Extratos vegetais na propagação in vitro de Epidendrum ibaguense Kunth / Plant extracts on spread in vitro epidendrum ibaguense Kunth

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / A micropropagação de orquídeas é uma técnica utilizada com vários objetivos, podendo-se destacar a multiplicação rápida de plantas, propagação de novos híbridos, produção em larga escala e obtenção de matrizes livres de patógenos. A utilização de extratos naturais com ação antimicrobiana na micropropagação mostra-se como uma forma alternativa, ecológica e econômica de obtenção de plantas isentas de micro-organismos. Este trabalho teve como objetivo avaliar a utilização de extratos de Allium sativum (alho), de Syzygium aromaticum (cravo) e de própolis para evitar a contaminação com micro-organismos durante o processo de subcultivo in vitro de Epidendrum ibaguense Kunth e as possíveis interferências desses extratos no desenvolvimento vegetativo das plântulas. Foram isolados e identificados os micro-organismos Aspergillus sp., Candida sp., bactéria Gram-negativa, bactéria Gram-positiva e Penicillium sp. presentes em frascos com meio de cultura contaminados, oriundos do cultivo in vitro e do ambiente. Avaliou-se a ação antimicrobiana dos extratos sobre os micro-organismos isolados e observou-se que, o extrato de cravo inibiu o crescimento dos mesmos. Foram utilizados sete tratamentos compostos pelo meio Murashige; Skoog (1962) com metade da concentração de macronutrientes, acrescido de carvão vegetal e dos extratos de alho a 1,0 mgL-1 (T1), alho a 0,5 mgL-1 (T2), cravo a 1,0 mgL-1 (T3), cravo a 0,5 mgL-1 (T4), própolis a 1,0 mgL-1 (T5), própolis a 0,5 mgL-1 (T6) e o controle (T7) em um delineamento inteiramente casualizado, com cinco repetições e três plântulas por repetição. Após sessenta dias do subcultivo nos meios com os extratos avaliou-se a altura da parte aérea, o comprimento da maior raiz, o número de raízes, o número folhas, o comprimento da maior folha, o peso da massa fresca, o peso da massa seca e a taxa de multiplicação das plântulas. Houve influência dos extratos de alho, cravo e própolis no desenvolvimento in vitro de plântulas de E. ibaguense. O extrato de cravo apresentou ação antimicrobiana frente aos micro-organismos Aspergillus sp., Candida sp., Escherichia coli, Penicillium sp. e Streptococcus pneumoniae. Já os extratos de alho e própolis não demonstraram atividade antimicrobiana sobre nenhuma das cepas testadas. Através deste trabalho foi possível estabelecer um protocolo para o cultivo in vitro de E. ibaguense visando o controle da contaminação. / The micropropagation of orchids is a technique used for various purposes, may be noted the rapid multiplication of plants, propagation of new hybrid, large-scale production and obtaining free arrays of pathogens. The use of natural extracts with antimicrobial activity in micropropagation shows up as an alternative, environmentally friendly and economical way to obtain plants free of microorganisms. This study aimed to evaluate the use of extracts of Allium sativum (garlic), Syzygium aromaticum (clove) and propolis to prevent contamination with micro-organisms during the process of in vitro subculture of Epidendrum ibaguense Kunth and the possible interference of these extracts on vegetative growth of seedlings. The microorganisms Aspergillus sp. Candida sp. Gram-negative bacteria, Gram-positive and Penicillium sp. present in the contaminated bottles with culture, derived from in vitro culture and the environment, were isolated and identified. We evaluated the antimicrobial activity of the extracts on the isolated microorganisms and it was observed that the extract of clove inhibited their growth. Seven compound treatments were used by Murashige; Skoog (1962) with half the concentration of macronutrients, plus charcoal and garlic extracts to 1.0 mgL-1 (T1), garlic 0.5 mgL-1 (T2), cloves 1.0 mgL- 1 (T3), cloves 0.5 mgL-1 (T4), propolis to 1.0 mgL-1 (T5), propolis 0.5 mgL-1 (T6) and control (T7) on a randomized complete randomized with five replications and three seedlings per replication. Sixty days of subculture in the media with the extracts evaluated the shoot height, the length of roots, number of roots, leaves number, the length of the longest leaf, the weight of the fresh weight, the weight of the mass drought and the multiplication rate of the seedlings. There were significant effects of the extracts of garlic, clove and propolis in the development of in vitro plantlets of E. ibaguense. Clove extract showed antimicrobial activity against microorganisms Aspergillus sp., Candida sp., Escherichia coli, Penicillium sp. and Streptococcus pneumoniae and the extracts of garlic and propolis showed no antimicrobial activity against any of the tested strains. Through this work, it was possible to establish a protocol for the in vitro cultivation of E. ibaguense for the contamination control.

La fonctionnalisation d'emballages biodégradables avec des antioxydants naturels est l'une des techniques prometteuses pour améliorer la conservation des aliments, diminuer la quantité de conservateurs chimiques utilises, protéger la dégradation aromatique des produits et ainsi conserver une meilleure qualité globale. Le contrôle du relargage de ces composés actifs de l'emballage vers l'aliment permet d'étendre l'efficacité de la fonctionnalisation en libérant progressivement les antioxydants à la surface de l'aliment. L'objectif global de ce travail était de fonctionnaliser le polymère HPMC afin de produire un film d'emballage coloré à activité antioxydante et d'évaluer son aptitude à servir d'emballage actif. Tout d'abord, des films d'HPMC contenant différents colorants synthétiques comme le bleu, le vert, le jaune, le rouge et le blanc ont été testés afin de déterminer la couleur la plus adaptée pour le contrôle de la photo-oxydation de produits gras. Ensuite, la couleur rouge synthétique, montrant un maximum de contrôle contre la photo-oxydation, a été remplacée par des composés actifs naturels de même couleur. Ces composés provenaient soit d'un mélange d'extraits de betterave et de carottes pourpres, d'un mélange de bétalaïnes soit d'un mélange d'anthocyanes. Le mode d'incorporation de ces composés actifs dans la matrice d'HPMC, leurs effets sur les propriétés thermiques, mécaniques, barrière et structurales des films ont été étudiés. Les résultats ont montré que l'intégration de ces différents composés actifs naturels a permis d'améliorer les propriétés des films. Les composés actifs utilisés ont la capacité de contrôler le photo-vieillissement de la matrice polymérique et que l'HPMC est un bon candidat pour incorporer ces molécules et permettre le contrôle de la dégradation de produits alimentaires riches en lipides / Biodegradable packaging functionalized with natural antioxidants is one of the promising techniques to enhance foods shelf-life, lower use of preservatives in food formations, higher protection of flavours and higher food qualities. Controlled release of these bioactive compounds from packaging to food surface provides longer food stability by continuously librating antioxidants at food surface. The overall objective of the present work was to functionalize the HPMC polymer as colored antioxidant packaging and investigate its suitability as active packaging for unsaturated lipids. Firstly, HPMC films containing different synthetic colours like blue, green, yellow, red and white were tested to chose a suitable color having control against photo-oxidation. Secondly, red synthetic color (showing maximum control against pho-oxidation) was replaced by natural active red compounds including "natural red color" (beetroot extract + purple carrot extract), betalains and anthocyanins to produce bioactive food packaging. Mode of incorporation of these active compounds in HPMC matrix and also their potential effects on thermal, mechanical, barrier and structural properties of films were investigated. Controlled release kinetics, antioxidant capacity and light stability of bioactive compounds in HPMC films were also investigated. The overall results showed that successful incorporation of different natural active compounds have capability to improve film properties. The active compounds under discussion have ability to control photo ageing of polymer matrix and HPMC has the capability for being a suitable carrier for antioxidant active packaging for some food products

Microbial and oxidative changes occurring in meat and meat products cause loss of quality and consequent commercial devaluation. The consumption of meat products has caused concern among consumers by the presence of chemical preservatives, which are often questioned because of the risk of harmful side effects to health. In an attempt to control microbial and oxidative changes, researches have been performed with the purpose of isolating natural phytochemicals obtained from various plant substrates, which besides having the preference of the consumers, also have the ability to act helping in the prevention of diseases. Antioxidant and antimicrobial activities have been attributed to the phenolic compounds; persimmon (Diospyros kaky L.) was used in this study since it is rich in polyphenols. Initially, we compared the antioxidant activity of hydroethanolic extracts of Diospyros kaki, L. of the cultivars Quioto and Rama Forte in chicken meat. The effect of these cultivars on the color and sensory characteristics of the samples was also verified. The crude hydroethanolic extract of persimmon cv. Rama Forte was used in subsequent tests since it showed a higher content of phenolic compounds, higher antioxidant activity, and it did not affect the sensory characteristics of chicken meat. The hydroethanolic extract of persimmon cv. Rama Forte was fractionated using solvents with different polarities (n-hexane, chloroform, and ethyl acetate) and subjected along with the hexane, chloroform, and ethyl acetate fractions and the residual fraction to the determination of total phenolic compounds as well as antimicrobial and antioxidant activity in vitro. The antioxidant activity in beef was verified and compared with the activity of the oily extract of rosemary (Chr. Hansen ®). The extract and/or the most efficient fractions were evaluated for antioxidant and antimicrobial activity as well as for the influence on the sensory characteristics and color of meat products (beef burgers and salami). Based on the results obtained, the crude hydroethanolic extract of persimmon cv. Rama Forte and the residual and ethyl acetate fractions showed antioxidant activity in beef during the storage period, however, no antimicrobial activity was observed. In the salami samples, the crude hydroethanolic extract of persimmon cv. Rama Forte also showed antioxidant activity and did not alter the sensory characteristics or microbial counts performed after the manufacturing period of the salami. In the frozen beef burgers, crude hydroethanolic extract of persimmon cv. Rama Forte and residual and ethyl acetate fractions, when used in the concentrations of 0.5 and 0.7%, did not show antioxidant potential or antimicrobial characteristics. Moreover, they did not interfere in the sensory samples. However, the ethyl acetate fraction (0.5 and 0.7%) contributed to the retention and stability of the red color of the samples of beef burgers during the storage of the frozen product. Thus, based on the results obtained, it seems appropriate to assume that the crude hydroethanolic extract of persimmon cv. Rama Forte as well as residual and ethyl acetate fractions have antioxidant potential. Further studies should be performed mainly in relation to the antioxidant activity in frozen meat products. / As alterações microbianas e oxidativas indesejáveis que podem ocorrer na carne ou nos produtos cárneos causam perda de qualidade e conseqüente desvalorização comercial. O consumo de produtos cárneos tem causado preocupação nos consumidores pela presença de conservantes sintéticos, os quais são questionados freqüentemente devido aos riscos de apresentarem efeitos colaterais maléficos à saúde. Com o intuito de controlar as referidas alterações, pesquisas têm sido desenvolvidas, com o propósito de estudar fitoquímicos naturais, obtidos de diversos substratos vegetais, os quais além de terem a preferência dos consumidores, também possuem a capacidade de atuarem como auxiliares na prevenção de doenças. Aos compostos fenólicos tem sido atribuída atividade antimicrobiana e antioxidante, e considerando tal capacidade foi utilizado neste estudo o caqui (Diospyros kaky L.) que é rico em polifenóis. Inicialmente foi comparada a atividade antioxidante dos extratos hidroetanólicos de Diospyros kaki L. das cultivares Quioto e Rama Forte em carne de frango e verificado o efeito dos mesmos sobre a cor e as características sensoriais das amostras. O extrato hidroetanólico bruto de caqui da cultivar Rama Forte, que apresentou maior teor de compostos fenólicos, maior atividade antioxidante e que não interferiu nas características sensoriais da carne de frango foi empregado nos testes posteriores. O extrato hidroetanólico de caqui da cultivar Rama Forte foi fracionado usando solventes com diferentes polaridades (n-hexano, clorofórmio e acetato de etila) e submetido juntamente com as frações hexânica, clorofórmica, de acetato de etila e a fração residual a determinação de compostos fenólicos totais, atividade antioxidante e antimicrobiana in vitro. Também foi verificada a atividade antioxidante dos mesmos em carne bovina e comparada com a atividade do extrato oleoso de alecrim (Chr. Hansen®). O extrato e/ou frações mais eficientes foram avaliados quanto a atividade antioxidante e antimicrobiana, bem como quanto a influencia sobre as características sensoriais e a cor de produtos cárneos (hambúrguer e salame). Com base nos resultados obtidos pode-se verificar que o extrato hidroetanólico bruto de caqui da cultivar Rama Forte, bem como as frações residual e a de acetato de etila apresentaram atividade antioxidante na carne bovina durante o período de armazenamento, mas não apresentaram atividade antimicrobiana. Nas amostras de salame, o extrato hidroetanólico bruto de caqui da cultivar Rama Forte também apresentou atividade antioxidante e não alterou as características sensoriais nem as contagens microbianas realizadas após o período de processamento do salame. Já no hambúrguer congelado o extrato hidroetanólico bruto de caqui da cultivar Rama Forte e as frações residual e de acetato de etila, quando utilizados nas concentrações de 0,5 e 0,7%, não demonstraram potencial antioxidante e antimicrobiano, e não interferiram nas características sensoriais das amostras, enquanto que a fração acetato de etila (0,5 e 0,7%) contribuiu na retenção e estabilidade da cor vermelha das amostras de hambúrguer de carne bovina durante o período de armazenamento do produto congelado. Desta forma, com base nos resultados obtidos parece adequado supor que o extrato hidroetanólico bruto de caqui da cultivar Rama Forte e as frações residual e de acetato de etila apresentam potencial antioxidante, mas estudos mais aprofundados devem ser realizados principalmente no que se refere a atividade antioxidante em produtos cárneos congelados.

Maîtrise du risque aflatoxique : utilisation d'extraits naturels et mise en évidence de leurs mécanismes d'action / Aflatoxin risk management : use of natural extracts and identification of their mechanisms of action

L’aflatoxine B1 (AFB1) est une mycotoxine contaminant de nombreux substrats agricoles, notamment les céréales, arachides, pistaches, graines de coton et autres fruits secs. Elle est génotoxique, agit comme un initiateur de la cancérogenèse et elle est classée par le CIRC (Centre International de Recherche sur le Cancer) dans le groupe I des molécules carcinogènes pour les hommes et les animaux. Cette toxine est produite par des espèces fongiques appartenant essentiellement au genre Aspergillus et à la section Flavi ; A. flavus étant l’espèce la plus préoccupante. Cette espèce peut se développer à la fois au champ ou lors du stockage des matières premières après récolte. Plusieurs études montrent qu’un grand nombre de denrées alimentaires destinées à l’alimentation humaine et animale sont contaminées par les aflatoxines mettant en évidence la faible efficacité ou l’insuffisance des stratégies actuelles utilisées pour la maîtrise de cette contamination. L’objectif principal de cette étude est le développement d’une stratégie alternative basée sur l’utilisation de produits naturels et visant à réduire la production de la toxine par les moisissures aflatoxinogènes. Grâce à l’élaboration d’un outil moléculaire regroupant un ensemble de gènes impliqués directement ou indirectement dans la biosynthèse de l’AFB1, ce travail permet de mieux comprendre le mécanisme d’action moléculaire de composés anti-aflatoxinogènes. Des extraits aqueux de plantes méditerranéennes ont été testés pour leur efficacité à inhiber l’aflatoxinogénèse. Ainsi, l’extrait aqueux d’hysope, Micomeria graeca, présente des propriétés anti-aflatoxinogènes importantes sans modification de la croissance fongique. Son effet inhibiteur sur plusieurs souches productrices d’aflatoxines ainsi que son mécanisme d’action ont été caractérisés. Ce dernier semble impliquer des gènes pivots dans la régulation des métabolismes fongiques primaire et secondaire ainsi que des voies impliquées dans la réponse cellulaire au stress oxydatif. Ces travaux permettent de valider l’utilisation d’extraits naturels comme stratégie alternative de lutte contre la contamination des aliments par les aflatoxines. / Aflatoxin B1 (AFB1) is a mycotoxin contaminating numerous agricultural commodities such as cereals, peanuts, pistachios, cottonseed and other dried fruits. It is a genotoxic initiating carcinogenesis and is classified by the IARC (International Agency for Research on Cancer) in the group I of molecules that are carcinogenic for humans and animals. This toxin is produced by fungal species mainly belonging to the Flavi section of the Aspergillus genus, A. flavus being the most preoccupying species. This species can develop both in the field and during storage of commodities following harvest. Several studies show that a significant number of food and feedstuffs are contaminated by aflatoxins, highlighting the feeble efficiency or the shortfall of the strategies currently used to prevent such contamination. The main objective of this study is the development of an alternative strategy based on the use of natural products and aiming the reduction of toxin production by aflatoxinogenic fungi. Through the elaboration of a molecular tool grouping a number of genes directly or indirectly linked to AFB1’s biosynthesis, this work allows a better understanding of the molecular mechanism of action of anti-aflatoxinogenic compounds. Aqueous extracts of Mediterranean plants were tested for their ability to inhibit aflatoxinogenesis. Thus, hyssop’s aqueous extract - Micomeria graeca – is shown to present significant anti-aflatoxinogenic properties without reducing fungal growth. Its inhibiting effect in other AFB1-producing species and its mechanism of action were both characterized. The latter seems to involve pivotal genes in fungal primary and secondary metabolisms as well as genes implicated in the fungal cellular response to oxidative stress. These works allocate the use of natural extracts as an alternative strategy to counteract aflatoxin contamination of food and feed.

Le suivi des xénobiotiques dans les extraits naturels destinés aux industries de la cosmétique, des parfums et des arômes alimentaires / Xenobiotics behavior in natural extracts intended for cosmetic, fragrances and flavor industries

Les extraits naturels tels que les huiles essentielles, les absolues ou les résinoïdes utilisés en parfumerie, arômes et cosmétique sont produits à partir de matières premières naturelles. La culture de ces matières premières est souvent basée sur la monoculture et implique l'utilisation de pesticides. La réglementation pour les matières premières destinées à ces industries devient de plus en plus stricte en termes de niveaux résiduels autorisés de pesticides, en raison de leur impact sur la santé publique et sur l'environnement. Dans ce manuscrit, un état de l’art montrant la problématique pour l’analyse des pesticides dans les extraits naturels et les travaux dédiés à cette étude, est présenté. Une méthode d’analyse multi-résidus de pesticides a ensuite été mise au point dans les roses et les extraits (concrètes et absolues) issus de la rose. La préparation d’échantillon s’appuie sur l’approche QuEChERS utilisant l’extraction et la purification par SPE-dispersive. Les échantillons ont ensuite été analysés par GC-MS/MS. Les méthodes ainsi développées ont permis d’étudier les devenir des pesticides au cours du procédé d’extraction des roses. Le travail a été fait dans un premier temps sur des roses issues d’un champ bio puis dopées avec 57 pesticides. Chaque intermédiaire de transformation, excepté les cires, a été analysé mettant en évidence la répartition des pesticides. Il y est montré que les pesticides présents dans les roses sont co-extraits durant le procédé d’extraction et sont concentrés entre 100 et 300 fois dans l’absolue. Cette étude a ensuite été réalisée sur des roses et leurs extraits issus de champs avec des traitements phytosanitaires « classiques ». / Natural extracts in form of essential oils, concretes, resinoids, as well as absolutes used in perfume, aroma, and cosmetic industries are produced from natural raw materials. The cultivation of natural raw materials is often based on monoculture and involves the use of pesticides. Legal regulations for raw materials intended for these industries become increasingly stricter in terms of the allowed residual levels of chemicals used for treatment because of their impact on public health and on the environment. In this manuscript, a review showing pesticide analysis problems in natural extracts and works dedicated to this study, is presented. Next, a method for multi-residue pesticides analysis was developed in roses and extracts (concrete and absolute) from roses. The sample preparation is based on the QuEChERS approach using extraction and purification by SPE-dispersive. The samples were then analyzed by GC-MS / MS. Thus the developed methods were used to study pesticides behavior during the extraction process of the roses. In a first time, the work was done on roses from organic field and spiked with 57 pesticides. Each transformation intermediate, except waxes, was analyzed showing the distribution of pesticides. It showed that pesticides which were present in rose flowers were coextracted during extraction process and concentrated by factor between 100 and 300 in absolute. This study was then performed on roses and extracts from fields with pesticide "classic" phytosanitary treatments. The trend observed for these samples was the same as for the model: pesticides were concentrated in the absolute reaching concentrations of several milligrams per kilogram.

The current approaches to reduce bacterial adhesion to various polymeric surfaces are discussed, as well as how multidisciplinary research on surface design and engineering may have an impact on both fundamental and applied microbiological science and technology.

No / This chapter describes the current approaches to reduce bacterial adhesion to various biomaterial surfaces, focusing on nonfouling surfaces through patterning and hydrophobicity plasma-assisted surface treatment and deposition; incorporation of antimicrobials, antibiotics, antibiofilms, and natural extracts that are either immobilized or released; dual function antimicrobial surfaces; incorporation of nonpathogenic bacteria, bacteriophages, and biofilm dispersal agents but also reduced bacterial adhesion through tissue integration. To facilitate the design of new materials, the role of physical, chemical, and biological surface properties on bacterial adhesion is reviewed in each case, as an insight into the chemical and physical cues that affect bacterial adhesion and biofilm formation can provide ideas for creating successful antifouling or antimicrobial surfaces. The application of these surfaces is explored based on the clinical needs and the market gaps. How multidisciplinary research on surface design and engineering may have an impact on both fundamental understanding of bacterial adhesion to biomaterials and applied biomaterial science and technology is finally discussed.

Le modèle cellulaire THP-1 : adaptation à l'étude de modulateurs de l'activité inflammatoire précoce implicant l'inflammasome

L’inflammation joue un rôle clé dans de nombreuses pathologies, telles que les maladies inflammatoires chroniques, les désordres métaboliques et le cancer. L’un de ses médiateurs le plus puissant est l’interleukine-1β (IL-1β), qui est une cytokine pro-inflammatoire participant à tous les stades de l’inflammation et de l’immunité. Son activation est régulée par un complexe multi-protéique nommé inflammasome, dont la caspase-1 active en découlant est responsable du clivage et de la maturation de l’IL-1β. Huit types d’inflammasomes activant et clivant la pro-caspase-1 ont été identifiés et contiennent tous la protéine ASC (apoptosis-associated speck-like protein containing a CARD). Les inflammasomes partagent un signal intracellulaire commun et le mécanisme menant à leur assemblage et leur activation n’est pas totalement élucidé. L’utilisation de la lignée cellulaire humaine monocytaire, THP-1, différenciée en macrophages grâce à un ester de phorbol, le TPA, a permis la mise en place d’un modèle d’étude de modulateurs de l’inflammasome en conditions stériles. Ce modèle a permis l’étude des mécanismes impliqués suite à des signaux issus de l’inflammation chronique, tels que l’ATP et les espèces réactives de l’oxygène (ROS). Ce travail montre qu’il existe une synergie entre ATP et ROS, qui agissent grâce à une boucle d’activation impliquant probablement plusieurs inflammasomes, dont NLRP3. Des donneurs de NO connus (trinitrine et isosorbide dinitrate) ou nouveau (dérivé de purine) ont montré une activité anti-inflammatoire. D’autres composés ont été identifiés comme de potentiels inhibiteurs d’inflammasome (extraits de dattes et dérivé de purine portant un acide lipoïque) / Inflammation has a pivotal role in several disorders, such as chronic inflammatory diseases, metabolic disorders and cancer. Interleukin-1β (IL-1β) is one of the most powerful mediators in this mechanism. IL-1β is a pro-inflammatory cytokine which is implicated in every step of inflammation and immunity. IL-1β is regulated by a multi-protein complex named inflammasome, in which active caspase-1 is responsible for IL-1β processing and maturation. Eight inflammasomes processing and activating pro-caspase-1 has been identified and all contain the adaptor protein ASC (apoptosis-associated speck-like protein containing a CARD). Inflammasomes share a common intracellular signal and the mechanism for their structural assembly and activation is not totally clear. The human monocytic cell line, THP-1, has been used and cells have been differentiated in macrophages by the phorbol ester, TPA. Based on this, a cellular pattern has been established for studying inflammasome modulators under sterile conditions. This pattern allowed the study of implicated mechanisms in chronic inflammatory signals, such as ATP and reactive oxygen species (ROS). This study shows that ATP and ROS synergize for activating inflammasomes and NLRP3 and act in an autocrine loop. Stimulation with others natural or synthetic compounds has been realized for characterization of their inflammatory activity. Therapeutic NO donors (trinitrin and isosorbide dinitrate) and a new NO donor (purine nitrate ester) have been characterized for their anti-inflammatory activity. Évaluation de l’approche métabolomique pour l’authentification des extraits naturels utilisés dans le secteur arômes et parfums / Metabolomics for the authentication of natural extracts used in flavour and fragrances

Certains extraits naturels sont très rares et onéreux. La tentation est alors forte pour les producteurs de matières premières ou intermédiaires d’avoir recours à des adultérations. Le mélange avec des extraits issus d’origines botaniques apparentées, d’origines géographiques différentes, l’ajout d’un composé synthétique présent dans l’extrait naturel ou d’un autre extrait végétal, l’utilisation de produits phytosanitaires réglementés, la mise en place de procédés d’extractions non standardisés, en sont quelques exemples. Ces différences de qualité sont de plus en plus difficilement décelables. Cette thèse a pour objet de mettre au point une méthodologie rapide, efficace et non ciblée. Une approche métabolomique en UHPLC-HRMS est développée pour identifier ces défauts ou pratiques frauduleuses pour les absolues destinées au secteur arômes et parfums. Cette mise en évidence est réalisée par la détection de métabolites marqueurs. Les absolues présentent un défi particulier : entre 50 et 95 % de l’extrait peut être constitué de composés non volatils, rarement décrits dans la littérature. La recherche d’authenticité de ces extraits est alors plus complexe que dans le cas d’un extrait volatil tel qu’une huile essentielle, dont la composition peut être plus facilement déterminée par des techniques analytiques éprouvées et l’utilisation de bases de données. Deux plantes emblématiques de la parfumerie ont été étudiées : la violette (Viola odorata) et la rose (Rosa damascena et Rosa centifolia). Des marqueurs d’origine géographique ont été identifiés dans le cas de la violette, et des marqueurs d’origine botanique dans le cas de la rose. / Some natural extracts are very scarce and expensive. The temptation is therefore very high for producers or brokers to resort to adulterations. Mixing of extracts from related botanical origins, from different geographical origins, addition of synthetic compounds with natural occurrence in the extract, or addition of another vegetal extract, use of phytosanitary products, non-standardized extraction processes, are some examples. The quality differences are more and more difficult to detect. The objective of this PhD study was to develop a fast, efficient and non-targeted methodology. Metabolomics approach in UHPLC-HRMS was developped to identify defects or fraudulent practices in absolutes used in flavour and fragrances. These identifications are realized by the detection of chemical markers. Absolutes are a great challenge: between 50 and 95 % of the extracts consist of non-volatile compounds; moreover these products are seldom described in literature. The quest for validation for authenticity is then much more complex than in cases of volatile extracts such as essential oils, whose composition can be more easily determined by robust analytical instruments and numerous databases. Two symbolic plants used in perfumery were studied: viola (Viola odorata) and rose (Rosa damascena and Rosa centifolia). Markers of French origin were identified for viola, and markers of R. centifolia were identified for rose. Their characterizations were nevertheless the fundamental limit for this technique being at trace level in the extract. This work demonstrated the performance and limitation of the non-targeted metabolomics approach on absolutes, which are specialties of perfumery.

ASSESSMENT OF THE INFLUENCE OF PROCESSING CONDITIONS ON THE ANTIOXIDANT POTENTIAL OF EXTRACTS OBTAINED FROM OLIVE OIL INDUSTRY BYPRODUCTS

Tesis por compendio / [EN] The main goal of this Thesis was to determine the influence of the main processing stages involved in obtaining natural extracts with high antioxidant potential from byproducts originating in the olive oil industry. Firstly, the effect of freezing and/or the drying methods applied to olive oil byproducts on the polyphenol content and antioxidant capacity of the extracts subsequently obtained was addressed. For this purpose, two byproducts were considered: olive leaves and olive pomace. Secondly, the feasibility of intensifying the extraction of olive leaf polyphenols by means of a new technology, such as power ultrasound, was approached taking both compositional and kinetic issues into account. Thirdly, how the processing conditions (drying and extraction) influence the extract's stability was evaluated. Thus, on the one hand, extracts obtained from olive leaves were subjected to in vitro digestion or dehydrated and stored at different conditions. Finally, the possibility of obtaining a dried vegetable matrix (apple) rich in olive leaf phenolic compounds was explored by addressing the influence of apple pretreatments (blanching and freezing) and drying on the final retention of infused phenolics. The antioxidant potential of extracts and the retention of infused polyphenols in apple were evaluated by means of the total phenolic content and antioxidant capacity analysis, as well as the identification and quantification of the main olive leaf polyphenols by HPLC-DAD/MS-MS. Moreover, in apple samples, the polyphenol oxidase and peroxidase activity and microstructure were also analyzed. The experimental results highlighted that both drying and freezing methods significantly (p<0.05) influenced the concentration of the main polyphenols identified in the olive leaf extracts. Thus, drying at the highest temperature tested was the best processing condition in which to obtain extracts with high antioxidant capacity and phenolic content. Ultrasound application was found to be a relevant, non-thermal way of speeding-up the antioxidant extraction from olive leaves. Thus, by appropriately tuning-up the process variables, the ultrasonic assisted extraction shortened the extraction time from the 24 h needed in conventional extraction to 15 min, without modifying either the extract composition or the antioxidant potential. As far as extract stability is concerned, the processing conditions used for obtaining the olive leaf extracts did not have a meaningful influence on bioaccessibility. Regardless of the method used, stabilizing the extracts by means of dehydration only reduced both the antioxidant capacity and the total phenolic content by around 10 %. Moreover, storage conditions did not show a significant (p<0.05) effect on the antioxidant potential of the extracts for 28 days of storage. A stable dried product (apple), rich in natural phenolic compounds (from olive leaves or tea extracts), was obtained by combining drying-impregnation-drying steps. However, it should be considered that the role of fresh apple drying on the retention of infused olive leaf polyphenols was more important than the further drying of the impregnated apple. In overall terms, olive leaves can be considered a potential source of natural phenolic compounds. Notwithstanding this, the previous drying and freezing steps applied in the raw material processing are decisive factors in the obtaining of natural extracts with high antioxidant potential. Moreover, enhancing the extraction by applying power ultrasound was stated as a non-thermal way of shortening processing times. The stability of olive polyphenols during storage and in vitro digestion was closely related to the individual component considered. Finally, the exploitation of olive leaf extracts as a means of enriching solid foodstuffs requires the use of porous solid matrices free of oxidative enzymes. / [ES] El objetivo principal de esta Tesis fue determinar la influencia de las principales etapas de procesado implicadas en la obtención de extractos naturales con alto potencial antioxidante a partir de los subproductos originados en la industria del aceite de oliva. En primer lugar, se evaluó el efecto de los métodos de congelación y/o secado de la materia prima (hojas y orujo), sobre el contenido polifénolico y la capacidad antioxidante de los extractos. En segundo lugar, se abordó la intensificación de la extracción de polifenoles de hoja de olivo con ultrasonidos de potencia, teniendo en cuenta: composición y la cinética del proceso. A continuación, se estudió cómo las condiciones de procesado (secado y extracción) podían influir en la estabilidad de los extractos. Así, extractos de hojas de olivo fueron sometidos a digestión in vitro o deshidratados y almacenados a distintas condiciones. Por último, se exploró la posibilidad de obtener una matriz vegetal deshidratada (manzana) y rica en compuestos fenólicos de hoja de olivo. Para ello, se evaluó la influencia de los pretratamientos de la manzana (escaldado y congelación) y del secado en la retención final de los polifenoles impregnados. El potencial antioxidante se determinó a través del contenido total en compuestos fenólicos y la capacidad antioxidante y la identificación y cuantificación (HPLC-DAD/MS-MS) de los principales polifenoles. Además, en manzana, se midió la actividad enzimática de la polifenol oxidasa y peroxidasa y se analizó la microestructura. Los resultados manifestaron que el método de secado y el de congelación influyeron significativamente (p<0.05) en la concentración de los principales polifenoles en los extractos. Así, el secado a mayor temperatura resultó ser el mejor tratamiento para obtener extractos con alta capacidad antioxidante y alto contenido fenólico. La aplicación de ultrasonidos resultó ser una alternativa no térmica muy interesante para acelerar la extracción de antioxidantes de hojas de olivo. Con la combinación adecuada de las variables del proceso, la aplicación de ultrasonidos redujo el tiempo de extracción de 24 h necesarias en extracción convencional a 15 min, sin modificar la composición de los extractos y su potencial antioxidante. En cuanto a la estabilidad del extracto, las condiciones de procesado no tuvieron una influencia significativa en la bioaccesibilidad de los extractos. Independientemente del método utilizado, la estabilización de extractos por deshidratación sólo redujo la capacidad antioxidante y el contenido total en compuestos fenólicos en torno a un 10 %. Además, las condiciones de almacenamiento no mostraron ningún efecto significativo (p<0.05) sobre el potencial antioxidante durante los 28 días de almacenamiento. Combinando secado-impregnación-secado, fue posible desarrollar un producto deshidratado (manzana), estable y rico en compuestos fenólicos naturales (de hojas de olivo o extractos de té). No obstante, cabe destacar que el secado de la manzana fresca jugó un papel más importante en la retención de los polifenoles de hoja de olivo infundidos que el secado final de la manzana impregnada. En términos generales, las hojas de olivo pueden considerarse como una fuente potencial de compuestos fenólicos naturales. No obstante, el secado y la congelación durante el procesado de la materia prima son factores decisivos para la obtención de extractos naturales con alto potencial antioxidante. Además, la aplicación de ultrasonidos de potencia durante la extracción puede resultar una alternativa no térmica muy interesante de cara a acortar el tiempo de procesado. La estabilidad de los polifenoles de la hoja de olivo, durante el almacenamiento y la digestión in vitro, dependió claramente del compuesto individual considerado. Finalment, es va explorar la obtenció d'una matriu vegetal deshidratada (poma) i rica en compostos fenòlics de fulla d'olivera considerant la influència del pretractament de la poma (escaldament i congelació) i de l'assecatge sobre la retenció final dels fenòlics introduïts en la poma. El potencial antioxidant es va avaluar determinant el contingut fenòlic total i la capacitat antioxidant, així com identificant i quantificant els principals polifenols (HPLC-DAD/MS-MS). A més, en poma l'activitat enzimàtica de la polifenoloxidasa i la peroxidasa i la microestructura. Els resultats experimentals van destacar que el mètode d'assecatge i el de congelació van influir significativament (p<0,05) en la concentració dels principals polifenols identificats en els extractes. L'assecatge a la temperatura més alta que es va provar va resultar la millor condició de processament per a obtenir extractes amb una alta capacitat antioxidant i un alt contingut fenòlic. L'aplicació d'ultrasons va ser una manera rellevant i no tèrmica d'accelerar l'extracció d'antioxidants de les fulles d'olivera. Així, amb la combinació adequada de les variables del procés, l'extracció assistida per ultrasons va escurçar el temps d'extracció, de les 24 h requerides en l'extracció convencional a 15 min, sense modificar la composició de l'extracte ni el potencial antioxidant. Quant a l'estabilitat de l'extracte, les condicions de processament utilitzades per a l'obtenció dels extractes de fulla d'olivera no van tenir una influència significativa en la bioaccessibilitat. Independentment del mètode utilitzat, l'estabilització dels extractes per mitjà de la deshidratació només va reduir la capacitat antioxidant i el contingut fenòlic total al voltant d'un 10 %. A més, les condicions d'emmagatzematge (temperatura i forma de l'extracte: líquid o pols) no van mostrar cap efecte significatiu (p<0,05) en el potencial antioxidant dels extractes durant els 28 dies d'emmagatzematge. Combinant etapes d'assecatge-impregnació-assecatge fou possible obtenir un producte assecat estable (poma) i ric en compostos fenòlics naturals (de fulles d'olivera o te). No obstant això, cal destacar que l'assecatge de la poma fresca va ser més important i determinant en la retenció dels polifenols de fulla d'olivera que no l'assecatge de la poma impregnada. En termes generals, les fulles d'olivera es poden considerar com una font potencial de compostos fenòlics naturals. No obstant això, l'aplicació d'assecatge i congelació durant el processament de la matèria primera són factors decisius per a l'obtenció d'extractes naturals amb un alt potencial antioxidant. A més, l'aplicació d'ultrasons de potència durant l'extracció resultà ser una forma no tèrmica de millorar el procés, tot reduint-ne el temps d'extracció. L'estabilitat dels polifenols d'olivera durant l'emmagatzematge i la digestió in vitro va dependre del compost individual considerat.
