When published, it was received as a book belonging to the chick lit genre but still got good reviews. This combination is really unusual and awoke my interest in the novel. My thesis is therefore an analysis whether </span><em><span lang="EN-GB">Ladies</span></em><span lang="EN-GB"> can be considered a chick lit novel or not, and in what way it is similar to or differs from the genre.</span></p><p class="MsoNormal"><span lang="EN-GB"> </span></p><p class="MsoNormal"><span lang="EN-GB">I start my thesis with an overview of former research of chick lit and what earlier was called trivial literature. Here Lisbeth Larsson’s <em>En annan historia</em>, </span><em><span lang="EN-GB"><span>[</span></span></em><em><span lang="EN-GB">Another Story</span></em><em><span lang="EN-GB"><span>]</span></span></em><em><span lang="EN-GB">,</span></em><span lang="EN-GB"> Janice Radway’s <em>Reading the Romance</em> and Suzanne Ferriss’ and Mallory Young’s anthology <em>Chick Lit The New Woman’s Fiction</em> were necessary to understand the background of the genre.</span></p><p class="MsoNormal"><span lang="EN-GB">After the research overview I continue with a narratological analysis, using the concepts of Maria Nikolajeva’s <em>Barnbokens byggklossar </em></span><em><span lang="EN-GB"><span>[</span></span></em><em><span lang="EN-GB">The Bricks of the Children Book</span></em><em><span lang="EN-GB"><span>]</span></span></em><span lang="EN-GB"> to look at the structure of a chick lit story. After finishing my chick lit genre analysis I try to complete my own definition of the genre.</span></p><p class="MsoNormal"><span lang="EN-GB">I find the themes love, sexuality and body focus in </span><em><span lang="EN-GB">Ladies</span></em><span lang="EN-GB"> especially interesting and therefore choose to look closer into how these areas are represented in the book. By using Judith Butler’s concepts the heterosexual matrix, the represented gender and Tiina Rosenberg’s hetero normativity I analyse the representation of the themes. </span></p><p class="MsoNormal"><span lang="EN-GB"> </span></p><p class="MsoNormal"><span lang="EN-GB">In the final discussion I compare my different analyses and my conclusion shows that </span><em><span lang="EN-GB">Ladies</span></em><span lang="EN-GB"> can’t be considered belonging to the chick lit genre. The biggest difference lies in how love and sexuality are represented in the book compared to chick lit novels. Development of roadway link screening criteria for microscale carbon monoxide and particulate matter conformity analyses through application of classification tree model

The impacts of emissions sources of carbon monoxide and particulate matter pollution levels for projected level conformity assessment and National Environmental Policy Act (NEPA) analyses are usually estimated through computer-aided models. Because of the involvement and interaction of a large number of variables that affect formation of CO and PM hot spots, exhaustive impact assessment studies can be time consuming. This is especially true for complex urban projects consisting of numerous roadways whose potential CO and PM impacts on surrounding neighborhoods must be disclosed. A highway project may consist of hundreds of roadway links, therefore undertaking project level conformity analysis without screening tools can be computationally resource intensive. CALINE4, a line source emission modeling tool, is used to predict downwind CO and PM concentrations for various receptors to generate a learning dataset for development of screening rules. This research has developed statistical screening criteria based on Classification and Regression Tree modeling that can be used to eliminate those links from the CALINE4 analysis whose contribution of pollutant concentration to a particular receptor site are insignificant. For the purpose of this study, any link that contributes a concentration of 0 ppm of CO or 0 µg/m3 of PM to a particular receptor site is termed insignificant for the corresponding pollutant. The model uses seven predictor variables, namely wind speed, wind directional variability, linear emission flux, link length and receptor polar coordinates. Response vector has two classes of pollutant concentrations namely significant and insignificant which are obtained by conversion of numerical values of pollutant concentration according to above mentioned criterion, thereby converting a regression problem into categorical or classification problem. The developed rules based on constructed model were validated through test samples and can be applied to future dataset to classify and screen out the insignificant links in highway planning analyses. The screening tool also allows analysts to prepare gridded pollution concentration predictions for use in environmental justice analyses.

<b>PROBABILISTIC ENSEMBLE MACHINE LEARNING APPROACHES FOR UNSTRUCTURED TEXTUAL DATA CLASSIFICATION</b>

<p dir="ltr">The volume of big data has surged, notably in unstructured textual data, comprising emails, social media, and more. Currently, unstructured data represents over 80% of global data, the growth is propelled by digitalization. Unstructured text data analysis is crucial for various applications like social media sentiment analysis, customer feedback interpretation, and medical records classification. The complexity is due to the variability in language use, context sensitivity, and the nuanced meanings that are expressed in natural language. Traditional machine learning approaches, while effective in handling structured data, frequently fall short when applied to unstructured text data due to the complexities. Extracting value from this data requires advanced analytics and machine learning. Recognizing the challenges, we developed innovative ensemble approaches that combine the strengths of multiple conventional machine learning classifiers through a probabilistic approach. Response to the challenges , we developed two novel models: the Consensus-Based Integration Model (CBIM) and the Unified Predictive Averaging Model (UPAM).The CBIM and UPAM ensemble models were applied to Twitter (40,000 data samples) and the National Electronic Injury Surveillance System (NEISS) datasets (323,344 data samples) addressing various challenges in unstructured text analysis. The NEISS dataset achieved an unprecedented accuracy of 99.50%, demonstrating the effectiveness of ensemble models in extracting relevant features and making accurate predictions. The Twitter dataset, utilized for sentiment analysis, demonstrated a significant boost in accuracy over conventional approaches, achieving a maximum of 65.83%. The results highlighted the limitations of conventional machine learning approaches when dealing with complex, unstructured text data and the potential of ensemble models. The models exhibited high accuracy across various datasets and tasks, showcasing their versatility and effectiveness in obtaining valuable insights from unstructured text data. The results obtained extend the boundaries of text analysis and improve the field of natural language processing.</p>

Revisão sistemática da reposição com testosterona em homens obesos com níveis séricos baixos de testosterona

Orientador: Vania dos Santos Nunes Nogueira / Resumo: Introdução: o uso de testosterona em homens obesos tem sido controverso em termos de perda de peso e controle de doenças relacionadas à obesidade. Objetivo: avaliar a efetividade da reposição de testosterona para perda de peso e prevenção de complicações cardiovasculares em homens obesos com baixos níveis séricos de testosterona. Métodos: foi realizada uma revisão sistemática de acordo com a Metodologia Cochrane de estudos randomizados que compararam em homens obesos com baixos níveis de testosterona a reposição de testosterona versus a não reposição. Dois revisores (ASM e LARB) independentemente fizeram a seleção dos estudos, avaliaram o risco de viés e extraíram os dados dos estudos incluídos. Os desfechos primários analisados foram: melhora dos parâmetros antropométricos (peso, índice de massa corpórea, circunferência de cintura e composição corporal), segurança, qualidade de vida, controle de complicações relacionadas a obesidade e melhora dos sintomas de hipogonadismo. As bases de dados eletrônicas para pesquisa foram Embase, Medline, LILACS e CENTRAL. Desfechos semelhantes em pelo menos dois estudos foram plotados na meta-análise usando o software Review Manager 5.3. Objective: To analyze the effectiveness of testosterone therapy for weight loss and prevention of cardiovascular complications in obese men with low serum testosterone levels. Methods: A systematic review was conducted according to the Cochrane Methodology of randomized trials comparing testosterone replacement versus non-replacement in obese men with low testosterone levels. Two reviewers (ASM and LARB) independently selected the studies, assessed the risk of bias, and extracted data from the included studies. The main outcomes analyzed were: improvement of anthropometric parameters (weight, body mass index, waist circumference and body composition), safety, quality of life, control of complications related to obesity and improvement of symptoms of hypogonadism. The electronic databases for search were Embase, Medline, LILACS and CENTRAL. Similar outcomes in at least two trials were plotted in the meta-analysis using Review Manager 5.3 software. O efeito dos hormônios glicocorticóides (GC) de suprimir o crescimento celular é exercido através de cascatas celulares nas quais a transcrição de genes de resposta primária, mediada direta ou indiretamente pelo receptor de GC, regulam a transcrição e a atividade de um conjunto de genes, incluindo fatores importantes para a progressão no ciclo celular. Entretanto, a conexão funcional entre diversos dos genes ativados ou inibidos por GC e a inibição da proliferação de determinados tipos celulares ainda não é completamente conhecida. Nosso laboratório isolou a variante ST1, a partir da linhagem C6 de glioma de rato, utilizando-a como modelo de estudo do mecanismo de ação de GC como agentes anti-tumorais. O tratamento com GC confere, às células ST1, um crescimento totalmente dependente de soro e ancoragem, morfologia em cultura semelhante à de fibroblastos normais e incapacidade de gerar tumor em camundongos da linhagem \"nude\", caracterizando uma completa reversão fenotípica tanto in vitro como in vivo. However, the functional connection between the GC-regulated transcriptional events and cell cycle arrest of determined cells is poorly understood at the molecular level. In this context, our lab has isolated the ST1 variant of the C6 rat glioma cell line to study the mechanism of action of GC as anti-tumor agents. GC treatment leads ST1 cells to a dramatic tumoral to normal phenotypic reversion, characterized by inhibition of their growth rate in monolayer, loss of their ability to form colonies in semi-solid medium and to induce tumor formation in nude mice, and morphological changes (flattening). As part of the strategy to understand the anti-tumor action of GC, differentially represented proteins, associated with the phenotypic reversion displayed by ST1 cells have been isolated through mRNA-based blind cDNA cloning and, more recently, by two dimensional electrophoresis (2D-PAGE). The sequences isolated by these two methodologies are potential targets for functional analysis. In the present study, we aimed at a) identifying genes which are differentially expressed during the GC-induced phenotypic reversion of ST1 cells, using the proteomic approach (2D-PAGE and mass spectrometry) and b) determining the functional role of a gene (Cyclin G) which had previously being identified as being induced during GC-treatment of the ST1 cells. The analytical methodology used relied on the comparison of sets of 2D nuclear protein profiles of ST1 cells, maintained in the absence (control) and in the presence of GC for 5 and 24h. After image analysis and visual validation, 33 polypeptides were considered as differentially represented 5 h after treatment, 16 of which were also differentially represented after 24 h. Six of those polypeptides were identified by peptide mass fingerprinting (PMF). Evidence obtained by Western blot analysis indicates that one of those polypeptides, Annexin 2 (ANX2), has its sub-cellular location modulated by GC-treatment of ST1 cells. The role of Cyclin G in the tumoral to normal phenotypic reversion induced by GC in ST1 cells was analyzed by over-expression of Cyclin G using a retroviral system and evaluation of the effects of this over-expression over ST1 cells response to GC, by growth curve and flow cytometry assays. The data suggest that Cyclin G over-expression leads to a more intense and prolonged effect of GC on ST1 cells.

Radium- und Radon-Isotopen-Untersuchungen als Hilfsmittel für die Aquiferdiagnose unter besonderer Berücksichtigung der geochemischen und hydrochemischen Verhältnisse im Grundwasserleiter

Zielsetzung der Arbeit war vorrangig die Entwicklung bzw. Weiterentwicklung von Methoden zur verbesserten Interpretation der hydraulischen Gegebenheiten in einem Grundwasserleiter mit Hilfe der Bestimmung von Radiumisotopenverhältnissen im Verlauf von Pumpversuchen. Daneben wurden 222Rn/226Ra-Verhältnisse betrachtet und interpretiert. Des Weiteren wurden Gesteinsuntersuchungen durchgeführt sowie Untersuchungen an Gesteinsoberflächen (Sekundärminerale) um Aussagen über das Löslichkeitsverhalten bzw. die Mobilität von Radium im Grundwasser zu erhalten. Die Grundwasser- und Gesteinsproben stammten vor allem aus dem Umfeld der kontinentalen Tiefbohrung (KTB Oberpfalz) sowie aus der Vorbohrung zur KTB, aber auch aus benachbarten Bundesländern und der Tschechischen Republik (Egergraben). Im Ergebnis zeigte sich, dass sich die Untersuchung der Isotopenverhältnisse zur vertiefenden Aquiferdiagnose gut eignet. Die geochemischen Untersuchungen erlaubten einen Einblick in das Löslichkeitsverhalten der untersuchten Radionuklide.

In the present study, we aimed at a) identifying genes which are differentially expressed during the GC-induced phenotypic reversion of ST1 cells, using the proteomic approach (2D-PAGE and mass spectrometry) and b) determining the functional role of a gene (Cyclin G) which had previously being identified as being induced during GC-treatment of the ST1 cells. The analytical methodology used relied on the comparison of sets of 2D nuclear protein profiles of ST1 cells, maintained in the absence (control) and in the presence of GC for 5 and 24h. After image analysis and visual validation, 33 polypeptides were considered as differentially represented 5 h after treatment, 16 of which were also differentially represented after 24 h. Six of those polypeptides were identified by peptide mass fingerprinting (PMF). Evidence obtained by Western blot analysis indicates that one of those polypeptides, Annexin 2 (ANX2), has its sub-cellular location modulated by GC-treatment of ST1 cells. The role of Cyclin G in the tumoral to normal phenotypic reversion induced by GC in ST1 cells was analyzed by over-expression of Cyclin G using a retroviral system and evaluation of the effects of this over-expression over ST1 cells response to GC, by growth curve and flow cytometry assays. The data suggest that Cyclin G over-expression leads to a more intense and prolonged effect of GC on ST1 cells.

Dendritic Cells in Head and Neck Cancer Microenvironment : From Mechanisms to Biomarkers / Les cellules dendritiques dans le micro-environnement tumoral des cancers ORL : des mécanismes aux biomarqueurs

L’objectif de ce travail était de comprendre l’état moléculaire des cellules dendritiques (CD) dans le microenvironnement tumoral. En intégrant l’analyse de tumeurs humaines par cytométrie en flux, de transcriptome, de secretome tumoral et l’analyse d’une base de données d’interaction CD-lymphocyte T générées in vitro, j’ai obtenus 2 résultats majeurs. Tout d’abord, nous proposons une nouvelle classification de CD activées humaines, qui sont soit « secrétantes », c’est-à-dire spécialisées dans la production de cytokines et chemokines, soit « aidantes » c’est-à-dire spécialisées dans l’induction de la sécrétion de nombreuses cytokines T helper après co-culture. Les CD infiltrant les tumeurs ORL inflammées correspondaient au type « sécrétantes ». Au-delà du nouveau concept biologique, cette classification est base théorique importante pour l’immunothérapie à base d’adjuvants. Deuxièmement, nous avons montré que l’inflammation tumorale n’était pas un facteur pronostic majeur des cancers ORL, mais que MMP2 et l’effraction extra-capsulaire étaient des facteurs pronostiques indépendants de la survie liée à la maladie. Nous avons pu classer les patients en 4 niveaux de risque et montré qu’ils avaient des chances équivalentes de réponse à l’immunothérapie. Nos données sont une base pour un essai clinique dirigé par biomarqueur, proposant de la chimiothérapie ou de l’immunothérapie néoadjuvantes, dans le but de diminuer le pourcentage de patients présentant des récidives sévères et précoces / The objective of the thesis was to decipher the molecular state of tumor infiltrating dendritic cell (DC) and their relation to the tumor microenvironment. By combining the analysis of human tumor samples by flow cytometry and RNA sequencing, of tumor secretome and of a large dataset of in vitro DC-Tcell interactions I obtained 2 main findings. First, we reported a novel classification of human activated DC, that are either “secretory” that is specialized in secreting cytokines and chemokines, or “helper” that is specialized at inducing the secretion of a broad range of T helper cytokines after cell co-culture. DC infiltrating inflamed human head and neck cancer matched the “secretory” phenotypic and transcriptomic signatures. Beyond this novel biological concept, this classification is of importance as a theoretical basis for adjuvant-based immunotherapy. Secondly, we showed that tumor inflammation was not the main prognostic factor for oral cavity cancer (OCC) patients, but that MMP2 and the presence of extra-nodal extension were independent predictors of reduced disease-specific survival. We could stratify OCC into 4 prognostic groups and showed that they had similar expected rates of response to immunotherapy. Our data may serve to design a biomarker-driven clinical trial proposing neoadjuvant chemotherapy or immunotherapy to high-risk patients, with the goal of reducing the percentage of OCC patients that will present with early and severe recurrences.
