Esta tese foi dividida em três capítulos. O capítulo I descreve o desenvolvimento do método Column switching UHPLC-MS/MS para a determinação simultaneamente de fármacos psicotrópicos em amostras de plasma de pacientes esquizofrênicos. A politerapia é uma prática comum no tratamento da esquizofrenia. Portanto, a monitorização terapêutica destes fármacos tem sido realizada para o ajuste das doses e individualização da terapia farmacológica. O método Column switching UHPLC-MS/MS apresentou linearidade na faixa de concentração de 0,025 a 1,25 ng mL-1 com R2 acima de 0,9950 e a falta de teste de ajuste (p > 0,05); precisão com coeficientes de variação inferiores a 12% e exatidão com erro padrão relativo inferior a 14%. Este método foi aplicado com sucesso para determinação de fármacos em amostras de plasma de pacientes esquizofrênicos para fins de monitorização terapêutica. No capítulo II, o desempenho cromatográfico de colunas C18 superficialmente e totalmente porosas com diferentes tamanhos de partícula foi avaliado para a análise de fármacos psicotrópicos por LC-MS/MS e LC-DAD. Com o sistema LC-MS/MS foram avaliados os seguintes parâmetros cromatográficos: altura do prato reduzido vs velocidade linear reduzida, impedância vs velocidade linear reduzida, tempo da corrida cromatográfica vs vazão, pressão vs vazão, resolução, capacidade de pico, assimetria e fator de retenção. Já com o sistema LC-DAD foram avaliados a hidrofobicidade, atividade silanol e impurezas metálicas também foram avaliadas. As colunas com superfície carregada apresentaram maior eficiência cromatográfica para os fármacos em sua forma ionizada. Já as colunas com partículas menores que 2 µm (Cortecs 1,6 µm, Acquity 1,7 µm, e Kinetex 1,7 µm) apresentaram maior eficiência cromatográfica para os fármacos na forma parcialmente ionizada. Os modelos matemáticos gerados foram capazes de prever a pressão e o tempo da corrida cromatográfica em diferentes vazões para todas as colunas. Considerando a eficiência, impedância, resolução, capacidade de pico, fator de retenção e hidrofobicidade, as colunas Cortecs 1,6 µm e Acquity 1,7 µm apresentaram melhor desempenho durante a análise dos fármacos em amostra de plasma. O capítulo III descreve o desenvolvimento e validação dos métodos SPME-UHPLC-MS/MS e Bio-SPME-Nano-ESI-MS/MS para a determinação dos endocanabinóides (AEA e 2-AG) em amostras biológicas. Para a otimização do processo SPME foram avaliadas as fases SPME (C18, C30 e HLB) e os solventes para dessorção (metanol, acetonitrila e isopropanol). Os aditivos modificadores de matriz, como cloridrato de guanidina, ácido trifluoroacético e acetonitrila foram avaliados por planejamento experimental. Os métodos SPME-UHPLC-MS/MS e Bio-SPME-Nano-ESI-MS/MS, com a fase HLB biocompatível, apresentaram para ambos endocanabinóides valores de LOQs de 1 ng mL-1 e 50 ng mL-1, respectivamente. O método Bio-SPME-Nano-ESI-MS/MS permitiu o direto acoplamento da fibra SPME ao espectrômetro de massas via dessorção/ionização nanoeletrospray que resultou em rápida determinação quantitativa dos endocanabinóides em amostras biológicas. / This thesis is divided into three chapters. Chapter I describes the development of a column switching UHPLCMS/MS method to determine psychotropic drugs in schizophrenic patients plasma samples simultaneously. Polytherapy is a common practice in schizophrenia treatment. Therefore, therapeutic drug monitoring has been applied to adjust doses and to customize pharmacological therapy. The column switching UHPLCMS/MS method developed here is linear at concentrations ranging from 0.025 to 1.25 ng mL-1 with R2 above 0.9950 and presents lack of fit test (p > 0.05), precision with coefficients of variation lower than 12%, and accuracy with relative standard error lower than 14%. This method was successfully applied to determine drugs in schizophrenic patients plasma samples for therapeutic drug monitoring. In chapter II, the chromatographic performance of C18 superficially porous columns and of C18 fully porous columns with different particle sizes were evaluated for analysis of psychotropic drugs by LC-MS/MS and LC-DAD. Within the LC-MS/MS system, the following chromatographic parameters were assessed: reduced plate height vs reduced linear velocity, impedance vs reduced linear velocity, chromatographic run time vs flow rate, backpressure vs flow rate, resolution, peak capacity, asymmetry, and retention factor. Within the LC-DAD system, hydrophobicity, silanol activity, and metal impurities were also examined. Columns with charged surface displayed improved chromatographic efficiency for drugs in the ionized form. Columns with particles smaller than 2 µm (Cortecs 1.6 µm, Acquity 1.7 µm, and Kinetex 1.7 µm) presented higher chromatographic efficiency for the drugs, which were in their partially ionized form. The generated mathematical models were able to predict the backpressure and the chromatographic run time at different flow rates for all the columns. Considering efficiency, impedance, resolution, peak capacity, retention factor, and hydrophobicity, columns Cortecs 1.6 µm and Acquity 1.7 µm provided the best performance during analysis of drugs in plasma samples. Chapter III describes the development and validation of the SPME-UHPLC-MS/MS and the Bio-SPME-Nano-ESI-MS/MS methods for determination of endocannabinoids (AEA and 2-AG) in biological samples. To optimize the SPME process, SPME coatings (C18, C30, and HLB) and solvents for desorption (methanol, acetonitrile, and isopropanol) were evaluated. Matrix modifier additives, such as guanidine hydrochloride, trifluoroacetic acid, and acetonitrile, were assessed by experimental design. The SPME-UHPC-MS/MS and the Bio-SPME-Nano-ESI-MS/MS methods with HLB biocompatible coating provided LOQ values of 1 ng mL-1 and 50 ng mL-1, respectively, for both endocannabinoids. The Bio-SPME-Nano-ESI-MS/MS method allowed direct coupling of SPME fibers to the mass spectrometer by desorption/ionization nanoelectrospray, which resulted in rapid quantitative determinations of endocannabinoids in biological samples.

Contribution à l'étude phytochimique d'orchidées tropicales : identification des constituants d'Aerides rosea et d'Acampe rigida : techniques analytiques et préparatives appliquées à Vanda coerulea et Vanda teres / Contribution to the phytochemical studies of tropical orchids

L’analyse de la composition chimique de deux orchidées de la sous-tribu des Aeridinae, Aerides rosea Lodd. ex. Lindl. & Paxton et Acampe rigida (Buch.-Ham. ex Sm.) P. F. Hunt, a été menée en ayant recours à des techniques de couplages et grâce à une stratégie de déréplication. Dix dérivés phénanthréniques ont ainsi été identifiés dans les tiges d’A. rosea, dont deux nouvellement décrits. Quatre stilbénoïdes, trois dérivés d'acides phénoliques et quatre esters d'acide cinnamique ont été identifiés dans les tiges d’A. rigida. Nous avons également effectué des dosages de traceurs dans différents échantillons de deux représentants de la tribu des Vandeae : Vanda teres (Roxb.) Lindl. et Vanda coerulea Griff. ex. Lindl., afin de mettre en évidence d’éventuelles variations de composition en fonction des facteurs environnementaux et du stade de croissance végétative. Enfin, nous avons mis au point des conditions d’isolement préparatif par chromatographie de partage centrifuge afin de purifier des marqueurs biologiques préalablement identifiés dans les tiges de Vanda teres : trois glucospyranosyloxybenzyl - malates ainsi que leur précurseur biosynthétique. / Phytochemical analyses of the composition of two orchids Aeridinae subtribe, Aerides rosea Lod . ex . Lindl . & Paxton and Acampe rigida (Buch. -Ham . Ex Sm.) P. F. Hunt were conducted using the state-of-the-art hyphenated techniques and a developed dereplication strategy. Ten phenanthrene derivatives have been identified in the stems of A. rosea, including two newly described structures. Four stilbenoids, three phenolic acids derivatives and four cinnamic acid esters were identified in the stems of A. rigida. We also performed quantitative assays of chemical tracers in different samples of two representative species from the Vandeae tribe: Vanda teres (Roxb.) Lindl. and Vanda coerulea Griff. ex. Lindl., in the aim to highlight possible variations in their composition, depending on environmental factors and vegetative growth stage. Finally, we have developed conditions for preparative isolation using centrifugal partition chromatography to purify biomarkers previously identified in the stems of Vanda teres: three glucosyloxybenzyl malates derivatives and their biosynthetic precursor.

Exposition fœtale à différentes familles de xénobiotiques en Bretagne : analyse de la matrice méconium / Fetal exposure to different families of xenobiotics in Brittany, France : analysis of meconium matrix

Le méconium constitue les premières selles du nourrisson. Cette matrice complexe est analysée dans le cadre du projet « PENEW » (Pregnancy Environment and NEWborn malformations) afin de déterminer si le degré d'exposition fœtale aux xénobiotiques joue un rôle dans la survenue de malformations congénitales. Le but de cette étude était le développement analytique pour la détection et la quantification dans le méconium d’une quarantaine de composés de familles différentes (composés organiques volatils, pesticides, éthers de glycol et les métabolites associés). Trois techniques analytiques différentes et une préparation d’échantillon spécifiques ont été développées pour la détection et la quantification de ces composés dans le méconium. L’application des ces méthodes à 246 échantillons de méconium a montré une exposition fœtale à plusieurs des substances recherchées. / Meconium is the earliest stool of newborns. This complex matrix was analyzed through the "PENEW" project (Pregnancy and Newborn malformations Environment) to determine if the degree of fetal exposure to xenobiotics has an influence in the occurrence of birth defects. The objective of this thesis was developed analytical methods for the detection and quantification of several different families of compounds (volatile organic compounds, pesticides, glycol ethers and their metabolites) in meconium. Three different analytical methods and a specific sample preparation have been developed for the detection and quantification of these compounds in the meconium. The application of these methods to 246 meconium samples showed a fetal exposure to several target compounds.

Contamination des wafers et de l'atmosphère des salles blanches de la micro-électronique : développement analytique et étude in-situ / Contamination of wafers and the atmosphere of microelectronic clean rooms : analytical development and field study

La miniaturisation et la complexification croissante des composants microélectroniques induit une sensibilisation et une fragilisation accrue des composants vis-à-vis des contaminations présentes dans les zones de productions appelées “salles blanches”. Dans ces espaces, le contrôle actuel de la contamination organique n’est pas suffisant puisqu’il ne permet pas d’éviter la contamination de surface des plaquettes de silicium et des optiques des robots de production utilisés pour la photolithographie. Un contrôle accru des concentrations des contaminants organiques dans les atmosphères des salles blanches devient donc nécessaire et de nouvelles méthodes analytiques doivent être développées et validées. Dans le cadre de ce travail, des méthodes d’analyse ont été développées et validées afin de disposer d’une gamme d’outils permettant un suivi rigoureux des contaminations. Ces outils permettent d’identifier et de quantifier les contaminations surfaciques des plaquettes de silicium par des composés organiques semi-volatils (phtalates et organophosphorés) mais aussi de déterminer les concentrations de composés organiques volatils présents dans l’atmosphère des salles blanches. Ces méthodes font appel aux technologies du WOS/ATD-GC-MS « Wafer Outgassing System/Automated Thermal Desorber–Gas Chromatography–Mass Spectrometry » et de la DART-ToF-MS « Direct Analysis in Real Time-Time of Flight–Mass Spectrometry » pour les analyses de surfaces et au PTR-ToF-MS « Proton Transfer Reaction – Time of Flight - Mass Spectrometry » pour l’analyse de l’atmosphère. / The recent advances in the miniaturization and complexification of microelectronic components induce an increase in the sensitivity of these components regarding the organic contamination present in the production zone called “clean room”. Although, the control of organic contamination in the clean room is very rigorous it does not avoid the contamination of silicon wafer surfaces and robot lenses used in the photolithography process. The later implies that new analytical methodologies should be developed and validated. In this work, analytical methods were developed and validated in order to have a panel of tools which allows careful monitoring of organic contaminants. These tools allow the identification and quantitation of the contamination of silicon wafer surface by semi-volatiles organic compounds (phthalates and organophosphates) and the determination of volatile organic compounds concentrations in the clean room atmosphere. These methods uses the WOS/ATD-GC-MS « Wafer Outgassing System/Automated Thermal Desorber–Gas Chromatography–Mass Spectrometry » technology and the DART-ToF-MS « Direct Analysis in Real Time-Time of Flight–Mass Spectrometry » technology for wafer surface analysis and the PTR-ToF-MS « Proton Transfer Reaction – Time of Flight - Mass Spectrometry » technology for gas-phase analysis.

Introduktion Antibiotikaresistens är ett globalt växande problem. Till gruppen β-laktamantibiotika hör piperacillin-tazobaktam och cefotaxim som båda verkar genom att försvaga cellväggen med kovalenta bindningar till peptidoglykanlagret som lyserar cellen. E. coli och K. pneumoniae tillhör gruppen Enterobacteriaceae, som är en del av den humana tarmfloran och ofta förekommande vid urinvägsinfektion och sepsis. Utvidgat Spektrum β-Laktamas (ESBL) är ett enzym som finns hos Enterobacteriaceae och som hydrolyserar β-laktamantibiotika. Matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF MS) är en kvalitativ analysteknik för detektion av kemiska föreningar i avseende på massa och laddning. Kännedom om antibiotikametaboliters molekylvikt vid hydrolys möjliggör detektion. Liquid Chromatography Tandem Mass Spectrometry (LC-MS/MS) är en högsensitiv kvantifieringsmetod som separerar molekyler i avseende på polaritet för vidare detektion i avseende på massa och laddning. Syfte Syftet med denna studie var att vidareutveckla en snabb och effektiv metod för att påvisa nedbrytning av piperacillin-tazobaktam och cefotaxim i blodplasma med LC-MS/MS. Material och Metod Tiofaldigt sjunkande koncentrationer av piperacillin-tazobaktam från 2000 till 2 µg/ml, och cefotaxim med koncentrationerna 500 till 0,5 µg/ml analyserades med MALDI-TOF MS, dels intakt men även med bakterierna E. coli och K. pneumoniae med uttryck av olika resistensmekanismer. Vid optimerade koncentrationer spikades plasmaprover med nedbrutet antibiotika som sedan kvantifierades med LC-MS/MS. Resultat Lägsta detektionsgräns med MALDI-TOF MS för intakt och hydrolyserat piperacillin-tazobaktam var 20/2,5 µg/ml. För cefotaxim var lägsta gränsen 5 µg/ml. Med kliniskt relevanta blodkoncentrationer gick hydrolys inte att detektera för. Med tre bakteriekolonier/50 µl kunde dock hydrolys detekteras och kvantifieras med LC-MS/MS. Slutsats Detektion av β-laktamantibiotika är möjligt med både MALDI-TOF MS och LC-MS/MS. För att påvisa hydrolys krävdes större mängder bakterier än förväntat med LC-MS/MS. / Introduction Antibiotic resistance is a global growing problem. Piperacillin-tazobactam and cefotaxime are parts of the group β-lactam antibiotics. The common feature is to inhibit the cell wall synthesis by covalent bindings to the peptidoglycan layer and thereby causing lysis of the bacterial cell. E. coli and K. pneumoniae are members of the Enterobacteriaceae which is a part of the human normal flora but also are commonly associated with urinary tract infections which sometimes develops into to sepsis. Extended Spectrum β-Lactamases (ESBLs) are enzymes with hydrolytic abilities acting on β-lactam antibiotics, expressed by Enterobacteriaceae. The qualitative, Matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF MS) can be used to detect chemical compounds in the ratio of mass to charge in accordance to their molecular weight. Liquid Chromatography Tandem Mass Spectrometry (LC-MS/MS) is a highly sensitive two-step method of quantification which first separate molecules by their polarity attraction force and then by the ratio of mass to charge. Aim The aim of this study was to develop a fast and efficient method to determine degradation of piperacillin-tazobactam and cefotaxime in blood plasma by LC-MS/MS. Method Tenfold dilution of piperacillin-tazobactam in concentrations of 2000 to 2 µg/ml, and cefotaxime in concentrations of 500 to 0,5 µg/ml where analyzed by MALDI-TOF MS, intact and also with the bacteria E. coli and K. pneumoniae with different expression of antibiotic resistance. Optimized concentrations where fixed in blood plasma and then quantified by LC-MS/MS. Result The detection limit by using MALDI TOF MS of hydrolyzed as well as non-hydrolyzed piperacillin-tazobactam was 20/2,5 µg/ml. The detection limit in cefotaxime was 5 µg/ml. Hydrolysis could not be detected in clinically fixed blood concentrations. Detection and quantification of hydrolysis by LC-MS/MS was possible in a concentration of three bacteria colonies/50 µl. Conclusion It is possible to detect hydrolysis in both MALDI TOF MS and LC-MS/MS. A larger amount of bacteria than expected was needed to demonstrate hydrolysis In LC-MS/MS.

Detection and Characterisation of Nanoparticles using Inductively Coupled Plasma Mass Spectrometry

In dieser Doktorarbeit wurde eine analytische Methode zur Charakterisierung metallischer Nanopartikel (NPs) entwickelt und die Methode bei der Untersuchung natürlicher Proben angewendet. Mit einem analytisches System bestehend aus einem Mikrotropfengenerator (microdroplet generator, MDG) zusammen mit einem pneumatischen Zerstäuber und einem induktiv gekoppeltem Plasma-Massenspektrometer (ICP-MS) konnte eine quantitative und qualitative Charakterisierung von NPs durchgeführt werden. Der MDG wurde verwendet um die Kalibrierungsfunktion für die massenspektrometrische Quantifizierung der Metalle in den Nanopartikelproben, die über den pneumatischen Zerstäuber eingeführt wurden, einzurichten. Der Hauptvorteil dieser Anordnung besteht darin, dass mit dem MDG für jedes Metall Tropfen einer gewünschten Größe hergestellt werden können und eine 100 %-ige Transporteffizienz gegeben ist. Die eingeführte Masse korrelierte mit der Signalintensität von Nanopartikeln, so dass die mit dem MDG generierten Tropfen für die Kalibrierung verwendet werden konnten ohne dass Referenzmaterial erforderlich war. Die aufwändige und fehleranfällige Bestimmung der Effizienz eines Zerstäubers, die für die Bestimmung des Metallgehaltes von NPs mittels eines Einzelpartikel-ICP-MS (spICP-MS) erforderlich ist, konnte dadurch vermieden werden. Unter Anwendung dieser dualen Einführungsmethode wurden Größen und Konzentrationen einer Reihe von Standard Silber (Ag) NPs und Referenz Gold (Au) NPs mit hoher Genauigkeit bestimmt. Zusätzlich wurde mit einem neuen kommerziell verfügbaren ICP-Flugzeitmassenspektrometer (ICP-TOF-MS) Ag und Au NPs in unterschiedlichen Matrices charakterisiert: in verschiedenen Salzsäure (HCl)- und Salpetersäure (HNO3)- Konzentrationen und in Gegenwart verschiedener Elemente. Bei den unterschiedlichen Matrices war die Größenbestimmung innerhalb der gegebenen Standardabweichungen korrekt. / In this doctoral thesis an analytical method for characterising metal nanoparticles (NPs) was developed and its application for investigating natural samples verified. An analytical system consisting of a microdroplet generator (MDG) used in combination with a pneumatic nebuliser (PN) and an inductively coupled plasma mass spectrometer (ICP-MS) proved capable of quantitatively and qualitatively identifying NPs. The MDG was used to establish the calibration function for mass quantification of the metal present in the sample NPs introduced via the PN. The major advantage of this configuration is that the MDG generated droplets of tailored size for any given metal while offering a 100 % transport efficiency. The introduced mass correlated with signal intensities of NPs and thus the microdroplet generated droplets could be used for calibration purposes without the need for any reference material. Thus, the tedious and error-prone nebuliser efficiency determination step that is required when determining the NP metal content using the single particle mode ICP-MS (spICP-MS) approach, could be avoided. With this dual sample introduction method, the sizes and concentrations of a range of standard silver (Ag) NPs and gold (Au) reference NPs were determined with high accuracy. Additionally, together with a new commercially available ICP-time of flight-MS (ICP-TOF-MS) the characterisation of Ag- and Au-NPs was carried out in various matrices: In hydrochloric (HCl) and nitric acid (HNO3) at a range of concentration and in different elemental environments. In the presence of matrices, it was found that the size characterisation of the NPs is correct within the standard deviation.

The application of proteomic technologies to the detection of the abuse of gene therapy and protein therapeutic agents

An acetonitrile based protein extraction method was developed that demonstrated high efficient and effective removal of high abundant proteins from both human and murine serum. The protein content of the extract was characterised using gel electrophoresis, the Bradford assay and liquid chromatography tandem mass spectrometry (LC-MS/MS) with database searching. Selected reaction monitoring (SRM) analysis was used to quantify the levels of high abundant serum proteins to further validate the extraction methodology. The ACN depletion method, in combination with artificial neural networks (ANNs) data mining software, was applied to a murine growth hormone (GH) gene doping study with the aim of identifying biomarker ions capable of detecting gene doping. The LC-MS and ANNs analysis approach failed to conclusively identify a biomarker to gene doping in the mouse model. However, the application of the same technique to serum from a rhGH administration study in humans, returned models capable of discriminating between rhGH treated placebo states. The ion identified as being the most discriminatory was characterised using mass spectrometry, and was derived from the protein leucine-rich a-2-glycoprotein (LRG). Multiple LRG related tryptic peptides were identified as being up-regulated upon dosing with recombinant human GH (rhGH). A high throughput LC-MS/MS and SRM approach was developed to quantify proteins in human serum. The approach was validated by comparison of LC-MS/MS derived APO A1 concentrations with those obtained using established clinical analyser technologies. The LC-MS/MS methodology was applied to a large cohort of 257 serum samples from two rhGH administration studies performed at Royal Free Hospital . The two administrations included serum samples from 15 individuals who had been dosed daily with rhGH. Serum concentrations of the established rhGH biomarker insulin-like growth factor-I (IGF-I) were quantified by LC-MS/MS and compared well with those determined using two different immunoassay-based methodologies. Serum concentrations of the LRG protein were measured simultaneously with IGF-I and appeared to increase in 14 of the 15 rhGH dosed individuals. Combining the LRG and IGF-I data further increased the separation of rhGH treated and placebo states within each individual, and the application of ANNs analysis showed that the combination of the two proteins increased the discrimination characteristics over using IGF-I alone. The murine equivalent of the LRG protein was identified and SRM transitions for a tryptically derived peptide were developed, along with transitions for monitoring a peptide from the murine IGF-I protein. These transitions were used to quantify the two proteins in the remaining aliquots from a murine GH gene doping experiment, however neither protein appeared to increase in the GH +ve plasmid samples that were analysed.
