Rob (Asteraceae) ? uma esp?cie de Asteraceae conhecida popularmente como arnica-do-mato, arnica-do-campo ou simplesmente arnica. E utilizada na medicina popular contra machucados e dores no corpo, porem a literatura carece de informa??es cient?ficas, encontrando-se apenas um artigo sobre estudo fitoqu?mico. Com o objetivo de contribuir para o estudo de P. brasiliensis, esse trabalho relata o estudo fitoqu?mico e ensaios biol?gicos que investigaram a atividade antiinflamat?ria e antioxidante de extratos das folhas da planta. As folhas foram coletadas na cidade de Diamantina, no Campus JK da UFVJM. Uma parte das folhas ainda frescas foi utilizada para extra??o de ?leo essencial, e outra parte do material vegetal foi seco e submetido ? extra??o com solventes de diferentes polaridades ? hexano, acetato de etila, etanol e ?gua ? obtendo-se quatro extratos. Os constituintes do ?leo essencial foram identificados por CG-EM, encontrando-se 25 terpenos, sendo os majorit?rios os monoterpenos ?-tujeno (17,21%) e ?-pineno (32,61%). Os extratos foram submetidos a t?cnicas cromatogr?ficas cl?ssicas e suas fra??es foram analisadas por CG-EM, IV, RMN de 1H e 13C, identificando-se o diterpeno acido cauren?ico; os triterpenos ?-amirina, acetato de ?-amirina, ?-amirina, acetato de ?-amirina, lupeol, acetato de lupeol, pseudotaraxasterol, taraxasterol; e possivelmente uma lactona sesquiterp?nica. Em ensaios de triagem fitoqu?mica realizados com os extratos, foram detectadas as classes de metab?litos secund?rios: cumarinas, flavon?ides, taninos condens?veis, antocianinas, antraquinonas, saponinas, compostos redutores e triterpenos/ester?ides. A esp?cie apresentou um potencial antiinflamat?rio, uma vez que os extratos aquoso e etan?lico modularam a produ??o da citocina IFN-?, envolvida diretamente na inicializa??o e amplifica??o da resposta inflamat?ria. Os extratos apresentaram baixo potencial antioxidante, nas concentra??es avaliadas e nos ensaios de atividade de retirada de radical, e de poder de redu??o do ?on met?lico Fe3+, apesar dos extratos aquoso e etan?lico possu?rem compostos fen?licos. Sendo assim, o presente trabalho foi uma contribui??o para o estudo fitoqu?mico e de atividades biol?gicas desta planta. / Disserta??o (Mestrado) ? Programa de P?s-Gradua??o em Qu?mica, Universidade Federal dos Vales do Jequitinhonha e Mucuri, 2012. / ABSTRACT Pseudobrickellia brasiliensis (Spreng.) R.M. King & H. Rob (Asteraceae) is a species of Asteraceae popularly known as arnica-do-mato, arnica-do-campo or arnica simply. It is used in folk medicine against wounds and body aches, but there lack of scientific literature and found only one article on phytochemical study. Aiming to contribute to the study of P. brasiliensis, this work reports the phytochemical and biological tests that investigated the anti-inflammatory and antioxidant activity of extracts of the leaves. The leaves were collected in the city of Diamantina, in the Campus JK of UFVJM. Part of the leaves still fresh, were used for extraction of essential oil, and another part of the plant material was dried and subjected to extraction with solvents of different polarities - hexane, ethyl acetate, ethanol and water - resulting in four extracts. The constituents of the essential oil were identified by GC-MS and was found 25 terpenes, of which the major was the monoterpenes ?-thujene (17.21%) and ?-pinene (32.61%). The extracts were subjected to classical chromatographic techniques and their fractions were analyzed by GC-MS, IR, 1H and 13C NMR, identifying the diterpene kaurenoic acid, triterpenes ?-amyrin acetate, ?-amyrin, ?-amyrin, ?-amyrin acetate, lupeol acetate, lupeol, pseudotaraxasterol, taraxasterol, and possibly a sesquiterpene lactone. In tests conducted with phytochemical extracts were detected classes of secondary metabolites, coumarins, flavonoids, condensed tannins, anthocyanins, anthraquinones, saponins, reducing compounds and triterpenes/steroids. The species had an anti-inflammatory potential, because the aqueous and ethanol extracts modulate the production of the cytokine IFN-?, directly involved in the startup and amplification of the inflammatory response. The extracts showed lower antioxidant potential, and concentrations evaluated in the trials of withdrawal of radical activity, and power reduction of Fe3+ metal ion, despite the aqueous and ethanol extracts possess phenolic compounds. Thus, the present work was a contribution to the study phytochemical and biological activities of this plant.

Nesse contexto, o uso de biomassas para a produ??o de combust?veis l?quidos que alimentem motores ? combust?o permite sustentar pr?ticas antigas com novos benef?cios. Dessa forma, a presente disserta??o teve por objetivo avaliar o potencial da torta de maca?ba, um coproduto da ind?stria de ?leos vegetais, como insumo para produ??o de bioetanol. Tal objetivo foi trabalhado atrav?s de estudos de sacarifica??o da torta de maca?ba com posterior processo fermentativo. A biomassa em quest?o foi caracterizada quimicamente, o que indicou a presen?a de aproximadamente 50% de carboidratos, distribu?dos em, 23,16 ? 0,95 % de amido, 11,49 ? 1,08% de celulose, 9,6 ? 0,79% de hemicelulose e 11,48 ? 0,62 % de a??cares sol?veis. Para a primeira interven??o da despolimeriza??o dos polissacar?deos presentes na torta de maca?ba foi realizado um planejamento experimental fatorial 25-1 com 5 fatores e 4 pontos centrais com as seguintes vari?veis: concentra??o de ?cido sulf?rico, tempo de pr?-tratamento ?cido, raz?o s?lido/l?quido, concentra??o de amiloglicosidase e concentra??o de celulase. Na sequ?ncia, o processo de sacarifica??o foi otimizado com o uso de delineamento composto central rotacional 23, com 3 fatores, 4 pontos centrais e 6 pontos axiais, onde foram avaliadas as concentra??es de ?cido sulf?rico, amiloglicosidase e celulase. Um hidrolisado obtido em condi??o otimizada com 92% de efici?ncia foi submetido ? destoxifica??o com carv?o ativado e, em seguida, foram realizados ensaios de fermentabilidade conduzidos como as leveduras Saccharomyces cerevisiae e Pichia stipitis. A levedura S. cerevisiae se mostrou resistente ao hidrolisado n?o destoxificado, apresentando YP/S de 0,49. P. stipitis mostrou-se suscept?vel ao meio n?o destoxificado, mas foi capaz de converter 99% dos a??cares redutores presentes no meio destoxificado. A torta de maca?ba apresentou potencial consider?vel para produ??o de bioetanol. A partir dos dados de bancada ? poss?vel estimar uma produ??o de 104l de etanol por tonelada de torta de maca?ba. / Disserta??o (Mestrado) ? Programa de P?s-Gradua??o em Qu?mica, Universidade Federal dos Vales do Jequitinhonha e Mucuri, 2011. / ABSTRACT For the maintenance of economic and socio-cultural practices built on the exploitation and processing of oil it is necessary to create alternatives that do not break so tight with current industrial practices. In this context, the use of biomass to produce liquid fuels that are fed into to combustion engines allows to sustain old practices with new benefits. Thus, this dissertation aimed to evaluate the potential of macaw palm coconut cake, a co-product of vegetable oil industry, as an input for bioethanol production. This goal was worked out through studies of the process of saccharification of macaw palm coconut cake with subsequent fermentation. The biomass in question was characterized chemically, what indicated the presence of approximately 50% of carbohydrates, among which, 23,16 ? 0,95% of starch, 11,49 ? 1,08% of cellulose, 9,6 ? 0,79% of hemicellulose and 11,48 ? 0,62% of soluble sugars. For the first intervention of deconstruction of the polysaccharides present in the macauba cake it was held a 25-1 factorial experimental design with five factors and four central points with the following variables: concentration of sulfuric acid, time of acid pretreatment, solid / liquid ratio, amyloglucosidase and cellulose concentrations. Following, the saccharification process was optimized using central composite rotational design 23, with three factors, four central points and six axial points, whereas the concentrations of sulfuric acid, cellulose and amyloglucosidase were evaluated. A hydrolyzed obtained in optimum condition with 92% efficiency was subjected to detoxification with activated charcoal and then, fermentability assays were carried out with yeasts Saccharomyces cerevisiae and Pichia stipitis. The yeast S. cerevisiae has proved resilient to the non-detoxified hydrolyzate and presented YP/S of 0.49. P. stipitis was found to be susceptible to the non-detoxified medium, but it was able to convert 99% of reducing sugars present in the detoxified medium. The macaw palm coconut cake showed considerable potential for bioethanol production. From the lab bench data it is possible to estimate a production of 104 l of ethanol per ton of macaw palm coconut cake.

Por?m, a hidrazina ? um material t?xico, sendo, portanto, necess?rio um m?todo sens?vel para a sua determina??o. Neste trabalho um sensor foi desenvolvido para determina??o de hidrazina mediante modifica??o de um eletrodo de carbono v?treo com ftalocianina de cobalto adsorvido em nanotubos de carbono de paredes m?ltiplas. A caracteriza??o eletroqu?mica e de superf?cie foi feita por voltametria c?clica, microscopia eletr?nica de varredura, FT-IR e Espectroscopia de Energia dispersiva de Raios-X. Os estudos de superf?cie deixaram evidente que os NTCPM e a CoPc formam um material comp?sito. Os estudos eletroqu?micos mostraram a ocorr?ncia de processos redox de superf?cie e ainda h? ind?cios de uma limita??o da transfer?ncia de carga. O n?mero de el?trons para a rea??o de oxida??o de hidrazina bem como a constante de velocidade heterog?nea foram estimados a partir dos coeficientes linear e angular das curvas de Koutecky-Levich sendo, respectivamente, 4,1 e 1,51 x 103 M-1s-1. Os par?metros operacionais de tr?s t?cnicas eletroanal?ticas (Amperometria, VPD e VOQ) foram otimizados em rela??o ? determina??o de hidrazina com o uso do ECV/NTCPM/CoPc, sendo que a VOQ apresentou maior sensibilidade, tendo sido, portanto, escolhida para a determina??o da hidrazina em ?gua de caldeira. Uma curva anal?tica final para a determina??o de hidrazina em amostra da ?gua de caldeira foi feita utilizando-se eletrodos impressos, sob condi??es experimentais otimizadas, apresentando resposta linear no intervalo de concentra??o de 99 a 7630 ?mol L-1, com R igual a 0,999. Foram obtidos valores de LD e LQ iguais a 50 e a 99 ?mol L-1, respectivamente. O sensor permitiu uma boa recupera??o para a hidrazina nas amostras de ?guas de caldeiras analisadas, com valores de recupera??o compreendidos entre 98,41 e 103,62%, atestando a exatid?o do m?todo proposto. / Disserta??o (Mestrado) ? Programa de P?s-Gradua??o em Qu?mica, Universidade Federal dos Vales do Jequitinhonha e Mucuri, 2011. / ABSTRACT Hydrazine is used as a corrosion inhibitor, antioxidant, catalyst, emulsifier, reducing agents, fuel for rocket propulsion, insecticides, and pharmaceutical preparation of various derivatives. However, the hydrazine is a toxic material and therefore requires a sensitive method for its determination. In this paper a sensor was developed for determination of hydrazine by modifying a glassy carbon electrode with cobalt phthalocyanine adsorbed on mutiwalled carbon nanotubes. The surface and electrochemical characterization was carried out by cyclic voltametry, scanning electron microscopy and energy dispersive spectroscopy X-ray. Surface studies shows that NTCPM and CoPc are a composite material. Electrochemical studies have shown the occurrence of redox processes of surface and also an indication of a limited transfer of load transfer. The number of electrons for the reaction of oxidation of hydrazine and the observed average rate constant of the process were estimated from the coefficients of linear and angular curves Koutecky-Levich being, respectively, 4,1 and 1,51 x 103 M-1s-1. The operational parameters of three electroanalytical techniques (amperometry, VOQ and VPD) were optimized for the determination of hydrazine with the use of ECV / NTCPM / CoPc, although the VOQ showed greater sensitivity and was therefore chosen for the determination of hydrazine in boiler water. A final analytical curve for the determination of hydrazine in boiler water sample was made using electrodes printed under optimized experimental conditions, with linear response over the concentration range of 99 mmol L-1 to 7630 ?mol L-1, with R equal to 0.999. We obtained values ??of LOD and LOQ equal to 50 and 99 ?mol L-1, respectively. The sensor allowed a good recovery for hydrazine in boiler water samples analyzed, with recovery values ??ranging between 98.41 and 103.62%, attesting to the accuracy of the proposed method.

Os extratos em hexano e em acetato de etila de B. coccolobifolia foram submetidos a m?todos cromatogr?ficos cl?ssicos e, atrav?s de t?cnicas espectrosc?picas (IV e RMN de 1H e 13C), foi poss?vel elucidar nos extratos das folhas as seguintes subst?ncias: friedelina, ?-sitosterol, mistura de ?-amirina e ?-amirina e derivados do ?cido olean?lico e urs?lico. Na identifica??o dos compostos presentes no extrato metan?lico de folhas e galhos de B. coccolobifolia, utilizou-se a t?cnica de espectrometria de massas por electrospray (ESI-MS), por inser??o direta dos extratos dissolvidos em metanol. Identificaram-se tr?s classes de metab?litos no extrato metan?lico: ?cidos fen?licos e derivados (galato de metila, ?cido qu?nico, ?cidos mono-, di-, tri- e tetragaloilqu?nico), proantocianidinas (d?mero e tr?mero epicatequina) e flavon?ides (quercetina e derivados glicosilados). As esp?cies B. coccolobifolia e B. intermedia apresentaram os melhores resultados para conte?do fen?lico e para atividade antioxidante de captura de radical e de poder redutor. Isso mostrou o potencial dos extratos polares dessas plantas como recurso para a busca de subst?ncias ?teis na preven??o ou no tratamento de doen?as associadas a radicais livres. No que diz respeito ? atividade inibit?ria da acetilcolinesterase (AChE), tamb?m investigada nas tr?s esp?cies de Byrsonima, o extrato hex?nico dessas plantas apresentou maior quantidade de compostos ativos frente a enzima AChE. E somente a esp?cie B. coccolobifolia apresentou atividade contra os microrganismos testados (Staphylococcus aureus, Bacillus cereus, Escherichia coli e Candida albicans). Sendo assim, o presente trabalho mostrou a import?ncia de continuar estudando a flora do cerrado brasileiro e as esp?cies do g?nero Byrsonima, sejam seus aspectos qu?micos ou medicinais. / Disserta??o (Mestrado) ? Programa de P?s-Gradua??o em Qu?mica, Universidade Federal dos Vales do Jequitinhonha e Mucuri, 2011. / ABSTRACT Byrsonima species are popularly known as "murici" and there are reports in previous work of many medicinal uses of these plants, some of them already confirmed as anti-inflammatory, antimicrobial and antioxidant activities. The aim of this study was to analyze phytochemically Byrsonima coccolobifolia Kunth and to make a study of biological activity in extracts of B. coccolobifolia, B. verbascifolia and B. intermedia, investigating the antioxidant activity, acetylcholinesterase inhibitory action and antimicrobial activity in these species. The aerial parts of plants were collected in Jequitinhonha Valley, Minas Gerais, Brazil, and dried at room temperature. Leaves and branches were separated, powdered and subjected to exhaustive extraction with solvents of increasing polarity (hexane, ethyl acetate and methanol), that were removed by rotary evaporator. The extracts in hexane and ethyl acetate of B. coccolobifolia were submitted at classical chromatographic methods and, by spectroscopic methods (IR and 1H and 13C NMR), it was possible to elucidate the following substances in leaves extracts: friedelin, ?-sitosterol, a mixture of ?-amyrin and ?-amyrin and oleanolic and ursolic acid derivatives. In the identification of compounds in methanol extracts of leaves and branches of B. coccolobifolia was used mass spectrometry with electrospray (ESI-MS) by direct infusion of the extracts dissolved in methanol. Three classes of metabolites were identified in methanol extracts: phenolic acids and derivatives (methyl gallate, quinic acid, mono-, di-, tri- and tetragalloylquinic acids), proanthocyanidins (epicatechin dimer and trimer) and flavonoids (quercetin and glycosides derivatives). The species B. coccolobifolia and B. intermedia showed better results for phenolic content and antioxidant activity of radical scavenging and reducing power. This showed the potential of polar extracts of these plants as a resource to search for substances useful in preventing or treating diseases associated with free radicals. Concerning the inhibitory activity of acetylcholinesterase (AChE), also investigated in three Byrsonima species, hexane extract of these plants had higher amounts of active compounds against AChE. And only B. coccolobifolia revealed activity against the microorganisms tested (Staphylococcus aureus, Bacillus cereus, Escherichia coli and Candida albicans). Thus, this investigation supported the importance of further studying the flora of Brazilian cerrado and the species of the genus Byrsonima, in their chemical or medicinal aspects.

For that, teaching-learning process related to basic chemical subjects which support the understanding of organic chemistry concepts was evaluated in a research period of two years. The education proposal linked to the theoretical content of the cited classes, pointed out the process of knowledge construction, in which educational commitment as well as dedication in the teaching-learning process was also valued. In that approach several didactic tools were applied, among them scientific articles were used as supplementary studies of the basic organic chemistry concepts and related. The acceptability of students, as well as their motivation, performance and learning process was justified by the data collection of the applied teaching methodology. Currently in Brazil, the treatment of Chagas disease is done, only using benznidazole, which is not effective for the disease?s chronic phase and causes aggressive side effects, which explains the necessity of researches to find novel anti-Chagas compounds. A strategy adopted for the development of bioactive compounds against T. cruzi consists on the inhibition of the sterol 14?-demethylase enzyme (CYP51), which is essential for the parasite?s cellular membrane integrity. The inhibition can be achieved by a complexation of heterocyclic ring-containing compounds with the iron atom of heme group, present on CYP51. Thus, molecular modeling techniques were used on this study to analyze the interaction of a heterocyclic compounds (with known activity) with T. cruzi CYP51 in order to obtain the necessary information to construct an effective model for the theoretical activity prediction of these and also novel compounds. The proposed model presented a good multiple correlation coefficient (r? = 0.84) with the terms used to its construction. The model was used to help the design of novel piperine derivatives with a triazole ring, that presented promising theorical activities against T. cruzi CYP51, calculated by the model. The most promising compounds were selected and synthesized with the purpose of being tested in vitro and in vivo against T. cruzi. / O Trypanosoma cruzi ? o parasito causador da tripanossom?ase americana (Doen?a de Chagas), uma doen?a tropical negligenciada antes restrita ? Am?rica do Sul, Am?rica Central e M?xico, mas que vem apresentando um n?mero cada vez maior de casos no mundo. Atualmente, o tratamento da Doen?a de Chagas no Brasil ? limitado ao uso do f?rmaco benzonidazol, que ? pouco eficaz para a fase cr?nica da doen?a e causa efeitos colaterais agressivos, o que torna a pesquisa por novos f?rmacos imprescind?vel. Uma estrat?gia adotada para o desenvolvimento de compostos bioativos contra T. cruzi consiste na inibi??o de uma enzima essencial para a integridade da membrana celular do parasito, a enzima esterol 14?-desmetilase (CYP51), causada pela coordena??o de compostos contendo an?is heteroc?clicos com o ?tomo de ferro do grupo heme presente na enzima, fundamental para a atividade. Dessa maneira, foram utilizadas nesse estudo t?cnicas de modelagem molecular, incluindo docagem molecular e c?lculos qu?nticos semi-emp?ricos, para analisar a intera??o de uma s?rie de compostos heteroc?clicos de atividade conhecida sobre a CYP51 do T. cruzi e com isso se obter informa??es necess?rias para a constru??o de um modelo efetivo para a previs?o te?rica da atividade destes compostos. O modelo proposto apresentou um bom coeficiente de correla??o m?ltipla com os termos utilizados para sua constru??o, com um r?=0,84. Esse modelo foi utilizado para o planejamento de novos triaz?is derivados da piperina, com atividade te?rica calculada promissora contra a CYP51 de T. cruzi. Alguns dos melhores compostos foram selecionados e sintetizados neste projeto, com a proposta de serem avaliados em testes in vitro e in vivo contra a doen?a de Chagas.

PN1 foi isolada como um ?ster de ?cido graxo ligado na posi??o C-3. Deste extrato foi obtida tamb?m a mistura dos ester?ides sitosterol e estigmasterol (PN3). O estudo fitoqu?mico conduzido com o extrato em acetato de etila das folhas e caules forneceu a flavona metoxilada nas posi??es C-6 e C-7, a cirsimartina (PN4). Todos os compostos obtidos foram descritos pela primeira vez na esp?cie. A elucida??o estrutural dos compostos isolados, baseou-se na an?lise espectrosc?pica de RMN 1D, CLAE, ESI e por compara??o com dados encontrados na literatura. O extrato metan?lico das folhas e caules de P. neochilus mostrou significativa atividade antioxidante quando avaliado pelo m?todo de DPPH, seguido pelo extrato em acetato de etila e hex?nico. Foi avaliado tamb?m o extrato metan?lico das flores da esp?cie, que apresentou atividade inferior aquele observado para o extrato metan?lico das folhas e caule, sendo estas atividades confirmadas pelo conte?do fen?lico dos extratos. Programa de P?s-Gradua??o em Qu?mica, Universidade Federal dos Vales do Jequitinhonha e Mucuri, 2011. / ABSTRACT The present work describes the phytochemical and pharmacological investigation on extracts of the leaves and stems and flowers of the species Plectranthus neochilus Schltr. belonged to the family Lamiaceae. The plant is aromatic herb popularly known in Brazil as ?boldo? or ?boldo-gamb??, and as the species P. barbatus Andrews (?boldo-brasileiro") e P. ornatus Codd. ("boldinho"), according to ethnopharmacological survey, is often used in the treatment of hepatic insufficiency and dyspepsia, replacing the Peumus boldus Molina (?boldo-do-chile?). The phytochemical investigation of the hexanic extract of the P. neochilus leaves and stems yielded two triterpenes: ?-amyrin (PN1) e friedelin (PN2). The compound ?-amirin was isolated as a fatty acid ester in position C-3. In this extract was also obtained the mixture of the steroids sitosterol and stigmasterol (PN3). The phytochemical study conducted with the ethyl acetate extract of the leaves and stems gave the methoxy flavone at positions C-6 and C-7, the cirsimartin (PN4). All compounds obtained were first described in the species. Structural elucidation of isolated compounds was based on spectroscopic analysis of 1D NMR, HPLC, ESY and by comparison with data found in literature. The methanol extract of leaves and stems of P. neochilus showed significant antioxidant activity when evaluated by the DPPH method, followed by the extract in ethyl acetate and hexane. We also evaluate the methanol extract of flowers of the species that showed activity lower than that observed for the methanolic extract from leaves and stems, and these activities confirmed for the content of phenolic extracts. The anticholinesterase activity of extracts of leaves and stems and flowers of P. neochilus was evaluated and found to inhibit the enzyme acetylcholinesterase to all extracts studied.

Diante disso, o presente trabalho teve como objetivo avaliar a composi??o qu?mica, o potencial antioxidante (in vitro) e hipolipid?mico (in vivo) da farinha das cascas do fruto de Myrciaria cauliflora. Para tanto, frutos (jabuticabas) maduros foram despolpados manualmente e suas cascas secas e trituradas at? a obten??o de uma farinha homog?nea (FCJ). Avaliou-se sua composi??o centesimal, conte?do mineral, fen?licos totais, flavon?ides e antocianinas, assim como sua atividade antioxidante in vitro atrav?s de diferentes m?todos (DPPH, ABTS?+, FRAP e ?-caroteno/?cido linol?ico). A FCJ foi submetida ? extra??o hidroalco?lica e os extratos fracionados em Cromatografia em Coluna (CC). Os fitoconstituintes isolados foram caracterizados por IV e RMN. O potencial hipolipid?mico foi avaliado em grupos de ratos machos alimentados com dietas semipurificadas, sendo a dieta Padr?o (PDR) baseada na AIN93M, a dieta Controle (CTRL) semelhante ? PDR e suplementada com 7% de banha de porco, e tr?s dietas experimentais semelhantes ? CTRL, por?m suplementadas com 7, 10 e 15% de FCJ (JAB1, JAB2 e JAB3, respectivamente). N?veis s?ricos e hep?ticos de colesterol, bem como n?veis s?ricos de HDL, triglicer?deos e glicose foram avaliados. A excre??o fecal de lip?deos foi avaliada tamb?m. A FCJ foi classificada como fornecedora de alto teor de fibras (15,26%). Os teores de fen?licos totais (1895 mg ?cido g?lico/100 g), flavon?ides (8,960 g de catequina /100g) e antocianinas (0,6823 g de cianidina-3-glicos?deo/100g) foram altos assim como a atividade antioxidante verificada pela captura de radicais-livres DPPH (3,184 g de FCJ/g DPPH) e ABTS?+ (1017 ?mol Trolox/g de FCJ), redu??o dos ?ons Fe+3 (167,7 mM de Fe2SO4/100g) e capacidade de inibi??o do branqueamento do ?-caroteno (75,96% de inibi??o para a concentra??o de 1000 mg/L). O ?cido c?trico, o sitosterol e o sitosterol-D-glicopiranos?deo foram isolados, sendo, os dois ?ltimos, citados pela primeira vez nesta esp?cie. A inclus?o da FCJ, nas tr?s propor??es, reduziu o colesterol total (CT) e os triglicer?deos (TG) s?ricos em rela??o ? dieta CTRL. A dieta JAB3 elevou os n?veis de HDL s?ricos e reduziu os de colesterol hep?tico em rela??o ? CTRL. A dieta JAB3 reduziu a glicose s?rica em compara??o ? JAB2, mas essa diferen?a n?o foi significativa em rela??o ? PDR, ? CTRL ou ? JAB1. N?o houve diferen?a na excre??o fecal de lip?deos entre os grupos experimentais. Programa de P?s-Gradua??o em Qu?mica, Universidade Federal dos Vales do Jequitinhonha e Mucuri, 2011. / ABSTRACT Myrciaria cauliflora (jabuticabeira) is a fruit specie, native in Brazil and created mainly in domestic gardens in Minas Gerais. However, little is known about its chemical composition and biological effects in the fruit residue. In front of this, the present work had as a goal to evaluate the chemical composition, antioxidant potential (in vitro) and hypolipidemic (in vivo) of Myrciaria cauliflora fruit peel flour (FCJ). For that, mature fruits (jabuticaba) were pulped manually and its peel was dried and crushed to obtain a homogeneous flour (FCJ). There had been evaluated the proximate composition, mineral content as well as total phenolic, flavonoids and anthocyanins as their antioxidant activity in vitro through different methods (DPPH, ABTS?+, FRAP and ?-caroten/acid linoleic). The FCJ was also subjected to extraction with water-alcohol extracts fractionated in column chromatography (CC). The phytochemicals isolated were characterized IR and NMR. The lipid-lowering activity was evaluated in groups of male rats, fed with semipurified diets, being a PDR diet based on AIN93M; a CTRL diet based on PDR, but added 7% of pork lard; and three experimental diets similar to CTRL, but added MPF at 7, 10 and 15% (JAB1, JAB2 and JAB3, respectively). Serum and liver cholesterol as well as serum levels of HDL, triglycerides and glucose were evaluated. Fecal output of lipids was also evaluated. The FCJ was classified as a supplier of high-fiber (15,26%). The levels of total phenolic (1895 mg gallic acid/100 g), flavonoids (8,960 g de catechin /100g) and anthocyanins (0,6823 g de cyanidin-3-glucoside/100g) were high as the antioxidant activity verified to capture of free radicals DPPH (3,184 g de FCJ/g DPPH) and ABTS?+ (1017 ?mol Trolox/g de FCJ), to reduction of ions Fe+3 (167,7 mM de Fe2SO4/100g) and capacity in inhibition of whitening of ?-caroten (75,96% of inhibition of concentration of 1000 mg/L). The citric acid, sitosterol and sitosterol-?D-glucopyranoside were isolated, being the last two, mentioned in this specie for the first time. The inclusion of the MPF, at the three ratios reduced serum cholesterol and triglycerides compared to CTRL. JAB3 diet raised serum HDL cholesterol and reduced liver cholesterol compared to CTRL. JAB3 diet reduced serum glucose compared to JAB2, but this difference was not significant compared to PDR, CTRL or JAB1. There was no difference in fecal output of lipids between the groups. The results obtained may serve as an aid to better utilization of residue and to understand the MFP metabolic effects in addition to ensure the safety of its intake as a food.

A prepara??o dos aceptores e doadores glicos?dicos envolveu sequ?ncias de prote??o e desprote??o de grupos hidroxila a partir da adenosina e da D-glicose, respectivamente, privilegiando-se a forma??o de doadores armados e a observa??o da estereoqu?mica no carbono anom?rico. Programa de P?s-Gradua??o em Qu?mica, Universidade Federal dos Vales do Jequitinhonha e Mucuri, 2012. / ABSTRACT This works discusses the synton?s synthesis of the, unpublished glycosidic donors and aceptors that have been used in a convergent synthesis of a precursor of adenophostin A. Furthermore, the convergence of syntons main purpose was to study the glycosylation, ie been explored two methods of forming glycosidic linkages, the method of trichloroacetimidate and method of isomerization of the allyl glycoside donor. The preparation of donors and acceptors glycosidic sequences involving protection and deprotection of hydroxyl groups from adenosine and D-glucose, respectively, favoring the formation of armed donors and observation of stereochemistry at anomeric carbon. The and anomers formed in this synthesis sequence were separated, but the only reason anomers were characterization by usual methods of analysis of 1D and 2D NMR spectroscopy, mass and infrared. Thus, we synthesized two glycoside acceptors (17) and (18) and five donor glycoside (9), (11), (11), (13) and (13), all substances are novel. Among the glycosylation assays only the method of tricloroacetimidate gave the desired product (glycosylation between 11 and 17) that is the object of characterizing and whose data has not yet been completed and conclusive.

Diante disso, o presente trabalho teve como objetivo avaliar os constituintes a??cares, s?lidos sol?veis totais, fen?licos, flavon?ides, cafe?na e determinar a acidez total, o pH e a atividade antioxidantepor diferentes m?todos (DPPH, ABTS?+ e FRAP) no gr?o de caf? cru. Tamb?m avaliar os efeitos do tratamento sub-cr?nico com cafe?na e caf? no peso corporal e dos ?rg?os (ba?o, cora??o, f?gado, rins, supra renais e test?culos), comprimento dos ossos (f?mur e t?bia), ingest?o de alimentos (s?lidos e l?quidos) e par?metros bioqu?micos (colesterol total, glicemia e triglic?rides) em ratos machos Wistar. Foram utilizados 30 ratos: Controle (C) ? receberam ra??o e ?gua; Cafe?na (CA) ? receberam ra??o e ?gua contendo 0,1% de cafe?na; Caf? 0,1% (CC1) ? receberam ra??o e caf? contendo 0,1% de cafe?na; Caf? 0,05% (CC5) ? receberam ra??o e caf? contendo 0,05% de cafe?na; caf? 0,025% (CC25) - receberam ra??o e caf? contendo 0,025% de cafe?na. As amostras de caf? das safras de 2010 e 2011 analisadas, apresentaram diferen?as significativas para os teores de fen?licos totais, flavon?ides, cafe?na, s?lidos sol?veis, a??cares totais e n?o-redutores, al?m de acidez total e pH. A atividade antioxidante total para os m?todos utilizados (ABTS, DPPH e FRAP) e teor de a??cares redutores n?o variaram significativamente de um ano para o outro. No ensaio biol?gico os animais CC1 pesaram menos do que os C, CC5 e CC25. Em rela??o ao ba?o, os CC5 pesaram menos do que os C e CC25; os CC1 menos do que os C. As supra renais dos CC1 pesaram menos do que as dos Ca e CC25. Os CC1 apresentaram menor peso dos test?culos do que os C, CA, CC5 e CC25 e os CC25 pesaram mais do que os C. Os ratos CC1 apresentaram menor comprimento do f?mur do que os C, CC5 e CC25 e menor t?bia do que os C, CA, CC5 e CC25. Os animais CC25 apresentaram maiores taxas de glicemia do que os CC1 e CC5 e os CC1 maiores taxas de colesterol total do que os C e CC25. Os animais CC1 comeram menos do que os C durante o tratamento e beberam menos na primeira semana. Programa de P?s-Gradua??o em Qu?mica, Universidade Federal dos Vales do Jequitinhonha e Mucuri, 2012. / ABSTRACT Coffee is among the most important agricultural products in international trade and its quality as a product is an important factor in price formation, besides being one of the most widely consumed beverages worldwide and therefore arouses interest for studies of the physiological effects of its constituents. Thus, the present study was to evaluate the constituent sugars, soluble solids, phenolics, flavonoids, caffeine and determine the total acidity, pH and activity antioxidantepor different methods (DPPH, FRAP and ABTS ? +) in raw coffee beans . The objective was also to evaluated the effects of sub-chronic treatment with coffee and caffeine on body weight and organs (spleen, heart, liver, kidneys, adrenals and testicles); bone?s length (femur and tibia); food intake (solid and liquid) and biochemical parameters (total cholesterol, glucose and triglycerides) in male Wistarrats.A total of 30 rats were used: Control (C) - received food and water; Caffeine (CA) - received food and water containing 1% caffeine; Coffee 1% (CC1) - received food and coffee containing 1% caffeine; Coffee 0.05% (CC5) - received food and coffee containing 0.05% caffeine; coffee 0.025% (CC25) - received food and coffee containing 0.025% caffeine. Samples of coffee crop in 2010 and 2011 analyzed showed significant differences in the levels of total phenolics, flavonoids, caffeine, soluble solids, total sugars and non-reducing sugars, and total acidity and pH. The total antioxidant activity for the methods (ABTS, DPPH and FRAP) and reducing sugars did not vary significantly from one year to another. In the biological tests the animals CC1 weighed less than the samples C, CC25 and CC5.Regarding the spleen, CC5 weighed less than C and CC25; the CC1 less than C.The adrenal glands of CC1 weighed less than that of the Ca and CC25.The CC1 testicles weighed less than C, CA, CC5 and CC25; and CC25 were heavier than C.The CC1 rats had lower femur length than C, CC5 and CC25; and lower tibia than the C, CA, CC5 and CC25.The CC25 animals showed higher rates of glucose than the CC1 and CC5; and CC1 higher total cholesterol than the C and CC25.CC1 animals ate less than C during the treatment and drank lessduring the first week.The results obtained may serve as an aid to improve the quality of coffee beverage and the understanding of its metabolic effects in the body.
