To increase their oxidative stability is necessary to add antioxidants, which are compounds capable of avoiding or delaying oxidation reactions and can be synthetic or natural, from the natural, we highlight a variety of plants with antioxidant activity, among them the mint (Mentha spicata L.) and Acerola (Malpighia glabra L.). The objective of this study was to evaluate the oxidative stability of biodiesel in the presence of natural antioxidants from extracts of Acerola and mint leaves obtained by conventional extraction and supercritical CO2 on temperature and various pressure using oxidative stability in Rancimat test, avaliou- is also the synthetic antioxidant TBHQ, to compare the results obtained with the use of the plant extracts. The effects of temperature and pressure on the yield and quality of the extract. The best induction times were obtained for biodiesel plus the supercritical extracts of Acerola 40 °C and 150 bar condition (1.38 h) and 60 °C and 250 bar (1.07 h) and the supercritical extract of mint conditions of 40 °C and 150 bar (1.08 h), 40 °C and 250 bar (1.71 h) and 60 °C and 250 bar (2.04 h) and soxhlet extract with hexane obtained from both acerola (2.84 h) and Mint (2.87 h), which showed greater induction time of the control (1.04 h). Biodiesel plus the synthetic antioxidant TBHQ got time higher induction to control and biodiesel plus extracts and from 36.95 hours. The oxidative stability of biodiesel was more increased when the extracts obtained by conventional extraction using Hexane as the solvent, which can be extracted from a larger quantity of compounds with antioxidant activity. / Biodiesel é um combustível que pode ser obtido de fontes renováveis e devido sua composição química é sensível à oxidação. Para aumentar sua estabilidade oxidativa é necessário acrescentar antioxidantes, que são compostos capazes de evitar ou retardar reações de oxidação e podem ser sintéticos ou naturais, dentre os naturais, destacam-se uma variedade de plantas com atividade antioxidante, dentre elas a Hortelã (Mentha spicata L.) e Acerola (Malpighia glabra L.). O objetivo deste trabalho foi avaliar a estabilidade oxidativa do Biodiesel na presença de antioxidantes naturais dos extratos das folhas de Acerola e Hortelã obtidos por extração convencional e com CO2 supercrítico em condições de temperatura e pressão variadas, utilizando teste de estabilidade oxidativa em Rancimat, avaliou-se, também, o antioxidante sintético TBHQ, para comparar com os resultados obtidos com a utilização dos extratos das plantas. Foram avaliados os efeitos da temperatura e Pressão no rendimento e da qualidade do extrato. Os melhores tempos de indução obtidos foram para o Biodiesel acrescido dos extratos supercríticos da Acerola nas condições 40 °C e 150 bar (1,38 h) e 60 °C e 250 bar (1,07 h) e os extratos supercríticos da Hortelã nas condições de 40 °C e 150 bar (1,08 h), 40 °C e 250 bar (1,71 h) e 60 °C e 250 bar (2,04 h) e os extratos soxhlet obtidos com hexano, tanto da Acerola (2,84 h) quanto da Hortelã (2,87 h), que apresentaram tempo de indução maior que o controle (1,04 h). O Biodiesel acrescido do antioxidante sintético TBHQ obteve tempo de indução superior ao controle e ao biodiesel acrescidos dos extratos, sendo de 36,95 horas. A estabilidade oxidativa do Biodiesel foi superior quando acrescido dos extratos obtidos por extração convencional utilizando Hexano como solvente, que, pode ter extraído uma maior quantidade de compostos com atividade antioxidante.

Phospholipides bio-sourcés riches en acides gras oméga 3 pour la formulation de liposomes / Naturally occurring phospholipids rich in omega 3 fatty acids for liposome formulation

Les liposomes, vésicules à base de phospholipides, sont des systèmes colloïdaux utilisés en recherche et dans différents domaines industriels (pharmaceutique, cosmétique, nutrition). Néanmoins, leur développement se heurte au manque de diversité des sources de phospholipides (soja et jaune d’oeuf). Parallèlement, les méthodes industrielles d’extraction des phospholipides sont basées essentiellement sur l’utilisation de solvants organiques. Dans ce contexte, mon sujet de thèse a eu pour but d’étudier l’extraction de PL, par le CO2 supercritique, à partir de différentes sources avec comme objectif final la formulation de ces PL en liposomes. La démarche a consisté à explorer différents sous-produits de l’huilerie (tourteau d’oléagineux) et de la pêche (sous-produit de la coquille Saint Jacques), potentiellement riches en phospholipides, puis à mettre en place différentes techniques analytiques pour discriminer et quantifier les types de lipides. Parallèlement, l’influence de différents paramètres de l’extraction par CO2 supercritique sur la pureté des fractions obtenues est abordée ainsi qu’une première approche de formulation de PL en liposomes et la caractérisation de ces vésicules. / Liposomes, phospholipids vesicles, are colloidal systems used in search and different industrial fields (pharmaceutical, cosmetic, nutrition). Nevertheless their development face lack of phospholipid sources (soya and egg yolk). At the same time, industrial methods to extract phospholipids use organic solvents. In this context, the phospholipid extraction were studied using a green technology from new different sources in order to formulate liposomes. Oil mill and fishery by-products (seed cake and scallop) were studied to know phospholipid quantities. Analytical techniques were established to discriminate and quantify lipid types. Simultaneously, varying the operating conditions of CO2 supercritical extraction allowed obtaining extracts with different purities and contents in phospholipids and a first study of liposome formulation was carried out.

O processo de imobilização térmica do PMODS foi otimizado aplicando-se planejamento composto central. O polímero, os suportes e as fases estacionárias foram caracterizados por testes físicoquímicos e cromatográficos. As fases estacionárias Si(PMODS), Si-Zr(PMODS) e Si-Ti(PMODS) apresentaram os melhores resultados quando foi aplicada a temperatura de 120 °C por 16 horas. As melhores FE foram submetidas ao processo de capeamento. Estas FE mostraram eficiências entre 82.000 e 90.000 pratos m e boas separações com picos simétricos para compostos apolares e picos com simetria adequada, segundo parâmetros estabelecidos pela Farmacopéia Americana (United States Pharmacopeia), para compostos polares, avaliados pelos testes de Engelhardt, Tanaka, Neue, SRM 870 e pelo modelo dos parâmetros de solvatação utilizando CFS. A presença do óxido metálico no suporte resultou em FE com maior estabilidade química em condições drásticas de fase móvel (pH 1,7 e pH 10 a 50 °C), quando comparada com fases similares baseadas em sílica nua. A reação de capeamento melhorou a estabilidade química das FE e diminuiu o número de silanóis residuais. As FE Si-Zr(PMODS), Si-Ti(PMODS), Si-Zr(PMODS)ec e Si- Ti(PMODS)ec apresentam potencialidade na separação de fármacos psicoativos, filtros ultravioleta (UV), xantinas e hidrocarbonetos policíclicos aromáticos (HPA), utilizando CLAE e CFS. / Abstract: This work presents the development of stationary phases (SP) for high performance liquid chromatography in the reversed phase (RP-HPLC) and for supercritical fluid chromatography (SFC) prepared by the sorption and thermal immobilization of poly(methyloctadecylsiloxane) (PMODS) onto silica and metalized silica supports modified with zirconium and titanium. The immobilization process was optimized using central composite design for determination of the best conditions of time and temperature. The polymer, supports and stationary phases were characterized with physico-chemical and chromatographic tests. The stationary phases Si(PMODS), Si-Zr(PMODS) and Si-Ti(PMODS) presented the best results when a temperature of 120 °C for 16 hours was applied. The best SP were submitted to endcapping processes. These SP showed efficiencies between 82,000 and 90,000 plates m and good separations with symmetric peaks for apolar and polar compounds evaluated by the Engelhardt, Tanaka, Neue and SRM 870 tests and the solvation parameter model using supercritical fluid chromatography (SFC). The presence of metallic oxide on the supports resulted in stationary phases with better chemical stability under drastic conditions of mobile phase (pH 1.7 and pH 10 at 50 °C), when compared to similar stationary phases based on bare silica. The endcapping reaction improved the chemical stability. The SP Si-Zr(PMODS), Si- Ti(PMODS), Si-Zr(PMODS)ec e Si-Ti(PMODS)ec presented potentialities for the separation different pharmaceutical compounds used as psychotropic drugs, ultraviolet (UV) filters, xanthines, and polycyclic aromatic hydrocarbons (PAH), using separations in HPLC and SFC. / Doutorado / Quimica Analitica / Doutor em Ciências

The lag between micro- and macro-mixing in compressed fluid flows

We report the application of a novel optical Raman-based measurement technique for the simultaneous determination of the progress of mixing on the micro- and on the macro-scale. The introduced measurement technique is applicable to mixing systems containing one compound, which potentially can form hydrogen bonds, such as water, alcohols or amines, and does not rely on the addition of traces of indicator compounds. Here we demonstrate its applicability by analyzing the lag of micro-mixing behind macro-mixing when liquid ethanol is injected into a supercritical bulk environment mainly composed of carbon dioxide (CO2). While the degree of mixing on the macro-scale is determined from the ratio of the intensities of characteristic Raman signals of ethanol and CO2, the degree of mixing on the micro-scale is determined from the shape of the OH stretching vibration Raman signal of ethanol, which is a function of the development of hydrogen bonds.

L'étude de l'injection d'un fluide dans des conditions de hautes pressions reste encore aujourd'hui un challenge. Lorsque la pression critique des fluides est dépassée, l'état supercritique est atteint, faisant disparaître la distinction entre liquide et gaz. Pour ces conditions extrêmes, les données expérimentales sont peu nombreuses et nécessitent d'être consolidées. Dans cette étude, un nouveau banc d'essai a été réalisé au laboratoire CORIA dans le but d'étudier des injections non-réactives d'éthane et de propane dans une atmosphère sub- et supercritique d'azote ou d'hélium. Les données ont été collectées à partir de quatre diagnostics optiques : l'ombroscopie, la DBI, la radiographie et la CBOS. Des informations qualitatives sur la topologie des jets et de leur couche de mélange sont apportées. Des mesures quantitatives de longueur de cœur dense, d'angle d'ouverture et de densité sont complétées par une étude phénoménologique à l'aide de la théorie des mélanges binaires. / Studying a fluid flow under high-pressure conditions through reliable experiments is still nowadays a challenge. When the chamber pressure exceeds the critical pressure of working fluids the supercritical state of matter is reached and the distinction between gas and liquid becomes blurred. For such special conditions, experimental data are scarce and need to be consolidated. In the present study, a new test bench has been designed at CORIA Lab to study the non-reactive injection of ethane and propane into nitrogen or helium under sub- and supercritical conditions. Experimental data are collected from four image-based techniques : shadowgraphy, diffused backlight illumination (DBI), radiography and color background oriented schlieren (CBOS). Qualitative information on topology of the jets and their mixing layer are provided. Quantitative measurements of dense core length, jet spreading angle and density field are supported by a phenomenological study based on binary mixing theory.

Charakterizace chirálních a achirálních chromatografických separačních systémů / Chromatograhic characterization of chiral and achiral separation systems

Dissertation thesis is a 5-publications' collection concerning characterization and application potential of cyclodextrins, polysaccharides and macrocyclic antibiotics based chiral stationary phases. The effects of stationary phase and mobile phase are studied. This approach ensures the complex insight into separation systems studied. Systems with different nature of chiral selector were studied by HPLC. Namely, macrocyclic antibiotics and derivatized polysaccharides were used for experiments. Former ones provided better results for enantioseparation of non-coded amino acids than latter ones. Dynamic coating procedure was used for preparation of a new chiral stationary phase. Characterization of new cationic cyclodextrin based chiral stationary phase was performed. Linear free energy relationship method was used for characterization of two different separation systems, i.e. newly prepared stationary phase and commercially available stationary phase. Based on results obtained, newly prepared stationary phase showed better results for separation of different achiral groups of analysts. New stationary phase prepared by dynamic coating was compared with chromatographic system, in which the chiral selector was used as a mobile phase additive. In this study, the effects of SCCD processing by different pressures 14, 16 and 18 MPa (35 ? 2 ?C / 10 min) on whey drink, whey drink and grape juice were investigated in comparison To conventional pasteurization (heat treatment at 72 ?C / 15 s). Physicochemical analyzes of pH, titratable acidity, total soluble solids, phenolic compounds, anthocyanins, antioxidant activity, angiotensin converting enzyme (ACE) inhibitory activity and volatile compounds were performed. The color, particle size, rheology, physical stability, as well as microbiological quality and sensory analysis of beverages were also smoothed. The results of this study evidenced the absence of differences between treatments in pH, titratable acidity, soluble solids, total anthocyanins and DPPH activity (p> 0.05). A direct relationship between SCCD pressure and ACE inhibitory activity was observed, with 34.63, 38.75 and 44.31% (14, 16 and 18 MPa, respectively). Few differences were found in the volatile compounds profile. The beverage processing by SCCD resulted in a product with lower particle diameter, lower consistency index and a reduction in pseudoplastic character compared to the beverage treated by the conventional process. No effect of high pressure CO2 on the sensorial attributes of the drink was observed for the studied levels. Consumers found no difference between CO2 treated beverages and heat-treated beverages. The results confirm the processing of SCCD as a promising technology for the non-thermal treatment of grape whey drink made available a health and wellness promoter beverage. Background: Non-thermal food processing is configured as an interesting alternative for the food industry due to the increased nutrient retention and minimal sensory changes in processed products. Scope and approach: The aim of this review is to address the potential of supercritical carbon dioxide technology, emphasizing milk and dairy processing, including the historical aspects, main advantages, microbial inactivation mechanisms, as well as effects in some quality parameters of dairy products. Key findings and conclusions: The use of supercritical carbon dioxide technology (SC-CO2) presents great potential application in dairy processing, since it is effective to reduce microbial load when compared to the pasteurization process, thus obtaining a product with greater shelf life and better organoleptic properties with minimal and sometimes positive changes in the intrinsic quality parameters The effect of supercritical carbon dioxide technology (SCCD, 140, 160, and 180 bar at 35 ? 2 ?C for 10 min) on whey-grape juice drink characteristics was investigated. Physicochemical characterization (pH, titratable acidity, total soluble solids), bioactive compounds ( phenolic compunds, anthocyanins , DPPH and ACE activity) and the volatile compounds were performed. Absence of differences were found among treatments for pH, titratable acidity, soluble solids, total anthocyanins and DPPH activity (p>0.05). A direct relationship between SCCD pressure and ACE inhibitory activity was observed, with 34.63, 38.75, and 44.31% (140, 160, and 180 bar, respectively). Regards the volatile compounds, it was noted few differences except by the presence of ketones. The findings confirm the SCCD processing as a potential promising technology to the conventional thermal treatment. The use of supercritical technology as a non-thermal pasteurization process of the whey-grape juice drink was investigated in this study. The effects of supercritical carbon dioxide at 14, 16, and 18 MPa (35 ? 2?C/10 min) on the physical and sensory properties of the beverage, when compared to conventional pasteurization (heat treatment at 72?C/15 s) were evaluated. High-pressure CO2 processing of whey-grape juice drink resulted in a product with lower particle diameter, lower consistency index, and a reduction in pseudoplastic character when compared to the beverage treated by the conventional process. No effect of high-pressure CO2 was observed on the sensory attributes of the beverage for the levels studied. Consumers did not find differences between the CO2-treated and heat-treated beverages. Eco-valorisation de la plante Kniphofia uvaria : de la plante à la galénique / Eco-valuation of the Kniphofia uvaria plant : from the plant to the galenic form

À l’heure où l’intégration des enjeux environnementaux dans le développement de procédés éco-efficients joue un rôle essentiel dans le moteur de l’innovation responsable, la chimie verte est devenue l’un des sujets de préoccupation majeure. Ainsi, le développement de nouveaux procédés éco-respectueux pour la production d’ingrédients naturels issus de matières premières végétales renouvelables est devenu une démarche incontournable dans le modèle de recherche. L’objectif de cette thèse a consisté au développement d’une stratégie d’éco-valorisation innovante employant les fluides sub/supercritiques pour l’extraction, la caractérisation, la production et l’imprégnation sur support cosmétique de produits naturels d’origine végétale. Pour cela, nous avons utilisé comme modèle végétal : les graines oléagineuses de la plante Kniphofia uvaria, sélectionnée pour des applications cosmétiques grâce à ses propriétés bioactives antioxydantes et anti-âge. Dans un premier temps, le développement de méthodes complémentaires en SFC ainsi que le développement du couplage SFC-MS a été réalisé à l’aide de la source APCI afin d’identifier les molécules responsables des activités bioactives des graines de Kniphofia uvaria. Ainsi, le développement d’un système hybride (U)HPLC/SFC-HRMS a été réalisé afin de mettre en place ce couplage. Des optimisations en termes de proportion et nature de solvant make-up ainsi qu’un travail au niveau des paramètres SFC et MS ont été faits afin de d’améliorer la sensibilité et la spécificité des analyses lipidiques. Dans un second temps, nous nous sommes attachés au développement d’une stratégie d’enrichissement en composés bioactifs à l’aide des méthodes : SFE et CPC. Ainsi, en SFE, des optimisations en termes de température, pression, nature/proportions de co-solvant dans le fluide ont été réalisées alors qu’en CPC, des optimisations au niveau de l’injection ont été faites. Des conditions optimales pour le fractionnement sélectif des anthraquinones et des triglycérides ont été déterminées en SFE et CPC. Dans un dernier temps, ce travail a consisté à développer un couplage en-ligne pour extraire et imprégner sélectivement sur silice cosmétique : les anthraquinones. Le développement et l’optimisation de ce procédé en-ligne ont été réalisés à l’échelle du laboratoire et ont démontré la faisabilité de ce couplage ainsi qu’un intérêt certain pour l’obtention de produits naturels sous une première forme galénique, destinée à une future incorporation dans la formulation de cosmétiques. / Nowadays, green chemistry is a great challenge. It seeks innovation in the development of eco-efficient processes. The production of natural products from renewable materials by these new environmentally friendly processes is more and more used. The aim of this Ph.D thesis is to develop an eco-valuation strategy to extract, characterize, produce and impregnate natural products onto a cosmetic support using sub/supercritical fluids. Consequently, we used oleaginous plant seeds from Kniphofia uvaria as a plant model, which was selected for its interesting cosmetic properties such as antioxidant or anti-ageing. Firstly, the SFC-MS hyphenation with the APCI as an ionization source was developed to screen bioactive molecules; responsible of cosmetic properties. This coupling was performed by the hybrid combination of (U)HPLC/SFC-HRMS. Various optimizations in terms of the solvent make-up (nature and proportion), modulation with SFC and MS parameters were carried out in order to improve sensitivity and selectivity of lipid analysis. Secondly, an enrichment strategy to concentrate bioactive compounds in the final extract was developed by SFE and CPC. Thus, in SFE, experimental parameters (temperature, pressure, nature/proportion of the modifier in the CO2 fluid) were optimized while in CPC, the injection optimization was realized. Methods for the selective fractionation of anthraquinones and triglycerides were obtained in CPC and SFE. Finally, an on-line sub/supercritical extraction-impregnation process was developed to extract and for simultaneously impregnating anthraquinones onto a cosmetic silica. Development and optimization of this process was realized on a laboratory scale. Consequently, this study demonstrated the feasibility of this concept and it presents a great interest to provide natural products as a galenic form, which could be used in the cosmetic formulation.

Optimisation de l’extraction des caroténoïdes à partir du persimmon (Diospyros kaki L.), de l’abricot (Prunus armeniaca L.) et de la pêche (Prunus persica L.) : étude photophysique en vue d’une application en thérapie photodynamique (PDT) / Optimization of carotenoids extraction from persimmon (Diospyros kaki L.), apricot (Prunus armeniaca L.) and peach (Prunus persica L.) : Photophysical study for photodynamic therapy (PDT) application

La thérapie photodynamique (PDT) est une technique utilisée cliniquement pour traiter certaines maladies de la peau, la dégénérescence maculaire liée à l’âge et certains types de cancer. Elle fait intervenir trois composants : une molécule photosensible ou photosensibilisateur (PS), la lumière et l’oxygène. Après administration du PS, celui-ci va se localiser plus ou moins sélectivement dans les zones tumorales où il est alors activé par irradiation lumineuse à une longueur d’onde et une puissance données. Ceci engendre la formation d’espèces réactives de l’oxygène (ROS) très réactives, dont l'oxygène singulet1 O2, qui entraînent la destruction des tissus tumoraux par nécrose ou apoptose. Afin d’améliorer la sélectivité du traitement, différentes pistes sont actuellement exploitées dont l’élaboration de « photodynamic molecular beacons » (PMB). Dans un PMB, le photosensibilisateur (PS) est associé via un peptide à un inhibiteur 1O2, appelé quencher. Ce quencher inhibe la formation d’1O2 tant que le composé n’a pas atteint sa cible. Une fois la zone cancéreuse atteinte, des enzymes spécifiques clivent le peptide, libérant ainsi le PS qui retrouve alors sa capacité à former de l’1O2. Trouver un couple PS/quencher adéquat reste un challenge en PDT. Les propriétés photophysiques particulières des caroténoïdes et leur aptitude à inhiber la production d’1O2 font de ces derniers des quenchers potentiellement utilisables pour l’élaboration de PMBs. Chez les plantes, les caroténoïdes (carotènes et xanthophylles) sont des pigments associés à la photosynthèse, qui ont deux rôles principaux : un rôle de collecteur de lumière et un rôle photoprotecteur en protégeant le(s) système(s) photosynthétique(s) contre les dommages photooxydatifs liés à une exposition trop intense à la lumière. Ceci s’opère, entre autre, via le cycle des xanthophylles. Cette aptitude à capter de l’énergie présente un intérêt potentiel à ne pas négliger dans la perspective de la conception de PMB utilisables en thérapie photodynamique. Dans le cadre de cette thèse en co-tutelle avec la Faculté des Sciences de Bizerte nous avons ciblé les caroténoïdes présents dans trois fruits produits en Tunisie à savoir les kakis (Diospyros kaki L.), les abricots (Prunus armeniaca L.) et les pêches (Prunus persica L.) connus pour leur richesse globale en ces pigments. Divers procédés d’extractions ont été étudiés : (i) L’extraction de type Soxhlet par solvants organiques à pression atmosphérique, utilisée comme référence, (ii) l'extraction accélérée par solvant organique (ASE : Accelerated solvent Extraction) effectuée sous pression, enfin (iii) l'extraction par CO2 supercritique avec l’éthanol comme cosolvant. Pour ces deux derniers procédés, une approche par plan d’expériences (surfaces de réponses) a été utilisée pour identifier les facteurs clé et les conditions optimales d’extractions de divers caroténoïdes (pression, température, débit, % de cosolvant, temps, nombre de cycles). L'analyse par chromatographie liquide à haute performance couplée à la détection UV-Visible et à la spectrométrie de masse a ensuite permis l'identification et la quantification des caroténoïdes présents dans les extraits obtenus, permettant ainsi de comparer les profils caroténoïdiques propres à chaque fruit et les performances de chaque procédé d’extraction. Cette étude ayant révélé un profil caroténoïdique particulièrement intéressant chez le kaki par rapport aux autres fruits, une extraction et une purification des caroténoïdes de ce fruit par chromatographie liquide haute pression préparative a ensuite été effectuée afin de disposer d’une quantité suffisante de chaque caroténoïde, et parfois de leurs isomères conformationnels, en vue de l’étude de leurs propriétés photophysiques (absorption, émission de fluorescence, inhibition d’1O2) et de l’évaluation de leur intérêt potentiel en tant que quencher d’1O2 dans un édifice de type PMB / Photodynamic therapy (PDT) is a clinically used technique for treating skin diseases, age-relatedmacular degeneration but mainly some types of cancer. PDT involves three components: a photosensitive molecule named photosensitizer (PS), light and oxygen. After administration of the PS, this one will be located more or less selectively in tumoral regions where it is activated by light irradiation at appropriate wavelength and power. This leads to the formation of highly reactive and cytotoxic reactive oxygen species (ROS), especially singlet oxygen, resulting in the destruction of the tumor by necrosis or apoptosis. To improve the treatment selectivity, different strategies are being exploited, one of which is the development of "photodynamic molecular beacons" (PMB). In PMB the photosensitizer is linked via a peptide to an inhibitor of 1O2 (quencher). This quencher inhibits the formation of 1O2 as long as the compound has not reached its target, namely cancer cells. In order to inhibit the toxicity of the PS in non-target cells and restore toxicity only close to the biological target, it is necessary to find an adequate PS/quencher couple. This remains a challenge for PDT. Carotenoids are interesting candidates due to their specific photophysical properties and ability to inhibit 1O2, which makes them potential quenchers for building PMBs. In plants, carotenoids (carotenes and xanthophylls) are pigments involved in the photosynthesis, in which they play two main roles: a light collecting role and a protecting role by preserving the photosynthetic systems against photoxydative damages induced by a too intense light exposure. This protection can for instance occur via the well-known xanthophylls cycle. This capacity to catch energy presents a potential interest that should not be neglected in the framework of the design of PMBs usable in photodynamic therapy. Within the framework as part of this PhD thesis in Cotutelle with the Faculty of Sciences of Bizerte, we focused on carotenoids from three fruits produced in Tunisia: persimmon (Diospyros kaki L.), apricot (Prunus armeniaca L.) and peache (Prunus persica L.), known for their global richness in these natural pigments. Three extraction processes were investigated: (i) the Soxhlet extraction based on the use of organic solvent at atmospheric pressure and used as reference, (ii) the accelerated solvent extraction (ASE) using organic solvent under high pressure, and (iii) the supercritical fluid extraction (SFE) using supercritical CO2 and ethanol as cosolvent. For these two last processes, a design of experiments (Surface Response Design) was used to identify the key factors and optimal extraction conditions of various carotenoids (pressure, temperature, flow, % cosolvent, time, number of cycles). Then, HPLC-PDA coupled with mass spectrometry (MS) enabled the identification and quantification of carotenoids from the extracts. Thus it was possible to compare the profiles in carotenoids content from each fruit as well as the performances of each extraction process. This study showed that the carotenoidic profile in the persimmon was the most interesting as compared to the profiles in the two other fruits. Extraction and purification of the carotenoids from persimmon by preparative high pressure liquid chromatography were then performed in order to have a sufficient amount of each carotenoid and sometimes of their conformational isomers. We finally performed a study of their photophysical properties (absorption, fluorescence emission, 1O2 inhibition) in order to evaluate their potential as 1O2 quencher in molecular construction such as a PMB
