United in Diversity : A Physiological and Molecular Characterization of Subpopulations in the Basal Ganglia Circuitry

The Basal Ganglia consist of a number of different nuclei that form a diverse circuitry of GABAergic, dopaminergic and glutamatergic neurons. This complex network is further organized in subcircuits that govern limbic and motor functions in humans and other vertebrates. Due to the interconnection of the individual structures, dysfunction in one area or cell population can affect the entire network, leading to synaptic and molecular alterations in the circuitry as a whole. The studies in this doctoral thesis aimed at characterizing restricted subpopulations of neurons in the Basal Ganglia circuitry and their importance in the wider function of the network. To this end, we identified subpopulations of neurons in the subthalamic nucleus (STN), substantia nigra (SN) and ventral tegmental area (VTA), characterized their molecular profile and investigated their physiological role in the circuitry. Within the mouse STN, reduction of glutamatergic neurotransmission in a subpopulation expressing Paired-like homeodomain transcription factor 2 (Pitx2) led to structural alterations in the nucleus as well as biochemical alterations of the dopaminergic system in the Nucleus accumbens (NAc) and changes in reward-related behavior. In the ventral midbrain, we identified and characterized novel marker genes selective to the VTA or SN. Of these, transient receptor potential cation channel subfamily V member 1 (TrpV1) marks a population of mainly glutamatergic neurons in the VTA which project to the NAc, while gastrin releasing peptide (Grp) is expressed in a population of dopaminergic neurons neuroprotected in Parkinson's disease. Furthermore, we discovered that disruption of glutamatergic co-release of dopaminergic neurons expressing dopamine transporter (DAT), diminishes fast EPSCs and glutamate release but does not affect the acquisition of reward-related behavioral tasks. To selectively quantify glutamate release from specific subpopulations, we devised a technique combining glutamate-amperometry and optogenetics. This was used to measure glutamate released from Pitx2-expressing synaptic terminals in the Globus pallidus as well as DAT- or TrpV1-expressing terminals in the NAc. In summary, this doctoral thesis has furthered understanding of the function and importance of specific subpopulations within the Basal Ganglia circuitry and provides a novel means to investigate glutamate in the intact rodent brain within clearly defined, restricted cell populations.

Développement de biocapteurs pour la détermination de substances biologiquement actives / Development of biosensors for the determination of biologically active substances

Les biocapteurs sont des moyens d’analyse en plein essor à la fois rapides, sélectifs et peu coûteux applicables à des domaines extrêmement variés (environnement, santé, agroalimentaire,…). Dans ce type d’outil, un élément sensible de nature biologique (anticorps, enzyme, microorganisme, ADN…) doté d’un pouvoir de reconnaissance pour un analyte ou un groupe d’analytes est associé à un transducteur pouvant être de type électrochimique, optique ou thermique.Dans ce travail, nous nous sommes intéressés au développement de trois biocapteurs pour la détection de substances biologiquement actives. Le premier permet la détermination simultanée de l’adénosine triphosphate (ATP) et du glucose par ampérométrie, le deuxième celle de la créatine kinase, et le troisième est un biocapteur conductimétique pour la quantification de l’ATP. Dans les deux premiers biocapteurs, deux enzymes (l’hexokinase et la glucose oxydase) sont immobilisées à la surface de microélectrodes constituées d’un disque de platine. Le troisième biocapteur est basé sur l’immobilisation de l’hexokinase sur des microélectrodes interdigitées en or. L’immobilisation est réalisée dans tous les cas par co-réticulation des enzymes en présence d’albumine de sérum bovin à l’aide de glutaraldehyde. Les caractéristiques analytiques des biocapteurs ont été déterminées et différentes procédures ont été développées pour l’analyse d’échantillons réels. Les biocapteurs ont pu être appliqués avec succès à la quantification de l’ATP, du glucose et de la créatine kinase dans des préparations pharmaceutiques et du sérum sanguin / Biosensors are rapid, selective and inexpensive devices that combine a biological recognition element, the so-called bioreceptor (e.g. enzymes, antibodies, DNA or microorganisms) to a physical transducer (e.g. electrochemical, optical, thermal or piezoelectrical). They can be used to detect one specific analyte or one family of analytes for a wide range of applications (e.g. environment, food, health). In this work, the detection of biologically active substances was targeted. A biosensor system for simultaneous determination of adenosine triphosphate (ATP) and glucose, a biosensor for creatine kinase analysis, and a novel conductometric biosensor for ATP determination were developed. In the first two biosensors, two enzymes (hexokinase and glucose oxidase) were immobilized at the surface of platinum disc microelectrodes for amperometric detection. The third biosensor was based on hexokinase immobilized onto gold interdigitated microelectrodes for conductometric detection. In all cases, the enzymes were co-immobilized with bovine serum albumin by cross-linking using glutaraldehyde. Analytical characteristics of the biosensors were determined and different procedures were developed for real samples analysis. The biosensors could be successfully applied to the determination of ATP, glucose, and creatine kinase in pharmaceutical samples and blood serum

Esta tese apresenta o desenvolvimento de metodologias analíticas em sistemas de solução em fluxo com detecção amperométrica e espectrofotométrica explorando a utilização de tecidos vegetais como fonte enzimática para a biocatálise de reações. Desempenhos satisfatórios foram obtidos com a utilização do mesocarpo fibroso do coco (Cocus nucifera, L.) e com a casca e/ou polpa de frutos da palmeira leque (Latania sp), fontes de polifenol oxidase. Uma metodologia, simples e rápida, para a determinação dos parâmetros bioquímicos da polifenol oxidase foi desenvolvida ao longo do presente trabalho com a enzima naturalmente imobilizada no tecido do coco, pela eliminação das etapas de extração e de purificação e associando-se análise em fluxo e espectrofotometria. Parâmetros cinéticos importantes como o efeito do pH, a constante de Michaelis-Menten (Km), a velocidade máxima (Vmax),a energia de ativação (Ea) e os parâmetros térmicos (valores D, z e Q10)foram determinados utilizando como substrato o catecol. A atividade relativa junto a diferentes substratos e o efeito de inibidores foram também avaliados. Aplicações envolvendo estes tecidos vegetais em reatores empacotados (associados com FIA) ou incorporados em eletrodos de pasta de carbono (medidas realizadas em batelada) para a determinação de produtos fenólicos como catecol, fenol e dopamina, inibidores da atividade enzimática como o sulfito, bem como para a quantificação do conteúdo de água, demonstram a sua versatilidade. Os produtos fenólicos foram quantificados com boa sensibilidade (na faixa de µmol L-1) pela redução amperométrica das respectivas quinonas, produzidas pela oxidação enzimática. Aplicações com amostras reais, dentre as quais água de rio e resíduos de uma fábrica de papel puderam ser implementadas. A determinação do conteúdo de água em meio aquo-restrito explorou a estimulação, pela água, da atividade catalitica da polifenoI oxidase naturalmente imobilizada no mesocarpo fibroso do coco na presença de catecol. Esta propriedade foi utilizada para a determinação rápida e reprodutível do conteúdo de água em amostras comerciais de álcool, utilizando acetonitrila como carregador. A quantificação de sulfito baseou-se no seu efeito inibidor sobre a polifenol oxidase naturalmente imobilizada em relação à catálise oxidativa do catecol. Diversos interferentes comumente presentes em amostras de vinho foram também investigados. Durante este trabalho, foi desenvolvida uma simples e interessante maneira de conservar o mesocarpo fibroso do coco como fonte de polifenol oxidase naturalmente imobilizada. O tecido desta fruta foi desidratado e moído. O pó deste tecido, estocado a mais de dois anos à temperatura ambiente, ainda apresenta boa atividade enzimática. / The development of analytical methodologies exploring plant tissues as enzymatic source for biocatalysis of many reactions is presented in this thesis. Amperometry and spectrophotometry was associated with flow analysis for analytical purposes and for biochemical characterization of naturally immobilized polyphenol oxidase. Good performance was achieved with tissues from the fruits of two palm trees: the fibrous mesocarp of green coconut fruits, Cocus nucifera, L., and the skin and the pulp of green fruits from Latania sp. Both tissues are very effective in the biotransformation of o-diphenols to the corresponding o-quinones. A simple, fast, and new methodology for the determination of the biochemical parameters of the poyphenol oxidase naturally immobilized on the fibrous tissue was developed (eliminating the extraction and purification of enzymes) by association of flow injection analysis and spectrophotometry. For coconut tissue, cinetic parameters like pH effect, Michaelis-Menten constant (Km) and maximum rate (Vmax), activation energy (Ea) and thermal parameters (D, z and Q10values) on catechol biotransformation were determined. Also the response for several substrates as too for various inhibitors was explored. Amperometric quantification of phenolic compounds was made using the vegetal tissue in form of packed reactors (associated with FIA) or incorporated in carbon paste electrodes (measurements in batch). Very good response for catechol, phenol and dopamine was registered for this compounds, with very high sensitivity (µmol L-1 range). Applicability to real samples as for river water and for a paper plant waste water was verified. The same amperometry-FIA system was employed for enzymology in non-aqueous medium. The activity of the polyphenol oxidase contained in coconut tissues are strongly dependent of water. The strategy involves the stimulation by the content of water on the biocatalytic activity of the enzyme in the presence of catechol substrate. This strategy was used for the determination of water content in alcohol samples, in medium of dry acetonitrile. The inhibition of the enzymatic activity produced by many compounds can be explored for an indirect quantification of the inhibitor. A flow injection amperometric procedure was developed for the determination of sulphite ion based in its inhibitory effect on the activity of polyphenol oxidase on the oxidation of catechol. The effect on the bioreactor performance of potential interferents commonly present in wine samples were also investigated. During this work, it was developed a simple and interesting way to preserve coconut tissue. The mesocarp of this fruit can be dried and grounded. These tissues still with very good activity after more than two years in \"shelf temperature\".

Novos materiais para aplicações analíticas nas determinações de compostos orgânicos de interesse farmacêutico e ambiental / New materials for analytical application in organic compound determination for pharmaceutical and environmental interests

Sensores de filme de carbono, construídos a partir de resistores com 2 Ω de resistência nominal, foram utilizados para aplicações em áreas de interesse ambiental e farmacêutico sem a modificação prévia de suas superfícies. Através de um tratamento eletroquímico em meio ácido, é possível obter uma faixa de potencial (para ambos os limites anódico e catódico) maior comparado aos eletrodos de carbono convencionalmente utilizados. Devido ao fato dos sensores serem confeccionados em escala industrial na Alemanha, seu custo é muito baixo, tornando possível sua utilização na forma de eletrodos descartáveis. Além disso, são caracterizados por uma grande versatilidade, reprodutibilidade e estabilidade. Para comprovar a viabilidade dos eletrodos de filme de carbono como transdutores voltamétricos, amostras de paraquat foram analisadas em águas de rio e potável. A associação com a voltametria de onda quadrada possibilitou obter um limite de detecção inferior ao limite máximo estabelecido para o herbicida em água potável. Estudos por voltametria cíclica mostraram dois picos para a redução do paraquat, similares aos descritos na literatura. Os sensores de filme de carbono também foram adaptados ao sistema de análise por injeção em fluxo para a determinação amperométrica de paracetamol e de ambroxol em formulações farmacêuticas. Em ambos os estudos, foi possível obter ampla faixa linear de concentração, baixos limites de detecção e sensibilidade elevada. Estudos de repetibilidade com os dois analitos apresentaram desvios padrões relativos de até 3%, comprovando a ausência de efeito de memória. Os resultados obtidos durante as análises em fluxo com os novos sensores amperométricos concordaram com as metodologias descritas nas farmacopéias. Ao longo dos quatro anos de doutoramento também foram desenvolvidos outros estudos, os quais estão descritos nos apêndices desta tese. / Carbon film sensors, built from resistors of 2 Ω nominal resistance, were employed for application in the environment and pharmaceutical fields without previous modification of the electrode surface. When these sensors are electrochemically treated they obtain a wider potential window (both anodic and cathodic regions) than many other forms of carbon. Due to the fact that sensors are produced on industrial scale in Germany, the cost of each unit is very low, a feature that enables the use of these electrodes in disposable form. Moreover, they have great versatibility, reproducibility and stability. To prove the suitability of the carbon film electrodes as voltammetric sensors paraquat samples were analysed in river and drinking water. The association with the square wave voltammetry led to a detection limit below the allowed upper limit for herbicide analysis in drinking water. Previous results with cyclic voltammetry showed two peaks for paraquat reduction, in agreement with the literature data. The carbon film sensors were also adapted to the flow injection analysis system for amperometric determination of acetaminophen and ambroxol in pharmaceutical formulations. It was possible to obtain a wide linear working range, low detection limit and high sensitivity for both analytes. During the repeatabilty studies, acetaminophen and ambroxol showed relative standard desviations lower than 3% without any memory effect. The results obtained with a new amperometric sensor associated to FIA were in good agreement with those recommended by pharmacopoeias. In the appendices of this thesis are described other studies which have been developed for four years of PhD.

Estudo da autoxidação dos complexos de Cu(II), Ni(II) e Co(II)/tetraglicina induzida por S(IV) / Study of the autoxidation of the complexes of Cu (II), Ni (II) and Co (II)/tetraglycine induced by S(IV)

O presente trabalho apresenta estudos espectrofotométricos relacionados à autoxidação dos complexos de Cu(II), Ni(II) e Co(II)/tetraglicina induzida por sulfito. Nossos estudos verificaram que a autoxidação de Cu(II)/tetraglicina (1,0x10-3 mol L-1) em pH = 9,0 (tampão borato) é afetada pela presença de traços de Ni(II) ou Co(II). Na ausência de Ni(II) ou Co(II), a reação é muito ineficiente e lenta com períodos de indução longos (aproximadamente 4 h). Ni(II) ou Co( II) em concentrações baixas ( 10-5 - 10-6 mol L-1) afetam significativamente a cinética: o período de indução diminui drasticamente (a menos de 2 s) e a formação de Cu(III) é fortemente acelerada com simultâneo aumento da eficiência da reação. A atividade catalítica e o sinergismo positivo de Co(II) e Ni(II) podem ser explicados pela oxidação mais rápida de Co(II) ou Ni(II)/tetraglicina pelo oxigênio dissolvido. O processo eletroquímico relacionado aos sistemas Cu(II)/Cu(III)/tetraglicina e Ni(II)/Ni(III)/tetraglicina são reversíveis, possibilitando monitorá-los adequadamente mediante o uso da técnica de eletrodo rotativo disco-anel. Entretanto, o sistema Co(II)/Co(III)/tetraglicina é irreversível. Esses estudos mostraram que as espécies de Cu(III) e Ni(III) geradas no eletrodo disco são instáveis nas condições experimentais empregadas. O efeito sinérgico positivo na presença de Ni(II) (que permitiu aumentar a sensibilidade) foi aproveitado para desenvolvimento de método espectrofotométrico e amperométrico simples e sensível para a determinação indireta de sulfito em meio aquoso. O método espectrofotométrico está baseado na medida de absorbância do complexo de Cu(III)/tetraglicina (gerado na presença de sulfito e traços de Ni(II)) em 365 nm. O método amperométrico em análise por injeção em fluxo baseia-se na medida de corrente (0,1 V vs Ag/AgCl) em função da concentração de Cu(III)/tetraglicina gerado quimicamente, na presença de sulfito e traços de Ni(II). Os métodos desenvolvidos foram empregados para a determinação de S(IV), em vinhos e sucos, após a sua extração da amostra acidificada, os resultados obtidos concordaram com aqueles obtidos pelo método iodométrico. / The present work presents spectrophotometric studies related to the sulfite induced autoxidation of Cu(II), Ni(II) and Co(II)/tetraglycine complexes. The sulfite induced autoxidação of Cu(II)/tetraglycine (1.0x10-3 mol L-1) at pH = 9.0 (borate buffer) is affected by the presence of small quantities of the Ni(II) or Co(II). In the absence of added nickel (II) or cobalt (II), the reaction is very inefficient and slow with one large induction period (about 4 h). Trace amounts of Ni(II) or Co(II) (10-5 - 10-6 M) affect the kinetic significantly: the induction period drastically decreases (less than 2 s) and the formation of Cu(III) is strongly accelerated. The effectiveness of Cu(III) formation becomes much higher. The catalytic activity and the positive synergism of Co(II) and Ni(II) may be explained by the faster oxidation of Co(II) or Ni(II)/tetraglycine complexes by dissolved oxygen. The electrochemistry of Cu(II)/Cu(III)/tetraglycine and Ni(II)/Ni(III)/tetraglycine systems are reversible, such as it was possible to monitor them by using the rotating ring-disk electrode technique. However, the Co(II)/Co(III)/tetraglycine system is irreversible. Those studies showed that the Cu(III) and Ni(III) species generated on the disk electrode are unstable in the employed experimental conditions. The positive sinergistic effect in the presence of Ni(II) (which allowed to increase the sensibility) was taken in advantage for development of one simple and sensitive spectrophotometric and amperometric method for indirect determination of sulfite in aqueous medium. The spectrophotometric method is based on the absorbance measurement of the Cu(III)/tetraglicyne complex (generated in the presence of sulfite and small quantities of Ni(II)) at 365 nm. The amperometric method by flow injection analysis is based on the current measurement (0.1 V vs Ag/AgCl) as function of Cu(III)/tetraglycine concentration chemically generated, in the presence of sulfite and Ni(II). The methods were employed for the determination of S(IV), in wines and juices, after its extraction from acidified samples and the results were in agreement with those obtained by the iodometric procedure.

RESUME Le travail de thèse porte sur le développement d’électrodes sélectives originales et performantes pour l’analyse de composés d’intérêt pharmaceutique. La partie introductive traite des notions relatives à l’électrochimie mais également de notions sur les molécules médicamenteuses étudiées, en l’occurrence les principes antipaludéens et l’iode. La partie expérimentale se subdivise en deux parties distinctes selon le type d’électrodes sélectives auxquelles font appel les techniques électrochimiques. La première partie concerne l’élaboration, la caractérisation et l’application des électrodes potentiométriques à membrane polymérique incluant une paire d’ions et sélectives à diverses molécules organiques pharmacologiquement actives (antipaludéens). Leur application aussi bien en analyse pharmaceutique qu’en cinétique de dissolution est décrite. La deuxième partie est consacrée à l’élaboration d’un type de senseur ampérométrique original à pâte de carbone à base d’argent micronisé ou colloïdal et à la comparaison de ses performances avec l’électrode d’argent métallique. L’intérêt analytique est mis en évidence par la détermination quantitative des iodures. Les différents aspects susceptibles d’influencer leur comportement, dont la nature des agents précipitants (tétraphénylborate de sodium et le tétrakis (4-chlorophényl) borate de potassium) et de plastifiants ont été investigués. Les bonnes performances des ces électrodes en analyse quantitative ont permis d’explorer les possibilités de leur utilisation à l’étude de la cinétique de dissolution. L’ampérométrie à électrode à pâte de carbone modifiée à base d’argent à l’échelle micronisée (35% m/m) couplée à la chromatographie liquide ionique s’est avérée très sensible vis-à-vis des iodures en particulier et des halogénures en général. Les facteurs susceptibles d’influencer les grandeurs de séparation et la réponse de l’électrode ont été investigués et l’exploitation du signal ampérométrique permet le dosage sélectif et rapide de faibles concentrations en iodures. Les informations fournies par les mesures réalisées en voltampérométrie cyclique à l’aide des mêmes électrodes permettent une bonne compréhension mécanistique quant au mode de détection ampérométrique évitant ainsi toute confusion à ce sujet et permettant l’optimisation du processus de détection. ------------------------------------------------ ABSTRACT This thesis describes the development of original and high performance selective electrodes for the analysis of several pharmaceutical compounds. The introduction describes the pharmaceutical compounds of interest (antimalarial drugs and iodine) and provides an overall understanding of the electrochemical groundwork pertaining to their analysis. The experimental aspect of the thesis is divided into two parts, each according to the type of electrode and electrochemical technique used for the analysis. The first part describes the design, characterization, and application of polymer membrane based ion selective potentiometric electrodes. Selectivity was provided by including ion pairs of several antimalarial drugs into the membrane. The feasibility of use of these electrodes in pharmaceutical analysis as well as in dissolution trials is also described in this part. The second part describes the design of an original silver-modified carbon paste amperometric sensor and compares its performances to those of a plain metallic silver electrode. The electrode has been modified by silver microparticles or by silver nanoparticles. Quantitative iodine determination serves to prove the usefulness of this new sensor in analytical chemistry. Different aspects, such as the nature of the counter ions (sodium tetraphenylborate and potassium tetrakis (4-chlorophenyl) borate) and the plastifying agents that are likely to influence electrode behaviour have been investigated. Since the electrodes have been shown to perform well in quantitative analysis, the possibility of use in dissolution trials was explored. Micronized silver-modified carbon paste electrode (35% Ag m/m) coupled to anionic-exchange liquid chromatography with amperometric detection was shown to be very sensitive with regards to the assay of halogenides in general and iodide in particular. After having investigated the various factors likely to influence chromatographic separation and electrode response, it was shown that the sensor could be used to rapidly and selectively determine low iodide concentrations in complex samples. Cyclic voltammetric analysis provided information concerning the mechanisms allowing amperometric detection, thus allowing an optimisation of the detection procedures.

[pt] O presente trabalho teve como objetivo o desenvolvimento de métodos eletroanalíticos empregando elétrodos de grafite-epóxi e diamante dopado com boro para a determinação do antibiótico sulfato de gentamicina, substâncias de interesse biológico (naftoquinonas) e do pesticida trifloxistrobina. O diamante dopado com boro (DDB) apresenta características atraentes de detecção eletroquímica que são úteis em aplicações analíticas baseadas em voltametria e amperometria. Possui uma ampla janela de potencial em soluções aquosas que permitiu a quantificação do fungicida trifloxistrobina (em +1744 mV versus Ag/AgCl(KClsat)) por voltametria de onda quadrada (SWV), em um tampão Britton-Robinson (0,04 mol L-1, pH 4,00 )/acetonitrila 70/30 por cento v/v. A curva de adição de analito foi obtida usando amplitude de pulso 40 mV, freqüência de 30 Hz, passo de potencial de 20 mV. O limite instrumental de detecção foi de 1,4 × 10-7 mol L-1 e a faixa linear dinâmica abrangeram três ordens de grandeza (10-7 a 10-5 mol L-1). As amostras foram analisadas com recuperações de cerca de 80 por cento em amostras de suco de laranja e de 92,4 a 104,0 por cento em amostras de água. Um estudo para avaliar potenciais interferentes foi realizado na presença de outros fungicidas. Estudos diagnósticos indicaram que a oxidação da trifloxistrobina em meio aquoso na superfície do DDB é irreversível, envolvendo duas etapas, cada um com dois elétrons. A degradação UV da trifloxistrobina foi avaliada utilizando o método eletroquímico proposto e a cinética de degradação estabelecida com meia-vida de 1.07 min. A amperometria por injeção em batelada (BIA do inglês batch-injection analysis) com o DDB (operando a +2000 mV) foi utilizada para desenvolver um método para a determinação de sulfato de gentamicina. A extração em fase sólida (SPE do inglês solid-phase extraction), utilizando um cartucho empacotado com um polímero impresso molecularmente, tendo a canamicina como molécula molde, foi utilizada para melhorar a seletividade. O tampão acetato (0,01 mol L-1; pH 4,4) foi utilizado como eletrólito suporte. A oxidação do sulfato de gentamicina ocorreu logo após a injeção do analito (60 microlitros) que entra em contato com a superfície do eletrodo. O cartucho de SPE foi muito eficiente na retenção do analito quando comparado com o polímero não-impresso correspondente (NIP). A curva analítica apresentou resposta linear, a precisão instrumental foi inferior a 3 por cento e o limite de quantificação foi de 2,7 × 10-6 mol L-1. A frequência analítica foi de 90 medições h-1 e nenhum efeito de memória foram observados entre as medidas sequênciais. O desempenho do método proposto foi comparado com o obtido por HPLC-UV e com espectrofotometria de absorção molecular (ambos após derivação química do analito com o-ftalaldeído ou com ninhidrina). O método desenvolvido neste trabalho foi aplicado em formulações farmacêuticas injetáveis e em amostras simuladas, com recuperações próximas de 100 por cento. Para a determinação do lapachol foi utilizada a SWV usando um eletrodo de grafite-epoxi feito no laboratório. O meio eletrolítico utilizado foi uma solução aquosa contendo o surfactante catiônico CTAB (1,2 × 10-4 mol L-1), tampão fosfato (4,0 × 10-2 mol L-1, pH 6,0) e KNO3 (1,0 mol L-1). O surfactante catiônico melhorou a difusão e a interação do analito com o eletrodo, produzindo um processo reversível que melhorou a corrente total medida pelo sistema. O sinal do lapachol foi medido a -470 mV, após pré-concentração a 400 mV durante 140 s, usando frequência de 30 Hz, amplitude de pulso de 40 mV com passo de potencial de 20 mV. O limite instrumental de detecção foi de 0,029 mg L-1 e a faixa linear dinâmica abrangeu duas ordens de grandeza. A determinação do lapachol em extrato etanólico de Tabebuia impetiginosa foi realizada após uma s / [en] The present work aimed to develop electroanalytical methods using graphite-epoxy and boron-doped diamond electrodes for the determination of the antibiotic gentamicin sulfate, substances of biological interest (naphthoquinones) and the pesticide trifloxystrobin. Boron doped diamond (BDD) features attractive electrochemical detection characteristics that are useful in analytical applications based on voltammetry and amperometry. It has a wide potential window in aqueous solutions that allowed the quantification of the fungicide trifloxystrobin (in +1744 mV versus Ag/AgCl(KClsat)) by square-wave voltammetry (SWV) in Britton-Robinson buffer (0.04 mol L-1, pH 4.00) / acetonitrile 70/30 percent v/v electrolyte medium. The analyte addition curve was constructed using 40 mV pulse amplitude, 30 Hz frequency, 20 mV step potential. The instrumental limit of detection was 1.4 × 10-7 mol L-1 and the linear dynamic range covered three orders of magnitude (from 10-7 to 10-5 mol L-1). Recoveries of about 80 percent in orange juice samples and 92.4 to 104.0 percent in water samples. A study to evaluate potential interferences was carried out in the presence of other fungicides. Diagnostic studies indicated that the oxidation of trifloxystrobin in aqueous medium, on the surface of the BDD, is irreversible, involving two steps, each of them with two electrons. UV degradation of trifloxystrobin was studied using the proposed voltammetric method and degradation kinetics with a half-life of 1.07 min was established. Batch injection amperometry (BIA) with BDD (operating at +2000 mV) was used to develop a method for the determination of gentamicin sulfate. Solid phase extraction (SPE) using a cartridge packed with a molecular imprinting polymer made with kanamycin as the template molecule, was used to improve the selectivity. The acetate buffer (0.01 mol L-1; pH 4.40) was used as supporting electrolyte. Oxidation of gentamicin sulfate occurred right after the analyte (60 microlitres solution) comes into contact with the surface of the electrode. The SPE cartridge packed with MIP was very efficient in retaining the analyte when compared to the one packed with the corresponding non-printed polymer (NIP). The analytical curve was linear, the instrument accuracy was less than 3 percent and the limit of quantification was 2.7 × 10-6 mol L-1. The analytical frequency was 90 measurements h-1 and no memory effect was observed between sequential measurements. The performance of the proposed method was compared with that obtained by HPLC-UV and molecular absorption spectrophotometry (both after chemical derivatization of the analyte with o-phthalaldehyde or with ninhydrin). The method was applied in injectable pharmaceutical formulations and in simulated samples, with recoveries close to 100 percent. For the determination of lapachol SWV was employed using a graphite-epoxy electrode made in the laboratory. The electrolytic medium used was an aqueous solution containing the cationic surfactant CTAB (1.2 × 10-4 mol L-1), phosphate buffer (4.0 × 10 -2 mol L-1, pH 6.00) and KNO3 (1.0 mol L-1). The cationic surfactant improved the diffusion and interaction of the analyte with the electrode, producing a reversible process that improved the total current measured by the system. The lapachol signal was measured at -470 mV after preconcentration at 400 mV for 140 s, using a 30 Hz frequency, 40 mV pulse amplitude with a step potential of 20 mV. The instrumental limit of detection was 0.029 mg L-1 and the dynamic linear range comprised two orders of magnitude. In the presence of beta-lapachone (structural isomer), the selectivity was obtained by the first-order derivative of the SWV signal. The determination of lapachol in ethanolic extract of Tabebuia impetiginosa was carried out after a simple separation of the analyte by thin layer chromatography. The results are statistically similar (with 95 percent confidence level) with those performed through HPLC-UV.
