Development of transaminases for the synthesis of enantiomerically pure chiral amines

Enantiomerically pure amines have a variety of industrial applications. They are valuable components within the pharmaceutical and agrochemical industry, resolving agents for separation of racemic mixtures by dimeric salt formation and ligands for transition metal catalysts and chemocatalysts. Biocatalysis is increasingly seen as the method of choice for the synthesis of chiral amines. New commercial enzymes being readily available and new screening/evolution technologies allow for enzyme optimisation towards a set of required conditions for chiral amine synthesis. Transaminases are a class of pyridoxal 5’-phosphate (PLP) dependant enzymes that catalyse the reversible transfer of ammonia from an amine donor (e.g. alanine) to a keto acceptor (e.g. acetophenone), allowing the potential for asymmetric methodologies. Transaminases are already well established for the industrial production of α-amino acids and now that research scientists have dealt with some of the problems with equilibrium and substrate/product inhibition, they are being investigated for the industrial application of chiral amines. A multi-enzyme kinetic assay has been utilised for characterisation of newly identified transaminase enzymes in solution phase. Identified transaminases that showed desirable characteristics were cloned and expressed and utilised in the synthesis of amines of interest to industry. Dual enzyme cascade reactions utilising transaminases and either galactose oxidase from Fusarium sp. or monoamine oxidase from Aspergillus niger were used to produce a number of primary and secondary amines in high e.e. and conversion.

Purification and properties of dolphin muscle glutamate-oxalacetate and glutamate-pyruvate transaminases and their possible roles in the energy metabolism of diving mammals

Mitochondrial and supernatant glutamate-oxalacetate transaminases (EC 2.6.1.1) and supernatant glutamate-pyruvate transaminase (EC 2.6.1.2) were purified 89, 204 and 240-fold respectively, from dolphin muscle. Starch gel electrophoresis of crude and purified perparations revealed that all three enzymes exist as single forms. Km values of a-ketoglutarate, alanine, pyruvate and glutamate for the glutamate-pyruvate transaminase were 0.45, 8.2, 0.87 and 15 mM, respectively. For the glutamate-oxalacetate transaminases, the Km values of a-ketoglutarate, aspartate, oxalacetate and glutamate were 0.76, 0.50, 0.10 and 9.4 mM, respectively, for the mitochondrial form and 0.13, 2.4, 0.06 and 3.2 mM, respectively, for the supernatant form. In all cases, as the assay pH was decreased from pH 7.3, the Km values of the a-keto acids decreased while those of the amino acids increased. This caused the apparent equilibrium constants for the glutamate-oxalacetate transaminases to remain independent of pH. These values were 9.2 and 6.8 for the mitochondrial and supernatant forms, respectively where K'eq = [asPartate][α-ketoglutarate]/[glutamate][oxalacetate]. Studies of the inhibition of the glutamate-oxalacetate transaminases by dicarboxylic acids indicated that these enzymes may be controlled by pools of metabolic intermediates. Three key roles are suggested for the transminases in the energy metabolism of the diving mammal. First, it is believed that a combined action of the transaminases could enhance energy production during hypoxia by providing (1) fumarate from aspartate for the ATP producing reversal of succinate dehydrogenase and (2) α-ketoglutarate from glutamate for the GTP producing succinyl thiokinase reaction. Next, diving mammals probably accumulate more NADH than other mammals during hypoxia. The glutamate-oxalacetate transaminases seem particularly well suited for restoring redox balance via the malate-aspartate cycle after aerobic metabolism is resumed. Finally, since migrating divers oxidize large amounts of stored fats, the combined reactions of the transaminases could be instrumental in providing increased supplies of oxalacetate to condense with the fat derived acetyl CoA in the citrate synthase reaction. / Science, Faculty of / Zoology, Department of / Graduate

Neste trabalho, foi avaliada a resolução cinética de ésteres carboxílicos β-borilados, catalisada por lipases, através de hidrólise e transesterificação. Foram sintentizados ésteres contendo diferentes cadeias carbônicas. A hidrólise enantiosseletiva catalisada por CAL-B forneceu o correspondente ácido carboxílico β-borilado com excesso enantiomérico moderado (e.e. 62%), enquanto os ésteres β-borilados metílico e etílico foram recuperados com excelente pureza enantiomérica (e.e.>99%). A reação de transesterificação enantiosseletiva entre ésteres carboxílicos β-borilados e álcoois, catalisada por CAL-B, demonstrou uma alta seletividade somente quando etanol e (β-pinacolilboronato)-butanoato de metila foram usados como substratos, levando a formação de (β-pinacolilboronato)-butanoato de etila enantioenriquecido (e.e.>99%). Entretanto, ésteres contendo longas cadeias carbônicas ligadas à carboxila não sofreram as reações desejadas, mesmo em temperaturas mais elevadas. Adicionalmente, foi investigada a reação de aminação redutiva assimétrica, catalisada por ω-transaminases, de cetonas pró-quirais contendo grupos borônicos (ácido, éster ou trifluoroborato de potássio). Após as sínteses de cetonas aromáticas e alifáticas, os melhores resultados para a biotransformação foram obtidos com ω-transaminases (R)-seletivas de Arthrobacter sp. e Aspergillus terreus e (S)-seletiva de Chromobacterium violaceum. Sob as condições otimizadas, foram obtidas altas conversões (94%) com perfeitas enantiosseletividades (e.e.>99%), proporcionando assim uma nova via de acesso a aminas (R) ou (S) contendo boro em condições reacionais suaves. / In this work, the kinetic resolution of β-borylated carboxylic esters via lipase-catalyzed hydrolysis and transesterification reactions was evaluated. Amine Transaminases in Multi-Step One-Pot Reactions

Amine transaminases are enzymes that catalyze the mild and selective formation of primary amines, which are useful building blocks for biologically active compounds and natural products. In order to make the production of these kinds of compounds more efficient from both a practical and an environmental point of view, amine transaminases were incorporated into multi-step one-pot reactions. With this kind of methodology there is no need for isolation of intermediates, and thus unnecessary work-up steps can be omitted and formation of waste is prevented. Amine transaminases were successfully combined with other enzymes for multi-step synthesis of valuable products: With ketoreductases all four diastereomers of a 1,3-amino alcohol could be obtained, and the use of a lipase allowed for the synthesis of natural products in the form of capsaicinoids. Amine transaminases were also successfully combined with metal catalysts based on palladium or copper. This methodology allowed for the amination of alcohols and the synthesis of chiral amines such as the pharmaceutical compound Rivastigmine. These examples show that the use of amine transaminases in multi-step one-pot reactions is possible, and hopefully this concept can be further developed and applied to make industrial processes more sustainable and efficient in the future. / <p>QC 20170113</p>

O primeiro capítulo apresenta o isolamento, identificação e a triagem enzimática de bactérias heterotróficas isoladas de rejeitos de mineração de cobre. A partir destas amostras foi possível isolar 189 bactérias, as quais apresentaram uma diversificada atuação catalítica. As bactérias isoladas foram identificadas por MALDI-TOF e por sequenciamento do gene RNAr 16S sendo encontrados diversos gêneros como Bacillus, Acinetobacter, Pseudomonas, Delftia, Stenotrophomonas, Hydrogenophaga, Rhodococcus, entre outros. O segundo capítulo apresenta o estudo do potencial catalítico de uma nova epóxido hidrolase de Aspergillus brasiliensis CCT1435, recombinante e expressa em E. coli. Esta EH é ativa em uma ampla faixa de pH e temperatura, apresentando um desempenho ótimo em pH 6,0 e 30 °C. Este trabalho ainda permitiu uma avaliação detalhada da aplicação biocatalítica desta EH na hidrólise do óxido de estireno em meio aquoso e bifásico. Por fim, o terceiro capítulo apresenta resultados preliminares envolvendo um processo quimio-enzimático para a produção de alcalóides pirrolidínicos a partir das respectivas 1,4-dicetonas. Esta metodologia é promissora uma vez que diversas 1,4-dicetonas foram convertidas nas suas respectivas iminas cíclicas, sendo que a subsequente etapa de redução com NaBH3CN promoveu a formação das pirrolidinas de interesse. Portanto, esta tese de doutorado apresenta um estudo amplo e diversificado envolvendo processos biocatalíticos aplicados em uma série de reações de interesse biotecnológico / Abstract: Biocatalytic processes have been addressed in this thesis from the microorganism screening for the selection of suitable biocatalysts to the use of isolated enzymes in biotechnological reactions. The first chapter presents the isolation, identification and enzymatic screening of heterotrophic bacteria isolated from copper mine drainages. From these samples, 189 bacteria were isolated, which showed diverse catalytic activities. The isolated bacteria were identified by MALDI-TOF and 16S rRNA gene sequencing and several genera were found: Bacillus, Acinetobacter, Pseudomonas, Delftia, Stenotrophomonas, Hydrogenophaga, Rhodococcus, among others. The second chapter presents the catalytic potential of a new epoxide hydrolase from Aspergillus brasiliensis CCT1435 (AbEH), recombinant and expressed in E. coli. This EH is active in a wide pH and temperature range, with a great performance at pH 6.0 and 30 °C. This work also presents the AbEH biocatalytic application for the styrene oxide hydrolysis in aqueous and biphasic media. Finally, the third chapter presents preliminary results involving a chemo-enzymatic method for the production of pyrrolidine alkaloids from the corresponding 1,4-diketones. This approach is promising since several 1,4-diketones were converted to their respective cyclic imines, and the subsequent reduction with NaBH3CN promoted the pyrrolidines formation. EFFECT OF ESTRADIOL SUPPLEMENTATION ON BLOOD ESTRADIOL AND METABOLITE LEVELS, AND HEPATIC PROTEIN EXPRESSION, IN GROWING, MATURE, AND SENESCENT BEEF CATTLE

Estradiol (Compudose®, COM) implants are extensively used in beef cattle production systems to alter body composition and feed efficiency. Little information exists about the physiological mechanisms affected by COM treatment in growing, mature, and senescent female cattle. Moreover, no reports describe the level of blood estradiol resulting from COM treatment. The effect of COM on levels of plasma estradiol and blood metabolites and proteins, and relative content of glutamine synthetase (GS) and other amino acid nitrogen-metabolizing enzymes in liver tissue, was studied using three experimental models relevant to cow-calf production regimens: senescent cows (Trial 1), young mature (young) versus senescent (old) cows (Trial 2), and growing heifers (Trial 3). In Trial 1, plasma estradiol concentrations were 222 % more after 14 and 28 d in COM-implanted than sham implanted (Control) cows. COM treatment did not affect measured blood metabolites and enzymes, but increased hepatic GS protein expression by 350% after 14 d and 200% after 28 d of implantation. In contrast, protein expression of alanine transaminase, aspartate transaminase, glutamate dehydrogenase, and two glutamate transporters was not affected by COM. In Trial 2, plasma estradiol concentrations of COM implanted young and old cows were 48% higher than Control groups, whereas blood metabolites were not affected. COM implantation did not affect GS protein expression in young cows, but tended to increase GS expression in the old cows by 283% after 14 d and 41% after 28 d. GS mRNA content was increased about 38% in both young and old COM-treated cows. Hepatic content of beta-catenin and G protein-coupled receptor 30 (GPR30) content was not affected by COM treatment, indicating that estradiol-mediated GS expression was not regulated by beta-catenin- or GPR30-controlled pathways. In Trial 3, plasma estradiol levels in COM-treated heifers were 70% higher in COM heifers, concomitant with increased levels of total bilirubin and creatine kinase, and decreased creatinine. Correlation analysis of plasma estradiol levels and blood constituents only identified a positive correlation between plasma estradiol and potassium. Collectively, these data describe positive estradiol-mediated effects on hepatic metabolism and blood parameters in female cattle.

O ensaio de digestibilidade foi realizado nas depend?ncias do Laborat?rio de Pesquisa Animal da Universidade Federal de Lavras, sendo utilizadas18 borregas Santa In?s com 6 meses de idade e 29kg, confinadas em gaiolas de metabolismo por 21 dias, sendo 14 dias de adapta??o de 7 dias de coleta de fezes, urina, sobras e amostras de ra??o. Os tratamentos foram constitu?dos das ra??es citadas acima, e no pen?ltimo dia do per?odo experimental realizou-se a coleta de sangue dos animais para avalia??o de constituintes sangu?neos indicadores do metabolismo energ?tico, prot?ico e fun??o hep?tica. Os dados foram avaliados pelo procedimento GLM do SAS, e as m?dias comparadas e analisadas pelo teste de t a 5% de probabilidade. Houve superioridade no desempenho, consumo e digestibilidade dos animais que receberam dieta controle em rela??o aos animais que receberam gr?o de soja, sendo que a dieta com ?leo n?o diferiu da controle na maioria dos par?metros analisados. Os animais que receberam monensina apresentaram menores coeficientes de digestibilidade, menor consumo de todos os nutrientes analisados, menor desempenho e apresentaram ind?cios de problemas hep?ticos. Os machos tiveram desempenho e cortes mais pesados que as f?meas. N?o se recomenda o uso de monensina a 78 ppm em base da mat?ria seca para cordeiros em confinamento. Programa de P?s-Gradua??o em Zootecnia, Universidade Federal dos Vales do Jequitinhonha e Mucuri, 2010. / ABSTRACT There was conducted a study to evaluate the use of two fat sources and a control diet, combined with two monensin levels on lambs feed. The performance test was accomplished at a sheep industry sector of the Federal University of the Jequitinhonha and Mucuri Valleys in the Moura Experimental Farm, Curvelo-MG. The work evaluated 36 lambs, 18 males and 18 females, Santa In?s x Texel finished in feedlot and fed with six diets: control diet (RC), soybean oil (SO), soybean (GS), control with monensin addition (CR + M), soybean oil with monensin addition (OS + M) and soybean with monensin addition (GS + M). The animals were slaughtered after 87 days of confinement, being evaluated for performance, carcass characteristics, weights and percentages of carcass and not carcass components. The digestibility trial was conducted on the premises of the Animal Research Laboratory of the Lavras Federal University evaluating 18 Santa Ines lambs with 6 months of age and 29kg, confined in metabolism cages for 21 days, being 14 days for adaptation counting 7 days of collection of feces, urine, and samples of feed. The treatments consisted of the diets mentioned above, and one day before the end of the experiment there was collected animals blood sampling for blood constituents evaluation, which are indicators of energy metabolism, protein and liver function. The data were analyzed by GLM procedure of SAS and the average were compared and analyzed by t test for 5% of probability. There were higher in performance, intake and digestibility of pigs fed with the control diet compared to animals fed with soybean, and oil diet did not differ from controls in most parameters. The animals fed with monensin had lower digestibility, lower consumption of all nutrients, lower performance and showed signs of liver problems. Males had cuts and performance heavier than females. There are not recommended the monensin use at 78 ppm in dry matter basis for confined lambs. Soybean oil has not improved performance, intake or digestibility of diets and the soybean, added 15% in the dry matter, worsened this parameters.

Apresentam diversas atividades biológicas devido às suas propriedades antioxidantes e habilidades em modular a atividade de diversas enzimas ou receptores celulares, tornando-os responsáveis pelo efeito protetor contra doenças relacionadas ao sistema cardiovascular; certas formas de câncer e doenças de fotossensibilidade e envelhecimento. As fosfatases ácidas, enzimas que hidrolisam ésteres fosfatos em meio ácido, encontram-se amplamente distribuídas no organismo. Estas enzimas são importantes no catabolismo de diversas substâncias, acreditando-se que a alteração da atividade destas enzimas esteja relacionada com modificações induzidas por drogas e por várias doenças. As transaminases são enzimas hepáticas cujo nível aumenta quando há lesão das células hepáticas (hepatócitos) provocada por qualquer tipo de agressão, como vírus, consumo excessivo de álcool ou drogas. O acetaminofeno é uma droga frequentemente usada como analgésico e antipirético. O dano hepático causado pelo uso frequente ou exagerado do acetaminofeno é comum hoje em dia. A manutenção correta dos sistemas metabólicos hepáticos é de grande importância para a manutenção da saúde. Desta forma, o estudo do flavonóide quercetina como possível protetor dos efeitos hepatotóxicos provocados pelo acetaminofeno pode ser muito promissor. O objetivo do presente trabalho foi estudar os efeitos preventivos e terapêuticos, in vivo, do flavonóide quercetina sobre as atividades de fosfatases ácidas (total, tartarato-resistente e de baixa massa molecular relativa) e de transaminases glutâmica oxalacética (TGO) e glutâmica pirúvica (TGP) no fígado de camundongos, tratados ou não com acetaminofeno. Para o desenvolvimento deste trabalho foi realizado um tratamento agudo (24 horas), de camundongos machos da linhagem Swiss, com quercetina e acetaminofeno, utilizando-se óleo de milho ou nujol como veículo para o flavonóide. A dosagem da TGO e da TGP confirmou que o acetaminofeno pode realmente ser considerado hepatotóxico, quando administrado ou ingerido em grandes concentrações no organismo. A quercetina, dissolvida em óleo mineral, reverteu a hepatotoxicidade induzida pelo acetaminofeno, nos tratamento terapêutico e, em conjunto com acetaminofeno. A quercetina dissolvida em óleo de milho, no tratamento preventivo, pode não ser a única responsável pela reversão da hepatotoxicidade causada pela administração do acetaminofeno, uma vez que quando se substituiu o veículo utilizado, óleo de milho pelo óleo mineral, este efeito não foi mais observado. Comparando-se os resultados obtidos entre fosfatases e transaminases pode-se observar que a atividade da FATR, no tratamento com nujol, demosntra uma semelhança muito grande com os resultados observados nos gráficos das atividades das transaminases e provavelmente pode se considerada uma marcadora de dano hepático. O alfa-tocoferol, presente no óleo de milho, pode ter exercido um efeito antioxidante, e mascarado o efeito protetor da quercetina e a hepatotoxicidade do acetaminofeno. Este estudo foi importante para mostrar que a atividade dos flavonóides no organismo vivo pode ser influenciada por diversos fatores, como: a natureza do veículo utilizado na sua administração, ordem de administração e o tempo de permanecia no organismo / Abstract: Flavonoids are polyphenolic phytocompounds chemically characterized as flavonic heterosides. These compounds present several biological activities due to their antioxidant properties and hability to modulate the activities of enzymes or cellular receptors, making them responsible for the protector effect against diseases related to the cardiovascular system, certain forms of cancer, photosensibility diseases and aging. Acid phosphatases, enzymes that hydrolyze phosphate esters at acid medium, are largely distributed in the organisms. These enzymes are important in the catabolism of several compounds and could be related to the modifications induced by drugs and diseases. Transaminases are hepatic enzymes which levels increase in consequence of hepatic cells (hepatocytes) lesions provoked by agressions such as virus and excessive alcohol and drug consumption. Acetaminophen is a drug frequently used as analgesic and antipyretic. It is common the hepatic damage caused by the frequent or exaggerated use of acetaminophen. The correct maintenance of the hepatic metabolic systems is of great importance for the health. In this way, the study of the flavonoid quercetin as a possible protector of the hepatotoxic effects provoked by acetaminophen seems to be promising. The objectif of the present work was to study the in vivo effects of the flavonoid quercetin on the activities of acid phosphatases (total, tartrate-resistant and relative low molecular weight) and of transaminases glutamic oxalacetic (GOT) and glutamic pyruvic (GPT) in mice livers treated with acetaminophen. To develop this work, it was performed an acute treatment (24 hours) of Swiss male mice, with quercetin and acetaminophen, using corn oil or nujol as a vehicle for the flavonoid. The determination of GOT and GPT activities confirmed that acetaminophen can be considered hepatotoxic, when administered or ingested in great amount. In the therapeutic treatments, when applied with acetaminophen, quercetin, solubilized in mineral oil, reverted the acetaminophen-induced hepatotoxicity. In the preventive treatment, quercetin, solubilized in corn oil, might not be the only responsible for the reversibility, since this effect was no more observed when the vegetal oil was replaced by the mineral oil. The results with FATR, in the treatment with nujol, showed great similarity with the results obtained with transaminases, suggesting that this class of phosphatases could be considered as markers of hepatic damage. The alphatocopherol, present in the corn oil, could be exerting an antioxidant effect, masking the protector effect of quercetin and the acetaminophen hepatotoxicity.
