Age sensitivity of the Barnes Maze and the Morris Water Maze: Associations with cerebellar cortical Purkinje neurons

The goal of this study was to examine age-related changes in spatial memory and its neurobiological substrates as assessed by the Morris water maze (MWM) and the Barnes maze. The MWM is one of the most widely used tests of spatial memory and numerous studies suggest that spatial memory abilities decline with age. To contrast, very few studies exist comparing different ages of mice in the less stressful Barnes maze, and no systematic life span analysis of performance has been published. As the cerebellum is one brain structure that undergoes a quantifiable change in anatomy across the life span (loss of Purkinje neurons), this study evaluated these behavioral tests in terms of sensitivity to both age-related changes in learning as well as age-related changes in the cerebellar cortex. A total of 65 CB6F1 mice were tested at one of five ages (4, 8, 12, 18, or 25 months) on the MWM and Barnes maze. Deficits in spatial memory acquisition were most apparent in both tasks when comparing a subset of good learning mice. Impairment began at 25 months in the MWM and 18 months in the Barnes maze. In all mice, retention was impaired at 25 months on the Barnes maze, but no clear retention deficits were found in the MWM. Unbiased stereology revealed an age-related loss in cerebellar cortical Purkinje neurons from 12-25 months, whereas hippocampus volume remained stable across the life span. Purkinje neuron number, but not hippocampus volume, was correlated with spatial learning in the Barnes maze, with impaired learning associated with lower neuron numbers. For the MWM, Purkinje neuron number was associated with performance in a subset of good learning animals. Overall, mice that learned the Barnes maze well had more Purkinje neurons than poor learners. Both tests were sensitive to heterogeneity in aging at each age tested. Comparisons of good and poor learners across the life span, as well as correlations between the two tasks and brain measures, suggested that the MWM and Barnes maze may be sensitive to different spatial learning abilities and mechanisms of aging. / Psychology

The development of jazz during the American industrial revolution represents a broader shift in the zeitgeist of the New World. With a rich heritage of rhythmic emphasis in both art and life, African American jazz musicians were able to internalize the increasingly polyrhythmic nature of the metropolis, and groove with the potentially oppressive presence of the machine. Their brazen embrace of the temporal encouraged artists of all media, replacing the burden of permanence and exactitude with the fearlessness of an improvising jazz soloist. The jazz-inspired works of Le Corbusier and Piet Mondrian, for example, explored a synesthetic relationship between the visual and the audible has captured the imagination of the great artists, musicians, architects, and philosophers throughout the history of culture. My thesis exploration attempts to continue this tradition in the context of an increasingly accelerating speed of life, and the new, environmentally sensitive role of the machine. Just as Jazz poeticized the hectic rhythms of the industrial age, I believe that architecture should be conceived of as a synchronizing element within the contemporary urban landscape. Through my design of an adaptive reuse transit hub, my intent was to embrace the temporal in a manner that not only reflects the spirit of the age, but also creates musical architecture. / Master of Architecture

La théorie standard de la consolidation postule que l’information est initialement encodée dans le réseau hippocampo-cortical, créant une trace mnésique au sein de l’hippocampe (HIP). Au cours du temps, la trace est transférée au cortex préfrontal médian (CPFm), et notamment au cortex cingulaire antérieur (CCA). À la suite de lésion des noyaux reuniens et rhomboide (ReRh), réciproquement connectés à l’HIP et au CPFm, le souvenir spatial se forme normalement mais ne persiste pas dans le temps. Ainsi, nous avons évalué l’impact de la lésion ReRh sur la plasticité structurale sous-tendant la persistance du souvenir spatial. Des rats lésés ReRh ont été entraînés en piscine de Morris et testés pour un rappel récent (5j) ou ancien (25j). La plasticité structurale a été évaluée par coloration de Golgi dans l’HIP et le CPFm. La lésion ReRh n'avait aucun effet sur l’apprentissage et le souvenir récent, mais a altéré celui du souvenir ancien. Dans le CA1 des rats Sham, le nombre d'épines dendritiques a été augmenté aux deux délais (5 et 25j) post-acquisition comparé au niveau basal. Après la lésion, cette augmentation n’a pas persisté entre 5 et 25j. Dans le CCA des rats Sham, le nombre d'épines dendritiques a été augmenté uniquement à 25j comparé au niveau de base, une modification non observée chez les rats lésés. Ainsi, à la lésion des noyaux ReRh perturbe la plasticité structurale sous-tendant le souvenir spatial ancien indiquant un rôle crucial de ces noyaux dans l’établissement d’un souvenir persistant. / The standard model of systemic consolidation posits that information is initially encoded in the hippocampo-neocortical network, the memory trace being first created in the sole hippocampus (HIP). Over time, the trace is progressively transferred to modules of the medial prefrontal cortex (mPFC), particularly to the anterior cingulate cortex (ACC). Following lesions of the thalamic reuniens and rhomboid nuclei (ReRh), which are reciprocally connected with both the Hipp and mPFC, a spatial memory forms normally but does not persist (Loureiro et al 2012). Therefore, we assessed the impact of ReRh lesions on structural plasticity underlying spatial memory persistence. Male Long-Evans rats subjected to NMDA lesions of the ReRh nuclei were trained in the Morris Water Maze and tested for retrieval of recent (5 days) or remote (25 days) memory. Structural plasticity was assessed on Golgi-stained material in the HIP and CPFm. ReRh lesions had no effect on learning and recent memory, but altered remote memory. In the HIP (CA1) of sham-operated rats, the spine number was increased at both 5 and 25 days post-acquisition vs baseline. After ReRh lesion, the increase did not persist from 5 to 25 days. In the mPFC (ACC) of sham-operated rats, the spine number was increased only at 25 days vs baseline, a modification not observed in ReRh lesioned rats. Thus, following lesion of ReRh nuclei, structural plasticity underlying remote spatial memory formation does not operate correctly in the mPFC and Hip, pointing to a crucial role of ReRh in memory persistence.

This thesis focuses on the formal and stylistic analysis and comparison of two works written by English authors, namely William Morris' poem The Story of Sigurd the Volsung and the Fall of the Niblungs (1876) and J. R. R. Tolkien's poetic work The Legend of Sigurd and Gudrún (published posthumously in 2009) with respect to how each of these works deals with the original Old Norse motives which they are based on. Both Sigurd the Volsung and The Legend of Sigurd and Gudrún can be described as poetic adaptations of the Old Norse tale of Sigurd Fafnisbani, which is recounted in the Saga of the Volsungs and in a cycle of poems found in the Poetic Edda. Both Morris and Tolkien borrowed this story to use it in their own works, yet each of them treats it in a different manner. Therefore, not only do both of the works differ from the original Old Norse texts on multiple levels, but they also differ one from another. The differences between them can be traced in the metrical properties of the individual poems, for instance, or in the use of specific stylistic elements. From this, it can be inferred that although it was the goal of both authors to evoke the atmosphere of the legendary heroic past where Sigurd's story takes place, each of them attempts to do so in a different way. Cette thèse avait pour objectif d’étudier le rôle du circuit composé de l’hippocampe (Hip), du cortex préfrontal médian (mPFC) et des noyaux reuniens et rhomboïde (ReRh) du thalamus dans les processus cognitifs qui sous-tendent la mémoire spatiale chez le Rat. Nous avons montré que les noyaux ReRh pourraient être impliqués dans la consolidation systémique, mécanisme nécessaire à la persistance des souvenirs et nécessitant un dialogue hippocampo-cortical. Nous avons mis en évidence que l’activité neuronale du mPFC durant le rappel d’une mémoire ancienne dépend des noyaux ReRh, ainsi que l’implication de ces noyaux dans une tâche de mémoire spatiale (dépendante de l’Hip) nécessitant une flexibilité comportementale (impliquant le mPFC). Enfin, nous avons montré un rôle du mPFC dans le rappel d’une mémoire spatiale récente. Ces résultats mettent en évidence l’importance de ce circuit hippocampo-cortico-thalamique dans le traitement et la persistance des informations spatiales chez le Rat. / This thesis aimed to investigate the role of a circuit encompassing the hippocampus (Hip), the medial prefrontal cortex (mPFC) and the reuniens and rhomboid nuclei (ReRh) of the thalamus in cognitive processes underlying spatial memory in rats. We first showed that ReRh nuclei may be involved in systemic consolidation, a mechanism necessary for memory persistence and requiring hippocampal-cortical interactions. We confirmed these findings in a second study showing that mPFC neuronal activity during recall of a remote spatial memory depends on ReRh thalamic nuclei. We also showed the involvement of the ReRh nuclei in a mnemonic task requiring the use of both spatial information (dependent on the Hip) and behavioral flexibility (involving the mPFC). Finally, we found a role of the mPFC in the recall of recent spatial memory. Taken together, these results highlight the importance of a hippocampo-cortico-thalamic circuit in the processing and persistence of spatial information in the Rat.

Etude expérimentale de neurones de Morris-Lecar : réalisation, couplage et interprétation / Experimental study of Morris-Lecar neuron : design, coupling and interpretation

Nous présentons dans ce manuscrit un neurone électronique expérimental basé sur le modèle complet de Morris-Lecar sans simplifications, afin d’obtenir une cellule de base pour étudier l’association collective de neurones couplés. La conception du circuit est donnée en détail selon les différents courants ioniques du modèle. Les résultats expérimentaux sont comparés aux prédictions théoriques, conduisant à un bon accord, ce qui valide donc notre circuit. Nous présentons les différents domaines de bifurcation selon les paramètres de contrôle, la capacité membranaire et le courant d’excitation. Nous avons mis en évidence le comportement du neurone pour chaque zone de paramétrage. Un couplage de ces neurones est introduit en utilisant des simulations Pspice (Multisim) où les neurones ont été conçus pour être les mêmes qu’expérimentalement. Premièrement, nous avons simulé une chaîne fermée de 26 neurones faiblement couplés le long de laquelle les ondes se propagent avec des phases en opposition 2 à 2. Pour cette première étude, on travaille dans une zone présentant uniquement un cycle limite stable. Deuxièmement, une dizaine de neurones sont couplés, avec un choix de paramètres correspondant à une deuxième zone où il y a deux attracteurs, un cycle limite stable et un point fixe stable, tandis qu’entre eux se trouve un cycle instable. Selon le nombre de neurones qui oscillent initialement et les conditions aux bords, nos études montrent que le système évolue vers un état où seuls 1, 2 ou 3 neurones restent à l’état oscillatoire, tandis que les autres sont retournés à un état de repos, ce qui met en évidence un phénomène de clusterisation. Il est à noter que certaines parties de notre circuit de base peuvent ainsi être utilisées dans d’autres modèles de neurones, car ces parties correspondent à la production des divers courants ioniques qu’on retrouve dans d’autres modèles. / We present in this manuscript an experimental electronic neuron based on complete Morris-Lecar model without simplifications, able to become an experimental unit tool to study collective association of robust coupled neurons. The circuit design is given in details according to the ionic currents of this model. The experimental results are compared with the theoretical prediction, leading to a good agreement between them, which therefore validates the circuit. We present the different areas according to the bifurcation control parameters, the membrane capacitance and the excitation current. We have highlighted the behavior of the neuron for each parameters zone. A coupling of such neurons is introduced by using Pspice simulations (Multisim) where neurons have been designed to be the same as the experimental one. First, we simulate a chain of up to 26 neurons weakly coupled along which anti-phase wave patterns propagate with phases in opposition 2 to 2. Second, about ten neurons are coupled, and we succeed to generate clusters based on the boundary conditions of theneurons and their initial conditions. For this study, we work in the region close to the fold bifurcation of limit cycles, where two limit cycles exist, one being stable and another one unstable. Our studies show that the system evolves to a state where only 1, 2 or 3 neurons remain in the oscillatory state, while others returned to a state of rest, which highlights a phenomenon of clustering. The use of some parts of the circuit is also possible for other neuron models, namely for those based on ionic currents.

Rôle de l'acétylation des histones dans différentes formes de mémoire impliquant l'hippocampe et le striatum chez la souris. : effet du vieillissement

Les modifications post-traductionnelles des histones jouent un rôle majeur dans la régulation de l’expression de gènes impliqués dans la plasticité et la mémoire. Parmi ces modifications, l’acétylation des histones permet le maintien de la chromatine dans un état « permissif », accessible pour la transcription. Nos travaux visent à identifier le rôle joué par l’acétylation de deux histones, H3 et H4, dans la formation de différentes formes de mémoire mettant en jeu les systèmes hippocampique et striatal chez la souris. Nous avons également recherché si des perturbations d’acétylation des histones sont responsables des déficits mnésiques observés au cours du vieillissement. Nous avons utilisé deux types d’apprentissage en piscine de Morris permettant de dissocier la mémoire spatiale, impliquant principalement l’hippocampe et la mémoire procédurale/indicée, impliquant le striatum. Nos résultats mettent en lumière une régulation différentielle de l’acétylation des histones dans l’hippocampe et le striatum selon la nature de la tâche et l’âge des animaux. L’apprentissage spatial induit une augmentation de l’acétylation des histones sélectivement dans l’hippocampe (CA1 et gyrus denté) alors que la tâche indicée augmente l’acétylation des histones spécifiquement dans le striatum. Nous montrons également que des changements opposés de l’acétylation de H3 (augmentation) et de H4 (diminution) dans l’hippocampe pourraient contribuer aux déficits de mémoire spatiale observés chez les souris âgées. Lors d’un test de compétition en piscine de Morris, durant lequel les souris ont le choix entre les stratégies spatiale et indicée pour résoudre la tâche, l’injection intra-hippocampique de Trichostatine A (TSA), un inhibiteur des histones déacétylases, immédiatement après l’apprentissage, perturbe la fonction striatale et favorise l’utilisation préférentielle de la stratégie spatiale hippocampique. Cependant, cet effet de la TSA est absent chez les souris âgées dont la fonction hippocampique est altérée. Dans une dernière série d’expérience, l’analyse des effets d’une injection intra-hippocampique de TSA, après un apprentissage spatial, a permis de préciser les contributions respectives des histones H3/H4 et du facteur de transcription CREB dans les déficits mnésiques associés au vieillissement. Dans leur ensemble, nos travaux apportent des éléments importants concernant l’importance de l’acétylation des histones dans la modulation des interactions entre systèmes de mémoire hippocampique et striatal. / Post-translational modifications of histone proteins play a crucial role in regulating plasticity and memory-related gene expression. Among these modifications, histone acetylation leads to a relaxed or “opened” chromatin state, permissive for transcription. Our work aims to identify the role played by histone H3 and H4 acetylation in the formation of different forms of memory involving hippocampal and striatal systems in mice. We also examined whether alterations of histone acetylation are responsible for age-associated memory deficits. We used two versions of the Morris water maze learning task to dissociate a spatial form of memory that relies on the hippocampus and a procedural/cued memory supported by the striatum. Our results highlight a differential regulation of histone acetylation within the hippocampus and striatum depending on the nature of the task and age of animals. Spatial and cued learning elicited histone acetylation selectively in the hippocampus (CA1 region and dentate gyrus) and the striatum, respectively. Age-related spatial memory deficits were associated with opposite changes in H3 acetylation (increase) and H4 (decrease) selectively in the hippocampus. During a water maze competition task in which mice can choose between spatial and cue-guided strategies, intra-hippocampal injection of Trichostatin A (TSA), an histone deacetylase inhibitor, immediately post-acquisition, impaired striatal function and promoted the use of a hippocampus-based spatial strategy. However, this effect of TSA was absent in old mice in which hippocampal function is impaired. In a final series of experiments, analysis of the effects of intra-hippocampal TSA injection immediately after a spatial training helped to clarify the respective contributions of histone H3/H4 and the transcription factor CREB in spatial memory deficits associated with aging. Taken together, our work provides important information regarding the importance of histone acetylation in modulating interactions between hippocampal and striatal memory systems.
