ANGLICKÝ ABSTRAKT Functional study of the putative nucleotidase encoded by spr1057 gene in Streptococcus pneumoniae, a likely homolog of Escherichia coli protein YjjG. Bacterial cells are constantly exposed to innumerable toxic substances, either in their external environment or by by-products of their own metabolism. For these reasons, the bacterial cells evolved several mechanisms to cope with this challenge. These mechanisms are represented by: blocking the uptake, export by specific transporters as well as specific inactivation of these substance by enzymes. A particular group of these toxic substances are noncanonica nucleotides, which can directly inhibit bacterial cell DNA replication or can result in increased mutation rate. Enzymes recognizing these modified derivatives are known as "house-cleaning" nucleotide phsphateses, which can inactivate the potentially mutagenic nucleotides and prevent their incorporation into DNA and RNA. Some of the "house- cleaning" enzymes belong to a group of haloacid dehalogenase enzymes (haloacid dehalogenase-like hydrolase superfamily), which are found in many bacterial species. Psykometrisk prövning av PG-13; ett självskattningsinstrument för ihållande sorg

Frågan om ”normal” respektive ”patologisk” sorg har under en lång tid varit ett omdiskuterat ämne inom psykologin och psykiatrin. En grupp forskare har tagit fram kriterier på ett syndrom som fått benämningen ihållande sorg (”prolonged grief”). Studier har visat på att ihållande sorg är ett distinkt syndrom separat från depression, posttraumatisk stressyndrom och generaliserat ångestsyndrom. Ihållande sorg föreslås ingå som en diagnos i de kommande versionerna av de diagnostiska manualerna DSM och ICD. Syftet med denna studie var att översätta självskattningsinstrumentet Prolonged Grief Disorder (PG-13) till svenska, och undersöka dess reliabilitet och validitet genom bland annat en explorativ och konfirmatorisk faktoranalys. Studien genomfördes på ett annonsrekryterat heterogent sample (n=162 varav 90 % var kvinnor) vad gäller typ av dödsfall och relation till den avlidne. Respondenterna besvarade en webb- eller pappersenkät som bestod av 75 items. PG-13 bedöms ha en hög reliabilitet, god begrepps- och samtidig validitet samt en måttlig diskriminativ validitet.

The thesis deals with the modeling and prediction of corrosion of radiation tube snake in the heating furnace. Specifically it is focused on vertical cylindrical furnace which is included in the catalytic hydrocracking unit and serves for heating aggressive circulation gas which is the cause of high temperature corrosion. An important basis for the creation of computational models are available records about the operation of the furnace and about the corrosion and degradation mechanisms during the lifetime of the tube system in furnace. Such information enables the creation of a computational model which is based on the prediction of high-temperature corrosive damage of radiation tube snake. The computational model involving all relevant factors may serve as the basis for a predictive life management system of radiation snakes in the heating furnace.

Had jako prostředek reflexe v textech o umělcích z období německého romantismu / The Snake as Means of Reflection in the Texts about Artists from the German Romantic Period

The submitted doctoral thesis examines the function and performance of the motive of a snake in four selected texts about artists of the German romantic period and perceives the snake as an attribute reflecting the problems of the artistic existence and a way to the artistic work, possibly of the artistic work at all. When interpreting the single motives of the snake, three-phase procedure as applied by Erwin Panofsky in description of the motives in the pictures has been chosen by analogy for literary texts. The interpretation is based on the structural position of the motive of the snake in the text and its performance in the text. Subsequently, the principal role is revelation of contexts which updates the given motive of the snake - in particular they are biblical snakes, mythical snakes and a serpentine line as a line of beauty which was the subject of the period aesthetic discourse. The meaning of the motive of the snake in the texts is given primarily by the mutual interaction of theme contexts which is in the single texts realized by various ways. The motive of the snake as formed in the single interpreted texts is a highly complex motive in all the cases. The aim is to analyze how Mekas?s work contributes to an understanding of the diary as a narrative form, and how the author constructs his process of self-representation by means of the diaries. This work seeks to establish a definition for the diary, by means of theoretical references related to the spheres of literature (Braud, 2006; Simonet-Tenant, 2004; Girard, 1986; Lejeune, 2015; Blanchot, 2005) and of audiovisual work (James, 2013; Renov, 1996). As object of study, we analyze the book I had nowhere to go (1991), the diary-films Lost Lost Lost (1976), Walden ? Diaries, Sketches & Notes (1969), Reminiscences of a Journey to Lithuania (1972), and video diaries from the 365 Day Project (2007). This thesis takes weavings (Daney, 2007; Benjamin, 1994) and rends (Daney, 2007; Didi-Huberman, 2014, 2015) as its methodology, grounding itself on the way in which Mekas elaborates his narrative choices and his historical condition. We conclude that Mekas?s work points to an understanding of the diary not solely as literary genre, but as a narrative that affirms itself in a plurality of forms, following the shifts done by the author in his relation to time, space, and the use of different technical devices. We also conclude that Mekas, in his diaries, builds an intrinsic relation between narrating the lived and living the narrated, which reveals how the diaries not only document a certain socio-historical context, but are also themselves part of that context and of the subjectivity built by their author. / Esta tese realiza um estudo sobre o di?rio, que aparece na obra do cineasta Jonas Mekas em tr?s formatos: o di?rio escrito, o filme-di?rio e o v?deo-di?rio. O objetivo ? analisar como essa obra contribui para um entendimento sobre o di?rio enquanto forma narrativa e como o autor constr?i o seu processo de autorrepresenta??o por meio dos di?rios. Analýza výskytu vybraných zoonóz v Jihočeském kraji / Analysis of occurrences of selected zoonoses in South Bohemian region

This diploma thesis is focused on the evaluation of six selected zoonoses with the occurrence in South Bohemia (campylobacteriosis, salmonellosis, listeriosis, tick-borne meningoencephalitis, Lyme borreliosis, tularemia) between the years 2003-2013. Campylobacteriosis and salmonellosis were evaluated the most common zoonoses in the South Bohemian Region during the monitored period. Their common feature is the alimentary transmission. Between 2003 to 2008 salmonellosis recorded the highest number of reported cases, on the contrary, campylobacteriosis has reported the highest number of occurrences since 2008. District of Czech Budweis showed the highest incidence of both zoonoses during the monitored period (4,139 cases of salmonellosis and 4,924 cases of campylobacteriosis). Another but not less important zoonotic is tick-borne meningoencephalitis, which incidence had the highest number in South Bohemian Region of all the regions of Czech Republic. The second part of the thesis is based on questionnaire studies focused on awareness of zoonoses and the issue of resistance and overuse of antibiotics. In total 479 questionnaires were evaluated. The survey findings may be equally described as interesting and positive, since the respondents had considerable awareness of the issue of antibiotic resistance.

The application of a technique for enhancing recall to improve learning in the science classroom

There has existed for many years a memory enhancement technique ("memory pegs") that although having dramatic demonstrable success in some individual cases has not been generally applied in education. The emergence of constructivist epistemology has emphasised the notion that learning occurs as a result of connecting new material with previously learnt concepts. There is, therefore, the implication that effective learning requires some previous knowledge upon which to attach new concepts - and thus realisation of the importance of learning with respect to acquiring factual information as a prerequisite to learning new processes and/or skills. This issue has focussed my attention on the need to ensure that the more physiological skills of accessing 'memory', both for learning and recall, are optimised for maximum learning. Further, there are some indications that the physiological skills of memory access (storage and retrieval) may respond favourably to training and 'exercise'. This study was designed to find out whether or not a repeated 'exercise' using a simple memory enhancement technique would lead to a determinable and statistically significant increase in overall performance in a range of cognitive skills (as indicated by science and mathematics examination results), whether learning such a technique would affect a student's attitudes towards science, whether there was a relationship between the amount of time spent practicing the technique and the degree of effect, and whether the memory technique did actually improve the ability to recall lists of objects.
