Why Does Media Marketization Reinforce Media Control in Post-Tiananmen China?: A Political Economic Theory of Media Control

The current Chinese media political literature ascribes China’s effective media control to Communist Party censorship. Up until now, scholars and authors have overlooked how the enormous social and economic changes that China has undergone since economic reform has affected media control. This dissertation explores how such changes influence media control in China. It first examines the Chinese political economy and then focuses on studying China’s media, which has gone through considerable change since economic reform. Previously, Party ideological indoctrination and violent suppression were rampant. Today’s situation, however, could be characterized as indoctrination mingled with entertainment or “indoctritainment” (Sun 2002), and repression with an absence of full freedom of the media. I argue that “repressive state capitalism” has propelled economic development in China, particularly since 1989. In the reform era, repression coexists with economic development and is actually productive to Chinese economic growth because repression has both ensured state intervention in the economy and safeguarded a stable environment that is pivotal to the flourishing of economic activities. Using this political economic approach, I propose a political economic theory of “marketizing media control” to account for effective media control after media marketization, beginning with an empirical investigation of the traditional media and ending with an investigation of the new media in China. In short, repressive state capitalism is my contribution to political economic theory and marketizing media control is my contribution to Chinese media politics. / Graduate

L’avènement des AAIRE a grandement contribué à la consécration d’un ordre répressif administratif aux côtés de l’ordre pénal, de même qu’il a contribué au renouvellement de la notion de sanction administrative. En tant que juristes, le pouvoir répressif des AAIRE nous interpelle à plus d’un titre, car il illustre parfaitement l’exorbitance du droit administratif et des pouvoirs de l’administration en matière économique. Si, pour les partisans de la régulation, sanctionner c’est encore réguler, il n’en demeure pas moins que ce pouvoir, qui sonne comme un retour du système de l’administration-juge, manquant de bases juridiques solides, altère les concepts du droit processuel et porte souvent atteinte aux droits fondamentaux du procès. C’est d’ailleurs ce qui explique la réorganisation interne de ces autorités, en distinguant les organes/fonctions de poursuites et d’instruction des organes/fonctions de jugement, et en soumettant l’exercice de ce pouvoir aux principes et règles régissant le droit pénal. Or, le phénomène de processualisation, qui accroît les garanties des administrés, est de nature à entamer l’efficacité tant recherchée à la création de ce type de régulateurs. Enfin, l’étude comparative des droits français et algérien sur ce sujet, nous révèle le hiatus existant entre eux, plus particulièrement à propos du respect des droits de la défense, de même qu’elle nous renseigne sur l’ampleur du phénomène mimétique en droit algérien, mais également ses limites. / The advent of the quasi autonomous non-governmental organizations of economic regulation has largely contributed to the consecration of an administrative repressive order beside the penal order, as well as it contributed to the renewal of the concept of administrative sanction. The repressive power of the Quango challenges us in many respects, because illustrates the exorbitance of the administrative law and the powers of the administration in economic matters.

Investigating the roles of arabidopsis polycomb-group genes in regulating flowering time and during plant development by (I) challenging silencing and (II) developing approaches to dissect Pc-G action

Polycomb-group (Pc-G) proteins regulate homeotic gene silencing associated with the repressive covalent histone modification, trimethylation of histone H3 lysine 27 (H3K27me3). Pc-G mediated silencing is believed to remodel chromatin, rendering target genes inaccessible to transcription factors. Pc-G mediated silencing might result in irreversible changes in chromatin structure, however, there has been little analysis addressing whether Pc-G mediated silencing is reversible. In this work we focused on CURLY LEAF (CLF), the first Pc-G homologue discovered in Arabidopsis. CLF mediated repression of the floral homeotic gene AGAMOUS (AG) was challenged during early and late leaf development. AG was activated by the late leaf promoter, revealing that Pc-G mediated silencing can be overcome in old leaves in the presence of CLF. AG was also activated in young leaf primordia, yet did not persist in older leaves, revealing that transient activation of a Pc-G target is not epigenetically stable. To address the mechanism of Pc-G action within an endogenous environment, the histone dynamics at the APETALA1 (AP1) locus were characterized by Chromatin Immunoprecipitation. Unexpectedly, we found that the activation of AP1 in leaves did not require the removal of H3K27me3, questioning whether H3K27me3 is sufficient to silence. The roles of CLF in leaf and flower development are masked due to partial redundancy with SWINGER (SWN). clf- swn- mutants form a callus-like mass on sterile-tissue culture with no distinguishable plant organs. The role of CLF in regulating flowering time in natural populations of A. thaliana was investigated by complementing clf- mutants with CLF alleles from two accessions. We found that natural variation in CLF did not affect flowering time. To dissect the roles of CLF and SWN in late leaf and flower development, two approaches were developed for targeted expression. Firstly, CLF was introduced into the LhG4/ pOp transactivation system to provide CLF during early plant development. For mosaic analysis, CLF was introduced into the CRE lox recombination system in order to create clf- sectors surrounded by CLF+ SWN+ and CLF+ swn- cells.

The thesis is focused on repressive coping style and self-reports of the anger. In the theoretical part are elaborated chapters concerning the definition of emotion, emotion regulation, coping and repressive coping style. The Repressors are characterized by unconscious denial of their own anxiety and self-image distortion within the low susceptibility to negative emotions. The research study focuses on the expression of anger by repressors and their comparison with other groups. Respondents (n=122) were divided into four groups using conventional questionnaires (STAI, MCSD): repressors (n=26), high-anxious (n=16), defensive high-anxious (n=47) and low-anxious (n=33). The aim of the study was find out differences in the expression of anger and trait anger. The hypotheses were partly supported, as the results in the expression of anger and trait anger pointed to differences between repressors and low-anxious and partly supported among other groups. The present study shows that repressors distort even other socially censored emotions, in this case anger.

TITLE: UNRAVELING THE MOLECULAR MECHANISMS OF THE CLASS II TRANSACTIVATOR IN SKELETAL MUSCLE MAJOR PROFESSOR: Dr. Judy Davie The inflammatory cytokine, interferon gamma, IFN-gamma orchestrates a diverse array of fundamental physiological processes and exhibits complex effects on myogenesis. IFN-gamma also induces the class II transactivator, CIITA, which is a critical mediator of IFN-gamma mediated repression and activation. The aims in my dissertation are directed towards understanding the role of IFN-gamma and CIITA in muscle. Stimulation by IFN-gamma in skeletal muscle cells induces CIITA expression as well as MHC class II gene expression. We show that the IFN-gamma induced inhibition of myogenesis is mediated by CIITA, which specifically interacts with myogenin. CIITA acts by both, repressing the expression and inhibiting the activity of myogenin at different stages of myogenesis. The IFN-gamma mediated repression is reversible, with myogenesis proceeding normally upon removal of IFN-gamma. We also show that CIITA is indispensible for the inhibition of myogenesis. To gain a mechanistic insight into the IFN-gamma induced repression of myogenesis, we have discovered that IFN-gamma and CIITA inhibit myogenesis by modifying gene regulation in a muscle cell subject to inflammation. We show that CIITA first interacts with JARID2, a non catalytic subunit of PRC2 complex, which induces a paused RNAPII, phosphorylated at serine 5 and then interacts with the catalytic subunit EZH2, in a JARID2 dependent manner. Our data show that both CIITA and IFN-gamma block myogenesis by the induction and recruitment of the PRC2 complex, which is normally silenced in a differentiating muscle cell. One of my dissertation aims sheds light on the silencing of CIITA in Rhabdomyosarcoma. Silencing of CIITA prevents the expression of MHC Class I and II genes. We have found that IFN-gamma signaling is intact in these cells, but pSTAT1 and IRF1 do not bind to the CIITA PIV promoter. The CIITA promoter is not hypermethylated in RD (ERMS) cells, but shows a modestly enhanced methylation status in SJRH30 (ARMS) cells. We have also observed that histone acetylation, which normally increases on the CIITA PIV promoter following IFN-gamma treatment, is blocked in both types of RMS cells. Further, our studies also impart a novel role for IFN-gamma and CIITA in inhibiting the IGF induced activation of muscle specific genes. Our data show that IFN-gamma does not block the signaling cascade of IGF. However, blocking exogenous IFN-gamma restores IGF activation of muscle specific genes. My dissertation also reveals an important role for the FACT complex in the early steps of gene activation through its histone chaperone activities that serve to open chromatin structure and facilitate transcription promoting muscle differentiation. We show that myogenin interacts with the FACT complex and the recruitment of FACT complex to muscle specific genes is dependent on myogenin. The final aim in my dissertation highlights the distinct binding profiles of the MRFs and E proteins during proliferation and differentiation. Our sequential ChIP assays show that MYOD, MYOG, and MYF5 co-occupy promoters. Taken together, my dissertation provides a comprehensive understanding of the molecular mechanisms during myogenesis and reveals the deleterious effects of chronic inflammation in skeletal muscle.

Adaptive Styles and Coping Strategies of Youth Diagnosed with Cancer: Relationship to Well-Being, Psychosocial and Educational Adjustment, and Parents’ Adaptive Styles

Although almost 16,000 children and adolescents are diagnosed with cancer each year, the incident rates have remained stable over recent years, and mortality has decreased consistently since 1975 (American Cancer Society, 2016). With increased survivorship, research and practice in pediatric oncology has focused more on the psychosocial well-being of patients during and after treatment. With research repeatedly indicating that patients and families appear to exhibit great resilience and adjustment, and low incidences of psychosocial difficulties (e.g., Kazak, 1994, Eiser et al. 2000), some researchers have examined adaptive style as a possible construct that may help explain the predominantly positive outcomes (e.g., Phipps and Srivastava, 1997). The current study examined adaptive styles and coping strategies in youth diagnosed with cancer to determine relationships between these variables, as well as between each of these constructs and measures of subjective well-being, and psychosocial and educational adjustment. This study was also the first to examine relationships between youth and parent adaptive styles. A total of 180 youth between 9 and 17 years old who have been diagnosed with cancer participated in this study. Each youth also had one parent who participated. Each youth participant completed a packet of self-report measures used to determine adaptive style group, coping strategy use, and subjective well-being scores. Each parent participant completed a questionnaire to provide sociodemographic information about the youth and parent, as well as information about their child’s illness and school experience. Parents also completed a packet of measures used to provide information about their child’s psychosocial adjustment and risk and to determine parent adaptive style. Data were analyzed to examine relationships between parent and youth adaptive style, group differences on measures of subjective well-being, psychosocial adjustment/risk, and academic variables, relationships between adaptive style and coping strategy use, and the predictive strength of adaptive style and coping strategies for the outcome variables. Although the distribution of adaptive styles was similar among participants in the current study compared to those in previous studies of adaptive style for Repressive, Low Anxious, and Defensive High Anxious adaptive styles, there was a higher percentage of participants with High Anxious adaptive style in the current study. A statistically significant bidirectional relationship was found between youth and parents with a Repressive adaptive style. Results revealed significant differences between groups on measures of subjective well-being and internalizing behaviors, with Repressive adaptive displaying the highest subjective well-being and lowest internalizing behavior scores. Repressive and Low Anxious adaptive style differed significantly from the two High Anxious groups on the school scale scores, with the Repressive and Low Anxious groups having more positive outcomes on the measure. Relating to coping strategies, youth in the two High Anxious groups used more strategies all together, including both adaptive and non-adaptive strategies. However adaptive strategies were more frequently related to positive adaptive styles and outcomes, while non-adaptive strategies tended to have a stronger relationship to High Anxious adaptive style and negative outcomes. Limitations are discussed, and suggestions for future research and practical implications are offered, based on the results of the current study.
