Atributos individuais, formas de manejo e contexto ambiental: quais fatores determinam a chance de cachorros visitarem remanescentes florestais? / Individual traits, management and environmental context: which factors determine the chance of dogs visiting forest remnants?

Invasões biológicas representam hoje a segunda maior ameaça à biodiversidade, e o homem desempenha papel fundamental na introdução de espécies exóticas potencialmente invasoras. O cachorro (Canis lupus familiaris) é uma dessas espécies. Presente em todos os continentes, é o carnívoro mais abundante do mundo, e pode causar impactos à fauna nativa através de efeitos letais e não letais da predação, competição, hibridização e transmissão de doenças, além de ser potencialmente importante na epidemiologia de zoonoses. Em áreas rurais, que concentram grande parte dos remanescentes de vegetação nativa no mundo, os cachorros são frequentemente criados soltos e circulam livremente, intensificando a chance de contato com a fauna nativa. Nesse trabalho, avaliamos a importância relativa de fatores associados a atributos individuais (sexo, idade, condição de saúde, comportamento exploratório, tamanho e raça), formas de manejo (incentivo do dono à movimentação, motivo para a criação, confinamento e frequência de alimentação) e contexto ambiental (proximidade à mata nativa) para determinar a chance de cachorros visitarem remanescentes florestais em paisagens fragmentadas de Mata Atlântica. Selecionamos oito paisagens de 2830 ha cada, nas quais visitamos todas as construções em áreas rurais a fim de entrevistar, através da aplicação de questionário, os responsáveis pela criação de cachorros e fotografar os cachorros. Utilizamos a imputação múltipla para estimar os dados faltantes (comuns em dados obtidos via questionários), gerando 10 conjuntos de dados imputados que foram analisados separadamente. Por meio de seleções de modelos através do Critério de Informação de Akaike, comparamos modelos candidatos com até cinco variáveis independentes para determinar a chance de cachorros visitarem remanescentes florestais. Nossos resultados nos permitem afirmar que quatro fatores - dois associados a atributos individuais e dois associados a formas de manejo - atuam em conjunto para determinar a chance de cachorros visitarem remanescentes florestais em paisagens rurais da Mata Atlântica. Cachorros maiores, mais exploradores, que recebem mais incentivo a se movimentar e que passam menos tempo confinados têm maior chance de visitar remanescentes florestais. A maior importância de atributos individuais e da forma de manejo está de acordo com a grande variação fenotípica existente entre cachorros e a variedade de modos como são manejados. Entre os atributos individuais, tanto características físicas como comportamentais são importantes, enquanto os aspectos chave da forma de manejo são aqueles mais diretamente relacionados à movimentação dos cachorros. Independentemente do tempo de confinamento, o incentivo do dono à movimentação do cachorro, em particular o estímulo para que o cachorro o acompanhe em visitas a remanescentes florestais, é fundamental. O contexto ambiental, em especial a proximidade do domicílio a áreas de mata nativa, por sua vez, é irrelevante dada a alta mobilidade dos cachorros. Nossos dados sugerem que a densidade de cachorros em paisagens rurais de Mata Atlântica é uma ordem de magnitude mais alta do que a de carnívoros de médio porte silvestres relativamente comuns, que muitos cachorros já tiveram contato direto com espécies silvestres, e que a vacinação e outras medidas profiláticas são relativamente incomuns. Todos esses dados indicam o potencial de efeitos negativos, tanto para a fauna silvestre como para o homem. Programas de redução destes impactos devem incluir tanto aspectos veterinários, como a expansão de campanhas públicas de profilaxia para além da vacinação antirrábica, quanto aspectos sociais, como a divulgação e conscientização dos potenciais problemas ocasionados pela entrada de cachorros em áreas de vegetação nativa, visando mudanças nas crenças, atitudes e comportamento da população humana / Biological invasions are today the second greatest threat to biodiversity, and humans play a significant role in the introduction of potentially invasive exotic species. The dog (Canis lupus familiaris) is one of such species. Distributed across all continents, the dog is the most abundant carnivore in the planet, and can impact wildlife through lethal and non-lethal effects of predation, competition, hybridization and disease transmission, besides being potentially important in the epidemiology of zoonoses. In rural areas where most remnants of native vegetation around the world are concentrated, most dogs are free-ranging, enhancing the chance of interactions with wildlife. Here, we evaluated the relative importance of factors associated with individual traits (sex, age, health condition, exploratory behaviour, size and breed), management (owner\'s incentive to movement, motive for raising, confinement and feeding frequency), and environmental context (proximity to native forest) to determine the chance of dogs visiting forest remnants in Atlantic Forest fragmented landscapes. We selected eight landscapes of 2830 ha each, where we visited all constructions in rural areas in order to interview dog owners by applying a questionnaire and to photograph dogs. We used multiple imputation to estimate missing data (common in data obtained through questionnaires), obtaining 10 imputed datasets that were analyzed separately. We compared candidate models with up to five independent variables to determine the chance of dogs visiting forest remnants through the Akaike Information Criterion. Our results indicate that four factors - two associated with individual traits and two associated with management - work together to determine the chance of dogs visiting forest remnants in Atlantic Forest rural landscapes. Larger dogs, and those that exhibit exploratory behaviour, are more stimulated to move or confined for shorter periods have greater chance of visiting forest remnants. The greater importance of factors associated with individual traits and management is in accordance with the ample phenotypic variation among dogs and the variety of ways they can be managed. Among individual traits, both morphological and behaviour characteristics are important, whereas the key aspects of management are those directly related to dog movement. Irrespective of the time confined, the owner\'s incentive to movement, in particular taking the dog to the forest, is a crucial aspect. In contrast, the environmental context, especially the proximity of the household to native forest, is irrelevant given the vagility of dogs. Our results suggest that the density of dogs across rural landscapes in the Atlantic forest is at least one order of magnitude higher than the density of relatively common medium-sized native carnivores, that several dogs have already had direct contact with wild species, and that vaccination and other prophylactic measures are relatively uncommon. All of these highlight the potential for negative effects on both wildlife and human population. Plans to reduce these effects should include not only veterinarian aspects, such as the expansion of public prophylactic campaigns beyond rabies vaccination, but also social aspects, such as the dissemination of information on the problems caused by dogs visiting native vegetation, aiming at changing people\'s beliefs, attitudes and behaviour

Karolina Suková Abstract People have been living with dogs (Canis familiaris) in close symbiotic relationship for many thousands of years. Such a kind of coexistence cannot get along without certain risk factors. Although aggression is a natural and wide spread phenomenon in the animal kingdom, manifestation of canine aggression toward people is a public health problem which has to be resolved. Dog bites can cause traumatic injuries to victims and decrease the quality of life or even cause the loss of life to the animal. Therefore there is a serious need to determine the risk factors of dog attacks to people and provide effective preventive strategies to this problem. We consider two kinds of risk factors to be fundamental in this respect: factors depend on the dog (e.g. age, breed, sex and size) and factors depend on the owner (e.g. method of training, prior experience with care of dogs, knowledge of natural behavior of dog, time dedicated to dog etc.). Our study examined this risk factors using a questionaire. Seventy two respondents (owners of both aggressive and non-aggressive dogs) participated on our study. Our results suggest that owner's knowledge of natural behavior of dog, time which the owner dedicates to his dog and sex of the dog correlate significantly with canine aggression toward people. Inicialmente, os sujeitos responderam a dois estímulos por sessão (um positivo[S+] e outro negativo[S-]), até que quatro das seis caixas apareciam com ambas as funções. Na fase seguinte, as quatro caixas foram apresentadas juntas, de maneira que duas caixas de uma classe eram S+ e as outras duas S-. Foram realizadas mudanças de função até que os sujeitos alcançassem o critério de aprendizagem em 11 de 12 sessões. Discute-se os efeitos da ênfase na discriminação visual e o uso de posições fixas das caixas sobre o desempenho dos sujeitos. / The present study investigated the possibility of teaching two domestic dogs (Canis familiaris) to respond adequately to two classes of three stimuli (boxes with pieces of food when having S+ function) through a procedure of successive changes of simple discrimination (SCSD). Sessions were carried out in a room in which two, four or six wooden boxes were placed side by side. Initially, the subjects responded to two stimuli (a positive [S+] and another negative [S-]) for session, until four of the six boxes appeared with both of the functions. In the following phase, the four boxes were presented together, whith two boxes of a class functioning as S+ and the other two, as S-. Changes were performed until the subjects reached the criteria of learning in 11 out of 12 sessions. After that, the subjects were trained to respond to the two boxes that had not been used before, and then, the training with all six boxes was initiated. The test took place after a session of change with four boxes in which the two remaining boxes had been re-inserted in the experimental room. Having negative results in this test, training of SCSD was conducted with the six boxes, after which the test was remade. Both of the subjects had adequately responded to all the trials. Other five tests were conducted with the others boxes, in the same format of the first test. One of the subjects adequately responded to two of these five tests and the other to three of them. It is argued about the effect of the emphasis in the visual discrimination and the use of fixed positions of the boxes on the performance of the subjects.

Conservation Genetics of Wolves and their Relationship with Dogs

<p>Management of wolves is a complex issue, and molecular genetics is an important tool in this work. Molecular genetics can provide important information at the species, population and individual level, which can be essential for the development of management programs aiming at the long term survival of wolf populations.</p><p>In this thesis I developed new genetic markers on the canine Y chromosome to estimate the number of founders of the Scandinavian wolf population. This knowledge is important to reconstruct the history of the population and to design the most appropriate conservation strategies. Next, genetic markers with different pattern of inheritance have been used to identify hybrids between wolves and dogs. This allowed us to determine the direction of hybridization and to evaluate its possible impact on the gene pool of a wolf population. Furthermore, I also developed a method for a more reliable identification of the predator responsible of an attack by using saliva remains left on the prey. Since predation on livestock is perhaps the main reason for the negative opinions about the predator, the correct identification of the responsible for an attack (wolf, dog or hybrid) is essential. </p><p>Finally, this thesis has also been focusing on the domestication of dogs. By using Y chromosome markers (paternally inherited), it has been possible to complement previous studies based on mtDNA sequences (maternally inherited) and autosomal markers (inherited from both parents). In this way I have obtained a more complete picture of the domestication process and of the origin of breeds. This has shown that there has been a bias in the contribution of the two sexes in the origin of dog breeds (fewer males then females contributing to each breed) and that the origin of dogs was not marked by extensive backcrosses with male wolves over the entire species range.</p>

Determining the antiquity of dog origins: canine domestication as a model for the consilience between molecular genetics and archaeology

Archaeologists have favored a date of 14,000-15,000 years before present (BP) for canine domestication. However, recent studies of mutations in the mitochondrial DNA sequence by molecular geneticists have implied that dogs were domesticated over 100,000 years ago, which has challenged traditional theory. Geneticists have further hypothesized that dogs originated from wolf ancestors based upon the number of substitutions observed in dog and wolf haplotypes. Although both disciplines provide substantial evidence for their theories, the origin of dog domestication remains controversial. Several areas continue to be debatable. First, both geneticists and archaeologists incorrectly use the term domestication to describe events that clearly can not be proven to under human control. Second, the evolutionary development of canines is viewed by molecular biologists as well as archaeologist to be indicators of domestication without any further exploration of other probable causes. Third, the studies in canine genetics are so complex that most archaeologists have difficulty in providing evidence that would be contradictory to molecular theory. Fourth, both fields of study continually ignore innate behavioral characteristics of wolves that would make domestication highly improbable. Fifth, geneticists rely heavily on data gathered from sequencing of mitochondrial DNA, which has been assumed to maternally inherited. However recent human studies have shown that this assumption has now been proven to be incorrect. And finally, not only are morphological traits of fossilized dogs and wolves so similar that making a taxonomic identification improbable, but also the amount of archaeological remains available are too sparse and fragmented for accurate affiliation. An alternate theory of canine domestication will be proposed utilizing data gathered from the archaeological record and molecular research. I hypothesize that dogs diverged naturally from wolves 100,000 years ago as a result of the natural course of evolution, not human intervention, and had already evolved into a dog prior to being domesticated by humans 14,000-15,000 years ago. Evidence will be presented to clearly show that this hypothesis is a more accurate scenario of canine domestication.

Behaviours and experiences as indicators for the result in a behavioural test for dogs

In 2005 Swedish Armed Forces (SAF) started a breeding program of military working dogs. The dogs leave SAF’s kennel at an age of 8 weeks and live with puppy raisers. To evaluate the suitability of dogs for military work the dogs conduct a behavioural test at an age of 15-18 months. An “Index value” is extracted from this behavioural test. The puppy raisers answered a modified version of Canine Behavioral Assessment and Research Questionnaire (C-BARQ) when the dogs were approximately 12 months old. Answered questionnaires and results from the behavioural test were obtained for 59 dogs. Dogs that had passed the behavioural test had tendency for higher scores for “Trainability” (p = 0.078) and “If lived with other animals” (p = 0.066). Failing dogs had significantly higher score for “Stranger Directed Fear” (p = 0.006), ”Non-Social Fear” (p = 0.005), “Dog Directed Fear” (p = 0.021), “Hours of daily activation” (p = 0.001), “Mounting objects” (p = 0.012), and a tendency for higher risk of “Urinating when home alone” (p = 0.058). In a regressions between the “Index value” and the values of the questions from C-BARQ, the “Index value” was negatively correlated to “Stranger Directed Fear” (p = 0.002), “Non-social Fear” (p = 0.003), and “Dog Directed Fear” (p = 0.006). The “Index value” was positively correlated to “Trainability” (p = 0.013), “Hours left home alone” (p=0.043), “Hyperactive” (p = 0.018), “Chases shadows/light spots” (p = 0.043), and a positive tendency for “Chewing on inappropriate objects” (p = 0.075). From a PCA at the categories in C-BARQ, 3 components were extracted. All three components had a correlation to the “Index value”. The results show that the use of C-BARQ can indicate whether the dog will pass the behavioural test or not.

Amostras de sangue de 43 cães foram coletadas nos municípios de Acari, Caicó, Caraúbas e Marcelino Vieira e avaliadas quanto a infecção pelo T. cruzi por meio de testes sorológicos e pela Reação em Cadeia da Polimerase (PCR). Inicialmente, testes sorológicos como o DPP® (Dual Path Platform, BioManguinhos/FIOCRUZ) e ELISA (Enzyme-Linked Immuno Sorbent Assay) foram realizados para verificar a presença de anticorpos anti-Leishmania. Para a detecção de anticorpos anti-T. cruzi foram realizadoso ELISA in house para IgM e IgG e a reação de Imunofluorescência Indireta (RIFI). As capturas de triatomíneos foram realizadas nas mesmas localidades em que foram coletadas as amostras de sangue de cão. A infecção foi avaliada por exame direto e/ou PCR. Na sorologia para T. cruzi, 16,3% dos cães apresentaram reatividade sugestiva de fase aguda e 11,6% de fase crônica. A sorologia para Leishmania foi reativa em 9,3% dos animais dos quatro municípios investigados, sendo mais elevado no município de Acari com 12,5% . A reatividade entre os testes para Leishmania e T. cruzi foi de 23,2% das amostras, relacionada provavelmente com sobreposição /de áreas endêmicas. Na detecção do kDNA pela PCR, 41,9% das amostras apresentam a banda 330pb, as quais 20,9% eram do município de Caraúbas, 9,3% de Marcelino Vieira, 9,3% de Acari e 2,3% de Caicó. Nesses municípios, ninfas e adultos de Triatoma brasiliensis, Triatoma pseudomaculata e Rhodnius nasutus foram capturados em ambientes peridomiciliar e/ou intradomiciliar. As análises do conteúdo intestinal avaliados pela PCR apontaram 59,5% de positividade. Os resultados apontam elevada taxa de infecção de cães e triatomíneos pelo T. cruzi em diferentes municípios do Rio Grande do Norte, o que pode favorecer a infecção em humanos e a manutenção da infecção ativa pelo T. cruzi no Estado. / The infection by Trypanosoma cruzi in Canis familiaris and Triatomines was evaluated in the home and peridomiciliary environment of rural communities in the State of Rio Grande do Norte. Blood samples from 43 dogs from rural areas in the municipalities of Acari, Caicó, Caraúbas and Marcelino Vieira were evaluated for T. cruzi infection by serological techniques and by Polymerase Chain Reaction (PCR) to identify T. cruzi kDNA. Serology was initially performed to identify anti-T.cruzi antibodies by ELISA (Enzyme-Linked Immuno Sorbent Assay) for IgM and IgG in house and the Indirect Immunofluorescence (IFR) reaction. The presence of anti-Leishmania antibodies with DPP® (Dual Path Platform, Bio-Manguinhos / FIOCRUZ) and ELISA were also used. The catches of triatomines were carried out in the same locations and in the same period in which the dog samples were collected and the infection evaluated by direct examination and/or PCR. The criterion for positivity in T. cruzi serology was the agreement between two different techniques, which resulted in 16.3% (7/43) of the samples suggestive of acute phase and 11.6% (5/43) of chronic phase. The reactive serology for Leishmania was 9.3% of the animals in the four municipalities studied, being more reactive in the municipalities of Acari. These results of reactivity for Leishmania and T. cruzi may be related to the overlap of endemic areas. In the identification of kDNA by PCR in blood samples from dogs, 41.9% (18/43) of the samples present the band 330pb, of these 6.8% (4/43) are in the municipality of Acari, 2.3% (1/43) in Caicó, 20.9% (9/43) in Caraúbas and 9.3% (4/43) in Marcelino Vieira. The catches of triatomines identified adults and nymphs of T. brasiliensis, T. pseudomaculata, R. nasutus and adults of P. lutzi in peridomiciliary and/or intradomiciliary environments of all the studied localities. The analysis of the intestinal contents evaluated by PCR found positivity of 78.5% (95/121) of the triatomines that cohabitated these environments with the dogs. The natural T. cruzi infection was identified in dogs and triatomines in all municipalities studied, probably helping to maintain active T. cruzi infection in the domiciliary area and increasing the risk of infection in the human population.

Resistance is caused by deletion of four nucleotids, that resulting in a frame shift and synthesis of nonfunctional transport of P-glycoprotein. The text describes a polymorphism of MDR1 (ABCB1) gene, which results in reduced resistance to drugs belonging to the group of macrocyclic lactones. It also describes inheritance of this phenomenon and it deals with the detection of mutation using PCR (polymerase chain reaction) and by fragmentation analyses. A review of literature study is a form of research solely from scientific publications. 128 dogs were included into the own analysis. The results confirmed that Collies had the highest presence of deletions (29,73 %) with a high number of carriers in the study population of dogs (54,05 %). The percentage of affected individuals in the breed of Australian Shepherd and Sheltie was significantly lower (7,32 % and 6 %), but the percentage of carriers were also high in both Australian Shepherds (34,14 %) and the breed Sheltie (48 %).

Desenvolvimento folicular em fragmentos ovarianos de cadelas (Canis lupus familiaris) cultivados em meios "MEM" suplementado com soro de cadela no proestro e cadela gestante / Follicular development in ovarian fragments of female dogs (Canis lupus familiaris) cultivated in "MEM" supplemented with dog serum and pregnant

Após a colheita dos ovários, foram obtidos fragmentos da zona parenquimatosa de cada ovário, um dos quais foi incluído fresco como controle, e os demais incubados por 48 horas em atmosfera de 5 % de CO 2 , 95 % de umidade relativa e temperatura de 39 oC, em diferentes tratamentos: T1: cultivados em Meio Essencial Mínimo (MEM); T2: cultivados em MEM associado a Soro de Cadela prenhe (MEM + SCpre) e T3: cultivados em MEM associado a Soro de Cadela no pró-estro (MEM + SCpro). Todos os fragmentos foram fixados por imersão em solução de Karnowisky por até 24 horas e processados segundo rotina para confecção de lâminas histológicas. Estas foram avaliadas quanto ao desenvolvimento folicular, sob microscópio de luz, em aumento de 100 vezes. A análise estatística foi feita pelo teste paramétrico de comparação de médias T Student. Todos fragmentos cultivados apresentaram aumento na proporção de folículos em desenvolvimento, assim conclui-se que o meio de cultivo utilizado associado à técnica de cultivo in situ foi suficiente para promover o desenvolvimento folicular em ovários de cadelas, sendo que a suplementação hormonal com SCpro e SCpre não influenciaram na taxa de desenvolvimento. / This present study evaluated different protocols of in vitro culture from ovarian fragments of domestic female dog (Canis lupus familiaris) in anestrus. For the study, ovaries from female dogs were used, obtained from the neutering of the stray animal population control of the municipal kennel program/ Veterinary Department - Universidade Federal de Viçosa (UFV). After the ovaries were collected, random fragments of the ovary parenchymal were obtained, one of them were used fresh as control, and the rest were incubated for 48 hours in 5 % of CO2 atmosphere, 95 % of relative humidity and 39 oC temperature, in different treatments: T1 cultivated in Minimum Essential Medium (MEM); T2 cultivated in MEM associated with pregnant dog serum and T3 cultivated in MEM associated with proestrus dog serum. Those ovaries cultivated in treatments T1, T2 and T3, were fixed by Karnowisky immersion for up to 24 hours, and processed following the normal laboratorial procedures for making histological slides. They were then evaluated under light microscope, using the 100X objective lens. The statistical analysis was made by the Student T`s parametric comparison measures test. All fragments of assessed ovaries showed a proportional increase of developing follicles, concluding that the culture medium used, associated to the in situ culture technique, was enough to promote the follicle development in dog’s ovaries, being that the hormonal supplementation with proestrus serum and pregnant serum did not influence the development rate.
