The Role of the Prefrontal Cortex and Stress in Huntington Disease-Mediated Aggression

Huntington Disease (HD) is a fatal neurodegenerative disorder that is characterized by motor, cognitive, and psychiatric symptoms. Although HD onset is determined by motor symptoms, psychiatric symptoms, like depression and aggression, can develop earlier, have a larger impact on quality of life, and are understudied due to stigma. Our lab has observed hyper aggression in our humanized HD mouse model (Hu97/18) compared to our knock-in HD mouse model (Q175FDN). We characterized these differences and found that the Hu97/18 mice overreact in neutral situations, behaving as if they are in threatening situations. We are now using this novel model of HD-related aggression to study its neurological basis. Increased reactive aggression has been linked to stress levels and the prefrontal cortex (PFC) due to its role in emotional regulation. This study seeks to determine if HD-related aggression is associated with increased stress levels and changes in the PFC. Our cortisol study shows that the Hu97/18 mice display significantly higher cortisol levels than baseline, suggesting a link between systemic stress and heightened aggression. Additionally, quantified PFC volumes show a moderate relationship between PFC volume and aggression in wild-type (WT) mice that is lost in the Hu97/18 mice. This data will help elucidate factors that modulate aggression in HD and may identify therapies with high potential to alleviate this devastating symptom in patients.

Aim: The aim of the study was to investigate the degree of adherence and non-adherence to treatment regimen in the population with heart failure, as well as to explore relationships to symptom burden and hospitalization-frequency. Introduction: Heart failure – a chronic disease with a somber prognosis and high mortality, incidence and prevalence increasing world-wide. Adherence to treatment regimen mitigates symptom flare-up, increases life quality and impedes hospitalizations. Non-adherence is a global problem, historically underestimated and difficult to measure. The level of non-adherence to medical treatment in the heart failure-population is about 50 %, degree of non-adherence from a wider perspective implicates a gap of knowledge. Methods: A cross-sectional survey-study was conducted via the electronic survey: ”Living with heart failure”. Selection was consecutive, based on patients visiting the Cardiac outpatient-clinic, in a Swedish University Hospital. Data was transferred to descriptive statistics using the statistical processing program SPSS. Results: 479 individuals participated, non-adherent were 73,6 % and adherent 26,4 %. There was a significant difference between symptom burden among the non-adherent and the adherent. A trend also appeared between low hospitalization-frequency, high degree of adherence and low symptom burden, an equivalent trend among the most frequently hospitalized which exhibited the lowest degree of adherence and the most severe symptom burden. Conclusion: The results pinpoint the need to further draw attention to non-adherence, repeatedly representing the majority of the population. With a patient-centered approach nurses/caregivers could have an empowering effect on the patient´s ability to health literacy and thus promote conditions for adherence.

Barn får inte bedömas i förskolan, men en uppfattning görs så till vida om barnet är redoför en överflyttning till skolan eller om barnet bör stanna ett år till. Syftet med studien äratt undersöka förskollärares kunskaper och erfarenheter av ADD och skolmognad, närdet gäller övergången till förskoleklass från förskolan. De frågeställningar som studienska besvara är: Varför stannar barn kvar på förskolan ytterligare ett år? Hur insatta ärförskolläraren i ADD och skolmognad? Det socialpsykologiska angreppsättet somstudien teoretiskt förankras i behandlar om övergången varit mjuk eller hård och desspåverkan på barn när de ska gå från förskolan upp till förskoleklass. I den här studienhar författaren använt en kvantitativ forskningsmetod. Data har samlats in genom enenkätstudie som skickats ut per mail till över ett trettiotal förskollärare i vissa fall viarektorerna/cheferna samt till en sluten Facebook grupp med cirka 50 000förskollärare/lärare. Totalt bygger studien på 45 svar. Resultatet från enkätstudienindikerar att det händer då och då att barn får stanna kvar på förskolan ett år till. Dethuvudsakliga resultatet tyder på att det kan finnas en mängd olika omständigheter till attdet blir så. Orsaken kan vara att barnet inte varit skolmoget, varit född sent på året ellerhaft en eller flera diagnoser som gjort att man kommit fram till att det ändå vore bäst förbarnet att stanna kvar i förskoleverksamhet.
