The results of this study revealed that it seems
possible for an orthodontist to establish an extra-skeletal
reference plane, if a specially designed analyzer is used.
It has been shown that by using this devise in his determination
of a base landmark, an orthodontist can have a high degree
of reliability within himself. It also appears likely
that the reference plane obtained in this manner by one
orthodontist is reproducible, to a high degree of reliability
by other orthodontists. A remarkably high reproducibility, as obtained if a group determination of a reference base was done. To the orthodontist, it means that there may
be a way of accurately producing a reliable reference
plane for static cephalocentric analysis. This could be

L’intestin humain contient une variété de microbes commensaux qui sont représentés par divers organismes appartenant aux trois domaines de la vie où les Eukarya sont essentiellement représentés par le règne des champignons. La levure commensale et opportuniste Candida albicans a été identifiée comme étant le champignon le plus commun dans l’intestin des humains sains. Des études récentes soutiennent que malgré leur faible abondance les levures du genre Candida peuvent altérer l'équilibre du microbiote et conduire à des dysbioses ou des pathologies récurrentes comme la maladie de Crohn et les colites ulcéreuses. Il a été démontré que le microbiote commensal joue un rôle essentiel dans la protection de l’intestin contre la colonisation par des bactéries pathogènes et des pathobiontes. Cependant, jusqu'à présent, on ignore si la prolifération ou la pathogénicité de C. albicans peuvent être contrôlées par d'autres microbiotes fécaux. Dans cette étude, nous avons démontré que le métabolome microbien de l'intestin humain exerce une activité antifongique contre C. albicans et d’autres levures qui résident au niveau intestinal. Ces métabolites inhibent plusieurs traits de virulence de C. albicans incluant la filamentation et l'invasion des cellules humaines. De plus, un crible génétique chez C. albicans a suggéré que TOR est la cible moléculaire de la ou des molécules antifongiques du métabolome microbien de l'intestin humain. / The human gut contains a variety of commensal microbes which are composed of diverse organisms that belong to all three domains of life with Eukarya primarily represented by fungi. The commensal / opportunistic yeast Candida albicans has been reported as the most common fungus in the gut of healthy humans. Recent evidences support that, this small fraction can alter the microbiota equilibrium leading to dysbiosis and diseases like inflammatory bowel diseases. It has been demonstrated that commensal microbiota plays a critical role in the protection of the gut against colonization by other bacterial pathogens and pathobionts. However, so far, whether C. albicans overgrowth or pathogenicity are controlled by other fecal microbiota is not known. In this study, we showed that the human microbial gut metabolome (GM) exerts an antifungal activity against different intestinal-resident yeasts including hyphal growth and the invasion of human enterocytes of C. albicans. Furthermore, a genetic screen in C. albicans suggested that TOR is the molecular target of the antifungal molecule(s) of the GM.

Effets de la dermaseptine-S 1 sur la croissance, la transformation, la formation de biofilms et l'expression de certains gènes de virulence de C. albicansBelmadani, Amine 25 July 2018 (has links)
C. albicans est un microorganisme opportuniste présent chez plus de 60% de la population. La pathogénicité de C. albicans est contrôlée par le système immunitaire de l’hôte, grâce entre autres aux peptides antimicrobiens (PAMs). Ces PAMs peuvent être de différentes origines. Notre étude a pour but d’évaluer l’effet d’un PAM d’amphibiens, la dermaseptine-S1 (DS1), sur la croissance, la transformation et l’expression de certains gènes de virulence de C. albicans. C. albicans (ATCC-SC5314) a été cultivé en présence ou en absence de la DS1 à 10, 50, et 100 μg/ml, pendant 24 h puis la croissance a été évaluée par un teste de numération cellulaire. Le passage de la forme levure à la forme hyphe a été évalué par microscopie optique et électronique à balayage après culture en présence ou en absence de la DS1 pendant 3 et 6 h à 37°C et ce en présence de 10% de sérum de veau foetal. L’effet de la DS1 sur la formation de biofilm a été évalué par un test MTT et par microscopie électronique à balayage. Enfin, l’expression de certains gènes protéase aspartique sécrétée (Saps 1, 2, 3, 9, 10 et Hwp1) a été évaluée par qRT-PCR. Nous avons montré que la DS1 réduit de façon significative la croissance de C. albicans. La DS1 inhibe le passage de C. albicans de sa forme blastospore à la forme hyphe, ainsi que la formation du biofilm. L’effet de la DS1 sur C. albicans pourrait passer par un contrôle de l’expression de plusieurs gènes impliqués dans la sécrétion d’enzyme protéolytique. En effet, la DS1 diminue de façon significative l’expression des gènes Saps 1, 2, 3, 9 et 10 ainsi que du gène Hwp1. L’ensemble de nos travaux démontre que la DS1 peut contrôler la pathogénicité de C. albicans. / Candida is an opportunistic microorganism present in more than 60% of the population. The pathogenicity of Candida is controlled by the host immune system, such as antimicrobial peptides (AMPs). These PAMs can be of different origins. Our study is to evaluate the effect of an amphibian-isolated antimicrobial peptide, dermaseptin-S1 (DS1) on the growth, transformation and the expression of certain C. albicans virulence genes. C. albicans (ATCC-SC5314) was cultured in the presence or the absence of DS1 at 10, 50, and 100 μg / ml for 24 h and then the growth of yeast was assessed by cell count. The transformation of yeast was evaluated at 3 and 6 h of culture under conditions conducive to transformation (37°C in the presence of 10% of serum) by optical and scanning electron microscopy. Biofilm formation was assessed using a collagen scaffold, after incubation for 30 min at 30°C, with MTT assay and scanning electron microscopy. Finally, the expression of some secreted aspartic protease genes (Saps 1, 2, 3, 9, 10 and Hwp1) was evaluated by qRT-PCR. The results showed that DS1 significantly reduces the growth of C. albicans. This effect is proportional to the concentration of the peptide. This peptide inhibits the passage of C. albicans from the blastospore to the hypha form. The effect of DS1 on C. albicans involves controlling the expression of several genes. Indeed, DS1 significantly decreases the expression of Sap 1, 2, 3, 9 and 10 genes as well as Hwp1 gene. All of our work demonstrates that DS1 can control the pathogenicity of C. albicans.

Étude de la perception et de l'adaptation à l'hypoxie chez la levure pathogène opportuniste Candida albicansBurgain, Anais 02 February 2024 (has links)
Candida albicans est une levure pathogène opportuniste responsable d’infections appelées candidoses. Ce type d’infection est particulièrement grave chez les personnes ayant un système immunitaire affaibli. Toutefois, peu de traitements sont disponibles et une augmentation des résistances font que les infections fongiques sont un problème de santé publique majeur. Au sein de l’hôte, cette levure doit s’adapter à l’environnement parfois hostile des différentes niches. Afin de persister chez l’être humain, ce micro-organisme doit être en mesure de percevoir les changements environnementaux et adapter son métabolisme en conséquence. L’adaptation aux nutriments disponibles ainsi que de la teneur en oxygène sont deux éléments importants pour le développement dans les différentes niches. La compréhension de l’adaptation de cette levure au sein de l’hôte pourra permettre l’identification de nouvelles cibles thérapeutiques. Nous avons effectué une analyse métabolomique à différents temps d’exposition à l’hypoxie dans le but de découvrir pour la première fois la réponse métabolomique de C. albicans lorsque cette levure est confrontée à une déplétion en oxygène. Nous avons observé un remodelage métabolique important en particulier concernant la glycolyse, la voie des pentoses phosphates et la paroi cellulaire. Le lipidome de C. albicans est également fortement affecté par l’hypoxie, en particulier les composants de la membrane plasmique tels que les phospholipides, les sphingolipides et l’ergostérol. Les données transcriptomiques corroborent les résultats observés lors de l’étude métabolomique. Enfin, nous avons testé la sensibilité de C. albicans à différents stress afin de confirmer l’altération physiologique provoquée par l’hypoxie comme suggérée par l’analyse du métabolome. Nous avons également réalisé un criblage de banques de mutants afin d’identifier des régulateurs de la flexibilité métabolique hypoxique. Nous avons identifié le rôle de Snf5, une sous-unité du complexe de remodelage de la chromatine SWI/SNF, permettant de coordonner la disponibilité en nutriments et en oxygène avec la réponse transcriptionnelle pour permettre l’adaptation aux changements environnementaux. Snf5 est nécessaire pour la réponse transcriptionnelle relative au commensalisme et aussi aux traits de virulence de C. albicans. Nous avons confirmé ces observations par des tests de colonisation intestinale dans un modèle murin et le mutant snf5 s’est révélé incapable de persister dans le tractus digestif des souris. Le mutant snf5 présente également une atténuation des traits de pathogénicité : il ne peut former des hyphes, ne cause pas de dommage aux cellules de l’hôte et il est avirulent dans un modèle d’infection des larves de Galleria mellonella. L’étude des interactions génétiques a permis d’identifier un lien entre SNF5 et le gène de l’adénylate cyclase CYR1. Ceci suggère que Snf5 et Cyr1 appartiennent à la même voie de signalisation afin de permettre la flexibilité métabolique hypoxique. Les travaux de cette thèse ont permis de connaitre la réponse métabolomique hypoxique chez C. albicans et apportent des connaissances fondamentales sur cette levure. Cette compréhension pourra par la suite servir à cibler les voies métaboliques importantes pour la survie au sein de l’hôte et ainsi permettre le développement de nouveaux antifongiques. Nous avons également identifié le rôle de Snf5 pour la flexibilité métabolique hypoxique. L’absence de Snf5 provoque un défaut majeur de virulence chez C. albicans, ceci renforce l’idée de cibler les voies métaboliques pour lutter contre cette levure. Ainsi Snf5 ou un autre membre du complexe SWI/SNF pourrait être une cible thérapeutique pour le développement de nouveaux traitements antifongiques.

Previous issue date: 2018-06-08 / Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES) / A estomatite protética é uma das afecções mais comuns e recorrentes em pacientes portadores de próteses totais. O fungo do gênero Candida, promotor dessa patologia, além de resistente se torna ainda mais complexo de ser combatido devido à dificuldade da ação de fármacos tópicos que só conseguem permanecer por um curto período no local da infecção, em virtude da dinâmica da cavidade bucal. O processo de Solution Blow Spinning permite a obtenção de fibras ultrafinas que podem ser aplicadas em vastas áreas, inclusive na bioengenharia. Uma das aplicabilidades das fibras ultrafinas é sua utilização para liberação controlada de fármacos de forma eficiente e duradoura. Dessa forma, o intuito do presente trabalho foi incorporar Cinamaldeído (CA), composto que possui propriedades antimicrobianas, a mantas de Poli(ácido lático) e Poli(etileno glicol) (PLA/PEG) e avaliá-las quanto à produção e caracterização por, Microscopia eletrônica de varredura (MEV), Termogravimetria (TGA), Calorimetria Exploratória Diferencial (DSC), Espectroscopia de infravermelho por transformada de Fourier (FTIR), Espectroscopia no ultravioleta visível (UV/vis), ensaios mecânicos e ação antifúngica. Para realização dos experimentos, foram fiadas as seguintes mantas: PLA, PLA/PEG e PLA/PEG 23,8% CA. As micrografias obtidas por MEV mostraram, que os diâmetros das fibras que não continham CA, apresentaram diâmetros semelhantes entre si, PLA (354±160 nm)a e PLA/PEG (428±250nm)a, sendo esses diâmetro menores dos que encontrados nas fibras de PLA/PEG 23,8% CA (749±370 nm)b . O ângulo de contato e tensão superficial não puderam ser verificados em virtude da proporção de polímeros nas blendas que apresentaram alta afinidade pelos solventes utilizados no teste. No ensaio de TGA, a curva de PLA/PEG com acréscimo de 23,8% CA exibiu uma maior estabilidade térmica. No teste de DSC o ponto de transição vítrea das mantas contendo 23,8% CA foi o que apresentou menor valor. A liberação de CA foi satisfatória ocorrendo até o 12° dia. No teste de ensaios mecânicos, o acréscimo de CA às mantas aumentaram significativamente o Módulo elástico (24,94±4,45) e a Tensão máxima de ruptura (0,99±0,16 MPa) com relação às mantas puras de PLA/PEG (18,74±3.41 MPa) and (0,85±0.09 MPa), esse acréscimo ainda promoveu redução estatisticamente significante (p˂ 0,05%) em mais de 50% nos biofilmes monotípicos de C. albicans e C. krusei e no multiespécie de C. albicans, C. krusei e C. glabrata. Mediante os resultados encontrados pode-se depreender que é possível se obter mantas de fibras ultrafinas de PLA/PEG contendo 23,8% de CA com propriedades antifúngicas e capacidade de liberação do agente antimicrobiano por cerca de 12 dias. / Denture stomatitis is one of the most common and recurrent conditions in patients with total dentures. The fungal of the genus Candida, the causer of this pathology, besides being resistant, becomes even more complex to be combated due to the difficulty of the action of topical drugs that can only remain for a short time at the site of infection due to the dynamic of the oral cavity. The Solution Blow Spinning process allows the production of ultrafine fibers that can be applied in large areas, including bioengineering. One of the applications of ultrafine fibers is their use for controlled release of drugs in an efficient and long-lasting manner. Thus, the aim of the present work was to incorporate Cinnamaldehyde (CA), a compound that has antimicrobial properties, to Poly (lactic acid) and Poly (ethylene glycol) blankets (PLA / PEG) and to evaluate them for the production and characterization by Scanning Electron Microscopy (SEM), Thermogravimetric (TGA), Differential Scanning Calorimetry (DSC), Fourier transform infrared spectroscopy (FTIR), Ultraviolet–visible (UV/vis) spectroscopy, mechanical properties and antifungal action. Antifungal activity was verified against C. albicans, C. krusey and C. glabrata by broth microdilution test, disk diffusion and anti-biofilm activity, in both multi-species and mono-species biofilms. For the experiment, three types of meshes were spun: pure PLA, PLA/PEG and PLA / PEG 23.8% CA. The micrographs obtained by SEM showed that the fibers that did not contain CA had similar diameters to each other and smaller than the fibers containing PLA / PEG 23, 8% CA. The contact angle and surface tension could not be measured by virtue of the proportion of polymers in the blends which showed high affinity for the solvents used in the test. In the TGA assay, the PLA/PEG curve with 23.8% CA increase exhibited a higher thermal stability while in the DSC test the glass transition point of the meshes containing 23.8% CA it was the one with the lowest value. The release of CA was satisfactory occurring until the 6th day. PLA membranes with fibres of diameter exhibited the lowest fibre diameter (354 ±160 nm)a followed by PLA/PEG (428±250nm)a and PLA/PEG/CA (749±370 nm)b. Addition of CA resulted in an increase in mechanical properties of the membranes from (24.94±4.85 MPa) the elastic modulus and (0.99±0.16 MPa) tensile strength in comparison to PLA/PEG (18.74±3.41 MPa) and (0,85±0.09 MPa). CA incorporation increased improved the thermal stability, with release of CA of 0.10 µg/mL over a 12 days period. The PLA/PEG CA membranes presented antifungal activities, showing reductions in more than 50% of the biofilm biomass, being statistically significant (p<0.05%) to the control group. Fibrous membranes of PLA/PEG/CA ultrathin fibres were produced by SBS that exhibited antifungal properties and release over a 12-day period.

No primeiro estudo, suspensões planctônicas de C. albicans (ATCC 90028) foram expostas a nove concentrações de curcumina (0,005; 0,01; 0,05; 0,1; 0,5; 1, 5, 10 e 20μM) e oito doses de luz (1,32; 2,64; 3,96; 5,28; 6,60; 13,20 e 26,4J/cm2). O efeito de diferentes tempos de pré-irradiação (PIT), a possível captação de curcumina pelas células fúngicas e participação do oxigênio singlete na fotoinativação também foram avaliados. Adicionalmente, a PDT mediada pela curcumina foi avaliada em biofilmes formados pela cepa de referência de C. albicans e sobre culturas celulares de macrófagos. Além disso, as características ópticas da solução de curcumina foram investigadas em função da dose de luz utilizada. Os resultados foram submetidos à análise descritiva, Análise de Variância (ANOVA) e Kruskal-Wallis (α= 5%). As suspensões planctônicas de C. albicans foram completamente inativadas com a utilização de 20μM de curcumina e 5,28J/cm2. Além disso, foi observada redução significativa na viabilidade celular dos biofilmes de C. albicans. O efeito fotodinâmico foi acentuado pela presença da curcumina durante a iluminação... / The aim of this investigation was to evaluate: 1. the Photodynamic Therapy (PDT) mediated by curcumin for in vitro inactivation of a reference strain of Candida albicans and its citotoxic effects against a macrofage cell line; 2. the effectiveness of PDT mediated by curcumin to inactivate clinical isolates of C. albicans, Candida tropicalis and Candida glabrata, both in planktonic and biofilm phases; 3. the photoinactivation of C. albicans in a murine model of oral candidiasis using different curcumin concentrations. In the first study, suspensions of C. albicans were treated with nine curcumin concentrations (0.005; 0.01; 0.05; 0.1; 0.5; 1, 5, 10 and 20μM) and exposed to LED light at different fluences (1.32; 2.64; 3.96; 5.28; 6.60; 13.20 and 26.4J/cm2). The effect of the pre-irradiation time (PIT) on PDT effectiveness, the uptake of curcumin by C. albicans cells and the possible involvement of singlet oxygen in the photodynamic action was also evaluated. In addition, curcumin-mediated PDT was assessed against biofilms. Similar protocols were tested on a macrophage cell line and the effect was evaluated by MTT and SEM analysis. The optical properties of curcumin were investigated as a function of illumination fluence. Data were analyzed with Analysis of Variance (ANOVA), and Kruskal-Wallis test (α= 5%). The results showed a statistically significant reduction in C. albicans viability after PDT (p<0.05), for both planktonic and biofilm cultures. Photodynamic effect was greatly increased with the presence of curcumin in the surrounding media and the PIT of 20 minutes improved PDT effectiveness. Although PDT was phototoxic to macrophages, the therapy was more effective in inactivating the yeast cell than the defense cell. The spectral changes showed a high photobleaching rate of curcumin. Microespectroscopia infravermelha por transformada de fourier : identificação e discriminação de cepas clínicas de Candida albicans e Candida glabrata /Cardoso, Michelle. January 2010 (has links)
Orientador: Luciane Dias de Oliveira / Banca: Célia Regina Gonçalves e Silva / Banca: Antonio Olavo Cardoso Jorge / Resumo: A técnica da espectroscopia infravermelha por transformada de Fourier (FT-IR) vem sendo largamente empregada como uma abordagem rápida e simples para a identificação de microrganismos, incluindo a do gênero Candida. A proposta deste estudo foi avaliar o emprego da microespectroscopia FT-IR na identificação e discriminação de 5 cepas clínicas de Candida albicans e 3 de Candida glabrata, as quais foram identificadas previamente por meio de métodos convencionais, e mais duas cepas ATCC de cada espécie. As amostras foram analisadas em triplicata, a partir de culturas independentes, por meio de filmes finos obtidos da suspensão em solução salina estéril a 0,85% da biomassa da colônia que ficou incubada por 24 horas em placas com ágar Saboraud Dextrose. Dez espectros com 32 varreduras foram obtidos na forma de ponto em 10 regiões selecionadas aleatoriamente nas amostras. A média foi obtida dos dez espectros de cada amostra para a análise estatística multivariada, dada pela análise de cluster. Além disso os espectros foram transformados em primeira derivada e três janelas espectrais foram determinadas (900-1250 cm-1, 1300-1600 cm-1, 2800-3000 cm-1). A representação dos resultados foi dada pela construção de um dendograma. Nesse, foi possível separar em dois grupos distintos as duas espécies de Candida estudadas. Dessa forma, pode-se concluir que a microespectroscopia FT-IR foi capaz de identificar e discriminar cepas clínicas de Candida albicans e de Candida glabrata, sendo um método promissor para identificação de leveduras / Abstract: The technique of infrared spectroscopy Fourier transform (FT-IR) has been widely used as a new approach for rapid identification and simple micro-organisms, including the genus Candida. The purpose of this study was to evaluate the use of FT-IR microspectroscopy for the identification and discrimination of 5 clinical strains of Candida albicans and Candida glabrata 3, which were previously identified by conventional methods, and two-standard strains of each species. The samples were analyzed in triplicate from independent cultures by means of thin films obtained from a suspension in sterile saline and 0.85% of the biomass of the colony that was incubated for 24 hours in Sabouraud dextrose agar plates. Ten spectra with 32 scans were obtained in 10 randomly selected regions in the samples.The average of ten spectra was obtained from each sample for the multivariate analysis, given by cluster analysis. In addition, three windows were determined spectral (900-1250 cm-1, 1300-1600 cm-1, 2800-3000 cm-1) and the spectra were transformed into first derivative. The representation of the results was given by the construction of a dendrogram. In this, we separated into two groups of two Candida species studied. Thus, one can conclude that the FT-IR microspectroscopy was able to identify and distinguish clinical isolates of Candida albicans and Candida glabrata is an important method for identification of yeasts / Mestre

Previous issue date: 2005 / Resumo: O objetivo deste trabalho foi avaliar a influência do método de polimerização e a presença da saliva humana na aderência de Candida spp. às superfícies de resinas acrílicas. Duzentas e cinqüenta e seis amostras retangulares (2,5 x 1,2 x 0,2 cm), confeccionadas com resinas polimerizadas por banho de água ou microondas foram avaliadas para rugosidade e energia livre de superfície e em seguida utilizadas para o ensaio de aderência de Candida spp. Para este propósito, as amostras foram aleatoriamente divididas em 8 grupos por resina, sendo quatro expostos durante 30 min a saliva humana. Em seguida, as amostras foram posicionadas verticalmente em tubos de plásticos estéreis contendo meio de cultura Sabouraud e uma entre quatro suspensões de Candida: Candida albicans, Candida tropicalis, Candida dubliniensis e Cândida glabrata (1 a 5 x 106 células / mL). A contagem das células aderidas foi realizada em microscópio óptico com 400 x de aumento. Os dados de rugosidade superficial e energia de superfície foram submetidos a ANOVA um fator e teste t. Não houve diferença estatística significante para rugosidade (p > 0,05), enquanto maiores valores de energia livre de superfície foram encontrados para a resina polimerizada por banho de água (p < 0,05). Para aderência de Cândida spp., os testes Wilcoxon-Mann-Whitney, Kruskal Wallis e Qui-quadrado demonstraram não haver influência dos valores de rugosidade e energia livre de superfície na aderência de Candida, enquanto houve uma diminuição geral na contagem de microrganismos em grupos expostos à saliva (p < 0,05). Concluiu-se que a saliva foi capaz de reduzir a aderência total de Candida, enquanto não houve correlação destes valores com rugosidade e energia de superfície / Abstract: The aim of this work was to evaluate the influence of polymerization methods and presence of human clarified saliva on Candida spp. adherence to acrylic resins surfaces. Two hundred and fifty six rectangular acrylic resins samples (2.5 x 1.2 x 0.2 cm) polymerized by water bath or microwave were evaluated for surface roughness and surface free energy and used in an adherence assay for Candida spp. For this purpose, acrylic samples were randomly divided into 8 groups for each resin, where 4 were exposed to saliva. For the adherence assay, the samples were placed vertically on test tubes containing yeast Sabouraud broth medium and one of the four Candida suspensions: Candida albicans, Candida tropicalis, Candida dubliniensis and Candida glabrata (1 to 5 x 106 cells / mL). Adhered yeasts were counted using an optic microscope at 400 x. Data from surface roughness and surface free energy were submitted to one-way ANOVA ant t test and Candida spp. adherence values were evaluated by the Wilcoxon-Mann-Whitney, Kruskal Wallis and Chi-square tests. No statistical significance was found for roughness (p > 0.05), while higher surface free energy values were found for water bath resin (p < 0.05). Roughness and surface free energy did not influence Candida adherence, while saliva generally decreased yeast counts (p < 0.05). It was concluded that polymerization methods did not interfere with adherence assay while saliva was capable of reducing Candida spp. adherence, while roughness and free energy did not influence the adherence rates / Doutorado / Protese Dental / Doutor em Clínica Odontológica
