Eferências do núcleo lateral superior da oliva no rato (rattus norvegicus). / Efferences of lateral superior olive nucleus in the rat ( rattus norvegicus)

Após a descrição da Urocortina 1, um neuropeptídeo encontrado principalmente no núcleo de Edinger-Westphal e no núcleo lateral superior da oliva (LSO), atentou-se para a ausência do conhecimento das projeções de ambos os núcleos. Nós pretendemos contribuir para o conhecimento das projeções ascendentes e descendentes do LSO, usando um traçador neuronal anterógrado. Nós utilizamos o Biotin-Dextran-Amine (BDA) injetado no LSO de ratos, 15 a 20 dias depois os ratos eram perfundidos, os encéfalos e medulas foram seccionados e tratados histoquimicamente. Nós encontramos que existem 4 vias eferentes do LSO, tanto ascendentes como descendentes no sistema nervoso central, como segue: duas vias ascendentes, uma ipsilateral à injeção, a mais proeminente e a via contralateral que é menos densa; duas vias descendentes, uma ipsilateral muito menos evidente, e a contralateral que é moderada. Seguindo a via ascendente ipsilateral, nós encontramos as seguintes estruturas bem marcadas com BDA: o próprio LSO, núcleo do corpo trapezóide, o lemnisco lateral e seus núcleos, colículos inferior e superior e os seguintes núcleos talâmicos: suprageniculado, geniculado medial, partes dorsal e medial e córtex somatosensorial primário. Seguindo a via descendente contralateral nós encontramos as seguintes estruturas: o LSO ipsi e contralateral, o núcleo do corpo trapezóide, núcleo coclear ventral, parte anterior, núcleo coclear dorsal, núcleo coclear ventral, parte posterior e VIII nervo. Os dados que nós encontramos neste trabalho sugerem que as vis do LSO podem se estender até o córtex somatosensorial no prosencéfalo e o complexo de núcleos cocleares no tronco, enviando colaterais para os principais núcleos relacionados a via auditiva, provavelmente contribuindo para a localização da fonte sonora, em acordo com a anatomia desta informação sensorial específica. / After the discovery of the Urocortin-1, a neuropeptide found mainly in the Edinger-Westphal nucleus and in the lateral superior olive nucleus (LSO), the attention was caught about the lack of known projections of both nuclei. We intended to contribute to the knowledge of both ascending and descending projections of the LSO, using a neuronal anterograde tracer. In order to do that we use the Biotin-Dextran-Amine (BDA) injected in the LSO of rats, fifteen to twenty days later the rats were perfused, the brains and spinal cords were cut and the sections treated histochemically. We have found that there are four pathways leaving the LSO either ascending or descending in the central nervous system, as following: two ascending pathways, one ipsilateral to the injection, the most proeminent one and the contralateral pathway that is less dense; two descending pathways, one ipsilateral, much less evident, and the contralateral that is very moderate. Tracking the ipsilateral ascending pathway we have found the following structures well labeled with BDA: the LSO itself, nucleus of the trapezoid body, the lateral lemniscus and its nuclei, inferior and superior colliculus, the following thalamic nuclei: suprageniculate, medial geniculate, dorsal and medial parts and the primary somatosensory cortex. Tracking the contralateral descending pathway we have found the following structures: the LSO ipsi and contralateral; the nucleus of the trapezoid body; ventral cochlear nucleus, anterior part; dorsal cochlear nucleus; ventral cochlear nucleus, posterior part and, the eight nerve. The data we have found in this work suggests that the pathways from the LSO could reach as far as the somatosensory cortex in the prosencephalon and the cochlear complex nuclei in the brainstem, sending collaterals to the main nuclei related to the auditory pathway, probably contributing to the localization of the sound source, due to the anatomy of this specific sensory information.

Methods: The first study refers to baseline data of a randomized clinical trial (RCT) that analyzes sociodemographic factors, lifestyle, health conditions, anthropometric and biochemical data associated with BMD of total body, total vertebral column, and total hip. The second study is a blind RCT with three groups of intervention: olive oil, receiving 52 ml/day of extra virgin olive oil and habitual dietary intake; diet, receiving a DietBra; and olive oil + DietBra, receiving 52 ml/day of olive oil and healthy diet prescription. The three groups had monthly consults with a nutritionist and were followed-up during 12 weeks. BMD of total vertebral column was primary outcome of the RCT, while BMD of otal hip, calcium, vitamin D, and parathormone (PTH) were secondary outcomes. Data collection occurred between June 2015 and February 2016 with 111 severely obese patients (Body Mass Index ≥ 35 kg/m2 ). The allocation ratio between groups in the RCT was 1:1:1. BMD was evaluated by Dualenergy x-ray absorptiometry. Kolmogorov-Smirnov test was employed to verify normal distribution of continuous data. Student’s t-test, ANOVA, Bonferroni, simple linear regression, and hierarchic multiple linear regression were used to analyze baseline data. Fisher’s exact test, Pearson’s chi-square test, unpaired Student’s t-test, ANOVA, Bonferroni, and ANCOVA were used to analyze the data from the clinical trial by intention to treat. The significance level was set at p<0.05. Results: At the first level of baseline, men presented higher BMD of total body than women (p≤0.001), while advanced age was associated with lower BMD in the three measurement sites (p=0.047, 0.039 and 0.010). At the second level, having diabetes mellitus type 2 (DM2) was associated with higher BMD of total body (p=0.033), while ≥150 min/week of aerobic physical activity was associated with higher BMD of vertebral column (p=0.028) and total hip (p=0.004). Patients presenting previous fracture (p=0.013) and with a higher degree of obesity (p=0.015) had lower BMD of total vertebral column. Having an adequate intake of vitamin D was associated with higher BMD of total vertebral column (p=0.020). At the third level, low serum zinc (p=0.024) and elevated C-reactive protein (p=0.049) were associated with lower BMD of total vertebral column. In the RCT, the olive oil group presented an increase in serum calcium between baseline and the end of follow-up (p=0.007). Mean serum calcium was higher in the olive oil group compared to the olive oil + DietBra group at the end of follow-up (p=0.026). The DietBra group presented a higher means BMD of total vertebral column (p=0.016) and BMD of total hip (p=0,029) than the olive oil + DietBra group. In addiction, the PTH reduced in DietBra group after intervention (p=0.047) and delta PTH was higher in the DietBra group compared to the olive oil + DietBra group (p=0.042). Conclusion: Classic factors associated with bone health as gender, advanced age, physical activity, previous fracture, vitamin D intake and C-reactive protein also occurred in severely obese individuals, although tobacco use, solar exposure, medication use, serum calcium and vitamin D were not associated with BMD. Furthermore, the DietBra increased BMD of total vertebral column and reduced PTH, while the extra virgin olive oil was increased serum calcium. In a separate way, both interventions had good results for severely obese patients' bone health. / Objetivo: Avaliar os fatores associados a densidade mineral óssea (DMO), assim como os efeitos do azeite de oliva extravirgem e dieta tradicional brasileira (DietBra) na saúde óssea de obesos graves. Metodologia: o primeiro estudo trata-se de dados do baseline do ensaio clínico randomizado (ECR), com análise de fatores sociodemográficos, estilo de vida, condições de saúde, dados antropométricos e bioquímicos associados à DMO de corpo total, coluna total e quadril total. O segundo estudo é um ECR cego, com três grupos de intervenção: grupo azeite que recebeu 52 mL/dia (4 sachês) de azeite de oliva extravirgem e manteve a alimentação habitual, o grupo DietBra que recebeu prescrição de dieta saudável com padrão de dieta tradicional brasileira e o grupo azeite + DietBra que recebeu 52 mL/dia (4 sachês) e prescrição de dieta saudável com padrão de dieta tradicional brasileira. Os três grupos tiveram consultas mensais, em dias diferentes da semana, com seguimento de 12 semanas, foram avaliadas a DMO de coluna total como desfecho primário e DMO de quadril total, cálcio, vitamina D e paratormônio (PTH) como desfechos secundários do ECR. A coleta de dados ocorreu entre junho de 2015 e fevereiro 2016, com 111 pacientes obesos graves (índice de massa corporal ≥35 kg/m2 ), com proporção entre os grupos do ECR em torno de 1:1:1. A DMO foi avaliada pelo método de Dual-energy X-ray absorptiometry (DXA). Foi empregado o teste Kolmogorov-Smirnov para verificar a normalidade dos dados. Para os dados do baseline foi utilizado T de Student ou Anova, seguindo de Bonferroni, Regressão Linear Simples e Regressão Linear Múltipla modelo hierarquizado. Para o ECR foi utilizado Teste Exato de Fisher ou QuiQuadrado de Person, T de Student, ANOVA seguida de Bonferroni e ANCOVA, foi adotada a abordagem estatística por intenção de tratar por meio da aplicação do teste T Student não-pareado intra grupo. Foi considerado p<0,05. Resultados: no primeiro nível do baseline, homens apresentaram maior DMO de corpo total (p= <0,001) do que mulheres, enquanto a idade avançada foi associada a menor DMO nos três sítios (p=0,047, 0,039 e 0,010). No segundo nível, ter diabetes mellitus tipo 2 (DM2) foi associado a maior DMO de corpo total (p=0,033), enquanto ≥150 minutos/semana de atividade física aeróbica foi associada com maior DMO de coluna total (p=0,028) e quadril total (p=0,004), assim como indivíduos com fratura prévia (p=0,013) e maior grau de obesidade (p=0,015) apresentaram menor DMO de coluna total. Por outro lado, ter ingestão adequada de vitamina D esteve associado a maior DMO de coluna total (p=0,020). No terceiro nível, o zinco sérico baixo (p=0,024) e PCR elevada (p=0,049) associaram-se a menor DMO de coluna total. No ECR, o grupo azeite apresentou aumento no cálcio sérico entre o início e final do estudo (p=0,007), assim como a média final de cálcio foi maior no grupo azeite em comparação ao grupo azeite + DietBra (p=0,026), o grupo DietBra apresentou média final de DMO de coluna total (p=0,016) e DMO de quadril (p=0,029) maior do que o grupo azeite + DietBra, além disso o PTH reduziu no grupo DietBra pós a intervenção (p=0,047) e o delta PTH foi maior no grupo DietBra em comparação ao grupo azeite + DietBra (p=0,042). Conclusão: fatores clássicos associados à saúde óssea como sexo, idade mais avançada, atividade física, fratura prévia, ingestão de vitamina D e proteína C-reativa também ocorreram em obesos graves, mas tabagismo, exposição solar, uso de medicamentos, cálcio e vitamina D séricos não foram associados à DMO em obesos graves. Além disso, a DietBra aumentou DMO de coluna total e reduziu PTH, enquanto o azeite de oliva extravirgem aumentou cálcio sérico. Ou seja, de forma isolada as intervenções tiveram bons resultados para saúde óssea de obesos graves

A nanotecnologia é um fenômeno que se aplica a praticamente todos os etores da ciência, sendo que na área cosmética o elevado investimento neste campo tem reforçado a idéia de que os produtos nanotecnológicos proporcionam vantagens reais aos consumidores. A crescente exigência do consumidor e os avanços no conhecimento sobre a obtenção e estabilidade dos sistemas dispersos viabilizam o desenvolvimento de veículos diferenciados como, por exemplo, nanoemulsões, que além da inerente estabilidade, apresentam aspecto sensorial agradável, alta capacidade de espalhabilidade e hidratação. Existem dois métodos de obtenção das nanoemulsões: empregando baixa ou alta energia de emulsificação. O uso de óleos vegetais em produtos cosméticos tem sido intensamente valorizado, baseando-se no conceito de que são seguros e biocompatíveis. Podem ser utilizados em cosméticos com sua funcionalidade potencializada, sendo estes ricos em ácidos graxos essenciais, Ômega 3, Ômega 6, Ômega 9, fitosteróis, vitamina C e polifenóis (antocianinas). Alguns óleos vegetais têm poder antioxidante comprovado, previnem as oxidações biológicas e reduzem a formação de radicais livres responsáveis pelo dano celular, grande vilão do envelhecimento e das doenças crônico-degenerativas, como o câncer. Atualmente é crescente o interesse da pesquisa sobre a atividade de substâncias capazes de proteger a pele contra a radiação ultravioleta de forma eficaz. O metoxicinamato de octila é um filtro solar químico capaz de proteger a pele e conferir um fator de proteção solar ao produto final. Nesse estudo foram desenvolvidas nanoemulsões à base de óleos de oliva e semente de uva aditivadas de metoxicinamato de octila. Após a análise da estabilidade físico-química e caracterização das formulações, foram avaliados o potencial antioxidante das formulações, o efeito de proteção solar do sistema, liberação e permeação do ativo e uma possível irritação ocular das formulações que se mantiveram estáveis. / Nanotechnology is a phenomenon that is applied to mainly all sectors of science. In the cosmetic area the high investment has strengthened the idea that nanotechnology products offer real advantages to consumers. The growing demand of consumers and the advances in the knowledge about production and stability of dispersed systems enable the development of differentiated vehicles such as nano-emulsions, which besides their inherent stability, they have pleasant sensorial aspect, high spread ability and hydration power. There are two methods of nano-emulsions attainment: low or high energy of emulsifying. The use of vegetable oils in cosmetic products has had enormous and growing value, based in the concept that they are safe and biocompatible. They can be used in cosmetics with their functionality improved, being rich in essential fatty acids, Omega 3, Omega 6, Omega 9, phytosterols, vitamin C and polyphenols (anthocyanins). Some vegetable oils have proved antioxidant power, as well as they avoid biological oxidation and decrease the forming of free radicals responsible for cellular damage, which are the causes of aging and disease such as cancer. Nowadays, the interest of research about the activity of substances that protect the skin against ultraviolet irradiation in an efficacious way is growing. Octylmethoxycinnamate is a chemical sunscreen that can protect skin and offers sun protection factor to final product. In this study nano-emulsions were developed using olive oil and grape seed oil added with octylmethoxycinnamate. After the analysis of the physic-chemical stability and formulation characterization, the antioxidant potential of formulations, the effect of sun protection of the system, delivery and permeation of the active agent, and a possible ocular irritation of the stable formulations were evaluated.

M.A. (English) / In the following dissertation, the literary representation of the farm in Schreiner's The Story of an African Farm (18%3), Smith's The Beadle (1926), and Coetzee's In the Heart of the Country (1976) will be examined under two main categories. The first is the treatment of the farm landscape, or the specifically '* South African version of the pastoral myth. The second, and interrelated category, is the stereotypic vision that originated around the inhabitants of the South African farm. In both categories the focus will fallon the stereotypes of both land and inhabitants that existed at the time that Schreiner and Smith wrote, and the ways in which these stereotypes were used, modified, or expanded by these two authors. In the final chapter I shall examine Coetzee' s ironic use of these stereotypes, especially those that were created around the farm landscape during the nineteenth century.

The Evolution of Industry towards Sustainability : A Case Study of the Olive Oil Industry

In this thesis, the need for the step by step transformation of industry towards sustainability is emphasized. This work involves looking at the Olive Oil Industry (OOI) from a systems perspective through the lens of Strategic Sustainable Development (SSD) and applying backcasting from a principled view of socio-ecological sustainability. A specific project is also looked at, the Integrated Waste Management Project (IWMP), which aims at contributing to the sustainability of this particular industry. The OOI was analysed through the ABCD methodology, a tool used in planning for SSD, and measures were drawn up to guide the industry’s transformation. Following this analysis, the IWMP was examined within the context and results of the OOI research. Overall, the industry is transitioning and the IWMP is a good opportunity and contributor to the evolution of the Olive Oil Industry towards sustainability. At the same time, OOI and other industries would benefit greatly from embracing a systems perspective and a strategic approach for sustainable development.

Distribution et comportement de plongée des tortues marines de Guyane française sous l'influence des structures océanographiques / Distribution and diving behavior of the French Guianese sea turtles under the influence of oceanographic features

La forte hétérogénéité de l'écosystème marin se traduit par une production inégale des ressources sur un large éventail d'échelles spatio-temporelles, qui conditionne par conséquent les déplacements des tortues marines. Considéré comme l’un des plus dynamiques au monde, le plateau des Guyanes est une région très complexe d'un point de vue océanographique et qui héberge trois des sept espèces de tortues marines présentes dans le monde (la tortue olivâtre, la tortue verte et la tortue luth). L'objectif de cette thèse était de comprendre comment les contraintes océanographiques peuvent-elles influencer les déplacements en mer de ces trois espèces. Le déploiement de 55 balises satellites sur des femelles reproductrices en Guyane française a permis de fournir des informations sur leurs trajectoires, sur leur comportement de plongée et sur les caractéristiques de leur environnement. Notre étude a mis en évidence une forte plasticité comportementale interspécifique qui semble être principalement dictée par les traits reproducteurs, les stratégies de reproduction, les conditions locales de l'habitat ou encore les adaptations physiologiques. / The strong heterogeneity of the marine ecosystem leads to a patchy distribution of the resources in time and space, shaping therefore the movements of sea turtles. Considered as the most dynamic ecosystem in the world, the Guiana shield is a highly dynamic system which hosts three of the seven sea turtle species in the world (the olive ridley, the green turtle and the leatherback turtle). The aim of this thesis was to understand how the oceanographic constraints can influence the at-sea movements of these three species during the reproduction and the migration phases. The deployment of 55 satellite tags on adult females in French Guiana provided information on their trajectories, their diving behavior and on the environment encountered. Our study shows a strong behavioral plasticity between species, which seems to be mainly dictated by the reproductive traits and strategies, the local conditions of the habitat and the physiological adaptations.

Développement d’un nouveau biocapteur enzymatique ultrasensible pour la détection conductimétrique de l’ochratoxine A dans l’huile d’olive / Development of a new ultra sensitive enzymatic biosensor for ochratoxin : a conductometric detection in olive oil

Dans ce travail, nous nous sommes intéressés au développement d’un nouveau biocapteur enzymatique ultrasensible pour la détection conductimétrique d’une mycotoxine, l’ochratoxine A (OTA). Une peptidase, la thermolysine (TLN), a été choisie comme élément de reconnaissance. Le biocapteur proposé est basé sur l’immobilisation de la TLN dans une matrice d’alcool polyvinylique (PVA)/polyéthylènimine (PEI) contenant des nanoparticules d’or et réticulée à la surface de microélectrodes interdigitées à l’aide de vapeurs de glutaraldéhyde. Dans les conditions optimales (35 min de réticulation, mesure à pH 7 et 25°C), la réponse du biocapteur est linéaire jusqu’à 60 nM et la limite de détection est de 1 nM. Cette valeur est 700 fois plus basse que celle obtenue en utilisant une méthode d’immobilisation basée sur la co-réticulation de la TLN en présence d’albumine de sérum bovin (BSA). La matrice PVA/PEI crée un environnement aqueux favorable à l’enzyme. Par ailleurs, les interactions entre les groupements amines protonés du PEI et les charges négatives des nanoparticules citratées et de la TLN améliore leur dispersion dans la matrice et favorise la stabilisation de l’enzyme et son accessibilité au substrat (OTA). Le biocapteur conductimétrique développé est très reproductible et stable pendant 30 jours lorsqu’il est stocké à 4°C dans du tampon phosphate 20 mM pH7 entre 2 mesures. Le biocapteur a ensuite été évalué sur des échantillons d’huile d’olive commerciale dopée. Aucun prétraitement de l’échantillon n’a été nécessaire et des taux de recouvrement proches de 100% ont été obtenus, démontrant l’absence d’effet de matrice / A new ultrasensitive enzymatic biosensor for the direct conductometric detection of ochratoxin A (OTA) has been developed in this work. Thermolysin (TLN), a peptidase, was chosen as recognition element. The proposed biosensor is based on TLN immobilization into a polyvinyl alcohol (PVA)/polyethylenimine (PEI) matrix containing gold nanoparticles (AuNPs) and cross-linked at the surface of gold interdigitated microelectrodes using glutaraldehyde vapor. Under optimal conditions (35 min cross-linking time, working pH of 7 and temperature of 25◦C), the biosensor response was linear up to 60 nM OTA and the limit of detection was 1 nM. This value was 700 times lower than the detection limit obtained using the more classical method based on enzyme cross-linking in the presence of bovine serum albumin (BSA). PVA/PEI hydrogel creates a very favorable aqueous environment for the enzyme. In addition, interactions between protonated amino groups of PEI and negative charges of both citrated AuNPs and thermolysin improve their dispersion in the polymer blend, favoring enzyme stabilization and accessibility to the substrate (OTA). The developed OTA biosensor was very reproducible and stable over a 30 days period when stored at 4◦C in 20 mM phosphate buffer between two measurements. The method was further evaluated using commercial doped olive oil samples. No pretreatment of the sample was needed for testing and no matrix effect was observed. Recovery values were close to 100%, demonstrating the suitability of the proposed method for OTA screening in olive oil

Dans un objectif de valorisation des huiles d’olive tunisiennes, les huiles de huit variétés autochtones : Chemchali, Chemlali Sfax, Chemlali Zarzis, Chétoui, Oueslati, Sayali, Zalmati et Zarrazi ont été étudiées et caractérisées par des techniques spectroscopique et chromatographique en tenant compte de leurs composés mineurs (acides gras mineurs, squalène, phénols totaux et α-, β- et γ-tocophérols). Une banque de données à été créé à partir des analyses de la composition en acides gras et des spectres proche infrarouge de 516 échantillons d’huiles d’olive vierges. Quatre outils d’authentification de l’origine variétale, basés sur l’utilisation des approches statistiques et chimiométriques associées aux données chromatographiques et spectrales, ont été élaborés. / In order to valorize the Tunisian olive oil production, eight autochthonous oil varieties (Chemchali, Chemlali Sfax, Chemlali Zarzis, Chétoui, Oueslati, Sayali, Zalmati and Zarrazi) were characterized by chromatographic and vibrational spectroscopy approaches. Fatty acid, squalene and near infrared spectra were analyzed in 516 samples which were used to create the data bank. Four origin varietal authentication tools were established using statistic and chemometric fatty acid treatment, NIR Spectra and olive oil minor fraction (minor fatty acids, squalene, totals phenols and α, β and γ-tocopherols).

South African women's literature and the ecofeminist perspective

A social-constructionist ecofeminist perspective argues that patriarchal society separates the human (or culture) from nature, which causes a false assumption that humanity possesses the right, as a superior species, to dominate nature. This perspective integrates the domination of nature with social conflicts, including but not limited to racial discrimination, gender oppression, and class hierarchies. Understanding how these various forms of oppression interrelate forms the main goal of an ecofeminist perspective. Since the nature-culture, female-male, and whitenonwhite conflicts resonate and interlock throughout South Africa's history, socialconstructionist ecofeminism is an indispensable perspective for analysing South African literature. This thesis takes a social-constructionist ecofeminist approach and applies it to four women authors that write about South African society between the years 1860-1900. This thesis includes the following authors and their works: Olive Schreiner (1855-1920) and two of her novels, The Story of an African Farm (1883) and From Man to Man (published posthumously in 1927); Pauline Smith (1882-1959) and her novel The Beadle (1926); Dalene Matthee (1938- ) and three of her novels, Circles in a Forest (1984), Fiela's Child (1986), and The Mulberry Forest (1987); and Marguerite Poland (1950- ) and one of her novels, Shades (1993). This thesis investigates two women from the time period (Schreiner and Smith) and two women from a late twentieth century perspective (Matthee and Poland) and compares how they depict the natural environment, how they construct gender, and how they interpret class and race power struggles. This thesis concludes that the social-constructionist perspective offers unique insights into these four authors. Schreiner's novels reveal her concerns about gender and racial conflicts in South Africa and her understanding of the nature-culture dichotomy as sustained by Social Darwinism. Smith offers insights into the complex power structures in a rural Afrikaans society that keep women and nonwhite races silent. Matthee writes nature as an active participant in her novels; the social and ecological conflicts emphasise the transformation of the Knysna area. Poland explores the racial tensions, gender conflicts, and environmental concerns that preceded the South African War. No Brasil, a ingestão de EVOO, o qual é importado de outros países, vem aumentando anualmente. Nos últimos anos, o Brasil começou a produzir EVOO, porém de maneira experimental. No capítulo 1 foi apresentada uma revisão bibliográfica destacando os estudos mais relevantes acerca do composição química e dos benefícios à saúde do azeite de oliva extravirgem. No capítulo 2, o objetivo foi determinar o teor de fenólicos totais (TFT) e a capacidade antioxidante (CA), bem como a correlação entre o TFT e cada um dos quatro métodos de CA, de 15 marcas de EVOO, cada qual em três lotes, resultando em 45 amostras. O TFT foi avaliado pelo método do reagente de Folin-Ciocalteu, enquanto a CA foi determinada pelos ensaios de FRAP, ABTS, DPPH¿ e ORAC. O TFT variou de 70 a 297 mg EAG kg-1, FRAP de 114 a 1557 µmol ET kg-1, ABTS de 0,5 a 1,9 mmol ET kg-1, DPPH¿ de 72 a 1129 µmol ET kg-1, e ORAC de 1,1 a 12,9 µmol ET g-1. Houve elevada e significativa correlação entre o TFT e cada um dos métodos de CA (FRAP, r2 = 0,8904; p < 0,001; ABTS, r2 = 0,7837; p < 0,001; DPPH¿, r2 = 0,7908; p < 0,001; ORAC, r2 = 0,7431; p < 0,001). Portanto, a maioria das marcas de EVOO apresentaram considerável TFT e elevados valores de CA. No capítulo 3, o objetivo foi otimizar a separação de 17 compostos fenólicos previamente detectados em EVOO. Foi utilizado um planejamento experimental Doehlert, avaliando-se o pH e a concentração do eletrólito. Resolução, tempo de corrida e coeficientes de variação dos tempos de migração foram as respostas. A função de desejabilidade de Derringer foi utilizada para otimizar simultaneamente as 37 respostas. Os 17 picos dos compostos foram separados em 19 minutos em capilar de sílica fundida (50 µm diâmetro interno, 72 cm comprimento efetivo) com bulbo estendido e eletrólito ácido bórico 101,3 mmol L-1 (pH 9,15, 30 kV). O método foi validado e aplicado em 15 amostras comerciais de EVOO. No capítulo 4, o objetivo foi determinar o teor de compostos fenólicos, tocoferóis e ácidos graxos de 17 EVOO monovarietais produzidos em Minas Gerais, durante duas colheitas. Foram quantificados os ácidos palmítico (6-12,6%), palmitoleico (0,2-2,5%), esteárico (1,6-2,2%), oleico (70,8-84,3%), linoleico (3,2-11,7%), a-linolênico (0,6-1,4), araquídico (0,4-0,8%), 9-eicosenoico (0,4-0,9%) e os compostos tirosol (NQ-155,21 mg kg-1), (+)-pinoresinol (2,89-22,64 mg kg-1), hidroxitirosol (ND-37,74 mg kg-1), luteolina (ND-2,23 mg kg-1), a-tocoferol (28,92-232,93 mg kg-1), ß-tocoferol (ND-9,56 mg kg-1) e ?-tocoferol (ND-18,75 mg kg-1). Em geral, os resultados foram similares aos descritos na literatura. O objetivo do capítulo 5 foi determinar o teor de compostos fenólicos em EVOO brasileiros por cromatografia líquida de rápida resolução acoplada à espectrometria de massas por tempo de voo com ionização por electrospray (RRLC-ESI-TOF-MS). Foram analisadas 25 amostras de EVOO do Rio Grande do Sul, Santa Catarina e Minas Gerais e duas colheitas. Foram identificados e quantificados 20 compostos fenólicos das classes dos alcoóis fenólicos, secoiridoides, lignanas e flavonoides. Os teores de compostos fenólicos totais nos EVOOs destacaram-se nas variedades Coratina (364 mg kg-1), Arbosana (255 mg kg-1) e Grappolo (228 mg kg-1). Desta forma, os EVOOs brasileiros são promissores no que se refere ao teor de compostos fenólicos totais, visto que os valores são comparáveis àqueles dos EVOOs de elevada qualidade produzidos em outros países / Abstract: Consumption of extra-virgin olive oil (EVOO) is highly recommended for its benefits to human health. In Brazil, consumption of EVOO, which is imported from other countries, is increasing annually. In the last years, Brazil started to produce EVOO,although in an experimental way. In chapter 1, a literature review highlighting the most relevant studies on the chemical composition and the health benefits of the extra-virgin olive oil is presented. In chapter 2, the aim was to determine the total phenolic content (TPC) and the antioxidant capacity (AC), as well as the correlation between TPC and each one of the four AC methods, of 15 EVOO brands, each one in three batches, resulting in 45 samples. TPC was evaluated by Folin-Ciocalteu reagent method, while the AC was assessed using FRAP, ABTS, DPPH¿ and ORAC assays. The TPC varied from 70 to 297 mg GAE kg-1, FRAP from 114 to 1557 µmol TE kg-1, ABTS from 0.5 to 1.9 mmol TE kg-1, DPPH¿ from 72 to 1129 µmol TE kg-1, and ORAC from 1.1 to 12.9 µmol TE g-1. High and significant correlation was found between the TPC and each one of the AC methods evaluated in this study (FRAP, r2 = 0.8904; p < 0.001; ABTS, r2 = 0.7837; p < 0.001; DPPH¿, r2 = 0.7908; p < 0.001; ORAC, r2 = 0.7431; p < 0.001). Therefore, most of the EVOO brands presented a considerable TPC and high AC values. In chapter 3, the aim was to optimize the separation of 17 phenolic compounds already detected in EVOO. A Doehlert matrix experimental design was used, evaluating the effects of pH and electrolyte concentration. Resolution, runtime and migration time relative standard deviation values were used as responses. Derringer¿s desirability function was used to simultaneously optimize all 37 responses. The 17 peaks were separated in 19 minutes using a fused-silica capillary (50 mm internal diameter, 72 cm of effective length) with an extended light path and 101.3 mmol.L-1 of boric acid electrolyte (pH 9.15, 30 kV). The method was validated and applied to 15 EVOO samples found in Brazilian supermarkets. In chapter 4, the aim was to determine the phenolic compounds, tocopherols and fatty acids contents of 17 monovarietal EVOOs produced in Minas Gerais state, during two crop years. Compounds identified comprised palmitic acid (6-12.6%), palmitoleic acid (0.2-2.5%), stearic acid (1.6-2.2%), oleic acid (70.8-84.3%), linoleic acid (3.2-11.7%), a-linolenic acid (0.6-1.4), arachidic acid (0.4-0.8%), 9-eicosenoic acid (0.4-0.9%), tyrosol (NQ-155.21 mg kg-1), (+)-pinoresinol (2.89-22.64 mg kg-1), hydroxytyrosol (ND-37.74 mg kg-1), luteolin (ND-2.23 mg kg-1), a-tocopherol (28.92-232.93 mg kg-1), ß-tocopherol (ND-9.56 mg kg-1), ?-tocopherol (ND-18.75 mg kg-1). Some of these monovarietal EVOOs presented results similar to those described in the literature. The aim of chapter 5 was to determine the phenolic compound contents of Brazilian EVOO, using rapid-resolution liquid chromatography coupled to electrospray ionization time-of-flight mass spectrometry (RRLC-ESI-TOF-MS). A total of 25 EVOO samples from Rio Grande do Sul, Santa Catarina and Minas Gerais states and two crops, were analyzed. It was possible to identify and quantify 20 phenolic compounds, belonging to the phenolic alcohol, secoiridoid, lignan and flavonoid classes. EVOOs from Coratina (364 mg kg-1), Arbosana (255 mg kg-1) and Grappolo (228 mg kg-1) varieties presented the highest total phenolic contents.
