Pesquisa do Estado da Bahia - FAPEB / The genus Lippia (Verbenaceae) is characterized by having species with medicinal and aromatic properties, its economic potential aims to the production of essential oils with important biological and pharmacological activities. Even with all known diversity and potential, few species are the target of agronomic studies. Considering the propagation as the first step in the domestication of a species and the influence of external factors on essential oil production, this study aimed to evaluate the effect of cutting type and auxin concentrations in the vegetative propagation of the species L. insignis, L. lasiocalycina and L. thymoides, as well as different types of fertilization and harvest season in production, and content and chemical composition of the essential oil. In propagation tests two types of cuttings were used (apical and middle) and five IBA concentrations (0, 250, 500, 750 and 1000 mg L-1). In cultivation three forms of fertilization were tested (control, manure and manure + NPK 10:10:10) and two harvest seasons (210 and 360 days after planting). The essential oils are extracted from dry leaves by the hydrodistillation method by Clevenger apparatus, and chemical composition determined by gas chromatography with flame ionization detector (GC / FID) coupled to mass spectrometer (GC / MS). It was concluded that apical cuttings are the most suitable for the propagation of L. insignis and both apical and middle cuttings for L. thymoides and L. lasiocalycina; treatment with IBA is not necessary in the propagation by cuttings of these species; chemical fertilizers (NPK) associated with cattle manure was the best treatment in growth and biomass production of the species L. lasiocalycina and L. thymoides, while for L. insignis to the control treatment was the best; pruning, followed by the second growing season (regrowth) is recommended in the cultivation of the studied species, aiming at the production of essential oil; ?-myrcene, limonene and E-ocimenone are the major components of L. insignis; ?-myrcene and E-ocimenone the major components of L. lasiocalycina and E-caryophyllene and caryophyllene oxide the major components of L. thymoides; fertilization and harvest season promote quantitative variations in the chemical composition of essential oils; the species L. insignis and L. thymoides are the most promising for essential oil production in the conditions of Feira de Santana, Bahia, Brazil. / O g?nero Lippia (Verbenaceae) ? caracterizado por apresentar esp?cies com propriedades medicinais e arom?ticas, sendo seu potencial econ?mico voltado para a produ??o de ?leos essenciais com importantes atividades biol?gicas e farmacol?gicas. Parmi les maladies affectant les pommes, la moisissure bleue causée par Penicillium expansum est une préoccupation majeure. Elle cause des pertes de rendement et de qualité dues également à la production de mycotoxines telles que la patuline (PAT) et la citrinine (CIT). La PAT est la plus alarmante en raison de ses propriétés cytotoxiques, génotoxiques et immunosuppressives.L'Union européenne (UE) a établi des réglementations spécifiques pour protéger la santé des consommateurs et des niveaux maximaux de 50 g / kg sont fixés pour les jus de fruits et les produits dérivés, 25 g / kg pour les purées de pommes et les compotes et 10 g / kg pour les aliments destinés aux bébés et aux jeunes enfants. En dépit de ces mesures, la PAT continue à être présente dans les aliments et / ou les boissons commerciaux, dépassant parfois les limites maximales. Des recherches supplémentaires sont par conséquent nécessaires pour minimiser la contamination des produits alimentaires par cette mycotoxine et son champignon producteur. Bien que la plupart des études sur P. expansum soient essentiellement centrées sur la PAT, le génomede ce champignon présente d'autres clusters de métabolites secondaires (SM) prédits dont certains peuvent être associés à des métabolites potentiellement toxiques. Afin de contrôler la synthèse de SM, l'étude des facteurs de transcription globaux régulant leur production est essentielle. Dans une première partie, le gène veA, appartenant à la famille des protéines du complexe velvet, a été caractérisé et son impact sur le développement du champignon, sa virulence et son métabolisme secondaire a été élucidé. La délétion de ce gène a conduit à une réduction de la production de PAT et de CIT et à une diminution de l'expression de leurs clusters de gènes. VeA a également un impact global sur le métabolisme secondaire, puisque 15 des 35gènes de structure présentent une régulation différentielle sur les milieux testés. Dans une deuxième partie, l’influence de l’acide ascorbique (AA) sur la concentration de PAT dans le jus de pomme trouble a été étudiée à la fois en laboratoire et en milieu semi-industriel. Une méthodologie analytique séparant la PAT et d'autres composés générés au cours de la réaction aété optimisée. Les conditions optimales d'action de l’AA sur la PAT ont été analysées. De plus,nous avons identifié des produits de dégradation moins toxiques que la PAT et résultant du traitement par l’AA. Pour conclure, cette thèse se rattache à la gestion des risques de la PAT dans le secteur des fruits ; elle apporte des connaissances et des améliorations significatives tant sur le plan fondamental que sur le plan pratique. Ces avancées résident principalement dans la description d'une souche mutée de P. expansum moins toxique que celle naturellement retrouvée dans la nature, et décrivant un additif alimentaire améliorant les qualités de nombreux produits transformés et diminuant la concentration de PAT en générant des composés moins toxiques. / Among diseases affecting apples, blue mould caused by Penicillium expansum is a major concern. It causes yield and quality losses, as well as food safety issues due to the production of mycotoxins such as patulin (PAT) and citrinin (CIT). PAT is the most worrying one and has cytotoxic, genotoxic and immunosuppressive properties. The European Union (EU) has established specific regulations to protect the consumer’s health and maximum levels of PAT of 5 g/kg is set for fruit juices and derived products, 25 g/kg for apple purees and compotes and 10g/kg for food intended for babies and young children. However, PAT is still found in commercial food and/or beverage products, sometimes exceeding the maximum limits and more research is needed to minimize contamination of food products by this mycotoxin and its fungus. Even though most studies on P. expansum have focused on PAT itself, the genome of this fungus exhibits other predicted secondary metabolite (SM) clusters, some of which may be associated with potentially toxic metabolites. In order to control the synthesis of SMs, the study of global transcription factors regulating their production is essential. In a first part, the veA gene, belonging to the velvet family, was characterised and its impact on the development of the fungus, its virulence and its secondary metabolism was elucidated. The disruption of this gene led to the failure in PAT and CIT production and a decrease in the expression of their gene cluster. It also revealed a global impact on the secondary metabolism, as 15 of 35 backbone genes showed differential regulation on the media tested. In a second part, the influence of ascorbic acid (AA) on the concentration of PAT in cloudy apple juice was studied on both lab and semi-industrial scale. An analytical methodology separating PAT and other compounds generated during the reaction was optimized. Optimal conditions of action of AA on PAT were studied. In addition, degradation products less toxic than PAT and resulting from AA treatment were identified. To conclude, this thesis is part of the risk management of PAT in the fruit sector; it provides significant improvements at both fundamental and practical levels. These advances are mainly characterized by the description of a mutated strain of P. expansum that is less toxic than that naturally occurring in nature, and the description of a food additive that improves numerous products qualities and affects PAT concentration, thusgenerating less toxic compounds

In dieser Arbeit wird die Eignung eines Haarwurzelsystems (HR) aus transgenen Sonnenblumen-hairy roots zur biotechnologischen Produktion von α-Tocopherol untersucht. Es wurden hairy roots mit und ohne Transgene erzeugt. Transgene HR exprimieren Tocopherolbiosynthesegene aus Arabidopsis thaliana. Die HR wurden unterschiedlichen Stressoren unterworfen, um die α-Tocopherolproduktion binnen eines Zeitraums zu überprüfen. Stressoren waren verringerte Kohlenstoffquelle, Beleuchtung und der Zusatz von Zytokininen. Die Produktionsleistung des Systems ist schlussendlich ungenügend zur kostengünstigen Produktion von α-Tocopherol.

Streptomyces bacteria are well-known producents of many bioactive metabolites. Its secondary metabolism is a source of many important groups of active compounds that are recently investigated by means of many new methods based on bioinformatic analyses of genome data, modern LC-MS techniques, and metabolic modeling methods. This thesis originates from the genetic screening for a specific gene (als) for cyclizing 5-aminolevulinate synthase. Based on earlier studies, we consider this gene as a genetic tag of the producers of secondary metabolites containing the C5N unit (2-amino-3-hydroxycyklopent- 2-enon). Such metabolites include several groups with variable structures and biological activities, which include manumycins as well. Manumycins are small polyketides with a weak antibiotic aktivity, especially against gram-positive bacteria. However, its cancerostatic and anti-inflammatory effects are of greater importance. Streptomyces monomycini BCCO10 1552 and Streptomyces capoamus BCCO10 1636 strains were found positive for the presence of the als gene in the targeted genetic screening. By the als phylogeny, they cluster near the producents of manumycin compounds. Genetic engineering of the primary/secondary metabolic interface in tobacco BY-2 cells

The supply of precursors from primary metabolism is often overlooked when engineering secondary metabolism for increased product yields. This is because precursor supply may be assumed to be non-limiting, and it is considered difficult to engineer primary metabolism, because control of carbon flow (flux) is generally distributed among most enzymes of the pathway. The aim of this thesis was to increase the production of sterols, part of the isoprenoid class of secondary metabolites, in tobacco (Nicotiana tabacum) Bright Yellow 2 (BY-2) cell cultures. This was achieved by genetically engineering increased activity of mitochondrial citrate synthase, an enzyme of the tricarboxylic acid (TCA) cycle that is involved in the provision of cytosolic acetyl coenzyme A, the primary metabolite precursor to sterols. Metabolic flux analysis revealed that citrate synthase exerts significant control over cyclic TCA cycle flux in BY-2 cells and suggested that increasing the activity of downstream enzymes within secondary metabolism could lead to a further redirection of TCA-cycle-derived precursors into sterol biosynthesis. Attempts were made to achieve this by genetically engineering increased activity of 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGR), a key enzyme of secondary metabolism involved in sterol biosynthesis. Consistent with previous research, transgenic lines had increased sterol levels. However, the high sterol phenotype was unstable, and attempts to co-express HMGR and citrate synthase genes were unsuccessful. The thesis demonstrates that increasing the provision of precursors to secondary metabolites can result in increased yields of those secondary metabolites but suggests that in most cases the activity of enzymes within secondary metabolism has a greater effect on those yields. It also reveals that single enzymes can exert significant control of flux within primary metabolism, although the control exerted by specific enzymes probably changes with the demands placed on metabolism.

Recentes estudos genômicos têm mostrado que vários fungos e bactérias possuem um potencial biossintético superior à quantidade de me tabólitos secundários já isolados desses micro-organismos. A descoberta de produtos naturais inéditos e bioativos é limitada pela impossibilidade dos micro-organismos expressarem to das as suas rotas biossintéticas em laboratório. Assim, estratégias alternativas para i nduzir a produção de produtos naturais microbianos são necessárias. A utilização de cultur as mistas de micro-organismos é uma estratégia que vem sendo recentemente utilizada, na tentativa de mimetizar condições mais naturais de crescimento. Além disso, a adição de mo duladores químicos e epigenéticos às culturas microbianas também pode potencialmente est imular a produção de compostos de interesse, seja por ativar mecanismos celulares em resposta à condição de estresse, ou por alterar a taxa de transcrição de certos genes, em f unção de mudanças no grau de enovelamento da cromatina. Alternativamente, a indu ção de certos genes, e até mesmo a diversificação do metabolismo secundário, podem ser conseguidos através de engenharia genética, pela manipulação direta de genes de inter esse. A linhagem endofítica Alternaria tenuissima SS77, selecionada para os experimentos de modulaçã o química e epigenética, teve seu metabolismo secundário alterado após o tra tamento com diferentes moduladores. Provavelmente, o efeito observado ocorreu em função de uma eliciação inespecífica dos diferentes moduladores. Além disso, o cultivo misto desse fungo com o fungo endofítico Nigrospora sphaerica SS67 , isolada da mesma planta hospedeira ( Smallanthus sonchifolius ), levou ao isolamento de dois novos policetídeos, da classe das perilenequinonas, juntamente com um já relatado na literatura científica. Para r ealizar os cultivos microbianos mistos, envolvendo uma linhagem bacteriana e uma fúngica, t rês linhagens de actinobactérias e cinco de fungos, todos endofíticos da planta Lychnophora ericoides , foram selecionadas. Alterações no perfil metabólico da cultura mista de Phomopsis sp FLe6 com Streptomyces albospinus RLe7 foram as mais evidentes e por isso a maioria das investigações foram focadas nessa cultura mista. Várias condições de cu ltivo foram testadas e diferentes resultados foram obtidos. Em alguns casos, o desenv olvimento da linhagem fúngica foi inibido pela bacteriana, e em outros, foi observado o inverso. Da mesma forma, houve acentuada inibição da produção de alguns metabólito s secundários na presença da linhagem desafiadora, mas também foi verificada a eliciação de outros. Os extratos das culturas simples desses micro-organismos também apresentaram relativas alterações nos perfis metabólicos em função das condições de cultivo. Os metabólitos produzidos pelo fungo Phomopsis sp FLe6 e pela actinobactéria Streptomyces albospinus RLe7 foram isolados e caracterizados. Os resultados mostram que as intera ções entre os micro-organismos endofíticos são bastante complexas, estando sujeita s a ação de diversos fatores externos que muitas vezes não podem ser pré-determinados. Po r isso, estabelecer um cultivo misto adequado, do ponto de vista da eliciação da produçã o de metabólitos secundários, pode requerer uma série de tentativas. Ainda assim, os r esultados almejados podem ser conseguidos utilizando essa estratégia. Diferenteme nte das linhagens endofíticas, manipuladas quimicamente através de diferentes estr atégias, a linhagem sequenciada de Fusarium heterosporum ATCC 74349, foi manipulada geneticamente para a co nstrução de um gene biossintético híbrido pks-nrps , contendo a porção nrps do gene híbrido da equisetina e um pks críptico de Aspergillus fumigatus . Era esperado que a linhagem hibridizada fosse capaz de produzir o metabólito se cundário geneticamente planejado, entretanto, após seu cultivo, esse produto não foi detectado nos extratos, e as possíveis razões são discutidas. Ainda que os resultados espe rados não tenham sido obtidos, estudos que contribuam para a ampliação do entendimento das megassintases fúngicas são de extrema valia. / Recently, genetic studies have shown that several b acteria and fungi hold a greater biosynthetic potential than the amount of secondary metabolites isolated from these microorganisms. The discovery of novel bioactive na tural products is limited by the inability of microorganisms to express all their biosynthetic pa thways in laboratory conditions. Therefore, alternative strategies to induce the production of microbial natural products are required. Mixed cultures of microorganisms are a strategy tha t has been used to mimic more natural conditions of growth. Furthermore, the addition of chemical and epigenetic modulators to the microbial cultures can also stimulate the productio n of compounds by activating cellular mechanisms in response to stress conditions or by c hanging the transcription rate of certain genes, due to changes in the chromatin folding. Alt ernatively, the induction of some genes, and even the diversification of secondary metabolis m, can be achieved by genetic engineering, by manipulating genes of interest. The endophytic strain Alternaria tenuissima SS77, which was selected for the experiments of che mical and epigenetic modulation, had changed its secondary metabolism after treatment wi th different modulators. Probably, the observed effect was due to a nonspecific elicitatio n of those modulators. Moreover, the mixed cultures of this fungus with the endophytic fungus Nigrospora sphaerica SS67, isolated from the same host plant ( Smallanthus sonchifolius ), led to the isolation of two new polyketides, belonging to perylene quinone class, along with ano ther one already reported in the scientific literature. Three strains of actinobacteria and fiv e fungi, all endophytes of Lychnophora ericoides , were selected to grow in microbial mixed cultures comprising one bacteria and one fungus. Changes in the metabolic profile of the mix ed culture of Phomopsis sp. FLe6 with Streptomyces albospinus RLe7 were the most obvious, and then further studi es were focused on this mixed culture. Many culture conditions were analyzed and different results were obtained. In some cases, the development of the fun gal strain was inhibited by bacteria, and in other cases was observed the opposite. Similarly , there was a remarkable inhibition of the production of certain secondary metabolites in the presence of the challenging strain, but the eliciting of others was also observed. The extracts of the single cultures of these microorganisms also showed changes in metabolic pro files due to culture conditions. The metabolites produced by the fungus Phomopsis sp. FLe6 and the actinobacteria S. albospinus RLe7 were isolated and characterized. The results show that interactions between endophytic microorganisms are quite complex and are influenced by various external factors that often can not be previously determined. Theref ore, establishing a suitable mixed culture to elicit the production of secondary metabolites m ay require some attempts. Still, the expected results can be achieved using this strateg y. Unlike the endophytic strains, that was chemically manipulated by different strategies, the sequenced strain Fusarium heterosporum ATCC 74349 was genetically manipulated to construct a hybrid PKS-NRPS biosynthetic gene containing the NRPS portion of the hybrid gene of e quisetin and a cryptic PKS gene of Aspergillus fumigatus . It was expected that hybridized strain could be a ble to produce the secondary metabolite genetically planned, however, after its cultivation, this product was not detected in any extracts, and some possible reasons are discussed. Evolution modularer Multienzymsysteme des bakteriellen Sekundärstoffwechsels

Modulare Polyketidsynthasen (PKS) sind Multienzymsysteme des bakteriellen Sekundärstoffwechsels. An ihnen läuft eine schrittweise Biosynthese vielfältiger Kohlenstoff-Gerüste ab, die von einfachen Carbonsäure-Einheiten ausgeht. Polyketid-Verbindungen zeigen eine große Bandbreite pharmazeutisch interessanter Aktivitäten. In dieser Arbeit wurde eine Reihe von Evolutionsstudien durchgeführt. Zunächst wurden die phylogenetischen Beziehungen zwischen modularen PKS und anderen PKS-Systemen sowie Fettsäuresynthasen untersucht, wodurch ihre zentrale Stellung innerhalb eines langen Evolutionsprozesses gezeigt werden konnte. Eine detaillierte Analyse der Phylogenien von Domänen bakterieller modularer PKS ergab, dass das Ausmaß an Genduplikationen, Genverlusten und Ereignissen horizontalen Gentransfers zwischen den verschiedenen Bakteriengruppen beträchtlich variiert. Aus der Genomsequenz des Actinobakteriums Streptomyces avermitilis wurden die Phylogenien aller Domänentypen rekonstruiert. Der Vergleich dieser Einzelphylogenien ermöglichte es, eine Reihe von homologen Rekombinationsereignissen aufzufinden. Homologe Rekombination scheint der Hauptmechanismus zu sein, auf dem die Strukturvielfalt der Polyketide in Bakterien beruht. Mit Hilfe eines „genome mining“-Ansatzes konnte im Genom des Cyanobakteriums Nostoc punctiforme eine Reihe von Biosynthese-Clustern, die zu den PKS und nichtribosomalen Peptidsynthetasen gehören, identifiziert werden. Durch chromatographische und massenspektrometrische Analysen von Zellextrakten und Kulturüberständen konnten einige der Biosynthese-Cluster bestimmten Metaboliten zugeordnet werden. Eines der Cluster wurde hinsichtlich des produzierten Metaboliten und der Regulationsstruktur eingehender charakterisiert. Die Folgerungen aus den gewonnen Ergebnissen werden im allgemeinen Zusammenhang der Evolution metabolischer Diversität ausführlich diskutiert. / Modular polyketide synthases (PKS) are multienzym systems of bacterial secondary metabolism. They perform a stepwise biosynthesis of diverse carbon skeletons from simple carboxylic acid units. Polyketide compounds possess a wide range of pharmaceutically interesting activities. In this study, a series of evolutionary analyses was performed. Initially, the phylogenetic relationships between modular PKS and other PKS systems as well as fatty acid synthases were investigated revealing their central position within a long evolutionary process. In detail reconstruction of the phylogenies of bacterial modular PKS domains demonstrated that the extent of gene duplications, gene losses and horizontal gene transfer events varies considerably between different bacterial groups. Using the genome sequence of the actinobacterium Streptomyces avermitilis the phylogenies of all domain types were reconstructed. Comparison of these phylogenies allowed for detecting numerous events of homologous recombination, which appears to be the main mechanism underlying polyketide structural diversity in bacteria. A genome mining approach revealed a number of biosynthesis clusters of the PKS and nonribosomal peptide synthetase type in the genome of the cyanobacterium Nostoc punctiforme. Cell extracts and culture supernatants were analysed by means of liquid chromatography and mass spectrometry and some of the biosynthesis clusters could be assigned to specific metabolites. One of the clusters was characterised in greater detail regarding the produced metabolite and the cluster’s regulatory structure. The implications of the results are extensively discussed within the general context of the evolution of metabolic diversity.
