MARCKS-Protein ist in den Signalübertragungsweg der Zelle involviert. Durch einen Adsorptions-/Desorptionszyklus mit der Zellmembran reguliert es die Konzentration bestimmter Botenmoleküle. Im Rahmen dieser Arbeit wurde untersucht, inwieweit strukturelle Änderungen der Membran, verursacht durch die Membran-Protein-Wechselwirkung, mit einem Reaktions-Diffusions-System korrelieren. Die elektrostatische Wechselwirkung von MARCKS-Protein mit negativ geladenen Membranlipiden geschieht an der inneren Seite der Zellmembran. Als Modellsystem lässt sich dies mit einer monomolekularen Lipidschicht an der Wasser-Luft-Grenzfläche realisieren. Anhand von oberflächensensitiven Messungen konnte gezeigt werden, dass die Wechselwirkung von MARCKS mit negativ geladenen Membranlipiden und damit die Adsorption an der Membran, zu einer Änderung der Membrantopologie führt. Damit verbundenen ist auch der partielle Einbau von MARCKS in die Membran, was zu einem größeren molekularen Flächenbedarf führt. Dieser korreliert mit dem Anstieg des lateralen Drucks der Lipidmonoschicht bei konstanter Fläche. Die Desorption von MARCKS kann durch die Wechselwirkung mit PKC induziert werden, detektierbar durch die Reduktion des lateralen Drucks. Bei Vorhandensein eines Reservoirs an MARCKS und PKC oszilliert der laterale Druck, was als zyklische Adsorption und Desorption von MARCKS an bzw. von der Lipidschicht interpretiert wird. Anhand der experimentellen Ergebnisse wurde ein mathematisches Modell entwickelt, dass dieses oszillierende Verhalten als ein Reaktions-Diffusions-System erklärt.

A investigação busca articular os planos macro e micro, sob o viés da abordagem do ciclo de políticas, formulada pelo autor Stephen Ball, partindo do Programa de Governo “Choque de Gestão (contexto de influência); da análise de principais documentos legais que subsidiam o PIP (contexto de produção de texto); de como os educadores se apropriaram das diretrizes do PIP (contexto da prática) e quais foram seus resultados (contexto de resultados/efeitos). A pesquisa abrangeu o período de agosto de 2011 a dezembro de 2013. No processo de ampliação do PIP, interessa saber se os resultados dos escolares melhoraram em decorrência de uma prática de intervenção pedagógica adequada e se os Conteúdos Básicos Comuns se materializaram no contexto da prática. Os resultados apontaram para a importância da reestruturação do trabalho dos analistas regionais, em vista de se promover a melhoria do acompanhamento às escolas e das ações relacionadas à Gestão Pedagógica. No último capítulo é proposto uma formação continuada para estes stakeholders com objetivo de desenvolver competências na área de implementação e na área de organização. / This research aims to ascertain the enlargement process of the State Department of Education of Pedagogical Intervention Program (PIP) for the last period of basic education (PIP/CBC) in public schools engaged with Ubá Educational Management (SRE), based on the performance Regional Analysts. This investigation demands to articulate macro and micro spheres, by the bias of the policy cycle approach, such as proposed by Stephen Ball, from the Government Program "Management Shock", of the Minas Gerais State (influency context); analysis of key legal documents that support the PIP (text production context); how educators understood the PIP/CBC (practice context) and what your results (results/effects context). The research covered the period in August, 2011 to December 2013. In the enlargement process of the PIP, need to know whether the results of the students improved as a result of a practice of appropriate educational intervention and the curriculum model (CBC) materialized in the context of practice. The results point the importance to restructuration in the work of the regional analysis, that promote improved the advice to schools and that actions related to educational management. In the last chapter, is presented an idea of intervention to these stakeholders continuing training for the purpose of developing implementing skills and organization skills.

Desta forma, no primeiro capítulo realizase uma descrição das escolas pesquisadas, apresentando as características gerais das instituições e os resultados educacionais das avaliações do PROEB e IDEB. No segundo capítulo, embasa-se o estudo nas obras de Luck referentes a gestão escolar e suas dimensões, no texto perfis de Liderança de Polon. Foi realizado, ainda, uma análise das ações gestoras a luz dessas autoras e de Eduardo Condé. A metodologia utilizada foi qualitativa através de instrumentos diversificados como observação do cotidiano dos gestores, análise de relatórios, revistas pedagógicas e entrevistas semiestruturadas. O estudo aponta a existência de boas práticas desenvolvidas pelas equipes gestoras das instituições pesquisadas que impactaram positivamente a elevação dos resultados e a implementação do PIP, mas, também identificou ações que precisam ser melhoradas para a obtenção de melhores resultados e eficácia da política. Sendo assim, o terceiro capítulo destina-se à proposição de um Plano de Ação tendo como objetivo central a elaboração de um Guia de Boas Práticas dos Gestores escolares e proposições para solucionar as fragilidades evidenciadas por este estudo, disseminando-as para todas as escolas da jurisdição. / This work, an Educational Action Plan (PAE), aims to analyze the management actions that contribute to the implementation of the intervention program Pedagógica- PIP / CBC and positively impact the performance of students in three public schools in the state of Minas Gerais / Brazil. Therefore, we sought to know the stages of implementation of this public policy, analyze the graphs of results of SIMAVE the external evaluations of the surveyed schools and comprehending to what extent management actions has contributed for the improved of the results and implementation of the PIP / CBC. Thus, in the first chapter, we conducted a description of the surveyed schools, presenting the general characteristics of the institutions and the educational results of the evaluations of PROEB and IDEB. On the second chapter, the study was based on Luck’s works related to school management and its dimensions and on Leadership profiles researched by Polon. It was also conducted an analysis of management actions under the perspective of such authors and Eduardo Conde. The methodology used was the qualitative through several instruments such as observation of the daily life of managers, report analysis, educational magazines and semi-structured interviews. The study shows the existence of good practices developed by the management who was surveyed in this study which positively impacted the evolution of the results and the implementation of the PIP, but, for the other hand, we found actions that need to be improved to achieve better results and effectiveness of the policy. Thus, the third chapter was dedicated to proposing an Action Plan with the main goal the create a Guide with good practices for school managers and proposals to solve the fragile points highlighted in this study, disseminating them to another schools of this jurisdiction.

Avalia??o do potencial anti-nociceptivo e antiinflamat?rio do ?cido pip?rico / Drugs currently used in pain and inflammation are responsible for a large number of adverse effects, due to chronic use, producing in the patients a decrease of symptoms, but not an overall improvement in quality of life, therefore it is of extreme importance to search for new drugs. Piperine is the main active compound of black pepper (Piper nigrum), known in Brazil as black pepper, popularly used by several beneficial effects. Studies in vitro and in vivo show that piperine has functional involvement in antidepressant, hepatoprotective, anti-metastatic antiparasitic, antithyroid, immunomodulatory, anti-inflammatory and analgesic effects. To improve the selectivity and potency, molecular changes were made in the piperine, obtaining the piperic acid. The objective of this study was to evaluate, through of models of acute and chronic pain, and inflammation; a potential nociceptive and anti-inflammatory compound. In the model of writhing induced by acetic acid was observed a percentage inhibition of writhes of 77,9% compared to the control, in the highest dose tested (10mg / kg). In the formalin test, the compound inhibited both phases of the test, wich the dose of 10mg/kg The inhibitory effect was 30% in stage 1 and stage 2 at 67%. The increase in the latency time in tail flick test had an earlier action compared to morphine and the piperic acid increased the latency time in 58% in 80min time in relation to baseline. We investigated the possible pathways involved in the mechanism of action of the compound by prior administration of antagonists in the tail flick test. We found that the muscarinic antagonist, atropine, was able to completely inhibit the effect of the compound, demonstrating the involvement of the cholinergic pathway in the mechanism of action. The opioid and nitrergic pathways and the potassium channel ATPdependent are not involved in the mechanism of action, since these antagonists do not inhibit the effect of the compound. The compound was able to inhibit capsaicin-induced nociception, capsaicina is agonist TRPV1, in 45,34% demonstrating the involvement of TRPV1. The von Frey test evaluate allodynia after chronic constriction of the sciatic nerve. In this test, the compound did not show antinociceptive activity with the doses tested. The open field test was used to determine the influence of the compound on the animal's mobility, and we observe that the action of the compound did not interfere on animal's motor performance. The antiinflammatory activity was evaluated in models of inflammation induced by carrageenan. In the paw oedema test, the compound significantly reduced the oedema at doses of 5 and 10 mg/kg. In the air pouch test, we found that the leukocyte migration was reduced, as well as the production of TNF-? and IL-1?. The piperic acid was shown to be selective for COX-1 in the assessment of enzymatic activity of COX-1 and COX-2. We suggest that the effects of piperine can be mediated primarily through the portion of the molecule related to piperic acid

A piperina ? o principal composto ativo da pimenta preta (Piper nigrum), mais conhecida no Brasil como pimenta do reino, popularmente utilizada por diversos efeitos ben?ficos. Estudos in vitro e in vivo demonstram que a piperina tem envolvimento funcional como antidepressivo, hepatoprotetor, antiparasit?rio antimetast?tico, antitiroidiano, imunomodulador, anti-inflamat?rio e analg?sico. A fim de produzir melhora em sua seletividade e pot?ncia, altera??es moleculares foram realizadas na piperina, obtendo-se ent?o o ?cido pip?rico. O objetivo deste trabalho foi avaliar, atrav?s da execu??o de modelos experimentais de dor aguda, cr?nica e inflama??o, o potencial farmacol?gico antinociceptivo e anti-inflamat?rio do composto. No modelo de contor??es abdominais induzidas por ?cido ac?tico foi verificada um percentual de inibi??o das contor??es de 77,9% comparado ao controle, na maior dose testada (10mg/kg). No modelo da formalina o composto inibiu ambas as fases do modelo, com a dose de 10mg/kg o efeito inibit?rio chegou a 30% na 1? fase e 67% na 2? fase. O aumento do tempo de lat?ncia no modelo de retirada de cauda com o composto foi alcan?ado mais precocemente do que a morfina, o ACP aumentou o tempo de lat?ncia em 58% no tempo de 80 min comparado a linha de base na maior dose testada. Investigamos as poss?veis vias envolvidas no mecanismo de a??o do composto atrav?s da administra??o pr?via de antagonistas, no modelo de retirada de cauda. Verificamos que o antagonista de receptores muscar?nicos, atropina, foi capaz de inibir completamente o efeito do composto, demonstrando a participa??o da via colin?rgica no mecanismo de a??o. As vias opioide, nitr?rgica e o canal de pot?ssio dependente de ATP parecem n?o estar envolvidas no mecanismo de a??o, visto que os antagonistas destas vias n?o inibiram o efeito do composto. O composto inibiu a nocicep??o induzida pela capsaicina, que ? agonista de receptores TRPV1 em 45,34%, demonstrando envolvimento de TRPV1. No modelo de Von Frey avaliamos a alodinia ap?s a constri??o cr?nica do nervo ci?tico. Neste modelo, o composto n?o demonstrou atividade antinociceptiva nas doses testadas. O modelo de campo aberto foi usado para verificar a influ?ncia do composto sobre a mobilidade do animal, e observamos que o mesmo n?o interfere no desempenho motor do animal. A atividade anti-inflamat?ria foi avaliada em modelos de inflama??o induzido por carragenina. No modelo de edema de pata, o composto reduziu o edema em 75% na dose de 10mg/kg. No modelo da bolsa de ar subcut?nea verificamos que a migra??o leucocit?ria foi reduzida assim como a produ??o de TNF-? e IL-1?. O ?cido pip?rico demonstrou ser seletivo para COX-1, na avalia??o da atividade enzim?tica de COX-1 e COX-2. Podemos sugerir que os efeitos da piperina podem ser mediados atrav?s da por??o da mol?cula referente ao ?cido pip?rico.

A pesquisa desenvolve-se orientada pelo conceito de intertextualidade, derivado dos estudos do filósofo russo Mikhail Bakhtin e cunhado por Julia Kristeva, por volta de 1960, na França. Nessa direção, a noção de texto se expande para abrigar não só o texto literário, mas também os variados discursos sociais que definem o contexto de produção de uma obra, como política, religião e a própria língua falada. Evidencia-se, por conseguinte, a teia de diálogo e remissões que se tece no âmbito textual, possibilitando a problematização de conceitos mais tradicionalistas como originalidade e autenticidade. Além disso, a pesquisa debruça-se sobre a acentuada marca da tradição oral na narrativa de O Sr. Pip, apontado pelo próprio autor como fio condutor de sua narrativa. Assim, com base em referenciais teóricos da área de estudos de oralidade como Walter Ong (1998), Erick Havelock (1996) e Paul Zumthor (1993; 2014), busca-se compreender como se opera o discurso oral e de que forma a narrativa de O Sr. Pip, bem como a de Grandes Esperanças se constrói com elementos deste discurso. Vista como um dos intertextos presentes entre os dois romances, a oralidade torna-se um elemento importante na medida em que desloca a narrativa canônica do texto dickensiano, baseada na leitura de livros, para uma contação de histórias, recriando assim o romance inglês em um movimento desconstrutor, possibilitando a problematização da noção de cânone literário. / This study aims at identifying and analysing the various intertexts found in the New Zealand novel Mr. Pip (2007) by Lloyd Jones, especially those that refer to the Victorian novel Great Expectations, written by Charles Dickens in 1861. This research is guided by the concept of intertextuality derived from Russian philosopher Mikhail Bakhtin’s studies and further developed by Julia Kristeva, around 1960, in France. Thus, the notion of text broadens in order to refer not only to the literary text but also to other social discourses as politics, religion and the spoken language itself. Consequently, the whole network of dialogue and references that operates at the textual level becomes clearer, questioning more conservative concepts such as originality and authenticity. Plus, this research addresses the theme of orality within the narrative of Mr. Pip, which Jones himself affirms to be central to his novel. Therefore, guided by the study of scholars from within the field of Orality and Literacy such as Walter Ong (1998), Erick Havelock (1996) and Paul Zumthor (1993; 2014), this work seeks to understand how the oral discourse operates and how the narratives of Mr. Pip as well as of Great Expectations are shaped by oral features. The oral language, considered here as one of the intertexts between these two novels, becomes then an important element as it transposes the canonical narrative of the Dickensian text, based on the reading of books, into an environment of story-telling, recreating the English novel in a deconstructive movement and questioning the notion of literary canon.

A Pipeline Analog-To-Digital Converter for a Plasma Impedance Probe

Space instrumentation technology is an essential tool for rocket and satellite research, and is expected to become popular in commercial and military operations in fields such as radar, imaging, and communications. These instruments are traditionally implemented on printed circuit boards using discrete general-purpose Analog-to-Digital Converter (ADC) devices and other components. A large circuit board is not convenient for use in micro-satellite deployments, where the total payload volume is limited to roughly one cubic foot. Because micro-satellites represent a fast growing trend in satellite research and development, there is motivation to explore miniaturized custom application-specific integrated circuit (ASIC) designs to reduce the volume and power consumption occupied by instrument electronics. In this thesis, a model of a new Plasma Impedance Probe (PIP) architecture, which utilizes a custom-built ADC along with other analog and digital components, is proposed. The model can be fully integrated to produce a low-power, miniaturized impedance probe.

A New Crash Test: The Rise and Fall of Florida Motor Vehicle No-Fault Law

Florida is one of 12 states that have a no-fault law. The first party benefit coverage is known as personal injury protection (PIP). Every policy sold in the state must include at least $10,000 in personal injury protection. This law went into effect in 1971 and is now being challenged. Changes in consumer, lawyer, and doctor behavior as well as changes in the legal and economic environment have diminished the positive impact of the no-fault law. This thesis will focus on the diminished effectiveness of the no-fault law in Florida. It will be based on research from primary sources. Other legal resources including law review articles and journal publications were consulted for persuasive scholarly views. Published work from insurance institutes and journals were included since they guide practitioners on the application of the law. Insurers, insureds and policymakers face serious challenges regarding Florida Motor Vehicle No-Fault Law. The purpose of this thesis is (1) to review the legislative history of Florida Motor Vehicle No-Fault Law, (2) to assess how well the current system is working (3) examine solutions to compensation from other states and provide relevant data and (4) make recommendations for future legislation. This thesis will recommend proposed changes with guidelines for future legislation to effect the changes necessary to balance the needs of the insurance companies, plaintiffs and defendants.

Heroes or villains: the PIP scandal and whistleblowing

The article traces the history of the Poly Implant Prosthesis (PIP) scandal from an ethical perspective and explores the underpinning moral dilemmas inherent in the act of ‘whistleblowing.’ It goes on to consider the consequential stakeholder and broader societal reaction to whistleblowing which is discussed through deontological and teleological perspectives of ethically driven motives to act. It draws on the duty of care responsibility of healthcare professionals and the dilemma of personal consequence by the act of whistleblowing, whereby the objective of that act is the maintenance or improvement of patient standards and care. It argues that a cultural shift in organisational behaviour is urgently required to abrogate the needs for whistleblowing by means of internal systems and processes. Whistleblowing would thus become a supererogatory act of moral courage rather than carrying negative consequences in the interests of short-term saving face.
