The influence of genetic polymorphisms of fibrinogen genes on changes in total fibrinogen and fibrinogen gamma prime concentrations over time in black South Africans / Ané Jobse

Jobse, Ané January 2014 (has links)
INTRODUCTION AND AIM - Cardiovascular disease is globally a major risk factor for morbidity and mortality. It is caused by various factors, one of which is an abnormal haemostatic process. Fibrinogen is a haemostatic factor that is considered to be an independent risk factor for cardiovascular disease. Elevated fibrinogen can be caused by environmental and genetic factors which increase the risk of the occurrence of thrombosis. The fibrinogen y' chain, which is one of the three chains of fibrinogen, has two different variants, the yA and y’. The presence of the fibrinogen y’ chain has been associated with thrombotic disorders. Many studies have investigated the fibrinogen variables in Caucasian individuals, but only a few such studies have been conducted on non-Caucasian individuals. The genetic diversity of ethnic groups differs and could cause differences in the fibrinogen variables between these groups. Fibrinogen is known to increase with age; therefore to explain changes over time in fibrinogen concentrations it was also important to investigate whether genetic determinants and possible gene–environment interactions influenced fibrinogen over time. In this study the main aim was to determine the change in the fibrinogen variables over a five-year period within a black South African cohort subdivided according to genotypes associated with fibrinogen variables, and to determine whether the observed changes were modulated by environmental factors. PARTICIPANTS AND METHODS - Data [baseline (n=2010) and follow-up (n=1288)] were collected in the Prospective Urban and Rural Epidemiology (PURE) study during 2005 and 2010 from apparently healthy black men and women aged between 35 and 65 years and residing in rural or urban settlements. Experimental methods included analysis of fibrinogen and fibrinogen y’ concentrations, single nucleotide polymorphisms (SNPs) and determination of environmental factors associated with the fibrinogen variables. RESULTS - The fibrinogen variables increased significantly from 2005 to 2010 in both the rural and urban participants, as well as in both men and women. The major environmental factors that affected the fibrinogen variables were C-reactive protein (CRP), interleukin-6 (IL-6), body mass index (BMI), glycated haemoglobin (HbA1c), age, blood lipids, human immunodeficiency virus (HIV) and tobacco use. Fibrinogen increased consistently from 2005 to 2010 in the respective genotypes of all SNPs analysed, except in the FGG 9340 T>C homozygous mutant carriers. Fibrinogen y’ also increased in general in most genotypes from 2005 to 2010, except in the FGG 10034 C>T mutant allele carriers, where a decrease was observed. It was determined that CRP was the only environmental factor that influenced the change in fibrinogen over time and that FGG 10034 C>T was the only SNP that influenced the change in fibrinogen y’ over the five years. Four gene–environment interactions also influenced fibrinogen on a cross-sectional level, i.e. FGA 2224 G>A with age, FGB Arg448Lys with HIV status, FGB 1643 C>T with urbanisation and FGB 1038 G>A with HbA1c. Only the FGG 9340 T>C with HbA1c interaction was found to predict change in fibrinogen concentrations over the five years. CONCLUSION - Both environmental and genetic factors significantly influenced the fibrinogen variables cross-sectionally as well as prospectively. It was clear that the influence of the environmental factors was mediated by genetic polymorphisms and vice versa, as can be seen by the gene–environment interactions found in this study. An important finding of this study was that the interaction of HbA1c with two SNPs on fibrinogen variables may explain the known inconsistent relationship found between fibrinogen concentrations and diabetes. / MSc (Dietetics), North-West University, Potchefstroom Campus, 2014

Kousis, Georgios, Reinholdz, Erik January 2019 (has links)
The purpose of this thesis is to clarify what a football agent does in Sweden and describe how legitimacy is created in an industry that is associated with crime and a lack of transparency. Qualitative interviews have been used to answer the two main issues that follows: What do football agents do in Sweden and what services do they offer football players and football clubs? And how do football agents construct legitimacy in an industry with a bad reputation? The respondents were picked from three different professions: three footballers, three football agents and three sports directors. The result shows that football agents in Sweden mainly worked with contracts negotiation, professional contact creation, scouting and giving career advice. Some agents even offered their players the opportunity for extra training sessions, match analysis and rehabilitation. The result also shows that some agents in Sweden uses work methods that could be considered unethical, for example giving bribes to players. When it came to how agents constructed legitimacy, it was mainly about creating a trust between themselves and other actors in Swedish football. This was done mostly through gaining external approval from other actors in football. In addition, it was about following the regulations that exist and staying away from anything that can be associated with the shadier side of the football agent industry.

Longevidade de restaurações em dentes decíduos submetidos à remoção parcial de tecido cariado e fatores associados ao insucesso : estudo retrospectivo / Longevity of primary teeth restorations undergoing partial caries removal and factors associated with failure : retrospective study approaching daily clinical life

Concha Melgar, Ximena January 2015 (has links)
Objective: to assess the longevity of direct adhesive restorations undergoing partial caries removal (PCR) in primary teeth and associated factors for failure, approaching the results to a daily clinical life. Methods: Retrospective analytical observational study which included 284 posterior and anterior primary teeth direct adhesive restorations that had been treated with PCR of 88 children with high caries experience. The teeth had clinical and radiographic follow-up through the information registered in the clinical records of the patients. In addition, factors potentially associated with treatment failure were investigated, such as age, gender, type of tooth involved, number of surfaces restored, type of capping material and restorative material used, visible plaque (VPI) and gingival bleeding indexes (GBI). Kaplan-Meier estimate and Log-Rank test were used to analyze the longevity of restorations and multivariate Cox´s regression model with shared frailty to evaluate the factors that could be related to treatment failure (p<0.05). Results: A clinical and radiographic success rate of the sample was 76% and an annual failure rate was estimated in 20% over 36 months. Only visible plaque index (p=0.014) and type of teeth, anterior or posterior (p<0.001) had a significant influence on the restorative failure. Conclusion:Adhesive restorations undergoing PCR in primary teeth in an environment that simulates a daily clinical life demonstrate good results, particularly in posterior teeth. However, the biofilm accumulation on the dental surfaces affects the longevity of them.

La décision conditionnelle en droit administratif français / The conditional decision in French Administrative law

Gallo, Carole 15 November 2017 (has links)
Wrongfully considered as the exclusive province of the specialist on unilateral administrative acts, conditional decisions have received the attention of public lawyers only in so far as they come to present a risk for the protection of aquired rights. This goes some way to explaining both the scarcity of doctrinal contributions to a definition of the concept of a conditional decision – autonomous from the paradigm of the obligations in French civil law –, and the difficulties encountered in trying to bring some clarity to the subject by attempting to determine precisely the boundaries of the notion. Nevertheless, recent turns taken by the administrative courts, and more recently by codifiers, do call for a clear and consistent definition. The case law has for long considered these legal acts as unilateral acts, thus subjecting conditional decisions to the corresponding legal principles. Despite this, the legal scholarship on the question has never managed to fully integrate conditional decisions in either branch of the main binary subdivision – contracts and unilateral acts –, ill-using both in order to stress the highly unusual character of the notion (for instance underlining the peculiar relevance of an analysis of this unilateral decision in contractual terms). This unusual object therefore forces us to reevaluate our traditional classifications, which otherwise systematically result in a severe inconsistency between substantive law and its doctrinal exposition. In order to do so, the theoretical tools and approaches used up to now must be renewed. This doctoral dissertation aims to reconcile the positive law and its doctrinal exposition, thus proposing a clear definition of the condition itself as a legal norm in its own right – both dependant on, and distinct from, the other provisions of the decision it is attached to – the function of which is to modulate the effects over time of the main decision. These proposed elements of definition come to reveal a unified and coherent legal status of the conditional decision in administrative law.

Ekvivalence měření nástroje SQWLI v čase / Equivalence of measurement SQWLI over time

Šeflová, Kristýna January 2016 (has links)
(in English): This paper is devoted to indicator of subjective quality of working life (SQWLI) equivalence of measurement and testing of this instrument over time. The theoretical part introduces the concept of equivalence, from its inception, through the definition, types, methods used to test different kinds of equivalence and programs that can be used for this purpose. Furthermore the tested data files from 2009, 2011, 2013 to 2014 are presented. The analytical part contains a description of confirmatory factor analysis for multiple groups which is the most common method for testing equivalence of measurement. Presented are the results of several testing methods - analysis of reliability, exploratory factor analysis and the confirmatory factor analysis for multiple groups. According to the results it is shown how it is possible to approach comparisons over time and finally the development of SQWLI and it's individual changes in terms of the functionality of the whole tool is discussed. 2
