Estabilidade da polpa de caju congelada obtida com o uso de conservantes, pasteurizada e concentrada / Physical and chemical stability of frozen cashew pulp obtained with the use of preservatives, pasteurized and concentrated

CoordenaÃÃo de AperfeiÃoamento de Pessoal de NÃvel Superior / O caju destaca-se dentre as espÃcies frutÃferas nativas do Nordeste, por possuir elevada potencialidade para o consumo in natura e processamento industrial. A polpa e o suco sÃo importantes componentes da dieta humana por serem considerados uma fonte natural de carboidratos, carotenoides, vitaminas e compostos fenÃlicos, substÃncias com alto potencial antioxidante, e tÃm despertado o interesse de diferentes grupos de pesquisa. Este trabalho teve como objetivo estudar a estabilidade da polpa de caju com conservantes, pasteurizada e concentrada congelada durante 12 meses de armazenamento, atravÃs de determinaÃÃes quÃmicas, fÃsico-quÃmicas e microbiolÃgicas e a elaboraÃÃo de nÃctar e suco tropical adoÃado, estudando a estabilidade sensorial dos mesmos durante 360 dias de armazenamento da polpa. As polpas nÃo apresentaram interaÃÃo significativa para pH, sÃlidos solÃveis, acidez titulÃvel e coordenada de cor a. O pH e os sÃlidos solÃveis variaram com o tempo mas mantiveram-se dentro dos padrÃes da legislaÃÃo. A acidez nÃo variou com o armazenamento. A coordenada de cor a* ajustaram ao modelo cÃbico. O Ãcido ascÃrbico variou de 190,65 a 308,45mg/100g na polpa com conservantes, 170,95 a 299,70 mg/100g na polpa pasteurizada e de 514,68 a 865,42mg/100g na polpa concentrada, valores estes bastante expressivos. Pigmentos escuros solÃveis, luminosidade, hue, croma, carotenoides, teor de polpa, aÃÃcares totais apresentaram interaÃÃo significativa e foram avaliados por regressÃo variando com o tempo de armazenamento, exceto a coordenada de cor b*. Os teores de aÃÃcares totais mantiveram-se em torno de 10,01 a 13,25% nas polpas com conservantes e pasteurizadas a 24 a 27% na polpa concentrada, sendo estes em sua maioria representados por aÃÃcares redutores. As anÃlises microbiolÃgicas confirmaram a eficÃcia das etapas de processamento, tratamento tÃrmico e concentraÃÃo na manutenÃÃo da qualidade microbiolÃgica, uma vez que nÃo foi observado crescimento de microrganismos durante o perÃodo de armazenamento. Os nÃctares e os sucos tropicais adoÃados de caju apresentam-se na faixa de aceitaÃÃo sensorial em todos os atributos avaliados. O congelamento das polpas de caju com conservantes, pasteurizada e concentrada à viÃvel uma vez que nÃo ocorrem perdas significativas na qualidade dos produtos. A polpa de caju concentrada apresentou as melhores notas nos atributos sensoriais avaliados. / The cashew (Anacardium occidentale, L.) has a great economic importance to the Northeast region, notably for the great acceptance by consumers, both for their sensory properties (color, aroma, flavor, texture) as for its nutritional and functional values. It stands out among the fruit species native to the Northeast because it has high potential for fresh consumption and industrial processing. The cashew and cashew apple juice are important components of the human diet because they are considered a natural source of carbohydrates, carotenoids, vitamins and phenolic compounds, substances with high antioxidant potential, and have attracted the interest of various research groups. This work aimed to study the stability of cashew apple pulp with preservatives, pasteurized and concentrated frozen storage for 12 months, through to chemical, physical-chemical and microbiological analysis. And production of cashew apple nectar and cashew apple sweetened tropical juice and sensorial stability during 360 days of storage of the pulp. The pulps showed no significant interaction for pH, soluble solids, acidity and color coordinated. The pH and soluble solids varied with time but remained within standards legislation. The acidity no change during the storage. The color coordinate set to the cubic model. The parameters vitamin C ranged from 190.65 to 308.45 in the pulp with preservatives, 170.95 to 299.70 in the pulp pasteurized and from 514.68 to 865.42 mg/100g in concentrated pulp, these values very expressive. Dark pigments soluble, brightness, hue*, chroma*, carotenoids, pulp content, total sugar showed significant interaction were assessed by regression with time varying storage except color coordinate b. The concentration of total sugars remained around 10.01 to 13.25% for pulps with preservatives and pasteurized,and to 24 to 27% in the concentrated pulp, which are mostly represented by sugars. The microbiological analyses confirmed the effectiveness of the processing, thermal treatment and concentration in the maintenance of the microbiological quality, once growth of microorganisms was not observed in the product during the storage period. The cashew apple nectars and juices sweetened have good acceptability in all sensory attributes. The frozen pulps cashew with preservatives, pasteurized and concentrated is viable since no significant losses occur in the quality of products.

Gestão do risco de granizo pelo seguro e outras alternativas: estudo de caso em pomares de maçã de Santa Catarina. / Hail risk management using insurance and other alternatives: case study on apple orchards in Santa Catarina, Brazil.

O prejuízo causado por chuvas de granizo é um dos principais problemas enfrentados pelos produtores de maçã no Brasil e em outros países. O trabalho apresenta, a partir de uma revisão abrangente da literatura e pesquisas de campo realizadas em região produtora do Estado de Santa Catarina, uma caracterização do problema e das alternativas existentes para a definição de estratégias ótimas para o gerenciamento do risco existente. As alternativas consideradas incluíram: seguro comercial, seguro mútuo, diversificação espacial, telas anti- granizo, foguetes anti- granizo e geradores de solo. O trabalho utiliza um modelo conceitual, especificado num diagrama de decisão e dados levantados na pesquisa, para evidenciar a relação qualitativa e quantitativa existente entre as diferentes alternativas consideradas e as variáveis mais relevantes para caracterização do problema. O modelo definido no diagrama de decisão foi implementado em software, em planilha eletrônica, de forma a facilitar o processo de seleção das melhores alternativas para gestão do risco de granizo, a partir da situação existente com relação a preços, custos e outras informações relevantes. Esse software foi utilizado em estudo de caso realizado junto a uma cooperativa de produtores de maçã de São Joaquim, SC, para a análise quantitativa das alternativas levantadas. O trabalho visa fornecer subsídios técnicos que auxiliem os agricultores no processo de tomada de decisão quanto à estratégia mais adequada para a administração do risco de chuvas de granizo em seus pomares, auxiliem as empresas de seguro na elaboração de novos contratos em suas carteiras agrícolas e, também, o governo no desenvolvimento de novas políticas voltadas ao setor agropecuário. / The damage caused by hailstorms is one of the most important problems faced by the apple producers, in Brazil, and in other countries. This work presents, from a literature review and local surveys within the apple producing region in Santa Catarina State, Brazil, a characterization of the hail risk problem and an evaluation of existing alternatives for risk management. The alternatives considered in the risk management process included: commercial insurance, mutual insurance, spatial diversification, antihail nets, hail rockets and ground burners. Besides, the research presents a conceptual model that uses decision diagrams to describe the qualitative relationship among the different alternatives for administration of the hail risk and the most important variables for the problem. The decision diagram guided the development of a software tool designed to help the selection of the best combination of alternatives for hail risk management. This software tool, implemented in a spreadsheet, was used in a case study involving an association of apple producers in São Joaquim, SC. The intention of this work is to aid apple producers in the selection of the most appropriate strategy for the administration of the risk of hailstorms in their orchards, aid the insurance companies in the design of new contracts in your agricultural portfolio and, also, aid the government in the development of new agricultural policies.

AgÃncia Nacional do PetrÃleo / CoordenaÃÃo de AperfeÃoamento de Pessoal de NÃvel Superior / In this work, the ethanol production from cashew bagasse was studied after acid followed by alkali pretreatment (CAB-OH) using the Separate Hydrolysis and Fermentation (SHF) and Simultaneous Saccharification and Fermentation (SSF) processes. In SHF process, the hydrolysate obtained from enzymatic hydrolysis of CAB-OH was used as carbon source for fermentation with different strains of Saccharomyces (S. cerevisiae CCA008, S. cerevisiae 01, S. cerevisiae 02 and Saccharomyces sp. 1238), Kluyveromyces (K. marxianus CCA510, CE025 and ATCC36907) and Hanseniaspora sp. GPBio03. The bioprocess was conducted at 30 ÂC and 50 g.L-1 initial glucose concentration. The K. marxianus ATCC36907 achieved ethanol concentration of 20 g.L-1 with consumption of all glucose in the hydrolysate. Similar results were obtained with Saccharomyces strains and higher ethanol concentration (23.43 g.L-1) was obtained by Saccharomyces sp. 1238. The maximum ethanol concentration of 24.54 g.L-1 was achieved by Hanseniaspora sp. GPBio03. Focused on further studies using SSF process, it was evaluated the temperature influence of thermotolerant yeast K. marxianus ATCC36907 in glucose and enzymatic hydrolysate from CAB-OH. The results showed that the temperature (30, 35, 40, 45 and 50 ÂC) did not affect the values of YE/G (0.45 to 0.46 gethanol/gglucose) using glucose as substrate. Moreover, the ethanol yields obtained with enzymatic hydrolysate were slightly influenced by temperature, 0.39 and 0.43 gethanol/gglucose were obtained at 30 and 40 ÂC, respectively. Based on this, the SSF of CAB-OH and K. marxianus ATCC36907 was conducted at 40 ÂC with cellulases from Celluclast 1.5L at 15 FPU/gcellulose. The highest ethanol concentration (24.90  0.89 g.L-1) was obtained with 76h of fermentation with 0.33 g.L-1.h-1, 0.34 gethanol/gglucose and 66.3% of productivity, YʹE/G and of ethanol efficiency, respectively. In enzymatic hydrolysis studies, the cellulase NS 22074 at 30 FPU/gcellulose without cellobiases supplementation resulted in glucose yield of 93.77  2.72% which is promising for studies of SSF with this enzyme complex. The temperature (40, 42 , 45 and 50 ÂC) influence in SSF process using microcrystalline cellulose, in contrast with SHF results, higher ethanol concentration, 19.86  0.32 g.L-1, was obtained at 40 ÂC. The SSF using CAB-OH, 30 FPU/gcellulose cellulases NS 22074 at 40 ÂC showed higher ethanol concentration of 37.35  0.64 g.L-1 at 80h, with productivity of 0.46 g.L-1.h-1. In this condition, there was an increase of YʹE/G from 0.34 to 0.49 gethanol/gglucose and the ethanol efficiency from 66.3% to 95.59% when compared to results obtained with SSF using Celluclast 1.5L. Based on the results of efficiency and ethanol yield (YʹE/G), the cashew apple bagasse showed as lignocelulose feedstock promising material for second generation ethanol production by SSF process using the yeast K. marxianus ATCC36907 and NS 22074 cellulases complex. / Nesse trabalho, estudou-se a produÃÃo de etanol de bagaÃo de caju apÃs prÃ-tratamento Ãcido seguido de Ãlcali (CAB-OH) atravÃs dos processos de FermentaÃÃo e HidrÃlise Separadas (SHF) e FermentaÃÃo e HidrÃlise SimultÃneas (SSF). No processo SHF, o hidrolisado obtido da hidrÃlise enzimÃtica de CAB-OH foi submetido à etapa de fermentaÃÃo com diferentes linhagens de Saccharomyces (S. cerevisiae CCA008, Saccharomyces sp. 1238, S. cerevisiae 01, S. cerevisiae 02), Kluyveromyces (K. marxianus CCA510, CE025 e ATCC36907) e Hanseniaspora sp. GPBio03. A fermentaÃÃo do hidrolisado foi conduzida a 30 ÂC com concentraÃÃo inicial de glicose de 50 g.L-1. ApÃs o screening de leveduras, a linhagem de K. marxianus ATCC36907 destacou-se com maior concentraÃÃo de etanol de 20 g.L-1 com consumo de toda glicose no hidrolisado. Resultados similares foram obtidos com Saccharomyces sp. 1238 e com a levedura isolada do caju (Hanseniaspora sp. GPBio03) com maiores concentraÃÃes de etanol de 22,41 g.L-1 e 24,54 g.L-1, respectivamente. Com o propÃsito de estudos posteriores de SSF, avaliou-se a influÃncia da temperatura da levedura termotolerante K. marxianus ATCC36907 em glicose PA e hidrolisado enzimÃtico de CAB-OH. Os resultados mostraram que para a glicose PA, a variaÃÃo da temperatura (30, 35, 40, 45 e 50 ÂC) nÃo influenciou nos valores de conversÃo de glicose em etanol (YE/G) obtendo-se valores na faixa de 0,45-0,46 getanol/gglicose. Por outro lado, os resultados de YE/G em hidrolisado enzimÃtico foram ligeiramente influenciados pela temperatura, obtendo-se 0,39 getanol/gglicose a 30ÂC e 0,43 getanol/gglicose a 40 ÂC. Em seguida, realizou-se a SSF de CAB-OH com K. marxianus ATCC36907 a 40 ÂC e celulases de Celluclast 1.5L a 15 FPU/gcelulose. A maior concentraÃÃo de etanol (24,90  0,89 g.L-1) foi obtida em 76h de fermentaÃÃo com produtividade de 0,33 g.L-1.h-1, conversÃo de glicose em etanol (YʹE/G) de 0,34 e eficiÃncia de produÃÃo de etanol de 66,3%. Contudo, visando aumentar a produÃÃo de etanol em estudos posteriores de SSF, realizou-se o estudo de hidrÃlise enzimÃtica com outros complexos de celulases (NS 22074) e celobiases (NS 50010). Os resultados de hidrÃlise enzimÃtica mostraram que a atividade de celulases NS 22074 a 30 FPU/gcelulose sem suplementaÃÃo de celobiase resultou no rendimento de glicose de 93,77  2,72% sendo resultado promissor para estudos de SSF com esse complexo enzimÃtico. Nos ensaios de SSF com celulases do complexo NS 22074, inicialmente realizou-se o estudo da temperatura (40, 42, 45 e 50 ÂC) com K. marxianus ATCC36907 utilizando celulose microcristalina; e, em contrapartida com os resultados SHF, na temperatura de 40 ÂC foi obtida a maior concentraÃÃo de etanol de 19,86  0,32 g.L-1, em 72h de fermentaÃÃo. Diante desses resultados, realizou-se o processo de SSF de CAB-OH nas seguintes condiÃÃes: 40 ÂC de temperatura e 30 FPU/gcelulose do complexo de celulases NS 22074. A maior concentraÃÃo de etanol (37,35  0,64 g.L-1) foi obtida em 80h de fermentaÃÃo, com produtividade de 0,46 g.L-1.h-1. Diante desses resultados, observa-se que a mudanÃa do complexo enzimÃtico de Celluclast 1.5L para NS 22074 proporcionou o aumento no valor de YʹE/G de 0,34 getanol/gglicose para 0,49 getanol/gglicose e no rendimento de etanol de 66,3% para 95,59%, o que torna o bagaÃo de caju prÃ-tratado promissor como matÃria-prima para produÃÃo de etanol de segunda geraÃÃo por processo SSF utilizando a levedura K. marxianus ATCC36907.

Processo de desenvolvimento das inovações na cadeia produtiva da maçã brasileira

A inovação exerce importância no negócio das empresas, compreende diferentes aspectos e contribui de formas diversas para o seu crescimento e desenvolvimento. Dentre os aspectos que podem ser levados em consideração para as inovações estão os aspectos internos relacionados com o contexto da organização e os aspectos externos que envolvem todo o ambiente em que a organização está inserida. Acredita-se que estes aspectos exercem influência tanto na decisão para o desenvolvimento das inovações quanto para a aplicação dos produtos ou processos de inovação no mercado. Este estudo teve como objetivo identificar como e porque as inovações emergem na cadeia produtiva da maçã brasileira e quais os resultados obtidos a partir das inovações implementadas nos segmentos produtores e packinghouses. Os procedimentos metodológicos empregados têm como estratégia de pesquisa o estudo de caso, o qual foi efetivado através das abordagens qualitativa e quantitativa de pesquisa. Em relação aos objetivos o estudo teve caráter exploratório, descritivo e explicativo. A etapa qualitativa foi executada através da pesquisa exploratória com especialistas da cadeia produtiva da maçã brasileira para identificar as inovações que foram implementadas e também para classificá-las de acordo com o encadeamento do anel tetralógico. Dentre os principais resultados obtidos ressalta-se a identificação dos aspectos que compuseram o framework e serviram de norteador para a realização da pesquisa qualitativa. Destaca-se também o encadeamento obtido entre os aspectos identificados de forma empírica e a junção com as teorias utilizadas, em especial os pressupostos de Edgar Morin, através da teoria da complexidade. A etapa quantitativa foi efetivada através do método estatístico, por meio da análise fatorial, cuja análise apresentou a identificação dos resultados que as inovações apresentam para os fatores sociais, econômicos, organizacionais, políticos e mercadológicos na amostra de produtores e packing-houses da cadeia produtiva da maçã brasileira definidas para esta investigação. Na análise quantitativa merece destaque a apresentação através de estatística descritiva da caracterização das amostras, bem como os processos de inovação que cada amostra tem implementado. A análise fatorial permitiu a apresentação das variáveis destacadas para cada fator nas quatro amostras (produtores e packing-houses de Santa Catariana e Rio Grande do Sul) individualmente e também para toda a cadeia incluindo as quatro amostras. Através da análise de regressão múltipla identificou-se a percepção sobre o impacto das inovações para toda cadeia produtiva da maçã brasileira, incluindo os produtores e packing-houses dos estados de Santa Catarina e Rio Grande do Sul. / Innovation plays importance in business enterprises, comprises different aspects and contributes in different ways to their growth and development. Among the aspects that can be considered for the innovations are the internal aspects related to the context of the organization and external aspects that involve the whole environment in which the organization operates. It is believed that these aspects influence both the decision for the development of innovations in the application for product or process innovation in the market. This study aimed to identify how and why innovations emerge in the productive chain of Brazilian apples and what the results obtained from the innovations implemented in the sectors producing and packing houses. The methodological procedures have employed as a research strategy case study, which was effected by means of qualitative and quantitative approaches to research. In relation to the objectives the study had an exploratory, descriptive and explanatory. The qualitative analysis was performed through exploratory research with experts in the productive chain of Brazilian apples to identify innovations that have been implemented and also to classify them according to the chain ring tetralogical. Among the main results emphasize the identification of aspects that compose the framework and provided a guideline for conducting qualitative research. Also noteworthy is the chain obtained among those identified empirically and the junction with the theories used, in particular the assumptions of Edgar Morin, by complexity theory. The quantitative stage was effected through the statistical method, by means of factor analysis, which analysis showed identification results show that the innovations to the social, economic, organizational, political and market in the sample of producers and packing-houses of the production chain Brazilian apple defined for this investigation. The quantitative analysis deserves the presentation through descriptive statistics of the characterization of the samples, as well as the processes of innovation that each sample has implemented. A factor analysis of the presentation highlighted variables for each factor in the four samples (producers and packing houses of Santa Catarina and Rio Grande do Sul) individually and for the entire chain including the four samples. Through multiple regression analysis identified the perception of the impact of innovations for the entire production chain of Brazilian apples, including growers and packing houses in the states of Santa Catarina and Rio Grande do Sul.

Analyse des conflits et synergies entre services écosystémiques multiples en vergers de pommiers / Analyzing multiple ecosystem services in apple orchards

Le concept de « service écosystémique », en plein essor depuis la publication du Millenium Ecosystem Assessment en 2005, a permis de souligner l'importance des performances non-marchandes des écosystèmes. En arboriculture, assurer une bonne productivité tout en préservant les ressources naturelles et la santé humaine est aujourd’hui un défi majeur, qui peut être analysé sous l’angle des services écosystémiques. Quels sont ces services au sein d’un verger de pommiers ? Comment les analyser ? Quels sont les liens - conflits et synergies - entre services écosystémiques multiples et comment les systèmes de culture modifient-ils les profils de services multiples? Ce travail de thèse vise à répondre à ces questions avec une démarche novatrice combinant mesures expérimentales, modélisation et analyse statistique. Suite à une analyse bibliographique des services écosystémiques pouvant être délivrés dans des vergers, cinq services ont été sélectionnés. Il s’agit de la production de fruits, de la disponibilité de l’azote dans le sol, de la régulation du climat reposant sur la prévention de la dénitrification de l’azote et sur la séquestration du carbone, de la maintenance et de la régulation du cycle de l’eau y compris de sa qualité, et de la régulation des bioagresseurs. Conjointement à ces services nous avons considéré les nuisances environnementales dues aux pesticides. Pour chaque service nous avons identifié les fonctions écosystémiques sous-jacentes ainsi que les pratiques agricoles et les conditions pédoclimatiques ayant un impact sur ces fonctions. Les services et fonctions ont été décrits par un ou plusieurs indicateurs et quantifiés à l’aide de modèles dans les cas suivants : (i) neuf systèmes de culture réels sur deux dispositifs expérimentaux dans le sud-est de la France et (ii) 150 systèmes de culture fictifs conçus à partir de la combinaison de cinq leviers de pratiques et de leurs modalités, dans des conditions pédoclimatiques identiques. Les deux modèles utilisés ont été STICS, un modèle générique de simulation du fonctionnement du sol et des cultures sous l’influence des pratiques, qui a nécessité une paramétrisation et une évaluation sur pommier à partir de mesures expérimentales, et IPSIM, une plateforme de modélisation simulant les effets des pratiques et des conditions du milieu sur les dégâts aux cultures causés par les bioagresseurs. IPSIM a été paramétré sur pommier, sur la base d’une importante étude bibliographique et à dires d’experts. Les simulations des modèles ont fait l’objet d’analyses statistiques simples dans le cas des systèmes de culture réels, et d’analyses multivariées à deux tableaux (analyse en composantes principales avec variables instrumentales) pour les systèmes de culture fictifs. Pour les neufs systèmes de culture réels, 14 liens importants entre services écosystémiques ont été identifiés, notamment des conflits comme celui entre la prévention de la dénitrification ou de la lixiviation de l’azote et la disponibilité de l’azote dans le sol à court terme, et des synergies, comme celle entre l’humidité du sol ou la séquestration du carbone et la disponibilité de l’azote dans le sol à court terme. Ces liens entre services sont expliqués par les fonctions écosystémiques sous-jacentes. La comparaison de profils de services entre systèmes a mis en évidence l’impact de pratiques agricoles sur certains services. Ainsi, sur un même site, une forte densité de plantation augmente la production de fruits et la séquestration du carbone. La fertilisation exclusivement sous une forme organique diminue la production de fruits via un stress azoté mais diminue également la lixiviation. Par ailleurs, les profils de services sont fortement influencés par les caractéristiques pédoclimatiques de chaque site. Ces résultats confortent la nécessité d’une prise en compte explicite de l’interdépendance ‘pratiques x conditions du milieu’ pour analyser les services. / The concept of « ecosystem service », which has been used increasingly since the publication of the Millennium Ecosystem Assessment in 2005, has highlighted the importance of ecosystem’s non-marketed performances. In orchards, ensuring high productivity while preserving natural resources and human health has become a real challenge that could be analyzed with the concept of ecosystem service. Which ecosystem services are delivered in an apple orchard? How to analyze them? What are the relationships - conflicts or synergies – among multiple ecosystem services and how do cropping systems change multiple ecosystem service profiles? This PhD work aims at answering those questions with an innovative approach combining experimental measures, modeling and statistical analysis.Based on a literature review of ecosystem services in orchards, five services were selected: fruit production, nitrogen availability in soil, climate regulation based on the prevention of nitrogen denitrification and on carbon sequestration, maintenance and regulation of water cycle, including water quality, and pest control. We also considered the environmental disturbances caused by the use of pesticides. For each service, we identified the underlying ecosystem functions as well as the agricultural practices and soil and climate conditions affecting these functions. Services and functions were described by one or multiple indicators and quantified using models in the case of (i) nine existing cropping systems on two experimental sites in southeastern France differing in terms of soil and climate conditions, and (ii) 150 virtual cropping systems designed out of the combination of five major agricultural practice levers and their modalities, in identical soil and climate conditions. The two models used were STICS, a generic soil-crop simulation model under the influence of practices which required a parameterization and an evaluation on apple orchards based on experimental measures, and IPSIM, a generic modeling framework simulating the impacts of agricultural practices and local conditions on crop injuries caused by pests. IPSIM was parameterized on apple orchards, based on an important literature review and expert opinions. Model simulations were analyzed with simple statistics in the case of the nine existing cropping systems and with two-table multivariate analyses (principal component analysis with instrumental variables) for virtual cropping systems.Concerning the existing cropping systems, 14 important relationships were identified among ecosystem services, especially conflicts, like the one between nitrogen denitrification or leaching prevention and soil nitrogen availability on the short term, and synergies such as the one between soil humidity or carbon sequestration and nitrogen availability on the short term. These relationships are explained by the underlying ecosystem functions. Comparing service profiles among cropping systems highlighted the impacts of agricultural practices on some services. That way, on a same site, a high planting density increases fruit production and carbon sequestration. An exclusively organic fertilization decreases fruit production through nitrogen stress but also nitrogen leaching in drained water. Furthermore, service profiles are strongly influenced by the soil and climate conditions of each site. These results strengthen the need to explicitly consider the ‘agricultural practices x soil and climate conditions’ interdependence in order to analyze ecosystem services. The results obtained with the virtual cropping systems simulations confirmed those of the existing ones and gave precision on the impacts of fertilization, irrigation and pest control for codling moth, rosy apple aphid and apple scab on ecosystem functions and services.

Os compostos fenólicos são substâncias amplamente distribuídas no reino vegetal, em particular nas frutas e em outros vegetais. Estes compostos, destacando-se flavonoides e os ácidos fenólicos, devido à estrutura molecular, podem apresentar a capacidade de inibir processos oxidativos. Além do mais, estão relacionados com a redução de risco de doenças crônicas não transmissíveis tais como: cardiovasculares, câncer, aterosclerose, entre outras. Considerando a elevada produção de caju em território brasileiro e a possibilidade da existência de compostos com potencial antioxidante no pedúnculo de caju , este trabalho teve por objetivo avaliar quantitativa e qualitativamente os compostos fenólicos, particularmente os ácidos fenólicos, e identificar a participação destes em processos metabólicos do organismo animal. Foram caracterizados quimicamente três clones distintos de pedúnculos de caju (CCP-76, CCP-09, BRS-189 e CCP-76 tratado) e na análise química, apresentaram um elevado teor de ácidos graxos monoinsaturados, predominando o ácido oléico, e de fenólicos totais. Os ácidos fenólicos identificados foram: gálico, protocatecuíco, p-cumárico, ferúlico, caféico e salicílico. Foram obtidos extratos aquoso (EAq) e alcoólico (EAlc) e frações de ácidos fenólicos a partir dos pedúnculos e, avaliados em sistemas modelo β-caroteno/ácidolinoléico e em Rancimat. As frações de ácidos fenólicos exibiram expressiva atividade antioxidante no primeiro sistema e os extratos, no segundo, demonstraram fatores de proteção superior ao antioxidante sintético BHT. Pôde-se também verificar a capacidade antioxidante dos extratos e frações do clone CCP-76 no sistema de varredura do radical DPPH. Em ensaio experimental com ratos, em condição normal, foi administrado EAq (80 e 240 mg/kg, v.o.) ou fração de ácidos fenólicos livres (40 e 120 mg/kg, v.o.) obtidos do pedúnculo de caju CCP-76. Neste estudo, não se observou potencialização de todos os antioxidantes enzimáticos (superóxido dismutase, catalase e glutationas peroxidase e redutase), contudo pôde-se verificar a redução dos níveis de lipoperoxidação no tecido cerebral dando indícios de aumento do estado antioxidante nos animais. Também foi avaliado o potencial antioxidante do EAq e da fração de ácidos fenólicos livres sobre o dano hepático em ratos tratados com tetracloreto (CCl4) de carbono. A administração deste teve seu efeito corroborado pela avaliação dos parâmetros bioquímicos, ou seja, aumento exacerbado das enzimas hepáticas no plasma: alanina transaminase (ALT) e aspartato transaminase (AST); decréscimo da atividades da enzimas antioxidantes no fígado e elevação da produção de peróxidos lipídicos no tecido hepático. Nos ratos que receberam EAq (480 mg/kg, v.o.) não se observou alteração comparando-os aos animais tratados apenas com CCl4 . No entanto, a administração de fração de ácidos fenólicos livres, nas duas doses (40 e 120 mg/kg, v.o.), evidenciou pronunciado efeito contra a lesão hepática, com níveis reduzidos de AL T e AST plasmáticas, aumento da atividade das enzimas antioxidantes no fígado e prevenindo a lipoperoxidação hepática mediada pelo radical CCl3• a partir do CCl4. Estudos histológicos do tecido hepático confirmaram as avaliações bioquímicas exibindo preservação tecidual, supressão de degeneração vacuolar macro e microgoticular e de sinais necróticos nos ratos tratados com a fração de ácidos fenólicos livres do pedúnculo de caju. / Phenolic compounds are widely distributed in the plant kingdom, particularly fruits and vegetables. Due to their chemical structure, these compounds, in particular flavonoids and phenolic acids, are able to inhibit oxidative processes. Furthermore, can be used to reduce the risk of non-transmissible chronic diseases such as cardiovascular diseases, cancer and atherosclerosis. Taking into consideration the large production of cashew in Brazil and the possible existence of potentially antioxidant compounds present in the cashew apples, the aim of this study was to quantitatively and qualitatively evaluate the presence of phenolic compounds in cashew apple, particularly phenolic acids, and identify their role in metabolic processes in animals. The cashew apples of three distinct clones (CCP-76, CCP-09, BRS-189 and CCP-76 (processed)) were studied. The determination of fatty acids yielded a high concentration of monounsaturated fatty acids, mainly oleic acid, and of total phenolic compound. The phenolic acids found were: gallic, proteocatechuic, p-cumaric, ferulic, caffeic and salicylic acids. Both aqueous (EAq) and ethanolic (EAlc) extracts and phenolic acid fractions were obtained from the cashew apples and were evaluated in a β-carotene/linoleate model system and Rancimat test. The phenolic acid fractions presented an expressive antioxidant activity in the β-carotene/linoleate model system and the extracts, by the Rancimat test presented a protection factor higher than that of antioxidant additive, BHT. We also observed the antioxidant capacity of the extracts and fractions of the CCP-76 clone in the DPPH radical scavenging assay. In an experimental assay with rats, the EAq (80 and 240 mg/kg) or the free phenolic acid fraction (40 and 120 mg/kg) obtained from the cashew apple of CCP-76 clone was administered via the oral route. In this study, the enhancement of enzymatic antioxidants (superoxide dismutase, catalase, glutathione peroxidase and reductase) was not observed, nevertheless, a decrease in the amount of lipoperoxidation in the brain tissue was observed, suggesting that the ingestion of cashew might increase the antioxidative state in animals. Also, the antioxidant activity of EAq and of the free phenolic acid fraction from the cashew apple of CCP-76 clone was verified on the liver damage induced by carbon tetrachloride. The liver damage caused by the administration of carbon tetrachloride was detected by biochemical parameters, namely, the increase in the serum concentrations of alanine transaminase (ALT) and aspartate transaminase (AST) as well as a decrease in the activities of antioxidant enzymes and an increase in peroxidation in the liver. Rats who received EAq (480 mg/kg, p.o.) did not present alterations in any of the parameters evaluated, compared to the animals treated with carbon tetrachloride. On the other hand, the administration of free the phenolic acid fraction in doses of 40 and 120 mg/kg, p.o., had a pronounced effect in protecting against hepatic lesion, which was evidenced by the decrease in plasma ALT and AST, enhancing the activity of antioxidant enzymes and preventing lipoperoxidation mediated by the CCl3• radical generated by carbon tetrachloride. Histological studies were able to confirm the biochemical alterations observed in that the liver tissue obtained from rats treated with phenolic acid fractions extracted from cashew apple of CCP-76 clone presented a preserved tissue structure and suppression of macro and microgoticular vacuolar degeneration as well as of signs of necrosis.

Third-party authentication services are becoming more common since it eases the login procedure by not forcing users to create a new login for every website thatuses authentication. Even though it simplifies the login procedure the users still have to be conscious about what data is being shared between the identity provider (IDP) and the relying party (RP). This thesis presents a tool for collecting data about third-party authentication that outperforms previously made tools with regards to accuracy, precision and recall. The developed tool was used to collect information about third-party authentication on a set of websites. The collected data revealed that third-party login services offered by Facebook and Google are most common and that Twitters login service is significantly less common. Twitter's login service shares the most data about the users to the RPs and often gives the RPs permissions to perform write actions on the users Twitter account.  In addition to our large-scale automatic data collection, three manual data collections were performed and compared to previously made manual data collections from a nine-year period. The longitudinal comparison showed that over the nine-year period the login services offered by Facebook and Google have been dominant.It is clear that less information about the users are being shared today compared to earlier years for Apple, Facebook and Google. The Twitter login service is the only IDP that have not changed their permission policies. This could be the reason why the usage of the Twitter login service on websites have decreased.  The results presented in this thesis helps provide a better understanding of what personal information is exchanged by IDPs which can guide users to make well educated decisions on the web.

Stanovení autenticity potravin rostlinného původu pomocí molekulárních metod / Determination of athenticity of plant foods by molecular techniques

The aim of presented diploma thesis was to determination of authenticity of fruit baby foods for early infant feeding using molecular methods. In the experimental part, isolation kit was used for isolation of plant DNA from fruits (strawberry, apricot, raspberry, apple) and from six commercial fruit products for children. Isolated DNA was characterized and verified using PCR methods with primers specific for plant rDNA (ITS2). Specific primer pairs were designed to amplify DNA for the detection of one fruit species. Primer specificity was assessed with four fruit species. A mixture of fruit puree from the two fruits was used to determine the sensitivity of the multiplex PCR assay. Six commercial fruit products were evaluated to verify the applicability of the multiplex PCR assay. The methodology of molecular detection of fruit DNA by qPCR and multiplex qPCR (duplex) includes approaches, which enable to detect two fruits (strawberry-raspberry, apricot-apple) in one reaction and thus reduces time and money requirements.

This diploma thesis deals with a problem of spores venturia inaequlis recognition. These spores are captured on a special tape which is then analyzed using a microscope. The tape can be analyzed by a laboratorian or by the program Sporedetect v3. This program provides functions for complete picture processing and object recognition. In this diploma thesis, there are also described ways to automatically control a sliding stage of a microscope utilizing motorized translation stages and linear actuators. The information about automatic control of a microscope stage was obtained from catalogues of the companies Standa and Edmundoptics.

Valorization of apple by-products by the extraction and purification of polyphenols : impact of the ultrasound / Valorisation des sous-produits de pomme par extraction et purification de polyphénols : impact des ultrasons

Cette thèse porte sur l'intensification de l'extraction de polyphénols à partir de produits à base de pomme (chair, peau et marc) par ultrasons (US) et sur la purification d'extraits de peau de pomme par adsorption/désorption et technologie membranaire. L'extraction sélective des polyphénols issus des produits de la pomme a été analysée. Les données obtenues ont démontré la possibilité d’une régulation fine de l’extraction sélective de la matière soluble, de la catéchine et des composés polyphénoliques totaux en utilisant différentes températures, protocoles UAE, mélanges éthanol/aqueux. La sélectivité de l'extraction de la catéchine dépendait également du type de tissu (chair, peau ou marc) et de la variété de pomme (verte ou rouge). Le phénomène de cavitation généré par les ultrasons pourrait augmenter l'extraction de composants précieux des pelures de fruits en endommageant les membranes cellulaires des échantillons et en accélérant ainsi le transfert de chaleur et de masse. D’autre part, les solvants eau-gaz pourraient améliorer l'efficacité d'extraction des polyphénols des peaux de pomme en renforçant le phénomène de cavitation généré par les ultrasons. L'efficacité de la purification de polyphénols d'extraits de pelure de pomme par adsorption/désorption assisté par ultrasons (adsorbant polyaromatique Amberlite XAD-16) et par électrofiltration sur membrane a été mise en évidence. Les données obtenues ont démontré que la sonication facilitait significativement la cinétique d’adsorption, la capacité d’adsorption accrue et augmentation de l’énergie d’activation de l’adsorption des polyphénols. En outre, le taux de désorption a été positivement affecté par la sonication au cours de l'étape d'adsorption. Par ailleurs, les résultats ont montré que l’électrofiltration sur membrane permettait de purifier les polyphénols dans l’espace anodique (+) et d’obtenir un volume plus important de filtrats. / This thesis focuses on the intensification of polyphenols extraction from apple products (flesh, peel, and pomace) by ultrasound (US) and the purification of apple peel extracts by adsorption/desorption and membrane technology. The selective extraction of phenolic contents from apple products has been analyzed. The obtained data evidenced the possibility of fine regulation of selective extraction of soluble matter, catechin and total polyphenolic compounds using different temperatures, ultrasound-assisted extraction (UAE) protocols, ethanol/aqueous mixtures. The selectivity of catechin extraction was also depended on the type of the tissue (flesh, peel or pomace) and apple variety (green or red). The cavitation phenomenon generated by ultrasound could increase extraction of valuable components from fruit peels by damaging cell membranes of samples and accelerating heat and mass transfer by disrupted cell walls of samples. Meanwhile, the gas water solvents could enhance the extraction efficiency of polyphenols and antioxidant activity from apple peels by enhancing cavitation phenomenon generated by ultrasound. The efficiency of polyphenols purification from apple peel extracts with adsorption/desorption process by ultrasound treatment with the polyaromatic amberlite adsorbent XAD-16 and with membrane electro-filtration were studied. The obtained data demonstrated that the sonication significantly facilitated adsorption kinetics and increased activation energy of polyphenols adsorption. In addition, the desorption ratio was positively affected by the sonication during the adsorption step. On the other hand, the results demonstrated that the membrane electro-filtration allowed the purification of polyphenols in the anode (+) space and obtaining larger volume of filtrates.
