Engineering strategies for ABD-derived affinity proteins for therapeutic and diagnostic applications

Small stable protein domains are attractive scaffolds for engineering affinity proteins due to their high tolerance to mutagenesis without loosing structural integrity. The albuminbinding domain is a 5 kDa three-helix bundle derived from the bacterial receptor Protein G with low-nanomolar affinity to albumin. In this thesis, the albumin-binding domain is explored as a scaffold for engineering novel affinity proteins with the possible benefit of combining a prolonged serum half-life with specific targeting in a single small scaffold protein. Previously, a library was created by randomizing surface-exposed residues in order to engineer affinity to a new target antigen in addition to the inherent albumin affinity. Here, phage display selections were separately performed against the tumor antigens ERBB2 and ERBB3. The ERBB3 selection resulted in a panel of candidates that were found to have varying affinities to ERBB3 in the nanomolar range, while still retaining a high affinity to albumin. Further characterization concluded that the clones also competed for binding to ERBB3 with the natural activating ligand Heregulin. The selections against ERBB2 resulted in sub-nanomolar affinities to ERBB2 where the binding site was found to overlap with the antibody Trastuzumab. The binding sites on ABD to albumin and either target were found in both selections to be mutually exclusive, as increased concentrations of albumin reduced the level of binding to ERBB2 or ERBB3. An affinity-matured ERBB2 binder, denoted ADAPT6, which lacked affinity to albumin was evaluated as a radionuclide-labeled imaging tracer for diagnosing ERBB2-positive tumors. Biodistribution studies in mice showed a high renal uptake consistent with affinity proteins in the same size range and the injected ADAPT quickly localized to the implanted tumor. High contrast images could be generated and ERBB2-expressing tissue could be distinguished from normal tissue with high contrast, demonstrating the feasibility of the scaffold for use as diagnostic tool. In a fourth study, affinity maturation strategies using staphylococcal cell-surface display were evaluated by comparing two replicate selections and varying the stringency. A sub-nanomolar target concentration was concluded to be inappropriate for equilibrium selection as the resulting output was highly variable between replicates. In contrast, equilibrium sorting at higher concentrations followed by kinetic-focused off-rate selection resulted in high output overlap between attempts and a clear correlation between affinity and enrichment. / <p>QC 20160510</p>

Although a prolific writer of history, Sakhawi is, primarily, a traditionist. As such, accuracy both in utterance and writing would, by the very nature of his training, be his first objective. Modern writers appear to have neglected the importance of his contribution to the understanding of the history of his century. accept for a few articles, comparatively little has been written. It is, therefore, strange that such a mine of information as Sakhawi's writing presents has remained so long in oblivion. In this thesis an attempt has been made to evaluate that contribution together with an edition of part of his work. The study has been divided into three sections, the first dealing with Sakhawi’s life and times. This part of the study is based largely on his autobiography which was written but a few months before he died. During research no reference was discovered to this most informative work. The section falls into three chapters, the first of which endeavours to show the political and educational aspects of Cairo during the early part of Sakhawi's lifetime. Cairo was his native city and, as such, made great impact on his early life. In the second chapter the position of his family, his Shaykhs, the academic journeys he made, his residence in Hijaz and the last phase of his life are portrayed. The third chapter deals with his activities as an adult, his reputation as a traditionist together with a survey of his works as presented in his autobiography. In the second part, the study deals exclusively with Sakhawi as a historian of the 9th/15th century. This part also is divided into two chapters, the first of which considers the following aspects: - I Sakawi's works on the century; II His motives, methods and literary style and III His treatment of the history of the century. The second chapter collates Sakhawi's methods of selecting his information and the painstaking efforts he made to verify them, together with his historical achievements, while the last two topics endeavour to evaluate his task as a historian in that century. Section three presents the hitherto unedited part of Wajiz al-Kalam... which deals with the history of the 9th/15th century. This section also falls into the three divisions of preface, text and annotations. The last divides again into two groups one of which deals with the textual variants mentioned in the footnotes and the other attempts to deal with the interpretation of most of the idiom, colloquial expressions and the names of places and personalities mentioned in the supplement to the text.

Depuis 1999, le Centre de recherches archéologiques de l'Université libre de Bruxelles a entrepris l'étude de la tombe thébaine numéro 29 à Cheikh Abd el-Gourna, construite pour le vizir et maire de la ville Aménémopé sous le règne d'Amenhotep II (vers 1425-1401 av. J.-C.). La fouille du monument, sous la direction du Professeur Roland Tefnin, a été menée suivant deux axes de recherches. Le premier, dans une perspective synchronique, visait à étudier les aménagements et le fonctionnement d'origine de la tombe, sous la 18e dynastie. Le second, dans une perspective diachronique, visait à reconstituer l'histoire du monument et de ses occupations successives jusqu'à l'intervention de la mission. La thèse porte sur les résultats de cette seconde approche, et plus particulièrement sur l'analyse des occupations de l'antiquité tardive et post-antiques. Celles-ci se distinguent par une affectation de l'espace différente de sa fonction d'origine : la tombe n'est plus utilisée comme lieu de sépulture et de culte funéraire mais comme habitation. La fouille a ainsi livré les vestiges d'une installation érémitique datée des VIIe et VIIIe siècles de notre ère, associés à un matériel archéologique et épigraphique particulièrement abondant, ainsi que les ruines d'une maison villageoise construite durant la seconde moitié du XIXe siècle.

Para tanto, foi construído um arcabouço teórico que articulou os conceitos de cultura, identidade cultural, midiatização e documentário. A pesquisa empírica, de cunho qualitativo, foi realizada a partir da observação de um corpus de cinco produções audiovisuais da Associação Brasileira de Documentaristas - PI (ABD-PI), e de entrevistas com produtores e participantes das produções. Os resultados da pesquisa permitem ver que as construções são marcadas pelo recorte dado pelos produtores audiovisuais e também a partir das informações obtidas com os sujeitos/ grupos sociais envolvidos no contexto das produções. As construções realizadas nos documentários permitem a expressão de sujeitos e de práticas culturais do contexto piauiense, particularmente de classes populares, apresentando possibilidades concretas de colaboração para a cidadania comunicativa destas culturas e identidades. / Considering the importance of cultures in the constitution of societies and communicative citizenship as a key element for raising and cultural recognition, this research investigated the construction of cultural identities from Piauí in audiovisual productions of the Brazilian Association of Documentary-Piauí and aiming to understand how they relate to the cultural communicative constitution of citizenship of these identities. Therefore, it was built a theoretical framework that articulated the concepts of culture, cultural identity, and documentary media coverage. The empirical research, a qualitative approach, was performed through the observation of a corpus of five audiovisual productions of the Brazilian Association of Documentary - PI (ABD-PI), and interviews with producers and productions participants. The survey results allow us to realize that those constructions are marked by clipping given by audiovisual producers and also from the information obtained with the subjects/social groups involved in the production context. The buildings made in documentaries allows the expression of subjects and cultural practices in Piauí context, particularly the popular classes, presenting concrete possibilities of collaboration for the citizens of these communicative cultures and identities.

Eight crossbreed mature horses were used with body weight (BW) average of 364kg, geldings, adapted to diet composed by grass hay and concentrate, in a 60:40 proportion. A complete randomized design was used with horses allocated in three treatments. Treatment I: (Control) (n=2) horses were slaughtered without overload carbohydrate; Treatment II (n=3) and III (n=3), horses subjected to overload carbohydrate, with gastric infusion of 17.6 g starch/kg BW, and slaughtered after 24 and 36 hours, respectively. Horses were subjected to clinical, hematological and fecal evaluations before the overload and 2, 4, 8, 12, 16, 20, 24, 28, 32 and 36 hours after overload. Four hours after overload horses became depressed and keeping until the end of evaluation, and one horse presented lameness 36 hours after overload. Any difference (P>0.05) were observed in heart rate, respiratory rate, body temperature and hoof temperature. Increase in packed cell volume and plasma protein concentration were observed 24 hours after overload, varying from 26.7 to 32.0% and 7.1 to 8.1 g/dL, respectively (P<0.05). Differences were observed (P<0.05) in plasmatic lactate concentration in zero, 20 and 28 hours after overload, with values of 0.7, 1.04 and 1.22 mmol/L, respectively. Plasma endotoxin concentration didn't cross 0.1000 EU/mL and may be not present. There were any difference (P>0.05) in fecal and digesta water content, however, fecal pH reduced along 36 hours post-overload (P <0.01), varying from 6.09 to 4.46. Content of large intestine in horses subjected to overload presented whitish-green color, milk aspect, with gas bubbles and acid odor. There weren t difference (P>0.05) in water content of feces and digesta, however, fecal pH reduced along 36 hours post-overload (P <0.01), varying from 6.09 to 4.46. Buffering capacity of ceco-colon digesta and feces were reduced in horses subjected to overload. Right dorsal colon, transverse colon and descendent colon were segments, except stomach, that presented lower pH values, varying from 4.49 to 4.56. Eosinophils infiltration were presented in mucosa and submucosa of all horses, however, only horses submitted to overload presented neutrophils and eosinophils leucocitoestase with neutrophils predominance restricted to large intestine. Tract gastrointestinal circulatory alterations observed were congestion, edema and lymphatic vessels dilatation, more evident in submucosa, with larger inflammatory cells infiltration in horses subjected to overload. Intestinal mucosa 36 hours after overload presented larger degree of imunorreactivity anti-myeloperoxidase, followed by horses evaluated at 24 hours after overload and control horses, varying from 2.7 to 4.0, 1.0 to 3.7 and 1.0 to 2.5, respectively. Overload carbohydrate in horses promoted intensive fermentation in ceco-colon, predisposing clinical disturbances, digesta alterations and mucosa and submucosa lesions at gastrointestinal tract of horses with light to moderate degree,36 hours after overload. / Este trabalho teve como objetivo avaliar as altera??es sist?micas, da mucosa e conte?do do trato gastrointestinal de eq?inos submetidos ? sobrecarga diet?tica com amido. Foram utilizados oito eq?inos adultos castrados, com peso vivo m?dio de 364 kg, adaptados a dieta composta por feno de Coastcross e concentrado, na propor??o de 60:40. Foi utilizado um delineamento experimental com tr?s tratamentos: Tratamento I (Controle) (n=2), eutan?sia dos animais sem sobrecarga com amido; Tratamentos II (n=3) e III (n=3), animais submetidos ? sobrecarga com amido, com infus?o g?strica de 17,6 g amido/kg de peso corporal e eutan?sia ap?s 24 e 36 horas, respectivamente. Nature and death in the poetry of al-Malā'ika, al-Shābbī and Shukrī, and certain English Romantic poets : a comparative study

The first part of this thesis, divided into two chapters, deals with the early background of European Romanticism; the reasons behind its appearance and problems of definition. There follows a discussion on the question of the originality of Arabic Romanticism, with ,a brief review of the roots and main literary groups of this movement in Arabic poetry. Part two examines the influence of English poetry and thought on three Arab Romantic poets: Nāzik Sādiq al-Malā'ika, Abū al-Qāsim al-Shābbī and Abd aI-Rahmān Shukrī. This is discussed parallel with the channels of this influence. The main focus of this research is however, to show the ways in which al-Malā'ika, al-Shābbī and Shukrī perceived and reflected nature and death in their poetry. Their attitudes towards certain phenomena in nature such as the countryside, night, the sea, childhood and moral and social lessons of nature are compared with certain attitudes of Wordsworth, Coleridge, Keats and Shelley. Themes such as life and death, fear of death, fatalism, immortality and death as a welcome experience are also the concern of this thesis, with a comparison of these themes in the poetry of the Arab and English Romantic poets. However, owing to the popularity of Keats and Shelley with the three Arab Romantic poets, this thesis concentrates on their poetry. This research has selected only certain phenomena and themes from nature--and death because of the dominance of these subjects in the poetry of al-Malā'ika, al-Shābbī and Shukrī. The translations of Arabic poetry in this thesis are intended to convey the general sense of the source texts, rather than to give a precise rendering of these texts into English.

A critical edition of Kitab Raf' shan al-hubshan by Jalal al-din al-Suyuti

The edition is based on nine manuscripts. The work deals with the virtues of noble Abyssinians and is based on the earlier work by ibn al-Jawzi which it partly extends. The Arabic writings on the black races are reviewed from the beginnings of the genre to works influenced by Suyuti. Attention is therefore particularly given to Suyuti's predecessors and successors from the 2nd to the 11th centuries with special reference to the relations between Suyuti's work and that of ibn al-Jawzi. The thesis is divided into two parts. The first part is the English introduction which consists of four chapters with the conclusions placed after Chapter Three. The first chapter deals with the works relevant to al-sudan and the Abyssinians. First, the two terms "al-Habashah" and "al-Sudan" are briefly discussed in an attempt to define their usage. The second chapter, which is a critical study of Kitab Raf Shan al-Hubshan, is divided into six sections, in the last section of which it will be shown how this book was more popular than ibn al-Jawzi's work' on which it was dependent. The third chapter provides biographical detail of al-Suyuti's life with some comments on the number of his works. The fourth chapter contains the description of the manuscripts and editing principles. Finally, the bibliography is provided at the end of this part. The second part consists of the list of works cited in the footnotes of the Arabic Text, the list of abbreviations used in these footnotes, the conventional signs used in the Text, the Text, and the indexes.

The first difficulty which confronts the student of the ideas of ʾAbd al-Raḥmān al-Kawākibi is the absence of a full biography of the man; al-Kawākibi did not have his Boswell, nor would his personality have attached a faithful and admiring disciple as Muḥammad Rashīd Riḍa. He is nevertheless a well thought of figure, and he appears in the standard literary and biographical dictionaries. It is to be noticed however that all the biographical notes which refer to him have one and the same source: the two official documents which Rashīd Riḍa, editor of al-Manār printed in his obituary notice about al-Kawākibi. These documents and the life of al-Kawākibi given in al-Manār were used by all subsequent writers and editors of biographical compilations.
