Structure, metamorphism, and tectonics of the northern Oman-UAE ophiolite and underlying metamorphic sole

Ophiolites - thrust sheets of oceanic lithosphere that have been emplaced onto the continental margin - provide the opportunity to explore the structure and genesis of oceanic crust. As many ophiolites formed above subduction zones, they also allow for the investigation of mantle wedge and subduction interface processes. This the- sis examines the Oman-United Arab Emirates (UAE) ophiolite, which is the largest and most intensely studied ophiolite on Earth. Three distinct problems are addressed. (1) Recent research has proposed that the architecture and tectonic evolution of the ophiolite in the UAE differs from in Oman. In Chapter 2, I test this hypothesis by integrating new geological mapping and field observations with previously published maps of the ophiolite in the UAE. My results indicate that the ophiolite is gently folded, but otherwise largely intact. I demonstrate that the architecture of the ophi- olite in the UAE is not significantly different from in Oman. Thus, there is no basis for a different tectonic evolution as recently proposed. (2) Observations from exper- iments and small-scale natural shear zones indicate that volumetrically-minor phases can control strain localization. In Chapter 3, I test the hypothesis that minor phases control strain-localisation at plate boundaries. To do so, I analyzed peridotites from the base of the ophiolite, a palaeosubduction interface. My results demonstrate that minor phases limited olivine grain growth, which led to rheological weakening. (3) The mechanisms by which metamorphic soles detached from the downgoing slab and accreted to the hanging-wall mantle is unclear. In Chapter 4, I examine a transect across the metamorphic sole in the UAE. My results reveal that granulite formation was more extensive than is typically considered. I propose that granulite formation resulted in rheological strengthening, which caused the subduction interface to migrate into the downgoing slab and accrete the metamorphic sole.

Neuroendocrine control of puberty in vertebrates : characteriization of the kisspeptin system in flatfish

The recently discovered decapeptide kisspeptin and its G-protein coupled receptor form a signaling system expressed ubiquitously and are implicated in a variety of still poorly characterized functions. In the brain, kisspeptin is secreted by specific neurons and its receptor is localized in GnRH neurons. Kisspeptin signaling has been fully established in the control of the onset of puberty in vertebrates, from fish to mammals. In this study, we characterized the kisspeptin gene in the Senegalese sole and characterized the kisspeptin receptor genes in both the Senegalese sole and in the Atlantic halibut. In contrast to other fish species, the two species analyzed here showed only the presence of one ligand and one receptor, probably as a consequence of the genome reduction characteristic of Pleuronectiformes. However, in both cases we found an alternative splicing mechanism based on intron retention that produces also non-functional isoforms, but whether this is part of a mechanism to control abundance of the active gene product is still not known. We document spatial and temporal changes of expression of kisspeptin and its receptor in the brain, pituitary and gonads related to the annual reproductive cycle. Finally, we present the first evidence of a possible link between energy balance and reproduction mediated by kisspeptin signaling in a non-mammalian vertebrate. / El recentment descobert decapèptid kisspeptina i el seu receptor associat a una proteïna G formen un sistema que s’expressa ubiqüitament i que està implicat en diverses funcions, moltes de les quals encara no estan ben caracteritzades. En el cervell, la kisspeptina és secretada per neurones específiques, mentre que el seu receptor es troba a les neurones GnRH. Aquest sistema s’ha relacionat amb el control de l’inici de la pubertat en diferents vertebrats, des de peixos fins a mamífers. En aquest estudi, hem caracteritzat el gen de la kisspeptina en el llenguado senegalès, i els gens del receptor de la kisspeptina tant a llenguado senegalès com en l’Halibut de l’Atlàntic. Al contrari del que ocorre en moltes altres espècies de peixos, aquestes dues espècies només presenten un gen pel lligand i un gen pel recep- tor. Aquest fet és probable que estigui relacionat amb la reducció de la mida del genoma que han sofert els Pleuronectiformes. Tot i així, en les dues espècies s’hi troba un mecanisme d’empalmament alternatiu conseqüència d’una retenció intrónica que produeix una isoforma no funcional. Ara bé, si aquest mecanisme està relacionat amb el control de l’abundància dels trànscrits de la isoforma funcional encara està per esbrinar. Per altra banda, hem trobat canvis en l’expressió gènica tant en l’espai com en el temps durant un cicle reproductiu dels gens de la kisspeptina i el seu receptor en el cervell, pituïtària i gònades. Finalment, també presentem la primera evidència, en un vertebrat no mamífer, d’una possible relació entre el balanç energètic i la reproducció controlada pel sistema kisspeptina.

Ultrassonografia Doppler do fluxo arterial digital associado à alteração do coxim digital dos bovinos / Doppler ultrasonography of digital blood flow associated with changes in bovine digital cushion

O objetivo do estudo foi avaliar, por meio da ultrassonografia bidimensional e da ultrassonografia Doppler, as inter-relações entre a alteração de espessura do coxim digital com os índices vasculares da artéria digital dorsal comum dos membros pélvicos dos bovinos. A avaliação ultrassonográfica Doppler é uma técnica não invasiva de avaliação de índices vasculares, assim como, indiretamente, do volume de sangue do leito vascular. Para isso, foram realizados exames ultrassonográficos da espessura do coxim digital e também avaliação ultrassonográfica Doppler da referida artéria de 13 vacas primíparas da raça Holandesa, em intervalos de 14 dias, durante 126 dias após o parto. Ao longo do experimento o coxim digital apresentou diminuição não significativa de espessura (p=0,1715) e apresentou forte correlação positiva com a média do escore de condição corporal (p=0,03, r=0,71). Ao longo do período avaliado, o fluxo de sangue aumentou significativamente (p<0,0001) e apresentou correlação negativa com os índices que denotam impedância ao deslocamento de sangue, isto é, índice de resistividade (p<0,0001, r=-0,34911) e pulsatilidade (p<0,0001, r=-0,5035). Além disso, realizou-se correlação entre a diferença diária do fluxo de sangue e a diferença da espessura do coxim digital para cada animal e constatou-se uma forte tendência à correlação entre as diferenças das variáveis (p=0,0515, r=-0,5729). Por meio dos resultados encontrados foi possível concluir que o escore de condição corporal teve forte correlação com a espessura do coxim digital, ao longo do estudo. Além disso, o fluxo de sangue, os índices referentes à velocidade de deslocamento das hemácias e o diâmetro da artéria aumentaram ao longo do trabalho. E, também, que a discreta diminuição do coxim digital não foi um fator que influenciou no aumento do fluxo de sangue e nos índices vasculares / The aim of this study was to evaluate, by bidimensional and pused Doppler ultrasound, the interrelationship between the changes of the thickness of digital cushion with vascular indices of the common dorsal digital artery of the hind limbs of cattle. Pused Doppler sonography is a non-invasive tool to assessment the vascular indices, as well as, the blood flow volume in vascular bed. So, were performed ultrasound exams of soft tissue thickness and also the Doppler ultrasonographic evaluation of that artery in thirteen first lactation Holstein cows and followed them up during 126 days in milk. Throughout the experiment the digital cushion showed a slight decrease without significance (p=0,1715), this also was strongly correlated with body condition score (p=0,03, r=0,71). The blood flow volume had significance increase (p<0,0001) and was negatively correlated with both impedance indices, resistance index (P<0,0001 r =-0,34911) and pulsatility index (p <0,0001 r =-0,5035). In addition, a correlation was performed with both daily differences of the blood flow and the thickness of the digital cushion for each animal and it was observed a strong tendency of correlation between the different variables (p=0,0515 r =-0,5729). Therefore, we can conclude that body condition score was strong correlated with the thickness of digital cushion, throughout the study. In addition, the blood flow volume, the velocity indices and the artery diameter increase throughout the days in milk. And, so, the slight decrease in the thickness of digital cushion was not a factor that influenced the increase of blood flow and vascular indices

Identification of novel cold-adapted nitrilase superfamily enzymes

Philosophiae Doctor - PhD / In bacteria, nitrile hydratases and enzymes of nitrilase and signature amidase superfamilies hydrolyse nitriles and amides to their corresponding carboxylic acids releasing ammonia. Bacteria expressing these enzymes are typically isolated where a sole nitrogen and/or carbon source is used to support their growth. The majority of characterised enzymes of industrial potential have been identified for their stabilities at elevated temperatures. To date, no reports of such enzymes have been isolated from cold adapted bacteria.In this study, an extensive screening program of cold-active microbial isolates for enzymes of this group led to the selection and detailed characterisation of an aliphatic amidase from Nesterenkonia.Nesterenkonia AN1, a new psychrotrophic isolate of the genus, was isolated from soil samples collected from the Miers Valley, Antarctica. AN1 showed significant 16S rRNA sequence identity to known members of the genera, but this is the only strain that had optimal growth at approximately 21oC. AN1, similar to known members, is an obligately alkaliphilic (pH 9-10) and halotolerant (Na+ 0- 15% (w/v)) strain.The genome of Nesterenkonia AN1, sequenced in-house, revealed two ORFs encoding putative nitrilases, referred to as Nit1 and Nit2. Based on analysis of their deduced protein sequences, both belonged to the nitrilase superfamily. Both sequences showed conserved catalytic residues (EKEC), glycine residues and contained the characteristic áââá monomer fold. Homology modelling using known structures suggested that both genes could encode N-carbamoyl D-amino acid amidohydrolases, although neither showed conserved residues implicated in the hydrolysis of carbamoyls.Nit1 and Nit2 were expressed in Escherichia coli BL21 (DE3) pLysS as Cterminal and N-terminal hexahistidine tagged fusion proteins, and purified using Ni-chelation chromatography. Nit1 showed no activity towards nitrile, amide and carbamoyl substrates. This protein, unlike members of the multimeric enzymes of the nitrilase superfamily, was a monomer ~30 kDa protein. It is possible that the C-terminal hexahistidine tag might have prevented Nit1 from forming multimeric proteins.Nit2 showed substrate specificity similar to known aliphatic amidases with a preference for small amides. Nit2 had maximal activity at 30oC and between pH 6.5 and 7.5, properties compatible with its cold-adapted alkaliphilic origins. In addition, the enzyme was irreversibly inactivated at temperatures above 30oC and had a half-life of approximately 7 mins at 60oC. The crystal structure of Nit2 was solved to 1.66 Å. It revealed a ~45.5 kDa dimer, composed of two tightly bound ~30 kDa monomers. These monomers associated along the A surface forming a áââá-áââá sandwich architecture that is conserved in known structures of the nitrilase superfamily.Nit2 is distinct from known aliphatic amidases in both its structure and enzymic activity: the enzyme did not possess an extended C-terminal region; is active in dimeric form; has high affinity for 3C amides rather than 2C amides; and has a low overall catalytic rate. The short C-terminal region of Nit2 may have contributed to the low stability of the enzyme at elevated temperatures. A dendrogram composed of protein sequences of members of the nitrilase superfamily and Nit2 further supported evidence that this aliphatic amidase falls within a distinct group of enzymes.This is the first report of the enzymic characterisation and structural analysis of an aliphatic amidase from a psychrotolerant, alkaliphilic and halotolerant extremophile.

Complete avulsion of the heel pad with talar and calcaneal fracture: salvage with multiple K‑wire anchorage, internal fixation and free ALT flap

Background Degloving of the sole of the foot is a rare and serious injury because the heel pad cannot be replaced by similar tissue. The management is challenging and only a few cases have been reported with different treatment regimens. - Methods Here, we report on a 46-year-old female patient with complex foot trauma consisting of complete avulsion of the heel pad at the hindfoot and a soft tissue defect at the posterior aspect of the heel accompanied by rupture of the anterior tibial tendon and fractures of the talus, calcaneus and midfoot. The sole of the foot was fixed to the calcaneus with multiple temporary Kirschner wires and moist wound dressings. The anterior tibial tendon was sutured. The soft tissue defect at the posterior heel was treated with a free anterolateral thigh flap. The fractures were fixed in staged procedures. - Results At 2-year follow-up, the patient had a durable soft tissue cover over the heel with full sensation over the sole and pliable flap over the posterior aspect of the heel. The patient was able to fully bear weight and was pain free during her daily activities in comfortable, custom shoes. All fractures had healed, the talar neck fracture after one revision and bone grafting. The foot was plantigrade and stable with preserved painless but limited range of motion at the ankle, subtalar and mid-tarsal joints. - Conclusion The unique tissue at the sole of the foot can be salvaged even in cases of full degloving at the hindfoot with the simple method of anchorage with multiple temporary K-wires. Traumatic defects of the vulnerable skin at the posterior aspect of the heel requires durable coverage with free flap coverage. With staged treatment of all bone and soft tissue injuries, a favorable result can be obtained even in case of a complex foot trauma.

Examen de la specificite du traitement des conflits familiaux / Analyse of the specific processing of the family disputes

Le droit de la famille est une vaste matière en constante évolution. Son histoire démontre que ce contentieux n’est pas facile à traiter. Depuis le XVIIème siècle, notre législateur a essayé par différents procédés d’apporter la meilleure réponse au conflit familial. Le tribunal de famille était composé de membres de la famille ou de voisins. Les juristes étaient totalement évincés de cette juridiction durant la Révolution française, et ont progressivement réapparu, pour anéantir cette juridiction au début du XIXème siècle. Le divorce et la filiation bénéficièrent également des changements de la société et connurent de nombreuses réformes. Les guerres mondiales du XXème siècle et la libéralisation de la femme ont amené les professionnels du droit à créer la chambre de la famille composée de juges habilités au traitement des divers conflits familiaux. Cette expérience conduit notre législateur a créer un juge spécialisé en la matière et notamment dans le divorce. Ses pouvoirs se développent en 1993 le remplaçant par le juge aux affaires familiales chargé de l’examen des conflits familiaux, en fonction des compétences qui lui sont confiées. Cependant, ce contentieux restant important, il se trouve dépassé. C’est ainsi que le législateur a introduit la médiation familiale, confiée au médiateur familial. Notre législateur utilise la procédure afin de trouver la meilleure solution au traitement de ces conflits familiaux. Actuellement, la commission Guinchard a soumis plusieurs propositions faisant l’objet de diverses lois intégrées dans notre droit français de la famille pour accroitre les compétences de ce juge et développer les modes alternatifs de règlement des conflits. / The family law is a vast subject which is in constant evolution. His history reveals that is litigation is not easy to treat. Since the XVIIth century our legislator tried different systems to bring the best solution to the familial litigation. The Family Court (tribunal de famille) was composed of persons who belong to the family in dispute, or of neighbors. Jurists were completely absent of this jurisdiction during the French Revolution. Finally, citizens realized that jurists were necessary in order to have a justice respecting each one’s rights. It disappeared at the beginning of the XIXth century. The divorce law and filiation law followed changes of the society and knew some reforms. The World Wars of the XXth century and the women’s liberation movement bring up jurists to create the family chamber (chambre de la famille) which is composed of judges who know families disputes. This experience was taken our legislator to invent a judge who is specialized in this subject and especially in the divorce. His powers were developed in 1993 with the “juge aux affaires familiales” who is charged to analyze family’s disputes which are in his competences. But this litigation is always to important and this judge doesn’t have enough time to treat it in a reasonable time, That’s why the legislator integrated into our legal system the familial mediation which is directed by a familial mediator. Our legislator uses the legal term to find the best solution to this litigation. Recently the Commission Guinchard is proposed some ideas which are legalizing in our French family law according more powers to this judge and developing alternative dispute resolutions.

La marche de l'enfant : évolution de la marche pieds nus et étude comparative de l'influence des éléments de conception de la chaussure / Young children gait : the evolution of barefoot gait and comparative study of shoe components influence on gait

Durant les premières années de marche indépendante, l'enfant voit sa stratégie de marche évoluer. De plus, il est rapidement amené à porter des chaussures dont l'influence sur sa marche est peu connue, même si documentée dans certains domaines particuliers (e.g. cas pathologiques, sport). Cette thèse consiste en la mesure de plus de 100 enfants sains, âgés entre 1 et 7 ans, marchant pieds nus et avec des chaussures spécifiquement développées pour l'étude (3 éléments variables : hauteur de talon, dureté de semelle et hauteur de tige). Des âges-clés, correspondant à l'âge où les paramètres biomécaniques sont semblables à ceux de l'adulte, ont été mis en évidence : 4 ans pour la cheville, 6 ans pour la hanche, et 7 ans pour le genou. Des régressions sur les paramètres biomécaniques en fonction de l'âge et de la vitesse de marche ont également été développées afin de servir de référence de population saine pour les études cliniques. Les mesures avec les chaussures ont révélé une influence prépondérante de la hauteur de talon sur la marche (e.g. mobilisation plus importante de la cheville avec un talon plus haut). À l'inverse, une tige haute sollicite moins la cheville au cours de l'appui. La dureté de semelle est relativement peu influente sur les paramètres de marche. Cette thèse a permis de préciser les résultats mis en évidence sur la maturation de la marche pieds nus et d'apporter des premières réponses aux industriels français de la chaussure enfant sur l'influence des éléments de conception de leurs produits sur la marche. Des futures mesures, plus nombreuses, permettraient de conclure sur l'influence croisée de ces éléments de conception / During first years of independent walking, children gait changes. Moreover, children wear shoes soon. Although gait of specific population was largely studied (e.g. pathological people, sports), shoe influence on young children gait is quite unknown. This thesis includes measurement of young children walking barefoot and wearing shoes especially designed for this study (3 variables components: heel height, upper height and sole hardness). For barefoot gait, a large database of biomechanical parameters was obtained, including more than 100 children aged between 1 and 7 years old. Key age were identified, corresponding to the age when biomechanical parameters are identical to adults’: 4 years for the ankle, 6 years for the hip and 7 years for the knee. Regressions between biomechanical parameters, age and speed were also established as a reference of healthy children for clinical application. Shoed measurements revealed leading influence of heel height on gait (e.g. more ankle mobilization with higher heel). Contrarily, high upper limits ankle movement during stance phase. Sole hardness poorly influences gait parameters. This thesis precises previous results obtained on gait maturation and delivers indications on shoe conception influence on gait for French children’s shoes manufacturers. Further measurements may allow conclusion about cross influence of shoe conception elements on children gait
