O discurso autobiogr?fico nos relatos de viagem de N?sia Floresta

Malgr? les particularit?s qui entourent chacun de ces r?cits, un aspect y est fortement pr?sent : les marques autobiographiques. Notre travail a comme principal objectif enqu?ter comment se manifeste ce discours autobiographique. A partir de l‟Analyse du Discours, nous avons travaill? quelques positionnements sur certaines oeuvres litt?raires, comme par exemple, le fait qu‟elles aient plus d‟un genre. Ce qui se passe dans les r?cits de voyage de N?sia Floresta est semblable, car au-d?l? de raconter le passage de N?sia par ces lieux, nous pouvons trouver des caract?ristiques qui appartiennent ? l‟?criture fragmentaire, autobiographique et ?pistolaire. Devant les propositions indiqu?es par Philippe Lejeune, nous avons pu confirmer que ces r?cits de N?sia font partie de l‟?criture autobiographique. Sous l‟optique de quelques th?ories, nous avons parcouru des points int?ressants qui enveloppent ces r?cits et, de cette fa?on, nous avons voyag? avec N?sia Floresta par l‟Allemagne et ult?rieurement par l‟Italie et Gr?ce, en connaissant ainsi, pas seulement un peu plus sur ces lieux, mais beaucoup de confidences sur cette ?crivain potiguar, qui a ouvert son coeur au plublic / Em pleno s?culo XIX, N?sia Floresta Brasileira Augusta escreveu sobre duas viagens empreendidas pela Europa. Alguns anos depois, essa norte-rio-grandense trouxe ao conhecimento do p?blico seus relatos de viagem: Itin?raire d un Voyage en Allemagne (1857) e Trois ans en Italie, Suivis d un Voyage en Gr?ce (1864/1872). Apesar das particularidades que envolvem cada um desses relatos, um aspecto est? fortemente presente neles: as marcas autobiogr?ficas. O nosso trabalho tem como principal objetivo investigar como se manifesta esse discurso autobiogr?fico. A partir da An?lise do Discurso, levantamos alguns posicionamentos acerca de algumas obras liter?rias, como o fato de, certas vezes, elas conterem mais de um g?nero. O que acontece nos relatos de viagem de N?sia Floresta ? semelhante, pois al?m de narrarem a passagem de N?sia por esses lugares, podemos encontrar caracter?sticas pertencentes ? escritura fragment?ria, autobiogr?fica e ao epistolar. Diante dos pressupostos apontados por Philippe Lejeune, pudemos confirmar que essas narrativas de N?sia fazem parte da escritura autobiogr?fica. Denna rapport redogör för utvecklingen av en applikation som används för att konfigurera integrationer som används av av Sigma Integration Architecture (SIA). Applikationen är riktad till Sigma-personal som inte är vana vid denna integrationsarkitektur, då den tillhandlahåller användaren med en rad olika hjälp-element för att underlätta konfigurationensprocessen och sänka tröskeln för att kunna skapa och redigera integrationer. Genom att använda Javas reflection API i kombination med dynamic proxies, kan applikationen utföra validering och simulering av integrationer innan de används i produktion. Vi innefattar även cache-metod för att snappa upp processen, och påvisar även testverifikation av cache-processen. Applikationen är utvecklad som två separata delar, bestående av en back end skriven i Java, och en webbaserad front end skapad med AngularJS. / This report describes the development of an application used to configure integrations used by Sigma Integration Architecture (SIA). The application is aimed towards Sigma personnel unfamiliar with the integration architecture, as it provides the user with various help elements in an effort to ease the configuration process and lower the entry barrier for creating and editing integrations. Using Java's reflection API in combination with dynamic proxies, the application is able to perform validation and simulation of integration pipelines before they are used for production. We also include a cache mechanism to speed up the process, and provide experimental verification of caching performance. The application is developed as two separate parts, consisting of a back end written in Java, and a web-based front end created with AngularJS.

Stanovení heparinu technikou SIA se spektrofluorimetrickou detekcí / Determination of heparine by SIA with spectrofluorimetric detection

This thesis was focused on a determination of heparin using sequential injection analysis with spectrofluorimetric and spectrophotometric detection. The principle of determination was based on the interaction of heparin with phenothiazine dye. A decrease of fluorescence intensity of dye in its emission maximum was detected. In the case of spectrophotometric detection a decrease of the absorbance of dye was measured. Azure A, azure B and methylene blue were used as representantive selection of phenothiazine dyes. The determination was performed on a laboratory made SIA apparatus, for which a control software in LabVIEW 7.1 graphical programming was created. Two types of flow configuration for spectrofluorimetric detection were implemented. Type 1: For deionized water as a carrier stream with a injection of heparin and dye zones there were found the following optimal conditions: cdye = 1×10-5 mol dm-3 ; vflow = 2500 µl min-1 ; reaction coil length of 0 cm; injected volume of dye 150 µl and injected volume of sample 150 µl. Dynamic range of calibration curves with an exponential course for the individual dyes in the range of LOQ - 1200, eventually up 1500 IU dm-3 were detected. This dissertation examines poets’ public performances in order to understand the social role of the poet in contemporary America. In the twenty-first century, poetry is increasingly disseminated through live events and digital media, and a rising number of people publicly share their poems. These changes present challenges for authors looking to attain credibility in the eyes of critics and audiences. The Role of the Poet examines how four poets perform their differences from non-authors, and thus form relationships with their audiences. In constructing roles for themselves, poets also make claims about the ontology of poems—whether they are primarily written, oral, or performative works of art. Each chapter focuses on an individual poet’s strategies for performing the role of the poet, and by extension, constructing the role of the audience. The chapters examine the ways poets define poetry; they include discussions of poetry’s ontology and how public poetry performances affect the artform. Performances of authorship are shaped through the vehicles of poets’ writings, poetry readings, interviews, teaching methods, and public programs. Chapter 2 examines Robert Pinsky’s performance of authorship as authority, relating that performance to Pinsky’s canonical ambitions and his affirmation that poetry is an oral, but not performative art. Chapter 3 focuses on Billy Collins’s performance of authenticity, investigating the apparent paradox of achieving popularity while maintaining artistic integrity. Beau Sia’s political poetry is the subject of Chapter 4; his ability to affect change in his audience is considered, as well as his goal of an author-audience alliance. Lastly, Patricia Smith’s performance of authorship as a means of survival is discussed in Chapter 5. It is a current theme, complex in itself, therefore involving the view of diverse disciplines, such as Biology, Philosophy and Sociology, which are represented in their constituent elements, such as common sense, science and culture, respectively. Religion stands out in this context, because the subject has the object of death and the treatment given to it. Theoretically, it was tried to characterize the evolution of the law in front of the social development resulting from the technological transformations and the emergence of Bioethics; analyzing ortotanasia in the context of the unfolding of the terminality of life; highlighting cultural aspects that can mobilize popular acceptance of controversial issues in general and ortotanasia in particular (common sense, science); to raise, among law students, objective and subjective aspects that may justify the non-undertaking of the artificial prolongation of life in terminal patients by the physician. Empirically, the results showed that there is still a lot of ignorance on the subject, which hinders or prevents answers that clearly point out the main difficulties of the social acceptance of ortotanasia. However, the tendency to recognize the right to refuse to therapeutic obstinacy was clear, which in itself already represents an advance against traditional culture. / O objetivo deste trabalho ?? verificar que principais quest??es podem envolver a aceita????o social do direito ?? implementa????o da ortotan??sia no contexto das transforma????es trazidas pela evolu????o da tecnologia, da Bio??tica e, consequentemente, do Direito. Trata-se de um tema atual, complexo em si, por isso mesmo envolvendo o olhar de disciplinas diversas, como o Biodireito, a Filosofia e a Sociologia, essas representadas em seus elementos constituintes, como senso comum, ci??ncia e cultura, respectivamente. A religi??o se destaca nesse contexto, pelo fato de o tema ter como objeto a morte e o tratamento dado a ela. Sia Furler som inspirationskälla till mitt eget musikskapande

I denna uppsats undersöks hur låtskrivaren och artisten Sia Furlers sound och låtskrivarmetoder kan anammas och appliceras på det egna musikskapandet. Ett antal låtar skrivna och framförda av Furler analyseras, tre av dem in i minsta beståndsdel. Det som undersöks är låtarnas uppbyggnad textligt, melodiskt och rytmiskt samt låtarnas sound. De små beståndsdelarna sätts sedan ihop i en ny konstellation i tre helt nya, egenskrivna låtar. Den känsla i Sias musik som eftersträvas är kontrasten mellan ”mörker” och ”ljus”. De nya låtarna blir tre original med klar inspiration från Sia Furlers musik. / This essay examines how composer and artist Sia Furler's sound and songwriting methods can be applied in my own music creation. A number of songs written and performed by Furler are being analysed, tree of them very closely. The parts which are being examined are the dynamic construction of the lyrics, the melodies and the rhythms as well as the sound. These small parts are then put together in a new constellation of three totally new songs. The feeling in Sia's music which is sought is the contrast between “darkness” and “light”. Three original songs inspired by Sia Furler's music are the results of this examination.

Values-led business : A study of the Ben and Jerry's concept and its applicability

The issue that is dealt with in this paper is the concept of values-led business. The idea behind the term is that businesses has a responsibility to the people and the society that make its existence possible and that social problems can not be solved unless business accepts taking on a leadership role which requires that business starts acting in the interests of the common good. This is specifically important since companies today are the main source of influence in people’s everyday life and the area is specifically interesting because of all the business scandals in recent years. (Cohen and Greenfield, 1997). The concept of values-led business arose mainly from the two companies the Body Shop and our chosen company; Ben and Jerry’s. The purpose of this thesis is to explore the concept of values-led business and find out how it is different from the procedures at a non-explicitly values-led organisation. We also aim to find out if there would be any benefits of introducing values-led business into the latter. In this paper we compare an explicitly values-led company, Ben & Jerry’s Ice Cream, with a non explicitly values-led company, SIA Glass on their internal and external performance on the study areas of Marketing, Sourcing, and HRM and Organisational culture. A qualitative research method has been used as method. The collection of primary data was done with the help of interviews done at our companies’ main offices in Sweden. In the case of Ben & Jerry’s, additional secondary sources have been used. Theories used serve the purpose of both a tool of analysis but also of setting the stage of values-led business. Furthermore, the theory has been divided into the four areas of study mentioned above. The first part of the conclusion states that we, throughout the thesis, have given a good overview of what the differences between values-led business and “ordinary” business really are. Also, we found that SIA Glass has the potential of becoming a values-led company as its foundation is not entirely based on financial goals but is to a large extent driven by a will to preserve something good for the local community. In order to develop a val-ues-led thinking a social mission should be formally put down in print and be held equal to the financial and quality missions. SIA Glass potentially has a lot to be gained especially in the marketing area as SIA Glass as it could create further leverage by using more creative marketing methods.

MTSR is a Dual Regulator that Controls Virulence Genes and Metabolic Functions in Addition to Metal Homeostasis in Group A Streptococcus

Group A Streptococcus (GAS) is a common pathogen of the human skin and mucosal surfaces and is capable of producing a variety of diseases. This dissertation investigates the function of a metalloregulator named MtsR in GAS physiology and disease process. An mtsR mutant was constructed and analyzed. Consistent with MtsR role in iron uptake regulation, the mtsR mutant accumulates more iron (80 ± 22.5%) than the wild type strain. Inactivation of mtsR results in constitutive transcription of the sia (Streptococcal Iron Acquisition) operon, which is negatively regulated by iron in the parent strain. We identified the promoter that controls the expression of the sia operon (Pshr) and used it as a model to study MtsR interaction with DNA. Electrophoretic mobility gel shift assays (EMSAs) demonstrated that MtsR binds to the shr upstream region specifically and in an iron and manganese dependent manner. DNase I footprint analysis revealed that MtsR protects a 69 bp segment in Pshr that includes 2 inverted repeats, overlapping the core promoter elements. A global transcriptional analysis determined that MtsR modulates the expression of 64 genes, of which 44 were upregulated and 20 were downregulated in the mtsR mutant. MtsR controls genes with diverse functions including immune evasion, colonization, dissemination, metal homeostasis, nucleic acid and amino acid metabolism, and protein stability. MtsR functions as a dual regulator as it binds to the promoters of the repressed genes ska, aroE, and nrdF.2, as well as the upstream region of the positively regulated genes mga, emm49, and pyrF. A 16 bp MtsR-binding consensus region was identified in all of the promoters that are directly regulated by MtsR. In conclusion, we have demonstrated that MtsR is a global regulator in GAS that controls the expression of vital virulence factors and genes involved in metal transport, virulence and metabolic pathways.

DISCONNECT: Assessing and Managing the Social Effects of Development in the Athabasca Oil Sands

This research investigated the system by which the social effects of oil sands development on Fort McMurray, a city in northeastern Alberta, are assessed and managed. The research focused on Social Impact Assessment (SIA), Strategic Environmental Assessment (SEA), and the work of an industry initiative, the Regional Issues Working Group (RIWG). The oil sands industry, which involves large, labour-intensive mining and drilling operations in a boom-bust cycle, places considerable pressure on Fort McMurray, a city of approximately 50,000 inhabitants and the only urban area within 350 km of the oil sands. The social effects experienced there include exorbitant housing prices, shortages in service industry labour, insufficient social services, at times, to assist individuals and families who can no longer cope with the difficult conditions in the area, and a variety of other negative effects. Sixteen key informant interviews were conducted with urban planners, municipal politicians, provincial employees, a spokesperson for one of the First Nations in the area, community NGOs, and oil sands industry representatives. Data from the interviews were combined with a literature review and a document analysis. A modified McKinsey 7S Integrated Management Framework was used as a structure for describing and analyzing the Social Effects Assessment and Management System (SEAMS) in Fort McMurray. The SEAMS was found to be weak in comparison to the needs of the community. Project-by-project assessment of oil sands development was found to downplay the cumulative nature of social effects. Furthermore, no legislation or regulation existed that demanded action based on the findings of SIA. As a result, mitigation and management of social effects was insufficient, often occurring only when it was directly in the interests of the oil sands industry. While government and industry have plans in place to resolve some of the negative social effects, their actions were criticized by informants as being uncoordinated, inconsistent and often ineffective. The findings indicate that a strategy for exploiting Alberta's oil sands is necessary. The project-by-project evaluation of oil sands development proposals is not addressing the important long-term and regional social issues that arise as a result of construction and operation of the mines and facilities. A tool recommended for incorporating resolutions to long-term, regional social effects into the development plan is SEA with an explicit Strategic Social Assessment component. This strategic assessment and planning process should be undertaken by a publicly-accountable government body empowered to rationalize the pace of oil sands development based on social, environmental and economic effects, and to coordinate long-term responses by government and industry.
