Optimalizace metod pro studium časných fází životního cyklu myšího polyomaviru / Optimization of methods for analysis of early steps of mouse polymavirus life cycle

Mouse polyomavirus is a type species of Polyomaviridae family and serves as model for studying viral infection of human pathogenic polyomaviruses. Minor proteins of viral capsid have been found to be necessary for effective initiation of infection. In order to study their role in the early steps of infection we utilized the novel Cre-LoxP system for production of the viral mutant lacking both minor proteins. Virus produced this way was compared with virus produced by standard method and we found that both systems facilitate production of mutant virus with the comparable quality and quantity. The mutant virus contained reduced amount of viral DNA and formed virions with impaired stability. For further studies of intracellular virion trafficking we prepared virions with genomes modified by thymidine analogues 5- bromo-2'-deoxyuridine (BrdU) and 5-Ethynyl-2'-deoxyuridine (EdU) and optimized the methods for analogue detection. The viral genome become accessible for detection 4 hours post infection. For ultramicroscopic analysis of translocation of virus to the nucleus we used freeze substitution. M.Ed. / However much ideologically diffuse argument there may be about the purposes of education, few would deny that it plays an integral part in the political and economical processes of societies. For the realities of power and the organisation of socio-economic structure are perhaps nowhere more clearly revealed than in a country's educational institutions. Education can maintain the existing social order as well as promote varying kinds of change (Toffler, 1971). Social scientists accept that "education is perhaps the most directly effective socialising activity serving the interests of dominant establishments" (Schlemmer, 1986: 1). Its curricula is a means of inculcating the political values as well as the skills required in the system of control and production in society (Wellington, 1987: 6). Education takes place relatively unobtrusively in homogenous societies but in deeply divided societies, it can become the focus of intense and often violent conflict. "In South Africa education is failing badly in what modern education is supposed to do draw different groups or classes together (Schlemmer, 1986: Preface). Hanf (1980), in addition, argues that education can do little to solve conflict in divided societies. He stresses that the significance of education in some societies lies in its negative role as a focus of conflict. Exploring the access and influence of Edu TV and radio programs on the learners' self-directed learning among the grade 12 learners enrolled on distance mode at Namibian College of Open Learning (NAMCOL )

This research study focused on the issue of access to flexible learning and the use of technology (radio and TV) to assist in the learning process. The study was aimed at measuring the influence of these technology services among grade 12 learners enrolled on distance mode at Namibia College of Open Learning (NAMCOL). The majority of distance education systems around the world have the same general structure of guiding students, which are based primarily on student-centered learning that demands a learner to work independently. Regular lecturing facilities for distance learners are only possible in scheduled workshops at study centers. These provisions are however, inconvenient since most distance learners are working people, as well as inadequate to provide the necessary educational guidance to distance learners. In situations where regular interaction between teacher and student is limited or not possible, the media plays a vital role in providing educational assistance to these learners. NAMCOL, as an institution, has embraced the concept of utilizing media to assist distance learners with programs such as Edu TV and Radio Programmes as well as Online resources. Self-directed learning theories and Malcolm Knowles's theory of Andragogy were used as the theoretical frameworks for the study. In exploring access and influence of Edu TV and radio program on the learner's self-directed learning among the grade 12 learners enrolled at distance mode at Namibian College of Open Learning (NAMCOL), the researcher employed several research strategies to measure the influence that flexible programs have on learners. The research strategies employed in the study included questionnaires and interviews, which were used to yield both qualitative and quantitative research data. The main findings of the study were that the flexible learning policy implemented by NAMCOL is proving successful in affording distance learners access to Edu TV and radio program services. All participants indicated that the lessons were informative. Furthermore, it helps learners to assume responsibility for their learning. However, additional efforts from all stakeholders are required to keep up with the ever-expanding demand for ODL platforms. The study further revealed that implementing appropriate Andragogy fostered self-directed learning. In light of the findings of the research and the Andragogy applied, even though learners have access to the Edu programs, the lack of physical and appropriate technologies can impact negatively. For example, you can be self-directed, but if there are physical constraints and challenges in the pedagogy, the learners are demotivated to be self-directed.

Syftet med studien är att studera, diskutera och analysera både om och på vilka sätt elever på fritidshem i större städer har likvärdig tillgång till material. Vi har använt oss av kvalitativ metod och har genom intervjuer samlat data. Datan har analyserats med utgångspunkt i sociokulturell teori och habitus. Vi fann att även om olika fritidshem har drastiskt olika mängder material finner lärarna det tillräckligt. Det finns god koppling mellan materialet på fritidshem och de intressen som eleverna presenterar. Lärares egna intressen och de populäraste aktiviteterna på fritidshemmet har störst påverkan på vilket material som köps in.

It was found that the consequences of labeling are generally permanent and irreversible, and that they result in isolation of the individual. Two literature studies were done. The first literature study dealt with the existing theories on self actualisation. After this study a list was drawn up that consisted of the characteristics of the actualiser. The second literature study dealt with the Seven Dimensions of Intelligence, as described by Edu-K. A connection was drawn between the characteristics of the actualiser and the Seven Dimensions. A clear connection was drawn theoretically between the characteristics of the actualiser and the seven dimensions. The empirical study also shows an improvement in the academical performance as well as the characteristics of the actualiser. / Die doel van hierdie studie is om te bepaal watter bydrae Edu-Kinesiologie (Edu-K) kan lewer tot die bereiking van selfaktualisering, veral op akademiese gebied Vir die verkenning is twee literatuurstudies gedoen. Die eerste literatuurstudie het gehandel oor die teoriee wat bestaan oor selfaktualisering. Na hierdie studie is 'n vraelys oor eienskappe wat kenmerkend van selfaktualiseerders is, saamgestel. Post-replicative resolution of under-replication

The evolutionary pressure to prevent re-replication by inactivating licensing during S phase leaves higher-eukaryotes with large genomes, such as human cells, vulnerable to replication stresses. Origins licensed in G1 must be sufficient to complete replication as new origins cannot be licensed in response to irreversible replication fork stalling. Interdisciplinary approaches between cellular biology and biophysics predict that replication of the genome is routinely incomplete in G2, even in the absence of external stressors. The frequency of converging replication forks that never terminate due to irreversible stalling (double fork stall), which result in a segment of unreplicated DNA, was modelled using high quality origin-mapping data in HeLa and IMR-90 cells. From this, hypotheses were generated that related an increase in unreplicated segments of DNA to reduced functional origin number. Presented in this thesis is the confirmation of this relation by quantifying chromosome mis-segregation and DNA damage responses when origin number was reduced using RNAi against licensing factors. The number of ultrafine anaphase bridges and 53BP1 nuclear bodies are in remarkable concordance with the theoretical predictions for the number of double fork stalls, indicating that cells are able to tolerate under-replication through such post-replicative cellular responses. 53BP1 preferentially binds to chromatin associated with large replicons, and functions synergistically with dormant origins to protect the stability of the genome. Additional candidates, inspired by common fragile site research, have also been characterised as responders to spontaneous under-replication, and include FANCD2 and MiDAS, which function in early mitosis to facilitate completion of replication before cells enter anaphase. In conclusion, a series of mechanisms that sequentially function throughout the cell cycle protects the stability of the human genome against inevitable spontaneous under-replication brought about by its large size.

Procedimentos modais na música brasileira: do campo étnico do Nordeste ao popular da década de 1960. / Modal Procedures in Brazilian Music

a)Objetivos: Traçar um perfil dos procedimentos melódicos modais presentes nas manifestações étnicas e populares do nordeste brasileiro afim de verificá-los em autores ligados à música popular do Brasil na década de 1960, cotejando os diferentes procedimentos harmônicos realizados. b)Métodos: O principal método utilizado foi o da análise a partir da transcrição de exemplos em áudio. A partir dessas análises uma série de comparações com materiais similares encontrados na bibliografia foi realizada. c) Resultados: Como resultado dessa pesquisa foram apontados os principais modos e traços cadenciais utilizados no repertório étnico e popular escolhido do nordeste brasileiro. Esses traços foram importantes na construção das canções aqui selecionadas, características da chamada MPB da década de 1960 apresentando, a partir dessas análises, os principais procedimentos harmônicos desses autores relacionados aos modos e traços cadenciais encontrados. / e)Objectives: Delineating a profile of the modal melodic procedures present in the ethnic and popular manifestations of the Northeast region of Brazil in order to verify them in authors connected to the Brazilian popular music from the 1960s, comparing the different harmonic procedures produced. f)Methods: The main method used was the analysis of transcripts of audio samples. From these analyses a series of comparisons with similar materials found in the bibliography was made. g)Results: As a result of this research, the main modes and cadential traces used in the ethnic and popular repertoire chosen from the Northeast of Brazil were indicated. These traces were important in the construction of the songs here selected, characteristics of the so-called MPB of the 1960s presenting, from those analyses, the main harmonic procedures of those authors related to the cadential modes and traces found.
