El trabajo está orientado tanto al estudio de los eventos y mecanismos por medio de los cuales la estrella adquiere la composición deficiente en H, como así también al estudio de su evolución posterior como estrella deficiente en H.

Využití mykorhizních hub ve školkařských substrátech

The aim of my thesis in the experimental part was to prove new composition of nursery substrate with the adding of mykorrhiza. The evaluation where there was the kind Forsythia x intermedia Z. 'Golden Times used was done in four varieties of decorative substrate with the adding of different amount of inoculant. In the checking Hample there was no inoculant used. I evaluated these parameters: height of plants, number of shoots, length of shoots, diameter of root --collar, volume of root ball and rate of mykorrhiza. This experiment was done in the experimental area of the Institution of cultivation and reproducing of garden plants of Garden faculty of the Mendel university in Lednice. The obtained results are worked statistically. The theoretical part describes other authors'experience with studying mycorrhiza's symbiosis

CoordenaÃÃo de AperfeiÃoamento de Pessoal de NÃvel Superior / The seaweed Gracilaria are important sources of agar sulfated polysaccharides that are commonly used in the food, pharmaceutical and biotechnology industries. For characterization of the media, the kelp was subjected to analysis of their chemical composition, which indicated their percentages of moisture, ash, protein and lipids. Extractions in which extraction by enzymatic digestion native extract (a seaweed is not subjected to pretreatments and obtaining the polysaccharides is by their boiling) is: For evaluation of red seaweed polysaccharides Gracilaria intermedia fractions were obtained for the following extraction methods seaweed is pretreated with NaOH and KOH in concentrations of 1% and 5%. The extracted material was subjected to colorimetric test chemical composition (protein content determination and quantification of carbohydrates, sulfate and 3,6-anhydro-galactose). The material was further analyzed by infrared spectroscopy with Fourier transform and Nuclear Magnetic Resonance ( NMR). Antioxidant activity with each of the fractions obtained tests were performed. Antioxidants tests (total antioxidant capacity, chelation of ferrous ion) indicated that such polysaccharides have the ability to scavenge free radicals and significant dose-dependent manner. Due to these results, the polysaccharide obtained from seaweed Gracilaria show potential intermediate that they will not be used as a synthetic antioxidant compound. Experiments to evaluate the rheological behavior of aqueous solutions of polysaccharides under study were performed at 25  1 ÂC, using stationary and dynamic oscillatory measurements, which indicated that the material obtained should be classified as pseudoplastic and highlighted the skills of such polymer solutions in acting as thickeners and gelling agents. / Algas marinhas do gÃnero Gracilaria sÃo importantes fontes de Ãgar, polissacarÃdeos sulfatados que sÃo comumente usados nas indÃstrias alimentÃcia, farmacÃutica e biotecnolÃgica. Para caracterizaÃÃo do material de trabalho, a alga marinha foi submetida à anÃlise de sua composiÃÃo centesimal, a qual indicou seus percentuais de umidade, cinzas, proteÃnas, lipÃdios e carboidratos totais. Para avaliaÃÃo dos polissacarÃdeos da alga vermelha Gracilaria intermedia foram obtidas fraÃÃes pelas seguintes metodologias de extraÃÃo: 1. extraÃÃo por meio de digestÃo enzimÃtica, 2. extraÃÃo nativa (a alga nÃo à submetida à prÃ-tratamentos e a obtenÃÃo dos polissacarÃdeos ocorre pela sua fervura) e 3. extraÃÃes nas quais a alga à prÃ-tratada com NaOH e KOH nas concentraÃÃes de 1% e 5%. Os materiais extraÃdos foram submetidos a testes colorimÃtricos de composiÃÃo quÃmica (determinaÃÃo do teor de proteÃnas e quantificaÃÃo de carboidratos, sulfato e 3,6 anidrogalactose). Os materiais foram ainda analisados por espectroscopia de infravermelho com transformada de Fourier e RessonÃncia MagnÃtica Nuclear (RMN). Foram realizados testes de atividade antioxidante com cada uma das fraÃÃes obtidas. Os testes antioxidantes realizados (capacidade antioxidante total e quelaÃÃo do Ãon ferroso) indicaram que tais polissacarÃdeos possuem capacidade de sequestrar radicais livres de maneira significativa e dose-dependente. Devido a estes resultados, os polissacarÃdeos obtidos a partir da alga marinha Gracilaria intermedia mostram potencial de virem a ser utilizados como um composto antioxidante nÃo sintÃtico. Experimentos para avaliar o comportamento reolÃgico de soluÃÃes aquosas dos polissacarÃdeos em estudo foram realizados a 25  1 ÂC, usando medidas estacionÃrias e dinÃmicas oscilatÃrias, que indicaram que os materiais obtidos devem ser classificados como pseudoplÃsticos e apontaram as habilidades de tais soluÃÃes polimÃricas em atuar como agentes espessantes e gelificantes.

Transmission of kalilo DNA in senescent strains of Neurospora intermedia

Senescence, the progressive loss of growth potential culminating in death, is common among Kauaian strains of Neurospora intermedia. Senescence is initiated by the insertion of kalilo DNA into the mitochondrial DNA. Mitochondrial DNA molecules carrying the insert accumulate and death occurs when the insert is equimolar with the mitochondrial DNA. The inserted form of kalilo DNA is referred to as mtlS-kalDNA. Studies on the somatic transmission of mtlS-kalDNA in ascospore series have revealed that kalilo DNA is capable of assuming new locations within the mitochondrial DNA. It is proposed that these novel insertions originate from intramitochondrial movement and an autonomous form of kalilo DNA, mtFF-kalDNA, is predicted to be an intermediate in movement. Novel insertion of kalilo DNA appears to depend on the form of mtlS-kalDNA transmitted sexually. If a mutagenic insert is transmitted, senescence is initiated at the onset of vegetative growth of the ascospores and no novel insertions are detected. The lifespans of these ascospores are quite short, death occurring in 10 subcultures or less. Transmission of a nonmutagenic insert delays the onset of senescence until either a novel insertion or a rearrangement of the transmitted insert occurs. The lifespans of these ascospores usually exceed 10 subcultures and are variable. Information obtained from tetrad analysis has revealed that novel insertion of kalilo DNA may also be under the influence of the host genome. A senescent Kauaian strain was identified which shows some but not all characteristics of kalilo senescence. In this strain and its derivatives, the behaviour of mtlS-kalDNA is erratic and in, some cultures the characteristic mitochondrial biochemical deficiencies, normally accompanying kalilo senescence, are not observed. It is suspected that kalDNA is not responsible for senescence in this strain and its derivatives but rather some other unknown factor is affecting the normal growth patterns of these cultures. Kauaian strains were surveyed for the presence of dsRNA to determine whether kalDNA has a viral origin. Only one senescent strain contains detectable amounts of dsRNA which was not homologous with a kalDNA probe. The survey identified six nonKauaian strains which contain dsRNA and seven dsRNA species were delineated. Although the presence of dsRNA is not relevant to kalilo senescence, analysis of dsRNA in a genetically-well defined organism like Neurospora may give insight into the significance of dsRNA in fungi in general. / Medicine, Faculty of / Medical Genetics, Department of / Graduate

La presente tesis doctoral emprende la investigación de lo monstruoso y sus implicaciones de carácter ideológico, estético y político, especialmente desarrollado en los vídeos musicales de Floria Sigismondi y Chris Cunningham en la década de los noventa. En ese período en concreto nace una nueva generación de directores de videoclips, que ya no se encuentran en el anonimato, como sucedía en los ochenta, ahora son conocidos y respetados, además de transgredir frecuentemente las reglas del marketing promocional del clip estandarizado. Directores entre otros como Michel Gondry, Chris Cunningham, Spike Jonze y Floria Sigismondi, estarían dentro de esta lista privilegiada. Por lo demás estos respectivos realizadores en general trabajan misceláneos ámbitos estilísticos fronterizos, la ultrafragmentación, la intertextualidad y las narraciones figurativas no lineales. Nuestro objetivo es proponer un corpus teórico interdisciplinar que determine las consecuencias que origina lo abominable en el lenguaje fluctuante del videoclip de esos respectivos autores, y sus analogías con otras corrientes del pensamiento postmoderno; a la vez que nos permiten ofrecer nuevas interpretaciones transversales sobre la ideación corporal inverosímil. A nivel metodológico hemos partido del estudio de la monstruosidad histórica y sobre todo de la cinematografía de terror como antecedente de la otredad somática en movimiento, de la cuál formulamos la hipótesis de que el concepto de lo teratológico contemporáneo aplicado al videoclip de factura grotesca; es similar en muchos aspectos al mencionado cine de género, especialmente desde la vertiente del psicoanálisis. The way of the Jedovnice creek: connection with the spring Falling into the underground halls and back to the light. The Water Way is an intermedial poem that is concerned to the element of water, character of its way through the landscape and its presence in the landscape. Since the type of work I most prefer is the work in the landscape, I am returning back to the natural landscape, strong places in Moravian karst near Brno. The audiovisual poem is primarily focused on the moment of melting. Sublimation of ice from the ice shelter, ephemeral phenomena on the surface, such as miniature whirls, waves, rays of sunlight and especially moonlight. The poem focuses on the possibilities of transferring the experience with a specific place, and the events in a unique, unrepeatable time and space into the theatrical scene and possibilities of the musical and audiovisual interpretation of such experience. I work with digital media (video mix and audiostop) to transfer the artist's physical experience in a unique time and unique natural setting into the scenic form of the work. Considering the physical and symbolic qualities of the water element, I have abandoned the composition of the event in the classical sense.

ANTIBIOTIC RESISTANCE OF PERIODONTAL PREVOTELLA INTERMEDIA/NIGRESCENS IN 2011 AND 2021

Prevotella intermedia/nigrescens group bacteria are Gram-negative, non-motile, anaerobic rods abundant in the subgingival microbiome of human periodontitis patients, and relatively sparse in persons with periodontal health. P. intermedia/nigrescens may be inadequately suppressed in periodontal pockets with conventional mechanical-surgical forms of periodontal therapy. Therefore, short-term systemic antibiotic therapy is often recommended in the treatment of recalcitrant (refractory) severe periodontitis patients where high numbers of P. intermedia/nigrescens persist in the subgingival microbiota. Limited available data suggests that the antibiotic sensitivity profile of periodontal P. intermedia/nigrescens has changed over time among severe periodontitis patients in the United States, with increasing levels of antibiotic resistance reported. These findings have potentially important clinical implications for dental professionals and their severe periodontitis patients which need further confirmation and clarification. To further expand knowledge on this clinically relevant issue, the purpose of the present study was to determine and compare over a 10-year period (2011 versus 2021) the prevalence of in vitro resistance of periodontal P. intermedia/nigrescens to the antibiotics amoxicillin, metronidazole, clindamycin, and doxycycline. / Oral Biology

This thesis' focal presentable object – to swim in air – is a mythosystem comprising six iteratively malleable experiential systems of intermedial musical and visual performance works composed by myself between the years 2018 and 2023. Conceived through the lens of Jennifer Walshe's New Discipline, created within my practice cycle's nodal context, and connected by a sub/conscious structure of perceptual timbre, the mythosystem and its parts form the centerpiece of this discussion of context, process, and method. As described in this document, the creative practice of nodal context and the adaptive intermedial methods used in the conceptualization and composition of to swim in air were developed through a personal and pragmatic application of feminist writer and independent scholar Sarah Ahmed's Queer Phenomenology: Orientations, Objects, Others, and composer, musicologist and trombonist George Lewis' curatorial decolonization guidelines as outlined in his "8 Difficult Steps to Decolonizing Music" towards the creation of presentable cultural objects which invite variable and continuous interaction from their participants through the exploration of the reciprocity of community, multi-practice creative strategy and malleable forms. Throughout this document I discuss how through the exploration of the reciprocity of community, multi-practice creative strategy and malleable forms I have addressed concerns of cost, access and participation in living culture with regards to my own work creating cultural objects. I also discuss phenomenological and practical issues to do with cost, access and living culture present within the creative, curatorial and institutional spaces and communities in which this work was created and is initially intended to exist, and how these concerns impact the pedagogical, practical and experiential potential of my own work. This document also reflects on how the pragmatic and personal adoption of these concepts of cultural object phenomenological ephemera and decolonized curatorial practice as presented by Ahmed and Lewis by other composers, artist, curators, educators, and other creative professionals might impact the nature of artistic institutional spaces, might encourage the engagement of new, speculative and/or more colloquial spaces, and how these spatial reorientations might encourage a more fluidic approach to creative practice, community cultivation and engagement.

P. gingivalis é um dos principais patógenos da doença periodontal, é encontrado em biofilmes orais com S. gordonii e P. intermedia e em células endoteliais da artéria coronária in vivo. P. gingivalis necessita de ferro em seu metabolismo e pode usar certas proteínas do hospedeiro como fontes deste íon em ambientes limitantes. Assim, este estudo investigou o papel dos genes PGN0741/PG0637 (receptor dependente de TonB) e PGN0531/PG1380 (fvW) de P. gingivalis na formação de biofilme em diferentes concentrações de ferro, em biofilmes mistos com S. gordonii e P. intermedia, e na adesão e invasão de células endoteliais da artéria coronária. Os resultados mostraram divergências no papel dos genes TonB e fvW na formação dos monobiofilmes e mistos e em diferentes concentrações de ferro, demonstrando uma relação cepa-dependente. Na adesão, fvW se mostrou importante para ambas cepas, mas na persistência apenas para P. gingivalis W83. Este trabalho enfatiza, assim, a necessidade do uso de mais de uma cepa de P. gingivalis no estudo do papel de genes em ensaios experimentais. / P. gingivalis is one of the major pathogens of periodontal diseases. It is found in oral biofilms associated with S. gordonii and P. intermedia, and inside of coronary artery endothelial cells in vivo. P. gingivalis requires iron for growth and can exploit iron-carrying proteins of the host as sources in limiting environments. Thus, this work aimed to study the role of genes PGN0741/PG0637 (TonB-dependent receptor) and PGN0531/PG1380 (fvW) of P. gingivalis in biofilm formation under different iron concentrations, in mixed biofilms with S. gordonii and P. intermedia, and in the adhesion and invasion of coronary artery endothelial cells. Our data showed discordance for the role of TonB and fvW in homo- and heterotypic biofilm formation and in different iron concentrations. The relevance of both genes was strain-dependent. Gene fvW was relevant for adhesion to endothelial cells, but only for strain W83 during persistence. Therefore, our study emphasizes the importance of using different strains for a better understanding of the role of genes in experimental assays.

Diese Veranstaltung gilt in zahlreichen Geschichten über die autonome bzw. inoffizielle Kunstszene der DDR als wichtiges Ereignis, an dem das ansonsten lose verbundene Netzwerk regionaler Szenen an einem Ort zusammen- und an dem Kunst- und Punkszene aufeinander traf. Musik, Malerei, Film und Tanz/Bewegung sind die Medien der Aufführungen, die als Intermedia, als Collagen in Zeit und Raum, beschrieben werden. Wie lässt sich die Vernetzung der Künste und die temporäre Vernetzung der Akteure anhand eines solchen ‚Knotens’ untersuchen und darstellen?
