Obtenção e caracterização de concentrados e hidrolisados proteicos a partir do coproduto arroz quebrado : da extração alcalina aos processos de separação por membranasSouza, Daiana de January 2016 (has links)
O Brasil é um grande produtor de commodities alimentícias, e hoje o desenvolvimento da indústria de ingredientes para a geração de insumos de maior valor agregado a partir destas matérias-primas é uma área pouco explorada pelas indústrias brasileiras. O arroz se destaca no agronegócio brasileiro, já que o país figura como o nono maior produtor mundial. O arroz quebrado é um coproduto do beneficiamento do arroz branco polido. Este estudo teve como objetivo propor e avaliar um processo de obtenção de concentrados e hidrolisados proteicos de arroz, contribuindo na oferta de estratégias para o desenvolvimento tecnológico do setor no Brasil. Adicionalmente, deseja-se auxiliar na valoração do coproduto arroz quebrado, alimento de baixo valor comercial, base para a geração de matérias-primas já encontradas internacionalmente, e que apresentam grande potencial de desenvolvimento no mercado brasileiro. Dentro deste contexto, buscou-se estudar a aplicação da tecnologia de membranas neste processo, por ser uma tecnologia limpa, de baixo consumo energético, já bem estabelecida e com aplicações crescentes na indústria de alimentos. O estudo iniciou com a avaliação da eficácia de um método rápido de extração alcalina de amido e proteína de arroz, no qual foi utilizada farinha de arroz derivada de arroz quebrado como matéria-prima, ao invés dos grãos quebrados, como no método tradicional. Nesse estudo, a partir do uso de uma razão substrato:solvente de 1:15, a etapa de extração foi realizada em estágio único a 30 oC em apenas 30 minutos – um curto tempo de extração se comparado ao método alcalino tradicional (24-48h). O rendimento da extração proteica com estas condições foi bastante elevado, atingiu 81,4 %. Observou-se ser possível realizar uma extração eficiente, com a minimização das reações indesejadas causadas pelo meio alcalino. Na segunda etapa foram avaliadas as propriedades funcionais do concentrado proteico obtido a partir do método rápido de extração alcalina. Além disso, suas propriedades foram comparadas com as de dois concentrados proteicos comerciais produzidos por extração enzimática. Foi observado que todos os concentrados proteicos avaliados apresentaram pobres propriedades funcionais, devido sobretudo a suas baixas solubilidades em condições de pH próximas à neutralidade e levemente ácidas, concluindo-se ser necessário estudos que visem a melhoria da solubilidade destas proteínas quando se busca uma aplicação mais abrangente das mesmas como ingredientes. Os resultados obtidos na primeira etapa deste estudo motivaram inicialmente o emprego da microfiltração na separação das correntes de amido e proteína do extrato alcalino, analisando a possibilidade de se obter um amido com elevada pureza e uma corrente de permeado contendo a proteína extraída. A análise global dos resultados obtidos nesse trabalho permite que seja destacado que a escolha da microfiltração para o processo de separação do amido e da proteína de arroz deve ser avaliada com cautela. Na quarta etapa deste estudo avaliou-se a aplicação da hidrólise enzimática parcial do concentrado proteico na solubilização da proteína de endosperma de arroz. A hidrólise parcial, realizada com Alcalase®, permitiu a solubilização de 40 % da proteína originalmente presente na suspensão de proteína extraída pelo método alcalino. Os peptídeos solúveis gerados apresentaram massa molar média de 1,18 ± 0,33 kDa, solubilidade proteica superior a 88,5 % na faixa de pH de 3 a 7 e boa capacidade de formação de espuma, apesar de formarem uma espuma de baixa estabilidade. Neste estudo o hidrolisado proteico foi concentrado e purificado por ultrafiltração, o que permitiu a redução de 68 % do conteúdo de sódio do produto. Concluiu-se que neste estudo foi possível propor um processo produtivo que permitiu a obtenção de ingredientes com adequadas propriedades nutricionais e funcionais a partir do coproduto arroz quebrado. / The Brazil is a large producer of food commodities, and today the development of the industry of ingredients to add value to these raw materials is an area little explored by the Brazilian industries. Rice stands out in the Brazilian agribusiness, as the country is the ninth largest producer. The broken rice is a byproduct derived from the processing of polished rice. This study aimed to propose and evaluate a process of obtaining rice protein concentrates and hydrolysates contributing to offer strategies for the technological development of the sector in Brazil. Additionally, it aims to assist in the valuation of the byproduct broken rice, a low commercial value food which is the basis for the generation of raw materials already found internationally, and which have great potential in the Brazilian market. Within this context, it sought to study the application of membrane technology in this process, because it is a clean, low-energy, well-established technology, with growing applications in the food industry. The study began with the evaluation of the effectiveness of a fast alkaline extraction method for obtaining rice starch and protein, using rice flour derived from broken rice as raw material, instead of broken grains, as in the traditional method. In this study, with the use of a substrate:solvent ratio of 1:15, the extraction step was performed in a single stage at 30 °C in 30 minutes - a short extraction time compared to traditional alkaline method (24-48h). The yield of protein extraction with these conditions was very high, reached 81.4%. It was possible to perform an efficient extraction, with the minimization of undesired reactions caused by the alkaline medium. In the second stage of this study were evaluated the functional properties of the protein concentrate obtained by the fast alkaline extraction method. In addition, their properties were compared with two commercial concentrated protein produced by enzymatic extraction. It was observed that all the evaluated protein concentrates showed poor functional properties, mainly due to their low solubility at pH conditions close to neutrality and slightly acidic, concluding that it is necessary studies aimed to improve the solubility of these proteins in order to seek more applications of this ingredient. The results obtained in the first step of this study initially motivated the use of microfiltration to separate the starch and protein chains of the alkali extract, analyzing the possibility of obtaining a starch with high purity and a permeate stream containing the extracted protein. The global analysis of the results obtained in this work allows it to be noted that the choice of microfiltration for the separation process of starch and rice protein should be evaluated with caution. In the fourth stage of this study was evaluated the application of the partial enzymatic hydrolysis in solubilization of rice endosperm protein. The partial hydrolysis performed with Alcalase allowed solubilization of 40% of the protein originally present in the protein suspension extracted by the alkaline method. The generated soluble peptides had molecular weight average of 1.18 ± 0.33 kDa, protein solubility higher than 88.5 % in the pH range 3 to 7, and good foaming ability, while forming a low stable foam. In this study, the protein hydrolyzate was concentrated and purified by ultrafiltration, allowing 68 % reduction in the sodium content of the product. It is concluded that in this study was proposed a production process allowing obtaining ingredients with suitable nutritional and functional properties from the byproduct broken rice.

Previous issue date: 2013-05-23 / CAPES / The aim of this study was to evaluate the process of protein concentration in bovine whey proteins by ultrafiltration process and subsequently the protein hydrolysate obtained by enzymatic hydrolysis to produce bioactive peptides with potential antimicrobial and antioxidant activities. For concentration process was used a ceramic ultrafiltration membrane with a molecular range cut-off of 10-20 kDa, transmembrane pressure of 5 bar and, temperature of 30 ?C, 40 ?C and 50 ?C . The optimum temperature condition was at 40 ?C. The Volume Concentrate Factor (VCF) parameter was used as a end-point of the ultrafiltration process and fixed at 2, corresponding on concentrating the initial volume twice, in volume. At the temperature of 40 ?C, VCF had a correspondence on final protein concentration on the concentrated fraction by ultrafiltration and confirmed by Bradford method. Two commercial enzymes were tested Alcalase, Flavourzyme and an equivalent mixture of both 50:50 (w/w) in the hydrolysis reaction. The hydrolysis conditions were determined according to manufacturer instructions and confirmed by other studies: 60 ?C and pH 8 for Alcalase; 50 ?C and pH 7 for Flavourzyme; 50 ?C and pH 8 for enzyme mixture with enzyme / substrate ratio (w / w) 5/100 for all enzymes. The reaction was monitored by pH Stat method. The final Degree of Hydrolysis (DH) achieved was 15%, 52% and 63% for Flavourzyme, Alcalase and enzyme mixture, respectively. Five aliquots were collected along the hydrolysis for each enzyme reaction corresponding to differents DH in order to evaluatethe antioxidant activity by ORAC and ABTS assays with final values between 597- 1092 m? TE (ABTS) and from 1615 to 2920 ?M TE (ORAC) for Flavourzyme; 998-6290 ?M TE (ABTS) and 3092-7567 ?M TE ( ORAC) for Alcalase and finally 913-2678 ?M TE (ABTS) and 2547-5903 ?M TE (ORAC) for the enzyme mixture. The samples from all hydrolysates showed no antimicrobial activity against strains of Salmonella choleraesuis subsp. A rea??o de hidr?lise foi monitorada pelo m?todo pH Stat. Os Graus de Hidr?lise (GH) finais alcan?ados foram de 15 %, 52 % e 63 % para Flavourzyme, mistura enzim?tica e Alcalase, respectivamente. Foram coletadas cinco al?quotas correspondentes a diferentes GH ao longo da rea??o para cada condi??o enzim?tica utilizada e avaliadas quanto a atividade antioxidante pelos m?todos ABTS e ORAC tendo valores entre 597 a 1092 ?M TE (ABTS) e 1615 a 2920 ?M TE (ORAC) para Flavourzyme, 998 a 6290 ?M TE (ABTS) e 3092 a 7567 ?M TE (ORAC) para Alcalase e por fim, 913 a 2678 ?M TE (ABTS) e 2547 a 5903 ?M TE (ORAC) para a mistura enzim?tica. Nenhuma das amostras de hidrolisado com diferentes GH apresentou atividade antimicrobiana contra cepas de Salmonella choleraesuis subsp. Enteritidis (ATCC 13076) e Listeria monocytogenes (ATCC 9117).

Cette thèse a pour but d'améliorer notre compréhension des processus d'altération des roches mafiques silicatées, de la spéciation des éléments et de leur migration dans le milieu boréal (bassin versant de la mer Blanche, Nord-Ouest de la Russie). Les objectifs principales du travail sont i) de caractériser et décrire les mécanismes responsables de l'altération chimique et de la formation des minéraux dans la zone subarctique, ii) évaluer le rôle de la lithologie (environnement granitique versus basaltique) dans la spéciation des éléments traces (ET) et la formation des colloïdes organo-minéraux dans les eaux de surface des bassins versants boréaux des hautes latitudes, et iii) révéler la dépendance de la spéciation des ET en fonction du pH de la solution pour prédire des changements éventuels dans la bioaccumulation des éléments dans les eaux naturelles à cause de leur acidification. L'originalité de cette thèse est de combiner, pour la première fois sur les mêmes objets naturels, les techniques physico-chimique, minéralogique et isotopique afin de mieux comprendre les facteurs qui contrôlent les cycles biogéochimiques des éléments dans les régions subarctiques.

Effect of ionic strength on the efficiency of heavy metal removal and recovery from aqueous solutions via continuous mode polymer enhanced ultrafiltration (PEUF) method was examined. Application of PEUF to divalent ions of cadmium, nickel and zinc after their prior linking with polyethylenimine (PEI) results in complete removal of metal ions from single component aqueous solutions at high pHs.
Binding ability and hence the extent of metal retention in high ionic strength medium exhibits differences between solutions containing single and multicomponent metal mixtures. In single component metal solutions, extent of retention decreases but
binding order of metals remains unaffected both in low and high ionic strength medium. But, in binary component metal mixtures, with increase in ionic strength the binding order of metals changes. Fractional separation of Cd, Ni and Zn ions from equimolar binary and ternary mixtures of these metals and effect of ionic strength on fractional separation efficiency
were investigated. Depending on pH and salt concentration and metal pairs present in the solution fractional separation can be achieved.Dynamic and static light scattering experiments were performed in order to gain insight about the conformational changes in PEI structure due to the pH and ionic strength alternations in solution. It was found that, the increase in ionic strength reduces the size of the macromolecules.
A chemical equilibrium model was developed in order to estimate the apparent binding constants of metal-PEI complexes. Based on the data obtained from continuous and batch mode PEUF experiments apparent binding constants were estimated and compared to reveal the performance differences between these operational modes.

Le but de cette thèse est d'étudier l'influence des propriétés d'une suspension colloïdale sur l'efficacité du procédé d'ultrafiltration appliqué au traitement de l'eau. Pour cela, les expériences, détaillées dans une première partie, sont menées avec une suspension colloïdale d'argile contenant des électrolytes. Les propriétés physico-chimiques d'un tel mélange font l'objet du deuxième chapitre où la stabilité de la suspension est définie à travers la théorie DLVO, c'est à dire en considérant les interactions électrostatiques. Le troisième chapitre analyse l'influence de la stabilité de la suspension sur le colmatage de la membrane. Des modélisations permettent d'expliciter physiquement ces résultats en prenant en compte l'influence des interactions électrostatiques sur les mécanismes de transfert qui régissent l'ultrafiltration: la cinétique de dépôt et la résistance hydraulique du dépôt. L'étape de décolmatage de la membrane après les filtrations est exposée dans un quatrième chapitre. Une cinquième partie présente le rejet de sel engendré par le dépôt de colloïdes. Ce phénomène est décrit par les répulsions électrostatiques entre les ions et les surfaces chargées du colloïde. En conclusion, cette thèse présente donc plus généralement l'influence de la stabilité d'une suspension ou des interactions électrostatiques sur l'ensemble des mécanismes de transfert intervenant lors du colmatage d'une membrane d'ultrafiltration par un colloïde.

Impact de la matière organique sur le comportement des terres rares en solution: étude expérimentale et modélisationPourret, Olivier 06 October 2006 (has links) (PDF)
Les objectifs de cette thèse ont été principalement de comprendre l'impact de la matière organique sur le comportement des terres rares en solution et notamment: (i) de comprendre pourquoi le développement d'anomalie de cérium négative dans des eaux organiques oxydantes ne se produit pas; (ii) de tester la capacité des modèles à prédire la spéciation des terres rares dans des eaux organiques; (iii) d'évaluer les réactions compétitives de complexation des terres rares avec les acides humiques et les carbonates.<br />La première partie a été consacrée à deux études expérimentales de l'impact de la complexation organique sur le comportement redox du cérium et l'adsorption des terres rares à la surface d'oxydes de fer et de manganèse. La complexation des terres rares par la matière organique inhibe à la fois, l'oxydation du Ce(III) en Ce(IV) et le développement d'un effet tetrad visible sur les spectres de terres rares des eaux. Cette inhibition est due au développement d'un écran organique entre les terres rares et les surfaces oxydantes/adsorbantes que représentent les oxydes de fer et de manganèse.<br />La deuxième partie présente une évaluation de deux modèles, Model VI et SHM, quant à leur faculté à prédire la spéciation organique dans les eaux naturelles. La comparaison entre les résultats calculés et les expériences d'ultrafiltration a permis de valider Model VI dans sa capacité à prédire la spéciation des terres rares dans les eaux organiques. Par contre, la divergence entre les résultats obtenus avec SHM et les résultats expérimentaux indique que SHM, dans l'état actuel du programme, n'est pas apte à décrire précisément la spéciation des terres rares dans une eau naturelle. Dans un deuxième temps, cette étude a permis de fournir un jeu de constantes terres rares - acides humiques spécifiques à Model VI, fiables et cohérentes.<br />La troisième partie de ce travail a été consacrée aux réactions de compétitions entre ligands, acides humiques et carbonates, pour la complexation des terres rares. En fonction du pH et du rapport entre acide humique et carbonate, les terres rares vont se répartir entre ces deux ligands. La compétition entre les deux ligands induit également la formation d'une anomalie positive de cérium sur les acides humiques et négative dans la solution. Un nouveau mécanisme capable de développer sans surface oxydante ou bactérie, une anomalie négative de Ce dans les eaux naturelles a ainsi été mis en évidence.<br />La matière organique est responsable d'une forte complexation des terres rares dans les eaux naturelles. Une plus grande attention doit donc être portée à cette forme complexée pour étudier le transport des terres rares dans les eaux. Cette étude remet également en cause l'utilisation de l'anomalie de Ce comme témoin des conditions redox dans des eaux riches en matières organiques.

Evaluation of Membrane Filtration and UV Irradiation for the Control of Flavobacterium psychrophilum in Recirculation Aquaculture SystemsHuyben, David C. 07 January 2013 (has links)
Flavobacterium psychrophilum, the causative agent of bacterial cold water disease (BCWD), is tolerant of recommended ultraviolet (UV) doses used in recirculation aquaculture systems (RAS). Membrane filtration (MF) is used to remove pathogens from wastewater in many industries, but has not been thoroughly tested in RAS. In this study, bacterial removal efficiencies were assessed between MF and UV treatments over 30 days in an RAS. Bacterial removal efficiencies were not significantly different (p > 0.05) between MF and UV treatments, which removed 98.5 ± 0.4 % and 99.6 ± 0.1 % of total bacteria and 93.2 ± 5.2 % and 92.5 ± 4.1 % of heterotrophic bacteria, respectively. Under laboratory conditions, a MF system was challenged with concentrated doses of F. psychrophilum and achieved 5.8 ± 0.2 log reductions of the pathogen. Therefore, MF represents a potential alternative to UV irradiation and could be used to reduce the prevalence of F. psychrophilum in RAS, potentially reducing the incidence of BCWD and its impact on the aquaculture industry. / Environment Canada and Ontario Ministry of Agriculture, Food and Rural Affairs.

Effect of elevated temperature on ceramic ultrafiltration of colloidal suspensionsCromey, Tyler 22 May 2014 (has links)
The inherent thermal resistance of ceramic membranes allows for treatment of industrial waters at elevated temperatures. Traditionally, the high temperature of wastewater has been an issue compromising the integrity of polymeric membrane systems or requiring the temperature to be lowered for further treatments. In ceramic membrane systems, a decrease in viscosity with increasing temperature, however, can be utilized, which increases the permeate flux. In this study, the fouling of ceramic ultrafiltration by feed solutions containing colloidal silica was evaluated at temperatures between 25 – 90 °C seen in various industries. Ceramic membranes were able to perform well at elevated temperatures up to 90 ºC with sustained mechanical and chemical integrity. Results showed net benefit of filtration at elevated temperatures on permeate flux in spite of increasing total fouling resistance with temperature. When the temperature increased from 25 to 90 °C, there was a 90% increase in steady-state permeate flux. The dominant resistance was physically removable fouling, and the increase in fouling with feed temperature was supported by force balance analyses. This study provides a foundation from which further studies can be developed including pilot-scale testing, use of real wastewater, and the effects of operating conditions.

Les réactions de type hypersensibilité parfois observées au début de séances d'hémodialyse ont été corrélées à la présence, dans les bains de dialyse, d'endotoxines ou lipopolysacharides (LPS) provenant de bactéries de type gram négatif. Nous nous sommes tout d'abord attachés au développement d'un système de production de bains de dialyse stériles et apyrogènes qui soit intégrable au matériel existant. Notre étude nous a permis de rejeter les dispositifs d'ultrafiltration utilisant des membranes organiques planes ou des membranes minérales tubulaires. Nous avons proposé l'emploi d'un hémofiltre à fibres creuses en polyamide qui s'avère un bon compromis puisque, dans des conditions de contamination normales, le bain produit est stérile et apyrogène et la faible perte de charge générée par le filtre permet son intégration aux générateurs de bains existants. Nous avons également déterminé plus précisément la masse moléculaire des LPS par électrophorèse et par filtrations successives sur des membranes de seuils de coupure décroissants. Nous avons ainsi pu mettre en évidence les LPS après filtration sur des membranes de 5 kDa pour les LPS de type R. Dans une dernière étape, nous avons cherché à modifier la membrane de dialyse pour augmenter sa rejection des LPS. Un revêtement de la face dialysat de la membrane par un polymère organique polycationique nous a permis d'obtenir des absorptions notables de LPS. La conjonction des deux méthodes que nous avons proposées, ultrafiltration des bains de transfert des LPS vers le compartiment sanguin. Des essais cliniques nous permettront de valider ce procédé.
