Ferrugem asiática da soja: métodos de preservação dos urediniósporos e fatores relacionados à infecção do hospedeiro / Asian soybean rust: methods for uredospores preservation and factors related to host infection

Furtado, Gleiber Quintão 02 May 2007 (has links)
Soybean is the agricultural crop with the largest planted area in Brazil. The country stands out in the world scenario as the second-largest producer and exporter of this oilseed crop. The Asian rust (AR), caused by the fungus Phakopsora pachyrhizi, is one of the most serious phytosanitary problems of soybean in Brazil, especially because no cultivars exist so far with satisfactory resistance levels. In order to acquire better knowledge about the biology of Phakopsora pachyrhizi and some factors related to its infectious process in soybean, the following were evaluated in the present work: (1) Uredospore preservation methods; (2) Influence of leaf wetness discontinuity on the infectious process of uredospores; (3) Influence of luminosity and leaf surface on the infectious process of P. pachyrhizi uredospores; (4) Influence of phenological stage of soybean on infection by P. pachyrhizi . The results obtained showed that spore dehydration provided greater spore viability with time, in all storage conditions tested. Spores could be preserved for up to 231 days in the deep freezer, regardless of their hydration condition. At room temperature, non-dehydrated and dehydrated spores remained viable for 16 and 22 days, respectively. Spore dormancy reversion was effective when hydration was used (24h in humid chamber), followed or not by thermal shock (40°C/5 minutes). In all treatments where discontinuous leaf wetting was applied, AR severity was always lower when compared with continuous wetting, especially when wetting was interrupted after 4 hours of initial treatment in the humid chamber. Temporary wetness interruption affected spores that had already germinated, as these were not able to infect after a new wetness period. As to luminosity, uredospores were capable of infecting soybean plants both in the light and in the dark. However, higher severity was observed when the adaxial surface was inoculated, with later incubation of the plants in the dark. The in vitro experiments showed that there was greater germination and greater formation of appressoria in the absence of light. Epicuticular wax content and its ultrastructural aspect did not show differences between the adaxial and abaxial surfaces in cultivar BRS 154. At the R5 reproductive phenological stage, cultivars BRS 154 and BRS 258 showed smaller severity in relation to stages V3 and R1. The AR mean latent period (MLP) was 8 and 9 days for cultivars BRS 154 and BRS 258, respectively. MLP differences due to phenological stage were not detected in any of the cultivars. As to the influence of leaf age on AR susceptibility, leaf 1 showed greater susceptibility and frequency of infection by P. pachyrhizi, considering the plant base-apex direction. The results herein presented are essential, either because they make information available on the biology and epidemiology of the fungus, or because they serve as a foundation for new studies on the most diverse areas about this important pathosystem.

Identificação de espécies de Botryosphaeriaceae e caracterização do monociclo da podridão apical da goiaba / Identification of Botryosphaeriaceae species and characterization of monocycle of stylar-end rot of guava

Nogueira Júnior, Antonio Fernandes 30 January 2013 (has links)
Nos experimentos in vivo goiabas cv. / Increased incidence of stylar-end rot in guava fruits have been observed in the São Paulo State - Brazil. However, little is known about its etiology and the influence of environment variables on the monocycle of this disease. This study aimed to identify Botryosphaeriaceae species that cause stylar-end rot in guava and to analyze the favorable environment conditions, in vitro and in vivo, for the monocycle development of this pathogen. We used 56 monosporic isolates from diseased fruit from guava producing regions from the São Paulo State - Brazil. The morphological and phylogenetic analyses performed with sequence data of the ITS region and β- tubulin allowed to identify the species Fusicoccum aesculi, Neofusicoccum parvum and Neofusicoccum ribis, as causal agents of the disease. The mycelial growth was evaluated at 10, 15, 20, 25, 30, 35 and 40 °C. Conidial germination was evaluated at 10, 15, 20, 25, 30, 35 and 40 °C with wetness periods of 4, 6, 12, 24 and 48 hours. In the in vivo experiments, guavas cv. Kumagai were inoculated with conidial suspensions of the three species identified by artificial wounds. The fruits were kept at 15, 20, 25, 30 and 35 °C with wetness periods of 6, 12, 24 and 48 hours after inoculation. We evaluated the incubation period, the lesion diameter on the seventh day after inoculation and the lesion growth rate. The optimum temperature for mycelial growth of N. ribis was 28.2 °C and there was no difference between the optimal temperatures (approximately 31 ºC) for N. parvum and F. aesculi. The optimal temperature for conidial germination was 30 ºC for the three species. There was an increase in germination with the increase of the wetness period. Within the optimal temperature range, values were higher 70% of germination after a wetness period of 6 hours. F. aesculi reached high germination values at 40 °C in wetness periods exceeding 12 hours. The optimal temperature for lesion growth rate was approximately 30 °C for the three pathogens and the wetness period of 48 hours caused higher disease severity as compared with the wetness period of 6 hours. The shortest incubation periods were obtained at 30 ºC with wetness periods of 48 hours. There was no difference between lesion growth rate for the three species and the average rate for three pathogens was 0.4/day. The best conditions for the disease development are high temperatures and prolonged periods of moisture. This is the first report of Neofusicoccum parvum and Neofusicoccum ribis associated with stylarend rot of guava fruits in Brazil.

Componentes monocíclicos e quantificação de danos no patossistema Corynespora cassiicola - soja / Monocyclic components and damage quantification of Corynespora cassiicola - soybean pathosystem

Mesquini, Renata Moreschi 28 January 2013 (has links)
Soybean is the most important grain crop in Brazil. The production of 2010/11 occupied an area of 24 million hectares, becoming the most important oilseed produced and consumed worldwide. Some factors, such as diseases, may limit soybean production. The target spot caused by Corynespora cassiicola is frequent in all soybean producing regions in Brazil and in the past years, an increasing loss has been assigned to it. The aims of the present study were to determine the ideal temperature range for mycelial growth of different isolates of C. cassiicola; verify the sporulation in relation to the culture medium; evaluate the conidia germination under different periods of leaf wetness; study the effect of the temperature and the leaf wetness on the monocyclic components and carry out studies of crop loss in the field. In order to evaluate the effect of the temperature on the mycelial growth, different isolates were cultivated at the temperatures of 5°C, 10°C, 15°C, 20°C, 25°C, 30°C and 35°C. The minimum and maximum estimated temperature by the beta function to provide C.cassiicola mycelial growth was 6.9 °C and 33 °C, respectively. The sporulation was analyzed in PDA and Czapek culture media. The results obtained were compared through the Scott-Knott test (P <0.05). The conidia production varied according to each isolate on both PDA and Czapek medias. The germination test was carried out in soybean leaves using different leaf wetness periods (0, 12, 24, 48 and 72 hours). The germinated conidia measurement was done in a scanning electron microscope. The conidia germination on the leaves surface was observed from 6 hours. The temperatures used to study the monocyclic components were 10 º C, 15 º C, 20 º C, 25 º C and 30 º C and the leaf wetness periods were 12, 24, 48 and 72 hours. The variables quantified were severity, number and diameter of lesions. The data were adjusted to the generalized beta and monomolecular functions. The minimum, maximum and optimum temperature able to promote the development of target spot in soybean leaves were 18 ° C, 32 ° C and 27 ° C, respectively. The 48- hour leaf wetness resulted in greater development of the disease. For damage quantification, commercial cultivars were sown in two sowing times. The components evaluated in the field were: severity, leaf area, normalized difference vegetation index (NDVI) and grain yield (kg ha-1). Using these data, it was calculated the area under disease progress curve (AUDPC), area under the leaf area index progress curve (AULIAPC), leaf area index (LAI), healthy leaf area index (HLAI), healthy area duration (HAD) and healthy area absorption area (HAA). The severity values up to 37% in the lower canopy do not cause crop loss in C.cassicola - soybean pathosystem .The variables HLAI and AULAIPC correlated with grain yield and may be useful for future studies of crop loss for this pathosystem.

Componentes monocíclicos do cancro cítrico no sistema Xanthomonas axonopodis pv. citri-limão Tahiti- Phyllocnistis citrella, sob condições controladas. / Monocyclic components of the asiatic citrus canker in the system: Xanthomonas axonopodis pv. citri-tahiti lime- Phyllocnistis citrella, under controlled conditions.

Christiano, Rock Seille Carlos 29 January 2004 (has links)
The Asiatic Citrus Canker (ACC), caused by Xanthomonas axonopodis pv. citri ( Xac) is one of the most serious problems of citrus in São Paulo State, Brazil. After the introduction of Citrus Leafminer (CLM), Phyllocnistis citrella, the behavior of ACC changed drastically, becoming more severe. To understand the interaction of Xac and CLM on Tahiti Lime, it were, under controlled conditions, carried three experiments: (A) influence of the way of Xac penetration (stomata, mechanical wound and CLM injury of the stage: egg, 1st instar, 3rd instar and pupa) and of the inoculum concentrations of 10 1 , 10 2 , 10 4 and 10 6 CFU/mL in the Xac infection; (B) effect of the age of mechanical wound, of CLM injury and of unwounded leaf in the progress of the ACC; (C) influence of the temperature and of the period of leaf wetness in the development of the ACC. In the experiment A, the minimum inoculum concentration to cause ACC by penetration of Xac through stomata was of 10 4 CFU/mL; 10 2 CFU /mL by mechanical wound and CLM injury of egg stage and 1st instar; and 10 1 CFU /mL by injury of 3rd instar and pupa stage. The injury of 3rd instar and pupa stage have generated higher severity in relation to other ways of penetration on all concentrations (about 6 times more than the severity in unwounded leaf on 10 6 CFU /mL). The severities in leaf with mechanical wound and with injury of egg stage and 1st instar have not differed from the unwounded leaf. In the experiment B, the mechanical wound between 0 and 6 days (0-6 days of age), the CLM injury between 0 and 32 days after eggs laying of P. citrella (0-32 days of age) and unwounded leaf between 5 and 35 days (5-35 days of age) have shown susceptibility to Xac. The severity decreased linearly with increasing age of the wound and age of unwounded leaf. In relation to age of injury, the severity varied in function of generalized Beta model and the inflection point was 18 days. The maximum severity in leaf with injury was 10 times bigger than in unwounded leaf. In the experiment C, the optimum conditions appearance of lesions and 100% of incidence were 25-35ºC under minimum wetness duration of 4 hours. The Beta-monomolecular function provided a good description of severity variation in relation to the temperature and duration of leaf wetness. The estimated minimum and maximum temperatures were 14.9 and 42ºC. The estimated maximum severity was determined under wetness of 24 hours and 31.7ºC. In general, the LMC injury (up to 3rd instar stage) has influenced in the Xac infection, it has increased the efficiency of bacterium penetration still on low inoculum concentration and it has allowed Xac infection for long period (32 days). In leaf with CLM injury, the severity can be 10 times bigger than in unwounded leaf. The Xac infection is favored under 25-35ºC and the Xac does not demand long periods of leaf wetness (more than 4 hours).

Plants go with the flow : predicting spatial distribution of plant species in the boreal forest

Zinko, Ursula January 2004 (has links)
<p>The main objectives of this thesis are to study if a topographic wetness index (TWI) could be used as a tool for predicting the spatial distribution of vascular plant species richness in the boreal forest as well as to study congruence in species richness between vascular plants, liverworts, mosses and lichens. A wetness index ln(a/tanβ) based on topography was used to assign a specific TWI-value to every 20 x 20m grid in two 25 km2 boreal forest landscapes (differing in average soil pH) in northern Sweden. Soil pH is known to be influenced by groundwater and to affect plant species richness in other biomes. Therefore, the relationships between plant species richness, TWI and soil pH were also studied. </p><p>The results showed that the majority of the investigated boreal forest landscapes were relatively dry and species-poor, whereas interspersed patches linked to areas with relatively high TWI had species-rich vegetation including the species of the drier parts of the landscape. There was a positive relationship between species richness of vascular plants and the TWI in both landscapes, but varied with average soil pH. TWI explained 30 % and 52 % of the variation in plant species richness in the landscape with lower and higher pH, respectively. The proportion of regionally uncommon plants also increased with TWI. Testing different calculation methods of the TWI resulted in a large variation in correlation strengths between the various TWI-values and different measured variables (species richness of vascular plants, soil pH, groundwater flow and soil moisture). The relationship between plant species richness and TWI could be further improved with some of the calculation methods. </p><p>When studying correlations in species richness using data sets from boreal forest in northern Sweden, strong positive correlations among vascular plants, mosses and liverworts were found, but no significant correlation between macrolichens and any of the other groups. This result could be explained by that the species number of each of the three related groups increases with ambient moisture, whereas the species number of macrolichens is weakly associated with moisture. </p><p>In conclusion, the TWI could become a useful tool in conservation management for identifying areas of special interest prior to field inventories.

[en] Summary - Weather-Radar Rainfall Measurement and Simulated Surface Wetness Duration for Septoria Leaf Blotch Risk Assessment. The persistence of free moisture on leaves, mainly as a result of precipitation in the form of rainfall or dew, plays a major role during the process of plant infection by most fungal pathogens. Acquiring rainfall and leaf moisture information is needed for accurate and reliable disease prediction and management. The objective of this research is to contribute to improve forecasting Septoria leaf blotch and other fungal pathogens on winter wheat in Belgium and Luxembourg./In the first part of this work, the potential of weather-radar rainfall estimates for plant disease forecasting is discussed. At first step, we focused on assessing the accuracy and limitations of radar-derived precipitation estimates, compared with rain-gauge data. In a second step, the Septoria leaf blotch prediction model PROCULTURE was used to assess the impact on the simulated infection rate of using, as input data, rainfall estimated by radar instead of rain gauge measurements. When comparing infection events simulated by PROCULTURE using radar-derived estimates and reference rain gauge measurements, the probability of detection (POD) of infection events was high (0.83 on average), and the false alarm ratio (FAR) of infection events was not negligible (0.24 on average). FAR decreased to 0 and POD increased (0.85 on average) for most stations, when the model outputs for both datasets were compared against visual observations of Septoria leaf blotch symptoms. Analysis of 148 infection events observed over three years at four locations showed no significant difference in the number of simulated infection events using either radar assessments or gauge measurements. This suggests that, for a given location, radar estimates are just as reliable for predicting infection events as rain gauges. As radar is able to estimate rainfall occurrence over a continuous space, unlike weather station networks that do observations at only a limited number of points, it has the great advantage of being able to predict the risk of infection at each point within an area of interest with an accuracy equivalent to rain gauge observations. This gives radar an important advantage that could significantly improve existing warning systems. In the second part, a physical model based on the energy balance, known as the Surface Wetness Energy Balance (SWEB), was applied for the simulation of Surface Wetness Duration (SWD) on winter wheat canopy. The model, developed in the United States on grapes canopies, was adapted for the winter wheat cultivars and was applied for use with agrometeorological data easily available from standard weather stations and weather-radar rainfall estimates. The SWEB model simulates surface wetness duration for both dew and rain events. The model was validated with data measured by sensors and with visual observations of SWD conducted in experimental plots during two cropping seasons in 2006 and 2007. The wetness was observed visually by assessing the presence or absence of surface water on leaves. Based on the results, the SWEB model appeared to underestimate surface wetness duration and especially for the dry-off events when compared statistically to visual observations. The error, on average, is generally less than 90 minutes. In order to establish a relationship between the surface wetness periods and Septoria leaf blotch development risk on the top three leaves, the SWEB model SWD outputs were compared with the number of hours of high probability of infection simulated by PROCULTURE as well as with visual plant diseases observations. A minimal surface wetness duration of favourable infection conditions for Septoria tritici was established. It is now required to develop an operational system that would integrate weather radar, surface wetness duration and foliar epidemic model. In this work, we have analyzed the advantages and limitations of the radar system as input to models and its ability for spatial interpolation of rainfall. We also tested the model for the determination of surface wetness periods required for Septoria Leaf Blotch Risk development. The proposed approach could be integrated in the existing system. Finally this approach shows once more the "happy marriage" between agrometeorology and plant disease management.

Plants go with the flow : predicting spatial distribution of plant species in the boreal forest

Zinko, Ursula January 2004 (has links)
The main objectives of this thesis are to study if a topographic wetness index (TWI) could be used as a tool for predicting the spatial distribution of vascular plant species richness in the boreal forest as well as to study congruence in species richness between vascular plants, liverworts, mosses and lichens. The results showed that the majority of the investigated boreal forest landscapes were relatively dry and species-poor, whereas interspersed patches linked to areas with relatively high TWI had species-rich vegetation including the species of the drier parts of the landscape. When studying correlations in species richness using data sets from boreal forest in northern Sweden, strong positive correlations among vascular plants, mosses and liverworts were found, but no significant correlation between macrolichens and any of the other groups. This result could be explained by that the species number of each of the three related groups increases with ambient moisture, whereas the species number of macrolichens is weakly associated with moisture. In conclusion, the TWI could become a useful tool in conservation management for identifying areas of special interest prior to field inventories. Since vascular plants can be used as an indicator taxon for species richness of mosses and liverworts, high TWI-values indicate areas of species hotspots of these taxa.

Preparation and characterization of vanadium oxides on carbon fiber paper as electrodes for pseudocapacitors

Cromer, Cynthia Eckles 10 April 2013 (has links)
Supercapacitors are important electrochemical energy storage devices for microelectronic and telecommunication systems, electric cars, and smart grids. However, the energy densities of existing supercapacitors are still inadequate for many applications. Vanadium oxides have been studied as viable supercapacitor alternatives, with varying results. Methods are often complicated or time-consuming, and electrode fabrication often includes carbon powder and binder. The objective of this work was to study the effect of processing conditions on specific capacitance of supercapacitors based on vanadium oxides coated on carbon fiber papers. This study was conducted to form easily-fabricated compounds of vanadium oxides which could offer promise as pseudocapacitor material, and to nucleate these compounds directly onto inexpensive carbon fiber without binder. The incipient wetness impregnation technique was used to fabricate the electrodes. Electrochemical performance of the resulting electrodes was tested in a Swagelok-type electrochemical two-electrode cell, and the electrodes were characterized by XRD and SEM. Interesting nanofeatures were formed and the vanadium oxides exhibited pseudocapacitance at a respectable level.

A GIS-based landscape analysis of dissolved organic carbon in boreal headwater streams

Andersson, Jan-Olov January 2009 (has links)
In boreal catchments, stream water chemistry is influenced and controlled by several landscape factors. The influence of spatially distributed variables is in turn dependent on the hydrological scale. Headwater streams have larger variability of water chemistry, and thus together represent a large biodiversity, and therefore need to be monitored in official environmental assessments. One objective of this study was, using Geographical Information Systems (GIS), to analyse co-variation between landscape variables and water chemistry and to determine which of the landscape variables have a major influence on the concentration of dissolved organic carbon (DOC) in headwater streams. Another objective was to find a simple method for predicting sources of DOC, using official map data and publically available GIS applications. Totally 85 headwater catchments (0.1-4 km2) in the county of Värmland, western south Sweden, were used in the study. Water chemistry was analysed for water sampled at low, medium and high flows, and landscape variables were extracted from official map data sources: topographic maps, a digital elevation model (DEM, 50 m grid), and vegetation data. Statistical analyses showed that topography (mean slope and mean topographic wetness index (TWI)) and wetland cover often correlated well with DOC in headwater catchments. Official map data could satisfactorily extract landscape variables (mean slope, mean TWI) that were useful in predicting stream water chemistry (DOC). A high-resolution elevation model, which was generated by interpolation of photogrammetric data, was used to calculate and evaluate two different wetness indices and their ability to predict the occurrence of wetlands in six catchments of different sizes and topography. The SAGA (System for Automated Geoscientific Analyses) wetness index (SWI) gave substantially better results than the TWI. The effects of resolution of DEMs on calculations of the SWI were investigated using 5, 10, 25 and 50 m grids. The results showed that SWI values increased with increasing cell size. The near linear increment of mean values for resolutions 10-50 m suggests a independence of terrain type and catchment size, which supported previous findings that indicated that mean slope and mean wetness index calculated from coarse elevation models may be used for prediction of DOC in headwater streams.

Aplicação de componentes principais e regressões logísticas múltiplas em sistema de informações geográficas para a predição e o mapeamento digital de solos / Application of principal components and multiple logistic regression in a geographical information system for prediction and digital soil mapping

Caten, Alexandre Ten 31 October 2008 (has links)
Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / Social demands on soil information have grown dramatically, meanwhile the soil surveys are seldom carried out in the country. Digital soil mapping techniques can be applied to infer the spatial distribution of soil from existing soil maps or from reference areas, extrapolating this information to areas not mapped. The purpose of this study was to apply in a Geographic Information System the Multiple Logistic Regressions (MLR) using Principal Components (PC) as explanatory variables to predict soil classes spatial distribution. The study area was the region of municipality São Pedro do Sul / RS. For the development of predictive models a set of nine terrain attributes were used. Model training was executed on an existing soil map and with a survey carried out in a reference area, both in a 1:50.000 scale. The first three retained PC explained 65.57% of the data variability. The predictive models which used PC had lower values of kappa index. The most accurate predicted map reached a kappa value of 63.20% and was generated by using the nine attributes of land as predictive covariates. The mapping accuracy is sensitive to similarities between the mapped classes, and mapping in a more homogeneous categorical level reduces the accuracy of the predicted maps. Soil classes relatively not representative in the training maps are not properly spatialized. The use of MLR allows spatializing of soil classes to areas not mapped, although the use of PC needs to be tested with a larger number of covariates.
