A dirofilariose canina é uma parasitose de distribuição mundial cujo agente etiológico é um verme nemátoda filarióide. Em sua fase adulta, os vermes produzem microfilárias por meio de reprodução sexuada. Sua transmissão ocorre por meio de mosquitos que atuam como hospedeiros intermediários. No Brasil, até o momento, a dirofilariose canina foi encontrada em 15 estados de todas as regiões. A maioria dos estudos está concentrada nas regiões sudeste e sul. A prevalência nacional é de 2%, embora alguns trabalhos tenham encontrado prevalências locais mais elevadas. Neste trabalho apresentamos o primeiro registro de dirofilariose canina para o estado de Rondônia com mapas de distribuição dos cães positivos e análise de mosquitos. Amostras de sangue de 727 cães foram coletadas aleatoriamente na cidade de Porto Velho. As amostras forma analisadas em busca de microfilárias e antígenos circulantes usando duas técnicas: microscopia ótica de gota espessa corada com Giemsa e imunocromatografia de fluxo lateral (ICT). As amostras positivas foram também testadas por PCR, as negativas forma testadas na mesma técnica em pools. Mosquitos foram coletados no domicilio e peridomicílio de todos os casos de cães positivos, estes mosquitos foram testado por PCR em busca de DNA de Dirofilaria immitis. Um mapa de distribuição dos casos de cães positivos foi elaborado. Noventa e três amostras de sangue foram positivas no ICT, representando 12,8% da amostra total, nenhuma amostra foi positiva na gota espessa. O PCR das amostras de sangue resultou em 10% para as positivas no ICT e 0% nas negativas no mesmo teste. Entre os 93 cães positivos, 89 (95,7%) nasceram em Porto Velho. Nenhuma diferença estatística foi observada entre cães que moram em quintais ou intradomicílio. O mapa de distribuição indica um hotspot na região norte da cidade. O PCR dos mosquitos resultou em apenas um pool positivo. A transmissão de dirofilariose canina ocorre na cidade de Porto velho e a frequência que ocorre nos cães é considerada moderada. A técnica de imunocromatografia e PCR são mais eficazes na detecção de dirofilariose comparadas a gota espessa. A confirmação de transmissão de dirofilariose canina em Porto Velho, coloca esta doença no ranking de diagnóstico diferencial de nódulos pulmonares em Porto Velho. / Heartworm is an infectious disease with worldwide distribution. Mosquitoes acts as intermediary host and vectors. In Brazil, until now, heatworm was found in 15 states with an national average of 2%. This present work aims to report for the first time canine heartworm in the state of Rondônia and confirms the transmission of the disease in the state. Blood samples were randomly collected from 727 dogs in the municipality of Porto Velho. The samples were analyzed for the presence of microfilariae and circulating antigens using two different techniques: thick blood microscopy stained with Giemsa and immunochormatography for the detection of filarial antigens. Aiming to test the efficacy of the immunoassay test, all the positive cases were examined by PCR and pools of negative samples were also examined. Mosquitoes were collected at the domiciles presenting positive cases and analyzed by PCR. A distribution map was made with positive cases. Ninety three blood samples out of 727 (12.8%) were positive by the immunoassay technique and none by the thick smear method. All the positive cases by the immunoassay technique were examined by PCR and pools of all negatives samples were also examined resulting in 10% and 0% positivity frequency, respectively. Among the 93 positive dogs, 89 (95.7%) were born in Porto Velho. No differences in the frequency of infection were observed between dogs raised indoor or in the yards. The distribution map indicates a hotspot in the north area of the city. Mosquitoes were analyzed by PCR, resulting in only one positive pool. This result shows that the transmission of canine heartworm is occurring in the municipality of Porto Velho and has moderate prevalence among dogs. The immunoassay technique and the PCR method are more efficient in detecting D. immitis infections in dogs when compared to the blood smear technique. The confirmation of heartworm transmission in Porto Velho also includes this disease among the ranks of differential diagnosis of pulmonary nodules in humans in Rondônia.

Use of Geographic Information System and Remote Sensing Technologies to Describe Mosquito Population Dynamics in the Ray Roberts Greenbelt, Denton County, Texas

A population survey was conducted from April through September 2002 on mosquito species occurring on the Ray Roberts Greenbelt, a riparian corridor used for public recreation on the Elm Fork of the Trinity River, in Denton County, Texas. ArcGIS software was used to set up a stratified random sampling design based on habitat parameters. Multivariate analyses of sampling data and climatic variables were used to describe spatial and temporal patterns of mosquito species. A total of 33 species were collected during this study belonging to the following genera: Aedes, Anopheles, Coquillettidia, Culex, Mansonia, Ochlerotatus, Orthopodomyia, Psorophora, Toxorhynchites, and Uranotaenia. Seasonal distributions of the dominant species revealed population fluctuations. Aedes vexans was the primary species collected in April and May, occurring in low numbers throughout the rest of the sampling period. Psorophora columbiae reached its highest population density in June, with a smaller peak occurring in late July. Present from May through the end of September, Culex erraticus was the most abundant species collected with major peaks in mid-June and the end of July. Abundance of Culex salinarius followed the same general trend as that for Cx. erraticus, but with smaller numbers. The specimens were tested for a variety of arboviruses by the Texas Department of Health. One pool of Cx. erraticus and Cx. salinarius, collected in August 2002, tested positive for West Nile virus. Variables that were important factors for determining dominant species abundance were temperature, wind speed, rain accumulation occurring one-week and two-weeks prior to sampling, number of day since last rain event, dew point, and average canopy coverage.

Os mosquitos (Culicidae) compreendem um grupo monofilético, mas algumas relações dentro da família ainda não estão totalmente resolvidas. O objetivo deste trabalho foi elaborar uma hipótese filogenética para os gêneros de Culicidae baseada nos caracteres de variabilidade genética e perfil proteico. Além disso, foi analisado como a forma da asa evoluiu dentro desse grupo. Utilizou-se 76 espécies diferentes abrangendo 20 gêneros. O formato alar mostrou-se um bom marcador taxonômico, já que as análises em cada tribo ou gênero revelaram grupos naturais. Análises com genes nucleares mostraram Anophelinae como grupo irmão de todos os outros Culicinae. A topologia construída com genomas mitocondriais revelou grupos bem suportados, com alguns discordantes da filogenia atual. Foram identificados 24 biomarcadores nas espécies analisadas usando espectrometria MALDI-TOF, que podem ter potencial na identificação taxonômica. A morfologia, a genética e os perfis proteicos não foram concordantes, e isso pode estar ocorrendo devido a taxas evolutivas distintas entre eles. / Mosquitoes (Culicidae) comprise a monophyletic group, but some relationships within the family are not fully resolved. The aim of this study was to elaborate a phylogenetic hypothesis for the genera Culicidae based on the characters of genetic variability and protein profile. In addition, it was analyzed how wing shape evolved within this group. It was used 76 different species covering 20 genera. The wing shape showed to be a good taxonomic marker, because the analyzes in each tribe or genus revealed natural groups. Analyzes with nuclear genes showed Anophelinae as sister group of all other Culicinae. The topology constructed with mitochondrial genomes revealed well supported groups, with some discordant of the current phylogeny. Twenty-four biomarkers were identified in the species analyzed using MALDI-TOF spectrometry, which may have potential in taxonomic identification. Morphology, genetics, and protein profiles were inconsistent, and this may be occurring due to distinct evolutionary rates between them.

Introdução: Este trabalho tem por objetivo identificar focos de filariose linfática na amazônia ocidental brasileira e constatar a infecção natural pela W. bancrofti do mosquito vetor, o C. quinquefasciatus. O estudo abrangeu as cidades de Porto Velho (RO), Guajará Mirim (RO) e Humaitá (AM). Método: foi utilizada a técnica da gota espessa de sangue, colhida após 22h00, corada com Giemsa para avaliação humana, e a Reação em Cadeia da Polimerase (PCR) para pesquisa de DNA de W. bancrofti em mosquitos vetores. Foram analisadas e avaliadas amostras do sangue de moradores dos bairros mais antigos dessas cidades e escolares noturnas e captura de mosquitos. Resultados: O inquérito hemoscópico em moradores resultou em 935 indivíduos (54,4%) examinados de um total de 1.720 cadastrados. As pesquisas entre escolares noturnos envolveram 2.709 indivíduos (75,2%) de um total de 3.601. Todos negativos para a presença de microfilaremia. Foram coletadas 7.849 fêmeas de C. quinquefasciatus, todas apresentaram resultados negativos para DNA de W. bancrofti. / Introduction: This work is aimed to identify possible lymphatic filariasis foci in the Western Brazilian Amazonian. Porto Velho and Guajara-Mirim (Rondonia State) and Humaita (Amazonas State) were the study target area. Methods: Blood thick film method with samples collected from 10 PM to 1 AM were stained using Giemsa to evaluate human infection and PCR for W. bancrofti DNA in mosquitoes vectors. Samples from the neighborhoods of these areas and from students attending public nighttime classes in the cities referred to above were analyzed and evaluated, as well as mosquitoes captured in the houses. Results: 935 individuals (54.4%) out of a total of 1,720 individuals engaged. Sample with night students involving 2,709 individuals (75.2%) out of the total of 3,601 previously selected. No individual examined was positive for the presence of microfilariae in the blood stream. 7,849 female C.quinquefasciatus specimens were captured and after evaluated by using PCRmethod, all of them were found negative for W. bancrofti DNA.

The mathematical model proposed here considers this seasonality and different treatment intensities accessible to the infected people. The numerical evidence the seasonal fluctuation and the relationship between the environment temperature and treatment efficiency, showing that the temperature increase strongly affects the extrinsic latent period,reducing the healthy care efficiency. Because malaria treatment already exists it should be import. For another hand, even the investment in treatment is an efficient form to block the epidemy, it is not always sufficient, because the protozoan has been more resistent to the medicine; then scientists are creating transgenic mosquitoes refractory to malaria to couple with wild one, generating descending transgenic. To avaliate this situation, we consider here a mathematical model that describes the relatioship between these populations. Then, we formulate and solve an optimal control problem indicating how the transgenic mosquitoes should be introduced in the environment. The numerical simulations show the effectiveness of the control. / Na Amazônia ocorre uma variação na incidência de malária que está intimamente relacionada à variação pluviométrica ao longo do ano. O modelo matemático aqui proposto considera esta sazonalidade e diferentes intensidades de tratamento acessíveis às pessoas infectadas. Experimentos numéricos descrevem a flutuação sazonal e evidenciam uma relação inversa entre a temperatura e eficiência do tratamento, mostrando que um aumento na temperatura afeta fortemente o período latente extrínseco, reduzindo a eficiência do investimento em saúde. Como o tratamento para os infectados existe, é importante concentrar esforços nesse sentido para obter sucesso no controle da malária. Por outro lado, embora o investimento em tratamento seja uma forma eficaz de impedir a epidemia, isso nem sempre é suficiente, pois é fato que o protozoário tem se mostrado cada vez mais resistente aos medicamentos; por esse motivo, cientistas estão criando mosquitos transgênicos refratários à malária que devem acasalar com os mosquitos selvagens, gerando descendência transgência. Para avaliar esta situação, consideramos neste trabalho um modelo matemático que descreve de maneira simplificada a relação entre estas duas populações. A partir desse modelo, formulamos e resolvemos um problema de controle ótimo indicando uma forma adequada de introduzir esses mosquitos transgênicos. Experimentos numéricos mostram a eficácia do controle adotado.

West Nile Virus preparedness in Multnomah County : efficacy, benefits, and limitations of adulticide use for mosquito-borne disease

The objectives of this study were to provide a comprehensive review of the risks and benefits of using adulticides to reduce risk of mosquito-borne disease (particularly West Nile Virus) transmission to humans, as well as to decrease annoyance from nuisance mosquitoes. The study was designed with two major research components, including: 1) an extensive literature review to determine the efficacy of adulticide use, the adverse effects of adulticide use, the impact of mosquitoes on community livability, and the risks and benefits of pesticide use in controlling mosquitoes; and 2) interviews with selected vector districts in seven states to determine effective and ineffective practices in mosquito management. This study has demonstrated that an integrated mosquito management program may be beneficial in reducing risk of disease transmission and mosquito annoyance when performed appropriately. The contribution of adulticiding to reducing mosquito-borne disease transmission, however, is unknown. Research is needed to: 1) further assess the ecological and human impacts of adulticides using the dose and exposure rates realistic to an adulticide program; 2) gain an understanding of the human and ecological impacts of aggregate and cumulative exposures to pesticides, especially for special populations, such as children; and 3) determine the contribution of adulticiding in interrupting or reducing the enzootic amplification of arboviruses, as well as the transmission of WNV to humans. / Graduation date: 2004

Mosquito-borne diseases like malaria, dengue fever, yellow fever and West Nile virus are serious problems in many parts of the world, especially in Africa, and partially in Ethiopia. Millions of people become infected and several hundred thousand people die worldwide from these diseases every year. The most conventional methods for mosquito vector control target female adult mosquitoes in a reactive way using insecticides. However, it is possible to perform proactive vector control through source reduction. The main focus of this study was to investigate if there are any associations between characteristics of managed still water bodies and the presence of mosquito larvae in Bahir Dar, Ethiopia. This was done by measuring twelve variables, both abiotic and biotic; pH, conductivity, dissolved oxygen, turbidity, biochemical oxygen demand, nitrate, phosphate, sulphate, carbonate, depth and presence of algae as well as collecting and counting mosquito larvae in ten different sites, during a time series of five weeks. The most common managed waters believed to serve as efficient breeding habitat for mosquitoes were ponds and ditches used for irrigation, drainage or cultivation. A total of 204 mosquito larvae were collected, where 95% were found in four of the ten sites, three of them were managed waters and the last one was unmanaged. Statistical analyses were performed in order to examine potential correlations and differences among the sites. Wilcoxon test was performed to investigate differences between managed and unmanaged waters. Simple linear regression analyses were performed to identify driving variables for the presence of mosquito larvae. The main results from the group tests were significant differences in mosquito larvae density between managed and unmanaged sites. Significant differences between mosquito sites and zero-sites for dissolved oxygen and sulphate were found. Simple linear regression revealed pH and dissolved oxygen as driving variables for mosquito larvae presence. It was concluded that resources should be put on treatment or manipulation of ponds, which were considered the preferred mosquito breeding habitats. However, if conflicts arise among interests concerning food production, measures must be planned and performed thoughtfully or focus should be put on less controversial breeding habitats. Among the driving variables, dissolved oxygen was the suggested variable to manipulate in order to reduce mosquito larvae populations. / Myggburna sjukdomar som malaria, denguefeber, gula febern och West Nile viruset orsakar allvarliga problem i många delar av världen, särskilt i Afrika, och till viss del Etiopien. Miljontals människor världen över blir smittade och flera hundratusen dör varje år till följd av dessa sjukdomar. De konventionella metoderna för att kontrollera och minska spridningen av myggburna sjukdomar handlar om att kontrollera vuxna myggor med insektsmedel. Det är dock möjligt att utföra en mer förebyggande vektorkontroll genom att minska populationen. Fokus för denna studie var att undersöka om det fanns några samband mellan mygglarvsförekomst och egenskaperna hos mänskligt reglerade eller oreglerade vatten i och kring Bahir Dar i Etiopien. Detta gjordes genom att mäta tolv variabler, både abiotiska och biotiska såsom; pH, konduktivitet, löst syre, turbiditet, biokemisk syreförbrukning, nitrat, fosfat, sulfat, karbonat, djup och algförekomst, samtidigt som mygglarver samlades in och räknades, vilket utfördes på tio olika platser under fem veckor. Den vanligaste typen av reglerade vatten som potentiellt skulle kunna utgöra effektiva mygglarvshabitat ansågs vara dammar och diken som används för bevattning, dränering eller odling. Totalt samlades 204 mygglarver in och 95 % av dem fanns i enbart fyra av de tio etablerade mätplatserna, där tre var reglerade och den sista var oreglerad. Statistiska analyser utfördes för att undersöka potentiella samband och skillnader i mygglarvsförekomst och uppmätta variabler bland de tio mätplatserna. Wilcoxons metod användes för att undersöka om det fanns skillnader mellan reglerade och oreglerade ytvattensamlingars larvförekomst och egenskaper. Enkel linjär regressionsanalys utfördes för att hitta eventuella drivvariabler som därmed anses styra mygglarvsförekomsten. De huvudsakliga resultaten i studien var att det förekom en signifikant skillnad i mygglarvsdensitet mellan reglerade och oreglerade ytvattensamlingar. Signifikanta skillnader i löst syre och sulfat förekom även mellan myggsiter och nollsiter. Enkel linjär regression visade på att pH och löst syre var de mest drivande variablerna för mygglarvsförekomsten i denna studie. Slutsatsen var att dammar utgjorde de mest tilltalande habitaten i samband med mygghonors äggläggning och därför borde prioriteras med avseende på resursfördelning vid planering och utförande av vektorkontroll. Om det skulle uppstå konflikter mellan olika intressen som kan äventyra matproduktionen bör insatser planeras och genomföras med försiktighet eller istället göras i andra mindre kontroversiella mygglarvshabitat. Den drivvariabel som ansågs vara den mest lämpliga att manipulera var löst syre.

The aim of this study was to identify culicid species in a conservation unit located in a preserved area of the Caatinga and the possible associations with local abiotic factors, as well as characterize the natural and artificial ecotopes present in the Caatinga of Rio Grande do Norte state. The Ecological Station of Serido (ESEC-Serid?) is a Caatinga Conservation Unit located in a rural area of Rio Grande do Norte. The hot and dry climate of the Caatinga, along with a short rainy season, favor the adaptation and resistance of plants, resulting in completely distinct landscapes over the course of the year. The study took place over the course of one year, with monthly collections between 10h-12h, 14h-16h and 18h-20h. Adult mosquitoes were collected in a forest area using a manual suction trap with human bait as attractant at the three times and a Shannon trap for night collections. During the rainy season ovitraps were randomly distributed at different sites. The different capture sites were observed and photographed. Immature and adult insects captured were taken to the laboratory for identification. Of the 5081 insects collected in human-bait and Shannon traps, 75% were Mansonia wilsoni. In addition, Aedeomyia, Aedes, Anopheles, Coquillettidia, Culex, Haemagogus, Mansonia and Psorophora were identified. Of the 92 Haemagogus eggs found, 71 were collected on the banks of a reservoir. Mosquito larvae were found and photographed in an artificial breeding site of a neighboring rural area. Humidity had the greatest influence on culicid density in the Caatinga. Rain was important in the appearance of Haemagogus, Anopheles, Aedes and Psorophora. Temperature had a negative influence on Anopheles triannulatus. Moustiques et dirofilariose : mise au point et utilisation d'outils innovants pour la détection et la surveillance / Mosquitoes and dirofilariasis : development and use of innovative tools for detection and monitoring

Dans ce travail, nous nous sommes intéressés à l’étude des dirofilarioses chez le réservoir canin « le chien » ainsi que chez les vecteurs « moustiques », en particulier, en ce qui concerne la détection, la surveillance et la prophylaxie. Le premier objectif était de développer une PCR duplex en temps réel ciblant le gène COI capable de détecter et de différencier simultanément D. immitis et D. repens. Ainsi, nous avons détecté par cet outil moléculaire, pour la première fois en France, D. immitis et D. repens chez des moustiques tigre "Aedes albopictus". Nous avons, de plus, confirmé la présence de l’infection à D. immitis chez les chiens du nord d'Algérie.Le deuxième objectif était d'évaluer l’intérêt de la spectrométrie de masse MALDI-TOF MS pour la détection de changements dans les profils protéiques d'Aedes aegypti infectés expérimentalement avec des nématodes filaires (D. immitis, Brugia malayi et B. pahangi). Les résultats obtenus montrent la capacité du MALDI-TOF MS à différencier des moustiques infectés et non infectés par les filaires. Ainsi, les meilleurs taux de classification correcte obtenus sont 94,1, 86,6, 71,4 et 68,7% pour les non infectés versus ceux infectés, respectivement, par D. immitis, B. malayi et B. pahangi.Le troisième objectif de ce travail était l’évaluation de l'efficacité anti-gorgement et insecticide d'un ectoparasiticide (Vectra® 3D) contenant trois principes actifs : le dinotéfurane, le pyriproxyfène et la perméthrine contre Ae. albopictus, l'une des principales espèces vectrices de Dirofilaria spp. Les résultats ont démontré que le DPP a une efficacité anti-gorgement et insecticide significative contre Ae. albopictus / In this work, we are interested in studying dirofilarial infections in dogs and vectors “Mosquitoes” especially detection, monitoring and prophylaxis. The first objective is to develop a real-time duplex PCR targeting the COI gene capable of simultaneously detecting and differentiating D. immitis and D. repens. Subsequently, we applied this tool to a canine dirofilariosis surveillance process in different endemic areas of Mediterranean basin (Corsica and Algeria). We have thus detected by this molecular tool for the first time in France, D. immitis and D. repens in Aedes albopictus mosquitoes. We reported, also, the presence of D. immitis in dogs from northern Algeria.The second aim was to assess whether the MALDI-TOF MS can detect changes in the protein profiles of Aedes aegypti infected experimentaly with filarial nematodes (D. immitis, Brugia malayi and B. pahangi). Obtained results showed the potential of MALDI-TOF MS as a reliable tool for differentiating non-infected and filariae-infected Ae. aegypti mosquitoes with a best correct classification rate obtained from the thorax-head part with 94.1 and 86.6, 71.4 and 68.7% for non-infected and D. immitis, B. malayi and B. pahangi infected mosquitoes respectively.The third aim of this work has focused on the evaluation of the anti-feeding and insecticidal efficacy of an ectoparasiticide (Vectra® 3D) containing three active ingredients: dinotefurane, pyriproxyfen and permethrin (DPP) against Ae. albopictus. Results demonstrated that the DPP combination has significant anti-feeding and insecticidal efficacy against Ae. albopictus for at least 4 weeks.
