Influência do laser de baixa potência (GaAlAs) na remodelação óssea, após a disjunção da sutura palatina mediana em ratos Wistar / Influence of low level laser (GaAlAs) on bone remodeling after midpalatal suture expansion in Wistar rate

Fundação de Amparo à Pesquisa do Estado do Rio de Janeiro / O propósito deste estudo foi analisar o comportamento do osso alveolar na remodelação óssea sob carga fisiológica e ortopédica após a disjunção rápida da sutura palatina mediana durante 14 dias, com e sem aplicação de laser de baixa potência e comparar imagens obtidas por técnica radiográfica convencional com microtomografias computadorizadas. O aparelho de laser utilizado foi de diodo de arsenieto de gálio-alumínio (AlGaAs) (830m; 100mW, onda contínua, 126J/cm2, 29 segundos) aplicado à sutura palatina mediana de ratos Wistar, machos, de 6 semanas de vida. Foram utilizados 65 ratos divididos em um grupo controle (GC) contendo 5 ratos e dois experimentais I e II (GE I e GE II), com 30 ratos cada, ambos GE submetidos à disjunção imediata, sendo que o GE II foi irradiado imediatamente após a disjunção. Os GE I e II foram subdivididos em períodos de disjunção imediata, 6, 24, 48 horas, 7 e 14 dias. Dois amostras de cada período dos GC e GE foram radiografadas (convencional) e processadas para captura de imagens em scanner micro-CT (Scanco Medical, Zurik, Suíça), com 24KV de energia e 3,8μm de resolução espacial e o restante foram processadas para serem analisadas histologicamente coradas com hematoxilina e eosina e tricrômico de Masson. Os resultados foram avaliados histológica e radiograficamente. O trabeculado ósseo recém-formado a partir dos bordos ósseos da sutura foi observado nos períodos de 2, 7 e 14 dias do GE II, 7 e 14 dias do GE I. O presente experimento demonstrou que a atividade osteoblástica aumenta a atividade da osteosíntese de modo mais expressivo após aplicação do laser de baixa potência, porem é necessários mais estudos em biologia molecular para desvendar os seus efeitos ao nível celular. As microtomografias computadorizadas mostraram maior riqueza de detalhes para analisar o rompimento da sutura palatina mediana e seu reparo com o depósito de tecido ósseo do que as radiografias convencionais. / The purpose of this study was to evaluate the behavior of the alveolar bone on bone remodeling under physiological load and on after rapid midpalatal suture expansion during a 14-day period, with and without low-level laser irradiation and to compare images obtained by conventional radiographic technique with microtomografias computed. The laser device was a gallium-aluminum-arsenide (GaAlAs) diode laser (830 m; 100 mW, continuous wave, 126 J/cm2, 29 seconds) applied to the midpalatal suture of 6-week-old male Wistar rats. Sixty-five rats were assigned in a control group (CG) containing 5 rats and two experimental I and II (EG I and EG II), with 30 rats each, both EG submitted to expansion immediately, while the EG II was irradiated pos expansion immediately. The EG I and II were divided into periods of immediate midpalatal suture expansion, 6 hours, 1, 2, 7 and 14 days). . Two samples of each period of CG and EG were radiographed (conventional) and processed to capture images in micro-CT scanner (Scancem Medical, Zurik, Switzerland) with 24kV of energy and 3.8 mm in spatial resolution and the rest were processed for are analyzed histologically stained with hematoxylin and eosin and Masson's trichrome.The results were evaluated histologically and radiographically. Bone trabeculae recently formed from the bone borders of the suture were observed in the periods of 2, 7 and 14 days of EG II, and 7 and 14 days of EG I. The present experiment demonstrated that the osteoblastic activity increases the osteosynthesis activity in a more manner after low-level laser irradiation. However, more studies in the field of molecular biology are necessary to determine its effects at cellular level. The computed microtomographs provided a more detailed analysis of midpalatal suture opening and its repair by bone tissue deposition than conventional radiographs.

Reparo do nervo facial com sutura epineural térmico-terminal e coaptação com adesivo de fibrina em ratos associado ou não a laserterapia / Repair of facial nerve with epineural end-to-end suture and coaptation with fibrin adhesive in rats associated or not laser therapy

As lesões que envolvem nervos periféricos, especialmente os traumatismos facias, são muito comuns e decorrentes principalmente de acidentes com veículos motorizados, lesões acidentais e quedas, que levam a fraturas do osso temporal ou lacerações da face e consequentemente lesões do nervo facial. A principal meta no estudo da regeneração nervosa é descobrir uma técnica adequada de reparo em lesões de nervos periféricos que traga como resultado a recuperação funcional das estruturas por eles inervadas. A sutura epineural é um método muito utilizado para recuperação de lesões nervosas, assim como o uso do adesivo de fibrina, que requer menor destreza do cirurgião. O adesivo derivado do veneno de serpente (CEVAP/UNESP, Botucatu-SP) é um selante biológico e biodegradável, pois não produz reações adversas, não contém sangue humano, apresenta uma boa capacidade adesiva, não transmite doenças infecciosas, e pode ser utilizado como coadjuvante em procedimentos de sutura convencional. Sendo assim, o objetivo deste trabalho foi comparar duas técnicas de recuperação de nervos periféricos lesionados: a sutura epineural término-terminal e o adesivo de fibrina derivado do veneno de serpente, e observar se o uso da laserterapia de baixa potência influencia esse processo de regeneração. Para isso, foram utilizados 42 ratos machos (Rattus norvegicus, Wistar), com 60 dias de vida, separados aleatoriamente em um Grupo Controle e quatro Grupos Experimentais, assim formados: Grupo Controle (GC, n=10), em que foi coletado o nervo facial íntegro aos 95 e 135 dias de vida; Grupo Experimental Sutura (GES, n=16) e Grupo Experimental Adesivo de Fibrina (GEF, n=16), onde no lado direito da face o ramo bucal do nervo facial foi seccionado e realizado a sutura epineural término-terminal e, no lado esquerdo da face, o ramo bucal do nervo facial foi seccionado e utilizado o adesivo de fibrina para coaptação das extremidades; Grupo Experimental Sutura e Laserterapia (GESL, n=16) e Grupo Experimental Adesivo de Fibrina e Laserterapia (GEFL, n=16), submetidos aos mesmos procedimentos de GES e GEF, adicionada a aplicação de Laser de Arseneto de Gálio Alumínio (GaAlAs) de pulso contínuo, comprimento de onda de 830 nm, 6J/cm2, por 24 segundos, três vezes por semana durante cinco semanas, em três pontos dos locais operados. Os animais dos Grupos Experimentais foram eutanasiados com 95 dias (cinco semanas pós-cirurgia) e 135 dias (dez semanas pós-cirurgia). As amostras coletadas foram submetidas à análise morfológica por microscopia óptica e eletrônica de transmissão, além de análise morfométrica da área e diâmetro da fibra, área e diâmetro do axônio, espessura e diâmetro da bainha de mielina. Observou-se brotamento axonal no coto distal do nervo facial em todos os Grupos Experimentais, com morfologia semelhante às fibras do Grupo Controle, e predomínio de fibras mielínicas sobre as amielínicas. Pode-se concluir que a reparação por meio da sutura epineural término-terminal apresentou melhores resultados em relação ao adesivo de fibrina e a laserterapia de baixa potência não influenciou o processo de regeneração. / The injuries involving peripheral nerves, especially facial traumatisms are very common and serious and longstanding facial paralysis lead to significant deterioration in the quality of the individuals life. The main goal in the study of nerve regeneration is finding a suitable repair technique for peripheral nerve injuries that bring results in the functional recovery of the structures innervated by them. Therefore, the aim of this study was to compare two techniques for recovery of injured peripheral nerves: the end-to-end epineural suture and fibrin adhesive derived from snake venom (CEVAP / UNESP, Botucatu-SP), and observeif the use of low-level laser therapy influences this regeneration process. For this purpose, 42 male rats (Rattus norvegicus, Wistar) were used , with 60 days of life, were randomly separated into a control group and four experimental groups, which were formed this way: Control Group (CG , n = 10), in which the intact facial nerve was collected at 95 and 135 days of life; Experimental Suture Group (ESG, n = 16 ) and Experimental Fibrin Adhesive Group (EFG, n = 16), where the right side of the face the buccal branch of the facial nerve was transectioned and the epineural end-to-end suture was performed and the left side of the face, the buccal branch of the facial nerve was transectioned and the fibrin glue was used for coaptation of the edges; Experimental Suture Laser Therapy Group (ESLG, n = 16) and Experimental Fibrin Adhesive Laser Therapy Group (EFLG, n = 16) underwent the same procedures of ESG and EFG , included the application of Laser Gallium- Aluminum-Arsenide (GaAlAs) by an 830 nm wavelength pulse continuous, 6 J/cm2, for 24 seconds, three times a week during five weeks at three points of the operated areas. The animals in the experimental groups were euthanized at 95 days (five weeks post-surgery) and 135 days (ten weeks post-surgery). The collected samples were subjected to morphological analysis by optical microscopy and transmission electronic microscopy, besides the morphometric analysis of the area and diameter of the fiber, area and diameter of the axon, thickness and diameter of the myelin sheath. It was observed axonal sprouting in the distal stump of the facial nerve in all experimental groups, with similar morphology to the fibers of the control group, and predominance of myelinated fibers to amyelinated. It can be concluded that the repair by epineural end-to-end suture showed better results in relation to fibrin adhesive and the low-level laser therapy did not influence the regeneration process

RESUMO A Expansão Rápida da Maxila é um recurso clínico adotado pelos ortodontistas no tratamento das más oclusões com deficiência transversa real ou relativa da maxila. Esta, inclui uma fase ativa, promovendo a desarticulação dos ossos maxilares por meio da sutura, e uma fase passiva, onde uma série de eventos levam à remodelação óssea e restauração da sutura, com o aumento clínico na largura do palato. Discute-se qual a melhor época para a disjunção e o tempo necessário para que haja a perfeita remodelação óssea. A radiografia oclusal assume aqui um importante papel, pois é por meio desta que se obtém a imagem da sutura palatina mediana permitindo uma análise descritiva de suas condições em todas as etapas do tratamento A nossa proposta no presente estudo, foi a de medir as densidades ópticas da sutura palatina mediana antes da disjunção (Fase I), imediatamente após a disjunção, (Fase II) e após contenção de três meses (Fase III), usando um sistema de imagem digital, comparando-as e correlacionando-as com a radiografia oclusal convencional. A amostra, constituída por 31 (trinta e um) pacientes que se submeteriam à disjunção palatina e que se encontravam em fase de crescimento, foi dividida em:grupo I, constituído pelos pacientes de dentadura mista e grupo II, pelos pacientes de dentadura permanente. Foram obtidas radiografias oclusais e imagens digitais nas fases Fase I, Fase II e na Fase III. Áreas de interesse foram demarcadas nas radiografias digitais, procedendo-se a leitura das densidades ópticas pelo programa Digora for Windows-2.1, e comparando os resultados entre si e com a avaliação subjetiva feita nas radiografias oclusais. Após a análise dos dados pelo teste t de Student pareado, pode-se concluir que 1) as densidades ópticas da sutura palatina mediana na fase final, mostram valores inferiores aos da fase inicial nos pacientes do grupo I; 2) as densidades ópticas da sutura palatina mediana na fase final, mostram valores semelhantes aos da fase inicial nos pacientes do grupo II; 3) as densidades ópticas finais da sutura palatina mediana não correspondem às iniciais nos pacientes do sexo feminino do grupo I; 4) as densidades ópticas finais da sutura palatina mediana são semelhantes às iniciais nos pacientes do sexo masculino do grupo II; 5) para os pacientes do grupo II não houve diferença significante entre os valores de densidade óptica inicial e final, tanto no sexo masculino como feminino; 6) os resultados encontrados nas imagens digitais são compatíveis com os encontrados nas radiografias oclusais convencionais. / SUMMARY The Fast Expansion of the Jaw is a clinical resource adopted by the ortodontists in the treatment of the bad occlusions with real or relative deficiency transversal, of the jaw. This, includes a phase, promoting the disarticulation of the maxilarys bones by means of the suture, and a passive phase, where a series of events takes to the bone remodelling and restoration of the suture, with the clinical increase in the width of the palate. The best time for the disjunction and the necessary time is argued which so that it has the perfect bone remodelling. The oclusal radiography assumes an important role here, therefore it is for way of this that if gets the image of the medium palatine suture allowing to a descriptive analysis of its conditions in all the stages of the treatment our proposal in the present study, was to measure the optic densities of the medium palatine suture before the disjunction (Phase I), immediately after the disjunction, (Phase II) and after containment three months (Phase III), using a system of digital image, comparing and correlating them with the conventional oclusal radiography. The sample, consisting of 31 (thirty one) patient that would submitted the palatine disjunction and that found in growth phase, divided in Grave I, constituted of the patients of mixing set of teeth and group II, for the permanent set of teeth patients. Oclusal radiography and digital images in the phases had been gotten Phase I, Phase II and in Phase III. Areas of interest had been demarcated in the digital x-rays, proceeding it reading from the optic densities for the Digora program for Windows-2.1, and comparing the results among themselves. With the subjective evaluation in the oclusal radiographies.(After the analysis of the data by student T test could be concluded that1) the opitic densities of the medium palatine suture in the final phase, show inferior values to the ones of the initial phase in the patients of group I;2) the optic densities of the medium palatine suture in the final phase, show similar values to the ones of the initial phase in the patients of group II;3) the final optic densities of the medium palatine suture do not correspond to the initials in the patients of the female groupI;4) the final opitic densities of the medium palatine suture are similar to the initials in the patients of the male group II;5) for the patients of group II it did not have significant difference betweenthe values of initial and final opitic density, as much in the male and female groups6) the results found in the digital images are compatible wit those found in the conventional oclusal radiographics.

Estudo comparativo entre a capsulorrafia com sutura simples e com âncora em quadris de coelhos / Comparative evaluation between capsulorrhaphy with simple suture and with anchor in rabbit hip joints

INTRODUÇÃO: A displasia do desenvolvimento do quadril (DDQ) é uma das patologias mais relevantes e polêmicas que acometem as crianças desde o nascimento. A anatomia tridimensional, a complexidade da articulação do quadril, o pouco conhecimento sobre o potencial de remodelação acetabular após luxação ou sub-luxação e as sequelas na marcha e na movimentação suscitam várias discussões sobre esse tema. A revisão bibliográfica a respeito dos diferentes tipos de capsulorrafia é muito pouco discutida entre os pesquisadores. Técnicas menos agressivas e que possibilitem maior resistência à recidiva da luxação após a redução cruenta devem ser pesquisadas. OBJETIVO: a presente pesquisa busca, por meio de ensaios biomecânicos, comparar as capsulorrafias com sutura simples e com âncoras, em quadris de coelhos. MATERIAL E MÉTODO: Foram utilizados 13 coelhos, 26 quadris, todos machos da raça Nova Zelândia albinos (Oryctolagus cuniculus). Inicialmente, realizamos um projeto piloto em três coelhos (06 quadris). Este experimento constou de 10 coelhos, divididos em 02 grupos: o grupo 1 submetido à capsulorrafia (quadris direito e esquerdo) com sutura simples utilizando fio absorvível de ácido poliglicólico e o grupo 2 submetido a capsulorrafia (quadris direito e esquerdo) com âncora de titânio. Após o período de quatro semanas de operados, todos animais foram submetidos à eutanásia e seus quadris congelados. Após um descongelamento prévio das peças no mesmo dia das análises biomecânicas, foram avaliados os parâmetros da rigidez (Rig), força máxima (Fmax), deformidade máxima (Dmax) e energia (E). RESULTADO: não houve diferença estatisticamente significante em relação à força no limite de proporcionalidade, rigidez e força máxima entre os grupos com sutura simples e com âncora. CONCLUSÃO: Por meio dos ensaios biomecânicos, tendo como parâmetro a rigidez (Rig), força máxima (Fmax), deformidade máxima (Dmax) e energia (E), ficou demonstrado que as capsulorrafias em quadris de coelhos com sutura simples e com âncora são semelhantes entre si. / INTRODUCTION:HDD (Hip Development Dysplasia) is one of the most important and controversial pathologies which affect children. The threedimensional anatomy and complexity of the hip joint, as well as the little understanding of the potential of acetabular reconstruction after luxation or sub-luxation and the later effects on the child\'s gait and movement, raise various points of discussion. Little literature exists about the different types of capsulorrhaphy. Techniques which are less aggressive or decrease risk of luxation after surgical reduction must be researched. OBJECTIVE: Using biomechanical studies, this research aims to compare hip capsulorrhaphy in rabbits, carried out with two different techniques: capsulorrhaphy with simple sutures and with anchors. MATERIAL AND METHOD: Thirteen New Zealand Albino (Oryctolagus cuniculus) male rabbits, twenty-six hip joints, were used. First, a pilot project was performed with three rabbits (six hip joints). The experimental group consisted of ten rabbits and was divided in two sub-groups: group 1 underwent capsulorrhaphy on both right and left hips with simple suture using polyglocolic acid absorbable thread, and group 2 underwent capsulorrhaphy with titanium anchors. After a four-week postoperation period, the animals were euthanized and the hip joints were frozen. On the same day the hip joints were unfrozen, a biomechanical study was carried out, evaluating the following parameters: rigidity, maximum force, maximum deformity and energy. RESULTS: There was no relevant statistical difference in rigidity, maximum force, maximum deformity and energy between the simple suture and anchor groups. CONCLUSION: Through biomechanical analyses, using parameters of rigidity, maximum force, maximum deformity and energy, it has been shown that a capsulorrhaphy with simple suture and with an anchor has similar results in rabbit hip joints.

Estudo biomecânico de duas técnicas extra-capsulares para estabilização do joelho com ruptura do ligamento cruzado cranial em cães / Biomechanical study of two extra capsular techniques for stabilization of the knee with cranial cruciate ligament rupture in dogs

A instabilidade do joelho resultante da ruptura do ligamento cruzado cranial apresenta-se como sendo uma das principais causas de dor, em membro pélvico, manifestada por animais da espécie canina. Tal instabilidade pode gerar alterações como o desenvolvimento de osteoartrose e lesão de menisco, principalmente o medial. Com o intuito de restabelecer a estabilidade da articulação, muitas técnicas vêm sendo desenvolvidas de forma que as alterações provenientes de tal afecção sejam minimizadas. Com uma dimensão similar de variedades estão os materiais utilizados em diferentes técnicas ou até mesmo para o mesmo tipo de procedimento. A técnica ideal, bem como o material de escolha ainda são muito discutidos dentre os autores pesquisados. O presente trabalho avaliou a biomecânica das técnicas de sutura fabelo-tibial lateral (SFTL) e a técnica de imbricação do retináculo modificada (SFTLM), em joelhos de cadáveres de cães, na estabilização do joelho com ruptura do ligamento cruzado cranial, utilizando-se fio de náilon monofilamento leader line (60 libras). Foram avaliados rigidez articular e deslocamento craniais e caudais da tíbia em relação aos côndilos do fêmur, proporcionados pelo tipo de material e técnicas cirúrgicas, sendo os resultados obtidos comparados com os resultados do joelho íntegro e entre si. A análise estatística encontrou diferença na gaveta cranial de todos os grupos (ligamento cruzado cranial íntegro (LCCr), ligamento cruzado cranial rompido (RLCCr), SFTL e SFTLM) e na comparação da gaveta caudal entre o grupo RLCCr e SFTLM e entre o grupo SFTL e SFTLM. Outras diferenças foram encontradas na rigidez cranial dos grupos LCCr, RLCCr e reconstruído, não sendo observado diferença entre a rigidez de SFTL e SFTLM. As duas técnicas testadas diminuem a instabilidade do joelho, sendo a SFTLM mais efetiva em tal promoção, porém tal técnica influencia de forma mais evidente na biomecânica do ligamento cruzado caudal, sendo importante a avaliação de tal interferência a longo prazo. Estas duas técnicas extra-capsulares testadas, apesar de diminuirem significativamente a translação cranial da tíbia em relação ao fêmur, não restauram a rigidez articular quando comparadas com a biomecânica do joelho íntegro. / Knee instability resulting from an insufficient cranial cruciate ligament is the major cause of hind limb lameness in dogs. This instability can result in osteoarthrosis and medial meniscal injury. Many surgical techniques have been developed to restore knee stability and to reduce degenerative joint desease. There is a similarly huge variability in materials utilized for different or the same techniques. An ideal technique and material arent established yet. This study analyzed the biomechanics of the lateral fabellar suture technique (SFTL) and the modified retinacular imbrication technique (SFTLM) in stabilizing cadaveric cranial cruciate ligament deficient stifles with 60 lb nylon leader line. Articular cranio-caudal stiffness and tibial translation were evaluated and compared between different techniques and with the intact knee. The statistical analysis revealed significant differences between all groups (intact cranial cruciate ligament (LCCr), ruptured cranial cruciate ligament (RLCCr), SFTL and SFTLM) in the cranial tibial translation. In the caudal tibial translation a significant statistical difference was seen between RLCCr and SFTLM groups and between reconstructed ones. Other differences were found in the cranial stiffness between LCCr, RLCCr and reconstructed groups, but not between SFTL and SFTLM. Both techniques tested reduced joint instability but SFTLM was more efficient, although it affected the caudal cruciate ligament biomechanics more evidently when compared with the SFTL technique. This long-term evaluation of the caudal influence induced by SFTLM will be important. Despite of the reduction in the tibial cranial translation by these two different extra capsular techniques, none have maintained normal stifle stiffness.

ENGLISH ABSTRACT The Late Devonian to Early Carboniferous kinematic evolution of the Teplá-Barrandian/Moldanubian boundary The Staré Sedlo complex (SSC) is a relic of meta-igneous arc-related pluton in the southern part of the Sedlčany-Krásná Hora roof pendant, intruded by granitoids of the Central Bohemian Plutonic Complex along the boundary of Teplá-Barrandian (TBU) and Moldanubian units (MU), Bohemian Massif. The SSC mainly comprises deformed orthogneisses of calc-alkaline granodiorite to tonalite protoliths of Late Devonian age (380−365 Ma; Košler et al., 1993) that were commonly mingled with minor basic magmas. Locally preserved subhorizontal intrusive contacts of the orthogneisses against their meta-sedimentary host rock indicate that these magmas intruded as a sill complex. The SSC preserves a rather unusual flat-lying subsolidus foliation (dip <40ř) associated with subhorizontal ~NE-SW-trending mineral lineation. Mesoscopic structures, anisotropy of magnetic susceptibility (AMS), and deformational microstructures indicate prolate shape of the strain ellipsoid with dominant coaxial pure shear regime. The solid state microstructures record cooling of the orthogneiss protolith down to the ambient greenschist facies conditions followed by its static recrystallization due to the intrusion of the younger...

Stem cells are crucial players during development, homeostasis and tissue regeneration and their interactions with the surrounding microenvironment are key to regulate stem cell fate. The skull's stem cell niches reside in the fibrous joints that connect flat bones of the skull. In the embryo, bone and sutures develop in concert to form a complex, multi-facted structure that requires interaction with multiple differentiating cell types to maintain balance between growth and differentiation. Disruption of this balance drives changes in size and shape of skull bones and can severely impact quality of life. Cranial sutures, often seen as simple extracellular matrix-rich structures bridging the rigid plates of the skull, are major actors in craniofacial morphogenesis of as they harmonize bone growth with expansion of the developing brain and participate in providing osteoblasts during repair. The complexity of the extracellular environment and the important role for sutures in skeletal development makes these niches a compelling structure to investigate how interactions with the surrounding microenvironment can modulate stem cells fate. The key role of sutures in development is highlighted by the numerous severe dysmorphisms arising from failure to maintain suture patency. The ability of the suture to respond to brain growth or trauma and the dysmophisms presented by patients with defective sutures is mediated by both biochemical and mechanical cues but the cell biology of these niches remains elusive, especially during their development. In particular, few studies have shed light on the underlying cellular behaviors behind microenvironmental regulation of cranial suture stem cell fate and what role mechanical inputs play in the establishment of this niche. In my thesis, I addressed gaps in our understanding of suture biology by characterizing the suture stem cell niche microenvironment and exploring how cell-ECM interactions serve as regulators of suture stem cell fate. Making use of various microscopy and analytical techniques I first characterized the composition of the microenvironment in a developing suture niche, such as organization of ECM, cytoskeleton and nuclear morphologies. My work builds on an incomplete transcriptional understanding of suture cell development, such that specific genetic markers are rarely useful for identifying distinct suture cell populations during its morphogenesis. By applying shape description tools to parse suture cells and test whether shape correlates to cell identity, we concluded that suture nuclei are distinct and less spherical than those of other cranial tissues. Using 'global' markers such as nuclear stains, I have also identified physical distinctions between suture nuclei and neighboring tissues, indicating that cell shape is an integral part of midline suture identity and can be used to explore coordination of fate choice and morphogenesis in this enigmatic structure. In addition, I present evidence that supports that maturation of extracellular matrix begins during early stages of suture development. In particular, embryonic midline sutures express high levels of fibrillary collagen, which contributes to the formation of a complex extracellular environment that provides the suture with physical properties distinct from those of developing bones. My work shows the presence of cell-ECM and cell-cell adhesions in the developing midline sutures, as well as a complex actin cytoskeleton that is, in part, mediated by physical stresses resultant from underlying brain expansion. Secondly, I aimed to address how perturbations in ECM composition can affect cell specification. To investigate the importance of ECM maturation in regulating suture cell fate I inhibited the function of lysyl oxidase, a collagen crosslinker, during embryonic development. Disruption of collagen crosslinking altered expression of collagen and ECM receptor encoding genes. In addition, this inhibition induced changes in the shape and size of collagen fibers in the embryonic midline suture and decreased tissue bulk stiffness relative to WT. These abnormal properties of the ECM impact tissue delineation in the cranial mesenchyme through nuclear shape analyses. This might be explained by observed changes in the composition of the nuclear envelop of suture cells as we find altered lamin concentration and localization upon lysyl oxidase inhibition. The work developed during myPhD steps away from the traditional genetic approaches used to study the embryonic suture and provides the first in-depth analysis of the physical properties of the developing midline suture at stages preceding known establishment of the niche. The various methods and analyses applied reveal a complex organization of embryonic suture ECM and its tight relationship with shape and fate in this tissue. This work serves as a foundation for future studies that can explore the mechanisms through which ECM regulates fate and development of the suture niche, and potentially skeletal development more generally.

In-vitro Untersuchungen zur Gelenkstabilität und Fadenspannung nach lateraler Fadenzügelung am Kniegelenk des Hundes

Zielstellung: Gegenwärtig wird nach Ruptur des vorderen Kreuzbandes (VKB) beim Hund häufig eine laterale Fadenzügelungen (LFZ) vorgenommen. In der vorliegenden Arbeit wurden folgende Ziele verfolgt: (1) Bestimmung der kranio-kaudalen Kniegelenkstabilität nach LFZ und passiver Kniegelenksbewegung. (2) Bestimmung der Schlaufenspannung nach Knotung mit der Hand. (3) Einfluss von vier häufig verwendeten Verankerungsformen auf die LFZ und von drei Kniegelenkwinkeln zum Zeitpunkt der Knotung des Fadenzügels auf den Spannungsverlauf innerhalb des Fadens. Material und Methode: Die kranio-kaudale Kniegelenkstabilität (KKStab) wurde anhand von latero-medialen Röntgenaufnahmen an 10 rechten Kniegelenken orthopädisch gesunder Hunde (>20 kg KM) ex-vivo bestimmt. Die Messung der KKStab erfolgte am intakten Gelenk, nach Durchtrennung des VKB und nach lateraler Ethibondfadenzügelung. Anschließend wurden die Gelenke 350 mal passiv gebeugt und gestreckt und die KKStab nach 50, 100, 250 und 350 Bewegungszyklen erneut bestimmt. Des Weiteren wurde die Spannung in 7 Ethibondfadenschlaufen an einer Materialprüfmaschine bestimmt. Die Spannung in der Schlaufe wurde nach dem ersten (FMax) und nach dem letzten Knoten (FEnd) gemessen. Mit Hilfe eines speziell konstruierten Kraftaufnehmers wurde die Spannung in der lateralen Fadenzügelung über den passiven Bewegungsradius des Kniegelenks in weiteren 9 Kniegelenken in-vitro gemessen. Dabei wurde nach Durchtrennung des VKB jedes Kniegelenk mit 4 Formen der LFZ bei 3 unterschiedlichen Kniegelenkswinkeln zur Zeit der Fixierung der Fadenschlaufe stabilisiert. Ergebnisse: KKStab nach Stabilisierung des Gelenkes betrug im Mittel 3,7 mm und war im Mittel um 2,7 mm größer als bei intaktem Kreuzband. Bereits 250 passive Bewegungszyklen bewirkten eine signifikante Destabilisierung um weitere 1,5 mm. FMax beim Knüpfen von sieben Fadenschlaufen betrug im Mittel 133 N und fiel auf 6 N (FEnd) im Mittel nach dem fünften Knoten ab. Alle 4 untersuchten LFZ-Formen zeigten eine Zunahme der Fadenspannung bei Beugung des Kniegelenkes. Die LFZ, bei der die Zügelung um die laterale Fabella geführt und in 2 parallelen Bohrungen in der proximalen Tibia fixiert wurde, zeigte die geringsten Spannungsänderungen über einen kompletten Bewegungsradius des Kniegelenks. Die Fixation der LFZ in einen Gelenkwinkel von 70° ergab einen signifikanten Abfall der Fadenspannung in Extension des Gelenkes. Schlussfolgerungen: Eine LFZ aus Ethibondfäden mit fünffacher Knotung erlaubt es nicht, die physiologische Gelenkstabilität nach Durchtrennung des VKB herzustellen. Bereits wenige passive Bewegungszyklen bewirken eine signifikante weitere Destabilisierung. Die in diesem Teil der Studie angewendete Knotentechnik erlaubte es nicht, die manuell in der Fadenzügelung aufgebaute Spannung dauerhaft zu konservieren und sollte deshalb keine Klinische Verwendung haben. Keine der getesteten LFZ-Formen zeigte einen gleichmäßigen Spannungsverlauf bei Bewegung des Kniegelenks. Aus diesem Grund sollten die aktuellen Empfehlungen hinsichtlich „isometrischer“ Verankerungspunkte kritisch betrachtet werden. Die Fixierung der LFZ bei gebeugtem Kniegelenk ist nicht zu empfehlen, da es in Extension zur Destabilisierung kommt.

High-P granulite facies metamorphism from the Tibetan Plateau and the Himalaya : metamorphic history and geochemistry of lower crustal and early subduction metamorphic rocks

Cette thèse porte sur deux suites de roches métamorphiques de haute-pression et haute température provenant de l'Orogène Tibeto-Himalayen. La première suite de roches consiste en des affleurements d'amphibolites à grenat et clinopyroxène se retrouvant sous la forme de blocs dans le mélange ophiolitique à matrice de serpentine sous-jacent aux ophiolites de la Zone de Suture du Yarlung Zangbo, au Sud Tibet. La Zone de Suture du Yarlung Zangbo est un linéament de plus de 2000 km de long situé à la bordure méridionale du plateau Tibétain, au nord de la crête Himalayenne. Elle contient les reliques du vaste océan qui séparait l'Inde du Tibet pendant le Jurassique et le Crétacé : la Téthys. Dans la suture, des fragments de lithosphère océanique ont été préservés sous la forme d'une ceinture ophiolitique discontinue sous laquelle se retrouve un mélange ophiolitique. Les roches documentées dans la première partie de cette thèse ont été échantillonnées dans les occurrences de Bainang et de Angren/Buma, près de Xigaze, et plus à l'ouest sous l'ophiolite de Saga. Les relations de terrain suggèrent que ces roches représentent une semelle sub-ophiolitique démembrée. Sur la base des teneurs en éléments majeurs et traces de ces roches, cette semelle métamorphique aurait une affinité de N-MORB ou de BABB très similaire à celle de la croûte des ophiolites sus-jacentes. La géochronologie en Ar/Ar sur hornblende indique un âge de refroidissement entre 130 et 123 Ma. Considérant les modélisations complétées pour d'autres semelles métamorphiques dans le monde, ces âges peuvent également être considérés comme datant de très près le pic métamorphique. Les conditions du pic métamorphique on été contraintes thermobarométriquement et sont supérieures à 13 kbar et 800°C avec des moyennes dans l'ordre de 15 kbar et 850°C. Les relations de terrain, les données de littérature concernant les unités associées ainsi que la géochimie, la géochronologie et l'histoire métamorphique de la semelle subophiolitique de la Zone de Suture du Yarlung Zangbo supportent le modèle géodynamique suivant. Pendant le Jurassique ou le Crétacé Inférieur, la croûte des ophiolites du Yarlung Zangbo et le protolithe de sa semelle métamorphique sont formés à un centre d'expansion situé dans une zone de supra-subduction comprenant un bassin d'arrière-arc mature. Vers 130 Ma, une perturbation tectonique majeure change la direction relative des plaques et force l'initiation d'une nouvelle subduction localisée sur la ride d'extension du bassin arrière-arc. Le résultat est une ophiolite d'affinité d'arrière-arc piégée en contexte d'avant-arc et sous laquelle se retrouve une semelle métamorphique du faciès des granulites de haute-P et d'affinité d'arrière-arc. La deuxième suite de roches étudiée dans cette thèse consiste en des migmatites alumineuses à kyanite retrouvées dans le coeur de l'Antiforme du Namche Barwa, au sein de la Syntaxie Himalayenne Orientale. Le coeur de l'Antiforme du Namche Barwa est un dôme métamorphique à extrusion très rapide montrant des taux de denudation et d'exhumation extrêmes (~10mm/a). Il est situé à l'extrémité orientale de la chaîne Himalayenne et comporte la gorge la plus profonde de la planète. Les roches étudiées se retrouvent sous la forme de lentilles enrobées dans le gneiss migmatitiques à sillimanite qui forme la majorité du coeur de l'antiforme. Les lentilles migmatitiques à kyanite, d'âge Éocène-Oligocène (Zhang et al. 2010), ont été interprétées comme représentant la croûte inférieure du plateau Tibétain mais leur pic métamorphique dans le faciès des granulites de haute-pression était jusqu'à aujourd'hui contesté. Dans la présente étude, ces roches ont été investiguées quant à leur minéralogie, leur géochimie, les relations texturales entre les minéraux qui les composent et leur chimie minérale. Les résultats ont été interprétés à l'aide de pseudosections. L'interprétation confirme que ces roches représentent des protolithes sédimentaires alumineux ayant été enfouis à des conditions de croûte inférieure de l'ordre de 15 kbar et 850°C où ils ont perdu une proportion de leur liquide anatectique. Cependant, une proportion significative de liquide anatectique est restée piégée dans le réseau cristallin réfractaire, donnant lieu à d'importantes modifications texturales pendant l'exhumation jusqu'à des conditions de l'ordre de 10 kbar et 800°C, correspondant à la solidification finale du liquide piégé. Les résultats des modélisations suggèrent également que le potentiel de fusion par décompression d'une croûte inférieure aussi chaude est très faible puisque la plus grande proportion du liquide anatectique est produite pendant l'enfouissement. Cette étude démontre que la croûte inférieure Tibétaine était déjà fortement épaissie et très chaude peu après la collision initiale Éocène.
