Effects of hyperoxia in alzheimers transgenic mice

An association between major surgery in the elderly and precipitation of Alzheimers disease (AD) has been reported. Hyperoxia (100%) oxygen is commonly administered after surgery to increase the oxygen content of blood. However, hyperoxia is a potent cerebral vasoconstrictor and generator of free radicals, as is [beta]amyloid (A[beta];). This study was aimed at examining behavioral, neuropathological, and neurochemical effects of hyperoxia treatments in APPsw transgenic mice (Tg+), which have elevated brain A[beta]; levels by 3-4 months of age but are not yet cognitively-impaired. At 3 months of age, Tg+ mice were pre-tested in the radial arm water maze (RAWM) task of working memory and found to be unimpaired. At 4.5 months of age, half of the Tg+ mice received the first of 3 equally-spaced hyperoxia sessions (3 hrs each) given over the ensuing 3 months. The other half of the Tg+ mice were exposed to compressed air during these 3 sessions. RAWM testing performed immediately following the final gas session at 7.5 months of age revealed significant working memory impairment in Tg+ mice exposed to hyperoxia. The Tg+ group that was exposed to placebo treatment showed a trend towards impairment, however, was not significantly different from the non-transgenic group. Hyperoxia-induced memory impairment in Tg+ mice did not involve changes in brain A[beta] deposition, degenerative cell numbers in hippocampus, neocortical lipid peroxidation, or hippocampal levels of APP, ApoE, COX-2, or GFAP. The combination of excess A[beta] and hyperoxia could have induced greater oxidative stress and cerebral vasoconstriction than either one alone, resulting in a pathologic cerebral hypoperfusion that triggered subsequent cognitive impairment.

The research to date, of transcutaneous electric nerve stimulation on cutaneous blood flow, is equivocal. The purpose of this report is to review the TENS body of knowledge, in particular synthesizing the literature on acupuncture stimulation of cutaneous blood flow with the two fold goal of creating a protocol to increase skin blood flow through the exogenous application of electrical stimulation, as well as creating an engaging engineering challenge for high school anatomy and physiology students. The hypothesis developed was TENS stimulation with electrode placement on specific acupuncture points would influence cutaneous blood flow as measured using laser Doppler flowmetry. The findings of this project did not support the hypothesis of TENS or Interferential electrical stimulation, in combination with acupuncture points or not, influencing skin blood flow. Perhaps this is due to the physiological differences between glabrous and non-glabrous skin and the different electrical resistances of each dermal layer, nerve stimulation, age and gender of subject or some combination thereof. These equivocal findings may also be the result of inconsistencies in testing protocols, such as subject preconditioning or not, subject’s position during administration of stimulation, electrode size and placement to name a few. Ultimately, this report provides a summary of the research to date, as well as outlining how this research could be adapted to supply engaging bio engineering challenges in the classroom including challenges to develop a model for delivering current to muscle; develop a model for skin blood flow management to name a few. / text

INGESTION OF ENDOPHYTE-INFECTED TALL FESCUE SEED INDUCES PERIPHERAL VASOCONSTRICTION BUT DOES NOT AFFECT CYCLICITY IN NON-PREGNANT MARES, AND A POPULATION OF BIOGENIC AMINE RECEPTORS RELATIVE TO VASOCONSTRICTION IS IDENTIFIED

Three experiments were conducted to explore the effect of fescue toxicosis on vasoconstriction and various parameters of the estrous cycle. In the experiment of Chapter 3, a 2x2 crossover experimental design with repeated measures was implemented to test whether the ingestion of endophyte-infected tall fescue seed would alter blood hormone concentrations of prolactin, progesterone, and estradiol, interovulatory intervals, and corpus luteum blood flow. Also, Doppler ultrasonography was used to assess whether palmar artery and palmar vein lumen diameter, area, circumference, and resistivity index could be altered by the ingestion of endophyte-infected tall fescue seed. Ingestion of endophyte-infected tall fescue seed significantly decreased palmar artery resistivity index, area, and circumference, and palmar artery and palmar vein lumen diameter, but did not alter interovulatory intervals, serum concentrations of prolactin, progesterone, estradiol, or corpus luteum blood flow. Experiments 1 and 2 from Chapter 4 profiled the dopaminergic, serotonergic, and α-adrenergic receptor types and subtypes, as expressed within the equine medial palmar artery, medial palmar vein, and uterine artery transcriptomes. Combined results from Chapters 3 & 4 imply that relative quantity of serotonergic receptor subtypes within a vessel may be related to with the severity of that vessel's vasoconstrictive response to endophyte-infected tall fescue.

INTERACTION OF ISOFLAVONES AND ENDOPHYTE-INFECTED TALL FESCUE SEED EXTRACT ON VASOACTIVITY OF BOVINE MESENTERIC VASCULATURE

Endophyte-infected tall fescue produces many ergot alkaloids, which have been shown to be vasoconstrictive in various vessel types of bovine. On the other hand, substantial evidence has been reported on the vasodilative effects of formononetin and biochanin A in different vessel types in humans and rats. So, a study was conducted using mesenteric vasculature collected from heifers shortly after slaughter. After 2-h incubation with formononetin (F), biochanin A (B), or an ergovaline-containing tall fescue seed extract (EXT) and their combinations, vessels were mounted in a multi-myograph to determine their ergotamine-induced contractility. Results indicated that F and B at 1 × 10-6 M and their combination did not impact the contractile response to ergotamine in mesenteric vasculature. The pre-myograph incubation of mesenteric vasculature with EXT altered the contractile response manner to ergotamine. However, at higher concentration, F and B may alleviate the reduction of vasoconstriction caused by prior exposure to EXT. To our knowledge, this study was the first to investigate the interaction of ergot alkaloids and isoflavones on in vitro bovine mesenteric vasculature. However, further investigations are necessary to understand the mechanism behind the interaction of ergot alkaloids and isoflavones on vasoactivity.

Benefits of Spontaneous Breathing : Compared with Mechanical Ventilation

When spontaneous breathing (SB) is allowed during mechanical ventilation (MV), atelectatic lung areas are recruited and oxygenation improves thereby. Whether unsupported SB at its natural pattern (without PEEP and at low pressure/small tidal volume) equally recruits and improves oxygenation, and if so by which mechanism, has not been studied. A porcine lung collapse model was designed to study this question. The cardiac output dependency of the pulmonary shunt was investigated with healthy lungs and with major shunt (during one-lung ventilation) and with SB, MV and continuous positive airway pressure (CPAP). The hypoxic pulmonary vasoconstriction (HPV) was blocked with sodium nitroprusside (SNP) to see whether HPV is the only mechanism available for ventilation/perfusion (VA/Q) matching during MV and SB. In all experiments, respiratory rate and tidal volume during MV were matched to SB. Oxygenation was assessed by serial blood gas measurements, recruitment by thoracic CTs; pulmonary shunt was assessed by multiple inert gas elimination or venous admixture. SB attained better oxygenation and lower pulmonary shunt compared with MV, although it did not recruit collapsed lung. Pulmonary shunt did not correlate with cardiac output during SB, whereas a correlation was found during MV and CPAP. With blocked HPV, pulmonary shunt was considerably lower during SB than MV. In conclusion, SB improves VA/Q matching as compared with MV, even when no recruitment occurs. In contrast to MV and CPAP, cardiac output has no major effect on pulmonary shunt during SB. The improved VA/Q matching during SB despite a blocked HPV might indicate the presence of a SB-specific mechanism that improves pulmonary blood flow redistribution towards ventilated lung regions independent of or supplementary to HPV.

Vasoconstrictor and Dilator Responses to Neurokinin a in Isolated Guinea Pig Heart

Effects of neurokinin A (NKA) and substance P (SP) on coronary resistance vessels were studied in isolated guinea pig hearts perfused with isotonic buffer containing 20 mM KCl. Injections of NKA and SP caused dose-dependent reductions in perfusion pressure with ED50 values of 14.0 and 0.326 pmol, respectively. Blockade of nitric oxide synthesis or removal of the endothelium inhibited vasodilator responses to neurokinins. Infusions of NKA or SP caused tachyphylaxis and cross-desensitization to the other neurokinin but not to acetylcholine. Injections of 2.5 nmol NKA increased perfusion pressure by 31 ± 8% when given after tachyphylaxis developed to infused SP (2.5 nmol/100 μl/min). It was concluded that 1) neurokinins cause an endothelium-dependent relaxation of coronary resistance vessels by stimulating NK-1 receptors on endothelial cells, and 2) desensitization of the receptor mediating vasodilation unmasks a vasoconstrictor response to NKA.

La thrombose est souvent liée à la survenue d’une rupture de la plaque d’athérosclérose qui expose le tissu sous endothélial thrombogène aux plaquettes sanguines circulantes. L’un des problèmes actuels est l’identification des mécanismes qui sont à l’origine de la fracture. Nous avons testé l’hypothèse que la vasoconstriction, induite par le LTC4 ou le thromboxane A2 (TXA2), joue un rôle dans la rupture de plaques d’athérosclérose et dans la formation de l’athérothrombose. Dans ce travail nous avons adapté la technique classique de la microscopie électronique pour observer les ruptures survenues sur une plaque d’athérosclérose. Avec cette méthode, nous avons montré qu’une vasoconstriction peut engendrer des ruptures de plaques, mais seulement lorsque ces plaques sont fragilisées (dites ‘vulnérables’) soit par un régime riche en graisse ou après exposition de la souris à un lipopolysaccharide extrait de membranes bactériennes. Nous avons montré aussi que la plaque produit du TXA2, et que l’absence de son récepteur chez les souris apoE-/- déficientes en TP prévient la rupture des plaques vulnérabilisées. La vasoconstriction constitue donc un élément déterminant dans la rupture des plaques vulnérables. Les résultats de ce travail identifient le TXA2 comme étant une éventuelle cible thérapeutique pour prévenir les ruptures des plaques. D’autre part, les analyses par microscopie électronique à balayage de la thrombose in vivo, et les analyses en microscopie électronique en transmission (MET) de l’ultrastructure des plaquettes agrégées in vitro, ont montré que les plaquettes activées fusionnent entre elle permettant ainsi la cicatrisation de l’endothélium lésé. / Rupture of atherosclerotic plaques exposes sub-endothelial thrombogenic material to circulating platelets, frequently leads to thrombus formation. At present, the mechanisms behind plaque ruptures are not well understood. We studied the hypothesis that vasoconstriction induced by LTC4 or thromboxane A2 (TXA2) plays an important role in plaque rupture and formation of atherothombosis. We used scanning electron microscopy to study rupture of the plaque in detail. We were able to show that vasoconstriction leads to plaque rupture, but only after plaques have been vulnerabilized by high fat diet, or by exposure of mice to lipopolysaccharide extracted from bacterial membranes. We could also show that plaques produce TXA2. As consequence the absence of TP, a receptor for TXA2 in apoE-/- mice prevented rupture of vulnerable plaques. Vasoconstriction is hence a determining factor in the rupture of vulnerable plaques. Our work has identified TXA2 as a potential therapeutic target in preventing plaque rupture. Moreover, analysis of the thrombosis in vivo by scanning electron microscopy, and the in vitro study of the ultra structure of aggregated platelets by transmission electron microscopy, showed that activated platelets fuse together to heal the injured endothelium.

EFFECT OF ERGOT ALKALOIDS ON BOVINE FOREGUT VASCULATURE, NUTRIENT ABSORPTION, AND EPITHELIAL BARRIER FUNCTION

Ergot alkaloids present in endophyte-infected (E+) tall fescue are thought to be the causative agent of fescue toxicosis, a syndrome affecting cattle in the eastern United States. Many of the observed signs of fescue toxicosis are thought to be attributed to peripheral vasoconstriction; however, there are data indicating that ergot alkaloids can alter blood flow to the gut. An experiment was conducted using right ruminal artery and vein collected from heifers shortly after slaughter. Vessels were mounted in a multi-myograph to determine the vasoconstrictive potentials of ergot alkaloids present in E+ tall fescue. Results indicated ergot alkaloids have the potential to induce vasoconstriction of foregut vasculature. A second experiment was conducted to determine if the additional ergot alkaloids present in E+ tall fescue increase the vasoconstrictive response above that of ergovaline. Results indicated that ergovaline is the main alkaloid responsible for vasoconstriction in bovine vessels. A third study was performed to determine the effect of ergot alkaloids on ruminal epithelial blood flow in the washed rumen of steers exposed to E+ or endophyte-free (E-) tall fescue seed. Steers were dosed with seed followed by a washed rumen experiment with differing levels of ergovaline incubated in the rumen. Results indicated that E+ tall fescue seed treatment reduced ruminal epithelial blood flow. Additionally, incubating ergovaline in the rumen during the washed rumen further decreased epithelial blood flow and volatile fatty acid (VFA) absorption. A final study was conducted to determine the acute effects of ergot alkaloids on isolated rumen epithelial absorptive and barrier functions and the potential for ruminal ergovaline absorption. Results indicate that acute exposure to ergot alkaloids does not alter the absorptive or barrier function of rumen epithelium and ergovaline is absorbed from the rumen. Data from this series of experiments have shown that ergot alkaloids from E+ tall fescue can induce vasoconstriction of blood vessels in the foregut of cattle, reduce blood flow to the rumen epithelium, and decrease VFA absorption. The decrease in nutrient absorption could contribute to the observed symptoms of fescue toxicosis, including depressed growth rates and general unthriftiness.
