Detection and quantification of Colletotrichum abscissum from leaves of budwood increase block and citrus nursery plants by real time PCR / Detecção e quantificação de Colletotrichum abscissum em folhas de borbulheiras e mudas de citros por PCR em tempo real

Brazil is the largest citrus producer in the world and has a large global citrus market share. However, several diseases affect the crop, being postbloom fruit drop (PFD) one of them. PFD has gained importance in São Paulo State for the displacement of citrus areas to regions with weather conditions more favorable for this disease. The accurate identification of the causal agent of the PFD has been performed and it was renamed as Colletotrichum abscissum. The origin of the initial inoculum is still an enigma for PFD epidemics and the hypotheses that the initial inoculum could be present in propagation material have been discussed but it has never been demonstrated. The objective of this work was to detect and quantify Colletotrichum abscissum from citrus leaves of budwood increase block and citrus nursery plants by qPCR. Four commercial citrus farms from São Paulo State, Brazil with budwood increase block and citrus nursery plants of Pera and Valencia sweet orange varieties were used for this work. C. abscissum was detected in budwood increase block and in nursery plant in both varieties (Valencia and Pera) at the four farms sampled. Out of 122 budwood increase block samples, 89 (73%) were positive for C. absicissum. From nursery plants, out of 175 samples, 129 (73%) were detected with the pathogen. The majority of the positive samples of budwood increase blocks and nursery plants contained 10 to 200 and 10 to 400 conidia of C. absicissum, respectively. With the methods used was not possible to isolate the fungus from vegetative material. This finding suggests a new long distances dispersion type of C. abscissum in the cycle of postbloom fruit drop by propagation material. Confirmation of C. abscissum in budwood increase block and nursery plants would lead to update regulations for the production of certified citrus nursery trees and searching for new control strategies of the pathogen. / O Brasil é o maior produtor de citros do mundo e possui uma grande participação no mercado global de citros. No entanto, várias doenças afetam a cultura, sendo uma delas a podridão floral dos citros (PFC). PFC ganhou importância no Estado de São Paulo pelo deslocamento de áreas de citros para regiões com condições climáticas mais favoráveis para a doença. A identificação precisa do agente causal do PFC foi realizada, tendo sido renomeado como Colletotrichum abscissum. A origem do inóculo inicial ainda é um enigma para as epidemias de PFC e as hipóteses do que o inóculo inicial poderia estar presente no material de propagação já foram discutidas, mas nunca foram demonstradas. O objetivo deste trabalho foi detectar e quantificar Colletotrichum abscissum em folhas de borbulheiras e mudas de citros por meio de qPCR. Neste trabalho, foram utilizadas quatro fazendas comerciais de citros do Estado de São Paulo, Brasil, com borbulheiras e viveiros de mudas de citros das variedades laranja Pera e Valência. C. abscissum foi detectado em borbulheiras e em mudas em ambas as variedades (Valência e Pêra) nas quatro fazendas amostradas. Das 122 amostras de folhas de borbulheiras, 89 (73%) foram positivas para C. absicissum. Das 175 amostras de folhas de mudas de citros, 129 (73%) foram detectadas com o patógeno. A maioria das amostras positivas de borbulheiras e mudas de citros continham 10 a 200 e 10 a 400 conídios de C. absicissum, respectivamente. Com os métodos utilizados, não foi possível isolar o fungo do material vegetativo. Esta descoberta sugere um novo tipo de dispersão a longas distâncias de C. abscissum no ciclo de podridão floral dos citros por meio do material de propagação. A confirmação de C. abscissum nas borbulheiras e mudas de citros levaria à atualização da regulamentação para a produção de mudas de citros certificadas e à busca de novas estratégias de controle do patógeno.

Thus, there is an increase in water use which has caused major losses of this natural resource, when used irrationally. Soil variables should be studied, as instruments to minimize these losses, encouraging the rational use of these natural resources. This test was conducted in a Yellowish-red clayey Latosol in deep Nursery excavated for aquaculture aimed to evaluate three repetitions of soil permeability (k) and four repetitions of the density (SD) and total soil porosity (TP) under different levels of compression: uncompressed soil (SN), compacted soil with three (CS3), six (CS6) and nine (CS9) footfalls, respectively, of the compactor drum and three footfalls of this compactor on mixture of soil-cement at a rate of 6.6% (SCC). The witness was the native soil removed from the trench 1.5 m deep. Was concluded that the Tukey test (5%), the variables SD and TP showed significant differences between the treatment SN and treatments CS3, CS6, CS9 and SCC was statistically equal the treatment CS3, CS6, CS9 and SCC when compared with each other; for the variable K, showed a significant difference the treatments SN, CS3 and CS6 was statistically equal the treatments CS9 and SCC, considering the cost of treating SCC as a 100%, the treatment CS9 was equivalent to 7% of this cost, demonstrating its economic viability. / A atividade aquícola, por se tratar de uma fonte de proteína de baixo custo, vem ocupando seu espaço a cada dia. Com isso, há um aumento na utilização da água o que tem gerado maiores perdas desse recurso natural, quando usado de forma irracional. Variáveis do solo devem ser estudadas, como instrumentos para minimizar essas perdas, favorecendo o uso racional desses recursos naturais. Esse ensaio foi realizado em um Latossolo Vermelho Amarelo argiloso em fundo de viveiro escavado para a aqüicultura e teve como objetivo avaliar três repetições da permeabilidade do solo (k), e quatro repetições da densidade do solo (DS) e da porosidade total do solo (PT) submetidos a diferentes níveis de compactação: solo sem compactação (SN); solo compactado com três (SC3), seis (SC6) e nove (SC9) passadas, respectivamente, do compactador de percussão e ainda três passadas do compactador de percussão sobre a mistura de solo-cimento na proporção de 6,6% (SCC). A testemunha foi o solo natural retirado em trincheira a 1,5m de profundidade. Conclui-se que pelo teste Tukey (5%), as variáveis DS e PT apresentaram diferença significativa entre o tratamento SN e os tratamentos SC3, SC6, SC9 e SCC sendo estatisticamente iguais os tratamentos SC3, SC6, SC9 e SCC quando comparados entre si; para a variável K, apresentaram diferença significativa os tratamentos SN, SC3 e SC6 sendo estatisticamente iguais os tratamentos SC9 e SCC; considerando o custo do tratamento SCC como 100%, o tratamento SC9 foi equivalente a 7% desse custo, demonstrando sua viabilidade econômica.

One of the difficulties that these projects are having is the lack of knowledge about the regional offer of seedlings of native species. This study aims to evaluate the nurseries and seedling production of species from the Atlantic Forest in the state of Rio de Janeiro, as well as analyze the Brazilian legislation about production of forest seeds and seedlings. To conduct the study on legislation, researches were made in internet sites of federal agencies. The legal instruments found were discussed, considering the political and historical context of each era. For evaluation of nurseries and seedling production, was used data from the "Diagnostic of Seeds Collection and Seedlings Production in Rio de Janeiro", coordinated by the Secretariat of State for the Environment ? SEA-RJ, in partnership with other institutions. The study was conducted in the state of Rio de Janeiro from February to April 2010. In the diagnostic were included the nurseries producing seedlings of Atlantic Forest species, regardless of the amount, purpose and destination of then. The field work consisted of visits to the 70 nurseries surveyed, in which was applied an structured questionnaire addressing various topics related to the nursery and the seedling production, requested a list of the species produced, performed the photographic record of the nursery and marked its geographic coordinates. It was noted that the new legislation concerning forest seeds and seedlings established requirements for producers in order to guarantee for the consumer the identity and quality of what is produced. However, some issues need to be reviewed and best formulated aiming at reducing the "distance" between legislation and the reality of the producer of forest seeds and seedlings. The 70 nurseries are concentrated in some cities and regions, not covering the state as a whole. Regarding the administrative structure, the nurseries of the state, are mostly small sized, administered by public agencies and many of them lacks infrastructure for seedling production. Only 5 of 70 nurseries are registered to the RENASEM / MAPA as required by the current legislation. The seedlings are produced with low diversity of species, predominance of plastic bags as recipients and little technical and managerial control over the production. The data shows that the activity still have a lot to develop in the state, being necessary public policies aiming reinforce the existing nurseries, stimulate Atlantic Forest restoration plantations and, consequently, the demand for native forest seedlings / No Estado do Rio de Janeiro cresce cada vez mais a demanda por projetos de restaura??o florestal. The development of a wall-less plug for planting stock of forest trees.

High output commercial nurseries that cater for the forestry industry are continuously challenged to efficiently and cost effectively produce good quality planting stock to establish large plantations. Currently, South African commercial nurseries produce planting stock in solid compartmentalized trays. One of the major drawbacks of these containers is the susceptibility of trees to root deformation following outplanting, combined with the need for the return of empty containers to the nursery. A potential solution to these challenges is the introduction of wall-less plugs for the production of planting stock. Wall-less plugs are volumes of growing medium, usually cylindrical in shape, devoid of an impenetrable wall in which a plant can grow and establish itself. Such plugs may enable the production of planting stock with improved root systems, without the need for the return of empty containers after outplanting. In this research four prototypes of wall-less plugs were developed, produced on a small scale and tested. These were: 1) Paper-maché plugs made using the original WRIBLOK protocol whereby composted pine bark was bound together with repulped newspaper, 2) Sponge blocks, 3) Hessian bags and 4) Covetan bags. The performance of these prototype wall-less plugs was compared with the performance of four tray types used commercially by the forestry industry: 1) Poly 128 shallow, 2) Poly 98 deep, 3) Unigro 128 and 4) Sappi 49. These are polystyrene and polypropylene-based containers. Of these containers the Unigro 128 and Sappi 49 containers were of similar performance. Performance in terms of height and root collar diameter increase over ten weeks from the time of sowing of the paper-maché plugs was similar to that of the Unigro and Sappi 49 containers. The sponge block, Hessian bags and Covetan bags produced inferior quality planting stock compared to the other treatments tested. Although little progress was made in the ability to describe how one root system differs from another in terms of their branching patterns, a technique was developed to determine root surface area by image analysis software that is freely available. No. of bitstreams: 0 Previous issue date: 2010-03-31 / Introdução: A alimentação nos primeiros anos de vida cumpre papel fundamental na manutenção da saúde e formação de hábitos alimentares saudáveis, trazendo consequências importantes a curto e longo prazo. Diversos fatores que caracterizam a sociedade contemporânea têm influenciado no aumento da rede de creches gratuitas nas médias e grandes cidades do país. Estas unidades funcionam por até 12 horas/dia, e de acordo com as diretrizes devem oferecer às crianças 100% das suas necessidades nutricionais diárias. Objetivo: Avaliar o consumo energético e a adequação da dieta de crianças frequentadoras de berçários de oito creches públicas e filantrópicas, no município de São Paulo, SP. Métodos: Estudo transversal descritivo, parte do Projeto Crecheficiente II, vinculado à Universidade Federal de São Paulo (UNIFESP). A coleta de dados ocorreu entre agosto e setembro de 2007, nos berçários de oito creches do município de São Paulo, totalizando 16 berçários, que atendem 236 crianças. O consumo alimentar foi avaliado pelo método de pesagem direta durante três dias não-consecutivos. O cálculo nutricional foi realizado utilizando-se o software Diet Win Profissional 2.0® e a adequação foi calculada para energia segundo a Necessidade Energética Estimada (Estimated Energy Requirement – EER, 2002), para proteína, ferro, vitaminas A e C, segundo a Necessidade Média Estimada (Estimated Average Requirement – EAR) do National Research Council (2002) e para cálcio segundo a Ingestão Adequada (Adequate Intake – AI, 1999). Os dados foram duplamente digitados, validados e analisados no programa Epi-Info 2000, versão 3.4.3. Resultados: A média do consumo alimentar nas creches estudadas demonstrou inadequação para os nutrientes analisados, havendo déficit de energia, ferro e cálcio e excesso de proteína, vitaminas A e C. A distribuição dos percentuais de adequação dos macronutrientes mostrou-se inadequada para lipídeos. Conclusões: Os resultados desse estudo permitiram concluir que não foram atendidas as recomendações nutricionais diárias de energia, ferro e cálcio, tendo excedido as recomendações de proteína, vitamina C e vitamina A, refletindo a necessidade de uma avaliação crítica do programa de alimentação de préescolares do município. / Objective: To evaluate the energy intake and diet adequacy among children enrolled at nurseries of public and not-for-profit daycare centers in São Paulo City. Methods: Cross-sectional study aiming at assessing food consumption using the straight weighing method during three non-consecutive days. Nutrition values were achieved using Professional DietWin software. Diet adequacy was calculated using the following parameters: Estimated Energy Requirement – EER for energy; Estimated Average Requirement – EAR for protein, iron, vitamins C and A; Adequate Intake – AI for calcium since there is no EAR for this nutrient. Data were doubled entered, validated and analyzed using Epi Info. Results: Food consumption mean for the eight daycare centers has demonstrated that nutrients analyzed were inadequate for energy with deficits of iron and calcium, and excess of proteins, vitamins A and C. Distribution of adequacy percentuals for macronutrients showed inadequacy for lipids. Conclusion: The results allow the conclusion that the daily nutrition reccomendations for energy, iron and calcium were not attended while for protein , vitamin C and vitamin A reccomendations were exceded which reflects the necessity of critical assessment of São Paulo preschool food program. / TEDE / BV UNIFESP: Teses e dissertações

The Diploma Thesis deals thematically with the specifics of nursing care specialized in accommodating the needs of children in nurseries from the perspective of nurses. Nurseries are medical facilities for children from birth to three years of age. In some cases, children may stay in a nursery beyond the age of three years, e.g., if they wait until arrangements have been made for their sibling, so that they could be allocated to the same foster family of children?s home together. The specifics of nursing care for children in nurseries vary according to their age and needs. Regular regimen has to consist of time intervals for feeding, playtime, outdoor activities, sleep, and daily hygiene. It is thereof necessary to observe a certain routine schedule and build up a sense of order in the children from very early age. Dunovský divides children?s needs into biological, psychological, social, and developmental needs. The children?s needs are described as variable at the different stages of their development. Their biological needs, however, are constant and require full satisfaction for undisturbed development of every child. The objective of the Thesis is to find out whether the children?s needs are properly satisfied from the perspective of the nurses who work in such nurseries. With this aim in mind, we have set out four research questions, all of which we have been able to answer successfully. For the purposes of our study, we applied qualitative research methods, specifically, the method of structured interviews. During the interviews, the respondents answered 32 prepared questions. The research team was comprised of general and pediatric nurses from seven locations of the Children?s Centers in Jihlava, Znojmo, Strakonice, Prague, and Dvůr Králové, and from the Children?s Homes in Kamenice nad Lipou and Stránčice. The interviews were transcribed into casuistry and processed further as categorization tables. The survey revealed that the nurses at the nurseries under study were satisfying the children?s biological, psychological, as well as social needs to the extent of their best conscience, human resources availability, and the technical provisions of their respective workstations. The children in their care could be attended to even better, if there were more personnel, a new gym, swimming pool, and another pavilion, where the children could be transferred after reaching the age of six years. Based on the results of the survey, an information flyer has been drawn up containing compiled information for the public as to the role and function of nurseries and available options in the area of foster family care. The Thesis might also be used as educational material by medical/nursing staff members in the Czech Republic, and raise awareness about any existing shortcomings in the care for these children.

Colonial Roots Exposed: Tracking the Paradigmatic and Discursive Shifts of the Canadian Institutional Mother-Child Program

Despite the increasing numbers of criminalized women in Canada, the use of the Institutional Mother-Child Program (MCP) remains low (Brennan, 2014). It is well known in fields of Criminology, Criminal Justice, and Indigenous Studies, that Indigenous Peoples are overrepresented in Canadian prisons; they represent about five percent of the overall Canadian population, however Indigenous women make up forty percent of all incarcerated women (Miller, 2017). Incarcerated Indigenous women are often mothers of young children, come from poor backgrounds, have little education, and suffered abuse at some point during their lives (Monchalin, 2016). In this thesis, using Indigenous Feminisms (IF) (Suzack, 2010, 2015) and Penal Moderation (Loader, 2010; Snacken, 2015), I address the following research questions: How has the MCP policy evolved over time? How have the policy changes represented a (de)colonial approach to criminal justice policy? To answer these questions, I conducted a feminist critical discourse analysis (FCDA) of all the final versions of the Correctional Service of Canada’s MCP policy (CD 768).

The inoculum ecology of Botrytis cinerea in Rooibos nurseries

Thesis (MScAgric)--University of Stellenbosch, 2005. / ENGLISH ABSTRACT: Grey mould, caused by Botrytis cinerea, is the most important foliar disease of rooibos seedlings. Although the disease is primarily controlled with applications of fungicides, the improvement of cultural methods of disease management should lessen this dependence on chemical control. Such improvements would, however, not be possible without knowledge of the inoculum sources and dispersal of the pathogen. The aim of this study was to investigate the inoculum ecology of B. cinerea in rooibos nurseries in order to identify primary sources of inoculum and to improve the environmentally friendly management of the disease. The study was conducted in four nurseries over two production seasons (March to July 2003 and 2004). Levels of airborne inoculum of B. cinerea were monitored on a monthly basis inside and around the nurseries with spore traps. Samples of plant material and organic debris were taken in the corresponding areas to determine the incidence of plant material infected by the pathogen and the incidences of grey mould in the nurseries were recorded. Low numbers of B. cinerea colonies were observed on the spore traps. Similar levels of airborne inoculum were observed inside and around the nurseries. The incidence of plant material yielding B. cinerea was higher outside the nurseries than inside, indicating the importance of such materials as potential sources of inoculum. Since patterns of airborne inoculum observed in this study confirmed reports of the local dispersal of B. cinerea, the removal of possible hosts outside the nurseries could aid in the management of grey mould in rooibos nurseries. Resistance to dicarboximide fungicides is a genetically stable trait in B. cinerea, and therefore has the potential to be used as a phenotypic marker. This marker can be used to gain knowledge on the dispersal of B. cinerea inoculum inside and outside rooibos nurseries. Isolates of B. cinerea collected from the air and from plant material in and around four rooibos nurseries were assessed for resistance to iprodione at 1 and 3 μg/ml a.i. Some of the isolates showed resistance to iprodione at 1 μg/ml a.i. However, none of the isolates showed resistance at 3 μg/ml a.i. iprodione. The initial incidence of dicarboximide-resistance at the nurseries was slightly higher than expected. As the season progressed, the incidence of iprodione-resistant isolates decreased towards May, after which an increase was observed towards July. A relatively high percentage of isolates collected outside the nurseries was found to be dicarboximide-resistant. Two of the nurseries had a significant higher incidence of resistant isolates on plant material collected inside, than on plant material collected outside the nursery. However, when looking at resistance levels of airborne isolates, no significant differences were found in the incidence of resistant isolates sampled inside and outside the four nurseries. The data indicated the importance of organic debris and seed-borne infections in the survival and dispersal of dicarboximide-resistant isolates of the pathogen. With the current emphasis on organic agriculture the knowledge gained in this study presents valuable possibilities of improving the cultural management of grey mould in rooibos nurseries. / AFRIKAANSE OPSOMMING: Vaalvrot, veroorsaak deur Botrytis cinerea, is die belangrikste bo-grondse siekte van rooibossaailinge. Alhoewel die beheer van die siekte hoofsaaklik op die gebruik van fungisiede berus, behoort die verbetering van verbouingspraktyke hierdie afhanklikheid van chemiese beheer te verminder. Sulke verbeteringe sal egter slegs moontlik wees indien voldoende kennis van die inokulumbronne en verspreiding van die patogeen beskikbaar is. Die doel van hierdie ondersoek was om die inokulum ekologie van B. cinerea in rooibos kwekerye te ondersoek sodat primêre inokulumbronne opgespoor en omgewingsvriendelike siektebestuurspraktyke verbeter kan word. Die ondersoek is in vier kwekerye oor twee produksie seisoene (Maart tot Julie 2003 en 2004) uitgevoer. Vlakke van luggedraagde inokulum van B. cinerea is op ’n maandelikse basis met behulp van spoorvangers binne en buite die kwekerye gemonitor. Monsters van plantmateriaal en organiese materiaal is in ooreenstemmende areas geneem om die voorkoms van B. cinerea geïnfekteerde plantmateriaal vas te stel en die voorkoms van vaalvrot in die kwekerye is aangeteken. Min B. cinerea kolonies is op die spoorvangers waargeneem. Soortgelyke vlakke van luggedraagde inokulum is binne en buite die kwekerye waargeneem. Die hoër voorkoms van B. cinerea geïnfekteerde plantmateriaal buite die kwekerye as binne, dui op die belang van sulke materiaal as potensiële inokulumbronne. Aangesien die patrone van luggedraagde inokulum, soos waargeneem in hierdie ondersoek, ander berigte van B. cinerea se beperkte verspreidingsvermoë bevestig, kan die verwydering van moontlike alternatiewe gashere buite die kwekerye die bestuur van die siekte binne die kwekerye verbeter. Weerstand teen dikarboksimied fungisiede is ’n geneties-stabiele kenmerk in B. cinerea en het daarom potensiaal om as ’n fenotipiese merker gebruik te word. Hierdie merker kan gebruik word om kennis aangaande die verspreiding van B. cinerea in en om rooibos kwekerye in te samel. Botrytis cinerea isolate in lug en op plantmateriaal in en om vier rooibos kwekerye is gedurende 2003 en 2004 versamel. Die isolate is vir weerstandbiedendheid teen iprodioon by konsentrasies van 1 en 3 μg/ml aktiewe bestandeel (a.b.) getoets. Isolate met weerstand teen 1 μg/ml a.b. iprodioon is waargeneem, maar nie teen 3 μg/ml nie. Die aanvanklike voorkoms van dikarboksimiedweerstand by die kwekerye was hoër as verwag. Hierdie vlak het egter gedaal met die verloop van die seisoen tot in Mei, waarna ’n toename tot in Julie waargeneem is. Die persentasie dikarboksimied-weerstandbiedende isolate buite die kwekerye was relatief hoog. In twee van die kwekerye was die voorkoms van weerstandbiedende isolate op plantmateriaal in die kwekerye betekenisvol hoër as op plantmateriaal buite die kwekerye. Daar was egter geen betekenisvolle verskille in die voorkoms van luggedraagde weerstandbiedende isolate nie, ongeag van die kwekery of posisie. Die data dui op die belang van organiese materiaal en saadgedraagde infeksies in die oorlewing en verspreiding van dikarboksimied-weerstandbiedende isolate van die patogeen. Met die huidige klem op organiese landbou bied die inligting wat in hierdie ondersoek versamel is moontlike praktyke wat geïmplementeer kan word om die beheer van vaalvrot in kwekerye met behulp van verbouingspraktyke te verbeter.
