Le CD40 est une glycoprotéine transmembranaire de type I, appartenant à la famille des TNFRs, exprimée à la surface des cellules immunitaires, hématopoïétiques, vasculaires, épithéliales, et d’autres types de cellules, y compris les cellules tumorales. Le CD40 ne possédant pas de domaine kinase, pour induire un signal il interagit directement ou indirectement avec des protéines adaptatrices telles que les TRAFs et les JAKs. L’interaction du CD40 avec son principal ligand, le CD154, joue un rôle primordial dans la régulation de la réponse immunitaire et le maintien de l’homéostasie. L’activation du CD40 à la surface des cellules B augmente leur capacité de présentation d’antigène, en plus d’induire la prolifération, la commutation isotypique et l’apoptose. Les patients souffrant de mutations au niveau du gène codant pour le CD40 ou de son ligand sont immunosupprimés et sensibles à des infections opportunistes. Des études ont montré que le CD40 comme d’autres membres de la famille des TNFRs est capable de former des homodimères. Plus récemment, on a montré que la formation du CD40 homodimère est le résultat de son engagement sur les cellules B. En plus, cette homodimérisation du CD40 est importante pour la phosphorylation de l’Akt. L’interaction CD40/CD154 peut avoir un rôle direct dans l’immunothérapie par l’induction de l’apoptose de certaines cellules cancéreuses ou un rôle indirect en activant les cellules présentatrices d’antigènes (CPA) afin d'augmenter l’efficacité de l’activation des cellules T cytotoxiques. Nos résultats montrent que l’induction de la mort cellulaire par le CD40 requiert la perméabilisation du lysosome, la libération de la cathepsine B, la présence de ROS et une interaction avec le TRAF6, cette mort cellulaire programmée est plus importante en présence de la forme monomérique du CD40, muté au niveau de la cystéine 238. Par ailleurs, l’homodimérisation du CD40 requerrait sa translocation vers les radeaux lipidiques et nécessiterait la présence des ROS. Cette homodimérisation du CD40 semble être importante pour l’activation des cellules B par le biais de l’induction de l’expression du CD23, CD69 et CD80. De plus, nos résultats montrent pour la première fois une implication du CD40 homodimère dans l’induction du CD23 par le biais du TLR4. Nos résultats soulignent l’importance du CD40 homodimère dans certaines voies de signalisation. Moreover, the homodimerization of CD40 requires its translocation to lipid rafts and the presence of ROS. This homodimerization is necessary for the CD40 B-cell activation via the induction of expression of CD23, CD69 and CD80. In addition, our results show for the first time the involvement of the CD40 homodimer in the induction of CD23 expression via TLR4. Our results emphasize the importance of CD40 homodimer in signaling pathways and highlight the role of Cys-238 in the cooperation between receptors of the innate and adaptive immune response. All together our results will allow a better understanding of CD40 signaling pathways involved in several autoimmune diseases, which give a rise to a better therapeutic trial design.

Analysis of electrogenerated chemiluminescence of PPV type conducting polymers

Mit Lösungen von 9,10-Diphenylanthracen und N(C2H5)4ClO4 oder N(C4H9)4ClO4 als Leitsalz im Lösungsmittel Acetonitril wurden Elektrochemilumineszenz (ECL)-Experimente durchgeführt. Dazu wurden die Elektroden mit Folgen von jeweils drei in bestimmten zeitlichen Abständen aufeinander folgenden Potentialsprüngen polarisiert. Es wird gezeigt, dass bei entsprechender Wahl der Potentiale und der Haltezeiten anodische und kathodische ECL-Emissionen gleicher Intensität erzeugt werden können. Sodann wurde ECL in den Derivaten von Poly(p-phenylen-vinylen), MEH-PPV und DB-PPV erzeugt. Diese leitfähigen Polymere wurden als dünne Schichten auf Platin-Elektroden aufgebracht und wie bei ECL aus der Lösungsphase in Acetonitril-Elektrolyten mit Tetralkylammonium-Leitsalzen Potentialsprüngen unterworfen. Bei geeigneter Einstellung der Potentialsprünge und Haltezeiten konnten anodische und kathodische ECL gleicher Intensität erhalten werden. Dies ist das erste Mal, dass symmetrische ECL mit polymerbeschichteten Elektroden erhalten wurde. Die Kinetik der ECL weicht deutlich von der aus der Lösungsphase ab. Der ECL-Prozess verläuft langsamer als in der Lösungsphase, und der Leitelektrolyt hat einen signifikanten Einfluss auf das elektrochemische Verhalten der Polymerschicht. Die Ursachen dafür wurden über Modellrechnungen analysiert, mit denen die Ladungstransportprozesse in der Polymerschicht simuliert wurden. In derartigen Simulationsrechnungen konnten die Geschwindigkeitskonstanten der ECL-Reaktion sowohl im Polymer als auch in der Lösung bestimmt werden. Um die Stabilität der Polymerschichten zu erhöhen, wurde versucht, die Polymerketten mit Synchrotronstrahlung zu vernetzen. Diese Experimente brachten nicht das erwartete Ergebnis. Die Ursachen dafür werden auf der Grundlage von Ex-Situ-Raman-spektroskopischen Untersuchungen diskutiert. / Electrochemiluminescence (ECL) has been generated in solution phase using 9,10-diphenylanthracene (DPA) with TEAClO4 (or TBAClO4) in acetonitrile solvent. Triple potential step was used for the generation of ECL. It was found that anodic and cathodic ECL of equal intensities can be generated by proper choice of potential step magnitude, width and the waiting period (tw) between successive triple potential steps. ECL was then generated in conducting polymers poly(2-ethylhexyloxy-5-methoxy-1,4-phenylenevinylene) (MEH-PPV) and poly(2,3-dibutoxy-1,4-phenylenevinylene) (DB-PPV) by coating them on Pt electrodes and subjecting to potential steps in tetraalkylammonium salt solutions with acetonitrile. Similar to the case of solution phase ECL, symmetrical anodic and cathodic ECL could be observed by the appropriate choice of the potential step parameters. But the kinetics of the ECL was found to be different from that of the solution phase ECL. The time scale of the ECL process was found to be longer than that in the solution phase ECL. The nature of supporting electrolyte had a remarkable impact on the electrochemistry of conducting polymers. The reasons were analyzed by theoretical calculations evoking the concept of charge transport characteristics of conducting polymers. The rate constants of the ECL process were calculated by separate simulation procedure in the solution phase as well as in the polymer phase ECL. To enhance the stability of conducting polymers, synchrotron radiation induced cross-linking was performed. The effects were different from expected which were analyzed and rationalized by ex-situ Raman spectroscopic studies.

Caractérisation et modélisation de l’évolution spectrale des étoiles naines blanches chaudes

Cette thèse présente une étude empirique et théorique approfondie de l'évolution spectrale des étoiles naines blanches, avec un accent particulier sur les naines blanches chaudes. La composition atmosphérique (et donc l'apparence spectrale) de ces cadavres stellaires peut changer drastiquement avec le temps à mesure qu'ils se refroidissent. Ce phénomène est généralement interprété comme le résultat d'une compétition entre divers mécanismes de transport des éléments dans l'enveloppe stellaire (tels que la diffusion, la convection, les vents et l'accrétion), mais demeure mal compris à plusieurs égards. Il est impératif de remédier à cette situation pour être en mesure d'exploiter le potentiel immense des naines blanches pour notre compréhension du passé de la Galaxie. Pour mieux caractériser l'incidence de l'évolution spectrale, nous effectuons tout d'abord une analyse spectroscopique exhaustive de près de 2000 naines blanches chaudes (Teff > 30,000 K) observées par le relevé SDSS. Nous déterminons les propriétés atmosphériques (notamment la température effective et la composition de surface) de ces objets à l'aide d'un nouvel ensemble de modèles d'atmosphère calculé spécifiquement à cet effet. En examinant la fréquence relative des étoiles riches en hydrogène et riches en hélium en fonction de la température, nous obtenons pour la première fois un portrait empirique détaillé de l'évolution spectrale des naines blanches chaudes. Plus spécifiquement, nous déduisons (1) qu'environ une étoile sur quatre arrive sur la séquence de refroidissement avec une atmosphère d'hélium, et (2) qu'environ deux tiers de ces objets développent ultérieurement une atmosphère d'hydrogène. En outre, nous accordons une attention particulière aux naines blanches hybrides (qui montrent à la fois des traces d'hydrogène et d'hélium) de notre échantillon et à ce que ces objets distinctifs nous apprennent sur l'évolution spectrale. Nous étudions ensuite l'évolution spectrale d'un point de vue théorique en modélisant les transformations chimiques qui s'opèrent dans les naines blanches. Pour ce faire, nous utilisons le code d'évolution stellaire STELUM, qui inclut un traitement cohérent et réaliste du transport des éléments et nous permet donc de réaliser les simulations numériques d'évolution spectrale les plus sophistiquées à ce jour. Nous modélisons la diffusion de l'hydrogène résiduel dans une enveloppe d'hélium à haute température, qui mène ultimement à la formation d'une atmosphère d'hydrogène. Nous simulons également le mélange convectif de cette couche superficielle d'hydrogène avec la couche sous-jacente d'hélium à basse température, qui produit à nouveau une surface dominée par l'hélium. En outre, nous étudions le transport du carbone dans les étoiles riches en hélium, incluant à la fois le tri gravitationnel à haute température et le dragage convectif à basse température. Ces calculs donnent lieu à plusieurs résultats astrophysiques d'intérêt. Nous obtenons notamment une contrainte inédite sur la quantité d'hydrogène résiduel contenue dans les naines blanches chaudes dominées par l'hélium. Nous démontrons aussi que la bifurcation observée dans le diagramme couleur-magnitude des naines blanches découvertes par le satellite Gaia est une signature du processus de mélange convectif à basse température. Par ailleurs, nos modèles fournissent de précieuses informations sur les propriétés des étoiles polluées par le carbone, en particulier sur leur masse et leur zone convective. Enfin, le résultat le plus important de cette thèse est la résolution définitive du problème le plus sérieux de la théorie de l'évolution spectrale, soit le problème de l'origine de l'hydrogène à la surface des naines blanches de type DBA. / This thesis presents an in-depth empirical and theoretical study of the spectral evolution of white dwarf stars, with a particular focus on hot white dwarfs. The atmospheric composition (and thus the spectral appearance) of these stellar remnants can change drastically over time as they cool. This phenomenon is generally interpreted as the result of an interplay between various element transport mechanisms in the stellar envelope (such as diffusion, convection, winds, and accretion), but remains poorly understood in several respects. It is imperative to remedy this situation to be able to exploit the immense potential of white dwarfs for our understanding of the past of the Galaxy. To better characterize the incidence of spectral evolution, we first carry out an exhaustive spectroscopic analysis of nearly 2000 hot white dwarfs (Teff > 30,000 K) observed by the SDSS survey. We determine the atmospheric properties (in particular the effective temperature and surface composition) of these objects using a new set of model atmospheres calculated specifically for this purpose. By examining the relative frequency of hydrogen-rich and helium-rich stars as a function of temperature, we obtain for the first time a detailed empirical picture of the spectral evolution of hot white dwarfs. More specifically, we infer (1) that about one in four stars enters the cooling sequence with a helium atmosphere, and (2) that about two-thirds of these objects eventually develop a hydrogen atmosphere. Furthermore, we pay special attention to the hybrid white dwarfs (which exhibit traces of both hydrogen and helium) in our sample and to what can be learned about spectral evolution from these distinctive objects. We then study spectral evolution from a theoretical point of view by modeling the chemical transformations that take place in white dwarfs. To do this, we use the stellar evolution code STELUM, which includes a consistent and realistic treatment of element transport and therefore allows us to perform the most sophisticated numerical simulations of spectral evolution to date. We model the diffusion of residual hydrogen in a helium envelope at high temperature, which ultimately leads to the formation of a hydrogen atmosphere. We also simulate the convective mixing of this superficial hydrogen layer with the underlying helium layer at low temperature, which once again produces a helium-dominated surface. Furthermore, we study the transport of carbon in helium-rich stars, including both gravitational settling at high temperature and convective dredge-up at low temperature. These calculations give rise to several astrophysical results of interest. In particular, we obtain an unprecedented constraint on the amount of residual hydrogen contained within hot helium-dominated white dwarfs. We also demonstrate that the bifurcation observed in the color-magnitude diagram of white dwarfs discovered by the Gaia satellite is a signature of the convective mixing process at low temperature. Furthermore, our models provide valuable information on the properties of carbon-polluted stars, in particular on their mass and convective zone. Finally, the most important result of this thesis is the definitive resolution of the most serious problem of the theory of spectral evolution, namely the problem of the origin of hydrogen at the surface of DBA-type white dwarfs.

En jämförelse i kostnad och prestanda för molnbaserad datalagring / A comparison in cost and performance for cloud-based data storage

I takt med att datakvantiteter växer och kraven på skalbarhet och tillgänglighet inom molntjänster växer, framhävs behovet av undersökningar kring dess prestanda och kostnadseffektivitet.  Dessa analyser är avgörande för att optimera tjänster och bistå företag med värdefulla rekommendationer för att fatta välgrundade beslut om datalagring i molnet. Detta examensarbete undersöker kostnad samt prestanda hos relationella och icke-relationella datalagringslösningar implementerade på Microsoft Azure och Google Cloud Platform. Verktyget Hyperfine används för att mäta latens och tjänsternas kostnadseffektivitet beräknas baserat på detta resultat samt dess beräknade månadskostnader. Studiens resultat indikerar att för de utvärderade relationella databastjänsterna uppvisar Azure SQL Database initialt en låg latens som sedan ökar proportionellt med datamängden, medan Google Cloud SQL visar en något högre latens vid lägre datamängder men mer konstant latens vid högre datamängder. Azure SQL visar sig vara mer kostnadseffektiv i förhållande till Google Cloud SQL, vilket gör den till ett mer fördelaktigt alternativ för företag som eftersträvar hög prestanda till lägre kostnader. Vid jämförelse mellan de två icke-relationella databastjänsterna Azure Cosmos DB och Google Cloud Datastore uppvisar Azure Cosmos DB genomgående jämförelsevis lägre latens och överlägsen kostnadseffektivitet. Detta gör Azure Cosmos DB till en fördelaktig lösning för företag som prioriterar ekonomisk effektivitet i sin databashantering. / As data volumes grow and the demands for scalability and availability within cloud services increase, the need for studies on their performance and cost-effectiveness is emphasized. These analyses are crucial for optimizing services and providing businesses with valuable recommendations to make well-grounded decisions about cloud data storage. This thesis examines cost and performance for relational and non-relational data storage solutions implemented on Microsoft Azure and Google Cloud Platform. The tool Hyperfine is used to evaluate latency and the cloud services cost efficiency is calculated using this result as well as their monthly cost. The study's results regarding relational data storage indicate that Azure SQL Database initially exhibits low latency, which then increases proportionally with the data volume, while Google Cloud SQL shows slightly higher latency at smaller data volumes but more consistent latency with more data. Azure SQL Database is more cost-effective, making it a more favorable option than Google Cloud SQL for companies seeking high performance at lower costs. Regarding evaluated services for non-relational data storage Azure Cosmos DB consistently demonstrates lower latency and superior cost efficiency compared to Google Cloud Datastore, making it the preferred solution for companies prioritizing economic efficiency in their database management.

O presente trabalho ocupa-se no desenvolvimento de um sistema computacional de gerenciamento da informação para auxiliar os estudos científicos sobre o sistema nervoso de chagásicos crônicos não-cardíacos. O objetivo é desenvolver o sistema requerido, pelo pressuposto de praticidade nas análises decorrentes da investigação. O método utilizado para desenvolver este sistema computacional, dedicado ao gerenciamento das informações da pesquisa sobre as alterações neurológicas de seus sujeitos, foi; compor o arquétipo de metas e a matriz de levantamento de requisitos das variantes do sistema; listar os atributos, domínios e qualificações das suas variáveis; elaborar o quadro de escolha de equipamentos e aplicativos necessários para sua implantação física e lógica e; implantá-lo mediante uma modelagem de base de dados, e uma programação lógica de algoritmos. Como resultado o sistema foi desenvolvido. A discussão de análise é; a saber, que a informatização pode tornar mais eficaz as operações de cadastro, consulta e validação de campo, além da formatação e exportação de tabelas pré-tratadas para análises estatísticas, atuando assim como uma ferramenta do método científico. Ora, a argumentação lógica é que a confiabilidade das informações computacionalmente registradas é aumentada porque o erro humano é diminuído na maioria dos processamentos. Como discussão de cerramento, estudos dotados de razoável volume de variáveis e sujeitos de pesquisa são mais bem geridos caso possuam um sistema dedicado ao gerenciamento de suas informações. / This is the development of a computer information management system to support scientific studies about the nervous system of non-cardiac chronic chagasic patients. The method used to develop this computer system, dedicated to information management of research about the neurological disorders of their human subject research, were; compose the archetypal matrix of targets and requirements elicitation of the system variants; list the attributes, qualifications and domains of its variables; draw up the choice framework of equipment and required applications for its physical and logic implementation, and; deploying it through a data modeling, an adapted entity-relationship diagram and programmable logic algorithms. As a result the required system was developed. The analytical discussion is that the computerization makes the data processing faster and safer. The more practical information management processes are: the operations of registration, queries and fields\' validations, as well as the advanced and basic queries of records, in addition to table formatting and exporting of pre-treated for statistical analysis. The logical argument is that the reliability of the recorded computationally information is increased because is insured that bias of human error is absent from most of the steps, including several the data processing operations. As end discussion, scientific studies with reasonable amount of variables and research subjects are better managed if they have a dedicated system to managing their information.

Fluidic Tuning of a Four-Arm Spiral-Based Frequency Selective Surface

Frequency selective surfaces (FSSs) provide a variety of spatial filtering functions, such as band-pass or band-stop properties in a radome or other multilayer structure. This filtering is typically achieved through closely-spaced periodic arrangements of metallic shapes on top of a dielectric substrate (or within a stack of dielectric materials). In most cases, the unit cell size, its shape, the substrate parameters, and the inter-element spacing collectively impact the response of the FSS. Expanding this design space to include reconfigurable FSSs provides opportunities for applications requiring frequency agility and/or other properties. Tuning can also enable operation over a potentially wider range of frequencies and can in some cases be used as a loading mechanism or quasi-ground plane. Many technologies have been considered for this type of agility (RF MEMS, PIN diodes, etc.). This includes the recent use of microfluidics and dispersions of nanoparticles, or fluids with controllable dielectrics, which have entered the design space of numerous other EM applications including stub-tuners, antennas, and filters. In this work they provide a material based approach to reconfiguring an FSS. An FSS based on a four-arm spiral with tunable band-stop characteristics is presented in this work. A thin colloidal dispersion above each element provides this tuning capability. The radial expansion and contraction of this dispersion, as well as the variable permittivity of the dispersion, are used to load each element individually. This design incorporates thin fluidic channels within a PDMS layer below the substrate leading to individual unit cells that provide a closed pressure-driven subsystem that contains the dispersion. With the capability to individually control each cell, groups of cells can be locally altered (individually or in groups) to create gratings and other electromagnetically agile features across the surface or within the volume of a radome or other covering. Simulations and measurements of an S-band tunable design using colloidal Barium Strontium Titanate dispersed Silicone oil are provided to demonstrate the capability to adjust the stop-band characteristics of the FSS across the S-band.

Lietuvos Respublikos muitinės vaidmuo smarkiai pasikeitė nuo 2004 m. gegužės 1 d. Po įstojimo į ES prekės valstybių narių teritorija juda be jokių prekybos apribojimų, o importuojamoms iš trečiųjų šalių prekėms taikomi vienodi tarifai ir kvotos. Lietuva atsakinga už ES išorinės sienos apsaugą, todėl padidėjo jos atsakomybė tikrinant iš trečiųjų šalių gabenamas prekes. Po 2004 m. pasikeitė ir muitinės, kaip mokesčių surinkėjos vaidmuo – kadangi muitai ir muito mokesčiai yra sudedamoji ES nuosavų išteklių sistemos dalis. Kadangi muitinis apmokestinimas siejamas su trimis pagrindinėmis charakteristikomis – prekių kilme, prekės kodu, prekių muitine verte, todėl muitinė atlikdama savo, kaip mokesčių surinkėjos vaidmenį, visų pirma, turi įvertinti ir atlikti patikrinimus, susijusios su šiomis charakteristikomis, kadangi daugiausia muitų teisės aktų pažeidimų kaip tik ir nustatoma šioje srityje. Customs plays a vital role in protecting borders from the entry of illegal and harmful goods. EU Customs Union is based on a main principle – free movement of goods, which means that the goods between EU territory can move without no restrictions applied. Since 2004 Customs as tax collector‘s role also change – customs duties form a part of Communities own resources system. The main characteristics that have an impact on customs duties applied are origin of goods, tarriff clasification and customs value and because of that the most of the customs law infringements accur then the import operations are performed. This is why the customs authorities should carry out the inspections on these elements very carefully. O presente trabalho ocupa-se no desenvolvimento de um sistema computacional de gerenciamento da informação para auxiliar os estudos científicos sobre o sistema nervoso de chagásicos crônicos não-cardíacos. O objetivo é desenvolver o sistema requerido, pelo pressuposto de praticidade nas análises decorrentes da investigação. O método utilizado para desenvolver este sistema computacional, dedicado ao gerenciamento das informações da pesquisa sobre as alterações neurológicas de seus sujeitos, foi; compor o arquétipo de metas e a matriz de levantamento de requisitos das variantes do sistema; listar os atributos, domínios e qualificações das suas variáveis; elaborar o quadro de escolha de equipamentos e aplicativos necessários para sua implantação física e lógica e; implantá-lo mediante uma modelagem de base de dados, e uma programação lógica de algoritmos. Como resultado o sistema foi desenvolvido. A discussão de análise é; a saber, que a informatização pode tornar mais eficaz as operações de cadastro, consulta e validação de campo, além da formatação e exportação de tabelas pré-tratadas para análises estatísticas, atuando assim como uma ferramenta do método científico. Ora, a argumentação lógica é que a confiabilidade das informações computacionalmente registradas é aumentada porque o erro humano é diminuído na maioria dos processamentos.
