Filogenia e evolução das espécies do gênero Phrynops (Testudines, Chelidae) / Phylogeny and evolution of the species of the genus Phrynops (Testudines, Chelidae)

O gênero Phrynops abrange quatro espécies válidas, incluindo o complexo da espécies Phrynops geoffroanus, e apresenta uma filogenia historicamente mal compreendida. Esse projeto objetivou estudar este gênero, com ênfase especial nas populações de P. geoffroanus, através de análises morfométricas, morfológicas, osteológicas e moleculares, a fim de entender sua taxonomia e definir suas relações filogenéticas. Para tanto, foram coletados e emprestados espécimes das populações de P. geoffroanus de dez bacias hidrográficas brasileiras, além de P. tuberosus, P. williamsi e P. hilarii para compôr a análise. A partir disso, foi feita uma análise morfométrica linear dos espécimes das populações de P. geoffroanus, bem como uma comparação do padrão de coloração desses indivíduos com o intuito de definir as populações taxonomicamente distintas dentro da área de distribuição da espécie. Para as análises filogenéticas, foram preparados esqueletos completos e sequenciados os genes 12S, cytb, ND4, R35 e Rag 2 das espeçies de Phrynops e do seu grupo externo, considerando todos os gêneros e quase todas as espécies de Chelidae, bem como representantes de Podocnemididae e Pelomedusidae. Foram conduzidos três conjuntos distintos de análises filogenéticas com os dados moleculares e morfológicos obtidos durante este estudo: 1) uma análise de parcimônia da matriz morfológica; 2) uma análise de máxima verossimilhança das sequências moleculares; 3) uma análise de parcimônia dos dados morfológicos e moleculares concatenados. Como resultados, os dados morfométricos e osteológicos mostram que a população de P. geoffroanus da bacia do rio Paraná representa uma espécie nova, distinta das demais populações pertencentes ao complexo geoffroanus. Além disso, a população de P. geoffroanus do Rio Doce é claramente distinta das demais populações nas hipóteses moleculares. O exemplar de P. williamsi do Rio Iguaçu se distingue molecularmente do clado formado pelos demais exemplares de P. williamsi do Rio do Peixe. Os dados morfológicos e moleculares concatenados mostram que o gênero Phrynops representa um grupo monofilético. As populações de P. geoffroanus apresentaram evidências de agrupamento por bacias. Nas análises moleculares e concatenadas, P. hilarii se posiciona dentro do clado formado pelas populações de P. geoffroanus. O conjunto de dados osteológicos posiciona Mesoclemmys como grupo-irmão de Phrynops, porém, no conjunto de dados moleculares Phrynops aparece como grupo-irmão dos demais gêneros, exceto Hydromedusa e Chelus que encontram-se em uma posição mais basal nessa hipótese. Desta forma, não foi possível definir com clareza o grupo-irmão do gênero Phrynops neste estudo. Os caracteres osteológicos e moleculares evidenciaram Mesoclemmys como um grupo parafilético, pela não inclusão de Mesoclemmys hogei nesse gênero. Por fim, neste estudo, fica evidente que P. geoffroanus e P. tuberosus pertencem à calha norte do rio Amazonas e não se distinguem através do conjunto de dados osteológicos e morfométricos empregados aqui. Entretanto, uma amostragem das localidades específicas será necessária para definir objetivamente o status taxonômico de P. tuberosus / The genus Phrynops covers four valid species, including the species complex Phrynops geoffroanus. The genus has a confuse and poorly resolved taxonomic and phylogenetic history. This project aimed to study the genus Phrynops, with particular emphasis on populations of P. geoffroanus and P. tuberosus, through morphometric, morphological, osteological and molecular analyzes, in order to understand their taxonomy and provide a phylogenetic hypothesis for their interrelationships. Specimens representing populations of P. geoffroanus from ten Brazilian river basins, as well as P. tuberosus, P. williamsi and P. hilarii, were collected or loaned from institutions. A linear morphometric analysis and comparison of color patterns were performed in individuals belonging to the populations of P. geoffroanus in order to define taxonomically distinct populations within the range of the species. For the phylogenetic analyzes, complete skeletons were prepared and tissue samples were sequenced for the genes 12S, cytb, ND4, R35 and Rag 2, for most species of Phrynops and its outgroup taxa, considering all genera and almost all species of Chelidae, as well as representatives of Podocnemididae and Pelomedusidae. Three distinct sets of phylogenetic analyzes were performed with the molecular and morphological data obtained during this study:1) a parsimony analysis of the morphological matrix;2) a maximum likelihood analysis of molecular sequences;3) a parsimony analysis of the combined morphological and molecular data. As a result, the morphometric and osteological data show that the population of P. geoffroanus from the Paraná River basin is a distinct species, and therefore new. The population of P. geoffroanus from the Doce River is clearly distinct from other populations in the molecular hypotheses. The specimen of P. williamsi from the Iguaçu River separates molecularly from the clade formed by the remaining populations of P. williamsi from the Peixe River. The combined morphological and molecular data show that the genus Phrynops corresponds to a well supported monophyletic group.The sampled populations of P. geoffroanus showed evidence of grouping by distinct river basins. In both molecular and combined analyzes, P. hilarii is rooted within the clade formed by the populations of P. geoffroanus. The set of skeletal data supports Mesoclemmys as the sister group of Phrynops, while the molecular data set shows Phrynops as the sister group of a clade composed by all the remaining genera, except Hydromedusa and Chelus that are positioned more basally in the tree. Therefore, the present analysis failed to define without ambiguity the sister-group relationship of the genus Phrynops. Both osteological and molecular datasets showed Mesoclemmys as a paraphyletic group, with the exclusion of Mesoclemmys hogei from the genus. Finally, it seems evident that P. tuberosus and P. geoffroanus belong to the north region of the Amazon River and are not distinguishable by the set of osteological and morphometric data used in the present study. However, a larger sampling of specific locations will be required to objectively define the taxonomic status of P. tuberosus

Hematologia, bioquímica do sangue, parasitologia, microbiologia, algas epizoárias e histopatologia de Phrynops geoffroanus (Schweigger, 1812) (Testudinata, Chelidae), expostos a diferentes influências antrópicas no rio Uberabinha, Minas Gerais.

One area drains regional agriculture (18º58'35 S and 48º17'11 W), while the second is predominantely urban (18º5'10 S and 48º18'09 W). From December, 1999 to January 2001, field and laboratory work were accomplished characterizing the two areas as to the geographical relief, the plant life on the river margins, the air temperature and relative humidity, microbiology and several physical-chemical parameters of the water and residues of organochlorine (agricultural chemical) from the river sediment. To test differences between the two areas, climatic data, twelve physical-chemical parameters and water instantaneous speed were analyzed using the Wilcoxon test at a 5% significance level. Significant differences in the relative humidity of the air, total dissolved solids, suspended solids, electric conductivity, temperature, transparency, nitrites and turbidity of the river water were all verified. The urbanized area presented smaller conservation in the margins vegetation, lowest air relative humidity, water with larger concentrations of total dissolved solids, suspended solids, nitrite, larger electric conductivity, smaller transparency, larger turbidity, larger variety of microorganisms and organochlorate residues, being that the transheptachlor, endossulfan and atrazine indexes were higher in the agricultural area.The different human-related influences on soil use were reflected in the composition of the flora on the river s margins and in the aquatic ecosystem, resulting in high levels mainly of phosphate, nitrogen, atrazine, transheptaclor, endosulfan and BHC, as well as water bacterial contamination. The fifty eight P. geoffroanus were captured in January of 2000, being 16 males and 16 females coming from the urban area and 12 males and 14 females from the agricultural area. To collect the Phrynops two funnel traps were used containing fish, chicken viscera and bovine heart as bait. The Phrynops were transported to the Reptiles Section at the Federal University of Uberlândia where they were sexed, marked individually and housed in concrete tanks. The ectoparasites and samples of the algae attached to the shells were collected and preserved. Blood samples were drawn for the glicemia determination. The morphometric data and the blood sample for hematozoary investigations together with hematological and biochemical blood analyses were also obtained from the turtles. Later, three males and three females from each subpopulation were sacrificed by decapitation for macroscopic analysis of the organs and histopathological studies. All the blood tests were performed during the summer. Analysis of the 15 morphometric characters and of the body mass data (ANOVA and ACP, 5% significance level) evidenced an accentuated sexual dimorphism; however, the different environmental conditions to which the two subpopulations were exposed didn't interfere in the size or in the form of the turtles. The microbiological analysis of the samples collected from the oral cavity, cloaca and the neck folds showed that the subpopulation from the urban area had a larger proportion of microorganisms in relation to that of the agricultural area (x2 = 31,0; P < 0,001). There were no significant differences among the proportions of microorganisms on body parts in relation to the geographical areas (x2= 0,017; P > 0,05). In both sample the oral cavity was the most infected (x2= 1,5897; P < 0,05). In the two subpopulations, the number of species of isolated microorganisms in the turtles was superior to the number of species found in the water of the river, with a few differences in the urban area. Although in this area a larger diversity of microorganisms, in the water and in the turtles, was found. A higher percentage of occurrence of the alga Basicladia chelonum, in the turtles shells was verified in the urban sample (43,8%) in relation to the agricultural sample (30,8%). Hemogregarine intraeritrocitary gametocytes were found in 27,6% of blood smears from P. geoffroanus, with a larger occurrence in the urban area. Ectoparasitism by leeches (Placobdella bistriata) was verified only in specimens from the urban area, and of the nine, six turtles were also infected by hemogregarine. Among the P. geoffroanus sacrificed, specimens from the agricultural area presented smaller amounts of fatty reserve tissue, more compact skeletal muscles, a more strong red color and dilation of some lung alveoli, indicating more physical activity compared with those from the urban area. The turtles from the urban area presented larger alterations as evidenced by the histopathological analysis, such as: discreet edema in the encephalon and the heart; cartilaginous formation in pulmonary alveolus and focal pneumonia; liver, pancreas, splein, kidney and stomach with inflamatory infiltrate and suggestive trematodes granulomas; hemossiderose in the liver and spleen. The hematological and biochemical data from the blood analysis were analyzed by analysis of variance (ANOVA, at a 5% significant level) for hypothesis testing and a difference of averages test (Tukey s test) to verify differences among the averages. Significant differences in relation to sex were obtained in 50% of the blood parameters (leucocyte numbers, eosinophyle percentages, hemoglobin, iron, phosphate, medium corpuscular hemoglobin, medium corpuscular hemoglobin concentration, prothrombin time, fibrinogen, ionized calcium, total proteins, albumin, alpha 1, alpha 2, gamma globulin, cholesterol high density lipoprotein and very low density lipoprotein, triglyceride, creatine kinase, gamma glutamil transferin, amilasys, lipase, uric acid, chloride and magnesium). In relation to the two geographical areas, 88% of the parameters were statistically significant (eritrocyte numbers, leucocytes, eosinophils, monocytes, heterophils, azurophils, hemoglobin, hematocrite, medium corpuscular hemoglobin and all the biochemical parameters, except to clotting blood). For the interaction area-sex analysis, 16% of the variables were significantly different (medium corpuscular hemoglobin, medium corpuscular hemoglobin concentration, leucocytes number, alkaline phosphatases, uric acid, beta globulin, iron and magnesium) between the two samples. The impact due to human intervention in terms of the different soil uses apparently caused environmental degradation interfering in most of the hematological and biochemical blood parameters between the two turtle subpopulations of the Uberabinha River. That, associated with the histopathological results, parasitological and the microbiological findings indicates that the environmental conditions, especially those related to the aquatic ecosystem, interfered in the physiology and could compromise the health of these turtles. Although the reference intervals of hemogram, electrophoresis and biochemical blood parameters used in the present study are important tools in the health control and diagnosis animal of diseases, they should not be considered as standard for this species, whose geographical distribution is wide, inhabiting ponds, streams and rivers with very diversified hidrogeochemical characteristics. Keywords: Phrynops geoffroanus, environmental contamination, morphometry, bacteria, epizoary algae, parasite infestation, hematology, histopathology, clinical biochemistry. / Foram realizados estudos comparativos da morfometria, microbiologia, hematologia, bioquímica do sangue, histopatologia, e pesquisa de hematozoários, de ectoparasitos e de algas epizoárias entre duas subpopulações do cágado Phrynops geoffroanus capturadas em duas áreas do rio Uberabinha com diferentes usos do solo no entorno, uma área com predomínio agropecuário (18º58 35 S e 48º17 11 W) e uma outra com predomínio urbano (18º55 10 S e 48º18 09 W). De dezembro de 1999 a janeiro de 2001 foram realizados trabalhos de campo e de laboratório, tendose caracterizado as duas áreas quanto ao relevo, composição da flora nas margens do rio, temperatura e umidade relativa do ar, microbiologia e vários parâmetros físicos e químicos da água e dos resíduos de defensivos agrícolas organoclorados no sedimento do rio. Para testar as diferenças entre as áreas foram utilizadas as amostragens de duas variáveis climáticas, de doze parâmetros físicos e químicos e de velocidade instantânea da água, utilizando-se o teste de Wilcoxon com nível de significância de 5%. Foram constatadas diferenças significativas na umidade relativa do ar, nos sólidos totais dissolvidos, nos sólidos suspensos, na condutividade elétrica, na temperatura, na transparência, nos nitritos e na turbidez da água do rio. A área urbanizada apresentou menor conservação da vegetação das margens, umidade relativa do ar mais baixa, água com maiores concentrações de sólidos totais dissolvidos, de sólidos suspensos, de nitritos, maior condutividade elétrica, menor transparência, maior turbidez, maior variedade de microorganismos e de resíduos organoclorados, e na área agropecuária os índices de transheptaclor, endossunfan e atrazina foram mais elevados. Os cinqüenta e oito P. geoffroanus foram capturados em janeiro de 2000, sendo 16 machos e 16 fêmeas procedentes da área de predomínio urbano e 12 machos e 14 fêmeas da área de predomínio agropecuário. Utilizou-se em cada área duas armadilhas tipo covo contendo peixes, vísceras de frango e coração bovino como iscas. Transportados para o Setor de Répteis da Universidade Federal de Uberlândia foram sexados, marcados individualmente e alojados em tanques de alvenaria. Foram coletados os ectoparasitos, as amostras das algas epizoárias da carapaça e as amostras de sangue para a determinação da glicemia. Em seguida, foram obtidos os dados morfométricos e da massa corpórea dos P. geoffroanus coletando-se novas amostras de sangue destinadas às investigações de hematozoários, às análises hematológicas, aos testes de bioquímica do sangue, à determinação das curvas glicêmicas e às eletroforeses de proteínas. Posteriormente, foram sacrificados por decapitação três machos e três fêmeas de cada subpopulação objetivando-se as observações macroscópicas dos órgãos e a confecção das lâminas para os estudos histopatológicos, sendo todos os exames de sangue realizados durante o verão. A análise de 15 caracteres morfométricos e da massa corpórea (ANOVA e ACP, nível de significância de 5%) evidenciou um acentuado dimorfismo sexual, sendo que as diferentes condições ambientais a que as duas subpopulações estavam expostas não interferiram no tamanho e na forma dos cágados. As análises microbiológicas de amostras coletadas na cavidade oral, na cloaca e no tegumento do pescoço dos P. geoffroanus mostraram que a subpopulação da área urbana tem maior proporção de microorganismos em relação aos da área agropecuária (&#967;2 = 31,0; P < 0,001), não tendo ocorrido diferença significativa entre as partes do corpo em relação às áreas (&#967;2 = 0,017; P > 0,05), sendo que nas duas áreas, a cavidade oral dos cágados foi a mais infectada (&#967;2 = 1,5897; P < 0,05). Nas duas subpopulações, o número de espécies de microorganismos isolados nos cágados foi superior ao número de espécies encontradas na água do rio, com pouca diferença na área mais urbanizada, embora nesta área tenha ocorrido maior diversidade de microorganismos, tanto na água quanto nos cágados. Maior porcentagem de ocorrência da alga Basicladia chelonum na carapaça dos cágados foi verificada na área de predomínio urbano (43,8%) em relação aos da área de predomínio agropecuário (30,8%). Nos esfregaços sangüíneos foram encontrados gametócitos intraeritrocitários de hemogregarinas em 27,6% dos P. geoffroanus, com maior predominância nos da área urbanizada. Ectoparasitismo por sanguessugas Placobdella bistriata foi constatado somente em espécimes da área urbanizada, sendo que dos nove cágados, seis estavam também infectados por hemogregarinas. Dos P. geoffroanus necropsiados, os da área agropecuária apresentaram menores quantidades de tecido adiposo de reserva, músculos esqueléticos mais compactos e com coloração mais avermelhada e dilatação de alguns alvéolos pulmonares, indicando serem mais ativos em relação aos da área urbanizada. As análises histopatológicas evidenciaram maior número de alterações nos espécimes da área urbanizada, tendo-se verificado a ocorrência de edema discreto no encéfalo e no coração, alvéolos pulmonares com formações cartilaginosas e pneumonia focal; fígado, pâncreas, baço, rins e estomago com infiltrado inflamatório e cistos com larvas sugestivas de trematódeos; fígado e baço com hemossiderose. Os dados de hematologia e de bioquímica do sangue foram analisados por análise de variância (ANOVA, nível de significância de 5%) para teste de hipótese e teste de médias (teste de Tukey) para verificar diferenças entre as médias. Diferenças significativas em relação ao sexo foram obtidas em 50% dos parâmetros sangüíneos (número de leucócitos, porcentagem de eosinófilos, hemoglobina, ferro, fósforo, hemoglobina corpuscular média, concentração de hemoglobina corpuscular média, tempo de protrombina, fibrinogênio, cálcio ionizável, proteínas totais, albumina, alfa 1, alfa 2, gama globulina, frações HDL e VLDL de colesterol, triglicérides, creatina quinase, gama glutamil transferase, amilase, lípase, ácido úrico, cloreto e magnésio). Em relação à área, 88% dos parâmetros foram estatisticamente significativos (número de eritrócitos, leucócitos, eosinófilos, monócitos, heterófilos, azurófilos, hemoglobina, hematócrito, hemoglobina corpuscular média e todos os parâmetros bioquímicos, exceto para o tempo de coagulação do sangue). Na interação área-sexo, 16% diferiram significativamente (hemoglobina corpuscular média, concentração de hemoglobina corpuscular média, número de leucócitos, alamina transferase, ácido úrico, beta globulina, ferro e magnésio). O impacto decorrente da intervenção humana pelos diferentes usos do solo aparentemente causou degradações ambientais, determinando diferenças significativas na maioria dos resultados hematológicos e bioquímicos do sangue dos cágados do rio Uberabinha. Estes resultados, associados as histopatologias, ao parasitismo e à microbiologia encontrada nestes cágados, indicam que as condições ambientais, especialmente as relacionadas ao ecossistema aquático, interferem na fisiologia, podendo comprometer a saúde destes animais. Embora os intervalos de referência do hemograma, do perfil eletroforético e bioquímica do sangue sejam uma importante ferramenta no monitoramento da saúde e no diagnóstico de doenças nos animais, eles não podem ser considerados padrão para esta espécie, cuja distribuição geográfica é ampla, podendo estes cágados habitar lagoas, riachos e rios com características hidrogeoquímicas muito diversificadas. O quelônio Phrynops geoffroanus aparece freqüentemente associado a cursos d\'água poluídos, sem que, no entanto, sejam conhecidos aspectos que descrevam sua atividade imune celular frente situações adversas (efeitos antrópicos). A citometria de fluxo permite mensurar características estruturais e imunológicas de células em suspensão submetidas a um fluxo contínuo, sendo capaz de analisar inúmeros tipos celulares. Neste trabalho foi desenvolvida metodologia para estudar e avaliar a função celular de leucócitos sangüíneos do Phrynops geoffroanus através da fagocitose e burst oxidativo espontâneo e induzido utilizando a citometria de fluxo. Para definir a metodologia foram utilizadas 86 amostras sangüíneas de Phrynops geoffroanus encontrados em ambientes antrópicos do rio Piracicaba, ribeirão Piracicamirim e Fundação Parque Zoológico de São Paulo como grupo de comparação. Estes resultados foram complementados pelo perfil hematológico. O processamento incluiu transporte e conservação das amostras em RPMI 1640 Gibco® para posterior utilização de Ficoll-PaqueTM PLUS como agente separador entre leucócitos e hemácias, utilizando-se centrifugações refrigeradas (18°C) com acelerações e desacelerações graduais. A fim de analisar o impacto da contaminação química e orgânica na espécie de estudo, foram realizadas análises ecotoxicológicas visando avaliar a ação de alguns dos principais componentes envolvidos com a proteção ao estresse oxidativo e metabolismo de detoxificação de fase I e II, e capacidade antioxidante. Os resultados indicam a influência de efluentes domésticos e industriais no metabolismo de detoxificação e estresse oxidativo. No entanto, apesar do aumento da atividade EROD e o efeito da atividade da GST, os valores médios de GST na área urbana estão dentro daqueles esperados em condição de hipóxia descritos na literatura. Esta observação sugere que o aumento da GST em resposta à produção de ERO pela presença de poluentes, aumenta a rede de defesa antioxidante, controlando os danos oxidativos causados pela hipóxia e pela reperfusão. Com o intuito de estimar a condição, que reflete a aptidão (fitness) individual do animal, foi avaliada a relação matemática entre peso e comprimento, que indicam alterações na forma do corpo e no incremento em peso, que permitem inferências sobre a saúde e o bem-estar animal. Os dados gerados informam diferenças em condição que estão associadas à área de avaliação, mas também ao sexo e período reprodutivo, e gradiente de contaminação indicando forte influência de estressores ambientais na fisiologia dos espécimes analisados. A partir da avaliação da estrutura genética entre populações do rio Preto e do córrego... / The objective of this study was to evaluate the role of human activity influence, such as environmental contamination and habitat changes as drivers for changing physiological, biochemical, and genetic diversity among Geoffroy’s side-necked turtle populations in one of the most impacted watersheds in southeastern Brazil. In order to analyze the impact of chemical and organic contamination, ecotoxicological analyses were performed to assess the action of some of the major components involved in oxidative stress protection and phase I and II of metabolism detoxification, and antioxidant capacity. The results indicate the influence of domestic and industrial effluents in detoxification metabolism and oxidative stress. However, in spite of increased activity and effect of EROD activity of GST, GST-average values in the urban area conform with those expected in hypoxia condition in the literature. This observation suggests that increased GST in response to the ROS production by the presence of pollutants increases the antioxidant defense network, controlling the oxidative damage caused by hypoxia and reperfusion. In order to estimate the condition, which reflects the individual ability (fitness), we evaluated the mathematical relationship between weight and length, indicating that changes in body shape and weight increase, allowing inferences about the health and well animal welfare. The data generated indicate differences in conditions that are associated with the area, but also with sex and reproductive period, and contamination gradient indicating strong influence of environmental stressors on the physiology of the specimens. The chemical communication sign may have a highest importance in the space orientation and sexual behavior displayed by the chelonian during the reproductive period. There are not histological and morphological studies of the Brazilian chelonian species tegumental appendices, like Hydromedusa maximiliani and Phrynops geoffroanus. So, the actual study had the goal to describe and compare morphologically and histologically the tegumental appendices from two Brazilian tortoise species which belong to the Chelidae family. For this study, it was used adult specimens from the Cole??o Herpetol?gica do Departamento de Zoologia da Universidade Federal de Juiz de Fora of H. maximiliani (2 males and 2 females) and P. geoffroanus (4 males and 4 females), totalizing 12 specimens. The analysis of the external surface neck, head and chin region was done. The animals were dissected using stereoscopic microscope to take samples out of the epidermis and dermis from the neck and head region. To the microscopic analysis, it was done the routine techniques that are used in the histological studies. In P. geoffroanus, the keratinized epidermis layer is thicker when compared to the same epidermis layer of H. maximiliani. It was observed that in P. geoffroanus and H. maximiliani these regions may present epidermis invaginations with the detachments of the most external keratin layers. The histological and histochemical study done in the epidermis cuts of the specimens H. maximiliani and P. geoffroanus haven?t evidenced the presence of submandibular gland. The PAS method hasn?t shown the presence of neutral mucopolysaccharides in this submandibular gland, which are products part of the secretions produced by the exocrine glands. In this study, it was observed that the dewlap presented in P. geoffroanus are cylindrical tegumental appendices presented in the mandibular symphysis posterior region, which occurs in pairs, constituting of epidermis and dermis, and with a great concentration of nervous terminations. In the actual study, the P. geoffroanus specimens macroscopically presented conical shape tubers, rarely pointed and also with verrucous shape, visible in the dorsal and lateral region, from the anterior part up to the posterior part of the neck, not presented in the ventral region of the neck, which presents slots, grooves and depressions in the epidermis. In the actual study, it wasn?t observed morphological and histological differences regarding the tubers when compared by gender. In H. maximiliani the tubers are irregularly distributed in the lateral and dorsal regions of the neck, being absent in the region of the head and in the ventral region of the neck. They present pointed and tray shape, different from what was observed in the P. geoffroanus specimens. The tubers presented in the two studied species are different in macroscopic terms, but all of them present a random disposition and are also absent in the ventral region of the neck. For the two types of tegumental annexes (tubers, dewlap) analyzed in this study, it was verified that there are not differences between the genders. / Os quel?nios empregam sinais t?teis, visuais, olfativos e qu?micos durante as intera??es sociais. A comunica??o usando sinais qu?micos pode ter uma import?ncia maior na orienta??o espacial e comportamento sexual exibidos pelos quel?nios durante o per?odo reprodutivo. S?o inexistentes os estudos histol?gicos e morfol?gicos dos ap?ndices tegument?rios das esp?cies de quel?nios brasileiros, como Hydromedusa maximiliani e Phrynops geoffroanus. Dessa forma, o presente estudo teve como objetivos descrever e comparar morfol?gica e histologicamente os anexos tegumentares de duas esp?cies brasileiras de c?gados pertencentes ? Fam?lia Chelidae. Para esse estudo foram utilizados esp?cimes adultos de H. maximiliani (dois machos e duas f?meas) e P. geoffroanus (quatro machos e quatro f?meas), totalizando 12 esp?cimes, provenientes da Cole??o Herpetol?gica do Departamento de Zoologia da Universidade Federal de Juiz de Fora Foi realizada a an?lise da superf?cie externa da regi?o do pesco?o, cabe?a e da regi?o mentoniana. Os animais foram dissecados sob microsc?pio estereosc?pico para retirada de amostras da epiderme e derme das regi?es da cabe?a e pesco?o. Para an?lise microsc?pica foram realizados as t?cnicas de rotina utilizadas nos estudos de histologia. Em P. geoffroanus a camada queratinizada da epiderme ? mais espessa quando comparada com a mesma camada na epiderme de H. maximiliani. Foi observado em P. geoffroanus e H. maximiliani que essas regi?es podem apresentar invagina??es da epiderme com o desprendimento das camadas mais externas de queratina. O estudo histol?gico e histoqu?mico realizado nos cortes da epiderme de ambas esp?cies de, n?o evidenciou a presen?a da gl?ndula submandibular. O m?todo de PAS n?o mostrou a presen?a de mucopolissacar?deos neutros nessa regi?o submandibular, que s?o produtos que fazem parte das secre??es produzidas por gl?ndulas ex?crinas. Nesse estudo observou-se que as barbelas presentes em P. geoffroanus s?o ap?ndices tegumentares cil?ndricos presentes na regi?o posterior da s?nfise mandibular, que ocorrem aos pares, constituindo-se de epiderme e derme, e com grande concentra??o de termina??es nervosas. No presente estudo, os esp?cimes de P. geoffroanus macroscopicamente apresentaram tub?rculos de forma c?nica, raramente pontiagudos, e tamb?m de formato verrucoso, vis?veis na regi?o dorsal e lateral desde a parte anterior at? a parte posterior do pesco?o, n?o estando presentes na regi?o ventral do pesco?o, que apresenta ranhuras, estrias e depress?es da epiderme. No presente estudo n?o foram observadas diferen?as morfol?gicas e histol?gicas em rela??o aos tub?rculos quando comparados por sexo. Em H. maximiliani os tub?rculos est?o distribu?dos de forma irregular nas regi?es laterais e dorsais do pesco?o, estando ausentes na regi?o da cabe?a e na regi?o ventral do pesco?o. Apresentam formato pontiagudo e de tabuleiro, diferente do que foi observado nos esp?cimes de P. geoffroanus. A espécie Phrynops geoffroanus, popularmente conhecida como “Cágado-de-Barbelas”, tem se proliferado em ambiente poluído, sujeita a condições adversas que podem influenciar nos seus hábitos de vida e condições fisiológicas. Estudos que monitorem os efeitos da contaminação ambiental são fundamentais para o conhecimento da biologia da espécie e delineamento de estratégias de conservação efetivas. Deste modo, as análises dos parâmetros hematológicos e bioquímicos mostram-se importantes na avaliação da saúde de animais silvestres e riscos para o animal e ecossistema. O objetivo do trabalho foi verificar a influência do ambiente na fisiologia de uma população de Phrynops geoffroanus por meio da avaliação da capacidade antioxidante e respostas aos estressores ambientais, em comparação com espécimes de local sob condições controladas. Foram avaliadas, por meio do hemograma completo, dosagem de espécies reativas ao ácido tiobarbitúrico (TBARS), ensaio da capacidade antioxidante em equivalência ao Trolox (TEAC) e atividades das enzimas antioxidantes catalase e glicose-6-fosfato desidrogenase (G6PDH), amostras sanguíneas de sessenta espécimes, sendo trinta provenientes do córrego Felicidade, ambiente poluído, no perímetro urbano de São José do Rio Preto-SP; e trinta do criatório “Reginaldo Uvo Leone”, Tabapuã-SP, local sob condições controladas, cujas amostras constituem o grupo controle dos experimentos. Houve grande variação nos parâmetros hematológicos de Phrynops geoffroanus de ambiente urbano. Os valores de glóbulos vermelhos e hemoglobina foram significantemente menores que os observados em animais do criatório (p = 0,0004; p = 0,0371, respectivamente)... / The turtles are among the most endangered vertebrates groups, and the main threats to populations are the environmental pollution and degradation. The species Phrynops geoffroanus, popularly known as Geoffroy's side-necked turtle, has proliferated in polluted environments, subject to adverse conditions that could influence their living habits and physiological conditions. Studies that monitor the effects of environmental pollution are the key to understand the species biology and design effective conservation strategies. Thus, the analysis of hematological and biochemical parameters showed to be important in assessing the health of wild animals and for the animal and ecosystem risks. This work aimed to assess the environmental influence on the physiology of a Phrynops geoffroanus population through the evaluation of antioxidant status and responses to environmental stressors, compared to specimens from a place under controlled conditions. They were evaluated by hemogram, measuring thiobarbituric acid reactive species assay (TBARS), Trolox-equivalent antioxidant capacity evaluation (TEAC) and the activities of antioxidant enzymes, catalase and glucose-6-phosphate dehydrogenase (G6PDH), blood samples of sixty specimens, thirty from the Felicidade stream, polluted environment, within the São José do Rio Preto city, and thirty from the Reginaldo Uvo Leone breeding farm, Tabapuã-SP, place under controlled conditions, whose samples constitute the control group of the experiments. There was a wide variation in hematological parameters of Phrynops geoffroanus from the urban environment. The red blood cells and hemoglobin values were significantly less than those observed in animals from breeding farm (p = 0.0004; p = 0.0371, respectively). A fim de analisar o impacto da contaminação química e orgânica na espécie de estudo, foram realizadas análises ecotoxicológicas visando avaliar a ação de alguns dos principais componentes envolvidos com a proteção ao estresse oxidativo e metabolismo de detoxificação de fase I e II, e capacidade antioxidante. Os resultados indicam a influência de efluentes domésticos e industriais no metabolismo de detoxificação e estresse oxidativo. No entanto, apesar do aumento da atividade EROD e o efeito da atividade da GST, os valores médios de GST na área urbana estão dentro daqueles esperados em condição de hipóxia descritos na literatura. Esta observação sugere que o aumento da GST em resposta à produção de ERO pela presença de poluentes, aumenta a rede de defesa antioxidante, controlando os danos oxidativos causados pela hipóxia e pela reperfusão. Com o intuito de estimar a condição, que reflete a aptidão (fitness) individual do animal, foi avaliada a relação matemática entre peso e comprimento, que indicam alterações na forma do corpo e no incremento em peso, que permitem inferências sobre a saúde e o bem-estar animal. Os dados gerados informam diferenças em condição que estão associadas à área de avaliação, mas também ao sexo e período reprodutivo, e gradiente de contaminação indicando forte influência de estressores ambientais na fisiologia dos espécimes analisados. (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: The objective of this study was to evaluate the role of human activity influence, such as environmental contamination and habitat changes as drivers for changing physiological, biochemical, and genetic diversity among Geoffroy's side-necked turtle populations in one of the most impacted watersheds in southeastern Brazil. In order to analyze the impact of chemical and organic contamination, ecotoxicological analyses were performed to assess the action of some of the major components involved in oxidative stress protection and phase I and II of metabolism detoxification, and antioxidant capacity. The results indicate the influence of domestic and industrial effluents in detoxification metabolism and oxidative stress. However, in spite of increased activity and effect of EROD activity of GST, GST-average values in the urban area conform with those expected in hypoxia condition in the literature. This observation suggests that increased GST in response to the ROS production by the presence of pollutants increases the antioxidant defense network, controlling the oxidative damage caused by hypoxia and reperfusion. In order to estimate the condition, which reflects the individual ability (fitness), we evaluated the mathematical relationship between weight and length, indicating that changes in body shape and weight increase, allowing inferences about the health and well animal welfare. The data generated indicate differences in conditions that are associated with the area, but also with sex and reproductive period, and contamination gradient indicating strong influence of environmental stressors on the physiology of the specimens. A espécie Phrynops geoffroanus, popularmente conhecida como "Cágado-de-Barbelas", tem se proliferado em ambiente poluído, sujeita a condições adversas que podem influenciar nos seus hábitos de vida e condições fisiológicas. Estudos que monitorem os efeitos da contaminação ambiental são fundamentais para o conhecimento da biologia da espécie e delineamento de estratégias de conservação efetivas. Deste modo, as análises dos parâmetros hematológicos e bioquímicos mostram-se importantes na avaliação da saúde de animais silvestres e riscos para o animal e ecossistema. O objetivo do trabalho foi verificar a influência do ambiente na fisiologia de uma população de Phrynops geoffroanus por meio da avaliação da capacidade antioxidante e respostas aos estressores ambientais, em comparação com espécimes de local sob condições controladas. Foram avaliadas, por meio do hemograma completo, dosagem de espécies reativas ao ácido tiobarbitúrico (TBARS), ensaio da capacidade antioxidante em equivalência ao Trolox (TEAC) e atividades das enzimas antioxidantes catalase e glicose-6-fosfato desidrogenase (G6PDH), amostras sanguíneas de sessenta espécimes, sendo trinta provenientes do córrego Felicidade, ambiente poluído, no perímetro urbano de São José do Rio Preto-SP; e trinta do criatório "Reginaldo Uvo Leone", Tabapuã-SP, local sob condições controladas, cujas amostras constituem o grupo controle dos experimentos. Houve grande variação nos parâmetros hematológicos de Phrynops geoffroanus de ambiente urbano. Os valores de glóbulos vermelhos e hemoglobina foram significantemente menores que os observados em animais do criatório (p = 0,0004; p = 0,0371, respectivamente)... The species Phrynops geoffroanus, popularly known as "Geoffroy's side-necked turtle", has proliferated in polluted environments, subject to adverse conditions that could influence their living habits and physiological conditions. Studies that monitor the effects of environmental pollution are the key to understand the species biology and design effective conservation strategies. Thus, the analysis of hematological and biochemical parameters showed to be important in assessing the health of wild animals and for the animal and ecosystem risks. This work aimed to assess the environmental influence on the physiology of a Phrynops geoffroanus population through the evaluation of antioxidant status and responses to environmental stressors, compared to specimens from a place under controlled conditions. They were evaluated by hemogram, measuring thiobarbituric acid reactive species assay (TBARS), Trolox-equivalent antioxidant capacity evaluation (TEAC) and the activities of antioxidant enzymes, catalase and glucose-6-phosphate dehydrogenase (G6PDH), blood samples of sixty specimens, thirty from the Felicidade stream, polluted environment, within the São José do Rio Preto city, and thirty from the "Reginaldo Uvo Leone" breeding farm, Tabapuã-SP, place under controlled conditions, whose samples constitute the control group of the experiments. There was a wide variation in hematological parameters of Phrynops geoffroanus from the urban environment. The red blood cells and hemoglobin values were significantly less than those observed in animals from breeding farm (p = 0.0004; p = 0.0371, respectively).
