Removal of emerging contaminants from water using green adsorbents

Water availability is presently under risk owing to the increased discharge of pollutants from both industrial and residential properties. A distinct category of pollutants known as "emerging contaminants" (ECs), whose hazards were either unknown before they were noticed, e.g., antibiotics, dyes, PFAS, etc. Most of the ECs are unregulated and pose a threat to aquatic and human life at even low doses. Our water treatment facilities are not designed to efficiently eliminate these toxic substances. Therefore, we need an economical tertiary treatment approach. Adsorptionis a sustainable, cost effective and simple technique, making it a viable technique for pollutants elimination on a worldwide scale. The removal of these ECs has been made possible by several commercially available adsorbents, however most of them are expensive. Adsorbents fabrication using, agricultural wastes is an effective waste management technique that helps reduce greenhouse gas emissions via carbon sequestration. The adsorption capability of adsorbents can be enhanced by further modification of its properties. This research study focuses on conversion of biomass into environmentally friendly adsorbents including biochar and nanocellulose aerogel. In the first study, a natural mineral dolomite (CaMg(CO3)2) modified biochar was fabricated from rice husk and used to remove anionic reactive dyes, Remazol Brilliant Blue (RBB) and Reactive Black 5 (RB-5) from synthetic wastewater. In the second study, a sustainable aminated/TEMPO cellulose nanofiber (Am/TEMPO-CNF) aerogel was fabricated and used to treat oxytetracycline (OTC) and chloramphenicol (CAP) contaminated water. The physiochemical properties of all adsorbents were studied using FTIR, SEM, TGA, elemental analyzer and N2 adsorption-desorption isothermal analyses. The contaminants were quantified using Ultraviolet–visible spectroscopy (UV-Vis) before and after the experiments. Kinetics, isothermal and thermodynamics modeling was applied to analyze the adsorption behavior and mechanism. In the third investigation, a novel In-situ UiO-66-NH2/TOCNF adsorbent was employed to remove the anionic azo dyes Orange II (ORII) and Congo Red (CR) from synthetic wastewater. We also looked at how pH, time, and initial concentration impacted CR and ORII adsorption. Research was conducted to assess the stability and adsorption potential under various situations by thermodynamics and regeneration.

Neste trabalho foram analisados os efeitos do BBG nos neurônios mioentéricos imunorreativos ao receptor P2X7 no íleo de ratos submetidos à isquemia e reperfusão (I/R). A isquemia intestinal foi obtida pela oclusão dos vasos ileais por 45 minutos, com reperfusão de 0 hora (h), 24 h e 14 dias. O BBG foi aplicado nos grupos I/R 24 h e I/R 14 dias nas dosagens de 50 e 100 mg/Kg. O grupo I/R 0 h é o grupo sem reperfusão. Os tecidos foram preparados para análises de duplas marcações, western blotting, histoquímica da mieloperoxidase, histologia e motilidade intestinal. Os resultados demonstraram a presença do receptor P2X7 nos neurônios mioentéricos. Houve uma diminuição da densidade e da área do perfil dos neurônios mioentéricos nos grupos I/R e nos grupos com o BBG houve uma recuperação dos neurônios. Nos grupos I/R houve aumento na expressão do receptor P2X7 e no número de neutrófilos e diminuição da motilidade. Os resultados sugerem que a I/R afetou os neurônios mioentéricos e que o BBG possa ter atenuado os efeitos da I/R, demonstrando uma provável neuroproteção. / We analyzed the effects of BBG antagonist on the P2X7 receptor and rats ileum enteric neurons subjected to I/R. Intestinal ischemia was obtained by ileal vessels obstruction for 45 minutes, followed by reperfusion of 0 h, 24 h and 14 days. The BBG was applied in I/R 24 h and 14 days groups in dosages of 50 and 100 mg/kg. The I/R 0 h is the group without reperfusion. Tissues were prepared by double labeling, western blotting, myeloperoxidase reaction, histology and intestinal motility analyzes. Our results demonstrated the presence of the P2X7 receptor in myenteric neurons. There was a decrease in density and in the area of the cell body profile of the myenteric neurons in the I/R groups and recovery in the BBG groups. In I/R groups there was an increase in the expression of P2X7 receptor and in the number of neutrophils and there was a decrease in intestinal motility and recover in the BBG groups. The results suggest that I/R affect the myenteric neurons and that the BBG may have attenuated the effects of ischemia, thus demonstrating a possible neuroprotection.

Evidências indicam uma interação na comunicação glia-neurônio. E sabido que a isquemia e reperfusão intestinal (I/R) são eventos clínicos graves. Este projeto visa estudar os efeitos de Brilliant Blue G (BBG) e de Probenecida (PB) nas células gliais entéricas (CGE). Os vasos ileais foram ocluídos por 45 min. Os períodos de reperfusão foram de 24 h, 14 e 28 dias.Os animais foram tratados com BBG, PB ou Salina. Os dados mostraram que o receptor P2X7 estava em CEG e neurônios, que há fenótipos diferentes de CGE e Panx-1 estava em CEG (GFAP). As densidades mostraram diminuição do número de células-IR ao receptor P2X7, Panx-1 e Hu no grupo I/R Salina, porém nos grupos tratados houve recuperação do número dessas células e com as CGE ocorreu um aumento no grupo I/R Salina. A área do perfil celular apresentou alterações em neurônios e CGE. Houve alterações na expressão proteica de P2X7 e Panx-1 e na atividade contrátil do intestino. O uso de BBG e de PB têm sido eficaz na recuperação de CGE e neurônios e podem ser alvos terapêuticos para doenças do trato gastrintestinal. / Evidence indicates an interaction in glia-neuron communication. It is known that intestinal ischemia and reperfusion (I/R) are serious clinical events. This project aims to study the effects of Brilliant Blue G (BBG) and Probenecida (PB) on enteric glial cells (CGE). The ileal vessels were occluded for 45 min. Reperfusion periods were 24 h, 14 and 28 days. Animals were treated with BBG, PB or Saline. The data showed that the P2X7 receptor was in CEG and neurons, that there are different phenotypes of CGE and Panx-1 was in CEG (GFAP). The densities showed a decrease in the number of cells-IR to the P2X7 receptor, Panx-1 and Hu in the Salina I/R group, but in the treated groups there was a recovery in the number of these cells and with the CGE there was an increase in the Salina I/R group. The cell profile area presented changes in neurons and CGE. There were changes in the protein expression of P2X7 and Panx-1 and in the contractile activity of the intestine. The use of BBG and PB has been effective in the recovery of CGE and neurons may be therapeutic targets for diseases of the gastrointestinal tract.

Avaliação do endotélio corneano de equinos após exposição ao corante azul brilhante 0,05% - estudo in vitro

Os corantes vitais têm sido utilizados em diversas áreas da oftalmologia para melhorar a visibilização de diferentes tecidos. Com o objetivo de avaliar os efeitos imediatos do corante Azul brilhante a 0,05% no endotélio da córnea de equinos, foram utilizadas 38 córneas de 19 cavalos, machos ou fêmeas, de diferentes idades e provenientes de abatedouro comercial licenciado. As córneas foram aleatoriamente divididas em dois grupos. Grupo1: o endotélio da córnea foi exposto a solução salina balanceada por 60 segundos. Grupo 2: o endotélio da córnea foi exposto ao corante Azul brilhante 0,05% (Opht-Blue, Ophthalmos, São Paulo, Brazil) por 60 segundos seguido por lavagem com solução salina balanceada (BSS, Ophthalmos, São Paulo, Brazil). As córneas então foram excisadas com trépano de 8mm e preparadas para avaliação com microscopia óptica (24 córneas) e microscopia eletrônica de varredura (14 córneas). As áreas de dano endotelial foram mensuradas. Devido ao aparecimento de resíduos não normais na utilização de comparação de médias via ANOVA, optou-se pelo modelo linear generalizado e, com 5% de significância, o teste Qui-quadrado demonstrou que o fator tratamento não difere do controle. Conclui-se que o corante Azul brilhante em concentração de 0,05% não provocou danos ao endotélio da córnea de equinos. / Vital dyes have been used in many areas of ophthalmology for improving visualization of different tissues. With the objective of evaluating the immediate effects of 0,05% brilliant blue on corneal endothelium of horses, 38 corneas of 19 horses, both male and female of different ages obtained from a licensed Brazilian commercial slaughterhouse were studied. Corneas were randomly divided into two groups. In group 1, corneal endothelium was perfused with BSS for 60 seconds. In group 2, corneal endothelium was perfused with 0.3 mL of brilliant blue 0.05% (Opht-Blue, Ophthalmos, São Paulo, Brazil) for 60 seconds followed by rinsing with a balanced salt solution (BSS, Ophthalmos, São Paulo, Brazil). The corneas were, subsequently, excised with an 8mm trephine and prepared to analyze posterior endothelial surface using a light microscope (24 corneas) and a scanning electron microscope (14 corneas). Due to non-normal residuals at ANOVA mean comparison, a generalized linear model was utilized at 5% level of significance. Qui-square test stated that treatment and control group were not different statistically. The 0.05 % brilliant blue did not damage equine corneal endothelium.

Jag har i mitt examensarbete gjort en kollektion kläder till den fiktiva Madame Brilliant, baserad på färgstarka personligheter som bl. a Anna Piaggi och Elsa Schiaparelli. Slår man upp ”glamourös” i en svensk-fransk ordbok finner man glamourös heta brilliant och det är just glamour mitt examensarbete handlar om, vardagsglamour.Jag har till Madame Brilliant skapat drömgarderoben. De plagg den excentriska kvinnan alltid drömmer om men aldrig hittar ute i butikerna; Färgstarka feminina kappor, klänningar och hattar att bära till vardags.Jag har i mitt arbete lagt stor vikt på färgkombinationerna, därför är stylingen och de små detaljerna mycket viktiga. Liksom färgerna i en tavla förhöjer varandra, förhöjer kläderna i min kollektion varandra och ger varandra karaktär.Färgsprakande kläder upplevs ofta som billiga, ofta för att de är gjorda i dåliga kvalitéer till tonåringar eller så är de tantiga och gjorda i linne för kulturtanten. Jag har därför gjort en kollektion till kvinnan som varken vill vara tonåring eller tant, kvinnan utan ålder, mina plagg har ingen ålder, bara personlighet. Jag har valt att använda mig av lyxiga material som man oftast inte använder till färgstarka kläder; siden, ull, sammet, viscosecrepe, fjädrar och hårfilt. Jag har även använt mig utav enklare material som bomull och syntetmaterial, men då just för att det ska ge en viss känsla åt plagget. Tygerna har jag gjort mer personliga genom applikationer och att blanda olika mönster och färger i plaggen.För att ge kollektionen en tidlös känsla har jag använt mig av klassiska feminina siluetter, timglasformen, prinsess-skärningen, fodralklänningen. Kläderna i min kollektion ska hålla i flera säsonger och vara bärarinnans favoritplagg. De plagg man aldrig slänger för att de är unika, tidlösa och gjorda i material av bra kvalité.Kollektionen är inspirerad av den kvinnliga kurviga kroppen, d v s kvinnor med midja , byst och rumpa. Mycket av modet ute i butikerna är gjorda till pojkflickan. Mitt mål är att framhäva kurviga kvinnors skönhet, men även personlighet hos var och en, att det inte handlar så mycket om kropp och ett vackert ansikte, med mina kläder spelar det ingen roll hur man ser ut, allt handlar om personlighet. Jag vill med min kollektion sprida färg, glamour, energi, humor och glädje. I have made my collection for the woman/girl without any age who likes clothes with a lot of personality and in exclusive fabrics. Development and characterization of a tunable ultrafast X-ray source via Inverse-Compton-Scattering

Ultrashort, nearly monochromatic hard X-ray pulses enrich the understanding of the dynamics and function of matter, e.g., the motion of atomic structures associated with ultrafast phase transitions, structural dynamics and (bio)chemical reactions. Inverse Compton backscattering of intense laser pulses from relativistic electrons not only allows for the generation of bright X-ray pulses which can be used in a pump-probe experiment, but also for the investigation of the electron beam dynamics at the interaction point. The focus of this PhD work lies on the detailed understanding of the kinematics during the interaction of the relativistic electron bunch and the laser pulse in order to quantify the influence of various experiment parameters on the emitted X-ray radiation. The experiment was conducted at the ELBE center for high power radiation sources using the ELBE superconducting linear accelerator and the DRACO Ti:sapphire laser system. The combination of both these state-of-the-art apparatuses guaranteed the control and stability of the interacting beam parameters throughout the measurement. The emitted X-ray spectra were detected with a pixelated detector of 1024 by 256 elements (each 26μm by 26μm) to achieve an unprecedented spatial and energy resolution for a full characterization of the emitted spectrum to reveal parameter influences and correlations of both interacting beams. In this work the influence of the electron beam energy, electron beam emittance, the laser bandwidth and the energy-angle-correlation on the spectra of the backscattered X-rays is quantified. A rigorous statistical analysis comparing experimental data to ab-initio 3D simulations enabled, e.g., the extraction of the angular distribution of electrons with 1.5% accuracy and, in total, provides predictive capability for the future high brightness hard X-ray source PHOENIX (Photon electron collider for Narrow bandwidth Intense X-rays) and potential all optical gamma-ray sources. The results will serve as a milestone and starting point for the scaling of the X-ray flux based on available interaction parameters of an ultrashort bright X-ray source at the ELBE center for high power radiation sources. The knowledge of the spatial and spectral distribution of photons from an inverse Compton scattering source is essential in designing future experiments as well as for tailoring the X-ray spectral properties to an experimental need. / Ultrakurze, quasi-monochromatische harte Röntgenpulse erweitern das Verständnis für die dynamischen Prozesse und funktionalen Zusammenhänge in Materie, beispielsweise die Dynamik in atomaren Strukturen bei ultraschnellen Phasenübergängen, Gitterbewegungen und (bio)chemischen Reaktionen. Compton-Rückstreuung erlaubt die Erzeugung der für ein pump-probe-Experiment benötigten intensiven Röntgenpulse und ermöglicht gleichzeitig einen Einblick in die komplexen kinematischen Prozesse während der Wechselwirkung von Elektronen und Photonen. Eine umfassende statistische Analyse, bei der ab-initio 3D Simulationen mit den experimentellen Daten verglichen und ausgewertet wurden, ermöglichte u.a. die Bestimmung der Elektronenstrahldivergenz mit einer Genauigkeit von 1.5% und erlaubt Vorhersagen zur zu erwartenden Strahlung der zukünftigen brillianten Röntgenquelle PHOENIX (Photon electron collider for Narrow bandwidth Intense X-rays) und potentiellen lasergetriebenen Gammastrahlungsquellen. Die Ergebnisse dienen als Fixpunkt für die Skalierung des erwarteten Photonenflusses der Röntgenquelle für die verfügbaren Ausgangsgrößen am Helmholtz-Zentrum Dresden - Rossendorf. Das Wissen um die räumliche und spektrale Verteilung der Röntgenstrahlung ist entscheidend für die Planung zukünftiger Experimente sowie zur Anpassung der Quelle an experimentelle Bedürfnisse.

Die so erfolgte vollständige Charakterisierung der Energie-Winkel-Beziehung erlaubt Rückschlüsse auf Parametereinflüsse und Korrelationen von Elektronen- und Laserstrahl. Eine umfassende statistische Analyse, bei der ab-initio 3D Simulationen mit den experimentellen Daten verglichen und ausgewertet wurden, ermöglichte u.a. die Bestimmung der Elektronenstrahldivergenz mit einer Genauigkeit von 1.5% und erlaubt Vorhersagen zur zu erwartenden Strahlung der zukünftigen brillianten Röntgenquelle PHOENIX (Photon electron collider for Narrow bandwidth Intense X-rays) und potentiellen lasergetriebenen Gammastrahlungsquellen. Die Ergebnisse dienen als Fixpunkt für die Skalierung des erwarteten Photonenflusses der Röntgenquelle für die verfügbaren Ausgangsgrößen am Helmholtz-Zentrum Dresden - Rossendorf. Das Wissen um die räumliche und spektrale Verteilung der Röntgenstrahlung ist entscheidend für die Planung zukünftiger Experimente sowie zur Anpassung der Quelle an experimentelle Bedürfnisse.

Avaliação do endotélio corneano de equinos após exposição ao corante azul brilhante 0,05% - estudo in vitro

Os corantes vitais têm sido utilizados em diversas áreas da oftalmologia para melhorar a visibilização de diferentes tecidos. Com o objetivo de avaliar os efeitos imediatos do corante Azul brilhante a 0,05% no endotélio da córnea de equinos, foram utilizadas 38 córneas de 19 cavalos, machos ou fêmeas, de diferentes idades e provenientes de abatedouro comercial licenciado. As córneas foram aleatoriamente divididas em dois grupos. Grupo1: o endotélio da córnea foi exposto a solução salina balanceada por 60 segundos. Grupo 2: o endotélio da córnea foi exposto ao corante Azul brilhante 0,05% (Opht-Blue, Ophthalmos, São Paulo, Brazil) por 60 segundos seguido por lavagem com solução salina balanceada (BSS, Ophthalmos, São Paulo, Brazil). As córneas então foram excisadas com trépano de 8mm e preparadas para avaliação com microscopia óptica (24 córneas) e microscopia eletrônica de varredura (14 córneas). As áreas de dano endotelial foram mensuradas. Devido ao aparecimento de resíduos não normais na utilização de comparação de médias via ANOVA, optou-se pelo modelo linear generalizado e, com 5% de significância, o teste Qui-quadrado demonstrou que o fator tratamento não difere do controle. Conclui-se que o corante Azul brilhante em concentração de 0,05% não provocou danos ao endotélio da córnea de equinos. / Vital dyes have been used in many areas of ophthalmology for improving visualization of different tissues. With the objective of evaluating the immediate effects of 0,05% brilliant blue on corneal endothelium of horses, 38 corneas of 19 horses, both male and female of different ages obtained from a licensed Brazilian commercial slaughterhouse were studied. Corneas were randomly divided into two groups. In group 1, corneal endothelium was perfused with BSS for 60 seconds. In group 2, corneal endothelium was perfused with 0.3 mL of brilliant blue 0.05% (Opht-Blue, Ophthalmos, São Paulo, Brazil) for 60 seconds followed by rinsing with a balanced salt solution (BSS, Ophthalmos, São Paulo, Brazil). The corneas were, subsequently, excised with an 8mm trephine and prepared to analyze posterior endothelial surface using a light microscope (24 corneas) and a scanning electron microscope (14 corneas). Due to non-normal residuals at ANOVA mean comparison, a generalized linear model was utilized at 5% level of significance. Qui-square test stated that treatment and control group were not different statistically. The 0.05 % brilliant blue did not damage equine corneal endothelium.
