Porém, não foram realizadas pesquisas que tenham verificado se a irradiação do esmalte dental ao redor de braquetes ortodônticos com esse laser é efetiva em reduzir a desmineralização nessa região numa situação de alto desafio cariogênico. Assim, esta dissertação objetivou verificar, in vitro, se a irradiação do esmalte dental com laser de CO2 (?=10.6 µm e 10.0 J/cm2), associada ou não a liberação de fluoreto pelo material de colagem, seria capaz de reduzir a perda mineral do esmalte, ao redor de braquetes ortodônticos, quando submetido a uma situação de alto desafio cariogênico. Nesse estudo, 24 blocos de esmalte bovino foram divididos em 4 grupos, em triplicata: 1 ¿ resina composta não liberadora de fluoreto Transbond (T); 2 ¿ cimento de ionômero de vidro modificado por resina Fuji (F); 3 ¿ laser de CO2 + resina composta não liberadora de fluoreto (TL); 4 ¿ laser de CO2 + cimento de ionômero de vidro modificado por resina (FL). Um grupo contendo blocos de esmalte foi incluído apenas para análise de microdureza. Após a colagem dos braquetes, os espécimes foram suspensos em água destilada deionizada esterilizada e esterilizados com radiação gama. A seguir, foram transferidos para o meio de cultura esterilizado de caldo de cérebro-coração (BHI) contendo sacarose a 5% e os 4 grupos experimentais foram inoculados com uma cultura overnight de Streptococcus mutans. Diariamente, o meio BHI foi trocado e analisado quanto à contaminação microbiológica. Após 6 dias de incubação (37ºC - 10% CO2), o biofilme foi coletado e submetido as análises microbiológica (UFC/mg) e bioquímica. Além disso, microdureza do esmalte seccionado longitudinalmente foi determinada. Os dados foram analisados pelos testes ANOVA e Tukey, com alfa a 5%. As concentrações de polissacarídeo insolúvel em água (µg/mg) no biofilme foram: T ¿ 213,206(±421,746)a, F ¿ 111,208(±43,501)a, TL ¿ 124,626(±37,488)a e FL ¿ 138,83(±118,893)a. As concentrações de cálcio (µg/mg) foram: T ¿ 340,5(±27,01)a, F ¿ 329,5(±143,97)a, TL ¿ 412,3(±228,80)a e FL ¿ 411,8(±252,59)a. As concentrações de fluoreto (µg/mg) no biofilme foram: T ¿ 0,001(±0,005) a, F ¿ 0,010(±0,021) a, TL ¿ 0,0009(±0,002)a e FL ¿ 0,002(±0,007)a. As concentrações de fósforo (µg/mg) foram: T ¿ 0,162(±0,134)a, F ¿ 0,149(±0,066)a, TL ¿ 0,170(±0,104)a a e FL ¿ 0,148(±0,029)a. Os resultados (expressos 107 UFC/mg) obtidos da análise microbiológica foram: T ¿ 2,54(±2,58)a, F ¿ ,90(±3,08)a, TL ¿ 2,59(±3,13)a e FL ¿ 2,30(±4,04)a. A média numérica da microdureza knoop (kg/mm2) variou de 195,5(±87,3)c, 209,8(±75,0)bc, 218,2(±113,6)ab e 229,1(±82,7)a para os grupos T, F, TL e FL, respectivamente. Em conclusão, esse estudo demonstrou que o uso do laser de CO2 (?=10.6 µm e 10.0 J/cm2) sozinho ou combinado com o material de colagem liberador de fluoreto foi capaz de reduzir a perda mineral do esmalte ao redor de braquetes ortodônticos, quando submetidos a uma situação de alto desafio cariogênico com biofilme de Streptococcus mutans. / Abstract: The application of carbon dioxide laser (CO2) on dental enamel structure modifies the chemical and/or morphologic composition of this surface and inhibits the development and progression of caries lesion. However, no research verified the if the irradiation of dental enamel around orthodontic brackets was able to reduce the enamel mineral loss in this region in a high cariogenic challenge. Thus, this dissertation aimed to verify in vitro, if the irradiation of the dental enamel with a laser of CO2 (? =10.6 µm and 10.0 J/cm2), associated or not with fluoride released from a bonding material, will be able to reduce the enamel mineral loss around orthodontic brackets, when submitted to a high cariogenic challenge situation. In this study, twenty four enamel slabs were divided into 4 groups in triplicate: 1. nonfluoride-releasing composite resin Transbond (T); 2. resinmodified glass ionomer cement Fuji (F); 3. CO2 laser + nonfluoride-releasing composite resin (TL); 4. CO2 laser + resin-modified glass ionomer cement (FL). One group with 6 specimens of sound enamel were used to determine the sound enamel microhardness. After brackets bonding, the specimens were immersed in sterile deionized distilled water and sterilized with gamma radiation. Following, the specimens were transferred to a sterile brain-heart infusion broth (BHI) with a 5% sucrose solution and the 4 experimental groups were inoculated with an overnight culture of Streptococcus mutans. The BHI medium was daily changed and analyzed to check for microbiological contamination. After 6 days of incubation (37ºC - 10% CO2), the biofilm was collected and submitted to microbiological (CFU/mg) and biochemical analyses. Additionally, microhardness assay of the enamel longitudinally sectioned was determine. The data were statistically analyzed by the ANOVA and Tukey¿ tests, with an alpha of 0.05. The concentrations of water-insoluvel polysaccharide (µg/mg) in biofilm were: T ¿ 213.206(±421.746)a, F ¿ 111.208(±43.501)a, TL ¿ 124.626(±37.488)a e FL ¿ 138.83(±118.893)a. The concentrations of calcium (µg/mg) were: T ¿ 340.5(±27.01)a, F ¿ 329.5(±143.97) a, TL ¿ 412.3(±228.80)a e FL ¿ 411.8(±252.59) a. Tal técnica baseia-se na ação simultânea de uma fonte luminosa e um agente fotossensibilizador, que na presença de luz, geraa produção de oxigênio singleto e espécies reativas de oxigênio que ao interagirem com componentes celulares, promovem a morte celular desses micro-organismos. Esse estudo teve como objetivo avaliar a ação da TFDa explorando nanopartículas de prata (com tamanho médio de 13 nm) e moléculas de riboflavina(157 μM) em conjunto com um LED (Light Emitting Diode) com comprimento de onda no azul (λ=455 ± 20nm), sobre cepas de Streptococcus mutans, Pseudomonas aeruginosae Candida albicans. As cepas de S. mutans foram repicadas em BHI (Brain Infusion Heart) e incubadas em estufa bacteriológica a 37ºC, em atmosfera de 5% de CO2 por 48 he os inóculos de P. aeruginosa foram cultivados e estocados em TSA (Tryptic Soy Agar), em presença de O2, a 37º C, por 24 horas. Decorrido o tempo de incubação, colônias dos micro-organismos foram suspensas em Phosphate Buffered Saline (PBS) a uma densidade de1x107 Unidades Formadoras de Colônia (UFC)/mL.As culturas de Candida albicansforam cultivadas em meio SAB+Y (Agar Sabouraud Dextroseenriquecido com extrato de levedura). A incubação foi realizada à temperaturade 37ºC, durante período de 24 horas. Em seguida,foram preparadassuspensões com concentração de 5x106 células/mL,correspondente à turbidez de 0,5 na escala McFarland.O experimento foi dividido em doze grupos–o grupo controle, o grupo apenas com o tratamento com a luz (L), tratamento com a riboflavina (Rb), utilização da prata (Ag) e o sistema NPsAgRb+L. Posterior aotratamento, foi realizado diluição seriada e as placasincubadas a 37 ºC por 24 horas para bactérias e 48 hs para Candida sp. Ao final foi realizada a contagem de UFC/mLde ambos os micro-organismos. Os resultados foram submetidos à análise estatística descritiva. Na redução de UFC/mL foi estatisticamente significante em culturas de S. mutans com redução de 3 logs no número de células viáveis após 6 minutos de irradiação (p ˂0,05).A inativação fotodinâmica contra P. aeruginosa não foi observada estatística significativa após tratamento (p > 0,05). O mesmo foi observado nas culturas de C. albicans(p > 0,05), concluindo-se que a utilização de nanopartículas próximas ao fotossensibilizador foi eficiente contra bactérias Gram-positivas. / Antimicrobial Photodynamic Therapy (TFDA) has been proposed as an alternative method for treating infections caused by microrganisms such as bacteria and fungi. This technique is based on the simultaneous action of a light source and a photosensitizing agent, in the presence of light, generates the singlet oxygen and reactive oxygen species to interact with cellular components and promote cell death of these microrganisms. This study aimed to evaluate the action of TFDa exploring silver nanoparticles (with an average size of 13 nm) and riboflavin molecules (157 mM) in conjunction with an LED (Light Emitting Diode) with a wavelength in the blue (λ = 455 ± 20nm) on Streptococcus mutans, Pseudomonas aeruginosa and Candida albicans. The strains of S. mutans were subcultured in BHI (Brain Heart Infusion) and incubated in bacteriological incubator at 37 ° C in an atmosphere of 5% CO2 for 48 h and the inocula of P. aeruginosa were grown and stored in TSA (Tryptic Soy Agar) in the presence of O2 at 37ºC for 24 hours. After the incubation time, colonies of microrganisms were suspended in Phosphate Buffered Saline (PBS) at a density of 1x107 colony forming units (CFU)/mL. The Candida albicans cultures were grown in SAB + Y (Sabouraud Dextrose Agar enriched with yeast extract). Incubation was carried out at 37°C temperature for 24 hours. Then, suspensions were prepared with a concentration of 5x106 cells/mL, corresponding to 0.5 on the McFarland turbidity scale. The experiment was divided into twelve groups - a control group, the only group with treatment with light (L), treatment with riboflavin (Rb), use of silver (Ag) and NPsAgRb + L system. After the treatment was carried out serial dilutions and the plates incubated at 37°C for 24 hours for bacteria and 48 hours for Candida sp. At the end it was performed CFU counts / ml of both microrganisms. The results were submitted to descriptive statistical analysis. The reduction of CFU/mL was statistically significant in cultures of S. mutans to 3 logs reduction in the number of viable cells after 6 minutes of irradiation (p ˂ 0.05). The photodynamic inactivation against P. aeruginosa was no statistically significant after treatment (p> 0.05). The same was observed in cultures of C. albicans (p> 0.05), it was concluded that the use of nanoparticles next to the photosensitizer was effective against Gram-positive bacteria.

Screening of Selected Libyan Medicinal Plants for the Synthesis of Metal Nanoparticles and their activity against Streptococcus mutans

It also has great potential to mitigate the challenges they face in various fields, especially medical sector. Nanodrugs are increasingly considered as a potential candidate to carry therapeutic agents safely into a targeted compartment in an organ, particular tissue or cell. In this study, twenty (20) Libyan plants were selected and evaluated for their potential to synthesis gold and silver nanoparticles. The screening of the different plant extracts was performed using 96 well plate method at 25 °C and 70 °C. The NPs formation was confirmed and characterized using UV- Vis, DLS, HR-TEM and EDX. A well-defined NPs were obtained at high temperature (70 °C). The Au NPs had an average diameter of 92 nm at 25 °C and 66 nm at 70 °C. The zeta potential values were observed to be negative (-14 to -24) and indicate the stability of the Au NPs. The HR-TEM showed polydispersity, which decreased at higher temperature (70 °C). The stability of Au NPs in nutrient broth prior was conducted as well. All the Au NPs under study showed stability, only minimal changes in the UV-Vis spectra can be observed. Two plant extract viz Pistacia atlantica, Junipers phoenicea showed consistent results and forming stable and smaller NPs compared to others, both of the plant extracts and the corresponding NPs were tested against Streptococcus mutans and showed MIC value ~ 49 g/mL. In case of silver NPs, two plant extracts viz J. phoenicea, Rosmarinus officinalis, showed superior results than the others; both plants produced stable and small Ag NPs. The antibacterial activity against S. mutans demonstrated MIC valus ~ 50 g/mL. The synthesised NPs showed a promising bioactivity for developments of new antibacterial agents against S. mutans strains. Dose-dependent activity was observed for the tested NPs.

La caries temprana de infancia, de inicio precoz en niños, es causada en forma frecuente por hábitos alimenticios inapropiados y la adquisición temprana de microorganismos como streptococcus mutans. El presente trabajo tuvo como objetivo principal determinar la relación entre los niveles colonización de streptococcus mutans en niños de 0 a 36 meses y el nivel de colonización de sus madres en el centro de salud Cerropón de Las Brisas, Chiclayo 2017. El diseño de estudio es de tipo analítico, transversal, observacional. La población de estudio fue determinada mediante los criterios de inclusión y exclusión a partir de la muestra. Se estableció la confiabilidad inter e intraexaminador mediante coeficiente de Kappa Cohen. El instrumento para la evaluación de las variables de estudio fue una ficha de recolección de datos. El análisis de datos se realizó en software estadístico SPSS versión 20. Se usó estadística analítica para presentar la información de las variables, mediante tablas y gráficos. La hipótesis fue evaluada mediante coeficiente de correlación de Spearman. Del total de los pacientes examinados 74.1% de niños tuvieron un nivel de Streptococcus mutans alto y 55.6% de las madres presentaron un nivel de Streptococcus mutans alto, además la ventana de infectividad de los niños se encontró en el rango de edad de 25 a 36 meses con un 86.7% encontrando así una relación del nivel de colonización en el binomio madre e hijo.

Human Leucocyte Antigen DRB1 in relation to colonization of Mutans Streptococci in a group of preschool children in the southern part of Sweden

Syfte: Målet med denna studie var att undersöka ett möjligt samband mellan de olika HLA-DRB1*-allelerna med mängden Mutans Streptococci (MS) i saliven, med fokus på HLA DRB1*04, i en grupp förskolebarn i södra Sverige, Skåne.Material och metod: Salivprover från 318 HLA-DRB1*-typade barn odlades på MSB-agarplattor och antalet MS CFU räknades. Resultaten analyserades statistiskt med chi-två-test.Resultat: Inget signifikant samband kunde fastställas mellan någon av DRB1*04-allelerna och mängden MS. Dock var höga MS-värden ungefär dubbelt så vanligt för homozygota DRB1*04 alleler (p = 0,354). Höga MS-värden var vanligare för DRB1*04:01 allelen (p = 0,717). Däremot var höga MS-värden procentuellt mycket mindre förekommande för DRB1*04:04 allelen (p = 0,098). Ett statistisk signifikant samband (p = 0,003) kunde ses för pojkar positiva för DRB1*07:01-allelen. Höga MS-värden var mycket vanligare för DRB1*07:01-allelen.Slutsats: Ett samband mellan HLA-DRB1-alleler och mängden MS i saliven kan finnas. Resultaten indikerar att barn positiva för DRB1*04:01-allelen och homozygota DRB1*04-alleler har en tendens att uppvisa ett högre MS-värde i saliven jämfört med barn negativa för allelen. HLA-typen är antagligen bara en av många faktorer som påverkar mängden MS. Resultaten från denna studie är delvis i linje med tidigare studier och ytterligare studier behövs. / Objective: The aim of the study was to investigate a possible relationship of the different HLA-DRB1* alleles, with focus on HLA-DRB1*04, and the amount of Mutans Streptococci (MS) in saliva from a group of preschool children in the Southern part of Sweden, in the County of Skåne.Material and method: Saliva samples from 318 HLA-DRB1* typed children were cultivated on MSB agar and CFU of MS was counted. The results were statistically analyzed using chi-square tests.Results: No statistical significant relationship could be established between any DRB1*04 allele and the amount of MS. However, high numbers of MS was found to be about twice as common for homozygote DRB1*04 alleles (p = 0.364). High numbers of MS was more common for DRB1*04:01 alleles (p = 0.717). On the contrary, high levels of MS was much less common for DRB1*04:04 alleles (p = 0.098). A statistical significant relationship (p = 0.003) could be seen for boys positive for the DRB1*07:01 allele. High MS count was much more common for DRB1*07:01 alleles.Conclusion: A relationship between HLA-DRB1 alleles and the amount MS in the saliva might exist. The results indicate that children positive for the DRB1*04:01 allele and homozygote DRB1*04 alleles have a tendency to display more MS in their saliva compared to children negative to the alleles. The HLA type might just be one of many factors affecting the amount of MS. The results are partly in line with earlier studies and further studies are needed.

O objetivo desse trabalho foi o de avaliar a atividade antimicrobiana utilizando-se óleos essenciais de Cymbopogon citratus (D.C.) Stapf, Curcuma zedoaria Roscoe e Tagetes minuta L. sobre cepas de Staphylococcus spp., Streptococcus mutans e Candida spp. em crescimento planctônico e em biofilme. Para estudo dos microrganismos em crescimento planctônico, foram determinadas a Concentração Inibitória Mínima e a Concentração Fungicida Mínima de nove cepas clínicas e uma cepa padrão para cada espécie: C. albicans, C. tropicalis, C. glabrata, S. aureus, S. epidermidis e S. mutans. Para avaliação dos efeitos dos óleos essenciais em biofilme, foram utilizadas cepas padrão de Candida albicans (ATCC 18804), Staphylococcus aureus (ATCC 6538) e Streptococcus mutans (ATCC 35688) isolados e em associações em corpos-de-prova durante cinco dias. A seguir, os corpos-de-prova em resina acrílica foram lavados e colocados em contato com óleos essenciais durante 5 min. O número de unidades formadoras de colônias obtidas em cada biofilme (UFC/mL) foram submetidos à análise estatística descritiva e teste t-Student utilizando-se o software Minitab considerando-se o nível de significância p ≤ 0,05. Também foi realizada microscopia eletrônica de varredura (MEV) nos corpos-de-prova em resina acrílica com biofilme com tratamento e controle. Foram observadas reduções significativas do número de unidades formadoras de colônias (UFC/mL) tanto no biofilme como em associação. As maiores reduções ocorreram no tratamento do óleo essencial de C. citratus, seguidas pelo óleo de T. minuta e C. zedoaria. Stapf, Curcuma zedoaria Roscoe and Tagetes minuta L. on strains of Staphylococcus spp., Streptococcus mutans and Candida spp. in planktonic and biofilm growth. For the study of microorganisms in planktonic growth, were determined Minimum Inhibitory Concentration and Minimum Microbicidal Concentration of nine clinical strains and one ATCC for each species: C. albicans, C. tropicalis, C. glabrata, S. aureus, S. epidermidis and S. mutans. To assess the effects of essential oils in biofilm, was used strains of Candida albicans (ATCC 18804), Staphylococcus aureus (ATCC 6538) and Streptococcus mutans (ATCC 35688) alone and in associations in acrylic resin discs immersed in sterile brain heart infusion broth (BHI) containing 5% sucrose, inoculated with microbial suspension and incubated for 5 days. On the fifth day, the discs were washed in sterile saline solution in order to remove loosely bound material. Next, the acrylic resin discs were washed and placed in contact with essential oils for 5 min. The number of CFU obtained in each biofilm (CFU/mL) were subjected to descriptive statistical analysis using Minitab software considering with significance level p ≤ 0.05. It was also performed scanning electron microscopy (SEM) acrylic resin disc with biofilm with treatment and control. It was identified significant reductions in the number of colony forming units (CFU/mL) in both planktonic and biofilm associated. The largest reductions occurred in the treatment of essential oil of C. citratus, followed by oil of T. minuta and C. zedoaria. We conclude that the essential oils of Cymbopogon citratus... O interesse clínico e laboratorial por essas técnicas tem aumentado em função da realização de estudos que comprovaram a eficácia da remoção manual de dentina infectada, bem como as vantagens da técnica conservadora e indolor. O objetivo desse estudo foi analisar e comparar os tratamentos mecânico manual (ART convencional) e químico-mecânicos (CarisolvTM e Papa-cárie®) na remoção de dentina cariada, por meio da contagem de Streptococcus mutans e Lactobacillus e avaliação clínica de sintomatologia dolorosa durante o procedimento restaurador. Para a realização do presente estudo foram selecionadas 32 crianças, de ambos os sexos, com idade entre 6 e 10 anos, que apresentavam no mínimo dois molares decíduos, com exposição de dentina cariada. As crianças foram divididas aleatoriamente em dois grupos homogêneos, totalizando 64 dentes. O grupo I foi submetido à remoção mecânica em um elemento dental e em outro dente a químico-mecânica com o uso de Carisolv e o grupo II, a remoção mecânica e a químico-mecânica com o uso do Papacárie®. Durante a realização dos tratamentos foram feitas coletas de dentina, antes e após a remoção de tecido cariado e aplicação de um questionário para a avaliação clínica. Posteriormente, foram realizadas diluições seriadas e cultivo das amostras em meios específicos, Mitis salivarius bacitracina sacarose para a contagem de Streptococcus mutans e Rogosa, para contagem de Lactobacillus ssp. Após o período de incubação, a 37º C e 72 horas, foi realizada a contagem de Unidades Formadoras de Colônias (UFC) e obtidos os valores médios do número de bactérias. / Abstract: In Dentistry, nowadays, it has been emphasizing the practice of techniques less invasive for the removal of decayed tissue that preserve the maximum the dental structure. The clinical laboratorial interest for those techniques have been increasing due to studies that proved the effectiveness of the manual removal of infected dentine, as well as the advantages of the conservative and painless technique. The objective of this study was to analyze and to compare the manual mechanic (conventional ART) and chemical-mechanic (Carisolv and Papa-carie®) treatments in the removal of decayed dentine, through the score of Streptococcus mutans and Lactobacillus and clinical evaluation of pain symptoms during the restorative procedure. For the performance of the present study 32 children were selected, of both genders, with age between 6 and 10 years that presented at least two deciduous molars, with exhibition of decayed dentine. The children were divided at random in two groups, summing 64 teeth. The group I was submitted to the mechanical and chemical-mechanical removal with the use of Carisolv and the group II, mechanical and chemical-mechanic removal with Papacárie®. During the performance of the treatments, dentine sample were collected, before and after the removal of decayed tissue and application of a questionnaire for the clinical evaluation. Later, it was carried out seriate dilutions and cultivation of the samples in Mitis- salivarius bacitracin sucrose for the score of Streptococcus mutans and Rogosa, for score of Lactobacillus ssp. Composición química del aceite esencial de Citrus paradisi “Toronja”, actividad antioxidante y determinación de la actividad antibacteriana frente a Streptococcus mutans

Determina la composición química del aceite esencial de Citrus paradisi Macfad. “toronja”, actividad antioxidante y antibacteriana in vitro frente a Streptococcus mutans. El aceite esencial se obtiene tratando aproximadamente 25 kg de cáscara de frutos en un sistema de hidrodestilación por arrastre de vapor de agua a temperatura y presión controlada, obteniéndose un rendimiento de 0,13 mL% v/p, realizándose asimismo el análisis preliminar del aceite esencial y sus propiedades fisicoquímicas: gravedad específica (0,852 g/mL), Indice de Refracción (1,351) y pH (5,9). Del análisis cualitativo de la composición química realizado en Cromatografía de Gases/ Espectrometría de Masas (CG/EM), se destacan los siguientes componentes químicos: dieciséis monoterpenos, cinco sesquiterpenos, dos ácidos orgánicos, cuatro aldehídos, un éster, cinco hidrocarburos, un fenilpropanoide. La evaluación de la actividad antioxidante in vitro del aceite esencial se realiza utilizando el método: Captación del radical 2,2-difenilpicrilhidrazil (DPPH), los resultados indican que el aceite esencial de Citrus paradisi Macfad no posee actividad antioxidante frente al patrón estándar de Trolox ®. La determinación de la actividad antibacteriana in vitro del aceite esencial, se realiza utilizando el método de difusión en agar mostrando actividad antibacteriana frente a Streptococcus mutans ATCC 35668 en 100, 50 y 25 por ciento, utilizando el antibacteriano ciprofloxacino como control positivo. / Tesis
