Expression and Splicing of Alzheimer’s Disease Risk Gene Phosphatidylinositol-Binding Clathrin Assembly Protein

Recent Genome Wide Association Studies (GWAS) have identified a series of single nucleotide polymorphism (SNP)s that are associated with Alzheimer’s disease (AD). One of the SNPs, rs3851179 (G/A), is near the gene phosphatidylinositol-binding clathrin assembly protein (PICALM). To evaluate whether this SNP is associated with PICALM expression, we quantified PICALM mRNA in 56 brain cDNA samples. Using linear regression analysis, we analyzed PICALM expression relative to rs3851179, AD status, and cell type specific markers. An association was detected between rs3851179 and PICALM, microvessel mRNA, glial fibrillary acidic protein (GFAP) mRNA, and synaptophysin (SYN) mRNA. To gain clarity into other possible SNP mechanisms, we searched brain cDNA for PICALM splice variants. We identified several PICALM splice variants involving exons 13-19. To identify and gain an estimation of relative abundance of splice variants, we PCR-amplified across exons 13-20 in cDNA from six individuals, three rs3851179 GG individuals and three rs3851179 AA individuals. Sequencing the cloned isoforms we found that PICALM lacking exon 13 (delta 13) is the most abundant isoform. Other isoforms detected included deletion of exon 18-19. We targeted the latter part of the gene, exon 17-20, to investigate unequal allelic expression using next generation sequencing. Individuals heterozygous for rs76719109 (n= 35), located in exon 17, were used to study the abundance of G/T allele in cDNA and genomic DNA. When we analyzed the T:G allelic ratio, the variant lacking exons 18 and 19 showed unequal allelic expression (p-value < 0.001) in a subset of individuals. One individual was an outlier, showing overall unequal allelic expression, which maybe be harboring a rare mutation capable of modifying PICALM expression. The PICALM intronic SNP rs588076 was associated with delta 18-19 isoform splicing (p-value < 0.001). In conclusion, this study gained a greater insight into the role of AD genetics in PICALM expression and splicing.

Das Rett-Syndrom ist eine postnatal progressiv verlaufende neurologische Entwicklungsstörung, die x-chromosomal vererbt. Das klassische Rett-Syndrom entsteht durch eine spontane Mutation des MECP2-Gens, welches für die Kodierung des Transkriptionsfaktors MeCP2 (methyl CpG binding protein 2) verantwortlich ist. Das Krankheitsbild verläuft in vier Stadien und ist vor allem von geistiger Retadierung, motorischer Dysfunktion und Unregelmäßigkeiten der Atmung geprägt. In verschiedenen Untersuchungen wurden bereits verschiedene zelluläre Dysfunktionen und Beeinträchtigungen der Mitochondrien bestätigt, weshalb wir weitere Untersuchungen anstrebten. Ziel dieser Dissertation war es, mögliche Einflüsse verschiedener Enzyme auf die ROS-Produktion zu untersuchen und somit die Erkenntnisse aus vorangegangenen Arbeiten zu erweitern. Im Fokus dieser Untersuchungen lagen zum einen die mitochondriale ROS-Produktion sowie die extramitochondriale ROS-Produktion. Durch unterschiedliche pharmakologische Modulationen wurden beide Systeme beeinflusst, um den jeweiligen Beitrag zur gesamten ROS-Produktion abzuschätzen.

The myth of prosperity: globalization and the South.

Despite many important advances since the “Bretton Woods” agreement, the state of Third World development remains extremely unsatisfactory. About a billion people live in extreme poverty and more than 800 million do not get adequate nutrition. Third World Debt looms large on the horizons of most underdeveloped nations and development continues to benefit the Third World elites, the developed nations, and their multinational corporations. The North-South divide continues to widen with very little “tricking down” to the poor majority in many nations. It is strongly believed that one-third of the world’s poor are getting poorer. The conventional approach to development theory and practice, focusing on economic growth, investment, trade and free markets continues to benefit developed nations. Even in nations where free trade and markets have spurred growth, the net results of globalization have not reached the majority. Globalization defined as the “economic, social, political and environmental integration of nations” creates some major imbalances in a world system increasingly based on liberal market economies. Ultimately, the IMF-WTO corporate globalization model has failed to deliver for developing countries, severely punishing those least capable of protecting themselves – the billions living on $ 400 million annually. Market access is only useful to countries at a stage of development that allows for their engagement in world markets, in a manner that promotes improved standards of living for their populations. The promise of market access is a distraction that has served to create a myth of prosperity. This research seeks to raise issues regarding the impact of globalization on North-South imbalances highlighting the crucial gaps in the globalization process. Initially, the research outlines the parameters and theories surrounding the globalization process and then progresses on to demonstrate the imbalances of the global system, highlighting the key areas of impact that adversely affect the development of Southern nations. Using India’s economic liberalization as a case in point, this thesis attempts to unveil the growing myth of prosperity that is being propagated in relation to the issues of globalization and the South. Maximum physical exercise is associated with increased production of reactive species and can be used as a physiological model for the study of redox imbalance. Pre-analytical conditions, such as sample freezing duration, may interfere with analyte integrity. Thus, this study evaluated the effect of sample freezing duration on the quantification of redox biomarkers in the gastrocnemius, heart and brain of Swiss mice submitted to a maximal exercise session. Twenty-six mice were divided into control group, who did not exercise, and exercise group, who performed a maximal swim test. The tissues were collected immediately after the experimental protocol and sectioned for fresh evaluation and after 1, 3 and 6 months of freezing at -80 ? C. The maximal exercise modified the redox status of fresh tissues, demonstrated bythe increase on levels of lipid peroxidation, activity of the antioxidant enzymes catalase and superoxide dismutase and reduction of the total antioxidant capacity, in all the analyzed tissues, and by the increase of protein carbonyl content in the gastrocnemius and brain. The effect of exercise on lipid peroxidation was reduced in gastrocnemius frozen for 6 months and in the brain and heart this occurred in samples after one month of freezing. On the other hand, the freezing duration did not alter the exercise effect on proteins carbonyl content and the total non-enzymatic antioxidant capacity. The catalase response to exercise in all tissues and the superoxide dismutase response in the gastrocnemius were reduced after one month of freezing. However in the brain and heart the response of superoxide dismutase to exercise was reduced only after three months of freezing. This study shows that freezing duration, depending on the tissue and marker under analysis, affects the evaluation of the redox state response to a maximal exercise session.

Estudo e desenvolvimento de sistema baseado em sensores de pressão aplicados à ortodontia

Este trabalho tem por objetivo o estudo e o desenvolvimento de uma solução de engenharia para o mapeamento e caracterização da oclusão dentária, não somente no âmbito de sua distribuição ao longo da arcada, mas também através da mensuração da intensidade de pressão nos pontos de contato oclusal interarco. Neste sentido, empregaram-se esforços no aprofundamento do conhecimento neste campo da Odontologia, assim como, na elaboração de um sistema experimental que, utilizando sensores de pressão/força resistivos (FSR) específicos para uso odontológico, viabilizou a análise do estado da oclusão, atuando como agente facilitador no diagnóstico de desequilíbrios oclusais. Inicialmente, são apresentados e esclarecidos os principais conceitos da esfera odontológica, os quais são tomados como base para todos os projetos deste trabalho, além da abordagem referente às formas de contato entre os dentes, aos diferentes tipos de oclusão e seus problemas. Em relação ao desenvolvimento do sistema, foram projetadas duas placas de circuito impresso para interface com os sensores e elaborados o firmware de acesso ao hardware, assim como o software de gerenciamento global do sistema. Todos os blocos do circuito de condicionamento foram avaliados e as suas funcionalidades validadas. Do mesmo modo, a caracterização da resposta dos sensores e a sua homogeneidade ao longo da extensão sensível foram analisadas, demonstrando que a resistência elétrica varia de forma análoga em todas as áreas do sensor. Através de ensaios realizados com sujeito envolvido no projeto, estabeleceu-se um comparativo entre a utilização do papel carbono e o sistema desenvolvido, mostrando as vantagens na utilização do segundo, principalmente atreladas à capacidade de avaliação da intensidade dos contatos oclusais. Por fim, ainda se valendo do uso paralelo do papel carbono, foram efetuados ensaios de análise da oclusão em sujeito envolvido no projeto utilizando placa miorrelaxante, possibilitando a utilização da capacidade do sistema desenvolvido para executar ajustes precisos na placa, com a finalidade de manter o balanço de pressões oclusais entre ambos os lados da arcada. / This essay has as objective the study and development of an engineer solution for the dental occlusion mapping and characterization, not only related to the interarch occlusal contact distribution, but also through the pressure magnitude measuring in each. In this sense, was employed effort in the Odontological knowledge increasing and also in the system elaboration allowing the occlusion state evaluation and acting as an easier agent in the occlusion disturbs diagnosing using a specific pressure/force sensing resistor (FSR) for dental use. Initially the main concepts of the Odontological sphere are presented, clarified and took as a base for all the projects in this essay. Besides this, different teeth contacts shapes are shown, as well as the occlusion’s types and its problems. In the system development, two printed circuit boards were designed for sensor interfacing. The hardware access through the firmware was elaborated, even as the global system managing software. All the conditioning circuit blocks and functionalities were evaluated and validated. In the same way, the sensor’s output and its homogeneity through all the sensible extension were evaluated, showing that the sensor’s electric resistance changes in the same form in all the sensor’s areas. In addition, it was showed the advantages of the developed system over the articulation paper through trials with a subject that is part of the project, showing the capacity of the system in the occlusion contacts magnitude evaluation. Ultimately, also in an articulation paper versus the developed system comparison, it was made occlusion analysis trials with subject using a night guard for teeth grinding (bruxism), showing the system capacity of precisely adjust the night guards for pressure balance between the both arch sides.

The diploma thesis deals with the examination of postural functions in pupils (11-15 years old) at chosen primary schools. There were two primary schools (Baarova in České Budějovice and 2nd primary school in Jindřichův Hradec) involved in the research. Standardized test were used for finding out the quality of the body to appraise the figure according to Jaroš and Lomíček and evaluating of body holding according to Matthias, functional tests of muscles that tend to be shortened and weaken. We expected the incidence of bad body holding and muscle unbalance in 30 % of the tested people within the intentionally chosen group. The research group achieved better results in the period given.

The diploma thesis deals with determination the influence of targeted motion activity on formation the right body holding habit in primary school pupils (teenagers). The research was realized at 2nd primary school in Jindřichův Hradec among the pupils in the 7th grade. The aim of the research was to compile an experimental as well as a control group. Within these groups an entrance examination in a form of standardized functional tests for the body holding appraisal and finding out the shortening and weakening the muscles was accomplished. Subsequently, an exercise programme for supporting the right body holding and elimination of muscle unbalance for the period of 6 weeks was put together. Afterwards, a final appraisal of the experimental and the control group was accomplished (the identical with the entrance one). When comparing the entrance and final examination we found out that within the experimental group all the monitored parameters improved. There was no positive change of monitored parameters within the control group that was not involved in the exercise programme.

Work-life balance among medical doctors in Nigeria : a gender perspective

This exploratory study examined the perceptions and realities of work life balance among medical doctors in Nigerian Public Hospitals. The study aimed to investigate the coping strategies adopted by medical doctors to manage work life balance and finally to examine the gender differences in the lived experiences of male and female medical doctors in Nigeria. The literature revealed that conceptualising work life balance models or employee flexibility in an African work setting is problematic when juxtaposed with primordial African cultural values where work and life are an indissoluble existential unit. The transition from an agrarian economy to new capitalist workplaces in many African countries is still relatively new. This study fills the gap by unveiling the implications this narrative has for understanding contemporary WLB. Forty-one semi-structured in-depth interviews were conducted for this study. It involved male and female doctors from public hospitals across three geopolitical zones of Nigeria. The sample of doctors interviewed were specifically resident doctors with a wide range of specialties. Thematic method of data analysis was adopted to provide major themes which were used to discuss the findings of the research investigations. The findings reveal that work in the Nigerian medical sector is notoriously intense as it underscores challenges of long working hours, intense work demand and staff shortage. The findings further suggest women must tread a ‘delicate balance’ between subordination to male authority, domestic responsibilities and ambition/achievement in a professional career. This study makes two key contributions to knowledge. Firstly, the study contributes to existing literature on work-life balance in the Nigerian context, elaborating the situation with work-life balance initiatives and how medical doctors in Nigeria manage to cope with the shortcomings of the organisations. Secondly, the findings fostered a useful extension of the work-family border theory. The border theory explains how individuals navigate between life domains and boundary management. However, the theory has provided little information on the factors that affect how employees manage and negotiate between the domains. This led to the development of the work spiritual life balance model.

Avaliação dos distúrbios ácido-base e eletrolíticos de cães com cetose e cetoacidose diabética / Evaluation of acid-base and electrolyte disturbances in dogs with diabetic ketosis and ketoacidosis

A cetoacidose diabética (CAD) é uma das complicações mais graves do diabetes mellitus (DM) em pequenos animais. A CAD é uma emergência médica caracterizada por alterações metabólicas extremas, incluindo hiperglicemia, acidose metabólica, cetonemia, desidratação e perda de eletrólitos. Embora seja um distúrbio comum e de alta mortalidade, os padrões dos distúrbios ácido-base de cães com CAD ainda não foram avaliados objetivamente. Muitas das assunções sobre a CAD em cães são generalizadas com base em dados de pacientes humanos e estudos experimentais em cães. O objetivo do presente estudo foi descrever os distúrbios ácido-base e eletrolíticos de cães com CAD e cetose diabética (CD) e caracterizá-los segundo a freqüência de ocorrência, adequação dos mecanismos de compensação e ocorrência de distúrbios mistos. Foram avaliados 40 cães diabéticos (22 animais recém diagnosticados e 18 cães em tratamento com insulina) atendidos apresentando cetonúria e hiperglicemia (&gt;250 mg/dL). De acordo com critérios clínicos, esses cães foram distribuídos em dois subgrupos: (CAD, n=22 e CD n=18) e foram determinados o pH e a hemogasometria arteriais e eletrólitos plasmáticos (sódio, cloro, potássio, cálcio ionizado) e o magnésio total e o fósforo inorgânico séricos. As alterações do equilíbrio ácido-base foram avaliadas sistematicamente pelo método de Van Slyke-Henderson-Hasselbalch. Os resultados foram comparados com os obtidos a partir de 37 cães clinicamente hígidos. Com relação aos distúrbios ácido-base, a acidose metabólica foi o mais comumente identificado (n = 27). A maior parte dos animais apresentava acidose normoclorêmica. A acidose hiperclorêmica foi observada em sete pacientes. Dos cães com acidose metabólica, 15 apresentavam alcalose respiratória concomitante. A distribuição dos valores de eletrólitos foi diferente entre o grupo de estudo e o controle, com exceção do magnésio. Não houve diferença na distribuição dos valores dos eletrólitos entre os subgrupos, com exceção do potássio plasmático. A hiponatremia e a hipocloremia foram os distúrbios eletrolíticos mais comumente observados nos 40 cães com DM. A hipocalemia ocorreu com maior freqüência no subgrupo CAD e a hipercalemia no subgrupo CD. Os valores do fósforo inorgânico sérico foram semelhantes entre os subgrupos de estudo. A hiperfosfatemia foi comum em ambos os subgrupos e nenhum paciente apresentou hipofosfatemia. A hipermagnesemia foi observada em sete pacientes com CAD e em apenas um com CD. A maior parte dos pacientes tinha hipocalcemia por ocasião do atendimento inicial. Distúrbios ácido-base mistos, principalmente a acidose metabólica normoclorêmica associada a alcalose respiratória são comuns em cães com cetose ou cetoacidose diabética, assim como distúrbios eletrolíticos como hiponatremia, a hipocloremia e hipocalemia e hiperfosfatemia. / Diabetic ketoacidosis (DKA) is one of the most serious complications of diabetes mellitus (DM) in small animals. DKA is a medical emergency characterized by extreme metabolic abnormalities, including hyperglycemia, metabolic acidosis, ketonemia, dehydration, and electrolyte losses. Despite it is a common disorder and with high mortality, the patterns of the acid-base disturbances in dogs with DKA were not evaluated objectively. Many of the assumptions about DKA in dogs are derived from studies in human beings and experimental studies in dogs. The objective of the present study was to describe the acid-base and electrolytic disturbances in dogs with DKA and diabetic ketosis (DK) according to their frequency, adequacy of the compensatory mechanisms e occurrence of mixed disturbances. Forty dogs with DM (22 with new onset diabetes and 18 insulin-treated dogs) with ketonuria and hyperglycemia (&gt; 250 mg/dL) were enrolled. On the basis of clinical criteria, the dogs were assigned to one of two subgroups: (DKA, n=22 e DK n =18). Arterial blood gases and plasma electrolytes (sodium, chloride, potassium and, ionized calcium), and serum total magnesium and inorganic phosphorus were determined in all dogs. The acid base abnormalities were evaluated systematically by the Van Slyke-Henderson-Hasselbalch method and the results compared to those obtained from 37 healthy dogs (control group). Metabolic acidosis was the most common acid-base disorder identified (n = 27) and most of the dogs had normochloremic acidosis. Hyperchloremic acidosis was observed in seven patients. Fifteen of the dogs with metabolic acidosis had coexisting respiratory alkalosis. The distribuition of the electrolytes values was different between the study group and the control group, with the exception of serum magnesium. The distribution of the electrolytes values was similar between the subgroups, with the exception of plasma potassium. Hyponatremia and hypochloremia were the most common observed electrolyte abnormalities showed in dogs with DK or DKA. Hypokalemia occurred more frequently in dogs with DKA and hyperkalemia in dogs with DK. Serum inorganic phosphorus values were similar between the subgroups. Hyperphosphatemia was a common finding and hypophosphatemia was not observed. Hypermagnesemia was detected in seven patients with DKA and in only one with DK. Most of the dogs were hypocalcemic on admission. Mixed acid-base disorders, mainly metabolic normochloremic acidosis with coexisting respiratory alkalosis are common in dogs with diabetic ketosis or ketoacidosis and electrolytic disturbances, mostly hyponatremia, hypochloremia, hypokalemia, and hyperphosphatemia, were also common.
