Evaluation of Knee Ligament Injuries in Occupants of Heavy Goods Vehicles by Simulating Frontal Impacts using THUMS HBM / Utvärdering av knäligamentskador hos åkande i tunga fordon genom att simulera frontala islag hos THUMS HBM

INTRODUCTION. Frontal collisions have been observed to cause the severe injuries on heavy goods vehicle occupants, and the lower extremities have been frequently injured. Injuries of knee joints are rarely life threatening, however they tend to give long-term consequences. AIM. Evaluate non-lethal frontal impacts towards the knee joint of Total Human Model for Safety (THUMS) v4.0 using a cylindrical barrier. The main objectives are to 1) create local injury risk functions of the knee ligaments restraining frontal impacts, 2) simulate frontal impacts towards the knee joints of THUMS and3) prepare the Hybrid III (HIII)-model for corresponding frontal impacts conducted on THUMS. The intention is for future HIII-simulations to be cross-correlated with the responses from THUMS for the ability to estimate knee ligament strains by investigating impacts on HIII. METHODS. 1) Ligament risk curves of PCL, MCL and LCL were formulated by assembling mean strain threshold values and standard deviations from literature. Virtual values were generated from these pooled strain thresholds, creating the risk curves. 2) THUMS lower body was impacted by a cylindrical steel barrier at four different locations - middle of patella, middle of knee joint, upper tibia and below tibia tuberositas. Four impact velocities ranging from 8-14 km/h were used at each location, giving a total of 16 impacts. 3) The HIII-model was prepared by removing the upper body and inserting the cylindrical steel barrier into the model file. RESULTS. The strain threshold at 50% rupture risk for PCL resulted in 23.6±4.4%, 34.2±6.0% for MCL and 26.6±6.5% for LCL. The simulated THUMS PCL strains reached between 36%-58% for the highest velocity at the impact locations where tibia was involved. Both MCL and LCL gave an approximate 5% strain outcome. The resultant knee displacement for these impacts ranged between 22 mm - 32 mm. The knee displacements at the PCL strain threshold ranged between 14 mm - 16 mm. DISCUSSION and CONCLUSION. Most of the maximal PCL strains exceeded the PCL threshold with large margins. However, the knee displacement at the PCL strain threshold resulted in outcomes comparable to the thresholds used for HIIImodel. These results supported the obtained PCL threshold to be within a reasonable range.

周邊注射安非它命能夠影響動物受試在表現與時間知覺有關的操作式制約行為作業,歷來被研究者認為是大腦多巴胺神經系統與動物時間知覺系統有關的主要證據之一。本研究所共同採用的研究方法為先注射多巴胺受體專屬拮抗劑再於大鼠受試周邊腹腔注射安非它命的方式探討安非它命影響大鼠時間知覺的大腦機制為何。實驗一利用區辨性增強低頻反應作業觀察周邊注射多巴胺受體專屬拮抗劑何者可以反制周邊安非它命對此作業的影響效果,結果發現多巴胺D1受體拮抗劑SCH23390與D2受體拮抗劑raclopride均可反制周邊安非它命的效果。實驗二同樣利用區辨性增強低頻反應作業,但是將SCH23390與raclopride分別注入海馬迴、背側中區紋狀體、腹側側邊紋狀體、依核、內側前額葉皮質以及腹側頂蓋區等六個部位,觀察何種多巴胺受體拮抗劑可在那些大腦部位產生反制周邊安非它命的效果。結果發現SCH23390可在海馬迴、依核、內側前額葉皮質以及腹側頂蓋區等四個部位產生反制周邊安非它命的效果,而raclopride可在腹側側邊紋狀體與內側前額葉皮質兩個部位產生同樣的反制效果。實驗三利用高峰時距作業觀察SCH23390在海馬迴與內側前額葉皮質是否能反制周邊安非它命對此作業的影響效果,結果發現SCH23390僅在海馬迴會影響大鼠受試的正常表現,特別是在與周邊安非它命同時注射的時候。綜合以上結果顯示,周邊注射安非它命能夠使大鼠受試在區辨性增強低頻反應作業當中表現出時間知覺變快的傾向,這個效果需要同時透過大腦內的海馬迴、依核、內側前額葉皮質以及腹側頂蓋區的多巴胺D1類受體和腹側側邊紋狀體與內側前額葉皮質的多巴胺D2類受體。 / The central dopaminergic system has been hypothesized to play a role in time perception based on the results that peripheral injections of d-amphetamine alter the responses in time-related operant conditioned behavioral tasks. The present study investigated the effect by injecting specific dopamine receptor antagonists before peripheral d-amphetamine injections in rats. Data from Experiment I showed that both peripheral the dopamine receptor D1 antagonist SCH23390 and D2 antagonist raclopride could attenuate the response alteration on differential reinforcement of low-rates responding task induced by peripheral d-amphetamine. By using the DRL task, Experiment 2 employed the microjeciton technique to determine the neural substrates for the DA receptor antagonist to attenuate the effect of peripheral d-amphetamine. The infusion sites for DA receptor antagonist were the hippocampus, the dorsomedial striatum, the ventrolateral striatum, the nucleus accumbens, the medial prefrontal cortex, and the ventral tegme ntal area. The results showed that SCH23390 infused into the hippocampus, the nucleus accumbens, the medial prefrontal cortex, the ventral tegmental area could attenuate the effect induced by peripheral d-amphetamine, and such attenuation effects were also observed for raclopride infused into the ventrolateral striatum, the medial prefrontal cortex. Experiment 3 tried to confirm the results of Experiment 2 by microinjecting SCH23390 in hippocampus and medial prefrontal cortex under peak-interval task. Only SCH23390 in the hippocampus altered the subject's normal performance in this task especially when combined with peripheral injection of d-amphetamine. In conclusion, that the response alteration on the DRL task induced by peripheral injection ofd-amphetamine suggests the subject's timing perception being accelerated. These effects of d-amphetamine were mediated by simultaneous activation of multiple dopamine receptor subtypes including D1 receptors located in the hippocampus, nucleus accumbens, medial pref rontal cortex, ventral tegmental area, as well as D2 receptors located in the ventrolateral striatum, medial prefrontal cortex.

The effect of using animated computer 3-D figures illustration in the learning of polyhedron in geometry

This study was carried out to investigate the effect of using animated computer 3-D figures illustration (ACTDFI) in the learning of polyhedron in geometry. By random sampling, intact group of four grade 9 classes in four different schools from a cluster of four educational district schools of Limpopo province in South Africa were selected. The study involved quasi-experimental and inquiry research approaches, the quasi-experimental approach involved pre and posttest design while the inquiry research approach involve classroom observation. There were three experimental groups and a control group with a total of 174 study participants. ACTDFI was used as an intervention for two weeks in the three experimental groups while in the control group, chalk-talk traditional teaching approach was used. Pre-test and post-test was used to collect quantitative data while classroom observation was used to collect qualitative data. A suitable observation checklist was developed for this purpose (Appendix 6 refers). Classroom observation checklist is a list of factors to be considered while observing a class. It gives a structure and framework for the observation. suggested that the use of ACTDFI might have improved academic achievement in learning of polyhedron during the intervention, while the qualitative data analysis indicated that the use of ACTDFI in the experimental groups might have facilitated the learning of the concepts of polyhedron. It is therefore recommended that further research is necessary on the application of ACTDFI in the teaching of 3-dimensional shapes at the primary schools / Mathematics Education / M. Sc. Behavioural and Structural Adaptation to Hippocampal Dysfunction in Humans

Die flexible Anwendung von Wissen in neuen Alltagssituationen ist eine notwendige kognitive Fähigkeit. Bisherige Studien betonen die zentrale Rolle des Hippocampus beim Lernen und Verknüpfen neuer Informationen mit bereits vorhandenem Wissen. Die funktionelle Integrität des Hippocampus ändert sich jedoch im Laufe des Lebens bzw. wird durch neuropsychiatrische Erkrankungen häufig beeinflusst. Die betroffenen Personen müssen deswegen adaptive Strategien entwickeln, um behaviorale Ziele weiter zu erreichen. Daher befasst sich meine Doktorarbeit mit Adaptationsprozessen im sich entwickelnden Gehirn und im vollständig entwickelten Gehirn mit einer hippocampalen Dysfunktion. Diese Synopsis umfasst dazu drei Studien: (1) zu behavioralen Strategien im sich entwickelnden Gehirn, (2) zu behavioralen Strategien im vollständig entwickelten Gehirn nach einer Läsion und (3) zu strukturellen Veränderungen im vollständig entwickelten Gehirn nach einer Läsion. Studie 1 zeigt einen altersgebundenen Wechsel beim assoziativen Gedächtnis: Kinder, Jugendliche und junge Erwachsene benutzen verschiedene Gedächtnisstrategien beim Integrieren von Gedächtnisinhalten. Studie 2 zeigt, dass die beobachteten Gedächtnisbeeinträchtigungen bei Patienten mit rechtsseitigen hippocampalen Läsionen sich nicht alleine durch ein Defizit des assoziativen Gedächtnisses erklären lassen, sondern auf einen zusätzlichen hippocampalen Beitrag zur Gedächtnisintegration zurückzuführen sind. Studie 3 zeigt, dass sich postoperative Adaptationsprozesse auf struktureller Ebene in überraschend kurzer Zeit ereignen und dass die strukturelle Reorganisation nicht nur im Hippocampus, sondern auch in entfernteren Hirnregionen, die mit dem Hippocampus verbunden sind, stattfindet. Zusammenfassend zeigen die Ergebnisse der drei Studien, dass Adaptationsprozesse im sich entwickelnden Gehirn sowie bei Erwachsenen mit einer hippocampalen Dysfunktion sowohl auf der behavioralen als auch auf der strukturellen Ebene auftreten. / Applying knowledge flexibly to new situations is a cognitive faculty that is necessary in every-day life. Previous findings emphasise the crucial role the hippocampus plays in learning and linking new information with pre-existing knowledge. However, the functional integrity of the hippocampus changes over the lifespan and is frequently affected by neuropsychiatric disorders. The affected subjects must, therefore, develop adaptive strategies to achieve behavioural goals. Thus, my doctoral thesis deals with adaptation processes in the developing brain and in adult brains with a hippocampal dysfunction. This synopsis encompasses three studies on: (1) behavioural strategies in the developing brain, (2) behavioural strategies in the lesioned fully developed brain, and (3) structural changes in the lesioned fully developed brain. Study 1 suggests an age-related shift in the associative memory: Children, adolescents, and young adults use different memory strategies when integrating information. Study 2 suggests that the memory deficits observed in patients with right-sided hippocampal lesions are not merely a consequence of an impaired associative memory but rather result from an additional hippocampal contribution to the memory integration. Study 3 suggests that postoperative structural adaptation processes occur on a surprisingly short time-scale, and this structural reorganisation happens not only in the hippocampus but also in distant brain areas connected to the hippocampus. In conclusion, findings from these three studies show that adaptation processes in the developing brain and in adult brains with hippocampal dysfunction occur on both the behavioural and the structural level.

Modulation centrale du fonctionnement cochléaire chez l’humain : activation et plasticité / Central modulation of cochlear functioning in human : activation and plasticity

Le système auditif possède deux particularités. En périphérie, les mécanismes cochléaires actifs (MCA), sous-tendus par la motilité des cellules ciliées externes (CCE), interviennent dans la sensibilité auditive et la sélectivité fréquentielle. Sur le versant central, le système efférent olivocochléaire médian (SEOCM), qui se projette sur les CCE et module les MCA, améliore la perception auditive en milieu bruité. Sur le plan exploratoire, ces deux processus peuvent être évalués grâce aux otoémissions acoustiques provoquées (OEAP) et leur suppression controlatérale. Par ailleurs, des résultats expérimentaux chez l’animal ont montré l’existence d’un rétrocontrôle exercé par le système auditif corticofuge descendant (SACD) sur la cochlée, via le SEOCM.Le présent travail comporte trois études réalisées chez l’humain, visant à explorer les interactions entre SACD, SEOCM et MCA. Les études 1 et 2, utilisant une méthodologie innovante chez des patients épileptiques réalisant une stéréo-électroencéphalographie, ont révélé un effet atténuateur différentiel de la stimulation électrique intracérébrale sur l’amplitude des OEAP, en fonction des modalités de stimulation, ainsi qu’une variabilité de cet effet selon les caractéristiques de l’épilepsie. L’étude 3 a montré un renforcement bilatéral de l’activité du SEOCM chez des musiciens professionnels.Pris dans leur ensemble, ces résultats fournissent d’une part, des arguments directs et indirects en faveur de l’existence d’un SACD fonctionnel chez l’humain. D’autre part, des phénomènes de plasticité à long terme, pathologique ou supranormale, seraient susceptibles de modifier l’activité de cette voie cortico-olivocochléaire. / The auditory system has two special features. At peripheral level, active cochlear micromechanisms (ACM), underlain by motility of outer hair cells (OHC), are involved in auditory sensitivity and frequency selectivity. At central level, the medial olivocochlear efferent system (MOCES), which directly projects onto OHC to modulate ACM, improves auditory perception in noise. From an exploratory point of view, both processes can be assessed through transient evoked otoacoustic emissions (TEOAE) and the procedure of contralateral suppression. In addition, experimental data in animals have disclosed a top-down control exerted by corticofugal descending auditory system (CDAS) on cochlea, via MOCES.The present work comprises three studies carried out in human, aiming to investigate interactions between CDAS, MOCES and ACM. The first and second studies, based on an innovative experimental procedure in epileptic patients undergoing presurgical stereoelectroencephalography, have revealed a differential attenuation effect of intracerebral electrical stimulation on TEOAE amplitude depending on stimulation modalities, as well as a variability of this effect depending on the clinical history of epilepsy. The third study has shown a bilateral enhancement of MOCES activity in professional musicians.Taking together, these results provide direct and indirect evidence for the existence of a functional CDAS in humans. Moreover, possible long-term plasticity phenomenon, either pathological –as in epileptic patients– or supernormal –as in professional musicians– may change cortico-olivocochlear activity.
