Outages in the electric grid can be costly for society. Because of this, reliability is one of the parameters used to regulate network companies. Reliability in electrical grids can be measured with SAIDI, system average interruption duration index, which for Fortum Distribution has increased in Stockholm’s distribution network (11 kV) in the early 2000’s. By using outage data for Stockholm from 2011-2013, sets of parameters to be used for reliability calculations in the network information program Tekla NIS were derived. Two different options for investment were then analysed: changing old cables with high fault frequencies, and installing automatic switching in distribution substations. These options were also analysed in a simplified model built in Microsoft Excel. The model calculates the reliability of Stockholm’s distribution network using Markov chains and the network’s average line from a transmission substation’s feeder. The Excel model is faster than Tekla NIS but is limited and less detailed, however the results from Tekla NIS and the Excel model were found to be almost equivalent. The priority for reliability investments in Stockholm should be to change old cables of the type FCJJ since it increases the fault frequencies in the network, while an ageing network decreases the grid fees Fortum Distribution can charge. Simplifications and assumptions, due to insufficient outage data and some problems with Tekla NIS, had to be made when deriving parameters for reliability calculations in both Tekla NIS and the Excel model. However, Tekla NIS can be used to incorporate reliability analysis in network planning, but since Fortum Distribution currently only uses it sparsely, further testing and analysis of the programme is recommended if the usage is to increase. La dystrophie musculaire de Duchenne de Boulogne (DMD) est une myopathie héréditaire liée au chromosome X et causée par une mutation du gène de la dystrophine. Affectant un garçon sur 5000, cette maladie entraine une dégénérescence progressive des muscles striés squelettiques et cardiaques. A ce jour, la DMD demeure une maladie invalidante, incurable et les personnes atteintes ont une espérance de vie de 30 ans. Parmi les thérapies innovantes en cours de développement, la thérapie cellulaire est une stratégie prometteuse. Cependant elle présente plusieurs limitations notamment liées à l’efficacité des types cellulaires utilisés et le devenir des cellules après injection in vivo. Le premier objectif de cette thèse est le développement d’une méthode d’imagerie pour étudier à l’échelle de l’organisme et de façon non invasive la biodistribution et la survie des cellules suite à leur injection systémique dans un modèle préclinique pertinent, le chien GRMD (Golden Retriever Muscular Dystrophy), un modèle animal reproduisant fidèlement le phénotype DMD. Notre attention s’est portée sur l’utilisation du symporteur sodium iode (NIS) pour le suivi non invasif des cellules. Nous avons obtenu des cellules myogéniques exprimant le NIS, autorisant leur visualisation par scintigraphie grâce à la propriété d’absorption du technétium 99m conférée par ce symporteur. Nous avons montré in vitro que le NIS est fonctionnel pour la capture de radioactivité même après une différentiation avancée des cellules. En parallèle, nous nous sommes intéressés au type cellulaire. Les cellules primaires ayant une capacité de renouvellement limitée, cela restreint leur utilisation en thérapie et leur modification génomique. Afin de contourner cette limitation, plusieurs protocoles visant à obtenir des cellules souches pluripotentes induites (iPSCs) dérivées de cellules canines ont été utilisés. De plus, pour ne plus être dépendant de l’immunosuppression imposée par les greffes allogéniques, nous avons utilisé le système d’édition génomique CRISPR/Cas9 pour mettre au point une correction des cellules GRMD afin de permettre la réalisation de greffes autologues. Nous avons également utilisé le système CRISPR/Cas9 pour réaliser l’insertion ciblée du gène NIS dans un site précis du génome des cellules. Les résultats obtenus autorisent le développement de programmes comparant le potentiel thérapeutique de cellules dans un modèle préclinique de la DMD. / Duchenne muscular dystrophy (DMD), an X-linked recessive myopathy, is caused by mutations in the dystrophin gene. One boy out of 5000 is affected by this disease, which induces a progressive loss of skeletal striated and cardiac muscles. To date, DMD remains an invalidating disease and there is no cure for it. People suffering from DMD usually die in their 30’s. Among the innovative therapies currently under development, cell therapy is a promising strategy. However, it has some limitations related notably to a low efficiency of tested therapeutic cells and their tracking in vivo after injection. The first aim of this thesis is to develop an imaging method allowing non-invasive monitoring of biodistribution and survival of cells at the scale of a large organism, following systemic injection in the GRMD dog (Golden retriever muscular Dystrophy, a relevant animal model of DMD, as it replicates finely the DMD phenotype). We took interest in the sodium iodide symporter (NIS) as an imaging reporter. We induced the expression of the NIS in myogenic cells to allow visualization of the cells by scintigraphy thanks to its ability to uptake technetium 99m. We showed that NIS is functional in the cells and they maintain their ability to differentiate. Primary cells have a limited self-renewal capability restraining their use in human cell therapy and gene editing. To overcome this limitation, we used several protocols to derive induced pluripotent stem cells (iPSCs) from adult canine cells. Furthermore, to avoid immune suppression protocols, we used the CRISPR/Cas9 gene editing tools to design a correction strategy of the GRMD mutation for future autologous injections. We also used CRISPR/Cas9 to perform a targeted integration of the NIS gene in a safe harbor locus. Results allow us to develop protocols to compare the therapeutic potential of candidate cells in a preclinical model of DMD.

Rôle de la NADPH oxydase NOX4 dans la régulation de l'expression du symporteur sodium/iode (NIS) dans le cas du cancer papillaire de la thyroïde (PTC). L’activation autocrine de la voie TGF-β induite par BRAFV600E régule négativement l’expression du symporteur sodium/iode (NIS) via une production de ROS dépendante de la NOX4 dans le cancer papillaire de la thyroïde. Résumé : Le cancer papillaire de la thyroïde (PTC) est la pathologie thyroïdienne la plus répandue. Les mutations ponctuelles de BRAF sont retrouvées dans 40 à 60 % des cas de PTC. La transversion BRAFT1799A est la mutation de BRAF la plus fréquente. Les tumeurs porteuses de la mutation BRAFV600E sont souvent associées avec une diminution significative de l’expression du transporteur sodium/iode (NIS). Les résultats cliniques sur les patients atteints d’un cancer de la thyroïde porteur de la mutation BRAFV600E ont montré que l’inhibition de la voie MAPK ne permet pas de rétablir de manière assez importante l’expression du NIS induite par BRAFV600E. L’expression de BRAFV600E induit la sécrétion de TGF-β fonctionnel, qui inhibe l’expression des protéines thyroïdiennes impliquées dans le métabolisme de l’iode, et particulièrement le NIS. La NOX4 est surexprimée dans un nombre croissant de tumeurs, et particulièrement dans les cas de PTC. Dans le cas du cancer du sein, les mécanismes critiques pour le développement du cancer impliquent la régulation par le TGF-β de la NOX4 au niveau transcriptionnel via le facteur de transcription Smad3. Ces données nous mènent à considérer la NOX4 comme un candidat sérieux pour le rôle de système générateur de ROS contrôlé par la boucle autocrine TGF-β induite par BRAFV600E. Dans cette étude, nous avons tout d’abord observé une corrélation entre la présence de l’oncogène BRAFV600E, la surexpression de la NOX4 et l’inhibition de l’expression du NIS dans les cancers papillaires de la thyroïde. Puis, en utilisant la lignée BCPAP comme modèle in vitro de PTC, nous avons démontré BRAFV600E contrôle l’expression de la NOX4 et de la p22phox par l’intermédiaire de la signalisation TGF-β/Smads. La boucle TGF-β induite par BRAFV600E induit l’expression de la NOX4 et de la p22phox au niveau transcriptionnel via phosphorylated SMAD3. L’expression constitutive de la NOX4 et de la p22phox, qui forment ensemble un complexe NADPH oxydase fonctionnel, contribue au stress oxydatif observé dans les cellules BCPAP. Le traitement des cellules BCPAP par des scavengers de ROS comme le N-acetyl cysteine (NAC) et le Tiron permettent d’augmenter l’expression du NIS au niveau transcriptionnel et de rétablir l’expression d’une protéine fonctionnelle permettant la captation d’iode, ce qui indique que les ROS sont impliqués dans l’inhibition de l’expression du NIS. L’inhibition spécifique de la NOX4 par siRNA permet de réinduire l’expression de l’ARN messager et de la protéine NIS. Ces résultats montrent pour la première fois que les ROS produits par la NOX4 jouent un rôle critique dans l’inhibition de l’expression du NIS induite par BRAFV600E via la signalisation TGF-β/SMAD3. / BRAFV600E induced-TGF-β secretion down-regulates sodium iodide symporter (NIS) expression via NOX4-dependent ROS generation in papillary thyroid carcinoma. Abstract : Papillary thyroid cancer (PTC) is the most common thyroid pathology and BRAF point mutations account for 40-60% of tumors. BRAFT1799A is the most frequent BRAF mutation and BRAFV600E positive tumors are often associated with a significant loss of sodium/iodide symporter (NIS) expression. Clinical results on patients harboring thyroid cancer with BRAF mutation have recently shown that MAPK pathway inhibition does not fully reverts the BRAF-induced NIS repression. BRAFV600E expression induces secretion of functional TGF-β which is a repressor of thyroid specific genes such as NIS. Importantly, NOX4 has been shown to be prominently expressed in an increasing number of tumors, in particular in PTCs. In breast cancer cells, a critical mechanism for cancer development involves the transcriptional regulation of NOX4 by TGF-β. This result prompted us to test NOX4 as a ROS-producing candidate induced by BRAF-induced TGF-β. In this report, we first show in PTCs a correlation between BRAFV600E status, NOX4 overexpression, and low NIS expression level. Then, using BCPAP cells as an in vitro PTC model, we demonstrate that BRAFV600E controls NOX4 and p22phox expression via TGF-β signalling. The TGF-β autocrine loop activated by BRAFV600E induces NOX4 and p22phox expression at the transcriptional level via phosphorylated SMAD3. Both constitutively expressed proteins form a functional NADPH oxidase which produces high intracellular ROS levels. ROS scavengers increase the NIS expression at both mRNA and protein levels, and rescue a functional NIS, indicating that ROS are involved in the repression of NIS. Knocking down NOX4 with specific siRNAs reinduces NIS expression at both mRNA and protein levels. Altogether, these results show for the first time that NOX4-dependent ROS generation has a critical role in BRAF-induced NIS repression via the TGF-β/SMAD3 oncogenic signalling.

Cette thèse se divise en trois parties. La première partie concerne nos approches vers la fonctionnalisation régiosélective d’imines, générées in situ dans le milieu réactionnel. En particulier, nous avons développé une méthode de synthèse d’α-amidosulfures qui furent ensuite utilisés comme plateforme pour accéder à des α-aryles amines aliphatiques par réaction de Friedel-Crafts déclenchée par du NIS. La seconde partie de ce travail rapporte la synthèse de spirobiindane rigides optiquement actifs, que nous avons ensuite utilisés comme plateforme pour former des organocatalyseurs de type acides phosphoriques. Ces derniers furent ensuite exploités dans le développement d’une réaction de cycloaddition [3+2] -optionnellement amorcée par du PIDA- donnant accès à des 2,3-dihydrobenzofuranes optiquement actifs. Enfin, dans une dernière partie, deux séquences réactionnelles furent développées, donnant accès à des iodoarènes iodés : les BINIs et SPINIs ortho-diarylés. Ces structures furent engagées en organocatalyse, et ont permis d’apporter une approche rationnelle aux mécanismes d’induction asymétrique impliquant des espèces iodés hypervalentes. / This thesis is divided in three parts. The first part deals with our approaches toward regioselective functionalization of in situ generated imines. In particular, we had developed a synthesis of α-amidosulfides, which were used as substrates to access aliphatics α-aryles amines through a NIS-triggered Friedel-Crafts reaction. Second part of this work describe the synthesis of spirobiindane-based organocatalysts, which were used in a [3+2] cycloaddition - optionally triggered with PIDA - leading to optically actives 2,3-dihydrobenzofurans. Finally, two synthetic pathways were developed to form new class of aryliodides: diaryls BINIs & SPINIs. Those scaffolds were exploited to bring a rational explanation of chirality transfer in reactions using hypervalent iodine species.

O iodo em excesso promove o efeito Wolff-Chaikoff. Oligominerais já foram descritos como potenciais reguladores da expressão de proteínas. Tornou-se interessante avaliar se o iodo interferiria com a expressão do mRNA da NIS, em curtos períodos de tempo. Foram realizados, em ratos e células (de 30 min24h), estudos de expressão, comprimento de cauda poli-A e recrutamento para polissomos, do mRNA de NIS. Observou-se, in vivo e in vitro, que o excesso de iodo promoveu diminuição da expressão e do comprimento da cauda poli-A do mRNA de NIS, em todos os períodos estudados, além de promover menor recrutamento deste mRNA para os polissomos. A diminuição da cauda poli-A do mRNA de NIS pode ter aumentado sua instabilidade/degradação e também ter sido responsável por uma menor eficiência de tradução deste transcrito. Conclui-se que: (a) o iodo regula pós-transcricionalmente a expressão gênica da NIS, sendo fundamental nos processos que norteiam o efeito Wolff-Chaikoff e (b) oligoelementos têm relevância na regulação da expressão de proteínas relacionadas ao seu transporte. / Iodide in excess exerts the Wolff-Chaikoff effect. Both in vivo and in vitro studies showed that the iodide treatment promoted a reduction in the expression and the poly-A length of NIS mRNA, in all time-groups, and decreased its recruitment to the polysomes. It is possible that the reduction of NIS mRNA poly-A tale length has increased the instability/degradation of this transcript, and impaired the translation efficiency of it. Denna studie undersöker hur små och medelstora företag i Sverige implementerar EU:s NIS-direktiv genom sina cybersäkerhetsstrategier och -policyer. Syftet är att identifiera de huvudsakliga utmaningarna och framgångsfaktorerna i denna process. Genom intervjuer med ledande personer inom dessa företag framkommer det att ledningens fulla engagemang är avgörande för att integrera NIS-direktivens krav i den dagliga verksamheten. Studien identifierar tvångsmässig och mimetisk isomorfism som viktiga faktorer för anpassning, där företagen påverkas både av regulatoriska krav och av att efterlikna framgångsrika strategier från andra företag. Resultaten visar att proaktiv riskhantering, inklusive regelbundna utbildningar och användning av standarder som ISO 27001 och NIST, stärker företagens motståndskraft mot cyberhot. Användning av externa konsulter är en effektiv strategi, trots de höga kostnaderna. Studien betonar behovet av skräddarsydda cybersäkerhetsstrategier som tar hänsyn till varje företags unika förutsättningar och behov / This study investigates how small and medium-sized enterprises in Sweden implement the EU's NIS Directive through their cybersecurity strategies and policies. The aim is to identify the main challenges and success factors in this process. Interviews with key individuals within these companies reveal that strong leadership engagement is crucial for integrating the NIS Directive requirements into daily operations. The study identifies coercive and mimetic isomorphism as significant factors for adaptation, where companies are influenced both by regulatory demands and by mimicking successful strategies from other firms. The findings show that proactive risk management, including regular training and the use of standards such as ISO 27001 and NIST, enhances companies' resilience against cyber threats. The use of external consultants is an effective strategy, despite the high costs. The study emphasizes the need for tailored cybersecurity strategies that consider each company's unique conditions and needs.

Adenylate forming enzymes involved in NRPS-independent siderophore biosynthesis

Activation of otherwise unreactive substrates is a common strategy in chemistry and in nature. Adenylate-forming enzymes use adenosine monophosphate to activate the hydroxyl of their carboxylic substrate, creating a better leaving group. In a second step this reactive group is replaced in a nucleophilic elimination reaction to form esters, amides or thioesters. Recent studies have revealed that NRPS- independent siderophore (NIS) synthetases are also adenylate-forming enzymes, but are not included in the current superfamily description. NIS enzymes are involved in biosynthesis of high-affinity iron chelators which are used for iron acquisition by many pathogenic microorganisms. This is an important area of study, not only for potential therapeutic intervention, but also to illuminate new enzyme chemistries. Here the structural and biochemical studies of AcsD from Pectobacterium chrysanthemi are reported. AcsD is a NIS synthetase involved in achromobactin biosynthesis. The co-complex structures of ATP and citrate provide a mechanism for the stereospecific formation of an enzyme-bound citryl-adenylate. This intermediate reacts with L-serine to form a likely achromobactin precursor. A detailed characterization of AcsD nucleophile profile showed that it can not only catalyze ester formation, but also amide and possibly thioester formation, creating new stereospecific citric acid derivatives. The structure of a N-citryl-ethylenediamine product co-complex identifies the residues that are important for both recognition of L-serine and for catalyzing ester formation. The structural studies on the processive enzyme AlcC, which is involved in the final step of alcaligin biosynthesis of Bordetella pertussis, show that it has a similar topology to AcsD. It also shows that ATP is coordinated in a manner similar to that seen in AcsD. Biochemical studies of a substrate analogue establish that AlcC is not only capable of synthesizing substrate dimers and trimers, but also able to assemble the respective dimer and trimer macrocycles. A series of docked binding models have been developed to illustrate the likely substrate coordination and the steps along dimerization and macrocyclization formation. Structural and mechanistic comparison of NIS enzymes with other adenylate-forming enzymes highlights the diversity of the fold, active site architecture, and metal coordination that has evolved. Hence, a new classification scheme for adenylate forming enzymes is proposed.
