The soluble fibers, as beta-glucan, present this property and have been studied about yours physiologic and technologic effects. The beta-glucan produce a gel with high viscosity, and research with the objective of alter the viscosity and swelling power for facilitate your incorporation in the foods has been accomplished. However, these modifications can alter the physiologic effects. Besides, don t exist studies about effect of oxidative treatment on the beta-glucan properties. The objective of this study was evaluate the effect of oxidative treatment with hydrogen peroxide in different concentrations (0,3; 0,6; 0,9%) and two times of reaction, 30 and 60 minutes, on the beta-glucan from oat and, later, apply in cheese bread in 2, 3, 4% levels. the oxidative treatment increase the carbonyl and carboxyl groups, affected the swelling power and increase glucose release after chemic digestion. The oxidative treatment increase bile acid binding capacity, however, didn t alter the fat binding capacity. Has decrease in hardness, adhesiveness and gumminess, as well as viscosity of gel with the oxidative treatment. The oxidized beta-glucan decrease the expansion factor and increase the firmness of cheese bread, with exception of the treatment with native beta-glucan at 2% and oxidized with 0,9% of H2O2/30 min at 3%. / Em vista da crescente preocupação com a saúde, a indústria e o mercado de alimentos buscam proporcionar aos consumidores alimentos com propriedades funcionais. As fibras solúveis, como a beta-glicana, apresentam essa propriedade e estão sendo estudadas quanto a seus efeitos fisiológicos e tecnológicos. A betaglicana forma um gel de alta viscosidade e, para facilitar a sua incorporação aos alimentos, são realizadas pesquisas de modificação da beta-glicana, com o intuito de alterar as suas características como viscosidade e poder de intumescimento. No entanto, com essas modificações, os seus efeitos fisiológicos podem também ser alterados. Além disso, não há estudos sobre o efeito de tratamentos oxidativos sobre as propriedades da beta glicana. O objetivo deste estudo foi avaliar o efeito do tratamento oxidativo com peróxido de hidrogênio em diferentes concentrações (0,3; 0,6 e 0,9%) com dois tempos de reação, 30 e 60 minutos em beta-glicana extraída da aveia e, posteriormente, utilizá-la na formulação de pães de queijo nos níveis 2, 3 e 4%. O tratamento oxidativo promoveu aumento de grupos carbonila e carboxila, alterou o poder de intumescimento da beta-glicana e aumentou a liberação de glicose após digestão química. A capacidade de ligação com ácidos biliares aumentou com os tratamentos oxidativos, entretanto, não houve alteração da capacidade de ligação com gordura. Houve diminuição da dureza, adesividade e gomosidade, bem como da viscosidade do gel com os tratamentos oxidativos. A adição de beta-glicana oxidada promoveu diminuição do índice de expansão dos pães de queijo, e aumento da firmeza, com exceção dos tratamentos com betaglicana nativa a 2% e oxidada com 0,9% de H2O2/30 min a 3%.

Rational Synthesis, Stabilization, and Functional Properties of Metal and Intermetallic Nanoparticles

The confluence of intriguing size and morphology dependent optical and chemical properties with versatile application in various fields, such as energetic and magnetic makes monometallic nonmaterial of high fundamental scientific interest. However, the challenge that needs to be addressed is to achieve their synthesis with a rational control on their dimensions, morphology and dispersion for the widespread applications of these materials. In addition to synthesis, achieving long-lasting stability of nonmaterial becomes imperative in order to realize their potential applications. Miniaturization in size of particles results in an increased surface to volume ratio, conducing especially reactive metal nanoparticals prone to oxidation. This thesis describes the synthesis of nearly monodiperse colloids of metallic and intermetallic nanoparticles using solvated metal atom dispersion method and digestive ripening facilitated interatomic diffusion process. Our aim is to understand the combinatiorial effects of nanosizing and stability on the functional properties of these nanomaterials. Towards this Direction, we investigated Co, A1 and Mg monometallic, and Au/Ag-In and Au-Sn intermetallic nanoparticle systems. Chapter 2 Describes the synthesis, detailed characterizations and magnetic properties of nearly monodisperse cobolt nanoparticles(<5nm) synthesized using a hydride synthetic protocol, solvated metal atom diserion method. The as-prepared cobalt nanoparticles in this size range exhibit intrinsic instability towards Oxidations. After 30 day of exposure to air, magnetic measurements showed drastic degration in saturation magnetization and complete conversion to antiferromagnetic cobalt oxide was confirmed. In order to achieve their stability, a heat treatment was applied to decompose the organic solvent and capping agent, resulting in carbonization of solvent/ligand around the surface of cobolt nano particles. Controlled and optimized annealing at different temperatures resulted in the formation of hexagonal closed packed (hcp) and fape-centered cubic (fcc) phases of metallic cobalt. Remarkably, the corresponding heat treated samples retained their rich magnetic behavior even after exposure to air for a duration of one year. Compared to un-annealed samples, magnetization values increased two-fold and the corecivity of nanoparticles exhibited strong dependence on the phase transformation of cobolt. Chapter 3 Deal with an exploratory study of the synthesis, characterization, and stabilization of nanometer-sized enegetic material, aluminum. Highly monodisperse colloidal aluminum nanoparticles (3.1‡ 0.6 mm) were prepared by using hexadecy amine (HAD) as the capping agent tetrahydrofurma as a coordinating solvent in the SMAD method. Since such small particles are highly prone to oxidation, a support materials is required for their stabilization. Stability has been achived by carbonization of the capping agent on the surface of A1 nanoparticles by carrying out thermal treatment of A1-HAD nanoparticles at a modest temperature. Presence of corbon was confirmed using Raman spectroscopy and TEM measurements evidencing that annealed A1 nanoparticles are encapsulated in a corbon matrix. The exhibition of robust stability was established using thermal analysis (TGA/DTA) wherein, oxidation of aluminum in air did not occur upto 500 0C. Indirectly, the successful passivation was further exploited in the synthesis and characterization of small sized monodisperse magnesium nanoparticles. The resulting samples were hybrided and nanosized MgH2 released hydrogen at much lower temperature than that of the bulk MgH2 (573 K). The observed hydrogen release was only partially reversible. This partial reversibility could be attributed to the coalescence of small sized Mg nanoparticles upon subsequent charging/discharging hydrogen cycles. In the next step, we exploed the intermetallic systes which are composed of more than one metallic species. Chapter 4 describes the synthesis and characterization of small sized, monodisperse (<10 nm) colloidal AuIn2 and Ag3In intermetallic nanoparticles. The formation of intermetallic nanoparticles could be explained by invoking digestive ripening facilitated atomic diffusion of Au/Ag and In nanoparticles followed simultaneously by their growth in te solution. The course of the reaction was followed using optical spectroscopy where the changes in UV-visible absorption band were correlated to the formation of AuIn/Ag3In intermetallic. Structural characterization, Performed using powder X-ray diffraction, brought out the formation of phase pure AuIn2 and Ag3In intermetallic compounds. Digestive ripening effects were clearly observed using transmission electron microscopy which showed the transformation of polydisperse physical mixture colloid of nanometallic species to uniform sized intermetallic nanoparticles. By invoking the phenomenon of interatomic diffusion at nanoscale favored by feasible thermodynamics ( G being negative) we were successful inrealizing the formation of these intermetallic nanoparticles. Optimization of temperature at which digestive ripening was performed, turned out to be a crucial factor in the successful synthesis of phase pure intermetallic nanoparticles. These promising results inspired us to study further the preparation of Au-Sn intermetallic system which is described in Chapter 5. The potential of such an unprecedented approach has been exploited in the synthesis of homogeneous intermetallic nanaocrystals of Au5Sn and AuSn. The two monometallic collids (Au and Sn), mixed in a stoichiometric amount were subjected to digestive ripening process. 1:1 stichiometry always led to the formation of eutectic mixture (Au5Sn and AUSn), The stoichiometry of monometallic nanocrystals. Therefore, by taking an extra equivalent of Au and Sn in two different experiments, phase pure Au5Sn and AuSn intermatillic nanocrsytals were obtained, respectively. This is the first observation that has been reported regarding the phase pure synthesis if Au5Sn intermetallic nanocrystals using solution based approach. Formation of different phases was established by structural characterization which elicited srystalline nature of the samples. A combination of TEM, HRTEM, and STEM-EDS mapping techniques employed here, brought and tailored phase. In conclusion, the careful selection of solvent, stoichiometry and growth directing agents is an important prerequisite for realizing distinct phases of Au-Sn system with a controlled morphology.

Les peptides : un levier pour orienter les fonctionnalités de matrices riches en caséines / The peptides : a lever to direct the functional properties of matrices highly concentrated in caseins.

Les aliments sont des matrices de composition complexe et variée au sein desquelles les protéines possèdent diverses fonctionnalités, pour stabiliser des mousses ou des émulsions, gélifier, solubiliser certains composés menant à différentes textures finales du produit. Une modification de ces protéines par hydrolyse enzymatique ménagée permet d’améliorer certaines de ces fonctionnalités et donc de changer les propriétés rhéologiques et structurales du produit. Les peptides dérivés des protéines sont présents dans de nombreux aliments soit par production in situ soit par incorporation lors de la formulation des produits sous forme d'hydrolysats. Des questions subsistent pour savoir i) si des règles générales d’assemblage existent ou non pour expliquer comment les peptides et les protéines interagissent entre eux et ii) et si une réorganisation se fait au sein des matrices ici laitières pour induire de nouvelles propriétés fonctionnelles ou les modifierL’objectif de la thèse était de déterminer comment et jusqu’à quel niveau les peptides sont capables de modifier la texture, la rhéologie et la microstructure des matrices à haute teneur en caséines.Nous avons ainsi pu montrer i) quels changements de structure et de texture avaient lieu à différentes échelles en variant le type de peptides produits, leur proportion et la concentration de la matrice dans laquelle ils étaient ajoutés et ii) les interactions entre peptides et caséines et la non-linéarité des phénomènes expliquant les évolutions parfois contradictoires des propriétés de texture des matrices caséiniques. / Food products are matrices of complex and varied composition, among which proteins are the main molecules possessing various functionalities, i.e. foam and emulsion stabilisation, gelation, solubilisation of some components. They led to various final textural properties of the food products. Modification of proteins by mild enzymatic hydrolysis, allowed improving some of the functionalities and in turn changing the structural and rheological properties of the food product. Protein-derived peptides are present in a huge number of food products either by in situ production or by incorporation as an ingredient during food formulation. Questions remains i) whether some general rules can be put forward or not to explain the building-up of the interactions between peptides and between peptides and proteins and ii) the subsequent reorganization of the food matrices and namely dairy ones that has to be done to induce new or modified functional properties in various dairy matrices.The objective of this thesis was to determine how and to what extent casein-derived peptides are able to modify the texture, the rheology and the microstructure of highly-concentrated casein matrices. We showed i) what kinds of structural and textural changes occurred at various scales from microscopic to macroscopic ones, when we varied the type of peptides produced, their percentage in the dairy matrices and the concentration of the matrices they were incorporated in; ii) interactions between peptides and caseins and the non-linearity of the phenomena explaining some adversely evolutions of the rheol

Os grãos de soja foram divididos em quatro lotes, sendo que dois foram irradiados em fonte de 60Cobalto, com doses de 2,0 ou 5,0 kGy, um lote foi irradiado em acelerador de elétrons com dose de 2,0 kGy e um lote foi utilizado como controle (não irradiado). Os lotes foram armazenados por um período de 12 meses. A cada quatro meses, uma porção de cada lote foi retirada para produção laboratorial de farinhas desengorduradas (FDSs) e isolados protéicos (IPSs). A composição centesimal nos grãos, FDSs e IPSs foi determinada. Nos grãos foram analisados a atividade de água, teor de isoflavonas e atividade antioxidante (IA) e nas FDSs, os teores de fitato e inibidores de tripsina (IT). O teor de acidez foi avaliado nos óleos obtidos do desengorduramento das farinhas. As características estruturais das proteínas dos IPSs obtidos foram avaliadas por análises de eletroforese em gel de poliacrilamida na presença de dodecil sulfato de sódio (SDS-PAGE) em condições não redutoras, das proteínas solúveis em diferentes sistemas, calorimetria diferencial de varredura (CDV), grupos sulfidrila livres (SH), fluorescência intrínseca (FI), cromatografia liqüida de alta eficiência de fase reversa (CLAE-FR) e cromatografia liqüida de alta eficiência de exclusão molecular (CLAE-EM). As propriedades funcionais avaliadas foram: solubilidade, gelificação, capacidade de retenção de água (CRA), capacidade emulsificante (CE), índice de atividade emulsificante (IAE) e estabilidade de emulsão (EE). O teor das isoflavonas totais e o índice de antioxidação, analisados nos grãos, não diferiu significativamente (p>0,05) e permaneceu estável ao longo do armazenamento, independente do tipo ou dose de radiação. No entanto, houve aumento das concentrações relativas dos b-glicosídeos e diminuição dos malonil glicosídeos ao longo do armazenamento. O teor de acidez aumentou em até 81,6% no óleo de grãos irradiados com dose de 5 kGy e armazenados por doze meses. As FDSs de grãos irradiados apresentaram os menores valores dos inibidores de tripsina e fitato. As análises de calorimetria diferencial de varredura, fluorescência intrínseca e cromatografia liqüida de alta eficiência de exclusão molecular indicaram não ter ocorrido alterações estruturais importantes nos isolados protéicos devido à irradiação ou ao armazenamento. Entretanto, os grupos sulfidrila livres aumentaram nos IPSs de grãos irradiados, mas sofreram pouca alteração ao longo dos 12 meses de armazenamento. Houve aumento da proporção relativa de frações hidrofóbicas observadas por CLAE-FR em todos os isolados após quatro meses de armazenamento dos grãos. Os IPSs de grãos irradiados apresentaram maior solubilidade protéica e capacidade emulsificante, enquanto que a estabilidade de emulsão e o índice de atividade emulsificante não diferiram significativamente (p>0,05). Os valores de dureza e capacidade de retenção de água dos géis dos IPSs não diferiram significativamente (p>0,05) em função do tratamento empregado. No entanto, os géis dos IPSs de grãos armazenados tiveram maiores valores de dureza. Os valores coesividade e elasticidade dos géis dos isolados, independente da dose e tipo de radiação, não diferiram significativamente (p>0,05). Nas condições estudadas, as radiações gama com doses de até 5 kGy e de feixe de elétrons com dose de 2 kGy podem ser utilizadas sem prejuízo as propriedades químicas de grãos e funcionais de seus isolados protéicos, durante o armazenamento / Abstract: The aim of this work was to study the effects of both irradiation and storage on the chemical composition of soybean grains and on the physicochemical and functional properties of their protein isolates. The soy grains were divided into four lots, two of them were irradiated with of 60Cobalt source in doses of 2.0 or 5.0 kGy, a third one was irradiated with an electron beam at 2.0 kGy and another was used as control (non-irradiated). All the lots were stored for a maximum of 12 months. Every four months, a portion of each lot was removed to produce deffated soy flour (DFS) and soy protein isolate (SPI). The proximal compositions of the grains, FDSs and SPIs were determined. In the grains, the water activity, total isoflavone content and antioxidant index (AI) were measured and in DFSs, the phytate content and trypsin inhibitor activity (TIA). The acidity content was measured in the oil obtained from the whole flours. The structural characteristics of SPIs were evaluated by polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulphate (SDSPAGE) under non reducing conditions of the soluble proteins using different systems. Additionally, differential scanning calorimetry (DSC), free sulfhydryl groups (SH), intrinsic fluorescence (IF), reversed-phase high performance liquid chromatography (RP-HPLC) and of size exclusion high performance liquid chromatography (SE-HPLC) were also used to evaluate the effects. The analyses of the functional properties in SPIs were: solubility, gelation, water holding capacity (WHC), emulsifying capacity (EC), emulsifying activity index (EAI) and emulsifying stability (ES). The total isoflavone content and antioxidant index analyzed in the grains did not differ significantly (p>0.05) and remained stable throughout storage, as a function of the dose or type of irradiation. However, there was an increase in the b-glycosides and decrease in the mean percentage of malonyl glycosides. The acidity content increased in the percentage of 81.6% in the oil grains irradiated with 5 kGy and stored for 12 months. DFSs of irradiated grains exhibited smaller values of the trypsin inhibitor and phytate. The analyses of differential scanning calorimetry, intrinsic fluorescence and size exclusion high performance liquid chromatography showed that no important structural alterations occurred in the protein isolates due irradiation or storage. However, the free sulfhydryl groups increased in SPIs of irradiated grains, without further significant changes along the storage. There was an increase of the proportion of the hydrophobic fractions, showed by reversed-phase high performance liquid chromatography profile, for all the isolated after four months of storage of the grains. SPIs of irradiated grains presented high solubility, emulsifying capacity while emulsifying stability and emulsifying activity index did not differ significantly (p>0.05). The values of hardness and water holding capacity of the SPIs gels did not differ significantly (p>0.05) for all treatment. However, SPIs gels of stored grains had high values of hardness. The values cohesiveness and elasticity of the SPIs gels did not differ significantly (p>0.05), independent of the dose and type of irradiation. A maca (Lepidium meyenii Walpers) é uma planta herbácea bienal da família Brassicae, cultivada principalmente na região dos Andes da América do Sul. A parte subterrânea vem sendo consumida por muito tempo devido a seu valor nutricional e energético, mas é mais conhecida no mercado peruano e internacional por alegadas propriedades terapêuticas. Esta raiz apresenta até 76% de carboidratos, dos quais 30% é amido. Este trabalho teve como objetivos estudar: as propriedades físico-químicas e funcionais do amido isolado; os parâmetros enzimáticos durante o armazenamento e a purificação parcial de enzimas amilolíticas. Em relação às propriedades do amido, este apresentou um teor de amilose de 20% valor semelhante aos encontrados em raízes e tubérculos similares. A turbidez das suspensões de amido apresentou estabilidade durante o armazenamento. A temperatura de gelatinização e a viscosidade da pasta foram a 45,7° e 46°C, respectivamente. Com base nos dados obtidos, o amido de maca seria indicado para alimentos que requeiram temperaturas moderadas no processamento, não sendo apropriado para o emprego em alimentos congelados. Os parâmetros enzimáticos medidos tais como teor de amido total, teor de açúcares solúveis, atividade amilolítica total, atividade de &#945; e &#946; amilases, não mostraram diferenças significativas entre as medidas durante um período de armazenamento de 16 dias. As microscopias eletrônicas de varredura (MEV) dos grânulos de amido mostraram grãos íntegros com superfícies lisas, com algumas depressões ao redor dos grânulos os quais poderiam indicar o inicio de ataque enzimático, ou fraturas na purificação. Em relação à purificação de enzimas amilolíticas, foi possível separar uma fração ativa com a carboximetilcelulose (CMC) seguida de cromatografia liquida de alta resolução (CLAE) que permitiu a separação de duas frações protéicas, analisadas por eletroforese SDS-PAGE e eletroforese bidimensional (2D). Os polipeptídeos identificados no gel 2D apresentaram massa molecular semelhante entre 22 a 27 kDa, e os pontos isoelétricos entre 4,8 e 7,3. / Maca (Lepidium meyenii Walpers) is a biennial herbaceous plant from Brassicae family, grown mainly in the Andes of South America. The gelatinization temperature and viscosity of the paste were 45.7 ° and 46 ° C, respectively. The polypeptides had a molecular mass between 22 and 27 kDa and isoelectric points ranging from 4.8 to 7.3.

Optimizacija tehnološkog procesa proizvodnje funkcionalnog fermentisanog mlečnog napitka / Optimisation of functional fermented dairy beverage technology

<p>U okviru doktorske disertacije ispitana je mogućnost proizvodnje<br />fermentisanog mlečnog napitka uz primenu različitih vrsta i koncentracija<br />nekonvencionalnog startera čajne gljive: a) nativnog inokuluma-I<br />(10% i 15%) b) koncentrovanog mikrofiltracijom - MFI (10% i 15%) i c)<br />koncentrovanog uparavanjem - UPI (1,5%, 3,0%, 10% i 15%). Varijante<br />fermentisanog mlečnog napitka dobijene su kori&scaron;ćenjem mleka sa 0,9%<br />i 2,2% mlečne masti. Za pobolj&scaron;anje fizičko-hemijskih i teksturalnih<br />karakteristika proizvoda u mleko pre inokulacije dodato je 0,02% enzima<br />transglutaminaze (aktiviran na 40&deg;C, a posle 2 sata je inaktiviran na<br />80&deg;C, 1 minut). Fermentacija mleka (pri temperaturi 42&deg;C) uz kori&scaron;ćenje<br />inokuluma čajne gljive praćena je do postizanja pH vrednosti 4,5.<br />Nakon proizvodnje analizirana je nutritivna vrednost fermentisanih<br />mlečnih napitaka: sadržaj masti, proteina, laktoze, galaktoze, glukoze,<br />fruktoze, mlečne i sirćetne kiseline, etanola, vitamina (B1, B2, B6 i<br />C), minerala (Ca, Mg, K, Na, Pb, Cd, Cu, Zn) i masnih kiselina. Sadržaj<br />odabranih komponenata u napicima praćen je tokom 10 dana skladi&scaron;tenja.<br />Fizičko-hemijske osobine (kiselost, sinerezis i sposobnost vezivanja<br />vode), teksturalne karakteristike (čvrstoća, konzistencija, kohezivnost i<br />indeks viskoziteta) i viskozitet uzoraka praćeni su nakon proizvodnje i<br />posle 10 dana skladi&scaron;tenja. Ukupan broj bakterija sirćetne kiseline (BSK)<br />i kvasaca u fermentisanim napicima analiziran je nakon proizvodnje i<br />tokom skladi&scaron;tenja. Varijante fermentisanih mlečnih napitaka senzorno<br />su ocenjene nakon proizvodnje.<br />Na osnovu rezultata ispitivanja utvrđeno je da fermentacija mleka<br />uz primenu nekonvencionalnog startera čajne gljive predstavlja složen<br />biohemijski proces pri čemu fermentacija traje od 9 do 12 sati i prosečno<br />je dva puta duža od fermentacije mleka u proizvodnji probiotskog i<br />tradicionalnog jogurta. Sadržaj laktoze tokom fermentacije mleka manji<br />je prosečno za 19,6% u odnosu na sadržaj laktoze u mleku. Uzorci<br />proizvedeni sa nativnim inokulumom uz primenu transglutaminaze i<br />koncentrata proteina surutke imaju za 15% niži sadržaj laktoze, a uzorak<br />15% I za 2% niži sadržaj u odnosu na uzorak 10% I. Uzorci proizvedeni<br />sa 10% MFI i 15% MFI imaju za 11%, odnosno 21% niži sadržaj laktoze<br />nakon proizvodnje u poređenju sa odgovarajućim uzorcima proizvedenim<br />uz primenu nativnog inokuluma. Sniženje sadržaja laktoze je<br />praćeno povećanjem sadržaja galaktoze i L-mlečne kiseline u svim uzorcima.<br />D-mlečna kiselina, sirćetna kiselina i etanol zastupljeni su u veoma<br />malim koncentracijama.<br />Imajući u vidu značaj vitamina C u ishrani i količinu detektovanu<br />u fermentisanom mlečnom napitku, konzumiranjem 500 g napitka<br />dobijenog primenom čajne gljive mogu da se zadovolje dnevne potrebe<br />za vitaminom C.<br />Tokom proizvodnje fermentisanih napitaka sadržaj vitamina B1<br />poraste za 5,09%, vitamina B2 za 2,65% i B6 za 2,03% u odnosu na<br />mleko. U fermentisanom mlečnom napitku iz mleka sa 2,2% masti<br />sadržaj vitamina B6 je povećan 24,76% u odnosu na sadržaj u polaznom<br />mleku. Fermentisani mlečni napici proizvedeni primenom čajne<br />gljive iz mleka sa 0,9% i 2,2% masti sa dodatkom 10% inokuluma<br />čajne gljive sadrže najvi&scaron;e vitamina B2 koji iznosi 108 (&mu;g/100g), odnosno<br />136 (&mu;g/100g). Mikrobiolo&scaron;ki sastav napitaka se razlikuje u zavisnosti<br />od vrste i sastava kori&scaron;ćenog inokuluma. Najveći broj BSK<br />(4,5 &middot; 104ćelija/mL) i kvasaca (9 &middot; 104 ćelija/mL) je u uzorku 10% MFI,<br />dok je manji u napitku 10% I (BSK: kvasci= 4,05 &middot; 104: 4,5 &middot; 104).<br />Dodatkom transglutaminaze u minimalnoj koncentraciji od<br />0,02% postižu se znatno bolje fizičko-hemijske i reolo&scaron;ke karakteristike<br />fermentisanih mlečnih napitaka dobijenih iz mleka sa 0,9% masti.<br />Generalno posmatrano, kori&scaron;ćenjem 10% nativnog inokuluma za<br />proizvodnju napitaka iz mleka sa 0,9% i 2,2% masti dobijaju se proizvodi<br />optimalnih hemijskih, mikrobiolo&scaron;kih i senzornih karakteristika.<br />Funkcionalni fermentisani mlečni napici proizvedeni uz primenu različitih<br />vrsta i koncentracija čajne gljive su proizvodi visoke nutritivne<br />vrednosti namenjeni različitim kategorijama potro&scaron;ača.</p> / <p> The possibility of fermented milk beverages manufacture<br /> by applying non-convenctional starter culture - tea fungus<br /> inoculum: a) natural inoculum (10% and 15%); b) concentrated<br /> by microfiltration (10% i 15%); c) concentrated by evaporation<br /> (1.5% and 3.0%) have been investigated. All fermented milk samples<br /> were produced from milk of 0.9% and 2.2 % fat content.<br /> Improvement of rheological properties of low fat fermented milk<br /> products by addition of enzyme transglutaminase (TG) was<br /> achieved. Transglutaminase (Activa MP, Ajinomoto Co. Physico-chemical characteristics (acidity, syneresis,<br /> water-holding capacity), textural characteristics (firmness,<br /> consistency, cohessivnes, and index of viscosity) and viscosity<br /> were measured at 5&deg;C after production and during 10 days of<br /> storage.<br /> On the basis of the obtained results it can be concluded<br /> that milk fermentation lasted from 9 to 12 hours and is two times<br /> longer compared to fermentation during probiotic and traditional<br /> yoghurt production. Lactose content decreased during fermentation<br /> for 19.6% compared to milk. Higher transformation of lactose<br /> was determined in samples produced by using microfiltration<br /> inoculum. Decreasing of lactose content affected the increased<br /> galactose and L-lactic acid content. Content of D-lactic acid,<br /> acetic acid and ethanol were detected in low concentrations. High<br /> concentration of ascorbic acid in beverage contributed that 500 g<br /> fermented milk beverage could fullfill recommended daily intake<br /> for vitamin C. Sample produced from milk of 2.2% fat content<br /> using 10% natural inoculum contained the highest concentration<br /> of vitamin B2 &ndash; 136 (&mu;g/100g).<br /> The highest total cell count of acetic acid bacteria was<br /> 4.5 &middot; 104 cfu/ml and yeasts 9 &middot; 104 cfu/ml in samples produced by<br /> using 10% microfiltrated inoculum. Propriedades funcionais da carne de ratitas : ema (Rhea americana) e avestruz (Struthio camellus) comparadas com a carne de frango (Gallus domesticus) /

Para tanto, analisaram-se a solubilidade das proteínas miofibrilares, a capacidade de retenção de água, a capacidade de emulsificação das proteínas totais, a estabilidade da emulsão e as perdas por cocção, por evaporação e por gotejamento. Obtiveram-se os seguintes resultados por meio das análises da composição nutricional do músculo Iliotibialis lateralis das aves: avestruz: 19,59 % de proteínas, 75,53 % de umidades, 1,37 % de lipídeos, 1,13 % de cinzas e 2,38 % de carboidratos; ema: 23,50 % de proteínas, 74,19 % de umidades, 1,11 % de lipídeos, 1,13 % de cinzas e 0,07 % de carboidratos e frango: 18,35 % de proteínas, 73,30 % de umidades, 5,92 % de lipídeos, 1,03 % de cinzas e 1,40 % de carboidratos. Por meio da análise das propriedades funcionais observou-se que as proteínas solúveis no NaCl (PS) e as proteínas extraídas com NaCl das proteínas totais (PT) apresentaram, respectivamente, os seguintes resultados: pH 7.0: avestruz (7,40 % e 38,06 %), ema (8,52 % e 36,11 %) e frango (7,68 % e 41,88 %); pH 5.5: avestruz (5,87 % e 30,18 %), ema (6,53 % e 28,54 %) e frango (5,65 % e 29,97 %) e pH 3.0: avestruz (3,04 % e 15,64%), ema (3,29 % e 13,97 %) e frango (2,85 % e 15,52 %). Na análise da capacidade de retenção de água obteve-se para avestruz (73,19 %), ema (66,25 %) e frango (67,13 %). Com relação à capacidade de emulsificação no pH 7.0 foram obtidos estes resultados: avestruz (21,42 mL óleo/ 100 mg de proteína), ema (18,98 mL óleo/ 100 mg de proteína) e frango (31,87 mL óleo/ 100 mg de proteína). Variation B-carotene content and physicochemical properties of orange-fleshed sweet potato (Ipomoea batatas (L.) Lam) cultivars grown in Limpopo Province, South Africa

Lam] cultivar, as a cheaper source of vitamin A. In Southern Africa, reports have shown that one in every three children has deficiency in vitamin A. This study investigated the variations in β-carotene (vitamin A precursor) content among selected OFSP cultivars grown in Limpopo Province, South Africa. In addition, the study investigated the effects of location on functional properties, total carotenoids and antioxidant profile of the sweet potato flour as well as their physicochemical, functional and nutritional properties. Four (4) sweet potato cultivars [Dagga, Bophelo, Impilo (orange fleshed) and Mvuvhelo (cream fleshed)] were obtained from two locations, namely, University of Venda (Univen) agricultural experimental farm and Tshiombo irrigation scheme (Tshiombo) and analysed for pasting, functional, physicochemical and nutritional properties. The sweet potato flours from these two locations were also analysed by computed tomographic (CT) scans for flour particle density. Mineral and microstructure of OFSP flours were analysed by scanning electron microscope (SEM). All measurements, except the tristimulus L* a* b* and other colour parameters from five replicates, were performed in triplicate. Statistical analysis was performed using a one-way analysis of variance and means were separated using Duncan’s multiple range test; P < 0.05 was considered to be statistically significant. Pearson’s correlation coefficients were used to determine the correlation among functional, physicochemical, nutritional and pasting properties of sweet potato flour. β-carotene content varied significantly between the two locations from 70.98 ± 0.8 μg/g (Tshiombo) to 86.09 ± 2.0 μg/g (Univen), among cultivars from 1.71 ± 0.0 μg/g (Mvuvhelo) to 201.50 ± 1.0 μg/g (Dagga) and among orange fleshed (Dagga, Bophelo and Impilo) flours from 28.38 ± 0.2 μg/g (Impilo) to 201.50 ± 1.0 μg/g (Dagga). There was a similar trend in the contents of α-carotene to that of β-carotene from the two locations, which ranged from 0.63 ± 0.0 mol/L (Tshiombo) to 1.01 ± 0.0 mol/L (Univen). Mvuvhelo had significantly the lowest α-carotene content (0.50 ± 0.0 mol/L) and Impilo the highest (1.28 ± 0.0 mol/L). The total carotenoids was highest for Impilo (7.56 μg/g) and lowest for Mvuvhelo (1.33 ± 0.1 μg/g) but did not vary significantly (P > 0.05) between ii the locations. The antioxidant activity, based on 2, 2-diphenyl-1picrylhydrazyl (DPPH) free radical scavenging activity, was significantly highest in Bophelo (63.37 ± 0.8 mMAAE ml-1) and lowest in Dagga (26.93 ± 1.4 mMAAE ml-1) flours. The ferric reducing antioxidant power (FRAP) varied significantly across the locations and among the cultivars and ranged from 14.45 ± 0.3 μMTE ml-1 (Tshiombo) to 17.40 ± 1.1 μMTE ml-1 (Univen). FRAP was significantly highest in Impilo (21.12 ± 2.1 μMTE ml-1) and lowest in Mvuvhelo (8.16 ± 2.0 μMTE ml-1). Total carotenoid content positively correlated with DPPH (0.486*), FRAP (0.830**), β-carotene (0.370*), and α-carotene (0.424*). The antioxidant content and the radical scavenging activity were significantly correlated (0.563**). Vitamin A content was significantly lowest (6.88 ± 0.38 μg/g retinol) in Dagga flour and highest (7.97 ± 0.25 μg/g) in Impilo flour. Protein content varied significantly across the two locations and ranged from 4.23% (Tshiombo) to 7.16% (Univen). Dagga had significantly the lowest (3.47%) protein content while Impilo had the highest (7.53%). The fat content of the flours varied significantly between the two locations ranging from 0.63% (Tshiombo) to 1.01% (Univen). The protein content was positively correlated 0.594** and 0.421* to vitamin A and fat contents respectively. The Bophelo colour luminosity was significantly lower (L* 76.2) than the other two OFSP, Dagga (L* 83.51) and Impilo (L* 82.07). Bophelo a* (14.09) also differed significantly across all sweet potato cultivars. All cultivars differed significantly in b* values. Colour intensity (ΔE*) ranged from 15.87 to 38.48 for Mvuvhelo and Bophelo, respectively. Chroma ranged from 87.52 to 84.95 for Dagga and Mvuvhelo respectively. Total starch content was significantly different in all sweet potato cultivars and ranged from 244.07 μg/100 g (Bophelo) to 325.04 mg/g (Dagga). Resistant starch differed significantly in all sweet potato cultivars ranging from 1.21 μg/100 g dwb (Bophelo) to 7.07 μg/100 g dwb (Impilo). Ash content was significantly different in all cultivars and ranged from 3.17% (Dagga) to 3.79% (Bophelo). Water holding capacity (WHC) varied significantly across the two locations and ranged from 1.44 g H2O/g DM (Tshiombo) to 1.72 g H2O/g DM (Univen). Bophelo had significantly the lowest (1.27 g H2O/g DM) WHC and Impilo the highest (1.99 g H2O/g DM). The ash content negatively correlated to total starch (-0.928**) but positively iii correlated to WHC (0.654**). Peak viscosity differed significantly across all cultivars under study and ranged from 126.54 RVU (Bophelo) to 302.36 RVU. (Dagga). There was significant difference in trough viscosity across all cultivars ranging from 55.97 RVU (Impilo) to 280.78 RVU (Dagga). There were significant differences in the final viscosity across all cultivars ranging from 78.99 RVU (Impilo) to 319.26 (Dagga). Peak time viscosity varied significantly across all cultivars ranging from 3.87 min (Impilo) to 8.47 min. (Bophelo). Dagga cultivar had significantly the highest pasting temperature (94.16oC) and Bophelo the lowest (77.75oC). The setback viscosity was significantly highest 51.10 RVU (Mvuvhelo) and lowest 23.01 RVU (Impilo). All cultivars from the two locations consistently showed high potassium content ranging from 0.46% (Impilo) to 1.38% (Dagga) as compared to other minerals such as magnesium, calcium and chlorine which ranged from 0.16% (Impilo) to 0.34%. The CT scans revealed that there was a possibility of evaluating the effect of location on the density of granules of sweet potato flour and also the inherent differences in granular distribution of various sweet potato cultivars. SEM revealed high amounts of potassium, magnesium and chlorine. A profile of physicochemical and functional properties of flour of some of the OFSP cultivars produced in Limpopo province, South Africa has been drawn. This study also highlights the nutritional quality of the flour from four sweet potato cultivars and provides a ranking of flour to help the producers in the selection process. Bophelo flour was ranked the highest in terms of β- and, α-carotenes, total carotenoids and antioxidant capacity while Impilo was ranked highest in terms of functional properties and ash content. Ash content was significantly different in all cultivars and ranged from 3.17% (Dagga) to 3.79% (Bophelo). Water holding capacity (WHC) varied significantly across the two locations and ranged from 1.44 g H2O/g DM (Tshiombo) to 1.72 g H2O/g DM (Univen). Bophelo had significantly the lowest (1.27 g H2O/g DM) WHC and Impilo the highest (1.99 g H2O/g DM). The ash content negatively correlated to total starch (-0.928**) but positively iii correlated to WHC (0.654**). Peak viscosity differed significantly across all cultivars under study and ranged from 126.54 RVU (Bophelo) to 302.36 RVU. (Dagga). There was significant difference in trough viscosity across all cultivars ranging from 55.97 RVU (Impilo) to 280.78 RVU (Dagga). There were significant differences in the final viscosity across all cultivars ranging from 78.99 RVU (Impilo) to 319.26 (Dagga). Peak time viscosity varied significantly across all cultivars ranging from 3.87 min (Impilo) to 8.47 min. (Bophelo). Dagga cultivar had significantly the highest pasting temperature (94.16oC) and Bophelo the lowest (77.75oC). The setback viscosity was significantly highest 51.10 RVU (Mvuvhelo) and lowest 23.01 RVU (Impilo). All cultivars from the two locations consistently showed high potassium content ranging from 0.46% (Impilo) to 1.38% (Dagga) as compared to other minerals such as magnesium, calcium and chlorine which ranged from 0.16% (Impilo) to 0.34%. (Dagga). The CT scans revealed that there was a possibility of evaluating the effect of location on the density of granules of sweet potato flour and also the inherent differences in granular distribution of various sweet potato cultivars. SEM revealed high amounts of potassium, magnesium and chlorine. A profile of physicochemical and functional properties of flour of some of the OFSP cultivars produced in Limpopo province, South Africa has been drawn. This study also highlights the nutritional quality of the flour from four sweet potato cultivars and provides a ranking of flour to help the producers in the selection process. Bophelo flour was ranked the highest in terms of β- and, α-carotenes, total carotenoids and antioxidant capacity while Impilo was ranked highest in terms of functional properties and ash content. This study has highlighted significant differences in a number of parameters measured among the sweet potato cultivars. Therefore, in order to derive the maximum benefit from a given cultivar careful selection of sweet potato cultivars would be necessary.

Influence of head-moisture treatment on functional, colour and thermal properties of bambara ground-nut starch

MSCFST / Department of Science and Technology / Heat-moisture treatment (HMT) is a physical modification that alters the physicochemical properties of starch without changing its molecular structure. The objective of the study was to investigate the influence of HMT on the functional, colour and thermal properties of bambara groundnut (BG) starch. A central composite rotatable design comprising two independent factors (temperature and time) was used for the study. The central composite rotatable design was generated using Design-Expert software version 8.0.1.0. Bambara starch extraction was done by milling BG into flour (5 Kg), suspension in 15 L, 0.3% sodium hydroxide and centrifugation followed by washing using distilled water. The starch was then HMT treated in an air oven at 80 - 120 °C for 30 - 90 min under 15 % moisture content (MC) (HMT 15), 25% MC (HMT 25) and 35% MC (HMT 35). The highest L* and WI values for HMT treated BG starch were observed at HMT 80 °C for 30 min under 15% MC; 100 °C for 60 min (25% MC); and 100 °C for 17.57 min (35% MC) while the lowest was observed in HMT 100 °C for 102.43 min (15% MC); 120 °C for 90 min (25% MC); and 120 °C for 90 min (35% MC). In HMT 15-BG starch, the gelatinisation parameters onset (To), peak (Tp) and concluding temperature (Tc) of the samples decreased as treatment time and temperature increased whereas gelatinisation enthalpy of BG starch increased with increase in HMT treatment temperature and time. In HMT 25-BG starch Tp, and gelatinisation enthalpy of the starch increased with increase in HMT treatment temperature and time. While in HMT 35-BG starch, To, Tp, Tc and gelatinisation enthalpy of the starches decreased with increase in HMT treatment temperature and time. In HMT 15-BG starch, the water absorption capacity (WAC), solubility and swelling power (SP) decreased as treatment time and temperature increased while oil absorption capacity (OAC) of the starch increased with increase in HMT treatment temperature and time. In terms of HMT 25-BG starch, the WAC and OAC increased as HMT treatment time and temperature increased while SP and solubility of the starch decreased with increase in HMT treatment temperature and time. In HMT 35-BG starch, OAC, solubility and SP decreased as treatment time and temperature increased while WAC of the starch increased with increase in HMT treatment temperature and time. The optimum HMT conditions for BG starch were found to be 80 °C for 30 min (HMT 15), 105.74 °C for 30 min (HMT 25), and 113.16 °C for 30 min (HMT 35). Desirability of the obtained optimum conditions were 0.63 (HMT 15), 0.56 (HMT 25) and 0.64 (HMT 35). Information obtained from scanning electron micrograph indicates oval and round shape for bambara starch granules, with varying sizes. The range of the granule size width was 4.2 – 4.7 mm and 10 μm for length. The modified starches showed some changes in granule morphology as they seem to disintegrate with application of HMT. Unmodified and HMT - BG starches showed characteristic FTIR bands linked with common starches. All the samples displayed complex vibrations in the region below 1000 cm-1 due to the skeletal vibrations of the glucose pyranose ring. Statistical analysis on colour, thermal and functional properties of HMT 15-BG, HMT 25-BG and HMT 35-BG starch showed that effects of temperature and treatment time had no significant (p ≥ 0.05) effect on these properties of HMT-BG starch. However, treatment time had a significant linear effect (p ≤ 0.05) on swelling power, for HMT 15-BG starch. In HMT 35-BG starch, WAC was significantly affected by quadratic effect of temperature and time while solubility was significantly affected by linear effect of time and quadratic effect of temperature. / NRF

QoS Contract Negotiation in Distributed Component-Based Software

Currently, several mature and commercial component models (for e.g. EJB, .NET, COM+) exist on the market. These technologies were designed largely for applications with business-oriented non-functional requirements such as data persistence, confidentiality, and transactional support. They provide only limited support for the development of components and applications with non-functional properties (NFPs) like QoS (e.g. throughput, response time). The integration of QoS into component infrastructure requires among other things the support of components’ QoS contract specification, negotiation, adaptation, etc. This thesis focuses on contract negotiation. For applications in which the consideration of non-functional properties (NFPs) is essential (e.g. Video-on-Demand, eCommerce), a component-based solution demands the appropriate composition of the QoS contracts specified at the different ports of the collaborating components. The ports must be properly connected so that the QoS level required by one is matched by the QoS level provided by the other. Generally, QoS contracts of components depend on run-time resources (e.g. network bandwidth, CPU time) or quality attributes to be established dynamically and are usually specified in multiple QoS-Profiles. QoS contract negotiation enables the selection of appropriate concrete QoS contracts between collaborating components. In our approach, the component containers perform the contract negotiation at run-time. This thesis addresses the QoS contract negotiation problem by first modelling it as a constraint satisfaction optimization problem (CSOP). As a basis for this modelling, the provided and required QoS as well as resource demand are specified at the component level. The notion of utility is applied to select a good solution according to some negotiation goal (e.g. user’s satisfaction). We argue that performing QoS contract negotiation in multiple phases simplifies the negotiation process and makes it more efficient. Based on such classification, the thesis presents heuristic algorithms that comprise coarse-grained and fine-grained negotiations for collaborating components deployed in distributed nodes in the following scenarios: (i) single-client - single-server, (ii) multiple-clients, and (iii) multi-tier scenarios. To motivate the problem as well as to validate the proposed approach, we have examined three componentized distributed applications. These are: (i) video streaming, (ii) stock quote, and (iii) billing (to evaluate certain security properties). An experiment has been conducted to specify the QoS contracts of the collaborating components in one of the applications we studied. In a run-time system that implements our algorithm, we simulated different behaviors concerning: (i) user’s QoS requirements and preferences, (ii) resource availability conditions concerning the client, server, and network bandwidth, and (iii) the specified QoS-Profiles of the collaborating components. Under various conditions, the outcome of the negotiation confirms the claim we made with regard to obtaining a good solution.
