The subject of this diploma thesis is a design documentation of multi-functional building. The building has a basement and four above-ground floors. The basement is served as an APS garage. The ground floor contains commercial premises as a bistro, copy centre and hairdresser's. The first floor is used for offices. The third and the fourth floors contain two maisonette with roof decks. Structural system is masonry systém of lightweight concrete block Ytong and RC’s elements. A roof is design as warm flat roof with waterproof membrane of m-PVC foil. Dominant elements are a ventilated cladding Cembrit and glass facade oriented on south-east.

L'explosion démographique a conduit à une augmentation intensive des contaminants environnementaux issus de différentes émissions anthropiques, par exemple, les hydrocarbures de pétrole, les pesticides, les biphényles polychlorés (BPC) ainsi que d’autres déchets ménagers tels que les détergents utilisés comme nettoyants ménagers. Du fait de leur persistance et du point de vue toxicologique, les résidus de ces contaminants organiques, qui peuvent être détectés dans les différentes matrices environnementales (eau, sédiments, air et biotes), parfois à très faible concentration (micropolluants), ont conquis l'attention du public. Cette étude vise à déterminer les niveaux de concentration de contaminants sélectionnés dans les sédiments de surface et à identifier la source de ces produits chimiques dans le ruisseau Cortiou. Les résultats ont montré une forte contamination par les hydrocarbures dans la zone étudiée. De plus, ces résultats ont probablement reflété le fait que les processus de biodégradation se sont produits parallèlement à une répartition chronique des charges d’épuration non traitées. Ces polluants organiques ont également confirmé un effet néfaste sur le biote marin et plus particulièrement sur les communautés benthiques pour certaines stations / Demographic explosion has led to increase intensively environmental contaminants issued from different anthropogenic release e.g. petroleum hydrocarbons, pesticides, polychlorinated biphenyls (PCBs) and also other domestic waste such as detergents as household cleaner. Due to their persistence and the toxicological point of view, the remnants of these organic contaminants, which can be detected in the different environmental matrices (water, sediment, air and biota), sometimes with very low concentration (micropollutants), has gained the public attention. This study aims to determine the concentration levels of selected contaminants in surficial sediments and identifying the source of these chemicals in Cortiou Creek. The results showed high contamination by hydrocarbons in the studied area. Additionally, these results probably reflected that biodegradation processes occurred concomitantly with a chronic apportionment of untreated sewage loading. These organic pollutants also confirmed an adverse effect for marine biota and more particularly for benthic communities for some stations

Neue Methoden und Anwendungen des Thermischen Spritzens

Die Habilitation befasst sich mit neuen Verfahren und Anwendungen des Thermischen Spritzens, beleuchtet anhand einer internationalen Umfrage den Forschungsbedarf der Branche und liefert zahlreiche Lösungen, die im Rahmen von grundlagenorientierten und industrienahen Forschungsvorhaben erarbeitet wurden. Der Fokus der Arbeit liegt auf der Verbesserung individueller Arbeitsschritte der Prozesskette des Thermischen Spritzens, wobei Ergebnisse aus den Bereichen Werkstoffentwicklung, Prozessoptimierung, Qualitätssicherung und Nachbearbeitung zusammengeführt und durch konkrete Anwendungsbeispiele hinterlegt werden. Im Detail werden die Aspekte Herstellung leistungsfähiger und preiswerter Spritzzusätze (Wasserverdüsung von Metallpulvern, Hochenergiekugelmahlen, Agglomerieren und Sintern sowie Fülldrahtherstellung), die Verbesserung der Prozessführung (numerisch optimierte Spritzbrenner und automatisierbare Online-Prozessdiagnostikmethoden) und die Steigerung der Leistungsfähigkeit der Beschichtungen durch mechanische Nachbearbeitung sowie Versieglung behandelt. Anwendungsbezogen werden das Beschichten von Hochleistungspolymeren und CFC-Leichtbaustrukturen untersucht. Um Anknüpfungspunkte für weiterführende Forschungsarbeiten zu schaffen, schließt die Arbeit mit der Darstellung von Entwicklungstrends und zeigt Arbeitsgebiete auf, die perspektivisch von thermisch gespritzten Schichten profitieren können.

Evaluation of particle release during cleaning of coated surfaces with pulsed Nd:YAG laser

The released particles during laser-based surface ablation of an epoxy resin coating were investigated for their number, size, mass and chemical composition. The laser used was a pulsed Nd:YAG laser with a maximum average power of 91 W. During the experiments, the applied laser power was investigated as a varying parameter. With the highest power, a maximum particle count of 4.3*108 particles/cm³ was measured, which corresponds to a mass of approximately 55 mg/m³. With decreasing laser power, the particle loading of the aerosol decreases. The particle sizes are bimodal distributed between 10 nm and approximately 3 μm. The first peak is at 34 nm and the second peak is at 630 nm. In terms of number, the nanoparticles dominate the distribution. During the analysis of the particles, fractal and spherical particles were determined, whereby the fractal particles are mainly in the nanometre range and the spherical particles mostly in the μm range. The chemical analysis of the particles showed that they consist of carbon and hydrogen.

Development-induced displacement and resettlement : a narrative inquiry into the experiences of community members physically displaced from a farm to a town in Mpumalanga, South Africa.

Although expected to be beneficial to society, development projects, such as dam construction and mining, often result in the physical displacement of marginalized groups. This is known as Development-Induced Displacement and Resettlement (DIDR) and often has far-reaching negative consequences for the affected peoples (APS). These include further marginalization, increased poverty, identity deprivation and socio-psychological stress. While decades of knowledge inform theory and practice on the mitigation of these consequences, the psycho-socio-cultural (PSC) impoverishment that may result from DIDR is often left unaddressed. Furthermore, qualitative knowledge of the experiences of people affected by DIDR is lacking. This research study aimed to produce an in-depth understanding of how DIDR affects the PSC well-being and identity construction of APS. The research took the form of a qualitative case study, whereby a social constructionist, narrative method of inquiry was used to analyse and gather data. This involved undertaking in-depth, individual interviews with a small sample of APS who had been resettled from a farm to a town in Mpumalanga by a mining house. Briefly, it was found that, contrary to the prevalent finding that DIDR causes further impoverishment, the resettlement actually resulted in development for the APS and as such enhanced their PSC well-being and contributed positively to their identity construction. However, the complexity inherent in resettlement was highlighted, as the participants revealed that the resettlement had not come without some socio-economic costs. It is believed that the findings from this study will complement existing knowledge on DIDR, and inform the design and implementation of Relocation Action Plans (RAPs) that better mitigate the negative PSC effects of DIDR.

Agencification in the Australian Public Service: the case of Centrelink

Agencification-the creation of autonomous agencies within the public service-has been occurring in many jurisdictions. It has usually had a rationale of improving the way in which government works. Generally, agencies are expected to provide more flexible, performance-oriented, responsive public services. The purpose of this work is to examine a particular example of agencification in the Australian Public Service (APS) and to compare it analytically with similar occurrences elsewhere. Specifically, it will examine the splitting of the former Department of Social Security (DSS) into two separate organisations, a policy department and a service delivery agency operating under a purchaser-provider arrangement, Centrelink. It will do this in the context of theories of agencification and of practical experience of agencification elsewhere. It will analyse why agencification has happened in this case and what the experience has shown, focusing on the role, governance, accountability and prospects for the new arrangements. This, the most prominent and substantial case of agencification in the Australian government, will be compared with the agencification experience reported in other jurisdictions-the United Kingdom and New Zealand. It will address why Centrelink came about, what the outcome has been of the change in institutional arrangements, and what the likely future is of the Centrelink arrangements. It will show that, when examined closely, the mechanisms bringing about agencification have been diverse. However, there are parallels in the experience. This leads to a conclusion that the current Centrelink arrangements are not stable in the long term, and some aspects-such as the purchaser-provider arrangement - should be set aside.

Avancerade planeringssystem som beslutsstöd vid produktions- och distributionsplanering / Decision support in production and distribution planning using advanced planning systems

<p>Logistik och Supply Chain Management som koncept har fått en ökad uppmärksamhet de senaste åren. Ett föråldrat synsätt på organisationen som en samling vertikala enheter som fungerar autonomt byts ut mot en vision om en flödesorganisation där ett helhetsgrepp om verksamheten och djupare samarbeten med leverantörer och kunder står i fokus. Ökade krav på konkurrenskraft genom effektivisering ställer nya ökade krav på en fungerande planering och styrning av varuflöden och där kan så kallade avancerade planeringssystem (APS) fungera som ett stöd och hjälpmedel. APS för produktions- och distributionsplanering är fortfarande ett nytt och relativt oprövat beslutsstödssystem. De senaste åren har företag som utvecklar affärssystem även börjat utveckla och sälja så kallade APS-moduler som genom sofistikerade matematiska algoritmer och optimeringsfunktionalitet kan stödja styrning och planering av komplexa system såsom försörjningskedjor. Under 2004 genom fördes ett antal strukturförändringar inom utsädesverksamheten hos Svenska Lantmännen, vilket medförde att två produktionsanläggningar och två logistikcenter avvecklades. Bakgrunden till dessa förändringar var överkapacitet och dålig lönsamhet. APS-modulen Movex Supply Chain Planner (SCP) implementerades samtidigt som stöd för huvudplanering för produktgruppen utsäde och planeringen lyftes upp på central nivå. Syftet med denna studie är att utvärdera vilka effekter strukturförändringar i försörjningskedjan och SCP som beslutsstöd vid huvudplanering har haft på totalkostnad och flöden i produktion och distribution. Genom intervjuer, observationer och datainsamling via Lantmännens affärssystem har en analys av totalkostnadsförändringar och förändringar på flöden i distribution och produktion genomförts. Resultaten av studien visar att Svenska Lantmännens utsädesverksamhet har effektiviserats med avseende på totalkostnad genom strukturförändringar och ett avancerat planeringssystem som stöd för en central huvudplanering. Totalkostnaderna för verksamheten har sjunkit med 11,8 miljoner kronor per år vilket motsvarar knappt 13 % samtidigt som försäljningskvantiteten har ökat något vilket ger en totalkostnadsminskning per fakturerad kvantitet på 15 %, eller 133 kronor per ton. Implementeringen av SCP och de investeringar som var nödvändiga vid strukturförändringarna har haft en återbetalningstid på drygt en månad. Lagernivåer i både produktion och logistikcenter har minskat med knappt 50 % vilket tyder på att ett snabbare genomflöde nu har realiserats och att SCP och central styrning har bidragit till denna förändring. Den potential som har identifierats i det nya planeringssystem och beslutsstödssystem som SCP utgör kan, om rätt förutsättningar ges, innebära att ytterligare effektivisering kan åstadkommas i utsädesverksamheten. During the least few years, companies that sell enterprise resource planning systems, have started developing and implementing APS-modules, which by the aid of sophisticated mathematical algorithms and optimization functionality, supports planning of complex systems such as supply chains. In 2004 structural changes were carried out in the seed production network that resulted in the termination of two production units and two central warehouses at the Swedish Farmers Supply and Crop Marketing Association. The background for these changes was overcapacity and therefore poor profitability. The APS Movex Supply Chain Planner (SCP) was implemented in the same time to support master planning for seed in a new centralized planning function. The purpose of this study is to evaluate the effects on total costs and the flow of goods after the structural changes and the implementation of the advanced planning system at the centralised master planning function. A total cost analysis and a flow analysis have been carried out regarding the seed production and distribution. Data have been collected through interviews, observations and the ERP database. The results of the study show that the structural changes and the implementation of the APS have streamlined the production and distribution network regarding the flow of goods, while the total costs have been reduced. Total costs have decreased by SEK 11,8 million per year, or 13 %, while at the same time the quantity of sold units have increased. This results in a total reduction of cost by some 15 %. The investments regarding the structural changes and the implementation of the SCP of have been paid off in just over one month. Furthermore, inventory levels in production facilities and warehouses have been reduced by almost 50 %. This study indicates that the SCP together with the centralized master planning function has contributed to these changes. Nevertheless, a further potential in the use of the SCP has been identified. There are opportunities to even further streamline the seed production network. The conclusions that can be made from this study is that positive effects on throughput and inventory levels and thus transportation, production and inventory cost has been realized with the aid of an advanced planning systems. However, a prerequisite is that the organisation employing the APS is operated effectively.</p>

Pteridine dependent hydroxylases as autoantigens in autoimmune polyendocrine syndrome type 1

<p>Autoimmune polyendocrine syndrome type I (APS) is a monogenous, recessively inherited disease characterised by endocrine and non-endocrine autoimmune manifestations. One fifth of APS I patients suffer from periodic intestinal dysfunction with varying degrees of malabsorbtion, steatorrhea and constipation. Alopecia areata is found in one third of APS I patients. By immunoscreening human cDNA libraries derived from normal human duodenum and scalp with APS I sera, we identified tryptophan hydroxylase (TPH) as an intestinal autoantigen and tyrosine hydroxylase (TH) as a dermal autoantigen. Forty-eight percent (38/80) of the APS I patients had TPH antibodies (Ab) and 44% (41/94) showed TH immunoreactivity. No reactivity against TPH or TH was seen in healthy controls. TPH-Abs showed a statistically significant correlation with gastrointestinal dysfunction (p<0.0001) and TH-Abs were significantly correlated to alopecia (p=0.02). TPH-Ab positive APS I sera specifically immunostained TPH containing enterochromaffin cells in normal duodenal mucosa. In affected mucosa a depletion of the TPH containing EC cells was seen. In enzyme inhibition experiments TPH and TH activity <i>in vitro</i> was reduced by adding APS I sera. TPH and TH together with phenylalanine hydroxylase (PAH) constitute the group of pteridine dependent hydroxylases. These are highly homologous enzymes involved in the biosynthesis of neurotransmitters. Immunoprecipitation of PAH expressed <i>in vitro</i> showed that 27% (25/94) of APS I patients had antibodies reacting with PAH, but no associations with clinical manifestations was observed. An immunocompetition assay showed that the PAH reactivity reflects a cross-reactivity with TPH.</p><p>In conclusion, we have identified TPH and TH as intestinal and dermal autoantigens in APS I, coupled to gastrointestinal dysfunction and alopecia. We have also demonstrated immunoreactivity against PAH in APS I patient sera reflecting a cross-reactivity with TPH.</p>
