Fluorescent and colorimetric molecular recognition probe for hydrogen bond acceptors

Yes / The association constants for formation of 1 : 1 complexes between a H-bond donor, 1-naphthol, and a diverse range of charged and neutral H-bond acceptors have been measured using UV/vis absorption and fluorescence emission titrations. The performance of 1-naphthol as a dual colorimetric and fluorescent molecular recognition probe for determining the H-bond acceptor (HBA) parameters of charged and neutral solutes has been investigated in three solvents. The data were employed to establish self-consistent H-bond acceptor parameters (β) for benzoate, azide, chloride, thiocyanate anions, a series of phosphine oxides, phosphate ester, sulfoxide and a tertiary amide. The results demonstrate both the transferability of H-bond parameters between different solvents and the utility of the naphthol-based dual molecular recognition probe to exploit orthogonal spectroscopic techniques to determine the HBA properties of neutral and charged solutes. The benzoate anion is the strongest HBA studied with a β parameter of 15.4, and the neutral tertiary amide is the weakest H-bond acceptor investigated with a β parameter of 8.5. The H-bond acceptor strength of the azide anion is higher than that of chloride (12.8 and 12.2 respectively), and the thiocyanate anion has a β value of 10.8 and thus is a significantly weaker H-bond acceptor than both the azide and chloride anions. / Supported by the ESPRC.

Optische Spektroskopie in der Ultraschallfalle

Die Kopplung von akustischer Levitation mit optischer Spektroskopie ermöglicht die Untersuchung von Reaktionen und intermolekularen Wechselwirkungen unter besonderen Bedingungen. Mit akustischer Levitation können Proben im µL-Bereich kontaktfrei gehaltert und Verunreinigungen durch Oberflächenkontaminationen ausgeschlossen werden. Durch die Verwendung verschiedener Methoden der optischen Spektroskopie wie Raman-Streuung, UV/Vis-Absorptions- und Fluoreszenz-Spektroskopie konnten die levitierten Proben untersucht werden. Die durchgeführten Untersuchungen zeigen, dass die Ultraschallfalle aufgrund der Wandlosigkeit sowie des Kontakts der levitierten Probe mit der Atmosphäre und der daraus resultierenden möglichen Aufkonzentrierung eine interessante Möglichkeit zur Probenhandhabung in der Mikrofluidik darstellt. Anhand zweier sonochemischer Reaktionen wurde mit Absorptionsspektroskopie gezeigt, dass das Ultraschallfeld des Levitators nur einen sehr geringen Einfluss auf die levitierte Probe ausübt. Mittels Absorptions- und Fluoreszenzspektroskopie wurde die durch die Verdampfung induzierte Aggregation eines Farbstoffes untersucht. Zudem wurde die relative Quantenausbeute des Farbstoffes im levitierten Tropfen bestimmt. Der Kontakt des levitierten Tropfens mit der umgebenden Atmosphäre kann auch zur Aufnahme von Substanzen aus der Atmosphäre führen. Dieser Effekt wurde anhand von levitierten ionischen Flüssigkeiten volumetrisch und schwingungsspektroskopisch bei verschiedenen Luftfeuchten untersucht und die Wechselwirkungen mit dem absorbierten Wasser detailliert charakterisiert. Die Kopplung und simultane Messung von Raman-Streuung und UV/Vis-Spektroskopie ermöglichte die Untersuchung der Bildung und Aggregation von Silber-Nanopartikeln und deren Auswirkungen auf die oberflächenverstärkte Raman-Streuung. Zudem wurde die Stabilisierung von Silber-Nanopartikeln in ionischen Flüssigkeiten und die Wechselwirkungen der Partikel mit den ionischen Flüssigkeiten untersucht. / Acoustic levitation in combination with optical spectroscopy allows for investigations of reactions and intermolecular interactions under specific conditions. Samples with microliter volumes can be handled without contact to solid surfaces resulting in the absence of impurities from surface contamination. Using different optical spectroscopy methods such as Raman, UV/vis, and fluorescence spectroscopy, different levitated samples were characterized in detail within this work. The investigations show that the acoustic levitator is an interesting tool for sample handling due to the wall-less fixture and because of the interaction of the levitated sample with the surrounding atmosphere. This interaction leads to an increase of a solved analyte upon the evaporation of the solvent. The ultrasonic field inside the trap was characterized by investigations of sonochemical reactions, which show a negligible influence of the ultrasonic field on the levitated sample. Absorption as well as fluorescence spectroscopy was used to study the aggregation of a dye due to an increasing concentration upon evaporation of the solvent. Furthermore, the relative quantum efficiency of the dye was determined from levitated droplet experiments. The interaction of the levitated droplet with the surrounding atmosphere can also lead to absorption of airborne substances. This effect was investigated for different levitated ionic liquids. Volumetric and vibrational studies were performed on levitated ionic liquids under different atmospheric humidity conditions to characterize the interaction of ionic liquids with water. Hyphenation of Raman scattering and UV/vis absorption spectroscopy enabled the investigation of the formation and aggregation of silver nanoparticles and the correlation of this information with the recorded surface-enhanced Raman spectra. In addition, the stability of silver nanoparticles in ionic liquids and the interactions of the particles with the ionic liquids were examined.

Optical characterization of potential drugs and drug delivery systems

This Thesis is a characterization study on substances having potency as drugs as well as on a lipid based drug-delivery matrix. The optical properties of newly synthesized molecules with proven pilicide properties have been characterized with several spectroscopic methods. These methods include optical absorption and fluorescence as well as time-resolved fluorescence. Upon covalently linking compounds with high quantum yields of fluorescence to specific parts of the pilicide, the biological impact was found to increase for some of the derivatives. Furthermore, by expanding the aromatic part of the pilicide molecule, a significant increase in the inherent fluorescence was obtained. The S0-S1 absorption band for these molecules was found to originate from an impure electronic transition, vibronically promoted by intensity borrowing from higher electronic states. Included in this Thesis is the measurement of how deeply some in this class of newly synthesized molecules become situated when placed inside ganglioside GM1 micelles, and how the molecules’ reorientation is affected. By means of radiation-less energy transfer, it was shown that the molecules place themselves close to the hydrophobic-hydrophilic interface inside the GM1 micelles. As a consequence they are exposed to a densely packed environment, which inhibits the free tumbling of the molecule. This restricted tumbling could be measured by means of time-resolved depolarization experiments. The release of drug-like fluorescent molecules is investigated from a lipid mixture, which upon equilibrium with water forms a mixture of inverted hexagonal and cubic phases. The lipid matrix displayed an extended release over the course of weeks, in vitro, for molecules having a large variation in hydrophobicity.

Les polyphénols sont abondamment trouvés dans de nombreux fruits, légumes, boissons etc. et ils possèdent de nombreux effets bénéfiques pour la santé. Les méthodes de calcul ont été utilisées dans le cadre de cette thèse pour rationaliser, décrire et prédire les propriétés physiques et chimiques des flavonolignanes et des pyranoanthocyanines pour la compréhension de leurs actions biologiques au niveau moléculaire. Tous les résultats des calculs théoriques ont été discutés par rapport aux données expérimentales. Les propriétés liées à l'activité antioxydante des flavonolignanes ont été étudiées par les méthodes de la théorie de la densité fonctionnelle (DFT). La dépendance au pH des propriétés d'absorption UV/Vis des flavonolignanes et des pyranoanthocyanines ont été évaluée par des méthodes DFT dépendante du temps (TD-), et les interactions non-covalentes ont été étudiées avec les méthodes de DFT incluant la correction de dispersion. Un bref aperçu est également donné sur l'interaction de ces composés avec des biomolécules. Le chapitre 6 présente des résultats non encore publiés de plusieurs systèmes non-covalents pigment: copigment. Cette partie des résultats constitue un bon point de départ pour la recherche du ‘meilleur copigment’. / Polyphenols are abundantly found in many fruit, vegetables, beverages, etc. and they possess many potential health benefits. Computational methods were thoroughly used through this thesis to rationalize, describe and predict physical chemical properties of flavonolignans and pyranoanthocyanins. Here, we aim at an understanding of polyphenol biological actions at a molecular level. All outcomes from the theoretical computations were discussed with respect to experimental data. The properties related to antioxidant activity of flavonolignans were investigated by density functional theory (DFT) methods. The pH dependence of ultraviolet/visible (UV/Vis) absorption properties of flavonolignans and pyranoanthocyanins were evaluated by time dependent (TD-) DFT methods, and noncovalent interactions were investigated within dispersion-corrected DFT methods. A short overview is also given on interaction of such compounds with biomolecules. Chapter 6 presents yet not published results of several noncovalent pigment: copigment systems. This part of the results serves as a good starting point to search for ‘the best copigment’.

Stanovení indikanu na uhlíkové pastové elektrodě a uhlíkové kompozitní elektrodě / Determination of indican on carbon paste electrode and on carbon composite electrode

This thesis is focused on the application of two working electrodes of graphite suitable for determination of oxidizable tumour biomarkers. It deals with the study of electrochemical behavior of urinary indican. For the determination the technique of differential pulse voltammetry (DPV) was used on carbon paste electrode and film composite paste electrode in Britton-Robinson (BR) buffer. In the first step optimal conditions were determined to find the lowest limit of determination (LQ) of this substance. BR buffer of pH 3 was chosen as the optimum medium for determination on a carbon paste electrode and BR buffer of pH 2 was used as the optimum medium for the determinaton using a film composite electrode. Performance for both electrodes was observed in a concentration range of from 1 · 10-6 to 5 · 10-5 mol·l-1 . The limit of determination for measurements on a carbon paste electrode was 0.7 µmol·l-1 and the film composite electrode was 1.7 µmol·l-1 . Possibility of the acummulation of indican was examined in order to increase sensitivity and decrease of limit of quantification. However, after 5 minute sorption, no increase in response was observed and the accumulation step was not inserted. L'antagonisme entre les besoins croissants d'énergie au niveau mondial et l'obligation de ralentir le réchauffement climatique fait partie des défis auquel l'humanité fait face. Dans le but d'assurer un confort thermique suffisant, les domaines de l'habitat, de l'automobile ou encore des bâtiments agricoles consomment de grandes quantités d'énergie. Pour réduire ces besoins, une des solutions proposées vise à améliorer l'isolation thermique de ces bâtiments grâce à l'utilisation de matériaux innovants. Un des objectifs majeurs est d'améliorer l'isolation des fenêtres qui représentent une partie importante des pertes énergétiques. Les recherches décrites dans cette thèse ont permis de développer des matériaux transparents aux rayonnements solaires visibles tout en étant des boucliers efficaces contre les rayonnements ultra-violets et proche-infrarouges. De tels matériaux sont le fruit de la combinaison entre des motifs à clusters de métaux de transition présentant des propriétés d'absorption recherchées et une matrice hôte permettant la mise en forme de ces matériaux (processabilité). La synthèse, ainsi que la méthodologie de modulation des propriétés d'absorption et d'intégration des clusters dans différentes matrices de type sol-gel ou polymères sont présentées. Un intérêt tout particulier est porté sur les relations qui s'établissent entre la structure des motifs à clusters et leurs propriétés d'absorption grâce aux comparaisons faites entre des études expérimentales et des études de chimie quantique utilisant la théorie de la fonctionnelle de la densité (DFT). In fine, l'augmentation du niveau de compréhension des relations structures-propriétés d'absorption des motifs à clusters étudiés permettra d'envisager l'étude de nouvelles compositions pour des matériaux encore plus efficaces à l'avenir pour le contrôle solaire. / The antagonism between growing global energy needs and the obligation to slow the global warming is one of the challenges humanity faces. In order to ensure sufficient thermal comfort, the housing, automotive or agricultural buildings sectors are major energy consumers. To reduce these needs, one of the proposed solutions aims to improve the thermal insulation of these buildings through the use of innovative materials. One of the major objectives is to improve the insulation of the windows which represent a significant part of the energy losses. The research described in this thesis has made it possible to develop transparent materials for visible solar radiation while being effective shields against ultraviolet and near-infrared radiation. Such materials are the result of the combination between transition metal cluster patterns having desired absorption properties and a host matrix for shaping these materials (processability). The syntheses as well as the methodology for modulating the absorption and integration properties of clusters in different sol-gel or polymer matrices are presented. Of particular interest is the relationship between the structure of cluster pattern and their absorption properties through comparisons between experimental studies and quantum chemistry studies using density functional theory (DFT). In fine, the increase in the level of understanding of the structure-absorption properties of the clusters studied will make it possible to consider the study of new compositions for even more efficient materials in the future for solar control.

3-(2-Benzylbenzoyl)-4(1H)-quinolinones : une nouvelle classe de composés photochromiques photoréversibles / 3-(2-Benzylbenzoyl)-4(1H)-quinolinones : a new class of photoreversible photochromic compounds

Actuellement, les photochromes photoréversibles présentent un intérêt important en vue de leurs applications éventuelles comme interrupteurs optiques ou pour le traitement et le stockage de données. L’objectif principal de ce travail de thèse a été d’évaluer les 3-­-(2-­-benzylbenzoyl)-­-4-­-quinolones en tant que système photochromique photoréversible. Afin d’étudier la relation entre la structure chimique et le comportement photochromique de ce système, une série de nouvelles 3-­-(2-­-benzylbenzoyl)-­-4-­-quinolones à substitution variée a été préparée à l’aide de procédures connues ainsi qu’élaborées lors de ce travail. La photoréaction de ces nouveaux molecules étant très complexe, une méthode d’étude a été établie avec des 2-­-benzyl-­-3-­-benzoyl-­-4-­-quinolones photoénolisables au comportement plus simple. Leurs spectres d’absorption modélisés à l’aide de fonctions pekariennes ont été comparés avec les résultats des calculs quantiques. Le mécanisme selon lequel la réaction de décoloration se fait par l’ionisation des photoénols fortement acides via protonation des molécules du solvant a été établi. Enfin, la troisième partie est consacrée à l’étude des nouveaux dérivés de quinolones. D’après l’analyse de leurs spectres d’absorption, ainsi que les résultats des calculs quantiques, nous proposons un mécanisme où le transfert d’hydrogène photo-­-induit conduirait à la formation d’un intermédiaire biradicalaire, capable de se cycliser en dibenzo[b]acrydinones hydroxysubstituées. Un tel mécanisme expliquerait la forte influence qu’ont la nature des substituants en positions 1 et 2, ainsi que la température et la présence d’oxygène sur la régio-­- et stéréo-­-sélectivité de cette photoréaction. / Photoreversible photochromic compounds are currently of considerable interest from the point of view of their potential applications as molecular switches and for data storage and processing. The main target of the present investigation is to evaluate the scope and limitations of 3-­-(2-­-benzylbenzoyl)-­-4-­-quinolones as a photoreversible photochromic system. In order to investigate the relationship between quinolone chemical structures and photochemical behavior, a large series of new 3-­-(2-­-benzylbenzoyl)-­-4-­-quinolones with different substituents has been prepared using known as well as newly elaborated synthetic procedures. In the second part of the work a series of simpler photoenolizable 2-­-benzyl-­-3-­-benzoyl quinolones is studied by the means of fitting their UV-­-Vis absorption spectra and comparison with the results of quantum mechanical calculations at the TD DFT level. It was concluded that the mechanism of the reverse reaction involves ionization of the strongly acidic photoenols via protonation of the solvent molecules. The third part of the work includes investigation of the new quinolone derivatives. From the analyses of their absorption spectra and the results of quantum mechanical calculations, we propose a tentative mechanism, according to which the photoinduced hydrogen transfer yields a biradical, capable of cyclization into the hydroxy substituted dibenzo[b]acrydinones. This mechanism accounts for the observation that the nature of substituents in positions 1 and 2, the temperature and the presence of oxygen strongly affect regio-­- and stereoselectivity of the cyclization.

B(C6F5)3-catalyzed reductions with hydrosilanes: scope and implications to the selective modification of poly(phenylsilane)

New complex silicon-containing molecules were made by B(C6F5)3-catalyzed hydrosilation, dehydrocoupling, and dealkylative coupling reactions starting from Si-H reagents. The scope of reactions starting from disilane was expanded to include the formation of silicon-sulfur1, silicon-oxygen and silicon-alkyl side-chains. Reaction inhibition was found with some heteroatom substrates, such as phenols and imines, that strongly bound to B(C6F5)3, and was consistent with the proposed mechanism (Chapter 2). B(C6F5)3 was found to be selective for Si-H activation in reactions of disilane and no competing Si-Si bond cleavage side-reactions were observed. This result will guide future studies and application of B(C6F5)3-catalyzed reactions with polysilanes. A different type of selectivity, the competing B(C6F5)3-catalyzed over-reduction, is evaluated and discussed in Chapter 3. This over-reduction reaction was classified into two distinct cases: alkyl groups for which over-reduction reaction was dependent on the steric bulk of the alkyl group and benzylic groups for which over-reduction was dependent on having an alpha-aryl group. These reactions are consistent with the proposed Piers-Oestreich mechanism (see Chapter 3) and suggest the rate-determining step for over-reduction is the nucleophilic attack of the alkoxysilane (R -O-SiR3) to the R3Si•••H•••B(C6F5)3 complex. Benzylic side-chains were over-reduced regardless of the steric bulk of the aryl groups. Literature precedents suggest that benzyl over-reductions must undergo an alternative mechanism to the Piers-Oestreich mechanism. A number of mechanisms have been proposed in the literature and in Chapter 3, suggesting conventional heteroatom substrate borane or silane-borane complexation. Furthermore, over-reduction of benzylic sulfur containing side-chains was found and this reaction was exploited in the B(C6F5)3-catalyzed synthesis of unique silicon-sulfur silicon-containing products. These over-reduction reactions highlighted the role of the silane for over-reduction and the challenges associated with the post-polymerization modification of poly(phenylsilane). The advances in B(C6F5)3-catalyzed synthesis of small silane molecules suggested reaction conditions and gave spectroscopic benchmarks that were applied to the post polymerization modification of poly(phenylsilane) (Chapter 4). New X-modified poly(phenylsilane) derivatives with thiolato (sulfur), alkoxy/aryloxy (oxygen), amido (nitrogen) and alkyl(carbon) side-chains were prepared with 10-40% incorporation of the ‘X’ group into poly(phenylsilane). These new polysilanes were characterized by the following methods: 1H/13C/29Si NMR, IR, MALS-GPC, EA, and UV-vis absorption spectroscopy. Together, these characterization methods showed that the polysilane had not undergone Si-Si cleavage and thus demonstrated the utility of B(C6F5)3 for the selective activation of Si-H bonds. Thermal decomposition of X-modified poly(phenylsilane) derivatives and parent poly(phenylsilane) showed interesting redistribution pathways (Chapter 5). The thermal decomposition products of poly(phenylsilane) were identified: volatile monosilanes, a structurally complex not-yet-identified phenylsilicon-containing material generated at 500 °C, and a mixture of silicon carbide (SiC) and elemental carbon generated at 800 °C. The B(C6F5)3-catalyzed post-polymerization method (Chapter 4) was evaluated based on the substitution percentage for X-functionalized poly(phenylsilane) derivatives. Reactions of highly electron-donating substrates gave a low amount of X incorporation (10%, e.g. aryloxy side-chains derived from phenol). Aryloxy groups were alternatively introduced via demethanative coupling, which gave a polymer with a greater substitution percentage (25%). The overall impact of the H-to-X substitution reactions was gauged by UV-vis absorption spectra and desirable UV absorption properties would require the modified poly(phenylsilane) to have a high degree of substitution. / Graduate / 2017-09-02

Pour notre travail de thèse, nous avons établi une méthodologie, basée sur la DFT et sa variante dépendante du temps, qui permet l'évaluation efficace et la prédiction rapide des couleurs de différentes familles de colorants carbonylés dans différents environnements. Ainsi, nous sommes capables de fournir des valeurs théoriques très précises pour les énergies de transition des dérivés de l'anthraquinone, de la coumarine et de l'indigo ainsi que du thioindigo. Notre stratégie prend en compte aussi bien les effets de solvatation, dans le cadre d'une modélisation en continuum, que les effets du pH, sur la position de la bande d'absorption maximale du spectre UV/VIS. / The project of the Ph. D. work consists in the elaboration of a theoretical methodology able to evaluate and predict the color of organic dyes (see the List of recent publications for more details). Our methodology takes into account the solvent effects as well as the pH impact on the color of the dyes. The theoretical tools used are the Density Functional Theory (DFT, for ground-state optimization) and the Time Dependent TDDFT for excitation spectra calculations. The solvent effects are taken into account via the PCM (Polarizable continuum Model).
