzlich fielen bei DEX F1, DEX F2 und DEX F3 im Vergleich zu den Kontrollen signifikant erniedrigte Gehalte an n3-FS auf, die u. a. für ihre antiphlogistische und kardioprotektive Wirkung bekannt sind (ALONSO et al. 2003, CALDER 2004, KINSELLA et al. 1990). Pränatale GC-Behandlung rief demzufolge über Veränderungen im Fettstoffwechsel ein erhöhtes Risiko für Herz-Kreislauf-Erkrankungen bei ihren weiblichen Nachkommen F1 bis F3 hervor. The mechanism by which negative environmental circumstances, first of all the influence of stress hormones, can alter the development of the fetus and cause a predisposition for diseases later in life is called prenatal or fetal programming (SCHWAB 2009, SECKL 2001). The aim of the present study was to investigate the consequences of stress during pregnancy on lipid metabolism of the offspring with special regard to known cardiovascular risk factors. Therefore, 28 common marmosets (F0) were given dexamethasone (DEX) – a synthetic glucocorticoid (GC) with the ability to pass the placenta easily (TEGETHOFF et al. 2009) – orally, once daily for one week during gestation (BEINDORFF et al. 2006, EINSPANIER et al. 2006c). The three female filial generations DEX F1 (n = 5), DEX F2 (n = 6) and DEX F3 (n = 3) of those monkeys were investigated. Only the F0 generation was treated with DEX, while all of the following pregnancies remained undisturbed. At the age of 3.3 up to 5.6 years (DEX F1) and from birth until 1.5 years (DEX F2, DEX F3), respectively, the animals were weighed weekly. Blood samples were analyzed on the one hand for fatty acids (FA), on the other hand for cholesterol (CHOL), triglycerides (TG) and lipoproteins using two different methods – enzymatic analysis after ultracentrifugation and direct assay. All results were compared to those of healthy controls of similar age (n = 12). Body mass of the offspring of dams prenatally treated with DEX was not significantly different from that of the controls at any point of time. Either there was no programming effect on weight or it was masked by individual fluctuations, maybe potentiated by hyperactivity or a higher sensitivity to stress of the DEX offspring (FRENCH et al. 2004, SCHWAB 2009) and hence a tendency to loose weight (KAPLAN and SHELMIDINE 2010). Both methods for lipoprotein analysis seemed to be suitable for the common marmoset and can be recommended for future investigations. In the controls, older animals showed significantly more LDL and VLDL CHOL, but significantly less HDL TG and n3 FA than younger ones, which points out to a cardiovascular risk rising with age as in humans (CARLSSON et al. 2010). DEX F2 and DEX F3 had significantly higher concentrations of LDL CHOL, significantly lower levels of HDL TG, more total CHOL and a higher ratio of CHOL : HDL CHOL in blood plasma than the controls. Those parameters are well-known human medicine cardiovascular risk factors and the aberrations detected indicate a higher probability of developing cardiovascular diseases (KANNEL et al. 1994, LUSIS et al. 2004, NCEP 2002). Additionally, compared to the controls, all DEX generations F1 to F3 showed significantly lower levels of n3 FA, which are known for their antiinflammatory and cardioprotective effects amongst others (ALONSO et al. 2003, CALDER 2004, KINSELLA et al. 1990). Consequently, prenatal treatment with GC caused an increased risk for cardiovascular diseases in the female offspring F1 up to F3 via alteration of lipid metabolism.
