Computational models for conformations of cell wall mycolates from Mycobacterium tuberculosisPrinsloo, Wilma 12 June 2009 (has links)
Literature highlights the effects of mycolic acid (MA) fine-structure on biological activity, pathogenicity, virulence and cell wall structure and permeability. Knowledge on MA-structure and how their conformations are dependent on their precise molecular composition becomes essential in exploiting these properties in drug-design and in advancing our understanding of the disease. In our group evidence for a structural or functional relationship between cholesterol and MAs have been discovered. The aim of the experimental part of this work was to study this relationship further by attempting to quantify the interaction between cholesterol and MAs in liposomes on an evanescent field biosensor. The binding profiles that were obtained could not be evaluated with kinetic software and the interaction between cholesterol and MAs was not linearly dependant on the concentration of cholesterol. However, novel insight into the interaction was gained when it was observed that cholesterol only accumulated on MA liposomes when cholesterol liposomes containing concentrations of cholesterol resulting in a suspected liquid ordered phase, were used. This is significant since it implies that cholesterol in membrane rafts of the host cell that exist in a liquid ordered phase would be able to interact with MAs under physiological conditions. The theoretical part of this work represents the first molecular modeling study in which MAs are allowed to fold with no conformational restrictions. It is proposed that MAs fold as a function of their functional groups, stereochemistry, and various chain lengths. It was also investigated whether methylation of the acid group changes conformational preferences. The effect of chain length on cyclopropane structure and the viability of systematic conformational searching in MAs were shown using quantum mechanics. Replicate molecular dynamics simulations were done for 4 ns in vacuo on alpha-; methoxy-; methoxy methyl ester- and keto-MAs. MAs had an open starting conformation without conformational restrictions. Results were analysed using eight distances characteristic of the conformational fold. Using these distances, W-, U- and Z-shaped folds were identified. Principal component analysis (PCA) and self-organising maps (SOMs) were used to evaluate differences and trends in MA-conformations. Quantum chemical results showed that chain length did not affect cyclopropane structure and that the systematic plotting of potential energy surfaces is an effective tool to analyse effects of changes in geometry on the energy of the molecule and to predict favoured conformations. Remarkably, single MAs assumed W-, U- and Z-folds in vacuo during molecular dynamics simulations that have previously been observed in monolayers. PCA and SOM plots showed that keto-MA folded faster than other MAs. Alpha-MA showed the highest frequency of W-, U- and Z-folds. Methoxy-MA did not readily fold at its cis-cyclopropane group. Methylation of the acid group of methoxy-MA did not show remarkable differences in the conformations assumed, but almost doubled the frequency of WUZ-structures obtained as compared to non-esterified methoxy-MA. The inherent structural differences between MA-subclasses clearly affect the trends in structural folds that they assume. Molecular modeling of MAs proved to be a versatile tool for resolving structure-function relationships at the molecular level. Copyright 2008, University of Pretoria. All rights reserved. Words about nothing: writing the ineffable in Calvino and Ma YuanTeichert, Evelyne 05 1900 (has links)
The thesis links the writings of the Italian Italo Calvino and the Chinese Ma Yuan through
the Taoist symbol of the Tao and the Borgesian concept of the Aleph, an imaginary point in
space containing all points in space and time. Based on Zhuangzi’s parable of the Emperor Hun-tun (Chaos) who lost his original state of chaos when he had sensory openings poked into him,
the vision of the Aleph/Tao represents the return to that chaotic state of undifferentiated
knowledge one experiences when one closes all sensory perceptions. This unnameable vision
allows one to transcend all apparent conceptual dichotomies as it lies in the realm of intuition
rather than language. Calvino, like Borges, posits that the chaos of the universe cannot be
represented through the sequential language system, but nevertheless demonstrates this
ineffability through language. Ma Yuan celebrates the chaos of life by writing about a
mythological Tibet, upholding the uniqueness of that culture as a subtle subversion to the Chinese
political and territorial takeover.
Chapter One and Two, respectively, discuss the “Overlapping Conceptual Spaces” in
Calvino’s Invisible Cities and Ma Yuan’s ‘The Temptation of the Gangdisi’. Chapter Three looks
in greater detail at the images of the Aleph and the Tao in the two main texts against the
backdrop of Borgesian thought. In accordance with the concept of the Aleph/Tao whose
definition is continuously unsettled by contradictory conjectures, the fourth chapter undoes the
conclusions reached in the previous chapters. This chapter discusses Calvino’s Cosmicomics and
Ma Yuan’s shorter Tibetan stories in the light of comic parody. That which was earlier posited as
the ineffable in these stories is elaborated in a profusion of words. The Conclusion discusses
from a Taoist point of view the predominantly male voice in the writings of the two authors.
While both advocate the spiritual sameness of all phenomena in an undifferentiated knowledge of
the world, they nevertheless write from the male perspective of the yang pursuing and wanting to
possess the yin. / Arts, Faculty of / English, Department of / Graduate

How institutions matter in MA research : a literature reviewHärström, Malin January 2019 (has links)
It is currently popular to study management accounting (MA) from an institutionalist perspective. Such a perspective is premised on that ‘institutions matter’; so that notions of social structure, agency and linkages in between them thus become central. While readings of the institutionally oriented MA-literature reveals interesting and insightful contributions about MA, they also unearth concerns regarding diversity and fragmentation in conceptualization and modelling. Therefore, the purpose of this licentiate thesis is to identify, classify and synthesize how ‘institutions matter’ in extant MA research and, based on this, suggest avenues for future research. To do so, this licentiate thesis entails a review of 34 empirical institutionally oriented MA studies. The findings entail ‘answers’ to 4 particular review questions regarding current (1) Conceptualizations of MA, (2) Overall explanatory models of agency, (3) Social roles of MA and, (4) Sources of MA practice – questions which are all derived from what I refer to as ‘A Basic Model of institutional theorizing (ABM)’. Overall, this review provides maps which are previously not pursued for this literature for the areas of each respective review question. Within such broader achievements, it identifies five ontological approaches to a relationship between social structure and agency instead of the established three. It also adds four social roles of MA previously not discussed for this literature. Finally, it suggests a uniform set of ‘sources of MA practice’ unlike the current literature that typically separates sources of continuity, change, homogeneity or heterogeneity respectively. The thesis concludes with a discussion of key findings and contributions, based on which a number of avenues for future institutionalist conceptualization and modelling of MA are suggested.

Ma Sicong's Violin Artistry and CompositionsYao, Xuan, 0000-0001-9986-1684 January 2022 (has links)
Ma Sicong was a violinist, composer, and educator of the highest caliber in China who devoted his entire professional life to the instrument. Known for producing violin music in various styles and forms for more than fifty years, Ma Sicong contributed to the violin's popularization in China through his performance and composition with Chinese elements. His violin music incorporates principal melodies from Chinese folk music, and his talents as a mature Western composer talents blend these melodies into violin solos with a distinct national identity. The Inner Mongolia Suite and First Rondo by Ma Sicong, both of which are considered among the most outstanding violin solos ever written for the instrument, are examined in this dissertation, as is the composer's own playing experience, to gain a better understanding of the compositions and their meaning. There are several ramifications for Chinese composition for the violin due to the inclusion of national features that affected the design and development of the violin. The author also hopes that this dissertation can serve as a beneficial resource for future students interested in comparable topics. / Music Performance

Down by the line : A study of the causes of girtingEkeroth, Patrik, Erngård, Johannes January 2015 (has links)
This essay studies the causes of girting, i.e. accidents where tugboats get pulled down and capsizes by the vessel it is towing. The study uses 14 official accident reports about girting accidents all over the world. The HFACS-MA model is used to categorize the causes of the accident into a causal chain, in order to find out which categories are most frequently present in the accident reports. The result shows that poorly planned tow operations (with inadequate planning and/or risk assessment), deficiencies in the organization’s documentation (i.e. manuals, instructions and procedures), and poor communication between the involved parties were the most frequently found factors for girting. In order to reduce the risk of girting, recommendations have been made that includes the importance of keeping documents up to date, maintain clear communication between the parties involved during towing operations and to plan each operation thoroughly, giving everyone involved the opportunity for feedback regarding the plan.

This research analysis the roll of the Brazilian Japanese descendant from the perspective of an artist of the body, with the scenic arts of the city of São Paulo as context: a Brazilian reality of immigration and diaspora, in a practicaltheoretical bias. Our start points are the processes of contamina-tion between West and East, represented here by Brazil and Japan, respectively, and the singular experiences that result from this relation, which are represented by the conversation with six Brazilian nikkei artists. We are considering the Miscegenation (mestiçagem) (PINHEIRO, 2013; LAPLANTINE & NOUSS, 2016; GRUZINSKI, 2001) that occurs from this rendez-vous, using terms from the Japanese culture, inclusive, Ma (?) and D? (?), properly displaced from its meaning and resignified in the context that we mentioned before. The Corpomídia Theory from Greiner and Katz (2001) is important for the comprehension of the co-relation and co-evolution of body and environment which is mutual and non-stagnant. The studies of Hashiguti (2008) and Takeuchi (2016) that reflect specifi-cally the Japanese descendant body also compose the theorical refer-ences of this study.

População de Mansonia humeralis (Diptera: Culicidae), sob o impacto de inundação da Represa Porto Primavera, Município de Presidente Epitácio, São Paulo, Brasil / Population of Mansonia humeralis (Diptera: Culicidae), under the Porto Primavera Dam flooding impact, Presidente Epitácio municipality, São Paulo State, BrazilPaula, Marcia Bicudo de 04 August 2005 (has links)
No sítio JB, no período do pré-impacto, Ma. humeralis representou 3,1%. / Objective. To assess the ecological and biological aspects of the population of Mansonia humeralis, in area under the Porto Primavera Dam flooding impact, Presidente Epitácio municipality, São Paulo State, Brazil, in order to understand the vector capacity of this population. This population dominance over other Culicidae species that constitute the local fauna represents public health concern because of the possibility of transmission of pathogenic agents and nuisance to human populations. Methods. In the JB Ranch adults Culicidae were monthly captured for 15 months, on each of the previous and posterior periods of the two flooding events of the Porto Primavera Dam, and for the 24 months after the second flooding in the Santo Antônio Farm. Captures were undertaken by morning Aspiration, Human Attraction Technique for 24 hours, Shannon traps, CDC light traps and Human Attraction Technique near sunset. Temperature and rainfall were accounted for temporal variation of the studied population. To environmental impact analysis on Ma. humeralis population richness, diversity, dominance, abundance and similarity, indexes and parametric and non-parametric statistics and correlation were used, blood feeding time activity was calculated by the Williams\' Mean, harmonic mean and simple linear regression model to flight range dispersal were used and to parity, chi-square was used. Results. In the JB Ranch, on the pre-impact period, Ma. humeralis represented 3.1%. After the first flooding Ma. humeralis represented 59.6% and after the second flooding it represented 53.8%, indicating significative dominance of this species and richness and diversity reduction of other Culicidae. Larger population densities of Ma. humeralis were found in the autumn and winter on the pre-filling of the Dam, in the winter and the spring after the first flooding and in the spring after the second flooding. Ma. humeralis was among the more abundant species and was similar on the fauna post-impact period. Blood feeding time activity of Ma. humeralis was practically of 24 hours, but its higher frequency was on the first post-crepuscular interval and on the 18:00 to 24:00 hours interval. In the Santo Antônio Farm, the largest populational density of Ma. humeralis occurred in the summer and in the spring and the means for species diversity, richness and dominance were close to the values found in the pre-filling in the JB Ranch, with captures undertaken by Shannon traps during sunset. As for parity, more parous females were found in the autumn. Ma. humeralis presented low flight radius dispersal. Conclusions. The construction of the Porto Primavera Hydroelectric Power Plant and the dam of the Paraná River, in the studies conducted in Presidente Epitácio, on the left margin of this river and the Peixe River, presented a reduction on the diversity and richness of the Culicidae fauna undertaken by different capture techniques. The dominance of Ma. humeralis showed that the environmental impact on the area supported its development as indicated by its abundance and permanence on the similar fauna, after the two flooding events. The highest densities of Ma. humeralis, associated with aquatic plants, seems to be connected with the renewable waters of the great reservoirs. Ma. humeralis showed a tendency to anthropophily, with the blood feeding activity preferably at sunset and at night. The low flight dispersal, does not allow to conclude if Ma. humeralis would not fly higher distances in search of blood sources. It is relevant to monitor some Culicidae populations favoured by dam constructions, once there is frequent man exposition on the rivers\' edges, due to fishing activities, hunting, leisure and dwelling.
