Centro de Pesquisas Gonçalo Moniz. Salvador, Bahia, Brasil / As fisalinas são seco-esteróides isolados de espécies do gênero Physalis que apresentam diversas atividades biológicas. Em estudos in vitro e em modelos de doenças mediadas pelo sistema imune, foi demonstrado que a fisalina F possui potente atividade imunossupressora, independente da ativação do receptor de glicocorticóide. Neste trabalho foi avaliado se a ação deste esteróide está relacionada à ativação do receptor de vitamina D (VDR), que é um fator de transcrição ativado pela vitamina D3. Inicialmente foi realizado um ensaio de translocação do VDR utilizando células COS-7 transfectadas com plasmídeo codificante para a proteína de fusão YFP-VDR, formada pelo VDR associado à proteína fluorescente amarela. As células transfectadas foram tratadas com 100 ηM da fisalina F e também da fisalina D, que não possui atividade imunossupressora, dexametasona e vitamina D3. Observou-se por microscopia de fluorescência o mesmo padrão de translocação do VDR do citoplasma para o núcleo em células tratadas com a fisalina F e com a vitamina D3, mas não com dexametasona ou fisalina D. Para comprovar a atividade do VDR como fator de transcrição sob ação da fisalina F, células COS-7 foram transfectadas com pTA-LUC (plasmídeo controle) ou pVDR-LUC que codifica a proteína luciferase sob o controle de elementos responsivos ao VDR. Após a transfecção, as células foram incubadas com os mesmos estímulos e na mesma concentração do ensaio de translocação ou apenas com veículo. As atividades da luciferase nos grupos tratados com fisalina F e vitamina D3 foram semelhantes e também superiores às do grupo tratado com veículo, fisalina D e dexametasona. Foi realizada a expressão do VDR em Escherichia coli BL21 (DE3) Rosetta utilizando o plasmídeo HS_VDR_EC1-pQE-T7, o qual codifica o VDR associado a uma cauda de 6 histidinas. Nas condições testadas, a proteína expressada encontrava-se em corpos de inclusão. Esta proteína purificada poderá ser utilizada em futuros estudos de interação entre VDR e fisalinas. Em conclusão, nossos resultados sugerem que a fisalina F interage com o VDR e que as atividades destas fisalinas podem ser atribuídas, ao menos em parte, à ativação deste receptor nuclear. / Physalins are secosteroids isolated from species of the genus Physalis endowed with several biological activities. Studies in vitro and in models of immune-mediated diseases have shown that physalin F has potent immunosuppressive activity independent of the glucocorticoid receptor. in this study we evaluated whether the action of this steroid is related to activation of the vitamin D receptor (VDR), which is a transcription factor activated by vitamin D3. Initially we evaluated the translocation of the VDR using COS-7 cells transfected with plasmid encoding a fusion protein YFP-VDR formed by VDR associated with yellow fluorescent protein. The transfected cells were treated with 100 ηM of physalins F and D, dexamethasone, and vitamin D3. We observed by fluorescence microscopy the translocation of VDR from the cytoplasm to the nucleus in cells treated with physalin F and with vitamin D3, but not with dexamethasone or physalin D. To prove the transcription factor activity of VDR under the action of physalin F, COS-7 cells were transfected with PTA-LUC (plasmid control) or pVDR-LUC that encodes the protein luciferase under de control of VDR responsive elements. After transfection, cells were incubated with vehicle or physalins F and D, dexamethasone and vitamin D3 at the same concentration of translocation test. The luciferase activity of cells treated with vitamin D3 and physalin F were similar, and higher than group treated with vehicle, dexamethasone or physalin D. The VDR expression was done in Escherichia coli BL21 (DE3) Rosetta. For this, we used the plasmid PQE-HS_VDR_EC1-T7, which encodes the VDR protein associated with a tail of 6 histidines. Under the conditions tested, the expressed protein was in inclusion bodies. This purified protein can be used in future studies of interaction between VDR and physalins. Our results suggest that physalin F interacts with the VDR and the activities of this physalin can be attributed at least in part to activation of this nuclear receptor.

In this sense, the work was divided into two chapters, where the objectives were: i) to evaluate the genetic variation in populations of physalis about the growth of reproductive structures measured over time; ii) evaluate the genetic variability within and among populations of physalis for selecting promising populations. The experiment was conducted at the State University of Santa Catarina (UDESC) on the experimental field of the Institute of Molecular Genetics and Breeding (IMEGEM). Populations originate from different regions, being Fraiburgo, Caçador, Vacaria, Lages, Colômbia and Peru. The field experiment was conducted in a randomized block design with two replications. The characters were the number of buds, number of flowers, number of fruits, fruit weight, mass capsule, thousand seed mass, diameter of fruits and fruit polar diameter. Data were analyzed considering information relating to the data with repeated measures in time to the first chapter and considering information regarding the error between and within populations in the second chapter. The different behavior regarding reproductive growth among populations suggests that assessments should be carried out over time. Unable to select populations for early emergence in the reproductive structures. The population of Fraiburgo showed higher emergence of fruit. There is variability among populations of physalis (p<0.05) for the characters of fruits evaluated. There is no variation in the experimental unit. It is recommended the hybridization of population Vacaria with populations Caçador, Lages and Peru, to obtain segregating populations of Physalis / Considerando ser uma alternativa de produção com potencial valor econômico e devido ao conhecimento incipiente da variação genética nas populações cultivadas, é necessário iniciar um estudo para o melhoramento genético da espécie, de forma que o conhecimento da variabilidade poderá possibilitar a seleção de populações produtivas e adaptadas às condições locais de cultivo. Nesse sentido, o trabalho foi dividido em dois capítulos, onde os objetivos foram: i) avaliar a variação genética em populações de physalis quanto ao crescimento de estruturas reprodutivas medidas ao longo do tempo; ii) caracterizar a variabilidade genética em populações de physalis e selecionar genitores para utilização em programas de melhoramento genético. O experimento foi realizado na Universidade do Estado de Santa Catarina (UDESC), no campo experimental do Instituto de Melhoramento e Genética Molecular (IMEGEM). As populações são originárias de diferentes regiões, sendo Fraiburgo, Caçador, Vacaria, Lages, Colômbia e Peru. O experimento foi conduzido a campo, em delineamento de blocos casualizados, com duas repetições. Os caracteres avaliados foram número de botões florais, número de flores, número de frutos, massa de frutos, massa de cápsula, massa de mil sementes, diâmetro equatorial de frutos e diâmetro polar de frutos. Os dados foram analisados considerando informações referentes a dados com medidas repetidas no tempo para o primeiro capítulo e considerando informações referentes ao erro entre e dentro para a seleção de populações no segundo capítulo. O comportamento diferenciado quanto ao crescimento reprodutivo entre as populações sugere que as avaliações devem ser realizadas ao longo do tempo. Não foi possível selecionar populações quanto a precocidade na emergência das estruturas reprodutivas. A população de Fraiburgo apresentou maior emergência de frutos no último período de avaliação. Existe variabilidade entre as populações de physalis no Planalto Serrano Catarinense (p<0,05), para os caracteres de frutos. Não existe variação dentro da unidade experimental. Recomenda-se o cruzamento da população de Vacaria com as populações de Caçador, de Lages e do Peru, para a obtenção de populações segregantes de Physalis

Efeito da fisalina e, isolada da Physalis angulata, em modelos de dermatite de contato induzida por tpa e oxazolona em camundongos / The effects of physalin e, isolated from Physalis angulata, on tpa and oxazolone induced contact dermatitis in mice

CoordenaÃÃo de AperfeiÃoamento de Pessoal de NÃvel Superior / A Fisalina E (FIS E), isolada das partes aÃreas da Physalis angulata (Solanaceae), foi avaliada em modelos de dermatite de contato induzida por 12-O-tetradecanoilforbol-13-acetato (TPA) e por oxazolona (OXA) em camundongos. A dermatite de contato irritativa aguda foi induzida pela administraÃÃo tÃpica de TPA (2,5&#956;g/orelha) e avaliada apÃs 4h. A FIS E(0,125;0,25 e 0,5 mg/orelha,por via tÃpica) reduziu de forma significativa o edema de orelha induzido por TPA em 33,1; 38,7; 39%, respectivamente, e dexametasona(0,05 mg/orelha,por via tÃpica) reduziu em 95,2%. Nas mesmas doses, a FIS E assim como a Dexa, reduziram em 45,7; 52,5; 67,6 e 83,4 %, respectivamente, a atividade da mieloperoxidase (MPO) tecidual. FIS E (3, 10 e 30 mg/kg,por via oral) e Dexa (1mg/kg,por via oral) reduziram o edema de orelha em 34,8; 40,7, 47,3 e 46,6% respectivamente. Nas mesmas doses, a FIS E assim como a Dexa, reduziram em 39,3; 46,7 e 54,3 e 81,7 %, respectivamente, a atividade da mieloperoxidase (MPO) tecidual. A FIS E (0.5 mg/orelha) foi capaz de reduzir os nÃveis teciduais de TNF-&#945; de 8,71&#61617;1.18 pg/ml (veÃculo) para 2,95&#61617;0.81 pg/ml e a dexametasona (0,05 mg/orelha) para 2,35&#61617;0.94 pg/ml, representando uma reduÃÃo de 66,2 e de 73 %, respectivamente. A administraÃÃo de Mifepristone (25 mg/kg,s.c), antagonista esteroidal, reverteu os efeitos da FIS E e da Dexa, por via oral e tÃpica, sobre a reduÃÃo do edema, atividade da mieloperoxidase e nÃveis de TNF-&#945;. Na marcaÃÃo imunohistoquÃmica para TNF-&#945; e NF-&#954;B, a FIS E (0,5 mg/orelha) e a dexametasona (0,05 mg/orelha) reduziram a intensidade de marcaÃÃo do TNF-&#945; e NF-&#954;B, e o Mifepristone, reverteu esse efeito. A aÃÃo antiinflamatÃria da FIS E no modelo da dermatite aguda induzida por TPA foi confirmada pelos achados histopatolÃgicos, com a reduÃÃo do edema, infiltrado inflamatÃrio de mono e polimorfonucleares. Na dermatite crÃnica foi induzida pela aplicaÃÃo tÃpica de 20 Âl de uma soluÃÃo de TPA (2,5Âg/orelha) em dias alternados durante 10 dias, a FIS E, por via tÃpica, nas doses 0,125; 0,25 e 0,5 mg/orelha, reduziu, de forma significativa, o edema de orelha, em 19, 23 e 30,3%, respectivamente, enquanto a Dexa (0,05mg/orelha) reduziu em 32,6 %. FIS E (0,125;0,25 e 0,5 mg/orelha,por via tÃpica) e a Dexa(0,05mg/orelha) reduziram a atividade tecidual da MPO em 30; 38,8; 50,8 e 56,6%, respectivamente. A administraÃÃo oral da FIS E (3, 10 e 30 mg/kg), reduziu, de forma significativa, o edema de orelha crÃnico, em 8; 14,8; 17 %, respectivamente, e a Dexa (1 mg/kg) reduziu em 20,7 %.Nas mesmas doses, a FIS E e a Dexa reduziram a atividade da MPO em 43,1; 54,5 ;59,3 e 65 %,respectivamente. O modelo de dermatite de contato induzida por oxazolona (OXA, 1%/orelha), provocou uma hiperplasia epidÃrmica onde o INF-&#947; teve papel crucial. A FIS E, por via oral e tÃpica, reduziu de forma significativa, a hiperplasia epidÃrmica do 7Â ao 19Â dia de observaÃÃo. A FIS E (0.5 mg/orelha, por via tÃpica) diminuiu os nÃveis de IFN-&#947; em 43,5% e a Dexa (0.05 mg/orelha, via tÃpica) em 37,5%.Os resultados encontrados, comprovados pelo estudo histolÃgico, demonstram o efeito antiinflamatÃrio da Fisalina no modelo de dermatite de contato alÃrgica crÃnica. ConcluÃmos que a Fisalina E demonstrou atividade antiinflamatÃria, por via tÃpica e oral, nos modelos de dermatite de contato induzida por TPA e oxazolona em camundongos, possivelmente atravÃs da interaÃÃo com receptores de glicocorticÃides. / The physalin E (PHY E), isolated from the aerial parts of Physalis angulata (Solanaceae), was assessed in models of contact dermatitis induced by 13-acetate-12-o-tetradecanoyl-phorbol (TPA) and oxazolone (OXA) in mice. The irritant contact dermatitis was induced by acute topical administration of TPA (2,5 mg/ear) and evaluated after 4h. The PHY E topically applied to ear at 0,125;0,25 and 0,5 mg/ear and dexamethasone (Dexa), topically, a dose of 0,05 mg/ear, reduced significantly the edema ear induced by TPA in 33,1; 38,7; 39 and 95,2%, respectively. In the same doses, PHY E as well as Dexa, reduced by 45,7; 52,5; 67,6 and 83,4%, respectively, the activity of myeloperoxidase (MPO) in tissue. Oral administration of PHY E (3,10 and 30 mg/kg) and DEXA (1 mg/kg) decreased the ear edema by 34,8; 40,7;47,3 and 46,6% respectively. The same doses, PHY E as well as Dexa, reduced by 39,3; 46,7;54,3% and 81,7%, respectively, the activity of myeloperoxidase (MPO) tissue. The PHY E (0,5 mg/ear) was able to reduce the tissue levels of TNF-&#945; of 8,71&#61617;1.18 pg/ml (vehicle) to 2,95&#61617;0.81 pg/ml and dexamethasone (0,05 mg/ear) to 2,35&#61617;0.94 pg/ml, showing a reduction of 66.2 and 73%, respectively.The administration of mifepristone (25 mg/kg,sc),a steroid antagonist, reversed the effects of PHY E and Dexa, orally and topically, on reducing the edema,myeloperoxidase activity and levels of TNF-&#945;. In the immunohistochemical analysis for TNF-&#945; and NF-&#954;B, PHY E (0,5 mg/ear) and dexamethasone (0,05 mg/ear) reduced the intensity of marking the TNF-&#945; and NF-&#954;B, and mifepristone reversed this effect. The anti-inflammatory action of the PHY E in the model of the dermatitis TPA-induced were confirmed by histopathological findings, with the reduction of edema and inflammatory infiltration of mono and polymorphonuclear. The chronic dermatitis was induced by application of 20 Âl solution of TPA (2,5 mg/ear) was applied topically every other day for 10 days, PHY E topically applied (0,125, 0,25 and 0,5 mg/ear) reduced significantly the edema of the ear, 19; 23; 30,3%,and Dexa (0,05 mg/ear) decreased by 32,6%.In the same doses, PHY E and Dexa reduced tissue activity of MPO in 30; 38,8;50,8 and 56,6%, respectively. Oral administration. PHY E (3, 10 and 30 mg/kg) reduced significantly the ear edema chronic in 8, 14,8 and 17%, respectively, and Dexa (1 mg/kg) reduced in 20.7%. In the same doses,PHY E and Dexa reduced activity of MPO in 43.1, 54.5, 59.3 and 65% respectively The model of contact dermatitis induced by oxazolone (OXA, 1%/ear) caused an epidermal hyperplasia where the INF-&#947; played crucial role. PHY E oral and topical, has significantly reduced the epidermal hyperplasia of 7 th to 19 th day of observation Physalin E (0,5 mg/ear topically) decreased levels of IFN-&#947; in 43,5% and Dexa (0,05 mg/ear topically) to 37,5% when compared to vehicle. Our results, as evidenced by histological study, demonstrate the anti-inflammatory effect of physalin in the model of contact dermatitis allergic. In conclusion, the results obtained showed that PHY E presented anti-inflammatory activity either when was used by oral and topical route in the models of contact dermatitis TPA and Oxazolone-induced in mice, possible trought the interaction with glucocorticoids receptors.

They were evaluated six populations goldenberries under three spacing between rows in six periods of harvest. A block design at random in subdivided plots with three replications. The experimental setup consists factorial 3 x 6 x 6 for yield components and 3 x 6 for the physicochemical characteristics. To yield components were evaluated: total yield (t ha-1), income accumulated fruits (kg plant-1), backlog of fruits (no plant-1), fruit weight (g plant-1), diameter Polar (mm) and equatorial (mm), and cracked fruit percentage (%). For physical and chemical characteristics were evaluated: total soluble solids (° Brix), titratable acidity (% citric ac.), Maturation index, firmness (N) and fruit color index. Statistical analysis of data was performed by adjusting matrix of variance and covariance residual by-AIC Akaike information for the items of income and both studies were submitted to analysis of variance (Anova) for description of the results, and when significant, for qualitative variables we used the comparison test of Tukey (p=0.05), and the quantitative variables we used linear and nonlinear regression. The spacing influenced mass, total soluble solids, and the color index of the fruit evaluated goldenberries populations. Populations showed variability for equatorial diameter and titratable acidity, which are important characters for determining the shape and flavor of the fruit. Yield components showed the variation over periods of crops / Considerando o potencial da cultura do fisális em Santa Catarina incorporada ao cultivo das pequenas frutas, o interesse de consumidores e produtores e a necessidade de maiores estudos para o desenvolvimento da cultura, o presente trabalho teve por objetivo caracterizar os componentes de rendimento e avaliar as características físico-químicas de frutos de populações de fisális, submetidos a diferentes espaçamentos entre filas. Foram avaliadas seis populações de fisális, sob três espaçamentos entre filas em seis períodos de colheita. Foi utilizado um delineamento em blocos ao acaso em parcelas subdividas com três repetições. O arranjo experimental compõe esquema fatorial 3 x 6 x 6 para os componentes de rendimento e 3 x 6 para as características físico-químicas. Para os componentes de rendimento foram avaliados: rendimento total (t ha-1), rendimento de frutos acumulados (kg planta-1), número de frutos acumulados (nº planta-1), massa de fruto (g planta-1), diâmetro polar (mm) e equatorial (mm), e percentual de fruto rachados (%). Para as características físico-químicas foram avaliados: sólidos solúveis totais (º Brix), acidez total titulável (% ác. cítrico), índice de maturação, firmeza (N) e índice de cor dos frutos. A análise estatística dos dados foi realizada pelo ajuste de matrizes de variância e covariância residuais pelo critério de informação de Akaike-AIC para os componentes de rendimento e ambos os estudos foram submetidos à análise de variância univariada (Anova) para descrição dos resultados, e quando significativos, para as variáveis qualitativas utilizou-se o teste de comparação de médias de Tukey (p=0,05), e para as variáveis quantitativas utilizou-se regressão linear e não linear. O espaçamento influenciou a massa, os sólidos solúveis totais e o índice de cor dos frutos das populações de fisális avaliadas. As populações apresentaram variabilidade para diâmetro equatorial e acidez titulável total, sendo estes importantes carácteres para determinação da forma e do sabor dos frutos. X-ray crystallography is one of the most powerful tools for the elucidation of the structure of biological macromolecules such as proteins and viruses. Crystallographic techniques are extensively used for investigations on protein structure, ligand-binding, mechanisms of enzyme catalyzed reactions, protein-protein interactions, role of metal ions in protein structure and function, structure of multi-enzyme complexes and viruses, protein dynamics and for a myriad other problems in structural biology. Crystallographic studies are essential for understanding the intricate details of the mechanism of action of enzymes at molecular level. Understanding the subtle differences between the pathogenic enzymes and host enzymes is necessary for the design of inhibitor molecules that specifically inhibit parasite enzymes. The current thesis deals with the application of biochemical and crystallographic techniques for understanding the structure and function of proteins from two pathogenic organisms – a plant virus Physalis Mottle Virus (PhMV), and a pathogenic bacterium, Salmonella typhimurium and also stress induced proteins from Oryza sativa. The thesis has been divided into seven chapters, with the first four chapters describing the work carried out on PhMV, while the rest of the chapters deal with the studies on stress response proteins from Oryza sativa and Salmonella typhimurium. The first part of the thesis deals with studies on viral capsids. Viruses are obligate parasites that have proteinaceous capsids enclosing the genetic material, which, in the case of small plant viruses, is invariably ss-RNA. X-ray diffraction studies on single crystals of viruses enable visualization of the structures of intact virus particles at near-atomic resolution. These studies provide detailed information regarding the coat protein folding, molecular interactions between protein subunits, flexibility of the N-and C-terminal segments and their probable importance in viral assembly, role of RNA in capsid assembly, nucleic acid (RNA)-protein interactions, the capsid structure and mechanism of assembly and disassembly. The present thesis deals with the capsid structure and analysis of the coat protein (CP) recombinant mutants of PhMV. Virus assembly, one of the important steps in the life cycle of a virus, involves specific interactions between the structural protein and cognate viral genome. This is a complex process that requires precise protein-protein and protein nucleic acid interactions. In fact, most of the biological functional units such as ribosomes and proteosomes also require highly co-ordinated macromolecular interactions for their functional expression. Viruses being simple in their architecture, serve as excellent model systems to understand mechanism of macromolecular assembly and provide necessary information for the development of antiviral therapeutics, especially in animal viruses. PhMV is a plant virus infecting several members of Solanaceae family. It belongs to the tymoviridae group of single stranded RNA viruses. Its genome is encapsidated in a shell comprising of 180 (architecture based on T = 3 icosahedral lattice) chemically identical coat protein (CP) subunits (~ 20,000Da) arranged with icosahedral symmetry. In an earlier phase of work, PhMV purified from infected plant leaves was crystallized in the space group R3 (a = 294.56 Å,  = 59.86). X-ray diffraction data to 3.8 Å resolution were recorded on films by screenless oscillation photography. Using this data of severely limited quality and poor completion (40%), the structure PhMV was determined by molecular replacement using the related turnip yellow mosaic virus (TYMV) structure as the phasing model. There was therefore a need to re-determine and improve the structure, which could be useful for understanding the earlier detailed studies on its biophysical properties. As a continuation of these studies, the present investigations were conceived with the goal of determining the natural top and bottom component capsid structures of PhMV. Investigations were also carried out to examine the possibility of enhancing the diffraction quality of PhMV crystals. The thesis begins with a review of the current literature on the available crystal structures of viruses and their implications for capsid assembly (chapter I). All experimental and computational methods used during the course of investigations are described in chapter II, as most of these are applicable to all the structure determinations and analyses. The experimental procedures described include cloning, overexpression, purification, crystallization and intensity data collection. Computational methods covered include details of various programs used during data processing, structure solution, refinement, model building, validation and analysis. Chapter III describes structural studies on top and bottom components of PhMV. Purified tymoviruses including PhMV are found to contain two classes of particles that sediment at different velocities through sucrose gradients and are called the top (sedimentation coefficient 54 Svedberg units(S)) and the bottom (115S) components. The top component particles are either devoid of RNA or contain only a small subgenomic RNA (5%) while the bottom component particles contain the full length genomic RNA. Only the bottom component is infectious. The top and bottom components were separately crystallized in P1 and R3 space groups, respectively. It is of interest to note that crystals of the bottom component obtained earlier belonged to R3 space group while recombinant capsids that lack of full length RNA as in natural top component crystallized in the P1 space group. A polyalanine model of the homologous TYMV was used as the phasing model to determine the structures of these particles by molecular replacement using the program AMoRe. The refinement of top and bottom component capsid structures were carried out using CNS version 1.1 and the polypeptide models were built into the final electron-density map using the interactive graphics program O. The quality of the map was sufficient for building the model and unambiguous positioning of the side chains. There is a significant difference in the radius of the top and bottom component capsids, the top component being 5 Å larger in radius. Thus, RNA makes the capsid more compact, even though RNA is not a pre-requisite for capsid assembly. Partially ordered RNA was observed in the bottom component. The refined models could form the basis for understanding the architecture, protein-protein interactions, protein-nucleic acid interactions, stability and assembly of PhMV. Chapter IV provides a detailed description of the mutations carried out on PhMV coat protein towards enhancing the diffraction quality of crystals. The gene coding for PhMV coat protein (PhMVCP) and several of its deletion and substitution mutants were originally cloned in pRSETC and pET-21 vectors by Mira Sastri and Uma Shankar in Prof. Savithri’s laboratory at the Department of Biochemistry. It was observed that the recombinant intact coat protein and several mutants lacking up to 30 amino acids from the N-terminal end could assemble into empty shells resembling the natural top component. None of these deletion mutants crystallized in forms that diffracted to high resolution. Based on the intersubunit contacts observed, three more site-specific mutants were designed. These three mutants were expressed in BL21 (DE3), purified and crystallized. Even these mutant crystals did not diffract to high resolution. The polypeptide fold of PhMV coat protein therefore was carefully examined for probable reasons. It was found that PhMV subunit has three major cavities. Three cavities are likely to increase the flexibility of protein subunits, which in turn may result in crystals of poor quality. Mutations V52W, S158Q and A160L were shown to fill up these cavities and with the view of obtaining better crystals. These site specific mutations were carried out the mutant proteins were purified. It was shown that the recombinant capsids are stable and possess T=3 architecture. Two mutants were crystallized and a data set for V52W extending to 6.0 Å resolution could be collected. Due to the limited resolution, further work was not pursued. It is plausible that the triple mutant will diffract to higher resolution. The second part of the thesis deals with stress response proteins from Oryza sativa and Salmonella typhimurium. It is known that viral infection and abiotic and biotic stresses induce a network of proteins in plants. Chapter V presents a review of the current literature on stress proteins, focusing mainly on Oryza sativa and S. typhimurium stress response proteins. Chapter VI describes the over expression of stress proteins SAP1 and SAP2 from rice. These stress related proteins confer tolerance to cold, dehydration and salt stress in rice. These proteins have been cloned in the expression vector pEt-28(a) and expressed in E. coli strain BL21 CodonPlus(DE3)RIL. The proteins were purified and crystallization trials were made. However, there were no hits. In an attempt to get crystals, nine deletion constructs of SAP1 were designed eliminating potentially disordered and unfolded regions based on a bioinformatics analysis. Crystallization trails are being carried out on three of the constructs. Structural studies on a universal stress protein from Salmonella typhimurium, which shares homology with the rice universal stress proteins, was initiated. Apart from this, several other stress related proteins of Salmonella typhimurium have also been selected for structural and functional studies. These include YdaA, YbdQ, Yic, Ynaf, Yec, Spy and Usb. All these were cloned and expressed in E. coli. Out of seven proteins, Ynaf, YdaA and YbdQ were found in the soluble fraction and were expressed in quantities suitable for structural studies. I could crystallize YdaA and Ynaf. X-ray diffraction data to resolutions of 3.6 Å and 2.3 Å were collected on crystals of YdaA and YnaF, respectively. A tentative structure of YnaF has been obtained. Further attempts to determine these structures are in progress. Biophysical, Biochemical functional characterization of YdaA and YnaF proteins are described. Structural studies on mannose-6-phosphate isomerase, an enzyme related to stress regulatory proteins from S. typhimurium are dealt with in Chapter VII. Mannose 6-phosphate isomerase (MPI) catalyzes the interconversion of mannose 6-phosphate and fructose 6-phosphate. The structure could be solved in its apo and holo forms (with two different metal atoms, Y3+ and Zn2+), and complexed with the cyclic form of the substrate fructose 6-phosphate (F6P) and Zn2+. Isomerization involves acid/base catalysis with proton transfer between C1 and C2 atoms of the substrate. Lys 132, His 131, His 99 and Asp 270 are close to the substrate and are likely to be the residues involved in proton transfer. Interactions observed at the active site suggest that the ring opening step is catalyzed by His 99 and Asp 270. An active site loop consisting of residues 130-133 undergoes conformational changes upon substrate binding. The metal ion is not close to the substrate atoms involved in proton transfer. Binding of the metal induces structural order in the loop consisting of residues 50-54. Hence, the metal atom does not appear to play a direct role in catalysis, but is probably important for maintaining the architecture of the active site. Based on these structures and earlier biochemical work, a probable isomerization mechanism has been proposed. The thesis concludes with a brief discussion on the future prospects of the work. Efecto del extracto etanólico del fruto de Physalis peruviana ("Aguaymanto") sobre la glucemia en animales de experimentación

Objetivo: Determinar el efecto hipoglicemiante en ratas normales y diabéticas (inducida con aloxano) al administrar el extracto etanólico de los frutos de Physalis peruviana (Aguaymanto). Materiales: frutos de Physalis peruviana (Aguaymanto) y 56 ratas machos cepa Holtzman. Método: Los animales de experimentación fueron divididos en 7 grupos (Normal, sobre carga de Glucosa, sobre carga de glucosa y glibenclamida, Insulina y tres concentraciones de Extracto) luego de realizado el experimento se dejó descansar 2 semanas y se produjo a randomizar y dividirlos en 7 grupos (Normal, aloxano, aloxano y glibenclamida, Insulina y tres concentraciones de extracto); después de 24 horas los animales presentaron un nivel de glucosa > 250 mg/dL y se procedió con el experimento; así mismo se realizó el estudio histológico del páncreas. Resultado: Al administrar el extracto en las 3 concentraciones se observa un efecto hipoglicemiante llegando hasta un 41.5% de disminución de la glucemia en ratas con sobre carga oral de glucosa. En el experimento de inducción de diabetes con aloxano se observa una disminución de 4.38% a las 2 horas con el extracto 600 mg/dL; así también se observa en el estudio histológico un menor daño del páncreas en el grupo experimental de inducción de la diabetes con aloxano. Conclusión: Se demostró el efecto hipoglicemiante del extracto etanólico de frutos de Physalis peruviana (Aguaymanto) administrado por vía oral en ratas con sobre carga de glucosa y aloxanizadas. / Objective: to determine the blood glucose lowering effect in normal and diabetic rats (alloxan-induced) to manage the ethanolic extract of the fruits of Physalis peruviana (Aguaymanto). Materials: fruits of Physalis peruviana (Aguaymanto) and 56 male Holtzman strain rats. Method: The experimental animals were divided into 7 groups (Normal, overload of glucose on glucose load and glibenclamide, insulin and three concentrations of extract) then performed the experiment was allowed to stand for 2 weeks and came to randomize and divide in 7 groups (Normal, alloxan, alloxan and glibenclamide, insulin and three concentrations of extract); after 24 hours the animals had glucose levels > 250 mg / dL and proceeded with experiment; likewise histological study of the pancreas was performed. Result: When administering the extract in 3 concentrations, a hypoglycemic effect reaching 41.5% decrease in blood glucose in rats with oral glucose load on is observed. In the experiment with alloxan diabetes induction decreased 4.38% was observed after 2 hours with the extract 600 mg / dL; well observed in the histological study of the pancreas less damage in the experimental induction of diabetes with alloxan. Conclusion: The hypoglycemic effect of ethanol extract of fruits of Physalis peruviana (Aguaymanto) administered orally in rats on overload glucose and alloxan diabetes was demonstrated. The principal means of spreading species from the genus Physalis are seeds. Physalis is considered easy to grow because of the quantity of seeds the genus produces, and its high rates of germination. This study aimed to characterize the developmental cycle and compare the growth of tomatillo 'purple' in two light conditions. For this purpose, P. ixocarpa 'purple' was grown in PVC pots with four replications of 10 samples for each treatment, a total of 40 plants per treatment. Evaluation of the seedling emergence was held in a range of 7, 14 and 21 days after sowing. The analysis of other phenophases started up to 26 days after sowing, was maintained throughout the cycle, and held in 14 days intervals. The estimated physiological parameters are the content of chlorophyll a and b, and leaf carotenoids. The plasticity of the species in the study was assessed using growth analysis with the variables height, stem diameter, number of leaves, buds, flowers and fruits, and dry weight of plant growth. Analysis was performed with the variables height, stem diameter, number of leaves, buds, flowers, fruits, dry mass of plants and leaf anatomy. The means of the data were assessed using analysis of variance (ANOVA) and the student T-Test with a 5% significance level. In this study the parameters that were influenced by light were the emergence rate of seedlings, the content of photosynthetic pigments, leaf thickness, period required for the establishment of phenophases, dry weight, fruit production and seed vigor. Although the plants grown in direct sunlight have higher rates of growth, greater production of fruits and viable seeds were obtained in plants grown under shade. / O centro de origem de Physalis ixocarpa ? o M?xico, sendo a quinta hort?cola mais produzida no pa?s, usada principalmente na prepara??o de pratos tradicionais da cultura mexicana. A quimioterapia é o único tratamento efetivo para a doença, mas essas drogas são, em geral, tóxicas e requer um longo período de tratamento. Produtos naturais provenientes de plantas oferecem novas perspectivas e representam uma importante fonte de novos agentes leishmanicidas. Assim, é de grande importância avaliar os efeitos do extrato aquoso da raiz de Physalis angulata, planta amplamente utilizada pela medicina popular, em formas promastigotas e amastigotas de Leishmania (Leishmania) amazonensis e sua ação sobre a célula hospedeira. As fisalinas D, E, F e G foram demonstradas pela primeira vez na raiz de P. angulata pela análise cromatográfica. Uma atividade antiproliferativa e uma inibição dose dependente de promastigotas 74,1% e 99,8 % (IC50 35,5 μg/mL) e amastigotas 70,6% e 70,9% (IC50 32.0 μg/mL) foram observadas quando os parasitas foram tratados com 50 e 100 μg/mL do extrato, respectivamente. A análise da atividade microbicida da célula hospedeira infectada com L. amazonensis mostrou que extrato foi capaz de reverter o efeito causado pelo parasito de inibir a produção de espécies reativas de oxigênio. O tratamento com o extrato também induziu alterações morfológicas importantes em formas promastigotas avaliadas por microscopia óptica, microscopia eletrônica de transmissão e varredura. Foram observadas alterações na morfologia, na divisão celular, principalmente na fase de citocinese, na membrana flagelar, na bolsa flagelar e alterações em organelas importantes, como o cinetoplasto, onde ocorreu duplicação irregular e alteração do seu tamanho. Já por citometria de fluxo foi possível confirmar que o tratamento induziu uma exposição de fosfatidilserina e diminuição no volume celular de promastigotas tratadas. Com relação à célula hospedeira, o extrato promoveu alterações no citoesqueleto, o aumento número de projeções citoplasmáticas, do volume celular e de vacúolos e da habilidade de espraiamento sem causar efeito citotóxico ou alteração ultraestrutural em macrófagos tratados com o extrato. Assim, estes resultados demonstram que o extrato aquoso da raiz de P. angulata foi eficaz na ativação da célula hospedeira e na inibição do crescimento do protozoário, o que representa uma fonte alternativa e promissora de agente leishmanicida. / Leishmaniasis is an infectious disease caused by various species of the protozoan parasites of the Leishmania genus. The chemotherapy is the only effective treatment for the disease, but these drugs are, in general, toxics and requires a longer treatment period. Natural products have been used as traditional medicine and offer new perspectives and represent an important source of new antileishmanial agents. Thus, it is of great importance to assess the effects of the aqueous extract of the root of Physalis angulata, a plant widely used in popular medicine, in promastigotes and amastigotes of Leishmania (Leishmania) amazonensis and its effect on the host cell. Physalins D, E, F and G were found present, for the first time, in the P. angulata roots using liquid chromatography/mass spectrometry analysis. Antiproliferative activity and a dose-dependent inhibition of promastigote growth 74.1% and 99.8 % (IC50 35.5 μg/mL), and intracellular amastigotes 70.6% and 70.8% (IC50 32.2 μg/mL) was observed when parasites were treated with 50 and 100 μg/ mL of extract, respectively. The analysis of the microbicidal activity of host cell infected, with L. amazonensis demonstrated that extract is able to reverse the effect caused by the parasite to inhibit the production of reactive oxygen species. This growth inhibition was associated with several morphological alterations assessed by optical microscopy, transmission electron microscopy and scanning such as alteration on cell division, especially in the phase of cytokinesis, alteration in flagellar membrane, in flagellar pocket and duplication of kinetoplast DNA. Already by flow cytometry was possible to confirm that the treatment induced a phosphatidylserine exposure and decreased cell volume of promastigotes treated. In the host cell were observed cytoskeleton alterations, high number of cytoplasmatic projections, increase of cytoplasm, vacuoles and spreading ability. No cytotoxicity towards macrophages was observed. We have demonstrated that aqueous extract effectively promotes antileishmanial activity and clearly demonstrate the induction of apoptosis and ultrastructural alterations in Leishmania parasites. Thus, aqueous extract may represent a promising natural alternative source for a new antileishmanial agent.

In addition to cultivated plants, the begomoviruses are also associated with a wide range of weeds. This study aimed to characterize the begomoviruses associated with weed families Malvaceae (Herissantia crispa, Waltheria indica, Sidastrum paniculatum, Sida rhombifolia e Triumfetta semitriloba), Solanaceae (Physalis sp.) and Lamiaceae (Hyptis sp.) In northeastern Brazil to assess their diversity and importance as sources of new virus to cultivated plants. Samples of weeds with symptoms typical of begomovirus infection were collected in the states of Alagoas, Pernambuco and Bahia during the years 2010-2012. We obtained a total of 28 genomic components (18 DNA-A and 10 DNA-B). Analysis of the sequences indicated the presence of eight species of begomovirus, six new species (HeYMV, TrYMV, PaYNV, PhYSV, HyRMV1 and HyRMV2). In phylogenetic analyzes between viruses obtained malvaceous weeds. indicate that all new species clustered with Brazilian begomoviruses. Evidence of recombination were detected manifold where the begomoviruses from Sida spp. and tomato were identified as parents. In weed Hyptis sp. and Physalis sp. Sequence analyzes indicated the presence of three new species of begomoviruses, the two found in the same sample Hyptis sp. (Mixed infection) and another found initially Physalis sp. and subsequently detected in Hyptis sp. In phylogenetic analyzes of these three new species formed a single monophyletic branch. The analyzes showed strong evidence of recombination between HyRMV1 and HyRMV2, and the like parental HyRMV1. The ToRMV was identified as a possible parental species HyRMV1 and PhYSV. Therefore, these results indicate that the weeds are important reservoirs of begomoviruses and that recombination events have apparently contributed to the emergence of new species in some hosts. / Os Begomovirus (família Geminiviridae) são caracterizados por infectarem plantas dicotiledôneas, apresentarem partículas icosaédricas geminadas, com genoma composto por um ou dois componentes de DNA circular de fita simples (DNA-A e DNA-B). Além de plantas cultivadas, os begomovírus estão também associados a uma ampla gama de plantas daninhas. Esse estudo teve como objetivo caracterizar os begomovírus associados a plantas daninhas das famílias Malvaceae (Herissantia crispa, Waltheria indica, Sidastrum paniculatum, Sida rhombifolia, Triumfetta semitriloba), Solanaceae Physalis sp.) e Lamiaceae (Hyptis sp.) no nordeste brasileiro para avaliar sua diversidade e importância como fontes de novos vírus para plantas cultivadas. As amostras de plantas daninhas com sintomas típicos de infecção por begomovírus foram coletadas nos estados de Alagoas, Pernambuco e Bahia durante os anos de 2010 a 2012. (infecção mista) e a outra encontrada inicialmente em Physalis sp. e posteriormente detectada em Hyptis sp. Nas análises filogenéticas essas três novas espécies formaram um único ramo monofilético. As análises de recombinação indicaram uma forte evidência de recombinação entre HyRMV1 e HyRMV2, tendo o HyRMV1 como parental. O ToRMV foi identificado como possível parental para as espécies HyRMV1 e PhYSV. Portanto, esses resultados indicam que as plantas daninhas constituem importantes reservatórios de begomovírus e que eventos de recombinação tem aparentemente contribuído para o surgimento de novas espécies nessas hospedeiras.

It is a species that produces edible fruits with a sour and sweet flavor, characterized as a fleshy berry. It has great potential for cultivation that has been attracting the attention of producers, traders and consumers. The objective of this work was to evaluate the liming requirement, to determine the tolerated aluminum saturation, the desired base saturation and the calcium and magnesium requirement for the physalis crop. A completely randomized design with five replications was used. Calcium oxide with a concentration of 380 g kg-1 of calcium oxide and 125 g kg-1 of magnesium oxide, with total neutralization power of 90%, was applied to liming requirements (LR) of 0, 0.6, 1.4 and 2.3 t ha-1 for Typic Quartzipsamment (TQ) and 0, 0.8, 1.8 and 2.8 t ha-1 for Rhodic Hapludox (RH). The height of the plants, shoot diameter, shoot dry mass, root dry mass, yield, longitudinal diameter and transverse diameter of the fruits of fisalis were submitted to multivariate analysis using the canonical variable of analysis of variance LR and soil type. Linear regression equations were adjusted for each variable as a function of LR for each soil type. Optimum values of fisalis development for soil acidity indexes and for optimal concentrations of macro and micronutrients in the diagnostic leaf were obtained by substituting the recommended LR in the regression equations. The maximum values for all evaluated attributes were obtained with the application of 1.6 t ha-1 for TQ and 2.0 t ha-1 for RH. Liming is an essential practice for the cultivation of Physalis peruviana L. in acid soils, where it is aimed to achieve higher yields. The determined standards for tolerated aluminum saturation, desired base saturation and calcium and magnesium requirements are: 5.0 %, 67 % and 2.5 cmolc dm-3, respectively.
