Flavor Chemistry of Regional Hops (Humulus lupulus L.) and Novel Aroma Application of Hop-derived Products

Hop (Humulus lupulus L.) is an indispensable raw material in beer brewing because it can provide unique aroma and bitterness to beer products. With growing consumer interests in locally-sourced ingredients and increasing number of microbreweries, hop production is emerging in many non-traditional U.S. growing regions like Virginia (VA). However, the lack of understanding on aroma chemistry of regional hops limited their prosperity. Moreover, suitable postharvest drying and packaging practices for VA hop producers are not available. This dissertation aims to address above issues by investigating the aroma chemistry of VA hops by varieties (Cascade, Chinook), growing locations (Meadowview/Petersburg/Blacksburg, VA), smaller-scale drying practices (oven drying, dehydrator drying, freeze drying) and packaging materials (PA/PE, OPP/Foil/PE, EVOH). Our efforts also extended to the novel application of hop-derived ingredients in non-beer drinks to promote value-added products. Solid phase microextraction and solvent-assisted flavor evaporation were applied for aroma extraction. Gas chromatography-mass spectrometry-olfactometry was used with stable isotope dilution analysis and standard addition method for accurate quantitation of aroma-active compounds. A total of 33 and 36 aroma-active compounds were identified in all fresh and dried hops, respectively. Geraniol, β-myrcene, linalool, methyl octanoate and trans-α-bergamotene were found to be the predominant compounds. Aroma profiles varied significantly with growing locations for both varieties. Individual aroma contents and total essential oil were the highest in dehydrator-dried hops, indicating the advantage of dehydrator-drying being a suitable practice to retain aroma power in hops for most smaller-scale producers in VA. Most volatile compounds in different packaged dried hops showed decreased concentrations over 8-month storage, but the variation was not statistically significant. Hop essential oil (HEO) microcapsules, manufactured by spray drying using modified starch CAPSUL® as the wall material, showed high flavor retention and controlled aroma release. The addition of HEO microcapsules significantly improved the aroma recovery and stability in hop tea. Our findings enhance the understanding of the aroma chemistry in regional hops as affected by multiple pre- and postharvest factors. The novel non-beer application of HEO was also successfully demonstrated. / Doctor of Philosophy / Hop (Humulus lupulus L.) is an essential raw material in beer brewing because it can provide beer products with unique flavor. With the growing "buy local" trend among consumers in recent years, hop production is moving to many non-traditional growing regions in the U.S. including Virginia. This resulted in an increasing number of local craft breweries operating on a smaller scale across the state. However, the aroma characteristics of Virginia-grown hops are unclear. Specific suggestions related to hop postharvest processing and storage are also unavailable for regional smaller-scale hop producers. Therefore, this study aims to study the aroma characteristics of Virginia-grown hops by varieties, growing locations, smaller-scale drying practices and packaging strategies. The novel non-alcoholic application of hop essential oil (HEO, mixture of many hop aroma compounds) was also demonstrated in hop tea. The results showed that both fresh and dried Virginia hops had a complicated profile of aroma compounds that primarily exhibited typical citrus, woody and fruity attributes. Both Cascade and Chinook varieties harvested from Meadowview showed significantly higher aroma contents than hops harvested from the other two locations in Virginia. Most aroma compounds in hops were better preserved by dehydrator drying than freeze an oven drying. No significant change was observed in aroma compositions from hops packaged in three different materials after storage. Finally, HEO particles with enhanced stability were prepared by engineering approaches, which were finally added to hop tea bags for the evaluation of aroma-intensifying effects and storage stability. The results indicated an overall desirable characteristic for the obtained HEO particles. The addition of HEO particles could significantly enhance the aroma stability of hop tea products during storage. Overall, the findings from this study enable a better understanding of flavor chemistry of regional hops from various origins and demonstrated a successful application of HEO particles as a flavoring agent in non-beer products.

Freedom, Music and the RIAA: How the Recording Industry Association of America Shapes Culture by De-politicizing Music

Since the development of widespread sound recording and distribution, the music industry has become increasingly consolidated among fewer companies. Max Horkheimer and Theodor Adorno described how the commodifying forces of the music industry lead to a predictable formulaic music that lacks any critical approach to society in their groundbreaking book, first published in 1944, Dialectic of Enlightenment (1972). Today, the patterns have not changed as there are now four major record labels known as the "Big Four" that produce commodified music with a business model that optimizes their profits at the expense of art, creativity and original style. Using the Recording Industry Association of America (RIAA) as its lobbying group and appointed vigilantly, the "Big Four" attempt to limit the access of independent artists and labels to music consumers. This thesis argues that in the process through which the music industry works to co-opt and commodify genres of music, the music is (de)politicized to appeal to a larger audience. While technological advances in digital media and the internet would seem to bring a decentralized (even democratized) structure that diverts the costly music distribution system allowing for more artists and labels to compete, the RIAA has acted to prevent these technologies from developing their greatest potential. First, I demonstrate how music is commodified and marketed towards consumers. The second part of this thesis uses hip hop as an example to demonstrate how the music industry co-opts a genre of music to sell to the largest number of consumers and in the process changes the political significance of that genre. Finally, I argue that the RIAA's attack on file-sharers in the name of copyright protection is a technique for the "Big Four" to stop competition from independent artists and labels. / Master of Arts

On Programmable Control and Optimization for Multi-Hop Wireless Networks

Traditionally, achieving good performance for a multi-hop wireless network is known to be difficult. The main approach to control the operation of such a network relies on a distributed paradigm, assuming that a centralized approach is not feasible. Relying on a distributed paradigm could be justified at the time when the basic technical building blocks (e.g., node computational power, communication technology, positioning technology) were the bottlenecks. Recent advances and breakthroughs in these technical areas along with the emergence of programmable networks with softwarized control plane intelligence allow us to consider employing a centralized optimization paradigm to control and manage the operation of a multi-hop wireless network. The programmable control provides a platform on which the centralized global network optimization paradigm can be supported. The benefits of a centralized network optimization lie specially in that a network may be configured in such a way that offers optimal performance, which is hardly possible for a network relying on distributed operation. The objectives of this dissertation are to fully understand the potential benefits of a centralized control plane for a multi-hop wireless network, to identify any new challenges under this new paradigm, and to devise innovative solutions for optimal performance via a centralized control plane. Given that the performance of a wireless network heavily depends on its physical layer capabilities, we will consider a number of advanced wireless technologies, including MIMO, full duplex, and interference cancellation at the physical layer. The focus is on building tractable computational models for these wireless technologies that can be used for modeling, analysis and optimization in the centralized control plane. Problem formulation and efficient solution procedures are developed for various centralized optimization problems across multiple layers. End-to-end throughput maximization is a key objective among these optimization problems on the centralized control plane and is used to demonstrate the superior advantage of this paradigm. We study several problems: • Integration of SIC and MIMO DoF IC. We propose to integrate MIMO Degree-of-Freedom (DoF) interface cancellation (IC) and Successive Interference Cancellation (SIC) in MIMO multi-hop network under DoF protocol model. We show that DoF-based IC and SIC can be jointly integrated to combat the interference more effectively and improve the end-to-end throughput significantly. We develop the necessary mathematical models to realize the idea in a multi-hop wireless network. • Full-Duplex MIMO Wireless Networks Throughput. We investigate the performance of MIMO full-duplex (FD) in a multi-hop network. We show that if IC is exploited, MIMO FD can achieve significant throughput gain over MIMO HD in a multi-hop network, which is contrary to the recent literature suggesting an unexpected marginal gain. Our proposed model handles the additional network interference by joint efficient link scheduling and interference cancellation. • PCP in Tactical Wireless Networking. We propose the idea of the Programmable Control Plane (PCP) for the tactical wireless network under the protocol model. PCP decouples the control and data plane and allows the network control layer functionalities to be dynamically configured to adapt to specific wireless channel conditions, customized applications and/or certain tactical situations. The proposed PCP functionalities are cast into a centralized optimization problem, which can be updated as needed and provide a centralized intelligence to manage the operation of a wireless MIMO multi-hop network under the protocol model. • UPCP in Heterogeneous Wireless Networks. We propose the idea of the Unified Programmable Control Plane (UPCP) for tactical heterogeneous wireless networks with interference management capabilities under the SINR model. The UPCP abstracts the complexity of the underlying network comprised of heterogeneous wireless technologies and provides a centralized intelligence over the network resources. Utility Accrual Real-time Channel Establishment in Multi-hop Networks

Real-time channels are established between a source and a destination to guarantee in-time delivery of real-time messages in multi-hop networks. In this thesis, we propose two schemes to establish real-time channels for soft real-time applications whose timeliness properties are characterized using Jensen's Time Utility Functions (TUFs) that are non-increasing. The two algorithms are (1) Localized Decision for Utility accrual Channel Establishment (LocDUCE) and (2) Global Decision for Utility accrual Channel Establishment (GloDUCE). Since finding a feasible path optimizing multiple constraints is an NP-Complete problem, these schemes heuristically attempt to maximize the system-wide accrued utility. The channel establishment algorithms assume the existence of a utility-aware packet scheduling algorithm at the interfaces. The route selection is based on delay estimation performed at the source, destination, and all routers in the path, from source to destination. We simulate the algorithms, measure and compare their performance with open shortest path first (OSPF). Our simulation experiments show that for most of the cases considered LocDUCE and GloDUCE perform better than OSPF. We also implement the schemes in a proof-of-concept style routing module and measure the performance of the schemes and compare them to OSPF. Our experiments on the implementation follow the same trend as the simulation study and show that LocDUCE and GloDUCE have a distinct advantage over OSPF and accrue higher system-wide utility. These schemes also react better to variation in the loading of the links. Among the two proposed approaches, we observe that GloDUCE performs better than LocDUCE under conditions of increased downstream link loads. / Master of Science

I denna undersökning har jag utforskat hur jag kan använda mig av mikrotiming som verktyg på elbas för att påverka ett groove. Jag utgick från två brasilianska rytmer som hade olika förskjutningar inom parametrarna för mikrotimingen, som sedan översattes till basgångar. Jag använde mig av två beats, ett till vardera vald rytm, som jag applicerade basgångarna på för att se hur och om jag kunde påverka den uppfattade känslan i groovet genom att spela basgångarna både med och utan applicerad mikrotiming.

Between-session reliability of four hop tests and the agility T-test

No / The purposes of this study were firstly to investigate whether learning affects were present in the administration of 4 hop tests and the Agility T-test and secondly to assess the between-session reliability of these tests. Twenty-two recreational athletes (11 women: age 22.3 ± 3.7 years, height 167.7 ± 6.2 cm, weight 59.2 ± 6.9 kg and 11 men: age 22.8 ± 3.1 years, height 179.8 ± 4 cm, weight 79.6 ± 10 kg) took part in the study. The subjects performed 6 repetitions of each hop test and 4 repetitions of the Agility T-test once a week over a period of 3 weeks. Distances were normalized to leg length and presented as a percentage value for the single, triple and crossover hop. Results showed that there were significant differences in scores between genders and that learning affects were present in all tests. Intraclass correlation coefficients ranged from 0.76 to 0.92 for the hop tests and 0.82 to 0.96 for the Agility T-test. The results indicated that the hop and Agility T-tests are reliable tests for use with subjects in a clinical or team sport environment. The error measurement statistics presented could be of help to practitioners to determine whether changes in individuals' scores in the hop and Agility T-tests are because of a true change in performance or measurement error. Of most importance was the fact that all subjects achieved at least 90% limb symmetry index on all 4 hop tests. Therefore, we recommend that a minimum limb symmetry value of 90%, rather than previously recommended 85%, should be adopted during rehabilitation and conditioning.

[pt] Este trabalho procura refletir sobre o crescimento e a consolidação do grafite no Rio de Janeiro. Uma das expressões da cultura hip-hop, o grafite surgiu nos Estados Unidos, na década de 1970. No início eram apenas assinaturas, que depois evoluíram para desenhos elaborados. São Paulo foi a primeira cidade brasileira onde este tipo de escrita urbana se desenvolveu nos anos 1980 e 1990. Mas no Rio, a atividade de pintar muros chegou mais tarde e somente se firmou nos anos 2000. O grafite começou a ser feito por grupos da Zona Norte, do subúrbio e do município de São Gonçalo. Porém, quando jovens da Zona Sul – alguns estudantes de design – levaram o grafite para os muros da área nobre da cidade houve uma mudança de conceito: o que era considerado marginal passou a ser visto como uma arte urbana legítima e moderna. O Rio repetiu um pouco a experiência ocorrida em Nova York, nos anos 1970, quando jovens artistas do circuito underground passaram a pintar prédios do SoHo e paredes do metrô. Com isso, o grafite adquiriu status e chegou às galerias de arte. Hoje, este tipo de intervenção urbana não só é aceita pela população carioca que antes a rejeitava como foi totalmente incorporada pelo mercado. Alguns discursos sobre a cultura que tomou conta das ruas do Rio – como os da imprensa, da publicidade e de grafiteiros – foram a base para a análise e reflexão deste trabalho. / [en] This work is a reflection on the rise and consolidation of graffiti in Rio de Janeiro. One of the expressions of hip hop culture, graffiti was born in the United States in the 1970s. At first, it comprised only signatures, which later evolved into elaborate drawings. São Paulo was the first Brazilian town to see the development of this sort of urban writing, around the 1980s and 1990s. The activity of painting walls came later to Rio and only became established in the 2000s. Graffiti in Rio began at the hands of groups from the North Zone, the suburbs and the town of São Gonçalo. Then, however, groups from the South Zone youth – some of them design students – took graffiti to the walls of the city’s noble areas, allowing a conceptual change: what was first considered unlawful became legitimate modern urban art. In truth, Rio was witness to a replication of the 1970s New York experience, when underground young artists such as Keith Haring and Jean- Michel Basquiat started painting Soho buildings and subway walls. Thus graffiti gained status and moved into art galleries. Nowadays, this kind of urban intervention has been accepted by the population of Rio, who used to reject it in the past. Not only that, it has also been completely incorporated by the market. Some of the discourses about this culture that has taken over the streets of Rio – such as from the press, advertising and graffiti artists themselves – have been the base for the reflection and analysis presented in this work.

Papel do complexo PrPc-HOP e vesículas extracelulares em câncer colorretal / Role of PrPC-HOP complex and extracellular vesicles in colorectal cancer

O câncer colorretal (CCR) é o terceiro tipo de câncer mais comum no mundo. Apesar dos avanços nos tratamentos convencionais, aproximadamente dois terços dos pacientes com CCR são submetidos à cirurgia potencialmente curativa. Entretanto, grande parte desses pacientes evolui mal, apresentando recidivas e/ou metástases. A busca de novos alvos moleculares para a terapia do CCR revelou a proteína celular Prion (PrPC) como um possível candidato. Trabalhos recentes sugerem participação direta ou indireta de PrPC no crescimento de tumores, na formação de metástases, na composição de complexos multiproteicos e na indução de vias de sinalização envolvidas em diversos processos biológicos, como proliferação. Além disso, PrPC foi descrito como um importante modulador do crescimento de tumor colorretal. Resultados prévios mostraram que a interação da proteína PrPC com a proteína HSP70/HSP90 Organizing Protein (HOP) induz proliferação em glioblastomas. HOP é uma proteína predominantemente citoplasmática, podendo também ser secretada associada às vesículas extracelulares. Assim, o presente estudo objetivou avaliar o papel do complexo PrPC-HOP e das vesículas extracelulares no desenvolvimento e progressão dos tumores colorretais. Os nossos resultados mostram que HOP induziu migração e invasão em linhagens de CCR de maneira dependente de PrPC, uma vez que o uso do peptídeo sem atividade que compete pelo sítio ligação de HOP a PrPC inibiu estes processos. Além disso, nossos dados apontaram que o aumento de migração e invasão das células de CCR induzida pela interação PrPC-HOP é mediada pela ativação da via ERK1/2. Os achados in vitro estimularam a avaliação do perfil de expressão de PrPC e HOP por imuno-histoquímica em tecidos de pacientes com diferentes tipos de tumores colorretais. Nossos resultados sugeriram que essas proteínas são importantes no início do desenvolvimento tumoral e na transição de adenomas para adenocarcinomas, não havendo correlação entre a presença de HOP e/ou PrPC com metástase, linfonodos acometidos, estadiamento, sobrevida ou região tumoral versus tecido normal. Em relação ao papel das vesículas extracelulares na progressão dos tumores colorretais, nossos resultados mostraram que linhagens celulares que apresentam padrões parecidos de agressividade tumoral podem ter perfis de secreção de proteínas e vesículas extracelulares bastante diferentes, induzindo, portanto, processos biológicos com intensidades distintas. O meio condicionado e as vesículas extracelulares da linhagem WiDr apresentaram maior potencial de indução de migração quando comparado com a linhagem HCT8. Além disso, a modulação negativa da proteína VPS4, uma das responsáveis pela formação dos corpos multivesiculares, mostrou-se uma abordagem interessante no estudo da secreção de vesículas por células de CCR, uma vez que o dominante negativo de VPS4 promoveu diminuição do cargo proteico e da secreção de vesículas extracelulares, redução da proliferação celular e do efeito indutor do processo de migração na linhagem WiDr. Assim, em conjunto, o presente trabalho indicou que o complexo PrPC-HOP pode ser um bom alvo terapêutico nos processos de migração e invasão em CCR. Ainda, essas proteínas se mostraram importantes nos estágios iniciais da formação dos tumores. A modulação da secreção de vesículas extracelulares pode contribuir para retardar a progressão dos tumores colorretais. / Colorectal cancer (CRC) is the third most common type of cancer in the world. Despite improvements in conventional treatments, approximately two-thirds of CRC patients undergo potentially curative surgery. However, most of these patients evolve poorly, showing recurrence and/or metastasis. Search of new molecular targets for CRC therapy revealed the cellular protein Prion (PrPC) as a putative candidate. Recent studies have shown that PrPC exhibit direct or indirect participation in tumor growth, formation of metastasis, composition of multiprotein complexes and induction of signaling pathways involved in many biological processes such as proliferation. Moreover, PrPC has been described as an important modulator of colorectal tumor growth. Previous findings showed that the interaction between PrPC and its ligand HSP70/90 heat shock organizing protein (HOP) induces gliobastoma proliferation. It is well known that HOP localizes mainly in the cytoplasm but HOP is also secreted associated with extracellular vesicles. In this way, the present study sought to evaluate the role of PrPC-HOP complex and extracellular vesicles in the development and progression of CRC. We demonstrate that HOP induces the migration and invasion of CRC cell lines in a PrPC-dependent manner because the use of HOP peptide, which is able to bind to PrPC, blocking PrPC-HOP complex formation, inhibited the migration and invasion processes. In addition, our data showed that the enhancement of migration and invasion induced by PrPC-HOP interaction is mediated by ERK1/2 pathway activation. These in vitro results lead us to evaluate the PrPC and HOP expression by immunohistochemistry in tissues from patients with different tumor types. Our data showed that these proteins could be important for the initial steps of tumor development, represented by the transition from adenoma to adenocarcinoma. No correlation was found among HOP and/or PrPC expression and metastasis, lymph node involvement, staging, survival or tumor area versus normal tissue. Regarding the role of extracellular vesicles in the progression of colorectal tumors, our results showed that cell lines exhibiting similar aggressive tumor behavior can have a different protein secretion pattern and a distinct profile of extracellular vesicles release, which could induce biological process with different intensities. The conditioned medium and the extracellular vesicles derived from WiDr cell line showed a higher potential to induce migration than HCT8 cell line. Moreover, the negative modulation of VPS4, one of the proteins responsible for multivesicular body formation, showed to be an interesting approach in the study of extracellular vesicles secretion secreted by CRC cells; the negative dominant of VPS4 promoted in the WiDr cell line a reduction in the protein cargo and secretion of the extracellular vesicles, a decrease of cell proliferation and induction of migration process. Therefore, taken together, our data highlights that PrPC-HOP complex can be considered a new therapeutic target in migration and invasion processes of CRC. Moreover, these proteins appeared to be important at onset of tumor formation. The modulation of extracellular vesicles secretion may contribute for delaying the progression of colorectal tumors.

Esta dissertação investiga o processo de formação da identidade, através do trabalho artístico/musical da banda de hip-hop norte-americano: Black Eyed Peas, formada na década de 80, nos Estados Unidos da América. Essa investigação diz respeito às questões de identidades discutidas pela temática do hip-hop, bem como a releitura que esta banda apresenta de elementos tidos como essenciais no gênero em questão. Ainda, visa uma maior compreensão da problemática envolvida na obra artística popular e racial e sua relação com a teoria de massa, para à partir daí, analisar seu potencial de resistência, de criatividade e de protesto. / This research ivestigates the process of meaning meaking/ signifyin(g) in the musical work of a North-american hip-hop band: Black Eyed Peas, founded in the decade of 1980, in the United States of America. This investigation is based on the identity questions discussed by the hip-hop themes, as well as the reelaboration/revisionism that this specific band shows about elements considered essences in the gender studied. Yet, this research intends a better understanding of the matters involved in the popular and racial artistic creation and its relation with the mass theories, in order to analyse its potencial of political resistance, criativity and protest.
