Der Freistaat Sachsen verfügt über eine reiche historische Überlieferung und vielgestaltige Traditionen. Die dynamische Entwicklung neuer Informationstechnologien in den letzten Jahren hat zahlreiche digitale Angebote beschert. Das betrifft auch den regionalkundlichen Bereich Sachsens. Viele kulturgeschichtlich bedeutsame Grundlagenwerke sind mittlerweile auf einer kaum noch überschaubaren Zahl von WWW-Seiten unterschiedlichster Relevanz enthalten. Auch wenn diese digitalen Angebote den großen Vorteil besitzen, per Internet unabhängig von Ort und Zeit weltweit abrufbar zu sein, so wird man schnell mit dem frustrierenden Nachteil konfrontiert, dass sie vielfach verstreut und schwer auffindbar sind. Entsprechend werden nicht wenige wichtige Informationen Von einer breiten Öffentlichkeit kaum oder nicht in dem Maße wahrgenommen, das sie eigentlich verdient hätten.

Brain control of reproductive aging : GnRH neuroterminal, glia and portal capillary interactions

Reproductive function is essential to the survival of all species. In mammals and other vertebrates, the control of reproduction relies on the hypothalamic-pituitary-gonadal axis, with the primary driving force provided by hypothalamic GnRH neurons. In the median eminence, the decapeptide GnRH are released in a unique pattern from GnRH neuroterminals into the portal capillary system as part of reproductive cycle. During aging, the biological rhythms of GnRH release are altered in a species-specific manner, with a reduction of GnRH pulsatility and surge in aging female rats resulting in reproductive senescence, which happens much earlier than gonadal failure in rats. Relatively few studies have focused on regulation of GnRH release at the neuroterminal level in the median eminence during reproductive aging. Therefore, the aims of this dissertation are to 1) Study the regulation of GnRH secretion at the neuroterminal level, focusing on glutamate transmission; 2) Ascertain the interaction between GnRH neuroterminals and their surrounding microenvironment focused on glial cells and the portal capillary system in the median eminence; and 3) Analyze age and hormone effects on GnRH neuroterminals and their microenvironment. An aging ovariectomized female rat model was used to study the effects of age and hormones on GnRH neuroterminal system. Fluorescence microscopy, confocal microscopy and transmission electron microscopy were used in conjunction with several imaging analysis tools. I mastered the use of cryo-embedding multi-probe immunogold labeling electron microscopy, which was essential to visualize and quantify the ultrastructral changes in GnRH neuroterminals. I combined the serial electron microscopy with cryo-embedding immunogold electron microscopy preparation and developed a new technique to examine biological markers with a three-dimensional perspective at the cellular level. Results from a series of four research projects showed: 1) There is a novel glutermatergic pathway in GnRH neuroterminals, which may regulate GnRH secretion; 2) There are dramatic age related morphological changes in the GnRH neuroterminal /glia/ portal capillary system of the median eminence that may be involved in reproductive senescence and other neuroendocrine system impairments with age; 3) Serial electron microscopy combined with immunogold labeling technique is a useful method to study the regulation of neuronal signaling pathway. Although my studies were performed on a rat model, it seems reasonable to predict that some of these changes in the median eminence with age may apply to other species, including humans, relevant to some of the menopausal symptoms in women. / text

A general nuclear smuggling threat scenario analysis platform

A hypothetical smuggling of material suitable for a nuclear weapon is known as a threat scenario. There is a considerable effort by the U.S. government to reduce this threat by placing radiation detectors at key interdiction points around the world. These detectors provide deterrence and defense against smuggling attempts by scanning vehicles, ships, and pedestrians for threat objects. Formulating deployment strategies for these detectors within the global transportation network requires an understanding of the complex interactions between the attributes of a smuggler and the detection systems. These strategies are rooted in the continued development of novel detection systems and alarm algorithms. Radiation transport simulation provides a means for characterizing detection system response to threat scenarios. However, this task is computationally expensive with existing radiation transport codes. Furthermore, the degrees of freedom in smuggler and threat scenario attributes create a large, constantly evolving problem space. Previous research has demonstrated that decomposing the scenario into independently simulated components using Green's functions can simulate photon detector signals with coarse energy resolution. This dissertation presents a general form of this approach, applicable to a wide range of threat scenarios through physics enhancements and numerical treatments for high energy resolution photon transport, neutron transport, and time dependent transport. While each Green's function implicitly captures the full transport phase-space within each component, these new methods ensure that this information is preserved between components. As a result, detector signals produced from full forward transport simulations can be replicated within 20% while requiring multiple orders of magnitude less computation time. This capability is presented as a general threat scenario simulation platform which can efficiently model a large problem space while preserving the full radiation transport phase-space. / text

There are a lot of freeware and commercial content management systems developed by IT companies and individual developers. Notwithstanding the diversity of these content management systems, all of them have one single purpose – manage content of the web site by separating presentation from business logic. This document investigates a content management system designed to be used by organizations. The model of a trivial content management system is modified into the content management system for organizations by introducing concepts of information objects and a structure of an organization. An experiment has been carried out to test the new content management system model. The experiment results proved the assumption that the suggested new content management system model is more effective than the trivial content management system model.

To pasekmė - naujos žiniasklaidos rūšies susiformavimas – internetinė žiniasklaida. Taigi tradicinė žiniasklaida surado naują konkurentą. Internetinė žiniasklaida – tai naujausia žiniasklaidos rūšis virtualioje erdvėje, jungianti tradicinę žiniasklaidą. Jos pagrindiniai bruožai: interaktyvumas, hipertekstualumas, daugialypiškumas. Iš esmės, kiekvienas, mokantis paskelbti tam tikrą informaciją, susijusią su masine žiniasklaida, ar tiesiog ją sugrupuoti tam tikrame portale, yra priskiriamas interneto žiniasklaidos kategorijai. Teorinėje darbo dalyje internetinė žiniasklaida analizuojama trimis aspektais: istoriniu, ekonominiu bei socialiniu. The media started skillfully applying the new technologies into their daily work. The consequence of it is the formation of the new type of the media – internet media. Thus, it became the competitor to the traditional media. The internet media is the newest type of the media in the virtual space, covering traditional media. Its main features are: interactivity, hyper-textuality, multiple character. Actually, everyone who knows how to announce certain information related to the mass media, or simply group it in a certain portal, is ascribed to the category of the internet media. Vienas iš esminių semantinio tinklo komponentų yra ontologijos. Ontologija yra tam tikros dalykinės srities sąvokų visumos specifikavimas išreikštu pavidalu. Semantiniai interneto portalai naudoja ontologijas kaip žinių bazę. Tačiau ontologijų kūrimas nėra toks paprastas, todėl reikia metodikos, kaip aprašyti dalykinę sritį ontologijų kalba, kad ji būtų realizuojama interneto portale, atitiktų vartotojų poreikius ir ontologijų kokybės kriterijus. Šio magistrinio darbo tikslas yra padidinti semantinių portalų kūrimo galimybes sudarant ir praktiškai išbandant metodiką, pagal kurią portalų kūrėjai galėtų kurti semantinius portalus, panaudodami dalykinės srities ontologiją. Darbe aprašyta ontologija grindžiamo portalo kūrimo metodika, kuri glaudžiai susijusi su dalykinės srities ontologijos sudarymo procesu; panaudotos ontologijos modelio saugojimo reliacinėje duomenų bazėje galimybės, atliktas ontologijos savybių rodiklių skaičiavimas. / Web portals collects the information scattered on the internet and organizes it for easy search and access. Current Web technologies employed to build up these portals present serious limitations regarding information search, access, extraction, interpretation and processing. The Semantic Web provides a common framework that allows data to be shared, reused and processed directly and indirectly by machines. To facilitate this process, OWL have been developed as standard formats for the sharing and integration of data in the form of rich conceptual schemas called ontologies. An ontology defines a common vocabulary for researchers who need to share information in a specific domain. There are several methods for facilitating development of ontologies. Although, neither describes how to build a Semantic Web portals step by step, using the latest Web portals development tools and technologies. The main goal of this work is to propose a development methodology for building Semantic Web portals from the scratch.
