Several diseases in dogs causing anemia, with the example of ehrlichiosis are thoroughly investigated. But in Brazil there are no studies to estimate the incidence of anemia in different species and so little, studies that assess the extent and causes related to the problem. Thus, it was developed this study to diagnose and assess the rates of anemia in male and female dogs of different breeds, ages and various conditions of health and nutritional management, but no clinical signs of anemia, and to evaluate the metabolic profile of these animals. We evaluated 148 dogs with anemia that was identified in 40% of the animals, most of then was normochromic normocitic (NNA) in type. It was concluded from the analyzed data that anemia is a significant occurrence, even in asymptomatic dogs. The nutritional deficiency especially of copper and iron, parasites and liver failure were not related to the occurrence of anemia among the animals studied. In this study we highlight the chronic infection with Ehrlichia canis and Anaplasma platys, and periodontal disease (PD) as chronic infectious/inflammatory diseases as likely causes of the NNA between the animals studied. The treatment of PD and evaluation of inflammatory markers would be useful for elucidation of the causes of anemia in the herd studied. / Instituto de Veterin?ria, Universidade Federal Rural do Rio de Janeiro, Serop?dica, RJ, 2009. Em c?es diversas enfermidades que cursam com anemia a exemplo da erliquiose s?o exaustivamente estudadas. Contudo, n?o existem no Brasil estudos para estimar a incid?ncia de anemia em diferentes esp?cies e, t?o pouco, estudos que avaliem a dimens?o e as causas relacionadas ao problema. Assim, desenvolveu-se o presente estudo visando diagnosticar e avaliar os ?ndices de anemia em c?es machos e f?meas, de diferentes ra?as, idades e condi??es diversas de manejo nutricional e sanit?rio, mas sem sinais cl?nicos de anemia, bem como avaliar o perfil metab?lico desses animais. Foram avaliados 148 c?es, sendo que anemia foi identificada em 40% dos animais, a maioria do tipo normoc?tica normocr?mica (ANN). Conclui-se pelos dados analisados que anemia tem ocorr?ncia significativa, mesmo em c?es assintom?ticos. A defici?ncia nutricional, especialmente de cobre e ferro, as parasitoses e insufici?ncia hep?tica n?o estiveram relacionadas ? ocorr?ncia de anemia entre os animais estudados. The objective of the present study was to evaluate the biological activity of commercial Dermatophagoides farinae and Blomia tropicalis allergenic extracts in dogs with atopic dermatitis and to identify the allergens present in these extracts by serological assays. For this purpose, 26 dogs were submitted to skin tests using commercial mite extracts (ALK-Abell? or FDA Allergenic) and skin reactions were evaluated after 15 min. Allergenic proteins present in the D. farinae and B. tropicalis extracts were identified by immunoblotting using sera from allergic animals and anti-dog IgE conjugate. The results showed that 0.95 and 1.68 BU/mL of the commercial D. farinae and B. tropicalis extracts (ALK-Abell?), respectively, are necessary to provoke a papule measuring 14 mm in diameter in susceptible animals. Immunoblotting analysis of antigens present in the D. farinae extract (FDA Allergenic) using sera from 10 allergic animals showed that 80% of the sera recognized a band of approximately 102 kDa, 80% two bands of 52 to 76 kDa, 70% two bands larger than 225 kDa, 50% one band of approximately 76 kDa, 50% one band of approximately 225 kDa, 40% one band of 31 to 38 kDa, and 20% one band of 12 to 17 kDa. Immunoblotting of antigens of the B. tropicalis extract (FDA Allergenic) showed that 50% of the sera recognized two bands of 52 to 76 kDa. These results demonstrate the importance of the two house dust mite species for the pathogenesis of canine atopic dermatitis in Brazil. The infection occurs by ingesting sporulated oocysts that were eliminated in DH feces, by bradyzoites when ingested by IH meat or by tachyzoites when occur vertical transmission. In this study, 529 canine blood samples were collected, 120 at the municipality of Itabuna, and 409 at the municipality of Ilh?us. All blood samples of Itabuna were consisted of animals from domiciled urban areas. From Ilh?us, 96 were domiciled from urban areas, 135 were urban but strayed dogs from Centre for Zoonosis Control (CZC) and 178 were domiciled and from rural area. The Indirect hemagglutination test was used to test the presence of anti-T. gondii and a questionnaire were applied to the domiciled dog owners to analyze risk factors associated to the infection. From all 529 dogs, 193 were positive (36.5%). At Ilh?us 37.7% (154/255) and at Itabuna 32.5% (39/81) of animals were positive. The risk factors associated to the infection were localization, rural dogs had higher chance than urban (p=0.001), the behavior, the strayed urban dogs had more chance than the urban domiciled dogs (p=0.01), home-made food and meat ingestion (p= 0.034 and 0.027 respectively), race, the undefined race dogs had more risk (p=0.0001) and age, were the animals under five years old had a higher risk to the infection (p=0.009). The presence of veterinary assistance was considered as a protection factor. The logistic regression confirmed the habitat and age of the animals as the main risk factor associsted with the infection. / Toxoplasma gondii ? um parasito intracelular obrigat?rio que tem como hospedeiro definitivo membros da fam?lia Felidae e animais homeot?rmicos como hospedeiros intermedi?rios. Neste estudo coletou-se sangue de 529 c?es, sendo 120 do munic?pio de Itabuna e 409 de Ilh?us. Todos os animais de Itabuna eram urbanos domiciliados. Foi realizado o Teste de Hemaglutina??o Indireta (HAI) para analisar a presen?a de anticorpos anti-T.gondii e um question?rio foi aplicado junto aos propriet?rios dos animais domiciliados para avalia??o dos principais fatores de risco associados ? infec??o. Do total de 529 c?es, 193 foram positivos (36,5%). No munic?pio de Ilh?us 37,7% (154/255) e em Itabuna 32,5% (39/81) foram positivos. A new real-time polymerase chain reaction method (qPCR) was patterned to target the citrate synthase gene (gltA) for the identification of A. platys in naturally infected dogs. The primers and probe were designed to amplify a fragment of 84 base pairs based on gltA gene sequences of A. platys available in GenBank. 186 blood samples of dogs from Itaguai microregion, Rio de Janeiro, Brazil, were tested by qPCR. The same samples were tested by cytology and nested polymerase chain reaction (nPCR, 16S rDNA) to determine the performance of qPCR front of these techniques. 17.20% of the samples tested positive by qPCR were significantly more than that detected by nPCR (13.98%). The qPCR technique was more specific than cytology, due to false-positive results obtained by optical microscopy. The prevalence of A. platys in dogs from Itaguai microregion was 14.4%. Dogs less than six months, infested by ticks, that spend the most of the time restrict to domestic environment and without shelter are factors associated with infection by this hemoparasite in dogs in the study area. During research, A. platys held in Cuba, 100 blood samples were collected from residents dogs in four cities located in the provinces of Havana and Mayabeque. When inspecting the animals, found ticks were collected, identified and carefully grouped, forming a total of 49 pools. DNA extracted from blood samples from dogs and ticks were subjected nPCR (16S rDNA). Positive samples in nPCR were also subjected to conventional PCR (gltA gene), and the products were sequenced. Only the species Rhipicephalus sanguineus sensu lato was found in Cuban dogs and 10.2% (n=5/49) of these ticks added to 16.0% (n=16/100) dogs were considered positive for A. platys. All sequences analyzed of the gltA and 16S rDNA genes, respectively, showed a 99-100% identity with sequences from A. platys reported in other countries. Phylogenetic analysis showed two clusters defined for the 16S rDNA gene and three clusters defined for the gltA gene. Based on the gltA gene, the deduced amino acid sequence showed two points of non-synonymous mutations at positions 88 and 168 compared to the reference sequence DQ525687. A preliminary study on the epidemiological aspects associated with infection with A. platys showed no statistical association with the variables studied (p> 0.05). This study also to report the first evidence of A. platys in both dogs and ticks in Cuba also presents for the first time the development of a new qPCR method that contributes to the advancement of research involving A. platys. The epidemiological study in Brazil allowed us to identify significant factors in the occurrence of canine anaplasmosis, while in Cuba, it can be concluded that more research is needed to assess what the deciding factors in the transmission and spread of A. platys in that country. / platys, e investigar a circula??o deste agente em c?es na microrregi?o de Itagua?, Rio de Janeiro, Brasil, e c?es e carrapatos em duas prov?ncias da ilha de Cuba, analisando aspectos epidemiol?gicos associados ? infec??o causada por esta bact?ria em c?es. Os oligoiniciadores e a sonda foram desenhados para amplificar um fragmento de 84 pares de base baseado em sequ?ncias do gene gltA de A. platys dispon?veis no GenBank. 186 amostras de sangue de c?es da microrregi?o de Itagua?, Rio de Janeiro, Brasil, foram testados pela qPCR. As mesmas amostras foram testadas pela citologia e rea??o em cadeia da polimerase nested (nPCR, 16S rDNA) para determinar o desempenho da qPCR frente ? essas t?cnicas. 17,20% das amostras testadas pela qPCR foram positivas, significativamente mais do que detectado pela nPCR (13,98%). C?es com menos de seis meses, infestados por carrapatos, que possam maior tempo restrito ao ambiente dom?stico e sem abrigo s?o fatores associados a infec??o por este hemoparasito em c?es na regi?o do estudo. Durante investiga??o de A. platys realizada em Cuba, 100 amostras de sangue foram coletadas de c?es residentes em quatro cidades localizadas nas prov?ncias de Habana e Mayabeque. Ao inspecionar os animais, carrapatos encontrados foram coletados, identificados e criteriosamente agrupados, formando um total de 49 pools. Amostras de DNA extra?das do sangue dos c?es e de carrapatos foram submetidas a nPCR (16S rDNA). Amostras positivas na nPCR foram tamb?m submetidas a PCR convencional (gene gltA), e os produtos foram sequenciados. Somente a esp?cie Rhipicephalus sanguineus sensu lato foi encontrada em c?es cubanos, e 10,2% (n=5/49) desses carrapatos somado aos 16,0% (n=16/100) de c?es foram considerados positivos para A. platys. Todas as sequ?ncias analisadas dos genes gltA e 16S rDNA, respectivamente, mostraram uma identidade de 99-100% com sequ?ncias de A. platys reportadas em outros pa?ses. A an?lise filogen?tica mostrou dois clusters definidos para o gene 16S rDNA e tr?s clusters definidos para o gene gltA. Com base no gene gltA, a sequ?ncia de amino?cidos deduzidos demonstrou dois pontos de muta??es n?o-sin?nimas nas posi??es 88 e 168 comparados com sequ?ncia de refer?ncia DQ525687. Um estudo preliminar sobre os aspectos epidemiol?gicos associados com a infec??o por A. platys demonstrou nenhuma associa??o estat?stica com as vari?veis avaliadas (p > 0,05). O presente estudo al?m de relatar a primeira evid?ncia de A. platys em ambos c?es e carrapatos em Cuba, tamb?m apresenta pela primeira vez o desenvolvimento de um novo m?todo de qPCR que contribui para o avan?o da pesquisa envolvendo A. platys. The affected animals show major changes as the increase in serum urea, creatinine and phosphorus. Currently, there is no specific treatment for nephropathic. Lactulose, a disaccharide formed by the reaction between fructose and galactose sacarol?ticas is assimilated by bacteria of the intestinal tract and these diminished competition by the population of urease-producing bacteria, limiting the production and absorption of ammonia and urea with consequent reduction in portal blood. Aiming to evaluate the effects of lactulose as adjuvant treatment of dogs with CRF were evaluated parameters indicative of liver function (enzymes alanine aminotransferase (ALT), aspartate aminotransferase (AST) and alkaline phosphatase (ALP) and renal (urea and creatinine) besides glucose and serum levels of Ca and P in dogs azot?micos not under normal handling and feeding, or not treated with lactulose orally for a period of 30 days, and serum urea and creatinine in dogs with CRF under different protocols therapeutic. feces showed normal color and odor, but diarrhea in animals treated with lactulose. animals as normal renal function, or not treated with lactulose showed no significant changes in blood glucose, serum activities of the enzymes ALT, AST and ALP, and values of urea and creatinine. animals treated with lactulose the phosphorus decreased progressively with a significant difference on days 21 and 30. biochemical analyzes for urea and creatinine nephropathic animals under different treatment protocols showed a reduction of these metabolites in animals of all groups , emphasizing the reduction of uremia and the effect of cetoan?logos and use of prebiotic lactulose in nephropathic dogs. / A Insufici?ncia Renal Cr?nica (IRC) ? caracterizada por uma crescente incapacidade dos rins em manter a homeostasia interna. As fezes apresentaram cor e odor normais, por?m diarr?icas nos animais tratados com lactulose. Os animais normais quanto ? fun??o renal, tratados ou n?o com lactulose n?o apresentaram altera??es significativas na glicemia, atividades s?ricas das enzimas ALT, AST e FA; e nos valores de ur?ia e creatinina.
