É um material resistente à degradação eletroquímica e fotoquímica. Com o aumento da produção industrial de corantes, há um aumento significativo da produção de rejeitos, sendo necessário o desenvolvimento de novas técnicas de degradação, a fim de reduzir a formação de efluentes. Dentre essas técnicas encontram-se os processos oxidativos avançados (POAs), que se baseiam na formação de radicais hidroxila para a degradação dos compostos liberados nos efluentes. A fotocatálise heterogênea utiliza um material semicondutor ativado por radiação ultra-violeta a fim de produzir os radicais hidroxila. Apesar de existirem estudos relacionados à utilização do TiO2 como fotocatalisador, há poucos dados com relação à sua aplicação na forma de filme suportado. Este trabalho teve por objetivos crescer filmes de TiO2 sobre borossilicato, por meio da técnica de deposição química de organometálicos em fase vapor, nas temperaturas de 400 e 500ºC por até 60 minutos, bem como proceder à caracterização microestrutural, morfológica e fotocatalítica desses filmes. Anatase foi a fase identificada em todos os filmes. Os filmes crescidos a 400°C apresentaram estrutura densificada, enquanto que os filmes crescidos a 500°C apresentaram estrutura colunar bem definida. A fotodegradação foi avaliada por meio da degradação do corante alaranjado de metila nos valores de pH 2,00; 7,00 e 10,00. Os resultados de degradação do corante mostraram que a maior eficiência do processo de degradação ocorre em pH = 2. Diploma thesis describes technology of pulse reactive magnetron sputtering of Al2O3 coating. Furthermore it deals with design of deposition process and testing new aluminium target. The technological window was determined by tests. As a result, it was possible to create Al2O3 coatings in the widest possible working range. The coatings created by apparatus PLS 160 were tested for optical properties in order to obtain transmittance. Besides the optical tests, the evaluation of mechanical properties and adhesion were performed. The obtained results from measurement were evaluated. Based on the results, some possible applications of the coating were suggested.

Phase transformation in tetrahedral amorphous carbon by focused ion beam irradiation

Ion irradiation of tetrahedral amorphous carbon (ta-C) thin films induces a carbon phase transformation from the electrically insulating sp3 hybridization into the conducting sp2 hybridization. In this work, a detailed study on the electrical resistivity and the microstructure of areas, irradiated with several ion species at 30 keV energy is presented. Continuous ion bombardment yields a drastic drop of the resistivity as well as significant structural modifications of the evolving sp2 carbon phase. It is shown that the resistivity lowering can be attributed to the degree of graphitization in the film. Furthermore, the structural ordering processes are correlated with the ion deposited energy density. It is therefore revealed that the ion-induced phase transformation in ta-C films is a combination of sp3-to-sp2 conversion of carbon atoms and ion-induced ordering of the microstructure into a more graphite-like arrangement. All experiments were done with focused ion beam (FIB) systems by applying FIB lithography of electrical van-der-Pauw test structures. Superhydrophobic Aluminum Surfaces: Preparation Routes, Properties and Artificial Weathering Impact

Among the materials that can be treated in order to impart superhydrophobic properties are many originally hydrophilic metals. For this, they must undergo a sequential treatment, including roughening and hydrophobic coating. This contribution presents various preparation routes along with various characterization methods, such as dynamic contact angle (DCA) measurements, scanning electron microscopy (SEM) and spectroscopic techniques (FT–IRRAS, XPS, EIS). Micro-rough surfaces of pure and alloyed aluminum were generated most easily by using a modifie Sulfuric Acid Anodization under Intensifie conditions (SAAi). This produces a micro-mountain-like oxide morphology with peak-to-valley heights of 2 μm and sub-μm roughness components. Additionally, micro-embossed and micro-blasted surfaces were investigated. These micro-roughened initial states were chemically modifie with a solution of a hydrophobic compound, such as the reactive f uoroalkylsilane PFATES, the reactive alkyl group containing polymer POMA, or the polymer Teflo ® AF. Alternatively, the chemical modificatio was made by a Hot Filament Chemical Vapor Deposition (HFCVD) of a PTFE layer. The latter can form a considerably higher thickness than the wet-deposited coatings, without detrimental leveling effects being observed in comparison with the original micro-rough surface. The inherent and controllable morphology of the PTFE layers represents an important feature. The impacts of a standardized artificia weathering (WTH) on the wetting behavior and the surface-chemical properties were studied and discussed in terms of possible damage mechanisms. A very high stability of the superhydrophobicity was observed for the f uorinated wet-deposited PFATES and Teflo ® AF coatings as well as for some of the PTFE layer variants, all on SAAi-pretreated substrates. Very good results were also obtained for specimens produced by appropriate mechanical roughening and PTFE coating.

Source Apportionment and Risk Assessment of Urban Diffuse Pollutants of Heavy Metals and Polycyclic Aromatic Hydrocarbons on Urban Watershed

In this Dissertation, systematic work has been carried out to study the road-deposited sediment and its adsorbed pollutants from a stormwater pollution perspective. Solid-phase concentration, surface load, source apportionment, risk assessment, and desorption dynamics of polycyclic aromatic hydrocarbons and/or heavy metals in road-deposited sediments (RDS) were investigated. In order to provide data to assist potential strategies of stormwater pollution mitigation and integrated catchment management to minimise the adverse impacts of RDS adsorbed pollutants on stormwawter quality, the following specific topics were addressed. ⑴ The influences of traffic load and antecedent dry-weather period on pollution level and ecological risk of heavy metals in RDS were analyzed. ⑵ The build-up dynamics and chemical fractionation of metals were determined. ⑶ The potential source contributions and risk assessment of polycyclic aromatic hydrocarbons in size-fractionated RDS were firstly determined by a Principal component analysis - Multiple linear regression receptor model. ⑷ The qualitative and quantitative source apportionments of polycyclic aromatic hydrocarbons were subsequently investigated through a combined qualitative Molecular Diagnostic Ratio and quantitative Positive Matrix Factorization source apportionment with an extended data set. ⑸ The exposure risk of polycyclic aromatic hydrocarbons were evaluated and calculated by incremental lifetime cancer risk models. ⑹ Then, a novel ecological risk assessment approach to the RDS adsorbed toxic substances was developed, which was explored exclusively for the study of RDS for a water pollution aspect. ⑺ Finally, the effects of rainwater, major wastewater constituents of dissolved organic matter and surfactant on the leaching of heavy metals from RDS were carried out.

Laser decontamination and cleaning of metal surfaces : modelling and experimental studies / Décontamination et nettoyage laser appliqués aux surfaces métalliques : études théorétiques et expérimentales

Le nettoyage des surfaces métalliques est nécessaire dans différents domaines de l'industrie moderne. L'industrie nucléaire cherche de nouvelles méthodes de décontamination des surfaces oxydées, et les installations thermonucléaires nécessitent le nettoyage des composants face au plasma pour enlever la couche déposée contenant tritium. L'ablation laser est proposée comme une méthode efficace et sûre pour le nettoyage des surfaces métalliques et leur décontamination. Le facteur important influençant le chauffage et l'ablation laser est la distribution en profondeur de l’intensité laser. Le modèle de propagation de la lumière dans une couche diffusant sur un substrat métallique est développé et appliqué pour analyser les caractéristiques de distribution de lumière. Pour simuler les surfaces contaminées, l'inox AISI 304L a été oxydé par laser et chauffé dans un four. La contamination radioactive de la couche d'oxyde a été simulée par l'introduction d’europium et / ou de sodium. Un facteur de décontamination de plus de 300 a été démontré avec le régime de nettoyage optimal trouvé. Une diminution de la résistance à la corrosion a aussi été montrée après un nettoyage laser. Les seuils d'ablation des surfaces ITER-like (représentatives d’ITER) ont été mesurés. Une vitesse de nettoyage de 0,07 m2/W∙h a été trouvée. Pour les surfaces miroir, les seuils de dommages étaient déterminés pour éviter les dommages lors du nettoyage au laser. La possibilité de restaurer la réflectivité après le dépôt d’une couche mince de carbone a été démontrée. Les perspectives de développement ultérieur de nettoyage laser sont discutées. / Metal surface cleaning is highly required in different fields of modern industry. Nuclear industry seeks for new methods for oxidized surface decontamination, and thermonuclear installations require the cleaning of plasma facing components from tritium-containing deposited layer. The laser ablation is proposed as an effective and safe method for metal surface cleaning and decontamination. The important factor influencing the laser heating and ablation is the in-depth distribution of laser radiation. The model of light propagation in a scattering layer on a metal substrate is developed and applied to analyse the features of light distribution. To simulate the contaminated surfaces, the stainless steel AISI 304L was oxidized by laser and in a furnace. Radioactive contamination of the oxide layer was simulated by introducing europium and/or sodium. The decontamination factor of more than 300 was demonstrated with found optimal cleaning regime. The decreasing of the corrosion resistance was found after laser cleaning. The ablation thresholds of ITER-like surfaces were measured. The cleaning productivity of 0.07 m2/hour∙W was found. For mirror surfaces, the damage thresholds were determined to avoid damage during laser cleaning. The possibility to restore reflectivity after thin carbon layer deposition was demonstrated. The perspectives of further development of laser cleaning are discussed.

Polymer intercalation of chemically bath deposited iron sulphide and nickel sulphide thin films

(Department of Chemistry, Faculty of Applied and Computer Sciences), Vaal University of Technology. / In chemical bath deposition (CBD) method, deposition of metal chalcogenide semiconducting thin films occurs due to substrate maintained in contact with a dilute chemical bath containing metal and chalcogenide ions. Semiconducting nickel sulphide (NiS) and iron sulphide (FeS) thin films have been prepared on a glass substrate by varying the deposition parameters such as the concentration of solutions, deposition time, temperature and pH. Multi-layered thin films were deposited on glass substrate and the spin-cast conductive polymer, poly (3.4-ethylenedioxythiopene) polystyrene sulfonate (PEDOT: PSS) was intercalated. The characterization of the films was carried out using UV-Vis spectroscopy, scanning electron microscopy (SEM) coupled with energy-dispersive X-ray spectroscopy (EDX), atomic force microscopy (AFM) and X-ray diffraction (XRD). Single layer nickel sulphide was deposited at room temperature, pH 10 and the deposition period of 3 hours, triethanolamine was used as the complexing agent. Iron sulphide was deposited for 6 hours at 70 °C with the pH of 2.5 using EDTA as a complexing agent. Generally the iron and nickel sulphide were prepared from their respective nickel or iron salt and the thiourea or thiosulfate as a source of sulphide ions in solution. SEM and AFM results show that the FeS film is evenly coated and has uniform grain size with the roughness of ~22.4 nm and thickness of ~23.8 nm. The optical absorption analysis of FeS showed the band gap energy of ~2.9 eV which blue shifted from the bulk. The EDX analysis confirms the compositions of iron and sulphur in FeS films. XRD pattern showed amorphous films for both FeS and NiS thin films due to the amorphous nature of the glass substrate. The optical data of NiS film were analysed and exhibited the band gap energy of ~3.5 eV and ~3.3 eV for successive ionic layer adsorption and reaction (SILAR), which is the modified CBD, both blue shifted from the bulk. The films were observed to have thickness value of ~35.7 nm and ~2.3 nm SILAR with the roughness of ~112.5 nm and ~35.4 nm SILAR from AFM results. SEM confirmed the uniformly distributed film presented by AFM analysis. The chemical composition of Ni and S were confirmed by EDX spectra. The PEDOT: PSS was intercalated between the FeS as the first layer and NiS as the top layer which gave the thickness of ~18.7 nm and roughness of ~115.2 nm from AFM analysis. PEDOT: PSS acted as a passive layer that protects and stabilize the FeS layer and NiS as the third active layer which enhanced the optical absorption of the film when using SILAR method for solar application.

mRNA localization and transcriptome dynamics in early zebrafish development

Die Lokalisierung von mRNA ist ein wichtiger regulativer Mechanismus in polarisierten Zellen und in frühen Embryonalstadien. Dort sind räumliche Muster maternaler mRNA für die korrekte Entwicklung der Körperachsen und die Spezifizierung der Keimzellen verantwortlich. Systematische Analysen dieser Prozesse wurden jedoch bisher limitiert durch einen Mangel an räumlicher und zeitlicher Auflösung von Einzelzell- Sequenzierungsdaten. Wir analysierten die Dynamik des räumlichen und zeitlichen Transkriptoms während frühen Embryonalstadien von Zebrafischen. Wir verbesserten Empfindlichkeit und Auflösung von tomo-seq und erfassten damit systematisch räumlich aufgelöste Transkriptome entlang der animal-vegetalen-Achse Embryonen im Einzell-Stadium und fanden 97 vegetal lokalisierte Gene. Außerdem etablierten wir eine Hochdurchsatz kompatible Variante der RNA-Markierungsmethode scSLAM-seq. Wir wendeten diese in Embryonen während der Gastrulation. Von den vegetal lokalisierten Genen waren 22 angereichert in Keimzellen, was eine funktionelle Rolle bei der Spezifizierung von Keimzellen nahelegt. Mit tomo-seq untersuchten wir die evolutionäre Konservierung der RNA-Lokalisierung zwischen Zebrafischen und gereiften Oozyten zweier Xenopus-Arten. Wir verglichen die lokalisierten Gene, suchten nach konservierten 3'UTR-Motiven, und fanden zum Teil überlappende Motive, was auf eine mögliche mechanistische Konservierung der Lokalisierungsmechanismen hinweist. Wir untersuchten auch RNA-Editierung von Adenin zu Inosin während der Embryonalentwicklung und in den Organen erwachsener Fische. In im Gehirn exprimierten Transkripten fanden wir 117 Editierstellen, die hauptsächlich für Ionentransporter kodieren und zum Teil zum Menschen konserviert sind. Die höchsten Editierraten konnten wir in Eierstöcken, Hoden und frühen Embryonen nachweisen, was auf eine mögliche Rolle bei der Regulierung der RNA-Stabilität hindeutet. / Subcellular localization of mRNA is an important regulatory mechanism in polarized cells. In early embryos of many species, spatial patterns of maternal mRNA are essential for the proper development of body axes and the specification of germ cells. These processes have been studied in zebrafish, but systematic analyses have been hindered by a lack of spatial and temporal information in single-cell RNA sequencing. We performed a spatial-temporal analysis of the zebrafish transcriptome during early embryonic development to systematically characterize localized mRNA and the fate of maternal transcripts until gastrulation stage. We enhanced sensitivity and resolution of the tomo-seq method and systematically acquired spatially-resolved transcriptomes along the animal-vegetal axis of one-cell stage zebrafish embryos, and found 97 genes to be localized vegetally. Furthermore, we established an in vivo and high-throughput compatible version of the single-cell RNA labeling method scSLAM-seq in gastrulation stage embryos. We followed localized transcripts until gastrulation and found transcripts of 22 of the vegetally localized genes enriched in primordial germ cells. We propose that these genes have a functional role in the early priming of the germ cell fate. To investigate the evolutionary conservation of vegetal RNA localization, we acquired tomo-seq datasets of mature oocytes of two xenopus species. We compared the pools of localized RNA and searched for conserved 3’UTR motifs. The resulting sets showed high similarity, possibly reflecting a mechanistic conservation of localization pathways. We also investigated RNA A-to-I editing during embryonic development and in organs of adult fish. Specifically, we identified 117 recoding editing sites in the brain that mainly encode for ion transporters and are partly conserved in humans. We detected the highest editing levels in ovary, testes and in early embryos, implicating a potential role in regulating RNA stability.
